Since some chemical compounds that are used in laboratories, especially in pathology laboratory are potentially mutagens, lab assistances that are working with chemicals such as Benzene, Xylazine and Formaldehyde for long period of time may be exposed to overload of these carcinogens. Therefore, it is necessary to use an indicator for detecting these occupational exposures. Ames test has been recommended in biomonitoring of environment that has high risk carcinogenicity characteristic."n"nMethods: A total of fifty seven urine samples of forensic medicine laboratory personnel's were extracted by C18 column and then tested by TA100 and TA98 standard strains of Ames assay. Each sample was analyzed with and without activator to detect mutagen and promutagen materials."n"nResults: Levels of mutagenicity were found by TA98 strain without activator in one case as well as with activator in two cases of urine samples of pathology laboratory personnel's. These cases were working in laboratory for long time in all of the workdays."n"nConclusion: Personnel's working in pathology laboratories may have greater risk of cancer and should be take care from these occupational exposures.

Summary: The study was designed to evaluate the effects of metformin on apoptosis and epididymal sperm quality in a rat testicular ischaemia/reperfusion (I/R) injury model. A total of 72 male rats were divided into four groups (n = 18 for each group): group 1 (sham‐operated group), group 2 (metformin group), group 3 (torsion/detorsion [T/D] + saline) and group 4 (T/D + 300 mg kg−1 metformin). Testicular torsion was achieved by rotating the right testis 720° in a clockwise direction for 1 hr. Tissue malondialdehyde (MDA) level and caspase‐3 activity increased and the activities of catalase, superoxide dismutase and glutathione peroxidase decreased in comparison with sham‐operated group 4 hr after detorsion (p < .001). In six rats of each group 24 hr after detorsion, histopathological changes and germ cell apoptosis were significantly deteriorated by measuring mean of seminiferous tubule diameters (MSTD) and TUNEL test. Moreover, 30 days after T/D, sperm concentration and motility were examined in six animals per group. Metformin pre‐treatment reduced MDA and caspase‐3 levels and normalised antioxidant enzyme activities 4 hr after detorsion, and germ cell apoptosis was significantly decreased, and the MSTD, as well as sperm functions, was significantly improved. We designed this experiment to evaluate the effects of nortriptyline on testicular injury after torsion/detorsion (T/D). Ninety-six adult Wistar rats were divided into six groups 16 each in control group (Group 1), sham operated (Group 2), T/D + saline (Group 3), and in groups 4-6; were administered 2, 10 and 20 mg kg−1, i.p. of nortriptyline 30 and 90 min after torsion respectively. The simultaneous analysis of dithiocarbamate fungicide residues on greenhouse and non-greenhouse tomatoes was performed by high-performance liquid chromatography with UV detection. A calibration curve with spiked samples was plotted to determine dithiocarbamate residues in tomato samples. Limits of detection and quantification were approximately 0.05 and 0.2 µg g−1, respectively. The ranges of mean recoveries at five spiking levels for all dithiocarbamates of interest were between 88.2%–95.7% and 99.5%–100.8% in standards and spiked samples, respectively. In this study, 40 greenhouse and 40 non-greenhouse tomato samples were analysed. None of the samples analysed contained dithiocarbamates in excess of the maximum residue levels established by the Codex Committee on Pesticide Residues, except for one greenhouse sample, with ethylenebisdithiocarbamates at 3.2 µg g−1. Aim: one hundred urine samples of abusers were examined for the presence of alkaloid substances in urine. Alkaloids of urine were confirmed with rapid and TLC methods. A thin-layer chromatography (TLC) procedure for the screening and confirmation of urinary codeine and morphine has been developed. Method: Urine samples were daily collected from abusers of Tabriz/Iran. Urine extracts were prepared using silica-gel absorbent resin in column. The resin was rinsed with two buffers. These procedures make PH appropriation, then residual water was removed by vacuum aspiration and adsorbed alkaloid substances were eluted with a organic solvent of methanol and concentrated. After evaporation to dryness with heater, the residue was dissolved in organic solvent of methanol to reach 10 to 40 fold concentration. For screening, the extraction residues were spotted on TLC plates which are developed in chloroform: methanol: Ammoniac (12:1.4:0.2). For confirmation the spots for the two opioids were visualized with acidified iodoplatinate. Codeine and morphine standards are well separated from one another and comparison with samples urinary substances on plates. The limit of detection was a concentration of 300ng/mL. Result: The result of this study has shown that 100 percent of urine samples from addicted in Tabriz (Iran) were contained alkaloid substances and 25 percent of these samples have shown drug interaction. Although aminoglycosides antibiotics are used in children and adult commonly, they have serious side effects such as nephrotoxicity and ototoxicity. In clinical practice, for renal function, the levels of serum creatinine and blood urea nitrogen routinely are measured. Since these parameters have limitations such as unreliability, insensitivity, and nonspecificity, the rapid assessment of renal function based on these patients is very important. Increase in N-acetyl-β-D-glucosaminidase (NAG), a hydrolytic lysosomal enzyme, suggests proximal tubular cell damage. In this study, 32 children aged 2 months through 2 years, treated with gentamicin and amikacin for suspected infections at the pediatric ward of Alborz hospital from September 2006 to February 2007, were enrolled. Serum and fresh urine before and after drug infusion were obtained on the 1st, 3rd, and 5th days of antibiotic treatment. Serum urea and creatinine with urinary creatinine, albumin, NAG, lactate dehydrogenase (LDH) and alkaline phosphatase (AP) activity were then determined. A statistically significant increase in urinary NAG, LDH, and AP on 5th day was found compared with before gentamicin administration (P < 0.001, P < 0.01, P < 0.05, respectively). The urinary NAG activity may be a useful indicator of renal injury in children treated with aminoglycosides drugs compared with other routine clinical indicators. [ABSTRACT FROM AUTHOR]

