The oxidative pyrolysis kinetics of coal before and after immersion effect was studied by thermogravimetric experiments. Non-isothermal thermogravimetric analysis data were analyzed at four heating rates of 5, 10, 20 and 30 K/min. Furthermore, temperature-programmed experiments and Fourier-transform infrared (FTIR) spectroscopy were employed to understand the oxidation activity and chemical structure of raw coal and soaked coal. After coal immersion effect, the increase in the content of active functional groups, the increase in the concentration of gaseous products, and the decrease in crossing point temperature during oxidation process all indicate that soaked coal is easier to oxidize and spontaneously combust. Lastly, four model-free methods such as Flynn-Wall-Ozawa (FWO), Kissinger-Akahira-Sunose (KAS), Friedman and Kissinger methods are employed to calculate the activation energy (Eα). Kinetic analysis reveals that Eα value obtained by the former three methods significantly depends on the conversion of coal oxidative pyrolysis process, and the average Eα value of soaked coal obtained by four methods is lower than that of raw coal. For the kinetic analysis of coal oxygen adsorption and combustion stages, it is more reliable to adopt the FWO and KAS methods in turn. This research helps to better understand the mechanism of enhanced oxidation activity of soaked coal and optimizes the calculation method of kinetic parameters in different oxidation stages of coal.

Emodin was one of the hydrophobic active molecules, the solubility bioavailability of which was improved by loading in a certain matrix. In this study, the microcapsules of emodin-casein complexes were prepared with casein as matrix with ultrasound treatment. The structural properties of the complexes were analyzed by fluorescence spectroscopy, Fourier transform infrared spectroscopy, scanning electron microscopy. The thermal properties, DPPH and ABTS+ free radical scavenging activities of the complexes were measured. In addition, the release profiles of emodin from the complexes were evaluated by simulated gastrointestinal digestion experiment. The results showed that the ultrasound treatment within 1~3 min duration had not significant effect on the fluorescence properties of the complexes with the addition of emodin at 8~10 μg/mg. Furthermore, there were no significant effect of ultrasound treatment on the FTIR spectra and morphology of emodin-casein complexes. In addition, the DPPH free radical scavenging activity of the complexes had the lowest value with 1 min-ultrasound treatment, but the highest value of ABTS+ free radical scavenging activity among other samples with the same emodin addition. The release rate of emodin from the emodin-casein complexes during simulated gastrointestinal digestion was improved by 1 min-ultrasound treatment, but retarded by 5 min-ultrasound treatment. The results would provide references for the application of ultrasound treatment in the properties' regulation of emodin-casein complexes.

Abstract Two major earthquakes (MW 7.8 and MW 7.7) ruptured left-lateral strike-slip faults of the East Anatolian Fault Zone (EAFZ) on February 6, 2023, causing >59,000 fatalities and ~$119B in damage in southeastern Türkiye and northwestern Syria. Here we derived kinematic rupture models for the two events by inverting extensive seismic and geodetic observations using complex 5-6 segment fault models constrained by satellite observations and relocated aftershocks. The larger event nucleated on a splay fault, and then propagated bilaterally ~350 km along the main EAFZ strand. The rupture speed varied from 2.5-4.5 km/s, and peak slip was ~8.1 m. 9-h later, the second event ruptured ~160 km along the curved northern EAFZ strand, with early bilateral supershear rupture velocity (>4 km/s) followed by a slower rupture speed (~3 km/s). Coulomb Failure stress increase imparted by the first event indicates plausible triggering of the doublet aftershock, along with loading of neighboring faults.

Summary: Subarachnoid hemorrhage (SAH) is a stroke subtype with high mortality, and its severity is closely related to the short-term prognosis of SAH patients. S100 calcium-binding protein A9 (S100A9) has been shown to be associated with some neurological diseases. In this study, the concentration of S100A9 in clinical cerebrospinal fluid samples was detected by enzyme-linked immunosorbent assay (ELISA), and the relationship between S100A9 and the prognosis of patients was explored. In addition, WT mice and S100A9 knockout mice were used to establish an in vivo SAH model. Neurological scores, brain water content, and histopathological staining were performed after a specified time. A co-culture model of BV2 and HT22 cells was treated with heme chloride to establish an in vitro SAH model. Our study confirmed that the expression of S100A9 protein in the CSF of SAH patients is increased, and it is related to the short-term prognosis of SAH patients. S100A9 protein is highly expressed in microglia in the central nervous system. S100A9 gene knockout significantly improved neurological function scores and reduced neuronal apoptosis. S100A9 protein can activate TLR4 receptor, promote nuclear transcription of NF-κB, increase the activation of inflammatory body, and ultimately aggravate nerve injury.

