Objective To explore the effects of circular RNA circATP2B1 on proliferation and invasion of gastric cancer cells and its potential molecular mechanism. Methods From July 2018 to February 2021, 44 cases of gastric cancer tissue specimens and paracancerous tissue specimens were collected from the Department of Gastrointestinal Hepatobiliary Surgery of Quanzhou First Hospital Affiliated to Fujian Medical University. Four gastric cancer cell lines (SGC7901, HS-746T, MGC803, BGC823) and normal gastric mucosal epithelial cells (GES-1) were selected. The expression of circATP2B1 in gastric cancer tissues and cell lines was examined by RT-PCR. The gastric cancer cells with the lowest expression of circATP2B1 were divided into control group (transfected with negative control plasmid) and experimental group (transfected with circATP2B1 over expression plasmid). MTT and Transwell experiment were used to detect the proliferation activity and invasion ability of gastric cancer cells in each group. Bioinformatics and dual luciferase reporter gene experiments were used to analyze the possible molecular mechanism of circATP2B1, and RT-qPCR and Western blot were used to detect the expression of circATP2B1 downstream genes. Results The expression of circATP2B1 in gastric cancer tissue was significantly lower than that in adjacent tissues (0.92±0.08 vs. 3.62±0.23, P＜0.01). The expression of circATP2B1 in gastric cancer cell lines was significantly lower than that in GES-1 cells (P＜0.01)and HS-746T cells had the lowest expression level (P＜0.01). Compared with the control group, the proliferation activity of HS-746T cells in the experimental group was significantly reduced (P＜0.05), and the invasion ability was significantly reduced (P＜0.01). Bioinformatics and dual luciferase reporter gene experiments showed that circATP2B1 can target miR-452-5p (P＜0.01), and miR-452-5p can target protocadherin 9 (PCDH9) (P＜0.01). Compared with the control group, the expression of miR-452-5p in HS-746T cells in the experimental group decreased significantly (1.00±0.04 vs. 0.24±0.05, P＜0.01), and the expression of PCDH9 gene increased significantly(P＜0.01). Conclusions The circATP2B1 is low expressed in gastric cancer tissues and cell lines, and circATP2B1 positively regulates PCDH9 gene expression through binding miR-452-5p, thereby reducing the proliferation and invasion of gastric cancer cells HS-746T.

With the widespread adoption of electric vehicles (EVs), their charging and discharging schedules pose new challenges for real-time load forecasting in commercial buildings. This study proposes a prediction model based on the integration of bidirectional long short-term memory (BiLSTM) networks and Transformer architecture, along with the introduction of a cognitive control system and cyber–physical systems (CPS) to address issues such as data loss and excessive computation time during the forecasting process. The BiLSTM–Transformer model significantly improves load-forecasting accuracy and real-time performance by combining time-series modeling with global feature extraction capabilities. Additionally, the cognitive control system includes user-aware cognitive control (UACC) and Microgrid Control Center Cognitive Control (MACC). UACC quantifies information gaps in real time and adaptively adjusts strategies during communication instability, while MACC employs Q-learning algorithms to evaluate the impact of data loss on scheduling and optimize power resource allocation. The synergy between these mechanisms ensures system stability and predictive performance in scenarios involving data loss or communication disruptions. Experimental results demonstrate that the model achieves outstanding predictive accuracy under complete data conditions and significantly reduces errors in scenarios with data loss, validating its superior accuracy and robustness. Mudflat wetland, one of the 27 surface elements identified by the International Geographic Data Committee, has undergone substantial transformations with the rapid growth of the social economy and marine hazards, resulting in significant changes in its area and distribution. Quick identification of mudflat wetland evolution is vital to improve the mudflat ecological service value. We employed object-oriented and decision tree classification methods to map the mudflat wetland in the Yellow Sea using the Landsat time series from 1983 to 2020. The Improved Spectral Water Index (IWI) was established by