ABSTRACT: This study investigated the effects of the early administration of enrofloxacin (E) or doxycycline (D) for the first 5 consecutive days of life, or the continuous administration of the coccidiostat monensin (M) throughout the rearing period on gastrointestinal function in turkeys infected with avian pathogenic Escherichia coli (APEC) in an early or later stage of rearing. Experiment 1 lasted 21 d, and turkeys in groups E, D, and M were infected with APEC on d 15. Experiment 2 lasted 56 d, and it had a factorial arrangement of treatments where birds in groups E, D, and M were infected with APEC on d 15 or d 50. In both experiments, control groups (C) consisted of infected and uninfected birds without antibiotic or coccidiostat administration. On d 21 (Experiment 1) and d 56 (Experiment 2), 8 birds from each subgroup were killed, and the ileal and cecal digesta were sampled to analyze the activity of bacterial enzymes and the concentrations of short-chain fatty acids (SCFA). The experimental treatments did not affect the final body weight or body weight gain of birds. Both experiments demonstrated that APEC contributed to an increase in ammonia levels of the cecal digesta (means from 2 experiments: 0.311 vs. 0.225 mg/g in uninfected birds) and ileal pH (6.79 vs. 6.00) and viscosity (2.43 vs. 1.83 mPa⋅s). Moreover, the E. coli challenge enhanced the extracellular activity of several cecal bacterial enzymes, especially in older turkeys infected with APEC in a later stage of life. The continuous administration of monensin throughout the rearing period resulted in a weaker gastrointestinal response in older birds, compared with the other 2 antibiotics administered for the first 5 d of life. The results of the study are inconclusive as both desirable and undesirable effects of preventive early short-term antibiotic therapy were observed in turkeys, including normalization of ileal viscosity and cecal ammonia concentration (positive effect), and disruption in cecal SCFA production (negative effect).

The aim of this review is to present and discuss the most recent literature about the processing of insect biomass and its impact on nutritive value, further implementation of meals and fats derived from invertebrates to livestock (poultry and swine), aquaculture (salmonids), and companion animal diets and their impact on growth performance, metabolic response, and gastrointestinal microbiota shifts. Additionally, the most important barriers to obtaining unified products in terms of their nutritive value are considered, i.e., to define insects' nutrient requirements, including various technological groups and further biomass processing (slaughtering, drying, and storage). Due to the current limitation in the insect production process consisting of the lack of infrastructure, there is stress on the relatively small amount of insect products added to the animal diets as a functional feed additive. Currently, only in the case of pet nutrition may insects be considered a full replacement for commonly used environmentally harmful and allergenic products. Simultaneously, the least information has been published on this topic. Thus, more scientific data are needed, particularly when the pet food branch and insect-based diets are rapidly growing.

ABSTRACT: It was assumed that early administration of enrofloxacin or doxycycline may impair immune function and alter the morphology of organs of the immune system in turkeys, and that diets containing the coccidiostat monensin, an ionophore antibiotic, can exert similar effects. The aim of this study was to determine whether early antibiotic administration or feeding a diet containing a coccidiostat affect immune function in young turkeys. The experiment had a completely randomized design, with 8 groups (a total of 3,080 one-day-old turkeys), 7 replicate pens per group and 55 birds per pen. The experiment had a 2-factorial design, with 4 treatments (C—control, M—monensin, E—enrofloxacin, and D—doxycycline) and 2 groups of birds (vaccinated and unvaccinated) per treatment. Control group birds did not receive the coccidiostat or antibiotics. Group M was administered monensin at 90 mg/kg feed for the first 5 d of life, group E received enrofloxacin at 10 mg/kg BW, added to drinking water, for the first 5 d of life, and group D received doxycycline at 50 mg/kg BW, added to drinking water, for the first 5 d of life. One-day old turkeys from groups C+, M+, E+, and D+ were administered live-attenuated vaccines against turkey rhinotracheitis (TRT) (Poulvac TRT; Zoetis, Parsippany, NJ) and Newcastle disease (ND) (Nobilis ND clone 30; Merck, Rahway, NJ) by coarse spray; 28-day-old birds were administered a subcutaneously injected inactivated vaccine against Ornithobacterium rhinotracheale (ORT) (Ornitin, Phibro, Poland). Turkeys from groups C−, M−, E−, and D− were not vaccinated. It was found that early administration of enrofloxacin or doxycycline, or feeding a diet containing monensin, did not weaken the immune system of turkeys. The administration of monensin, in particular when combined with vaccination, was least effective in inhibiting inflammatory responses. Histological changes in immunocompetent organs (fatty degeneration) were also most severe in birds receiving monensin, followed by those administered doxycycline and enrofloxacin. The observed changes were exacerbated by vaccination.

The effects of the different dietary levels of Arginine (Arg) in low- and high-methionine (Met) diets on the meat quality and myofibrillar protein profile of breast muscles from turkeys were determined. The experiment had a completely randomized 3 × 2 factorial design with three levels of Arg (90%, 100%, and 110%) relative to the dietary Met levels (30% or 45%). At 42 days of age, eight turkeys from each treatment were sacrificed; the meat pH value was measured at 48 h post-slaughter, and meat color was measured according to the CIE L*a*b* system. The SDS-PAGE method was performed to investigate the myofibrillar protein profile of the breast muscle. The analysis of variance showed a significant effect of the Arg or Met dietary levels on the color parameter b* and the profile of myofibrillar proteins in muscles. The results of the cluster analysis of the myofibrillar protein profile showed that, with a high level of Arg (i.e., 110%), the level of Met 35% or 45% was less important. It can be concluded that the increase in the share of Arg and Met in the diet of turkeys increases the content of some myofibrillar proteins (actinin, desmin, actin) and reduces degradation during the post-slaughter proteolysis of proteins that are considered tenderization indicators.