BackgroundThe relationship between plant-based diets and gallstone disease has been debated. This study aimed to shed light on the association between plant-based dietary index and the risk of developing gallstone disease.MethodsEligible participants were selected from National Health and Nutrition Examination Survey (NHANES) 2017-2020. Three plant-based diet indexes (PDI, healthy PDI, unhealthy PDI) were calculated using data from two NHANES 24-h dietary recall interviews. Restricted Cubic Spline and multivariate logistic regression were used to analyze the associations. Subgroup analysis was adopted to make the results more robust.ResultsA total of 5673 eligible participants were analyzed. After adjusting for various confounding variables, uPDI was positively associated with gallstone disease (OR = 1.53, 95%CI: 1.02-2.29). No association was found between PDI/hPDI and gallstone disease (p > 0.05). The results of subgroup analysis did not show any positive association between uPDI and gallstones in specific groups.ConclusionOur study shows that the elevated uPDI are linked to a higher risk of gallstone disease.

On 19 September 2022, a major earthquake struck the northwestern Michoacán segment along the Mexican subduction zone. A slip model is obtained that satisfactorily explains geodetic, teleseismic, and tsunami observations of the 2022 event. The preferred model has a compact large‐slip patch that extends up‐dip and northwestward from the hypocenter and directly overlaps a 1973 Mw 7.6 rupture. Slip is concentrated offshore and below the coast at depths from 10 to 30 km with a peak value of ∼2.9 m, and there is no detected coseismic slip near the trench. The total seismic moment is 3.1 × 10^20 N·m (Mw 7.6), 72% of which is concentrated in the first 30 s. Most aftershocks are distributed in an up‐dip area of the mainshock that has small coseismic slip, suggesting near‐complete strain release in the large‐slip patch. Teleseismic P waveforms of the 2022 and 1973 earthquakes are similar in duration and complexity with high cross‐correlation coefficients of 0.68–0.98 for long P to PP signal time windows, indicating that the 2022 earthquake is a quasi‐repeat of the 1973 earthquake, possibly indicating persistent frictional properties. Both the events produced more complex P waveforms than comparable size events along Guerrero and Oaxaca, reflecting differences in patchy locking of the Mexican megathrust.

Abstract Strong tsunami excitation from slow rupture of shallow subduction zone faults is recognized as a key concern for tsunami hazard assessment. Three months after the 22 July 2020 magnitude 7.8 thrust earthquake struck the plate boundary below the Shumagin Islands, Alaska, a magnitude 7.6 aftershock ruptured with complex intraplate faulting. Despite the smaller size and predominantly strike-slip faulting mechanism inferred from seismic waves for the aftershock, it generated much larger tsunami waves than the mainshock. Here we show through detailed analysis of seismic, geodetic, and tsunami observations of the aftershock that the event implicated unprecedented source complexity, involving weakly tsunamigenic fast rupture of two intraplate faults located below and most likely above the plate boundary, along with induced strongly tsunamigenic slow thrust slip on a third fault near the shelf break likely striking nearly perpendicular to the trench. The thrust slip took over 5 min, giving no clear expression in seismic or geodetic observations while producing the sizeable far-field tsunami.

C-X-C chemokine receptor type 4 (CXCR4) is critical for homeostasis of the adaptive and innate immune system in some CNS diseases. Bruton's tyrosine kinase (BTK) is an essential kinase that regulates inflammation in immune cells through multiple signaling pathways. This study aims to explore the effect of CXCR4 and BTK on neuroinflammation in the pathogenesis of early brain injury (EBI) after subarachnoid hemorrhage (SAH). Our results showed that the expression of CXCR4 and p-BTK increased significantly at 24 h after SAH in vivo and in vitro. Ibrutinib improved neurological impairment, BBB disruption, cerebral edema, lipid peroxidation, neuroinflammation and neuronal death at 24 h after SAH. Inhibition of BTK phosphorylation promoted the in vitro transition of hemin-treated proinflammatory microglia to the anti-inflammatory state, inhibited the p-P65 expression and microglial pyroptosis. NLRP3 deficiency can significantly reduce pyroptosis in SAH mice. Moreover, CXCR4 inhibition can suppress NLRP3-mediated pyroptosis, NF-κB activation and NOX2 expression in vitro, and ibrutinib can abolish CXCR4-aggravated BBB damage and pyroptosis in EBI after SAH. The levels of CXCR4 in CSF of SAH patients is significantly increased, and it is positively correlated with GSDMD and IL-1β levels, and have a moderate diagnostic value for outcome at 6-month follow-up. Our findings revealed the effect of CXCR4 and P-BTK on NLRP3-mediated pyroptosis and lipid peroxidation after SAH in vivo and in vitro, and the potential diagnostic role of CXCR4 in CSF of SAH patients. Inhibition of CXCR4-BTK axis can significantly attenuate NLRP3-mediated pyroptosis and lipid peroxidation by regulating NF-κB activation in EBI after SAH.