combining the characteristics of many ratio indices and using ratio operation and quadratic power operation. The coefficient of variation (CV) of the IWI was calculated, and the range of the intertidal zone in 1983, 1990, 2000, 2010, and 2020 was obtained by using a threshold method. The results indicate that the mudflat wetland area decreased continuously from 1983 to 2020, with a reduction of 337.38 km2/10a. Among the total area, the natural wetland experienced a decline of 446.9 km2/10a, with the most drastic changes occurring between 2000 and 2010. In contrast, the area of the human-made wetland increased by 109.56 km2/10a. Over the 38 years, the tidal flat has undergone the most drastic reduction, with an average of 157.45 km2/10a. From 1983 to 2020, the intertidal zone area decreased, with a reduction of 429.02 km2/10a. Human activities were the key factors causing mudflat wetland loss. Based on these findings, we propose several policy suggestions. This study provides a scientific basis for understanding the synergetic evolution mechanism of coastal resources utilization and mudflat wetland protection under global change. Heavy-weight concrete (HWC) is a widely adopted radiation shielding material in the nuclear industry. In this study, two kinds of high-density aggregate, namely, iron sand (IS) and steel slag coarse aggregate (SSCA), were adopted to fully replace river sand and natural coarse aggregate, respectively, producing ultra-heavy-weight concrete (UHWC) with a unit weight >3800 kg/m3, to ensure excellent radiation shielding performance. However, IS and SSCA seriously impaired the passing ability of UHWC as the cement paste could not hold the IS and SSCA as firmly as natural aggregates, owing to their high density. To overcome this, the rheology of UHWC should be optimised by partially replacing the cement with superfine silica fume (SSF) and using a suitable amount of superplasticiser (SP) to enhance the wet packing density (WPD). A total of 28 UHWC mixes with different replacement ratios of SSF and different SP dosages were tested for segregation width, flowability, L-box passing ability, unit weight and WPD. Results revealed that incorporating an appropriate quantity of SSF and SP improved the WPD of UHWC, resulting in improved segregation resistance, enhanced flowability, increased passing ability and a higher unit weight. Currently, one in three adults worldwide may be obese or overweight. Obesity is characterized by an excess of energy‐storing white fat. An ingenious and sought‐after strategy against obesity is to activate energy‐consuming beige fat, which is converted from white fat or brown fat. However, existing tools for assessing brown or beige fat activation in vivo have certain limitations, such as being cumbersome and expensive. Optical imaging is a relatively straightforward and economical imaging technique that utilizes light to peer into the structural–functional information of living organisms at multiple scales. Despite the availability of various optical imaging modalities for detecting brown or beige fat, there is a dearth of literature summarizing relevant studies. Accordingly, this review focuses on these optical modalities and elaborates on their imaging principles, characteristics, and recent research advances in the detection of brown or beige fat. Their imaging targets, advantages, and disadvantages are further concluded. As a methodological reference, this review can guide the selection of optimal optical modalities to noninvasively profile brown or beige fat activation from a specific biological perspective, maximizing the potential of optical imaging in anti‐obesity assessment. Transition metal oxide-based catalysts are known as the most active oxygen evolution reaction (OER) catalysts due to their intrinsic electronic structures and binding strength of oxygenated intermediates. However, even the most active transition metal oxide-based OER catalysts have a turnover frequency far lower than that of the oxygen-evolving complexes in biological systems for intermolecular site aggregation. Herein, we implement the hybridization of a metal–organic framework and tungstic acid (WO3·xH2O-in-MIL-88) to achieve ordered FeNi site isolation at the molecular level and avoid intermolecular decomposition during the catalysis process. The optimized hybrid with 15% tungstic acid (15%-WO3·xH2O-in-MIL-88) achieves a much-enhanced OER activity and stability in terms of a low overpotential