This study describes a new species, Sinocyclocheilus xiejiahuai sp. nov., discovered within a cave located in Hongguo Town, Panzhou City, Guizhou Province, southwestern China, with the type locality in the Nanpanjiang River basin. Phylogenetic trees reconstructed based on mitochondrial genes show that the new species represents an independent evolutionary lineage with large genetic differences, 1.9%–13.8% in mitochondrial Cyt b, from congeners. Morphologically, this species can be differentiated from the 79 species currently classified under the genus Sinocyclocheilus by several characteristics: absence of horn-like structures and indistinct elevation at the head-dorsal junction, absence of irregular black markings on the body lateral and scaleless, eyes large, eye diameter 13% of head length, dorsal-fin rays, iii, 6½, last unbranched ray strong, with serrations along posterior margin, pectoral-fin rays, i, 13, anal-fin rays, iii, 5, pelvic-fin rays, i, 7, lateral line pores 74, gill rakers well developed, nine on first gill arch, pectoral fins short, tip not reaching to pelvic-fin origin. The number of Sinocyclocheilus species has been increased from 79 to 80 since the description of this new species.

Previous studies indicated that adipose tissue significantly influences cancer invasion and lymphatic metastasis. However, the impact of neck adipose tissue (NAT) on lymph node metastasis associated with head and neck cancer remains ambiguous. Here, we systematically assess the classification and measurement criteria of NAT and evaluate the association of adipose tissue and cancer-associated adipocytes with head and neck cancer. We delve into the potential mechanisms by which NAT facilitate cervical lymph node metastasis in head and neck cancer, particularly through the secretion of adipokines such as leptin, adiponectin, and Interleukin-6. Our aim is to elucidate the role of NAT in the progression and metastasis of head and neck cancer, offering new insights into prevention and treatment.

Abstract Objective Systemic chemotherapy is the first-line therapeutic option for head and neck squamous cell carcinoma (HNSCC), but it often fails. This study aimed to develop an effective prognostic model for evaluating the therapeutic effects of systemic chemotherapy. Methods This study utilized CRISPR/cas9 whole gene loss-of-function library screening and data from The Cancer Genome Atlas (TCGA) HNSCC patients who have undergone systemic therapy to examine differentially expressed genes (DEGs). A lipid metabolism-related clustered polygenic model called the lipid metabolism related score (LMRS) model was established based on the identified functionally enriched DEGs. The prediction efficiency of the model for survival outcome, chemotherapy, and immunotherapy response was evaluated using HNSCC datasets, the GEO database and clinical samples. Results Screening results from the study demonstrated that genes those were differentially expressed were highly associated with lipid metabolism-related pathways, and patients receiving systemic therapy had significantly different prognoses based on lipid metabolism gene characteristics. The LMRS model, consisting of eight lipid metabolism-related genes, outperformed each lipid metabolism gene-based model in predicting outcome and drug response. Further validation of the LMRS model in HNSCCs confirmed its prognostic value. Conclusion In conclusion, the LMRS polygenic prognostic model is helpful to assess outcome and drug response for HNSCCs and could assist in the timely selection of the appropriate treatment for HNSCC patients. This study provides important insights for improving systemic chemotherapy and enhancing patient outcomes.