Background: Swertia spp. (Gentianaceae) grow widely in eastern and southern Asian countries such as Japan, China and India and are used as traditional remedy for gastrointestinal complains because of their bitter principles. Several studies have been carried out on hypoglycemic, hepatoprotective, mono amino oxidase inhibitory and antidepressant effects of these plants and it has been shown that xanthones and iridoids are responsible for their activities. Purpose of the study: In order to gain better knowledge of endemic plants of Flora Iranica, Swertia longifolia Boiss. growing in the northern parts of Iran, was subjected to phytochemical studies. Methods: Dried and milled aerial parts of the plant were extracted with petroleum ether and ethanol of which results of petroleum ether extract has been reported previously. For purification of ethanol extract, it was acidified with acetic acid and subsequently extracted with chloroform and then with n-butanol. The n-butanol extract was analyzed using different chromatographic methods and the structures of the isolated components were established by means of spectroscopic techniques. Results: Four components including an iridoid glycoside (loganic acid), a secoiridoid glycoside (gentiopicroside), a secoiridoid dilactone (gentiolactone) and a nucleoside (uridine) were isolated from n-butanol extract of the plant. Gelatin–hydroxyapatite (HAp) nanocomposites have been prepared by particulate leaching technique using glutaraldehyde (GTA) as cross-linking agent for polymer. The porosity in the scaffolds was controlled using sodium chloride as porogen agent. Microstructural investigation by scanning electron microscopy (SEM), revealed the formation of a well interconnected porous scaffold with pore size in the range of 100–200 μm. X-ray diffraction and Fourier transform infrared spectroscopy were used to confirm the formation of crystalline HAp as well the presence of both constituents in the composite samples. The bioactivity of the samples was evaluated by conducting MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay and cell adhesion tests. The results suggest that the use of GTA in excess of 025% can be detrimental to cell survival. Cell attachment on the nanocomposite scaffold was verified by microscopic analysis. Paraquat (PQ) is a highly toxic herbicide that is used in most of the countries without restriction. The cytotoxic effect of PQ is mediated by radicals, which are the products of PQ reduction in cells. The anti-oxidative action of captopril, an angiotensin-converting enzyme inhibitor, appears to be through its ability to scavenge reactive oxygen species. In this study, the heptoprotective effect of captopril against PQ-induced hepatotoxicity was evaluated using primary cultured rat hepatocytes. Hepatocytes were isolated from male Wistar rats using a two-step collagenase perfusion, following incubation in the presence of captopril at 0.1, 0.2, 0.4 and 0.8 mM with or without PQ (5 mM). Hepatoprotective effects of captopril were studied indicating glutathione level intensity, thiobarbituric acid reactive substances (TBARs) formation, lactate dehydrogenase (LDH) leakage and cell viability every 70 min for 210 min. Captopril at 0.2 mM concentration maintained the LDH leakage, glutathione level and cell viability in the presence of 5 mM PQ. In spite of a significant elevation in TBARs formation in the PQ group, captopril did not show any significant protection. In conclusion, our data reveals that incubation of freshly isolated rat hepa-tocytes with captopril (0.2 mM) significantly protected the hepatocytes against the cytotoxicity of PQ (P < 0.05). Abstract: In this study, five heavy metals in canned tuna fish were determined after digestion by the Association of Official Analytical Chemists methods. Mercury and arsenic levels in canned tuna fish were determined by hydride generation atomic absorption spectrophotometery while cadmium and lead levels were determined by graphite tube atomic absorption spectrophotometery and tin levels were determined by flame atomic absorption spectrophotometery. The metal contents, expressed in μgg−1 wet weight, varied from 0.043 to 0.253 with an average value of 0.117 for mercury, from 0.0369 to 0.2618 with an average value of 0.128 for arsenic, from 0.0046 to 0.0720 with an average value of 0.0223 for cadmium, from 0.0126 to. 0726 with an average value of 0.0366 for lead and non detectable for tin. Several samples were spiked with known amounts of metals. Recoveries of the metals were in the range of 91.7±2.89–99.3±4.03%. The results of this study indicate that tuna fish from the Persian gulf area of Iran have concentrations well below the permissible FAO/WHO levels for these toxic metals. Their contribution to the body burden can be therefore considered negligible and the fish seem to be safe for human consumption. [Copyright &y& Elsevier]