of 263 mV to reach a current density of 10 mA cm−2, a low Tafel slope of 39 mV dec−1, and long-term stability, which is much superior to that of the commercial RuO2 catalyst. Background/Aims: Data on the natural course of Chinese patients with ulcerative colitis (UC) was lacking. This study aimed to evaluate the natural history and prognosis of patients with UC in the past 15 years in China. Methods: This cohort study included patients with UC in a tertiary hospital in southern China from 2007 to 2021 (cohort I: 2007–2011, cohort II: 2012–2016, cohort III: 2017–2021). Patients' clinical characteristics and natural history were analyzed retrospectively. Results: Of 1,139 included patients, 683 patients presented with proctitis or left-sided colitis at diagnosis and 38.5% of them (263/683) developed proximal disease extension. Fifty-eight percent of patients experienced relapse, chronic continuous and intermittent active course. Five patients (0.4%) developed colorectal tumors/dysplasia. The overall surgery rate was 8.6%, and the rates were 14.2%, 7.8%, and 8.0% in the 3 cohorts, respectively (P=0.059). Average time from diagnosis to surgery decreased from cohorts I to III (144 months vs. 36 months, P<0.001), so did the use of glucocorticoids (58.2% vs. 43.5%, P<0.001) and immunosuppressants (14.1% vs. 13.4%, P=0.016), and days of hospitalization (13 days vs. 9 days, P<0.001). Biologics were used more frequently during the first year (0.8%, 2.1%, and 13.7% for cohorts I to III, respectively; P<0.001). The rate of mucosal healing increased over time. Conclusions: In Chinese UC patients, one-third of patients experienced proximal disease extension. The rates of malignancy and mortality were low. More biologics were used, while use of immunosuppressants and glucocorticoids were reduced over time. This paper presents a few-mode fiber (FMF) various fault-detection method for long-reach mode division multiplexing (MDM) based on multi-mode transmission reflection analysis (MM-TRA). By injecting unmodulated continuous light into the FMF, and measuring and quantitatively analyzing the transmitted and reflected or Rayleigh backscattering power of different spatial modes, it is possible to accurately detect and locate reflective and non-reflective fault events. This paper discusses the localization accuracy of fault types such as FMF break, FMF link connector mismatch, and FMF bending. Theoretical analysis and simulation experimental results demonstrate that the proposed MM-TRA can provide an effective characterization of various faults and can achieve high fault localization accuracy. In addition, the influence of mode crosstalk of mode multiplexer/demultiplexer and mode coupling in FMF on the localization accuracy of various faults are considered. The results indicate that when using the combination of LP01 and LP21 modes, the localization errors for the FMF break, connector mismatch, and bending are 3.42 m, 1.97 m, and 3.29 m, respectively, demonstrating good fault localization performance. The traditional single-mode fiber (SMF) optical time domain reflectometer (OTDR) may not be able to accurately detect and locate fault events in the dead zone of few-mode fiber (FMF) links. This paper introduces the concept of higher-order spatial mode detection dimensions unique to FMF, combined with the spatial mode coupling characteristics between modes. The Fresnel reflection from the end face of the fiber, the interior of the circulator, and the connector only occurs in the spatial mode of the injected optical pulse. The Rayleigh backscattering, which reflects the fault distribution characteristics of FMF links, can be detected by non-excited higher-order spatial modes. The proposed method can completely overcome the traditional OTDR dead zone. In this paper, the six-mode fiber is taken as an example for experimental verification. The detection optical pulse is injected into the fundamental mode LP01, and the Rayleigh backscattering of LP11a, LP11b, LP21a, LP21b, and LP02 higher-order spatial mode are collected and analyzed to accurately detect and locate the fusion splice fault event at 100 m and 500 m in the dead zone. CuS/Bi2O3 composite photocatalyst was prepared by calcination and in situ precipitation, and peroxymonosulfate (PMS) was applied to the degradation of tetracycline (TC) wastewater under visible light. The microscopic morphology, chemical composition, and optical properties of the composites were investigated