15. Clavibacter tessellarius causing bacterial mosaic of wheat establishes in the Old World.

Abstract

The genus Clavibacter (family Microbacteriaceae) includes gram‐positive actinobacterial species infecting diverse agricultural crops such as alfalfa, maize, pepper, potato, tomato and wheat. Clavibacter tessellarius, causing bacterial mosaic of wheat, was reported for the first time in 1976 in Nebraska (United States). Since then, the pathogen was thought to be restricted to North America (Canada and the United States). While bacterial strains suspected to be C. tessellarius have occasionally been isolated from wheat samples outside North America, occurrence of the pathogen in the Old World has not yet been confirmed by the plant protection authorities. In spring 2020 (January to May) dozens of wheat fields with young plants showing leaf chlorosis and mild mosaic symptoms were surveyed and sampled in southern Iran. Among 192 surveyed wheat fields, gram‐positive, orange‐pigmented actinobacterial strains were isolated from samples of 53 fields. Altogether, 61 bacterial strains were identified as C. tessellarius based on the phenotypic features, PCR‐based molecular tests and pathogenicity assays. Multilocus sequence analysis of five housekeeping genes (atpD, dnaK, gyrB, ppk and rpoB) using 10 representative strains from diverse geographic areas in southern Iran confirmed the phylogenetic status of the strains within C. tessellarius. Our data indicate that the bacterial mosaic pathogen occurs in four southern Iranian provinces including Bushehr, Fars, Kerman and Khuzestan. Thus, occurrence of C. tessellarius outside North America is confirmed. Potential risks associated with the emergence of the pathogen in the Old World, and the need for strict quarantine measures in the area, is further discussed. [ABSTRACT FROM AUTHOR]

Abstract

Cataract is a leading cause of vision loss in the world. Several factors are involved in the development of cataracts, of them; diabetes and uncontrolled plasma glucose have great importance. Therefore, controlling glycemic reduces the risk of related to microvascular complications of diabetes diseases such as cataract. This study aimed to compare fasting plasma glucose (FPG), HgbA1c, and oral glucose tolerance test (OGTT) between Diabetic patients with and without cataract in comparison with the healthy group as control. Among 150 subjects, 50 diabetic patients, 50 diabetic cataracts, and 50 healthy individuals without a history of specific disease were compared with each other. Fasting plasma glucose, OGTT, and HgbA1c were measured by colorimetric methods. SPSS23 software was used to analyze data. P-values less than 5% or lower were considered to be statistically significant. In this study, the mean fasting plasma glucose, HgbA1c, and OGTT in the group of diabetic patients, diabetic with cataract, and healthy groups fasting plasma glucose were as follow, 162.46 mg dL -1 and 184.76 mg dL -1, 92.88 mg dL -1 (P˂0.05), HgbA1c: 7.8, 8.0, 5.8% (P˂0.05) and OGTT: 228 mg dL -1, 246.12 mg dL -1 and 114.32 mg dL -1 (P˂0.05) respectively. Our findings showed that in diabetic patients with and without cataract, fasting plasma levels, HgbA1C, and OGTT are higher than the healthy group. There was a significant relationship between the studied factors and cataracts. As a result, high blood glucose has a definitive role in the development of cataracts. [ABSTRACT FROM AUTHOR]

Abstract

Citrus tristeza virus (CTV) is one of the economically destructive viruses affecting citrus trees worldwide, causing significant losses in fruit production. Comparative genomic studies have shown genetic diversity in various regions of the genome of CTV isolates, which has classified the virus into several genotypes. In recent years, some orange citrumelo-tolerant rootstocks showed yellowing, decline, and vein clearing in northern Iran (Mazandaran province, Sari). We confirmed the presence of CTV in the symptomatic trees by reverse transcription PCR (RT-PCR). The complete genome of a Sari isolate of CTV (Sari isolate) was sequenced using next-generation sequencing (NGS) technology. In addition, phylogenetic analysis, differential gene expression of the virus and identification of its variants in a population were studied. We obtained the final contigs of the virus (nt) and annotated all genomes to viral ORFs, untranslated regions (UTRs), intergenic regions, and 5' and 3' ends of the genome. Phylogenetic analysis of the Sari isolate and other genotypes of CTV showed that the Sari isolates were placed in a distinct cluster without a sister group. Based on the number of specific transcripts (TPM) in CTV RNA -Seq, P13 was the most highly expressed gene related to the host range of the virus and its systemic infection. The ORFs of the polyprotein, P33, and P18 showed variation in a single population of the sari isolate. The CTV has a potential for variation in a population in a host, and these variations may contribute to the best fit of the CTV in different situations. In Iran, whole genome sequencing of the CTV was performed for the first time, and we gained new insights into CTV variation in a population. [ABSTRACT FROM AUTHOR]