This study investigated the effects of dietary replacement of soybean meal (SBM) with graded levels of pea seeds (PS) on the gastrointestinal function of turkeys. Seeds of 2 pea varieties, a colored-flowered variety and a white-flowered variety (CFP and WFP, respectively) were fed to 56-d-old birds for 8 wk. A total of 539 female Hybrid turkeys were allocated to 7 groups, each group consisted of 7 pens with 11 birds per pen. The experiment had a 2-factorial design, with 3 dietary inclusion levels of PS (100, 200 and 300 g/kg) and 2 pea varieties (CFP and WFP). The control group (diets without PS) was compared with CFP and WFP treatments by simple contrast analysis. In comparison with CFP seeds, WFP seeds contained 7-fold less tannins (0.67 vs. 4.66 g/kg) and less non-starch polysaccharides (NSP, 117.8 vs. 132.7 g/kg), but more trypsin inhibitors (1.34 vs. 0.98 g/kg) and starch (489 vs. 455 g/kg). A rise in the PS content of diets from 100 to 200 and 300 g/kg increased the weight of the small intestine (P = 0.031) and the dry matter (DM) content of intestinal digesta (P = 0.001), but it had no effect on the pH of digesta. Only the highest PS content differentiated the concentrations of short-chain fatty acids (SCFAs) in the small intestinal digesta (WFP > CFP, P = 0.008), whereas PS did not cause any changes in the morphological parameters of the small intestinal mucosa. The dietary inclusion of PS had no influence on the levels of acetate, butyrate, putrefactive SCFAs or total SCFAs in the cecal contents. Apart from increasing the activities of β-glucosidase (P = 0.017) and β-galactosidase (P = 0.025), pea varieties did not affect the activities of the analyzed cecal microbial enzymes. However, CFP seeds decreased the DM content (P = 0.041) and increased the pH of cecal digesta, compared with WFP seeds (P = 0.013). The results of this study, pointing to a few differences in the functional parameters of the small intestine and cecum, indicate that tannins are not a factor differentiating the suitability of CFP and WFP seeds in the nutrition of finisher turkeys. The inclusion of PS at 200 and 300 g/kg of the diet reduces the content of SBM and wheat in turkey diets, which has a positive effect on gastrointestinal function.

Abstract Arginine (Arg), lysine (Lys), and methionine (Met) can be used to support the health status of turkeys. The present study investigated selected performance, gut integrity, and immunological parameters in turkeys reared in optimal or challenge conditions. The experiment lasted for 28 days, and it had a completely randomized 2 × 3 factorial design with two levels of dietary Arg, Lys and Met (high or low) and challenge with Clostridium perfringens (C. perfringens), Escherichia coli lipopolysaccharide (LPS) or no challenge (placebo). Increased dietary levels of Arg, Lys and Met had a beneficial effect on turkey performance and immunological parameters, and it improved selected indicators responsible for maintaining gut integrity in different challenge conditions. Under optimal conditions (with no challenge), high ArgLysMet diets did not compromise bird performance and they improved selected performance parameters in challenged birds. The immune system of turkeys was not excessively stimulated by high ArgLysMet diets, which did not disrupt the redox balance and had no negative effect on gut integrity. High ArgLysMet diets increased the expression levels of selected genes encoding nutrient transporters and tight junction proteins. However, the influence exerted by different dietary inclusion levels of Arg, Lys and Met on gut integrity was largely determined by the stressor (C. perfringens vs. LPS). Further studies are required to investigate the role of Arg, Lys and Met levels in the diet on the immune response, gut function and performance of turkeys in different challenge conditions.

ABSTRACT: The tissue-specific profile of alternatively spliced genes (ASGs) and their involvement in reproduction processes characteristic of turkey testis, epididymis, and ductus deferens were investigated for the first time in birds. Deep sequencing of male turkey reproductive tissue RNA samples (n = 6) was performed using Illumina RNA-Seq with 2 independent methods, rMATs and SUPPA2, for differential alternative splicing (DAS) event prediction. The expression of selected ASGs was validated using quantitative real-time reverse transcriptase-polymerase chain reaction. The testis was found to be the site of the highest number of posttranscriptional splicing events within the reproductive tract, and skipping exons were the most frequently occurring class of alternative splicing (AS) among the reproductive tract. Statistical analysis revealed 86, 229, and 6 DAS events in the testis/epididymis, testis/ductus deferens, and epididymis/ductus deferens comparison, respectively. Alternative splicing was found to be a mechanism of gene expression regulation within the turkey reproduction tract. In testis, modification was observed for spermatogenesis specific genes; the changes in 5’ UTR could act as regulator of MEIG1 expression (a player during spermatocytes meiosis), and modification of 3’ UTR led to diversification of CREM mRNA (modulator of gene expression related to the structuring of mature spermatozoa). Sperm tail formation can be regulated by changes in the 5’ UTR of testicular SLC9A3R1 and gene silencing by producing dysfunctional variants of ODF2 in the testis and ATP1B3 in the epididymis. Predicted differentially ASGs in the turkey reproductive tract seem to be involved in the regulation of spermatogenesis, including acrosome formation and sperm tail formation and binding of sperm to the zona pellucida. Several ASGs were classified as cilia by actin and microtubule cytoskeleton organization. Such genes may play a role in the organization of sperm flagellum and post-testicular motility development. To our knowledge, this is the first functional investigation of alternatively spliced genes associated with tissue-specific processes in the turkey reproductive tract.

