ObjectiveAortic dissection (AD) is a severe aortic disease with high mortality, and its pathogenesis remains elusive. To explore the regulatory mechanisms of AD, we integrated public RNA sequencing (RNA-seq) and single-cell RNA sequencing (scRNA-seq) datasets to screen the hub genes of AD and further analyzed their functions, which may provide references to the diagnosis and treatment of AD.MethodsFour AD-related datasets were obtained from the Gene Expression Omnibus (GEO) database. Weighted gene co-expression network analysis and differential expression analysis were applied to identify overlapping genes in dataset GSE153434. Protein–protein interaction (PPI) network was constructed based on overlapping genes. Five methods (closeness, degree, EPC, MCC, and MNN) were used to pick hub genes. The receiver operating characteristic curve was used to evaluate the diagnostic efficiency of the hub genes in extra datasets GSE98770 and GSE52093. scRNA-seq dataset GSE213740 was used to explore the expression and function of the hub genes at the single-cell level. Quantitative real-time polymerase chain reaction was used to verify the expression of hub genes in beta-aminopropionitrile (BAPN)-induced mouse thoracic aortic aneurysm and dissection (TAAD) model.ResultsA total of 71 overlapping genes were screened by intersecting the significant genes in the pink module and the differentially expressed genes. A PPI network with 45 nodes and 74 edges was generated, and five top hub genes (HIF1A, HGF, HMOX1, ITGA5, and ITGB3) were identified. All the hub genes had area under the curve values above 0.55. scRNA-seq data analysis showed that HIF1A was significantly upregulated in macrophages and HGF was significantly upregulated in vascular smooth muscle cells (SMCs) of the ascending aortas in AD patients. HIF1A may transcriptionally regulate multiple downstream target genes involving inflammation (TLR2, ALOX5AP, and MIF), glycolysis (ENO1, LDHA, and GAPDH), tissue remodeling (PLAU), and angiogenesis (SERPIN and VEGFA). HGF may participate in the signaling among SMCs, fibroblasts, and endothelial cells through binding to different receptors (MET, EGFR, IGF1R, and KDR). The mRNA expression of Hif1a, Hgf, and their target genes, including Alox5ap, Serpine1, Tlr2, Plau, Egfr, and Igf1r, was significantly upregulated in aortic tissues of BAPN-treated mice.ConclusionBy integrating RNA-seq and scRNA-seq data, we identified HIF1A and HGF as two hub genes with good diagnostic efficiency for AD. HIF1A in macrophages may promote AD formation by promoting inflammation, glycolysis, tissue remodeling, and angiogenesis, and HGF may mediate signaling among SMCs, fibroblasts, and endothelial cells in the development of AD.

ABSTRACT: The timing of the initial Indo-Asian collision is a subject of debate for a long time. Besides, the magmatic trace of the collisional process is also unclear. In the present study, the authors report Early Eocene leucocratic sill/dike swarms in the northern edge of the Nymo intrusive complex of the Gangdese belt, southern Tibet. The Nymo intrusive complex was emplaced at ca. 50 –47 Ma and surrounded by the metamorphosed Jurassic-aged Bima Formation volcano-sedimentary sequence along its northern side. At outcrops, the leucocratic sills/dikes intruded along or truncated the deformed foliations of the host Bima Formation, which has been subject to high-temperature amphibolite-facies metamorphism at ca. 50 –47 Ma. Detailed cathodoluminescence image analyses reveal that the zircon grains of the leucocratic sills/dikes have core-mantle textures. The cores yield the Jurassic ages comparable to the protolith ages of the Bima Formation. In contrast, the mantles of zircon grains yield weighted mean ages of ca. 49–47 Ma, representing the crystallization timing of these leucocratic sills/dikes. The coeval ages for the Nymo intrusive complex, the high-temperature metamorphism, and the leucocratic sills/dikes indicate that a close relationship exists among them. The authors tentatively suggest that these leucocratic sills/dikes were generated from partial melting of the Jurassic-aged Bima Formation volcanic rocks, triggered by the high heat from the magma chamber of the Nymo intrusive complex. This Early Eocene tectono-thermal event of coeval magmatism, metamorphism and partial melting was most likely formed during the Indo-Asian collisional setting.©2021 China Geology Editorial Office.

