Abstract Pleural effusion (PE) is excess fluid in the pleural cavity that stems from lung cancer, other diseases like extra-pulmonary tuberculosis (TB) and pneumonia, or from a variety of benign conditions. Diagnosing its cause is often a clinical challenge and we have applied targeted proteomic methods with the aim of aiding the determination of PE etiology. We developed a mass spectrometry (MS)-based multiple reaction monitoring (MRM)-protein-panel assay to precisely quantitate 53 established cancer-markers, TB-markers, and infection/inflammation-markers currently assessed individually in the clinic, as well as potential biomarkers suggested in the literature for PE classification. Since MS-based proteomic assays are on the cusp of entering clinical use, we assessed the merits of such an approach and this marker panel based on a single-center 209 patient cohort with established etiology. We observed groups of infection/inflammation markers (ADA2, WARS, CXCL10, S100A9, VIM, APCS, LGALS1, CRP, MMP9, and LDHA) that specifically discriminate TB-PEs and other-infectious-PEs, and a number of cancer markers (CDH1, MUC1/CA-15-3, THBS4, MSLN, HPX, SVEP1, SPINT1, CK-18, and CK-8) that discriminate cancerous-PEs. Some previously suggested potential biomarkers did not show any significant difference. Using a Decision Tree/Multiclass classification method, we show a very good discrimination ability for classifying PEs into one of four types: cancerous-PEs (AUC: 0.863), tuberculous-PEs (AUC of 0.859), other-infectious-PEs (AUC of 0.863), and benign-PEs (AUC: 0.842). This type of approach and the indicated markers have the potential to assist in clinical diagnosis in the future, and help with the difficult decision on therapy guidance.

Pleural effusion (PE), excess fluid in the pleural space, is often observed in lung cancer patients and also forms due to many benign ailments. Classifying it quickly is critical, but this remains an analytical challenge often lengthening the diagnosis process or exposing patients to unnecessary risky invasive procedures. We tested the analysis of PE using a multiplexed cytokeratin (CK) panel with targeted mass spectrometry–based quantitation for its rapid classification. CK markers are often assessed in pathological examinations for cancer diagnosis and guiding treatment course. We developed methods to simultaneously quantify 33 CKs in PE using peptide standards for increased analytical specificity and a simple CK enrichment method to detect their low amounts. Analyzing 121 PEs associated with a variety of lung cancers and noncancerous causes, we show that abundance levels of 10 CKs can be related to PE etiology. CK-6, CK-7, CK-8, CK-18, and CK-19 were found at significantly higher levels in cancer-related PEs. Additionally, elevated levels of vimentin and actin differentiated PEs associated with bacterial infections. A classifier algorithm effectively grouped PEs into cancer-related or benign PEs with 81% sensitivity and 79% specificity. A set of undiagnosed PEs showed that our method has potential to shorten PE diagnosis time. For the first time, we show that a cancer-relevant panel of simple-epithelial CK markers currently used in clinical assessment can also be quantitated in PEs. Additionally, while requiring less invasive sampling, our methodology demonstrated a significant ability to identify cancer-related PEs in clinical samples and thus could improve patient care in the future.

The Alcathoe bat (Myotis alcathoe von Helversen, Heller, 2001) is a newly described species of the cryptic complex of whiskered bat species living in sympatry across Europe. The habitats preferred by this species are natural, moist and deciduous forests with old trees and water streams. The majority of known locations of this species come from highlands, foothills and mountainous regions while practically not occurring in the lower elevations. They come mainly from cave areas, where bats were caught during autumn swarming, while records in other seasons are much less frequent. Currently, individual sites in Poland also come from lowlands, beyond the range of the mountains and uplands, which indicates a possible wider range of this species. The bats were captured using mist-nests in the Silesian Lowlands for three consecutive years (2013–2015). The localities were placed in various forest environments, ranging in elevation from 100 to 260 m a.s.l. Bats were initially identified based on morphological features, then the correctness of identification was confirmed by molecular methods. Two females and five males of Myotis alcathoe were captured into four new locations from the lowlands of Poland: Przemkow, Glebowice, Chodlewo, and Dalkowskie Jary (Silesian Lowland). Old trees and a small watercourse were important elements in all these locations. Currently, these findings are the northernmost locations of the species in Poland and one of the northernmost locations in Central Europe. The latest findings of the species are located about 50 km to the south, but the vast majority of the rest of locations is from the foothills belt. This finding suggests that the species may inhabit regions much further north than previously suggested, and the limiting factor is the presence of old trees in wet environments.

Successful evacuation of a peculiar ‘colony’ of the wood ant Formica polyctena Först., for years trapped within an old bunker previously used for storing nuclear weapons (see Czechowski et al. 2016), is reported. Using an experimentally installed boardwalk, the imprisoned ants managed to get through the ventilation pipe to their maternal nest on the top of the bunker. In our previous report, we left open the question of how the ‘colony’ could survive seemingly without food. Here we show that the ‘colony’ in the bunker survived and grew thanks to an influx of workers from the source nest above the bunker and mass consumption of corpses of the imprisoned nestmates.

The presented studies are the first one on the occurrence of mites in bat boxes and focuses on Uropodina (Acari: Mesostigmata). Investigation was carried out in Western Poland in October 2015. Guano was collected from 58 bat boxes occupied by 10 species of bats. Excrements from particular bat boxes were placed separately in string bags and transported to the laboratory. The extracted fauna was preserved in 75% ethanol and mites were identified with stereoscopic light microscope. The study revealed only one species of Uropodina mite on bat guano in the studied bat boxes, namely Leiodinychus orbicularis (C.L. Koch, 1839). DOI: http://dx.doi.org/10.5281/zenodo.3835917

Warmer winters may lead to changes in the hibernation behaviour of bats, such as the barbastelle Barbastella barbastellus, which prefers to hibernate at low temperatures. The species is also known for its large annual fluctuations in the number of wintering individuals, so inference about population trends should be based on long-term data. Prior to 2005, analyses indicated stable or even increasing barbastelle population in Poland. We analysed the results of 13 winter bat counts (2005-2017) of the species from 15 of the largest hibernacula, and additional site of 47 small bunkers, in Poland. The total number of wintering individuals remained stable during the study period, because the barbastelle is not a long-distance migrant, this likely reflects the national population trend. On the basis of mean winter air temperatures we divided the country into four thermal regions. Analyses of barbastelle abundance in hibernacula in the four regions revealed a 4.8% annual mean increase in numbers in the coldest region, where mean winter temperatures were below -2°C, annual mean declines of 3.3% and 3.1% in two warmer regions of western Poland, but no trend in the region of intermediate mean winter temperatures of between -1°C and -2°C. Overall, there was a significant, but weak, negative correlation between the abundance of hibernating individuals and the mean winter temperature. On the other hand, the number of individuals hibernating in small bunkers increased, even though the site was located in one of the warm regions. The results indicate a warming climate will likely reduce the use of large, well-insulated winter roosts by species that prefer colder conditions-and that this is already happening. For forest-dwelling bats, such as the barbastelle, for which monitoring schemes are primarily based on winter surveys of large hibernacula, estimations of population trends may consequently become less reliable.