ABSTRACT: In this study, the effects of the Arginine/Lysine (Arg/Lys) ratio in low- and high-methionine (Met) diets on the sarcoplasmic protein profile of breast muscles from turkeys reared under optimal or challenge (Clostridium perfringens infection) conditions were determined. One-day-old Hybrid Converter female turkey poults (216 in total) obtained from a commercial hatchery on hatching day, and on the basis of their average initial body weight were randomly allocated to 12 pens (4 m2 each; 2.0 m × 2.0 m) containing litter bedding and were reared over a 42-day experimental period. Diets with high levels of Lys contained approximately 1.80% and 1.65% Lys and were offered in two successive feeding periods (days 1–28 and days 29–42). The supplemental levels of Lys were consistent with the nutritional specifications for birds at their respective ages as established in the Management Guidelines for Raising Commercial Turkeys. The experiment was based on a completely randomized 3 × 2 × 2 factorial design with three levels of Arg (90%, 100% and 110%) relative to the content of dietary Met (30 or 45%) and without (−) or with (+) C. perfringens challenge at 34, 36, or 37 d of age. Meat samples were investigated in terms of pH, color, and sarcoplasmic protein profile. The experimental factors did not influence meat quality but the dietary Arg content affected meat color. The sarcoplasmic protein profile was influenced by all studied factors, and glycolytic enzymes were the most abundant. This study evidenced strong association between the challenge conditions and the involvement of glycolytic enzymes in cell metabolism, particularly in inflammatory processes, and DNA replication and maintenance in turkeys. The results showed an effect of C. perfringens infection and feeding with different doses of Arg and Met may lead to significant consequences in cell metabolism.

ABSTRACT: This study investigated the effects of early and short-term administration of an antibiotic or feeding a diet containing a coccidiostat on gastrointestinal function and the blood levels of selected hormones in young turkeys. A total of 1540 Hybrid Converter turkeys were allocated to 4 groups on the day of hatch. Each group consisted of 7 pens with 55 birds per pen. Group ENR was treated with enrofloxacin for the first 5 d of life, group DOX received doxycycline for 5 d and group MON was administered monensin for 84 d. CON birds served as a control group without any antibiotic treatment or MON administration. An analysis of the activity of bacterial enzymes revealed that the cecal microbiota of turkeys were less sensitive to MON than to the other 2 antibiotics. Turkeys subjected to ENR and DOX treatments were characterized by lower (P < 0.05) extracellular activity of cecal bacterial β-glucosidase, compared with groups CON and MON. The extracellular activity of cecal bacterial α-galactosidase and β-galactosidase decreased significantly in response to the experimental treatment with DOX (P < 0.05 vs. CON). Turkeys treated with ENR had higher total activity of bacterial β-galactosidase than those administered DOX or MON. Despite the differences in the enzymatic activity of microbiota, the use of antibiotics did not affect the concentrations of total short-chain fatty acids or ammonia in the cecal digesta of turkeys. A diet containing MON and the early administration of ENR or DOX induced an increase in blood noradrenaline levels (P = 0.004) in 56-day-old turkeys. Early DOX use increased plasma cortisol concentrations (P < 0.001) and decreased plasma serotonin levels (P = 0.006) in 56-day-old turkeys. Over the entire experiment (up to 12 wk of age), the use of MON improved the BW gain of turkeys (P = 0.055) and feed conversion (P = 0.016), compared with the DOX treatment.

Postoperative cerebral venous sinus thrombosis (CVST) is a rare complication of the retrosigmoid approach. To address the lack of literature, we performed a retrospective analysis. The thromboses were divided into those demonstrating radiological (rCVST) and clinical (cCVST) features, the latter diagnosed during hospitalization. We identified the former by a lack of contrast in the sigmoid (SS) or transverse sinuses (TS), and evaluated the closest distance from the craniotomy to quantify sinus exposure. We included 130 patients (males: 52, females: 78) with a median age of 46.0. They had rCVST in 46.9% of cases, most often in the TS (65.6%), and cCVST in 3.1% of cases. Distances to the sinuses were not different regarding the presence of cCVST (p = 0.32 and p = 0.72). The distance to the SS was not different regarding rCVST (p = 0.13). However, lower exposure of the TS correlated with a lower incidence of rCVST (p = 0.009). When surgery was performed on the side of the dominant sinuses, rCVSTs were more frequent (p = 0.042). None of the other examined factors were related to rCVST or cCVST. Surgery on the side of the dominant sinus, and the exposing of them, seems to be related with rCVST. Further prospective studies are needed to identify the risk factors and determine the best management.

Abstract In the present experiment, it was assumed that the appropriate dietary ratio of arginine (Arg) to lysine (Lys) can improve the immune status and growth performance of turkeys. The aim of this study was to evaluate the effects of two inclusion rates of Arg relative to Lys in turkey diets with Lys content consistent with National Research Council (NRC) recommendations or 10% higher on the immune status of birds and indicators of protein and DNA damage due to oxidation, nitration or epigenetic changes. Another goal was to determine which dietary Arg:Lys ratio stimulates the immune response of turkeys vaccinated against Ornithobacterium rhinotracheale. The experiment was performed on 576 turkeys randomly assigned to four groups with two levels of Lys (low = NRC recommendation or high = NRC + 10%) and two levels of Arg (95% or 105% Arg relative to the content of dietary Lys). It was found that the Lys content of turkey diets should be 10% higher than that recommended by the NRC and combined with the higher Arg level (105% of Lys). Although the above Arg:Lys ratio did not improve the growth performance of birds, it stimulated their immune system and reduced protein nitration as well as protein and DNA oxidation.