PurposeCardiac fibrosis is characterized by the excessive deposition of extracellular matrix (ECM) proteins and leads to the maladaptive changes in myocardium. Endothelial cells (ECs) undergoing mesenchymal transition contributes to the occurrence and development of cardiac fibrosis. CD146 is an adhesion molecule highly expressed in ECs. The present study was performed to explore the role of CD146 in modulating endothelial to mesenchymal transition (EndMT).MethodsC57BL/6 mice were subjected to subcutaneous implantation of osmotic minipump infused with angiotensin II (Ang Ⅱ). Adenovirus carrying CD146 short hairpin RNA (shRNA) or CD146 encoding sequence were infected into cultured human umbilical vein endothelial cells (HUVECs) followed by stimulation with Ang II or transforming growth factor-β1 (TGF-β1). Differentially expressed genes were revealed by RNA-sequencing (RNA-Seq) analysis. Gene expression was measured by quantitative real-time PCR, and protein expression and distribution were determined by Western blot and immunofluorescence staining, respectively.ResultsCD146 was predominantly expressed by ECs in normal mouse hearts. CD146 was upregulated in ECs but not fibroblasts and myocytes in hearts of Ang II-infused mice and in HUVECs stimulated with Ang Ⅱ. RNA-Seq analysis revealed the differentially expressed genes related to EndMT and Wnt/β-catenin signaling pathway. CD146 knockdown and overexpression facilitated and attenuated, respectively, EndMT induced by Ang II or TGF-β1. CD146 knockdown upregulated Wnt pathway-related genes including Wnt4, LEF1, HNF4A, FOXA1, SOX6, and CCND3, and increased the protein level and nuclear translocation of β-catenin.ConclusionsKnockdown of CD146 exerts promotional effects on EndMT via activating Wnt/β-catenin pathway and the upregulation of CD146 might play a protective role against EndMT and cardiac fibrosis.

AIM: To investigate the curative effect of phacoemulsification and intraocular lens(IOL)implantation combined with goniosynechialysis in the treatment of age-related cataract merging with primary angle-closure glaucoma(PACG).METHODS: Totally 80 patients with age-related cataract merging with PACG were in our hospital from January 2014 to January 2016. The preoperative average intraocular pressure(IOP)was 33.22±3.17mmHg; the average depth of anterior chamber was 2.07±0.15mm; the dynamic situation of primary angle closure ≤1/2 cycle by gonioscope. They were randomly divided into Group A and B for doing a study. All the two groups were treated with phacoemulsification and intraocular lens implantation. And the Group A was with goniosynechialysis. The following up period was 2mo, and we observed the IOP, chamber depth and the anterior chamber angle.RESULTS: The change of chamber depth and intraocular pressure about the two groups: the average intraocular pressure of the Group A was 15.11±3.67mmHg,the chamber depth was 3.11±0.08mm; those of the Group B were 17.24±1.67mmHg, 2.76±0.15mm respectively; the differences had statistical significance(PPCONCLUSION: The phacoemulsification and intraocular lens implantation combined with goniosynechialysis in the treatment of age-related cataract merging with primary angle-dosure glaucoma is safe and reliable. It's simple to operate, and do not increase the risk of surgery.

Abstract Five new polychlorinated bibenzyls (1–5) along with 3 known compounds (6–8) were isolated from the stems and leaves of Rhododendron minutiflorum. The chemical structures of all the isolates were determined by spectroscopic methods, and compounds 1 and 2 were further verified by single-crystal X-ray diffraction analyses. Compounds 1–5 were halogenated compounds which bear three to five chlorine atoms in their chemical structures. Biologically, compounds 2, 5 and 6 showed varying degrees of toxicity toward the Asian citrus psyllid (Diaphorina citri) with LD50 values 27.15, 17.02 and 16.20 mg/L, respectively. These values were comparable to the positive control matrine (LD50 = 11.86 mg/L), which were calculated using observations on day 6. Meanwhile, compound 4 had α-glucosidase inhibitory activity with IC50 value of 17.87 ± 0.74 μM. Graphical Abstract