Human dental pulp stem cells (hDPSCs) promote recovery after ischemic stroke; however, the therapeutic efficacy is limited by the poor survival of transplanted cells. For in vitro experiments in the present study, we used oxygen-glucose deprivation/reoxygenation in hDPSCs to mimic cell damage induced by ischemia/reperfusion. We found that miRNA-34a-5p (miR-34a) was elevated under oxygen-glucose deprivation/reoxygenation conditions in hDPSCs. Inhibition of miR-34a facilitated the proliferation and antioxidant capacity and reduced the apoptosis of hDPSCs. Moreover, dual-luciferase reporter gene assay showed WNT1 and SIRT1 as the targets of miR-34a. In miR-34a knockdown cell lines, WNT1 suppression reduced cell proliferation, and SIRT1 suppression decreased the antioxidant capacity. Together, these results indicated that miR-34a regulates cell proliferation and antioxidant stress via targeting WNT1 and SIRT1, respectively. For in vivo experiments, we injected genetically modified hDPSCs (anti34a-hDPSCs) into the brains of mice. We found that anti34a-hDPSCs significantly inhibited apoptosis, reduced cerebral edema and cerebral infarct volume, and improved motor function in mice. This study provides new insights into the molecular mechanism of the cell proliferation and antioxidant capacity of hDPSCs, and suggests a potential gene that can be targeted to improve the survival rate and efficacy of transplanted hDPSCs in brain after ischemic stroke.

Abstract Background Peutz-Jeghers syndrome (PJS) is a rare, autosomal dominant disorder caused by germline mutations of STK11/LKB1, with an increased risk of tumors at multiple sites. Intraductal oncocytic papillary neoplasm (IOPN) is a unique subtype of intraductal papillary neoplasm of the bile duct (IPNB) defined by a premalignant neoplasm with intraductal papillary or villous growth of biliary-type epithelium. IOPN has a distinct mutation profile compared with both IPNB and intraductal papillary mucinous neoplasm (IPMN). Case presentation We herein describe the case of a 44-year-old woman who presented as polyps in the intestinal lumen of sigmoid colon and a 3.1 × 2.1 cm mass in the left lobe of liver. Gross feature revealed a cystic papillary mass and the neoplasm had a clear boundary with the surrounding liver tissue. Histology revealed complex papillary structures, a small amount of fine fibrovascular cores and immunohistochemistry showed extensive positive for MUC5AC, MUC6, CD117. Therefore, histological and immunohistochemical examination of the liver tumor suggested the diagnosis of IOPN. Next-generation sequencing (NGS) revealed other than STK11 germline mutation, the tumor also harbors GNAS somatic mutation at codon 478 and EGFR amplification. Conclusion To our knowledge, this is the first report of IOPN arising in PJS. This case enlarges the spectrum of PJS related tumors and genetic rearrangements in IOPN.

Abstract Background Neuroinflammation and neuronal apoptosis are closely associated with a poor prognosis in patients with subarachnoid hemorrhage (SAH). We investigated the role of C–C motif chemokine receptor 2 (CCR2) in SAH. Methods Pre-processed RNA-seq transcriptome datasets GSE167110 and GSE79416 from the Gene Expression Omnibus (GEO) database were screened for genes differentially expressed between mice with SAH and control mice, using bioinformatics analysis. The endovascular perforation model was performed to establish SAH. RS504393 (a CCR2 antagonist) and LY294002 (PI3K inhibitor) were administered to explore the mechanism of neuroinflammation after SAH. SAH grading, neurological scoring, brain water content and blood–brain barrier (BBB) permeability determination, enzyme-linked immunosorbent assay (ELISA), western blotting, and immunofluorescence were performed. An in vitro model of SAH was induced in H22 cells by hemin treatment. The protective mechanism of CCR2 inhibition was studied by adding RS504393 and LY294002. Clinical cerebrospinal fluid (CST) samples were detected by ELISA. Results Expression of CCR2 was upregulated in both datasets and was identified as a hub gene. CCR2 expression was significantly upregulated in the cytoplasm of neurons after SAH, both in vitro and in vivo. RS significantly reduced the brain water content and blood–brain barrier permeability, alleviated neuroinflammation, and reduced neuronal apoptosis after SAH. Additionally, the protective effects of CCR2 inhibition were abolished by LY treatment. Finally, the levels of CCR2, inflammatory factors, and apoptotic factors were elevated in the CSF of patients with SAH. CCR2 levels were associated with patient outcomes at the 6-month follow-up. Conclusion CCR2 expression was upregulated in both in vitro and in vivo SAH models. Additionally, inhibition of CCR2, at least partly through the PI3K/AKT pathway, alleviated neuroinflammation and neuronal apoptosis in vivo and in vitro. CCR2 levels in the CSF have a moderate diagnostic value for 6-month outcome prediction in patients with SAH.