by characterization means of XRD, FTIR, SEM, XPS, and UV-Vis DRS. The results showed that the introduction of CuS increased the specific surface area of Bi2O3 and increased the visible absorption boundary of Bi2O3 from 455 to 524 nm, which effectively inhibited the complexation of photogenerated electron-hole pairs. The experimental results showed that the introduction of PMS strengthened the removal of TC from the composites, and 95% of TC could be removed under visible light irradiation, and the reaction rate was 8.22 times higher than that of the unspiked PMS, indicating that the BC-15+vis/PMS catalytic system could degrade the pollutants efficiently. The radical capture experiments showed that several radicals, including ·OH, SO4·−, ·O2−, h+, and 1O2, were present in the catalytic system as the main active species to degrade TC, and the mechanism of photocatalytic activation of PMS by Z-type heterostructures of CuS/Bi2O3 composites was proposed. Aims: Acute kidney injury (AKI) has been associated with a variety of neurological problems, while the neurobiological mechanism remains unclear. In the present study, we utilized resting‐state functional magnetic resonance imaging (rs‐fMRI) to detect brain injury at an early stage and investigated the impact of microglia on the neuropathological mechanism of AKI. Methods: Rs‐fMRI data were collected from AKI rats and the control group with a 9.4‐Tesla scanner at 24, 48, and 72 h post administration of contrast medium or saline. The amplitude of low‐frequency fluctuations (ALFF) was then compared across the groups at each time course. Additionally, flow cytometry and SMART‐seq2 were employed to evaluate microglia. Furthermore, pathological staining and Western blot were used to analyze the samples. Results: MRI results revealed that AKI led to a decreased ALFF in the hippocampus, particularly in the 48 h and 72 h groups. Additionally, western blot suggested that AKI‐induced the neuronal apoptosis at 48 h and 72 h. Flow cytometry and confocal microscopy images demonstrated that AKI activated the aggregation of microglia into neurons at 24 h, with a strong upregulation of M1 polarization at 48 h and peaking at 72 h, accompanying with the release of proinflammatory cytokines. The ALFF value was strongly correlated with the proportion of microglia (|r| > 0.80, p < 0.001). Conclusions: Our study demonstrated that microglia aggregation and inflammatory factor upregulation are significant mechanisms of AKI‐induced neuronal apoptosis. Background Small bowel involvement is related to poor prognosis in Crohn's disease (CD), which may be a potential marker to stratify patients with a high risk of progression. This study aimed to establish a novel location classification system for CD and to develop a predictive model for disease progression. Methods Consecutive patients with non-stricturing/non-penetrating CD were retrospectively included in the Sixth Affiliated Hospital, Sun Yat-sen University (Guangzhou, P. R. China) between January 2012 and January 2018. Patients were classified into two groups according to disease location: small bowel involvement group and isolated colon group. The primary outcome was disease progression to stricturing or penetrating phenotypes. Progression-free survival was estimated using Cox proportional hazards regression analysis and Kaplan–Meier method. Results A total of 463 patients were analysed, with a median follow-up time of 55.3 months. Patients with small bowel involvement had a higher risk of disease progression than those with isolated colon disease (hazard ratio = 1.998, P  =   0.007), while no differences were found between Montreal location classification and disease progression. Median progression-free survival was higher in the isolated colon group than in the small bowel involvement group (84.5 vs 77.3 months, P  =   0.006). Four independent factors associated with disease progression were identified: small bowel involvement, duration of onset of >1 year, deep mucosal ulcer, and C-reactive protein levels of ≥10 mg/L (all P  <   0.05). The nomogram model based on these factors showed good performance in predicting disease progression, with a C-index of 0.746 (95% confidence interval, 0.707–0.785). Mycelium-bound composites (MBCs) are innovative materials created by combining lignocellulosic sub-products with fungal mycelium. These composites possess a remarkable ability to transform waste fragments into a continuous material