A practical and straightforward access to pyrazolo[3,4-c]quinolines by molecular sieve mediated dehydrogenative [2 + 2 + 1] heteroannulation of N-(o-alkenylaryl)imines with aryldiazonium salts is described using a sp3-hybrid carbon atom as a one-carbon unit. The reaction enables the formation of three new chemical bonds, a C-C bond and two C-N bonds, in a single reaction and features simple operation and excellent functional group tolerance. The Litang fault system that crosses the Litang Plateau, a low relief surface at high elevation (∼4200-4800m above sea level) that is not affected by regional incision, provides the opportunity to study exhumation related to tectonics in the SE Tibetan Plateau independently of regional erosion. Combining apatite and zircon fission track with apatite (U-Th)/He thermochronologic data, we constrain the cooling history of the Litang fault system footwall along two transects. Apatite fission track ages range from 4 to 16 Ma, AHe ages from 2 to 6 Ma, and one zircon fission track age is ∼99 Ma. These data imply a tectonic quiet period sustained since at least 100 Ma with a slow denudation rate of ∼0.03 km/Ma, interrupted at 7 to 5 Ma by exhumation at a rate between 0.59 and 0.99 km/Ma. We relate that faster exhumation to the onset of motion along the left-lateral/normal Litang fault system. That onset is linked to a Lower Miocene important kinematic reorganization between the Xianshuihe and the Red River faults, with the eastward propagation of the Xianshuihe fault along the Xiaojiang fault system and the formation of the Zhongdian fault. Such strike-slip faults allow the sliding to the east of a wide continental block, with the Litang fault system accommodating differential motion between rigid blocks. The regional evolution appears to be guided by the strike-slip faults, with different phases of deformation, which appears more in agreement with an "hidden plate-tectonic" model rather than with a "lower channel flow" model. Electrochemical behavior of a carbon paste electrode (CPE) modified with nickel(II)-5, 11, 17, 23-tetra-tert-butyl-25, 27-bis(diethylcarbamoylmethoxy) calix[4]arene (Ni (II)-L) complex and its electrocatalytic activity towards the oxidation of hydrogen peroxide were investigated by cyclic voltammetric technique in a 5.0×10−2 mol/L NaClO4+1.0×10−3 mol/L NaOH solution. It was found that Ni(II)-L acts as an effective catalyst for the oxidation of hydrogen peroxide. The modified electrode exhibited a linear response over a hydrogen peroxide concentrations in the range of 2.0×10−5−1.0×10−4 mol/L with a detection limit as low as 1.0×10−6 mol/L. The relative standard deviation was 3.5% for 5 successive determinations of H2O2 at 1.0×10−5 mol/L. The modified electrode was used successfully in rainwater analysis. [ABSTRACT FROM AUTHOR]

A novel metal-free oxidative 1,2-alkylarylation of unactivated alkenes with the α-C(sp³)-H bonds of acetonitriles for the synthesis of 5-oxopentanenitriles is presented. In the presence of TBPB (tert-butyl peroxybenzoate), a variety of α-aryl allylic alcohols underwent the 1,2-alkylarylation reaction with acetonitriles, giving 5-oxopentanenitriles in good to excellent yields. This method proceeds via the C(sp³)-H oxidative coupling with the C-C double bond and 1,2-aryl-migration, and represents a new access to acyclic molecules through metal-free oxidative alkene 1,2-alkylarylation. A new oxidative coupling of unactivated terminal alkenes with amides using peroxides is described, in which mono- and difunc-tionalization of alkenes are selectively achieved. In this reaction with amides, the chemoselectivity toward the functionalization of the C(sp³)-H bonds adjacent to the nitrogen atom or the functionalization of the carbonyl C(sp²)-H bonds across alkenes relies on the reaction conditions. Since carbon-based structures of various dimensions, including one-dimensional (1D) carbon nanotubes, two-dimensional (2D) graphene and three-dimensional (3D) carbon foams, have attracted significant attention as microwave absorption fillers, we present an exceptional hetero-junction filler with a 1D–2D–1D feature, achieved by manipulating 2D graphene into 1D carbon fibers in the fiber-extruding process under the electric field. The as-fabricated 1D–2D–1D structural fillers exhibited much-improved dielectric properties and promoted microwave absorption performance in their composites, which is linked to the establishment of enhanced polarization capability, the generation of increased electric loss pathway and the creation of more favorable electromagnetic energy consumption conditions. The results suggest that employing 2D graphene in the 1D–2D–1D nanostructures played the critical role in tuning the electromagnetic response ability, because of its intrinsic electric advantages and dimensional features. To broaden the effective absorption bandwidth, periodic pattern-absorbing structures were designed, which showed combined absorption advantages for various thicknesses. Since manipulating electromagnetic waves with electromagnetic active materials for environmental and electric engineering is a significant task, here a novel prototype is reported by introducing reduced graphene oxide (RGO) interfaces in carbon fiber (CF) networks for a hierarchical carbon fiber/reduced graphene oxide/nickel (CF-RGO-Ni) composite textile. Upon charaterizations of the microscopic morphologies, electrical and magnetic properties, the presence of three-dimensional RGO interfaces and bifunctional nickel nanoparticles substantially influences the related physical properties in the resulting hierarchical composite textiles. Eletromagnetic interference (EMI) shielding performance suggests that the hierarchical composite textiles hold a strong shielding effectiveness greater than 61 dB, showing greater advantages than conventional polymeric and foamy shielding composites. As a polymer-free lightweight structure, flexible CF-RGO-Ni composites of all electromagnetic active components offer unique understanding of the multi-scale and multiple mechanisms in electromagnetic energy consumption. Such a novel prototype of shielding structures along with convenient technology highlight a strategy to achieve high-performance EMI shielding, coupled with a universal approach for preparing advanced lightweight composites with graphene interfaces.