Previous studies have found that miR-335 is highly expressed in type II diabetes mellitus (T2DM) models and is related to insulin secretion, but there are few studies on the regulatory effects of miR-335-3p on insulin resistance and macrophage polarization in T2DM patients. This study aims to explore the effects of miR-335-3p on insulin resistance and macrophage polarization in T2DM patients. Blood glucose (insulin tolerance tests, glucose tolerance tests) and body weight of the T2DM model were measured; macrophages from adipose tissue were isolated and cultured, and the number of macrophages was detected by F4/80 immunofluorescence assay; the Real-time quantitative polymerase chain reaction (qPCR) assay and Western blot assay were used to detect the miR-335-3p expression levels, insulin-like growth factor 1 (IGF-1), M1-polarizing genes (inducible nitric oxide synthase [iNOS] and TNF-α) as well as M2-polarizing genes (IL-10 and ARG-1). The targeting link between miR-335-3p and IGF-1 was confirmed using bioinformatics and dual luciferase assay. The results showed that miR-335-3p expression level in adipose tissue of the T2DM model was significantly decreased, and the mice’s body weight and blood glucose levels dropped considerably, miR-335-3p inhibited the number of macrophages, inhibiting the iNOS and TNF-α relative mRNA expression levels, and up-regulated the IL-10 and ARG-1 relative mRNA expression levels, miR-335-3p negatively regulated target gene IGF-1, IGF-1 significantly increased the iNOS and TNF-α mRNA and protein expression levels, decreasing the IL-10 and ARG-1 mRNA and protein expression levels, indicating that miR-335-3p could affect the T2DM process by regulating macrophage polarization via IGF-1.

Cholesterol is the most abundant sterol molecule in mammalian cells, which not only constitutes the cell membrane but also plays essential roles in the synthesis of important hormones, synapse formation, and cell signal transduction. The effect of hypercholesterolemia on hearing has been studied extensively, and multiple studies have demonstrated that hypercholesterolemia is a risk factor for hearing loss. However, the impact of cholesterol homeostasis within auditory cells on peripheral auditory development and maintenance has not been evaluated in detail. Mutations in certain cholesterol metabolism-related genes, such as NPC1, SERAC1, DHCR7, and OSBPL2, as well as derivatives of cholesterol metabolism-related ototoxic drugs, such as β-cyclodextrin, can lead to disruptions of cholesterol homeostasis within auditory cells, resulting in hearing loss. This article aims to review the impact of cholesterol homeostasis within auditory cells on the peripheral auditory function from the following two perspectives: (1) changes in cholesterol homeostasis regulatory genes in various hearing loss models; (2) mechanisms underlying the effects of some drugs that have a therapeutic effect on hearing loss via regulating cholesterol homeostasis. This article aims to summarize and analyze the impact of disruption of cellular cholesterol homeostasis within auditory cells on hearing, in order to provide evidence regarding the underlying mechanisms.

In order to provide the basic data of lithology modeling with high resolution in the early stage of coal precise mining geological guarantee, this paper established the p-wave impedance model of wedge-shaped coal seam according to the thickness and storage conditions of coal measure strata based on geostatistical inversion theory. The influence of different probability density functions and transverse range parameters on thickness inversion of coal seam are analyzed through multiple inversion experiments. The effect of inversion under different amount and location constraints is further discussed. Experimental results show that geostatistical inversion parameters have a great influence on the inversion results. Reasonable probability density function, lateral range and constrained well selection can improve the accuracy of inversion. Results shows that geostatistical inversion can predict about 1 m thin coal seam, and the error range between the inversion coal seam thickness and the logging data is 1.82 % ~12.24 %. It is therefore feasible to predict the thickness of thin coal seam, and can provide valuable modeling data for the early lithologic geological modeling of "transparent mine".

Abstract Objective We aim to review the roles of plasmacytoid dendritic cells (pDCs) in head and neck squamous cell carcinoma (HNSCC) and explore the effects of hypoxia on the tolerogenic transformation of pDCs. Background pDCs, best known as professional type I interferon‐secreting cells, play key roles in immune surveillance and antitumor immunity. Recently, pDCs have been shown to be tolerogenic and correlate with poor prognosis in a variety of cancers, including HNSCC. However, it remains unclear what drives the tolerogenic transformation of pDCs in the HNSCC microenvironment. Hypoxia, a prominent hallmark of the tumor microenvironment (TME) of HNSCC, can interfere with multiple immune cells and establish an immunosuppressive TME. Methods In this review, we summarize the antitumor and protumor functions of pDCs, explore the effects of hypoxia on the migration and maturation of pDCs, and discuss related mechanisms in HNSCC. Conclusions pDCs mainly display protumor functions in HNSCC. The hypoxic TME in HNSCC can enhance the migration of pDCs and inhibit the differentiation and maturation of pDCs, promoting the tolerogenic phenotype of pDCs.