without requiring additional energy input or generating further waste. The production process of MBCs involves utilising different fungal species, substrates, and pressing techniques, resulting in composites with diverse physical, mechanical, and functional properties. A comprehensive evaluation of MBCs' properties is crucial to explore their potential applications in the construction sector and ensure their suitability for specific purposes. This study provides a critical evaluation of the physical and mechanical properties of engineered mycelium-bound composites under various manufacturing conditions. Additionally, the analytic hierarchy process (AHP) and fuzzy comprehensive evaluation (FCE) methodologies were applied to investigation the optimum conditions for mycelium composites in the construction industry. The outcomes of FCE show the most promising fungal species, offering an optimal balance between material performance and production efficiency. Furthermore, the future development of MBCs manufacturing techniques was reviewed, providing a valuable reference for future research endeavours and showcasing the potential of MBCs applications within the field of civil engineering. Modern cloud native applications are generally built with a microservice architecture. To tackle various performance problems among a large number of services and machines, an end‐to‐end tracing tool is always equipped in these systems to track the execution path of every single request. However, it is nontrivial to conduct root cause analysis of anomalies with such a large volume of tracing data. This paper proposes a novel system named TraceRank to identify and locate abnormal services causing performance problems with dis‐aggregated end‐to‐end traces. TraceRank mainly includes an anomaly detection module and a root cause analysis module. The root cause analysis procedure is triggered when an anomaly is detected. To fully leverage the information provided by the tracing data, both the spectrum analysis and the PageRank‐based random walk methods are introduced to pinpoint abnormal services. The experiments in TrainTicket and Bookinfo microservice benchmarks and a real‐world system show that TraceRank can locate root causes with 90% in Precision and 86% in Recall. TraceRank has up to 10% improvement compared with several state‐of‐the‐art approaches in both Precision and Recall. Regenerative braking technology is a viable solution for mitigating the energy consumption of electric vehicles. Constructing a distribution strategy for regenerative braking force will directly affect the energy saving efficiency of electric vehicles, which is a technical bottleneck of battery-powered electric vehicles. The distribution strategy of the front- and rear-axle braking forces of electric vehicles that possess integrated front-wheel-drive arrangements is established based on the Economic Commission of Europe (ECE) regulations, which enables the clarification of the total braking force of the front axle. The regenerative braking torque model of the motor is adjusted to optimize the ratio of motor braking force to the whole front-axle braking force. The regenerative braking process of electric vehicles is influenced by many factors, such as driving speed and braking intensity, so regenerative braking presents characteristics of nonlinearity, time variability, delay, and incomplete models. By considering the impact of fuzzy controllers having better robustness, adaptability, and fault tolerance, a fuzzy control strategy is employed in this paper to accomplish the regenerative braking force distribution on the front axle. A regenerative braking model is created on the Simulink platform using the braking force distribution indicated above, and experiments are run under six specific operating conditions: New European Driving Cycle (NEDC), World Light-Duty Vehicle Test Cycle (WLTC), Federal Test Procedure 72 (FTP-72), Federal Test Procedure 75 (FTP-75), China Light-Duty Vehicle Test Cycle-Passenger (CLTC-P), and New York City Cycle (NYCC). The findings demonstrate that in six typical cycling road conditions, the energy saving efficiency of electric vehicles has greatly increased, reaching over 15%. The energy saving efficiency during the WLTC driving condition reaches 25%, and it rises to 30% under the FTP-72, FTP-75, and CLTC-P driving conditions. Furthermore, under the NYCC road conditions, the energy saving efficiency exceeded 40%. A cross‐1,3‐dipolar