The application of transcriptomics to the study of the reproductive tract in male turkeys can significantly increase our current knowledge regarding the specifics of bird reproduction. To characterize the complex transcriptomic changes that occur in the testis, epididymis, and ductus deferens, deep sequencing of male turkey RNA samples (n = 6) was performed, using Illumina RNA-Seq. The obtained sequence reads were mapped to the turkey genome, and relative expression values were calculated to analyze differentially expressed genes (DEGs). Statistical analysis revealed 1,682; 2,150; and 340 DEGs in testis/epididymis, testis/ductus deferens, and epididymis/ductus deferens comparisons, respectively. The expression of selected genes was validated using quantitative real-time reverse transcriptase-polymerase chain reaction. Bioinformatics analysis revealed several potential candidate genes involved in spermatogenesis, spermiogenesis and flagellum formation in the testis, and in post-testicular sperm maturation in the epididymis and ductus deferens. In the testis, genes were linked with the mitotic proliferation of spermatogonia and the meiotic division of spermatocytes. Histone ubiquitination and protamine phosphorylation were shown to be regulatory mechanisms for nuclear condensation during spermiogenesis. The characterization of testicular transcripts allowed a better understanding of acrosome formation and development and flagellum formation, including axoneme structures and functions. Spermatozoa motility during post-testicular maturation was linked to the development of flagellar actin filaments and biochemical processes, including Ca2+ influx and protein phosphorylation/dephosphorylation. Spermatozoa quality appeared to be controlled by apoptosis and antioxidant systems in the epididymis and ductus deferens. Finally, genes associated with reproductive system development and morphogenesis were identified. To the best of our knowledge, this is the first genome-wide functional investigation of genes associated with tissue-specific processes in turkey reproductive tract. A catalog of genes worthy of further studies to understand the avian reproductive physiology and regulation was provided.

We postulated that the use of appropriate levels and proportions of arginine (Arg) and methionine (Met) in compound feed with high lysine content (Lys) would make it possible to fully exploit the growth potential of modern fattening turkey crossbreds, without compromising their immune system. The aim of this study was to determine the effect of different ratios of Arg and Met in diets with high Lys content on the performance and immune status of turkeys. The turkeys were assigned to 6 groups with 8 replicates per group and 18 birds per replicate. Six feeding programs, with 3 dietary Arg levels (90, 100, and 110%) and 2 dietary Met levels (30 and 45%) relative to dietary Lys content, were compared. During each of 4 feeding phases (weeks 0–4, 5–8, 9–12, and 13–16), birds were fed ad libitum isocaloric diets containing high level of Lys, approximately 1.83, 1.67, 1.49, and 1.20%, respectively. The dietary treatments had no effect on daily feed intake or body weight at any stage of the study. The protein content of the breast meat was higher in the treatments with the highest Arg level (110%) compared with the lowest Arg level (90%). Similarly, protein content was higher in the treatments with the higher Met level compared with the lower Met level. Higher plasma levels of tumor necrosis factor, interleukin 6 (IL-6), and immunoglobulin Y were found in turkeys fed diets with the lowest Arg content. An increase in Met content resulted in a decrease in plasma content of IL-6. In growing turkeys fed diets high in Lys, an Arg level of 90% relative to Lys can be used without negatively affecting production results and immune system. Regardless of dietary Arg levels, an increase in Met content does not stimulate the immune defense system and shows no effect on growth performance of turkeys in current trial.

Abstract Background Fruit pomaces are by-products rich in polyphenol compounds and dietary fiber. They seem to play an important role in regulating the gut microbiota, morphology and physiology. The aim of this study was to assess whether apple (A), blackurrant (B) or strawberry (S) pomaces could be suitable ingredients in broiler diets and their effect on gut health. A total of 480 male broilers were randomly allotted to 8 dietary treatments with lower (3%-L) or higher (6%-H) dietary fiber content: two control groups (CL/CH), two A diets (AL/AH), two B diets (BL/BH), two S diets (SL/SH). Diet and fruit pomaces were chemically analyzed to assess polyphenol concentration and fibre fraction content. After the evaluation of growth performance, 6 birds/group were slaughtered at 35 days of age. Morphometric and histopathological investigations were performed on duodenum, jejunum and ileum. Excreta were collected to perform microbiota evaluation by 16S DNA sequencing. Weight, viscosity, enzymatic activity, short chain fatty acid (SCFAs) and ammonia concentration were determined in ileum and/or ceca content. Results A pomace and A diets showed the lowest polyphenol content and the highest content of soluble fibre fraction. No significant differences were observed for growth performance, gut morphometry and histopathology (P > 0.05). Dietary fruit pomace inclusion increased the weight of ileum and ceca and the ileum digesta viscosity (P 0.05). This study was conducted to determine the effect of probiotic Pediococcus acidilactici (PA) strain MA18/5M supplementation of diets with different dietary energy levels on productive performance, egg quality, and body composition in Hy-Line Brown hens during a 16-week period from 32 to 47 wk of age. The experimental treatments with a 2 × 2 factorial design received a 2 wheat–corn–soybean diet: a moderately low energy density diet with 2,650 kcal ME/kg (M-LED) and a low energy density diet based on the M-LED diet with 2,550 kcal ME/kg (LED), each diet without and with probiotic supplementation (M-LED, LED, M-LEDp, and LEDp, respectively). Abstract Two experiments were performed to investigate the effect of different ratios of arginine (Arg) to lysine (Lys) in diets with low (30% Lys; Experiment 1) and high (45% Lys; Experiment 2) methionine (Met) levels on selected metabolic parameters, oxidative and epigenetic DNA damage, and the mechanisms underlying intestinal barrier integrity in turkeys challenged with Clostridium perfringens. In each experiment, 108 one-day-old Hybrid Converter female turkeys were placed in 6 pens (18 birds per pen) and reared for 42 days. At 34, 36 and 37 days of age, half of the birds were subjected to C. perfringens challenge. A 3 × 2 factorial design with three levels of Arg relative to Lys (90, 100 and 110%; Arg90, Arg100 and Arg110, respectively) and C. perfringens infection (−, +) was employed. Challenging birds with C. perfringens increased lipid oxidation and the oxidation and methylation of DNA of intestinal mucosa, and down-regulated the activities of DNA-repairing enzymes. Neither the dietary treatment nor the challenge affected the markers of liver function or metabolism. Arg110 diets with the high Met level induced DNA oxidation and methylation whereas these processes were downregulated in birds fed Arg90 diets. The results indicate that Arg90 diets with high Met levels have a beneficial influence on the indicators of intestinal barrier integrity in turkeys with necrotic enteritis (NE). Despite the analyzed amino acid ratios interacted with the systems responsible for the maintenance of gut integrity in the host organism, this dietary intervention probably enabled birds to cope with NE.

