Abstract

Abstract: A new method for the analysis of anthocyanins in grapes based on a systematic study of the extractability of eleven anthocyanins from grapes has been developed. Microwave assisted extraction was applied as a prior stage to the chromatographic determination of anthocyanins in the extracts. The stability of anthocyanins under the extraction conditions was checked using a standardised extract from grape skins. Temperatures from 50°C up to 150°C were evaluated. A fractional factorial experimental design was developed to analyse the influence on the extraction process of six different extraction variables: solvent (mixtures of methanol and water), stirring, extraction temperature, extraction time, microwave power and extraction volume. The extraction solvent was the most important variable for the recovery of most anthocyanins from grapes. Finally, the influence of the extraction time was also studied. With this new method, anthocyanins can be extracted from grapes in 5min, using 100°C as extraction temperature and 40% methanol in water as the extraction solvent. Repeatability and reproducibility were also checked, the resulting RSDs (n =9) were lower than 7% for glucosides, the main components, and lower than 9% for the acyl derivatives, the compounds found in the lowest concentrations. Abstract

In this work, the feasibility of two commercial products enriched in hydroxytyrosol (HT) as alternative to sulfur dioxide in Syrah red wines was evaluated. The HT enriched products came from synthesis and from olive waste. Wines treated with HT were compared with wines treated with sulfur dioxide at two winemaking stages: bottling and after 6 months of storage in bottle. Minor differences were found in enological parameters and volatile composition (esters, alcohols and acids). Significant differences were observed in color related parameters and sensory analysis. HT wines improved color parameters as well as scents and tasting at bottling. However, after 6 months of storage in bottle HT wines were more oxidized than SO 2 wines. The olfactometry profile of HT wines supported sensory analysis. HT wines showed new odorant zones from both the added product and oxidation. [ABSTRACT FROM AUTHOR]

Abstract

Whole-genome sequencing (WGS) holds great potential as a diagnostic test. However, the majority of patients currently undergoing WGS lack a molecular diagnosis, largely due to the vast number of undiscovered disease genes and our inability to assess the pathogenicity of most genomic variants. The CAGI SickKids challenges attempted to address this knowledge gap by assessing state-of-the-art methods for clinical phenotype prediction from genomes. CAGI4 and CAGI5 participants were provided with WGS data and clinical descriptions of 25 and 24 undiagnosed patients from the SickKids Genome Clinic Project, respectively. Predictors were asked to identify primary and secondary causal variants. In addition, for CAGI5, groups had to match each genome to one of three disorder categories (neurologic, ophthalmologic, and connective), and separately to each patient. The performance of matching genomes to categories was no better than random but two groups performed significantly better than chance in matching genomes to patients. Two of the ten variants proposed by two groups in CAGI4 were deemed to be diagnostic, and several proposed pathogenic variants in CAGI5 are good candidates for phenotype expansion. We discuss implications for improving in silico assessment of genomic variants and identifying new disease genes.