cycloaddition reaction of α‐halohydroxamates (in situ generated azaoxyallyl cations) with N,N′‐cyclic azomethine imines was developed. We use Sentinel‐1 Interferometric Synthetic Aperture Radar observations to derive the first complete map of slip‐rate distribution at km‐scale along the 1,000‐km‐long Haiyuan fault system, and further to analyze its earthquake potential. Rate distribution reveals four highly locked asperities (slip rate <1 mm/yr) and three creeping regions (slip rate ranges from 2 to 5 mm/yr) at depths of 0–20 km. Based on the rate distribution, we estimate seismic moment accumulation rate, and by combining historical earthquake records, we calculate the moment deficits on each fault segment. Results show that many fault segments have accumulated a seismic moment that equates to an Mw > 7.0 earthquake. The westernmost fault segment has been rarely mentioned in previous studies, but our calculations show evidently that it is capable of producing Mw > 7.0 earthquakes given the high seismic moment accumulation rate and a lack of small‐to‐moderate earthquakes. Plain Language Summary: Faults accumulate strains over long periods of seismic quiescence between great earthquakes, that is, interseismic strain accumulation, and release them either seismically by producing earthquakes or aseismically as creep. Studies of such strain accumulation and release processes are critical for understanding regional seismic hazard. We focus here on the 1,000‐km‐long Haiyuan fault system in northeastern Tibet. By using the most recent geodetic data sets, we quantify strain accumulation along the fault and evaluate its seismic potential. We derive fault slip rate distribution and calculate along‐strike seismic moment accumulation rate. Combining our knowledge of previous earthquake ruptures, including the time, extent and size, we compute the total seismic moment that has accumulated on each fault segment since the last major event. We find high seismic potentials along many fault segments, such as Lenglongling, Jinqianghe‐Maomaoshan and Gulang, which can generate an Mw > 7.0 earthquake anytime from now. Toward the western end, earthquake history remains poorly understood, but our analysis shows that the westernmost segment can produce Mw > 7.0 earthquakes, which deserves more attention. As widely acknowledged and targeted in Transforming Our World: The 2030 Agenda for Sustainable Development, talents education and training is an important measure to systematically solve the problem of economically, societally, resource, and environmentally sustainable development, and so as the post-graduate geographical talents education and training. Whether post-graduate geographical talents education and training can meet the need of sustainable development is an increasingly significant issue in geography science. Therefore, from the perspective of population scale, education quality, and education input, taking Chinese post-graduate geographical education as an example, the paper empirically investigated the spatial differentiation and talents production mechanism. With the support of spatial analysis tools by ArcGIS and GeoDa software, the strong inter-regional differentiation and imbalance characteristics of post-graduate geographical talents education were detected, outlining a general east-west geographical pattern in China. Moreover, the spatial production mechanism of post-graduate geographical talents has its own global and national scale, regional comprehensive and province-related characteristics, and production of the talents education and training in human geography, physical geography, and cartography. Summary: Background: Ustekinumab is effective in treating Crohn's disease (CD) and ulcerative colitis (UC). However, the loss of response (LOR) to ustekinumab and the efficacy of dose escalation have not been systematically explored. Methods: Databases were searched for eligible studies from inception through July 2021. Summary estimates were pooled, and subgroup analyses were performed to explore heterogeneity. Results: We included 14 studies (CD: 13; UC: 1). In CD patients, the annual risk of LOR to ustekinumab and dose escalation among primary responders was 21% (95% confidence interval [CI] 12–31%, 1530 person‐years, n = 9) per person‐year and 25% (95% CI 12–32%, 657 person‐years, n = 5) per person‐year respectively. Clinical response was regained in 58% (95% CI 49–67%, 279 patients, n = 8) of secondary