The research hypothesis postulated that the optimal dietary inclusion levels and ratios of lysine (Lys), arginine (Arg), and methionine (Met) can increase the growth potential of hybrid turkeys and limit metabolic disorders that weaken immune function. The experiment was carried out in a full rearing cycle, from 1 to 16 wk of age, in a two-factorial randomized design with 3 levels of Arg and 2 levels of Met (90, 100 and 110% of Arg, and 30 or 45% of Met, relative to the content of dietary Lys), with 6 groups of 8 replicates per group and 18 turkeys per replicate. In the first and second month of rearing, a significant dietary Arg-by-Met interaction was noted for daily feed intake and body weight gain, and a more beneficial effect was exerted by higher Met content and medium Arg content. Throughout the experiment, the higher dietary Met level increased the final body weight (BW) of turkeys (P = 0.001). Different dietary Arg levels had no influence on the growth performance of turkeys, but the lowest level decreased dressing yield (P = 0.001), and the highest level increased the percentage of breast muscles in the final BW of turkeys (P = 0.003). The lowest Arg level (90% of Lys content) undesirably increased the concentration of the proinflammatory cytokine IL-6 (P = 0.028) and decreased globulin concentration (P = 0.001) in the blood plasma of turkeys. The higher dietary Met level (45% of Lys content) increased plasma albumin concentration (P = 0.016). It can be concluded that higher dietary levels of Met (45 vs. 30% of Lys content) and Arg (100 and 110 vs. 90% of Lys content) have a more beneficial effect on the growth performance and immune status of turkeys.

Abstract Background Haemorrhagic enteritis (HE) of turkeys was first described in 1937 in the USA, while in Poland it was first diagnosed in 1987. Polish haemorrhagic enteritis virus (HEV) isolates are usually low pathogenic and trigger a subclinical disease. Unfortunately, even the low- pathogenic HEV strains cause severe immunosuppression leading to secondary bacterial infections and huge economic losses. The objective of this study was to evaluate if the influence of Met on HEV infected turkeys immune response can be differentiated by both its level and source. Met is one of the amino acids that not only play a nutritional role but also participate in and regulate key metabolic pathways and immune response. In our study, the birds were assigned to 4 dietary treatments which differed in Met levels (0.55 and 0.78% in weeks 1–4 of age and 0.45 and 0.65% in weeks 5–8 of age, respectively) and sources (DL-methionine (DLM) or DL-methionine hydroxy analogue (MHA)). Results The HEV added the percentage of CD4+ cells and decreased the percentage of IgM+ cells in the blood, spleen and caecal tonsils (CTs) of turkeys. In addition, it increased the percentage of CD4+CD25+ cells in blood, and interleukin-6 (IL-6) level in plasma. The higher dose of Met led to a significant decrease in the percentages of CD4+, CD8+ and CD4+IL-6+ cell subpopulations in the blood of HEV-infected and uninfected turkeys and to an increase in the percentage of IgM+ B cells in CTs. Turkeys administered feeds with an increased Met content displayed a decrease in plasma IL-6 levels and an increase in plasma IgA levels. Conclusions The results of this study indicate that HEV infection impairs the immune function in turkeys. Met content in the feed has a moderate effect on the immune response in HEV-infected turkeys. The source of this amino acid appears not be as important as its dose, because value of the analysed parameters did not differ significantly between turkeys receiving feeds with DLM or MHA. In the uninfected turkeys, the higher by 40% (than recommended by NRC) level of Met in the feeds had a positive effect on humoral immunity parameters.

Abstract Distinct enzymes, including cyclooxygenase 1 and 2 (COX-1 and COX-2), lipoxygenase (LOXs), and cytochrome P450 monooxygenase (CYP450), produce different stress mediators and mediate inflammation in birds. Bioactive agents such as acetylsalicylic acid (ASA) and vitamin E (vE) may affect enzyme activities and could be used in poultry production to control the magnitude of acute phase inflammation. Here, we characterized COX, LOX, and CYP450 mRNA expression levels in chicken immune tissues in response to Escherichia coli lipopolysaccharide (LPS) challenge and investigated whether ASA and vE could alter gene expression. Additionally, for the first time in chickens, we evaluated oxygen consumption by platelet mitochondria as a biomarker of mitochondria function in response to ASA- and vE. LPS challenge compromised bird growth rates, but neither dietary ASA nor vE significantly ameliorated this effect; however, gradually increasing dietary vE levels were more effective than basal levels. ASA regulated arachidonic acid metabolism, providing an eicosanoid synthesis substrate, whereas gradually increasing vE levels evoked aspirin resistance during challenge. Gene expression in immune tissues was highly variable, indicating a complex regulatory network controlling inflammatory pathways. However, unlike COX-1, COX-2 and CYP450 exhibited increased mRNA expression in some cases, suggesting an initiation of novel anti-inflammatory and pro-resolving signals during challenge. Measuring oxygen consumption rate, we revealed that neither the ASA nor vE levels applied here exerted toxic effects on platelet mitochondria.