A great number of patients with Coronavirus Disease 2019 (COVID-19) experience olfactory dysfunction, typically of a short duration and with a high incidence rate, during the early stages of infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). This kind of olfactory dysfunction appears more likely in young people and women. This study presents a review of the clinical features and pathogenic mechanism of the olfactory dysfunction related to SARS-CoV-2 infection, aiming to provide a clinical reference for the diagnosis, differential diagnosis, treatment, and prevention of olfactory dysfunction in COVID-19 patients.

The transcriptomic landscape of mice with primary auditory neurons degeneration (PAND) indicates key pathways in its pathogenesis, including complement cascades, immune responses, tumor necrosis factor (TNF) signaling pathway, and cytokine-cytokine receptor interaction. Toll-like receptors (TLRs) are important immune and inflammatory molecules that have been shown to disrupt the disease network of PAND. In a PAND model involving administration of kanamycin combined with furosemide to destroy cochlear hair cells, Tlr 2/4 double knockout (DKO) mice had auditory preservation advantages, which were mainly manifested at 4–16 kHz. DKO mice and wild type (WT) mice had completely damaged cochlear hair cells on the 30th day, but the density of spiral ganglion neurons (SGN) in the Rosenthal canal was significantly higher in the DKO group than in the WT group. The results of immunohistochemistry for p38 and p65 showed that the attenuation of SGN degeneration in DKO mice may not be mediated by canonical Tlr signaling pathways. The SGN transcriptome of DKO and WT mice indicated that there was an inverted gene set enrichment relationship between their different transcriptomes and the SGN degeneration transcriptome, which is consistent with the morphology results. Core module analysis suggested that DKO mice may modulate SGN degeneration by activating two clusters, and the involved molecules include EGF, STAT3, CALB2, LOX, SNAP25, CAV2, SDC4, MYL1, NCS1, PVALB, TPM4, and TMOD4.

Presbycusis contributes to cognitive decline and Alzheimer's disease. However, most research in this area involves clinical observations and statistical modeling, and few studies have examined the relationship between hearing loss and the molecular changes that lead to cognitive dysfunction. The present study investigated whether hearing loss contributes to dementia in the absence of aging and noise using a mouse model of severe bilateral hearing loss induced by kanamycin (1000 mg/kg) and furosemide (400 mg/kg). Immunohistochemistry, silver staining, immunofluorescence analysis, and Western blotting were used to observe pathological changes in different regions of the hippocampus in animals with hearing loss. Changes in the cognitive function of animals with hearing loss were assessed using the Morris water maze test. The results showed that neurons began to degenerate 60 days after hearing loss, and this degeneration was accompanied by structural disorganization and decreased neurogenesis. The level of phosphorylated tau increased over time. Increases in escape latency and distance traveled during the training phase of the Morris water maze test were observed 90 days after hearing loss. Activated microglia and astrocytes with increased levels of inflammatory cytokines, such as tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) were detected in the hippocampus. These results suggest that hearing loss alone causes neuronal degeneration, inhibition of neurogenesis, increased tau protein phosphorylation, and increased neuroinflammation in the hippocampus. Early intervention in individuals with hearing loss may reduce the risk of cognitive decline.

