OBJECTIVE: Sepsis has a high morbidity and mortality and is prone to cause acute kidney injury (AKI). Here, we aimed to demonstrate the function and molecular mechanism of microRNA-543 (miR-543) in septic AKI. MATERIALS AND METHODS: MiR-543 inhibitor or NC was transfected into LPS-treated HK-2 cells to observe lipopolysaccharide (LPS)-induced inflammation and apoptosis. The detection of inflammation and apoptosis of HK-2 cells relies on Western blot, quantitative Reverse-Transcription Polymerase Chain Reaction (qRT-PCR), enzyme-linked immunosorbent assay (ELISA), Cell Counting Kit-8 (CCK-8) assay, flow cytometry, and terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick end labeling (TUNEL) staining. RESULTS: MiR-543 expression was increased in LPS-treated HK-2 cells. By transfecting miR-543 inhibitor into HK-2 cells, miR-543 expression was dramatically reduced. The downregulation of miR-543 remarkably inhibited the inflammation and apoptosis, which was manifested by the reduction of inflammatory cytokines (TNF-a, IL-6, IL-1ß), the reversal of apoptosis-related proteins expression (Bcl-1, Bax), the increase of cell viability and the decrease of the proportion of apoptotic cells. The result of Luciferase activity assay demonstrated that miR-543 directly targets Bcl-2. CONCLUSIONS: MiR-543 expression was increased in LPS-treated HK-2 cells, and silencing miR-543 could inhibit LPS-induced inflammation and apoptosis in HK-2 cells via targeting Bcl-2. Electromagnetic shielding fabric (ESF) is a novel electromagnetic shielding product with portability, flexibility, and good mechanical properties. However, the existing ESFs suffer from poor washing fastness of coating and poor comfort performance in terms of hand-feel characteristic. In this study, a core layer-coated yarn (CLCY) was successfully spun using a carboxylic acid-functionalized multi-walled carbon nanotube/polypyrrole/Fe3O4 composite suspension with polyvinyl butyral as an adhesive agent. To better explore the properties, the original fabric and treated fabrics, viz., core layer-coated fabric (CLCF) and surface layer-coated fabric (SLCF), were characterized by several methods. Scanning electron microscopic observations revealed that the coating was on the core layer in CLCF. In addition, the Fouriertransform infrared spectroscopy and X-ray diffraction spectroscopy results revealed that the composition of the coating corresponds with that in the multi-composite suspension. Moreover, the coating of CLCY formed a conductive path with good conductivity in the core layer, but the conductivity of the coating on the surface layer of SLCF deteriorated sharply after washing. Further, compared with the original fabric and SLCF, CLCF has highest breaking strength (after 10 washes), and keeps a relatively good hand-feel characteristic. Finally, the evaluated electromagnetic interference shielding characteristics reveal that the fastness of coating affects the electromagnetic shielding effectiveness, suggesting that the wrapped protection of outside staple fibers in CLCF reduce the loss of coating in the core layer during washing. Ultraviolet upconversion fluorescence band (260–350 nm) has been observed from Pr3+:Y2SiO5 pumped by Ar+ ion laser (488 nm). Power dependence of the fluorescence emitted from 4f5d, 3P0 and 1D2 were measured. The upconversion mechanism was analyzed using the rate equations with a simplified three level model. It appears that excited state absorption (ESA) is the dominant upconversion process for lower Pr3+ concentration and energy transfer upconversion (ETU) is dominant for higher Pr3+ concentration. High power laser interactions with metal targets are of great theoretical significance and are widely encountered in engineering applications. However, there are few efforts to investigate the liquid cooling effect when the metal target is in contact with a liquid, as is usually the case of the fuel tank of a missile. To study the flow physics and heat transfer characteristics during the interaction, a 3-D unsteady conjugate conduction-natural convection model is developed in this paper and an experiment is also performed to validate the model. The simulation results, using the parameters of the experiment, show that the heat exchange takes place mainly in the vicinity of the metal/liquid interface. At the initial stage of laser irradiation, the natural convection effect is not remarkable and the heat transfer intensity is weak. After that, the heat transfer coefficient increases gradually and approaches to a constant value. Moreover, the temperature variation of the metal plate and the velocity field variation of the liquid are shown to be in well agreement with the ones obtained by experiment, which validates the numerical model. Objective Naturally acquired anti–hepatitis E virus (HEV) immunity can protect against new HEV infections. The aim of this study was to analyse the persistence of naturally acquired anti-HEV immunoglobulin (Ig) G and anti-HEV IgG concentrations after vaccination. Methods We examined the seropositivity rates of participants included in a phase 3 clinical efficacy trial (67 months' follow-up) for a HEV vaccine (Hecolin; Xiamen Innovax Biotech, China) and predicted long-term persistence using mixed-effect models. Results The analysis focused on 2242 baseline seropositive participants in a control group (placebo recipients) and 2031 baseline seropositive participants in an vaccine group (vaccine recipients) who received 1 to 3 doses of Hecolin. Naturally acquired anti-HEV IgG levels decreased steadily independent of the initial antibody level; 50% of the placebo recipients were expected to have undetectable antibody concentrations after 14.5 years. After immunization with Hecolin, the power-law model and the modified power-law model predicted that 82.1 and 99.4% of the participants, respectively, would remain seropositive for anti-HEV IgG for 30 years after vaccination. Conclusions Whereas naturally acquired anti-HEV IgG levels decrease steadily, HEV vaccination induces long-lasting, high-level anti-HEV IgG concentrations. [ABSTRACT FROM AUTHOR]
