In this paper, we study a new kind of generalized Caputo fractional differential quasi-variational inequalities in Hilbert spaces. We prove the uniqueness and the stability of the abstract inequality by using generalized singular Gronwall's lemma, projection operators, and contraction principle. The experience of a physician at a clinical center is among the critical factors in managing chronic myeloid leukemia (CML) during its treatment with tyrosine kinase inhibitors (TKIs). The authors conducted a cross-sectional questionnaire to investigate barriers to physician use of published evidence-based guidelines in CML management in a real-world setting. Among the participating physicians (N = 407), 99.8% of physicians reported that CML guidelines were useful; however, only 62.9% of physicians reported that they follow guidelines in real-time. Although 90.7% of physicians prefer second-generation TKIs as the first-line treatment, imatinib (88.2%) remains the most widely administered TKI in the first-line setting. Only 50.6% of physicians switched the treatment when patients failed to achieve early molecular response (at 3 months), whereas 70.3% of physicians switched the treatment when patients' response to TKI was inadequate at 6 months and/or 12 months. Moreover, only 43.5% of physicians considered treatment-free remission (TFR) as one of the top 3 goals for their patients. The major concern to obtain TFR was patients' adherence. This study demonstrated that CML management was generally in line with the current guidelines, but some of the details at the point of care are needed to be improved in CML. Leukemia stem cells (LSCs) constitute the critical barrier to the cure of acute myeloid leukemia (AML) due to their chemoresistance and immune evasion property. Herein, the role of anlotinib, a multiple tyrosine kinase inhibitor, in killing LSCs and regulating chemoresistance and immune evasion was explored. Anlotinib treatment induced apoptosis of LSC-like cells as well as primary AML LSCs, while sparing the normal mononuclear cells in vitro. Moreover, anlotinib could impair the regeneration capacity of LSCs in the patient-derived leukemia xenograft mouse model. Mechanistically, anlotinib inhibited phosphorylation of c-kit, JAK2/STAT3, and STAT5, and downregulated STAT3 and STAT5 expression. In addition, anlotinib downregulated the anti-apoptotic proteins Bcl-2 and Bcl-xL, and upregulated Bax, thereby enhancing the sensitivity of LSCs to idarubicin in vitro. Intriguingly, anlotinib could also partially rescue the interferon-g production of T cells cocultured with LSCs by downregulating PD-L1 expression. In conclusion, anlotinib showed anti-LSC activity and the potential to enhance the sensitivity to idarubicin and inhibit the immunosuppressive feature of LSCs via JAK2/STAT signaling pathway downregulation in the preclinical study. The aim of this paper is to investigate the topological properties of mild solution sets for a control system described by Hilfer fractional delay evolution equations with nonlocal initial conditions. We firstly obtain the nonemptiness, the compactness and R δ -property of the mild solution set by applying Schauder's fixed point theorem, a fixed point theorem of nonconvex valued maps and the fractional calculus. Then we apply this obtained result to show that the presented control problem has a reachable invariant set under nonlinear perturbations. Furthermore, we also apply the obtained results to characterize the approximate controllability of the presented control problem. In this paper, we introduce and study a new class of fractional differential hemivariational inequalities ((FDHVIs), for short) formulated by an initial-value fractional evolution inclusion and a hemivariational inequality in infinite Banach spaces. First, by applying measure of noncompactness, a fixed point theorem of a condensing multivalued map, we obtain the nonemptiness and compactness of the mild solution set for (FDHVIs). Further, we apply the obtained results to establish an existence theorem of the mild solution of a global attractor for the semiflow governed by a fractional differential hemivariational inequality ((FDHVI), for short). This article investigates optimal feedback controls of a system driven by fractional evolution hemivariational inequalities. An existence result with respect to feasible pairs is obtained by applying the Cesari property and a fixed‐point theorem for multivalued maps. Introduction Osmanthus fragrans flowers are used as folk medicine and additives for teas, beverages and foods. The metabolites of O. fragrans flowers from different geographical origins