Purpose: Head and neck squamous cell carcinoma (HNSCC) is a highly invasive malignancy with poor survival. Perforin (PRF1) plays essential roles in host immunity. Our research intended to identify the correlations of PRF1 with clinical prognosis and tumor immune infiltration in HNSCC. Methods: We explored PRF1 expression and its associations with the clinical features of HNSCC via the Tumor Immune Estimation Resource (TIMER), Oncomine and The Cancer Genome Atlas (TCGA) databases. The prognostic value of PRF1 for HNSCC was further explored by Kaplan–Meier plotter and TIMER. Finally, the relation between PRF1 and immune infiltration in HNSCC was estimated via CIBERSORT and TIMER. Results: PRF1 expression was remarkably elevated in HNSCC and associated with clinical stage and HPV infection. High PRF1 expression predicted favorable outcomes in HNSCC, especially in HPV+ HNSCC. Moreover, higher infiltration of CD8+ T cells and CD4+ T cells were found in the PRF1high group of HNSCC. PRF1 expression in HNSCC was strongly correlated with infiltrating CD8+ T cells and dendritic cells (DCs), with higher relevance in HPV+ HNSCC. Conclusion: Our findings suggested that PRF1 could be a novel prognostic biomarker in HNSCC and that its expression was related to immune cell infiltration, which was impacted by HPV status.

Current methods for the early diagnosis of cancer can be invasive and costly. In recent years, exosomes have been recognized as potential biomarkers for cancer diagnostics. The common methods for quantitative detection of exosomes, such as nanoparticle tracking analysis (NTA) and flow cytometry, rely on large-scale instruments and complex operation, with results not specific for cancer. Herein, we present a tri-channel electrochemical immunobiosensor for enzyme-free and label-free detecting carcino-embryonic antigen (CEA), neuron-specific enolase (NSE), and cytokeratin 19 fragments (Cyfra21-1) from exosomes for specific early diagnosis of lung cancer. The electrochemical immunobiosensor showed good selectivity and stability. Under optimum experimental conditions, the linear ranges were from 10−3 to 10 ng/mL for CEA, 10−4 to 102 ng/mL for NSE, and 10−3 to 102 ng/mL for Cyfra21-1, and a detection limit down to 10−4 ng/mL was achieved. Furthermore, we performed exosome analysis in three kinds of lung cancer. The results showed a distinct expression level of exosomal markers in different types. These works provide insight into a promising alternative for the quantification of exosomal markers in specific diseases in the following clinical bioassays.

In order to resolve the contradiction between the high demand for water resources,water ecology and water environment in the Guangdong-Hong Kong-Macao Greater Bay Area (GBA) and the insufficient supervision capabilities in the water conservancy industry,and achieve the transformation from passive management to active management of key drinking water sources in the GBA,this paper carries out supervisory monitoring of key drinking water sources through remote sensing macro monitoring,UAV cruise monitoring,fixed-point monitoring and other intelligent monitoring technology system.The results show that:①After application of a supervisory monitoring technology system for key drinking water sources,the full-coverage,dynamic and quasi-real-time monitoring of 35 key water sources in the GBA is realized,and the risk sources and water problems can be identified efficiently.②For identifying five suspected risk sources in water source areas,namely construction projects,key river (reservoir) tributaries,temporary shipping stops,material stacking places,agricultural production activities by remote sensing macro monitoring and UAV cruise monitoring,the accuracy rate is 65.7%.③For the chlorophyll a and CODMn monitoring of the water source of main Guangzhou-Dongguan-Huizhou Dongjiangbei River by remote sensing macro monitoring and artificial fixed-point monitoring during the same period,the monitoring results of the two technical means are in good agreement.Therefore,with application of the technology system in supervision and management of key drinking water sources,the smart water conservancy supervision and business management are deeply integrated,and the effective technical means for supervision and management are available.

Noise-induced hearing loss (NIHL) is characterized by damage to cochlear neurons and associated hair cells; however, a systematic evaluation of NIHL pathogenesis is still lacking. Here, we systematically evaluated differentially expressed genes of 22 cochlear samples in an NIHL mouse model. We performed Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis and weighted gene co-expression network analysis (WGCNA). Core modules were detected using protein–protein interactions and WGCNA with functional annotation, diagnostic value evaluation, and experimental validation. Pooled functional annotation suggested the involvement of multiple inflammatory pathways, including the TNF signaling pathway, IL-17 signaling pathway, NF-kappa B signaling pathway, rheumatoid arthritis, and p53 signaling pathway. The core modules suggested that responses to cytokines, heat, cAMP, ATP, mechanical stimuli, and immune responses were important in NIHL pathogenesis. These activities primarily occurred on the external side of the plasma membrane, the extracellular region, and the nucleus. Binding activities, including CCR2 receptor binding, protein binding, and transcription factor binding, may be important. Additionally, the hub molecules with diagnostic value included Relb, Hspa1b, Ccl2, Ptgs2, Ldlr, Plat, and Ccl17. An evaluation of Relb and Hspa1b protein levels showed that Relb was upregulated in spiral ganglion neurons, which might have diagnostic value. In conclusion, this study indicates that the inflammatory response is involved in auditory organ changes in NIHL pathogenesis; moreover, several molecules and activities have essential and subtle influences that have translational potential for pharmacological intervention.