non‐responders after dose escalation (interval reduction or intravenous reinduction). In UC patients, no studies provided data on LOR, but only one study showed that 35% (100/284) of patients underwent dose escalation (or sham dose adjustment), leading to an annual risk of dose escalation of 18% per person‐year. After dose escalation, 58% (14/24) of the patients regained symptomatic remission. Conclusions: Primary responders with CD experienced LOR to ustekinumab at a risk of 21% per person‐year and required dose escalation at a risk of 25% per person‐year. Combination therapy based on different mechanisms of cell death has shown promise in tumor therapy. However, when different modalities are integrated, the maximum synergy of the therapeutic effects is often lacking in the design. Herein, we report a cancer theranostic nanomedicine formula developed by considering the mechanisms of action of ferroptosis and the photothermal effect in combination therapy. The croconaine molecule was encapsulated as both a photothermal converter and an iron‐chelating agent with BSA, thus leading to biocompatible and stable Cro‐Fe@BSA nanoparticles (NPs). The Cro‐Fe@BSA NPs in the tumor milieu showed an activated photothermal effect leading to enhanced radical formation owing to the temperature‐dependent Fenton reaction kinetics, while radical formation during ferroptosis in turn prevented the heat‐induced formation of heat shock proteins and thus the self‐protection mechanism of cancer cells in response to heat. The activatable photoacoustic and magnetic resonance imaging performance of the Cro‐Fe@BSA NPs also enabled safe and reliable cancer theranostics. [ABSTRACT FROM AUTHOR]

Torreya grandis seed oil, with a feature of Δ5‐sciadonic acid (SA, 20:3 Δ5, 11, 14 ω‐6), is an attractive woody oil as its potential health benefits in lowering blood and hepatic lipids and anti‐inflammatory properties. In the present study, its oxidative stabilities at both room temperature and heating conditions were investigated by analysing oxidation stability index (OSI), shelf‐life, changes of fatty acids, acid value and peroxide value during stir‐frying tests. Also, its effects on the sensory qualities of stir‐fried shredded potatoes were evaluated. The OSI of T. grandis seed oil was 2.97 h at 120 °C, which was 11.24% higher than that of the control soybean oil, contributing to a longer predicted shelf‐life of 135–195 d for T. grandis seed oil than 127–180 d for soybean oil. T. grandis seed oil also exhibited superior thermal stability compared to soybean oil. The feature fatty acid, SA, only lost 2.48–5.73% (decreasing from 14.14 to 13.33–13.79%) under the optimised stir‐frying conditions at 140–180 °C for 3–5 min. For the stir‐frying shredded potatoes, T. grandis seed oil made it more pleasing in colour, odour and palatability. Due to its desirable oxidative stability at room temperature and stir‐frying performance at high temperature, T. grandis seed oil is suitable for use as a domestic cooking oil. Serum infliximab trough level(S-IFX) and antibody were documented to correlate with clinical response. The aim of this study was to identify the relationship between early S-IFX, early mucosal healing (MH) and one-year outcome in a cohort on maintenance IFX therapy in Crohn's disease (CD). The study group comprised of retrospectively enrolled patients diagnosed for Crohn's disease (n = 108). Patients received scheduled maintenance therapy after response to IFX induction, and had undergone the early S-IFX test and endoscopic examination at week 14. Clinical outcomes were evaluated during maintenance therapy until week 52. Early S-IFX was 4.78 ± 6.16 ug/ml in all the patients and 19% (21/108) of them developed antibodies, and 52 patients reached early MH. During 52 weeks' follow-up. Twenty-eight percent (30/108) of patients showed loss of response to IFX. Patients who lost response had lower early S-IFX than those who had sustained response (3.01 ± 3.66 vs. 5.47 ± 6.79 ug/ml, p =.02; 48 vs. 23%, p =.02). At week 52, 73 patients had repeated endoscopy and 42% of them reached MH. Early S-IFX had a predictive value on MH at week 52. When early S-IFX > 2.5 ug/ml, the sensitivity for predicting MH at week 52 was 87%, and the specificity were 61% (AUC = 0.73, p <.01). The combined predictive value of early S-IFX and early MH became stronger. Only 6% (1/18) of those patients who had low early S-IFX and had not reached early MH could reach MH at week 52.