were inconsistent in some extent. Chromatography and mass spectrometry combined with multivariable analysis methods provides an approach for discriminating the origin of O. fragrans flowers. Objective To discriminate the Osmanthus fragrans var. thunbergii flowers from different origins with the identified metabolites. Methods GC-MS and UPLC-PDA were conducted to analyse the metabolites in O. fragrans var. thunbergii flowers (in total 150 samples). Principal component analysis (PCA), soft independent modelling of class analogy analysis (SIMCA) and random forest (RF) analysis were applied to group the GC-MS and UPLC-PDA data. Results GC-MS identified 32 compounds common to all samples while UPLC-PDA/QTOF-MS identified 16 common compounds. PCA of the UPLC-PDA data generated a better clustering than PCA of the GC-MS data. Ten metabolites (six from GC-MS and four from UPLC-PDA) were selected as effective compounds for discrimination by PCA loadings. SIMCA and RF analysis were used to build classification models, and the RF model, based on the four effective compounds (caffeic acid derivative, acteoside, ligustroside and compound 15), yielded better results with the classification rate of 100% in the calibration set and 97.8% in the prediction set. • An UHPLC-MS/MS method was developed to determine OPN in milk. • A consensus signature peptide was selected as the representative of OPN from different species' milk. • Stable isotope dimethyl labeling was used to prepare internal standard during determination of OPN. • HLB cartridge was applied to purify and enrich OPN signature peptide. • The method was able to measure OPN content in bovine, buffalo, yak, goat and sheep milk. Osteopontin (OPN) is a multifunctional protein present in different tissues, body fluids and milk. Different milk has different level of OPN content. To determine the amount of osteopontin in bovine, buffalo, yak, sheep and goat milk, we developed an ultra-high performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS) method to detect an osteopontin signature peptide. The signature peptides selected by searching Uniprot database for trypsin digested osteopontin. The sample preparation procedure includes trypsin digestion, dimethyl labeling of tryptic peptides, purification and concentration of labeled tryptic peptide with solid phase extraction. The limit of detection and limit of quantification are 0.5 mg L−1 and 2.0 mg L−1, respectively. The method has satisfactory analytical performance with a linearity of R 2 ≥ 0.998, recoveries of 103.7–111.0%, and precision of 1.8–6.2%. Ethnopharmacological relevance Osmanthus fragrans var. thunbergii ( O. fragrans ) flower has been consumed as folk medicine for thousands of years. O. fragrans flower extract is a well-characterized phenylethanoid glycoside-rich extract, which has been used as a natural anti-oxidant. The aim of this study was to evaluate the safety of O. fragrans flower phenylethanoid glycoside-rich extract (OFFE). Materials and methods The OFFE was extracted by 80% (v/v) aqueous ethanol with 0.01% sodium isoascorbate (w/v) from the O. fragrans flower and purified on HPD300 resins. The total phenylethanoid glycosides content and individual phenylethanoid glycosides was determined by photocolorimetric method and reversed phase UPLC respectively. An acute oral toxicity study, reverse mutation test, bone marrow cell micronucleus test, and sperm abnormality test as well as a 90-day oral toxicity study were performed on experimental animals. Results The total content of phenylethanoid glycosides in OFFE was 73.4 g acteoside equivalent per 100 g of extract, include acteoside (52.5 g per 100 g of extract), salidroside (13.8 g per 100 g of extract), and isoacteoside (2.6 g per 100 g of extract) and so on. No acute lethal effect at the maximal tested OFFE dose of 10 g/kg body weight (bw) in either rats or mice was observed, suggesting that OFFE can be considered nontoxic. No evidence for mutagenicity was detected in any of the three mutagenic tests. Administration at levels of 0.50, 1.00, and 2.00 g/kg bw to rats for 90 days failed to induce any significant hematological, clinical, chemical, or histopathological changes. The no-observed adverse-effect-level for OFFE was >2.00 g/kg bw for the study on subchronic toxicity. Conclusion The results showed that consuming OFFE has no adverse effects and poses no health risk in the acute oral toxicity study, subchronic oral toxicity study, and in the micronucleus test, which may provide supportive evidence for the safety of OFFE powder that has been used in medicine as well as in functional foods, and dietary supplements.