In the present study, the effect of cholecystokinin agonists and antagonists on dependence to morphine in mice has been investigated. Mice were treated subcutaneously with morphine (50, 50 and 75 mg/kg) three times daily for 2-4 days, and a last dose of morphine (50 mg/kg) was administered on day 3, 4 or 5. Withdrawal syndrome (jumping) was precipitated by naloxone (2.5, 5 and 10 mg/kg) which was administered intraperitoneally 2 hr after the last dose of morphine. To study the effects of cholecystokinin receptor agonists or antagonists, 10 injection of morphine (3 administrations each day) for dependence and a dose of 5 mg/kg of naloxone for withdrawal induction were employed. Cholecystokinin-8 (0.001-0.01 mg/kg), low doses of the cholecystokinin agonists caerulein (0.00001 and 0.0001 mg/kg) and, unsulfated cholecystokinin (but not high doses) as well as the antagonists MK-329 (0.5-1 mg/kg) and L-365,260 (0.5-1 mg/kg) elicit reduction of the nalaxone-induced jumping. The inhibition of jumping induced by caerulein was reduced with the selective cholecystokinin antagonists MK-329 and L-365,260. It is concluded that cholecystokinin mechanism(s) may be involved in morphine dependence, that the agonists may act on a presynaptic receptors and that the antagonists may work on postsynaptic receptors. Occupational inhalation exposure to noxious agents is not uncommon. Herein, we present a 26-year-old male student who had accidental acute inhalation exposure to a large quantity of titanium ethanolate and hydrogen chloride in chemistry lab. He was referred to the emergency department of our hospital with low-grade fever, dyspnea, headache, fatigue and myalgia. After 24 hrs of symptomatic treatment (oxygen therapy and acetaminophen), the fever was subsided and the patient discharged home in a good clinical condition. The presented symptoms could be interpreted as a form of metal fume fever. It can therefore be concluded that organo-metallic compound of titanium metal may have the potential to produce metal fume fever in human. [ABSTRACT FROM AUTHOR]

The isocyanates are widely used as precursors of polyurethane products, as well as carbamate insecticides. Toluene 2,4-diisocyanate (TDI) is one of the most important commercially used isocyanates. Humans may be exposed to TDI by inhalation, ingestion, dermal and eye contact. TDI is a powerful irritant to the mucosal membranes of the gastrointestinal and respiratory tracts, eyes and the skin. Pulmonary manifestations, especially occupational asthma, are the predominant manifestations after TDI toxicity. Herein, we present intestinalobstruction as an extraordinary manifestation of acute TDI toxicity after occupational exposure. TDI toxicity may cause intestinal obstruction. [ABSTRACT FROM AUTHOR]

Central nervous system (CNS) inflammation in cases such as head trauma, infection and stroke has been associated with the occurrence of epileptic seizures. Microglia, the principal immune cells in the brain, readily become activated in response to injury, infection or inflammation. The bacterial endotoxin lipopolysaccharide (LPS) induces the activation of microglia and the production of proinflammatory factors including nitric oxide (NO) and prostaglandins (PGs). We examined the effect of LPS on seizure susceptibility of mice, by using the sensitive test, threshold of clonic seizures induced by i.v. infusion of pentylenetetrazole. LPS decreased the seizure threshold in a dose- and time-dependent manner. Pretreatment of mice with the NO synthase inhibitor, NG-nitro-l-arginine methyl ester or cyclooxygenase inhibitor, piroxicam or the opioid receptor antagonist, (−)-naloxone completely reversed the proconvulsant effect of LPS.These results indicate that NO, PGs and endogenous opioid peptides seem to be involved in LPS-induced decrease in seizure threshold. However, the protein expression of Bcl-2 is greater in tissue from rats exposed to paraquat alone and paraquat together with pre- and posttreatment with captopril compared to tissue from untreated control rats and from those treated with captopril alone, which can be due to inflammatory responses of lung tissue. Background: Hypertension is a very common and important disease. There are conflicting reports about mercury, a trace element, in the genesis of hypertension.Methods: In this study we examined the relationship between blood mercury levels and hypertension prevalence in a population-based sample of hypertensive and normotensive patients at the Shariati Hospital and the Tehran Heart Center in Tehran, Iran. A cross sectional sampling of 224 patients, aged 40-80 years, who participated in physical examinations conducted in 2006 were included in this study. The population that participated in this study were sample of hypertensive (n=112) which had essential hypertension and normotensive (n=112) patients which had no history of essential hypertension at the Shariati Hospital and the Tehran Heart Center in Tehran. The consent of all the patients were taken in the written form before the experiments. After selecting the patients the range of blood mercury levels were measured with Flame atomic absorption. Results: The range of blood mercury levels was 0 to 39.55 µg/dL. The mean blood mercury level of hypertensive patients (10.75 +1.23 µg/dL) was higher than that of normotensive patients (1.6 +1.02 µg/dL). There was a significant difference in the mean blood mercury level of normotensive men (1.74 +1.56 µg/dL) versus that of hypertensive men (11.9 +1.38 µg/dL).