Autophagy, a highly conserved cellular mechanism, plays an essential role in the development and pathology of many central and peripheral nervous system diseases. The auditory system, especially hair cells (HCs) and spiral ganglion neurons (SGNs) in the inner ear, are postmitotic cells, which are extremely reliant on cellular homeostasis and energy supply. Therefore, autophagy may be involved in contributing to and facilitating the normal function of inner ear cells. Recently, studies on hearing loss induced by ototoxic drugs, noise exposure and other factors have revealed that autophagy could serve in an antioxidative capacity and could possess the potential to treat sensorineural hearing loss (SNHL). Therefore, here we review previous studies concerning autophagy and SNHL to gain insight into the role of autophagic mechanisms in inner ear disorders.

The incidences of presbycusis and dementia are high among geriatric diseases. Presbycusis is the general term applied to age-related hearing loss and can be caused by many risk factors, such as noise exposure, smoking, medication, hypertension, family history, and other factors. Mutation of mitochondrial DNA in hair cells, spiral ganglion cells, and stria vascularis cells of the cochlea is the basic mechanism of presbycusis. Dementia is a clinical syndrome that includes the decline of cognitive and conscious states and is caused by many neurodegenerative diseases, of which Alzheimer’s disease (AD) is the most common. The amyloid cascade hypothesis and tau hypothesis are the two major hypotheses that describe the AD pathogenic mechanism. Recent studies have shown that deposition of Aβ and hyperphosphorylation of the tau protein may cause mitochondrial dysfunction. An increasing number of papers have reported that, on one hand, the auditory system function in AD patients is damaged as their cognitive ability declines and that, on the other hand, hearing loss may be a risk factor for dementia and AD. However, the relationship between presbycusis and AD is still unknown. By reviewing the relevant literature, we found that the SIRT1-PGC1α pathway and LKB1 (or CaMKKβ)-AMPK pathway may play a role in the preservation of cerebral neuron function by taking part in the regulation of mitochondrial function. Then vascular endothelial growth factor signal pathway is activated to promote vascular angiogenesis and maintenance of the blood–brain barrier integrity. Recently, experiments have also shown that their expression levels are altered in both presbycusis and AD mouse models. Therefore, we propose that exploring the specific molecular link between presbycusis and AD may provide new ideas for their prevention and treatment.