We postulated that the use of optimal levels and proportions of Arg and Met relative to a low or high concentration of Lys in diets for meat turkeys would reduce the occurrence of metabolic disturbances in the nervous tissue that can lead to neurodegenerative changes. The aim of the study was to determine the effect of various proportions of Lys, Arg, and Met in diets for turkeys, with a low content of Lys in accordance with NRC (Experiment 1) recommendations, and in diets with high Lys levels that are close to the recommendations of breeding companies (Experiment 2) on selected indicators of potential neurodegenerative effects in the brain and liver of turkeys. The Experiment 1 and Experiment 2 was conducted using 864 day-old turkey chicks randomly assigned to six groups, in eight replicates (6 groups × 18 birds × 8 replicates). A full description of the methodology can be found in previously published papers using the same experimental design. Indicators informing about the presence or advancement of neurodegenerative processes in the nervous tissue were determined in the brain and liver (level of: AChE, amyloid-β, GAChE, Tau protein, LRP1, and the degree of DNA methylation). It was established that in the case of both a low (National Research Council, NRC) and a high (British United Turkeys, BUT) level of Lys in the diet of turkeys, the Arg level can be reduced to 90% of the Lys level and Met to 30% of the Lys level, because this does not cause neurodegenerative changes in turkeys. Unfavorable neurodegenerative changes may appear if the Arg level is increased from 100 to 110% of the Lys level recommended by the NRC. However, due to the lack of such a relationship when Arg is increased from 100 to 110% of the Lys level recommended by BUT, at this stage of research no definitive conclusions can be drawn regarding the risk of neurodegenerative changes caused by increasing Arg in the diet of turkeys.

The aim of the study was to compare the effect of the administration of antibiotics or probiotics on chickens in their first week of life, on selected parameters of humoral and cellular immunity, and on the bursa of Fabricius and spleen indices. The experiment was carried out on 90 one-day-old male broilers. The control group received no additive in the drinking water; the group GP received a probiotic providing Enterococcus faecium and Bacillus amyloliquefaciens; and the group GA received 10% enrofloxacin in the drinking water on the first five days of life. Administration of the antibiotic enrofloxacin or a probiotic containing E. faecium and B. amyloliquefaciens strains to chickens in their first week of life exerts pronounced immunomodulatory effects on humoral and cellular defense mechanisms in these birds. The changes in the subpopulations of B and T cells immediately following early administration of enrofloxacin or the probiotic were not observed at the age of 35 days. Early administration of enrofloxacin can pose a risk of suppression of humoral immunity, as indicated by the significant decrease in the total IgY concentration in the plasma of the chickens.

The aim of the study was to determine whether the level and form of Cr in the diet of chickens influences its accumulation in tissues as well as intestinal absorption of P and its deposition in tissues. The experiment was carried out on 405 one-day-old male Ross 308 chickens that were randomly divided into five treatment groups. Control group was fed the diet without supplemental chromium; experimental groups were fed the diet with 3 or 6 mg/kg chromium picolinate (Cr-Pic) and with 3 or 6 mg/kg chromium nanoparticles (Cr-NP). Chromium was found to accumulate in the tissues of the ileum, liver, breast muscle, bones skin and in feathers of chickens. Chromium deposited in the ileum of chickens does not affect the ex vivo estimated intestinal absorption of P. The use of Cr in the diet of chickens carries the risk of lowering P levels in femur.

A 2 × 3 factorial experiment was conducted to evaluate the effects of two dietary methionine levels (Met; 30% and 45% of Lys content) and three arginine levels (Arg; 90%, 100%, and 110% of Lys content) on plumage development in 4- and 16-week-old female turkeys. One-day-old turkey poults were assigned to six groups (eight replicate pens per group and 18 birds per pen) and fed experimental diets containing 1.6%, 1.5%, 1.3%, and 1.0% of Lys in four successive four-week periods. After weeks 4 and 16 of feeding, eight turkeys per group were selected for plumage evaluation. Feathers were collected from the outer side of one thigh and from an area of 4 cm2 in the interscapular region. Plumage was evaluated based on an established pattern of five feather development stages in turkeys, from stage I (pinfeathers covered in sheaths) to stage V (mature feathers). An increase in the Met inclusion rate to 45% of Lys content had no significant effect on feather growth in 4-week-old turkeys, but it accelerated the development of feathers in 16-week-old birds. A lower percentage of stage II (p = 0.035), stage III (p = 0.019), and stage IV (p = 0.003) immature feathers, and a higher percentage of stage V (mature) feathers (p = 0.001) were observed. Methionine exerted a greater effect on the development of thigh feathers (p = 0.001) than interscapular feathers (p = 0.074). Unlike Met, different Arg concentrations had no influence on plumage development in turkeys. Overall, the present results indicate that supplemental Met has a potential for accelerating feather development in 16-week-old turkeys via an increased supply of total sulfur amino acids.

Abstract Background The results of experiments involving broiler chickens and turkeys indicate that increased dietary methionine (Met) levels may improve the antioxidant protection of tissues in fast-growing birds. This is an important consideration since viral infections induce oxidative stress. The aim of this study was to verify the hypothesis that turkey diets with increased Met content can suppress oxidation processes induced by infection caused by the haemorrhagic enteritis virus (HEV), and that the noted effect is determined by the chemical form of this amino acid: DL-methionine (DLM) or DL-hydroxy analogue of Met (MHA). Results Dietary Met content above 40% higher than the level recommended by the NRC (1994) intensified lipid peroxidation in the small intestine, leading to an increase in malondialdehyde (MDA) and lipid peroxide (LOOH) levels, but it also stimulated antioxidant mechanisms in the blood and liver of turkeys infected with HEV. In comparison with DLM, MHA contributed to more severe symptoms of oxidative stress, such as elevated MDA levels in the intestines, and a decrease in glutathione peroxidase (GPx) activity and ferric-reducing ability of plasma (FRAP). Conclusions In HEV-infected turkeys, diets with increased Met content did not exert a clear antioxidant effect, which was noted in uninfected birds. The prooxidant activity of Met observed in the small intestinal wall was suppressed in the blood and liver of turkeys, most likely due to intensified synthesis of uric acid and glutathione. In comparison with MHA, DLM had a more beneficial influence on the analysed parameters of the redox status in the small intestine, blood and liver of turkeys.

