The accuracy of dynamic-force spectroscopy (DFS), a promising technique of analyzing the energy landscape of noncovalent molecular bonds, was reconsidered in order to justify the use of an atomic-force microscopy (AFM) cantilever as a DFS force probe. The advantages and disadvantages caused, for example, by the force-probe hardness were clarified, revealing the pivotal role of the molecular linkage between the force probe and the molecular bonds. Room-temperature ionic liquids (RTILs) are a vast class of organic non-aqueous electrolytes whose interaction with biomolecules is receiving great attention for potential applications in bio-nano-technology. Recently, it has been shown that RTILs dispersed at low concentrations at the water-biomembrane interface diffuse into the lipid region of the biomembrane, without disrupting the integrity of the bilayer structure. In this letter, we present the first exploratory study on the effect of absorbed RTILs on the mechanoelasticity of a model biomembrane. Using atomic force microscopy, we found that both the rupture force and the elastic modulus increase upon the insertion of RTILs into the biomembrane. This preliminary result points to the potential use of RTILs to control the mechanoelasticity of cell membranes, opening new avenues for applications in bio-medicine and, more generally, bio-nano-technology. Dry adhesion is a common strategy utilized throughout nature, allowing attachment of animals to a large range of surfaces. Crucially, it enables the animal to switch rapidly and safely between attachment and detachment in order to facilitate motion. Here we investigate the magnitude and directionality of the dry adhesion systems of a number of spider species. Atomic force microscopy (AFM) was used to measure the normal adhesion strength of a single setule. Secondly, scanning electron microscopy (SEM) was used to directly observe the interaction of a spider seta with an AFM cantilever tip and then, based on the known stiffness of the cantilever and the deflection distance, to estimate the adhesion force between them. The AFM-measured values for normal adhesion of single spider setules to a surface (9–16 nN) were similar to values predicted by the Johnson–Kendall–Roberts (JKR) model. In contrast, the force of adhesion, estimated from the SEM images, between the seta and the cantilever reached values as high as 10 μN, much higher than predicted by the JKR theory alone. With spiders, as with geckos, the principle of contact splitting leads to an increase in adhesion force. In addition, it was observed that significantly higher adhesion occurred when the setules were dragged laterally across a spherical surface in the proximal direction (i.e. towards the spider's tarsus). The fluid mosaic model of biological membranes is that of a two-dimensional lipid bilayer in which both lipids and associated membrane proteins diffuse freely. More recently, the raft hypothesis proposed that membranes contain small, dynamic, functional domains (rafts), which act as platforms for membrane protein attachment and interaction. Although experimental evidence supporting the raft hypothesis is growing, very little is known of the structure of the membrane-fluid interface of lipid raft systems. Here, we report the direct submolecular-scale imaging of model raft membranes using ultrahigh resolution atomic force microscopy. We characterize the heterogeneous nature of crystalline hydration layers at the membrane-fluid interface. The association of crystalline hydration layers with raft membranes would significantly affect the mechanism and kinetics of both inter-raft interactions and those between rafts and external biomolecules, and therefore this finding has important implications for membrane biology. The composition and nanoscale mechanical characteristics of the adhesive from two species of subaerial green unicellular microalgae (Chlorophyta), Coccomyxa sp. and Glaphyrella trebouxiodes, have been studied using Raman spectroscopy, chemical staining, and atomic force microscopy (AFM). Raman spectroscopy confirmed the adhesive proteins of both species to be predominantly in ss-sheet conformations and composed of a number of hydrophobic amino acid residues. Chemical staining with Congo red and thioflavin-T dyes further confirmed the presence of amyloid-like structures. Probing the adhesives with AFM revealed highly ordered and repetitive mechanical responses indicative of highly ordered structures within the adhesive. The repetitive nature of the sawtooth response is typical of a “sacrificial bond” and “hidden length” mechanism, and what we -propose is the result of mechanical manipulation of individual molecules within an intermolecular ss-sheet that makes up the generic amyloid structure. The mechanical data show how amyloid provides cohesive strength to the adhesives, and this intrinsic mechanical property of an amyloid-based adhesive explains the ecological success of attachment of these subaerial microalgae on various surfaces in urban environments. It is unknown to what extent amyloid fibrils occur in algal adhesives, but we postulate that the amyloid structure could provide a widespread mechanism for mechanical strength. We have developed a dynamic force microscope (DFM) working in a novel operation mode which is referred to as phase modulation atomic force microscopy (PM-AFM). PM-AFM utilizes a fixed-frequency excitation signal to drive a cantilever, which ensures stable imaging even with occasional tip crash and adhesion to the surface. The tip-sample interaction force is detected as a change of the phase difference between the cantilever deflection and excitation signals and hence the time response is not influenced by the Q factor of the cantilever. These features make PM-AFM more suitable for high-speed imaging than existing DFM techniques such as amplitude modulation and frequency modulation atomic force microscopies. Here we present the basic principle of PM-AFM and the theoretical limit of its performance. The design of the developed PM-AFM is described and its theoretically limited noise performance is demonstrated. Finally, we demonstrate the true atomic resolution imaging capability of the developed PM-AFM by imaging atomic-scale features of mica in water. [ABSTRACT FROM AUTHOR]