Obtaining information on tectonic tendencies is a prerequisite for intelligent and accurate mining in mines. In the special mine environment, the co‐polarized ground‐penetrating radar can only identify the spatial location of faults, and it is difficult to analyse the inclination information of fault structures. This paper proposes a mine full‐polarimetric ground‐penetrating radar fault tendency detection method based on this. First, based on the stacking characteristics of the coal depositional, this paper analyses the propagation law of the pulse electromagnetic wave in the coal seam and puts forward the assumption of the overlapping echo reflection of the fault structure. The reasonableness of the fault reflection assumption is verified through a numerical simulation study. Second, based on the cutting relationship of the fault to the coal seam, we divided the reflection structure of the fault structure into plane scattering and dihedral angle scattering. We realized the mingled echoes' decomposition using the improved Yamaguchi decomposition technique. To analyse the applicability of the modified Yamaguchi and Freeman decomposition methods in the identification of fault inclination, we use the upright fault simulation data for the discussion, and we find that the improved Yamaguchi decomposition method is more advantageous in the identification of fault inclination in the mine. The decomposition results based on the simulation data of fault models with different dip angles found that when the dip angle of the fault is less than 90°, the scattering of the fault structure is dominated by planar scattering and dihedral angle scattering; when the dip angle of the fault is greater than or equal to 90°, the scattering of the fault structure is dominated by planar scattering, and the scattering power of the dihedral angle model is zero. By analysing the effect of fault strike on the decomposition results, it is found that the fault strike angle has little effect on the identification of fault tendency. Finally, the application potential of this paper's method is tested by constructing complex numerical models and probing experiments. This research significantly extends the previous studies by providing a quantitative analysis of the impact of defect size on the mechanical integrity of 5A90-T3S Al–Mg–Li alloy joints via refill friction stir spot welding (RFSSW). It particularly elucidates the pronounced effects of the defects at both vertical and horizontal interfaces on the mechanical properties (such as shear and tensile strengths). Notably, larger defects at the vertical interface are found to critically undermine the tensile strength, whereas the defects at the horizontal interface primarily diminish the shear strength. Moreover, the presence of cavity defects at the interface junctures is identified as a pivotal factor severely compromising joint performance. Comprehensive mechanical testing coupled with detailed statistical analysis underscores the critical importance of precise control over welding parameters and the implementation of rigorous inspection protocols to minimize defect impacts, thereby bolstering the structural integrity of aerospace components. Damage to the intestinal mucosal barrier and dysbiosis of the gut microbiota are critical factors in HIV progression, reciprocally influencing each other. Besides bacteria, the fungal microbiota, a significant component of the gut, plays a pivotal role in this dysregulation. This study aims to investigate changes in the gut mucosal barrier and mycobiota during the initial stages of HIV infection, focusing on the involvement of intestinal fungi and their secretions in mucosal damage. Peripheral blood, intestinal mucosa, and fecal samples were collected from 13 asymptomatic HIV‐infected individuals at the non‐AIDS stage and 13 healthy controls. Assessments included colonoscopy, immune function analysis, and measurement of mucosal damage markers (LPS, I‐FABP, and D‐LA) and inflammatory cytokines (IL‐6 and IL‐18). Additionally, Claudin‐1 levels in mucosal samples and fungal profiles in fecal samples were evaluated. The study found that colonic abnormalities were significantly more prevalent in the HIV group compared to healthy controls (p < 0.001) and Claudin‐1 levels were notably lower in the HIV group (p < 0.001). Candida albicans (p = 0.0084), its secretion SAP1 (p = 0.023), and the levels of IL‐18 (p = 0.0016) and IL‐6 (p < 0.001) were all significantly higher in the HIV group. CD4+ T‐cell counts were positively correlated with Claudin‐1 expression (p = 0.034, r = 0.417). Candida albicans showed negative correlations with several virulence factors, while other fungi exhibited varied correlations. Additionally, Claudin‐1 levels were significantly negatively correlated with Candida albicans (p = 0.013, r = −0.668), SAP1 (p = 0.027, r = −0.609), IL‐18 (p < 0.001, r = −0.922), and IL‐6 (p < 0.001, r = −0.920). Overall, these findings suggest that asymptomatic HIV‐infected individuals have already exhibited intestinal mucosal damage in the early stage and highlight the critical role of Candida albicans and its secretions in early‐stage intestinal mucosal barrier damage. The article in the Australasian Journal of Dermatology discusses a case of a 57-year-old female diagnosed with pemphigus foliaceus (PF) with prominent neutrophilic pustules. The patient was treated with Acitretin after initial treatments with antihistamines and traditional Chinese medicine. The use of Acitretin led to a significant improvement in the patient's condition, with no recurrence of skin lesions observed three months post-discharge. The article highlights the effectiveness of Acitretin in treating isolated PF with prominent neutrophilic pustules, emphasizing the importance of accurate diagnosis and appropriate treatment.