Arginine (Arg) and lysine (Lys) may be important for the overall health of turkeys. The aim of this study was to determine whether low (consistent with the guidelines) and high (10% higher than recommended) levels of dietary Arg and Lys can modulate performance and the functional status of the gut. Female turkeys were allocated to four dietary treatments (two levels of Lys (low or high) and two levels of Arg (low or high)) for a 16 wk feeding period. The treatments did not affect turkey performance determined separately for four feeding phases and for the entire 16 wk experiment (p > 0.05). They had no significant influence on carcass yield, meat characteristics or the associated traits either (p > 0.05). High-Lys diets contributed to a decrease in cecal pH, a significant increase in the concentrations of short-chain fatty acids (SCFA) and a decrease in the concentrations of putrefactive SCFA and ammonia in the cecum. High dietary levels of both amino acids significantly enhanced the activity of cecal microbiota evaluated based on extracellular enzyme activity. These findings indicate that the higher dietary level of Lys was more effective in modulating the physiological status of the gut in turkeys than Arg.

The velocity potential of the fluid satisfies the Laplace equation with nonlocal boundary conditions on a free surface. This differential problem is transformed to an evolution equation in Fourier variables. The Fourier transform images of boundary functions are approximated by Picard’s iterations and the method of lines on meshes related to roots of Hermite polynomials. Due to convolutions of sine and cosine functions the integral terms of Picard’s iterations reveal unexpected instabilities for wave numbers in a neighborhood of zero.

The aim of this study was to compare the effect of the use of enrofloxacin and a probiotic containing Enterococcus faecium and Bacillus amyloliquefaciens strains in the first week of life of chickens on oxidative and epigenetic changes in molecules and intestinal integrity. The three treatments were as follows: the control group received no additive in the drinking water (GC); the second group (GP) received a probiotic preparation in the drinking water during the first five days of life, providing E. faecium strain 4a1713 at 1.0 × 107 CFU/L water and B. amyloliquefaciens 4b1822 at 1.0 × 107 CFU/L water, the third group (GA) received an antibiotic (enrofloxacin 0.5 mL/L water) in the drinking water during the first five days of life. The use of both enrofloxacin and a probiotic containing E. faecium and B. amyloliquefaciens strains in chickens’ first week of life improved intestinal integrity and reduced inflammation and oxidative and epigenetic changes in the small intestine. This effect was evident both at 6 days of age and at the end of the rearing period.

The present study investigated the effects of apple (A), blackcurrant (B) and strawberry (S) dried pomaces on meat quality, blood chemistry and redox status of broiler chickens. A total of 480 Ross-308 male broilers were divided into 8 dietary treatments containing 3% and 6% of cellulose preparation (C), A, B or S. Six birds/group were slaughtered at 35 days of age and blood samples were collected. Carcass traits and meat quality were determined on the Pectoralis major muscles, recording nonsignificant differences. Antioxidant activity was evaluated in serum, liver and breast muscle. In serum, fruit pomaces lowered triglycerides, creatinine and atherogenic index (p < 0.05). Regarding redox status, in serum, ACW (antioxidant capacity of water-soluble substances) and ACL (antioxidant capacity of lipid-soluble substances) were greater in A (p < 0.001). In breast, ACW and ACL were higher in B and S compared to C (p < 0.05). In liver, ACL was greater in B and S compared to C (p < 0.001) and in higher dosage compared to low (p = 0.036). GSSG (oxidized glutathione) concentration was lower in A, whereas A, B and S presented a higher GSH (reduced glutathione)/GSSG ratio. The results showed that fruit pomaces could represent promising feed ingredients for broilers, improving serum, meat and tissue antioxidant parameters.

We hypothesized that full-fat insect meal from Hermetia illucens (HI) larvae can be an acceptable source of protein and energy in the diet of young turkeys, in an amount adapted to the nutritional needs of these birds, and at the same time can improve their antioxidant status and metabolism. The turkeys were fed a control diet (HI0) without the insect meal, and three diets with increasing HI content of 5%, 10% and 15% (treatments HI5, HI10 and HI15, respectively). The use of 10% or 15% HI in the diet of young turkeys, while beneficially raising levels of P, Fe and Hb, has a negative effect on lipid metabolism, increasing TC levels, lipid oxidation, and fat deposition in the liver. The inclusion of 5% HI in the diet of young turkeys has no adverse effect on the lipid status and histology of the liver, but it does not improve antioxidant status. To conclude, the level of HI meal in the diet of turkeys should not exceed 5%. However, as similar studies on turkeys have not yet been published, overly general conclusions should not be drawn from the results of the present study, and further research is necessary.

The aim of the study was to determine whether the use of Mn nanoparticles would make it possible to reduce the level of this micronutrient added to turkey diets without adversely affecting growth performance, antioxidant and immune status, or apoptosis. The experiment was conducted on 6 groups of turkeys with 10 replications in a two-factor design with 3 dosages of manganese, 100, 50 and 10 mg/kg, and 2 sources, manganese oxide (MnO) and manganese nanoparticles (NP-Mn2O3). The study showed that irrespective of the form of Mn used, reducing the Mn level recommended by British United Turkeys for supplementation of the diet of young turkeys from 100 mg/kg to 10 mg/kg increases the content of this element in the blood with no adverse effect on growth performance or the immune system. The reduction in Mn supplementation in the form of NP-Mn2O3 from 100 to 50 and even to 10 mg/kg of turkey diet has no negative effect on antioxidant defence in young turkeys. A 50% reduction of the recommended Mn level in the form of MnO enhances lipid oxidation processes. Replacing MnO with NP-Mn2O3 in the turkey diet probably can increase apoptosis in young turkeys. On the other hand, irrespective of the form of Mn used, reducing supplementation of the turkey diet with this element from 100 to 50 and even to 10 mg/kg probably can reduce apoptosis.