We have developed a liquid-environment frequency modulation atomic force microscope (FM-AFM) with a low noise deflection sensor for a wide range of cantilevers with different dimensions. A simple yet accurate equation describing the theoretical limit of the optical beam deflection method in air and liquid is presented. Based on the equation, we have designed a low noise deflection sensor. Replaceable microscope objective lenses are utilized for providing a high magnification optical view (resolution: <3 μm) as well as for focusing a laser beam (laser spot size: ∼10 μm). Even for a broad range of cantilevers with lengths from 35 to 125 μm, the sensor provides deflection noise densities of less than 11 fm/√Hz in air and 16 fm/√Hz in water. In particular, a cantilever with a length of 50 μm gives the minimum deflection noise density of 5.7 fm/√Hz in air and 7.3 fm/√Hz in water. The measurement of short-range forces with the atomic force microscope (AFM) typically requires implementation of dynamic techniques to maintain sensitivity and stability. While frequency modulation atomic force microscopy (FM-AFM) is used widely for high-resolution imaging and quantitative force measurements in vacuum, quantitative force measurements using FM-AFM in liquids have proven elusive. Here we demonstrate that the formalism derived for operation in vacuum can also be used in liquids, provided certain modifications are implemented. To facilitate comparison with previous measurements taken using surface forces apparatus, we choose a model system (octamethylcyclotetrasiloxane) that is known to exhibit short-ranged structural ordering when confined between two surfaces. Force measurements obtained are found to be in excellent agreement with previously reported results. Frequency modulation atomic force microscopy utilizes the change in resonant frequency of a cantilever to detect variations in the interaction force between cantilever tip and sample. While a simple relation exists enabling the frequency shift to be determined for a given force law, the required complementary inverse relation does not exist for arbitrary oscillation amplitudes of the cantilever. In this letter we address this problem and present simple yet accurate formulas that enable the interaction force and energy to be determined directly from the measured frequency shift. These formulas are valid for any oscillation amplitude and interaction force, and are therefore of widespread applicability in frequency modulation dynamic force spectroscopy. © 2004 American Institute of Physics. A modified atomic force microscopy (AFM) system, based on a force modulation technique, has been used to find an approximate value for the elastic modulus of a single peptide molecule directly from a mechanical test. For this purpose a self-assembled monolayer built from two kinds of peptides, reactive (able to anchor to the AFM tip) and nonreactive, was synthesized. In a typical experiment a single C[sub 3]K[sub 30]C (C=cysteine, K=lysine) peptide molecule was stretched between a Au(111) substrate and the gold-coated tip of an AFM cantilever to which it was attached via gold–sulfur bonds. The amplitude of the cantilever oscillations, due to an external force applied via a magnetic particle to the cantilever, was recorded by a lock-in amplifier and recalculated into stiffness of the stretched molecule. A longitudinal Young’s modulus for the α-helix of a single peptide molecule and for the elongated state of this molecule has been estimated. The obtained values; 1.2±0.3 and 50±15 GPa, for the peptide α-helix and elongated peptide backbone, respectively, seem to be reasonable comparing them to the Young’s modulus of protein crystals and linear organic polymers. We believe this research opens up a means by which scientists can perform quantitative studies of the elastic properties of single molecule, especially of biologically important polymers like peptides or DNA. © 2001 American Institute of Physics. Atomic force microscopy (AFM) is widely used in liquid environments, where true atomic resolution at the solid–liquid interface can now be routinely achieved. It is generally expected that AFM operation in more viscous environments results in an increased noise contribution from the thermal motion of the cantilever, thereby reducing the signal-to-noise ratio (SNR). Thus, viscous fluids such as ionic and organic liquids have been generally avoided for high-resolution AFM studies despite their relevance to, e.g. energy applications. Here, we investigate the thermal noise limitations of dynamic AFM operation in both low and high viscosity environments theoretically, deriving expressions for the amplitude, phase and frequency noise resulting from the thermal motion of the cantilever, thereby defining the performance limits of amplitude modulation, phase modulation and frequency modulation AFM. We show that the assumption of a reduced SNR in viscous environments is not inherent to the technique and demonstrate that SNR values comparable to ultra-high vacuum systems can be obtained in high viscosity environments under certain conditions. Finally, we have obtained true atomic resolution images of highly ordered pyrolytic graphite and mica surfaces, thus revealing the potential of high-resolution imaging in high viscosity environments.