The aim of this study was to determine the efficacy of different dietary fruit pomaces in reducing lipid oxidation in the meat of turkeys fed diets with a high content of n-3 polyunsaturated fatty acids (PUFAs). Over a period of 4 weeks before slaughter, turkeys were fed diets with the addition of 5% dried apple, blackcurrant, strawberry and seedless strawberry pomaces (groups AP, BP, SP and SSP, respectively) and 2.5% linseed oil. Pomaces differed in the content (from 5.5 in AP to 43.1 mg/g in SSP) and composition of polyphenols Proanthocyanidins were the main polyphenolic fraction in all pomaces, AP contained flavone glycosides and dihydrochalcones, BP contained anthocyanins, and SP and SSP-ellagitannins. The n-6/n-3 PUFA ratio in all diets was comparable and lower than 2:1. In comparison with groups C and AP, the percentage of n-3 PUFAs in the total fatty acid pool of white meat from the breast muscles of turkeys in groups BP, SP and SSP was significantly higher, proportionally to the higher content of α-linolenic acid in berry pomaces. The fatty acid profile of dark meat from thigh muscles, including the n-6/n-3 PUFA ratio, was similar and lower than 3:1 in all groups. We have postulated that supplementation with Cr can increase serotonin levels and improve the antioxidant status of chickens, with no adverse effect on the secretion of other hormones. The study aimed to determine what form and dose of Cr more favorably affect the level of selected hormones (insulin, glucagon, serotonin, dopamine, noradrenaline, histamine, T3 and T4) and the antioxidant status (level of malondialdehyde and lipid peroxides, activity of superoxide dismutase and catalase) of chicken tissues. The experiment was carried out on chickens randomly divided into five treatment groups. The basal diets (control group) were supplemented with two levels of Cr (3 and 6 mg/kg) and two Cr sources: Cr-picolinate (Cr-Pic) and Cr-nano (Cr-NP) to obtain four experimental diets: 3 mg/kg Cr-Pic, 6.0 mg/kg Cr-Pic, 3.0 mg/kg Cr-NP. and 6.0 mg/kg Cr-NP. The addition of Cr in both forms increased the level of serotonin at a dose of 3 mg/kg and, at the same time, reduced the level of noradrenaline. The addition of Cr at 3 mg/kg, irrespective of the form used, regulated the level of hormones of carbohydrate metabolism (increasing insulin levels and reducing glucagon levels) and had an adverse effect on the antioxidant status of the liver and breast muscle. Due to the adverse effect of Cr at 3 mg/kg on the antioxidant status of chickens, this level of Cr should not be considered in both forms Cr-Pic and Cr-NP as a feed additive for broiler chickens. In the future, studies on the potential beneficial effects of Cr on the organism should take into account doses lower than 3 mg/kg.

The aim of the study was to determine the effectiveness of the combined use of Cu, Zn, and Mn nanoparticles in the diet of turkeys with regard to potential reduction of the levels of these elements added to feed. The experiment was carried out on turkeys’ hens assigned to four groups. Turkeys from the PC group received feed with the addition of inorganic forms of Cu, Zn, and Mn in the B.U.T. (British United Turkeys) recommended levels, from group IR received the addition in amounts reduced to 10% of the recommended levels, and from group NR received the addition of elements in the form of nanoparticles in the same amounts as in group IR. The turkeys from group NC received feed without the addition of these elements. The research showed that the addition of Cu, Zn, and Mn to turkey diets, in both inorganic forms and as nanoparticles, in quantities covering only 10% of B.U.T. recommendations had no adverse effect on growth performance or on the antioxidant and immune defense of turkeys. The changes in the redox status of the turkeys whose diet was not supplemented with Cu, Zn, and Mn indicate reduced oxidation processes in the tissues.

The problem describing a ship motion in waves comprises the Laplace equation, boundary condition on wetted surface of the ship, condition on the free surface of the sea in the form of a differential equation, the radiation condition, and a condition at infinity. This problem can be transformed to a Fredholm equation of second kind, and then numerically solved using the boundary element method, if the fundamental solution of the problem is known. This paper presents the derivation of the fundamental solution. In physical interpretation, the fundamental solution represents the moving and pulsating source under free surface of the sea. The free surface elevation, generated by the source for different forward speed and frequency of pulsation, is presented in this paper.

A method is presented for the real-time quantitative determination of isocyanic acid (ICA) in air using proton transfer reaction-mass spectrometry (PTR-MS). Quantum mechanical calculations were performed to establish the ion-polar molecule reaction rate of ICA and other isocyanates. The PTR-MS was calibrated against different ICA air concentrations and humidity conditions using Fourier transform-infrared spectroscopy (FT-IR) as quantitative reference. Based on these experiments a simple humidity dependant model was derived for correction of the PTR-MS response for ICA. The corrected PTR-MS data was linearly correlated (R² > 0.99) with the data acquired by FT-IR. The PTR-MS instrumental limit of detection (LOD) for ICA was 2.3 ppb. Humid atmospheres resulted in LODs of 3.4 and 7.8 ppb, at an absolute humidity (AH) of 4.0 and 15.5 g m-3, respectively. Furthermore, off-line sampling using denuder and impinger samplers using di-n-butylamine (DBA) as derivatization reagent was compared with PTRMS measurements in a dynamically generated standard ICA atmosphere. Denuder (n = 4) and impinger (n = 4) sampling subsequent to liquid chromatography mass spectrometry (LC-MS) determination compared to corrected PTR-MS data resulted in recoveries of 79.6 (8.1% RSD) and 99.9 (9.3% RSD)%, respectively. Measurements of ICA from thermally decomposed cured 1,6-hexamethylene diisocyanate (HDI)-paint was performed using PTR-MS and denuder (n = 3) sampling. The relation between the average ICA responses using denuders (34.4 ppb) and PTR-MS (42.6 ppb) was 80.6%, which coincided well with the relative recovery obtained from the controlled laboratory experiments using dynamically generated ICA atmospheres (79.6%). The variability in ICA air concentration during the welding process (170% RSDPTR-MS) illustrated the need for real-time measurements. [ABSTRACT FROM AUTHOR]