The bismuth sulfide nanostructure has become a promising gas sensing material thanks to its exceptional intrinsic properties. However, pristine Bi2S3 as a room-temperature sensing material cannot achieve the highly sensitive detection of ppb-level NO2 gas. Herein, 1D nanorods with self-assembled hierarchical Bi2S3 nanostructures were obtained via a simple hydrothermal process. The as-prepared hierarchical Bi2S3 nanostructures exhibited outstanding NO2 sensing behaviors, such as a high response value (Rg/Ra = 5.8) and a short response/recovery time (τ90 = 28/116 s) upon exposure to 1 ppm NO2. The limit of detection of hierarchical Bi2S3 was down to 50 ppb. Meanwhile, the sensor exhibited excellent selectivity and humidity tolerance. The improved NO2 sensing properties were associated with the self-assembled hierarchical nanostructures, which provided a rich sensing active surface and accelerated the diffusion and adsorption/desorption processes between NO2 molecules and Bi2S3 materials. Additionally, the sensing response of hierarchical Bi2S3 nanostructures is much higher at 100% N2 atmosphere, which is different from the chemisorption oxygen model.

Background: As a crucial economic characteristic and a major indicator of reproductive performance in layers, egg production is controlled by a series of complex regulatory heredity basis. In particular, the interacting regulatory function between noncoding RNAs (ncRNAs) and coding RNA plays important roles in regulating laying performance. Methods: In this study, the RNA sequencing (RNA-seq) of ovarian tissues from Lohmann hens (n = 3) and Chengkou Mountain chicken (n = 3) under the laying peak period was performed to identify RNA transcriptional differences among different laying-performance populations. Results: Results showed that the expression level of 303 mRNAs, 68 long ncRNAs (lncRNAs), 533 circular RNAs (circRNAs), and 79 microRNAs (miRNAs) was significantly different among the groups. Functional enrichment analysis of these differentially expressed (DE) mRNAs revealed that the laying process was implicated in numerous significantly enriched pathways (p < 0.05), such as the neuroactive ligand-receptor interaction, steroid hormone biosynthesis, and calcium-signaling pathway. Furthermore, the lncRNA/circRNA–miRNA–mRNA regulatory networks related to the regulation of laying performance were constructed. Some randomly selective DE RNAs were verified by Real Time Quantitative (RT-qRCR), indicating that the bioinformatics analysis results of RNA-seq data were credible. Conclusions: This study could increase our understanding of the heredity basis of transcriptome in the laying performance of chicken.

Horns are an important breeding trait for sheep. However, no widely recognized viewpoint on the regulatory genes and mechanisms of horns is available, and the genetic basis of the four-horn phenotype (FHP) is unclear. This work conducted a genome-wide association study with 100 sheep genomes from multiple breeds to investigate the genetic basis of the FHP. The results revealed three significant associations (corrected as p < 1.64 × 10−8) of the InDels (CHR2: g.133,742,709delA, g.133,743,215insC, and g.133,743,940delT) for FHP in the intergenic sequence (IGS) between the MTX2 and the LOC105609047 of CHR2. Moreover, 14 significant associations (corrected as p < 1.42 × 10−9) of SNPs with the FHP phenotype were identified in CHR2 and CHR16, including five (e.g., CHR16: g.40,351,378G > A and g.40,352,577G > A) located in the intron of the ADAMTS12 gene, eight (e.g., CHR2: g.133,727,513C > T and g.133,732,145T > G) in the IGS between MTX2 and LOC105609047, and only one (CHR2: g.133,930,761A > G) in the IGS between HOXD1 and MTX2. Obvious divergence was also observed in genotype patterns between the FHP and others (two horns and hornless) in the HOXD1 and ADAMTS12 gene regions. An extremely significant linkage also occurred between Loci I and Loci II within 100 individuals (LD = −156.02186, p < 0.00001). In summary, our study indicated that the genomic sequences from CHR2 and CHR16 contributed to the FHP in sheep, specifically the key candidate genes HOXD1 and ADAMTS12. These results improved our understanding of the Mendelian genetic basis of the FHP in sheep.

Abstract Background Pitayas are currently attracting considerable interest as a tropical fruit with numerous health benefits. However, as a long-day plant, pitaya plants cannot flower in the winter season from November to April in Hainan, China. To harvest pitayas with high economic value in the winter season, it is necessary to provide supplementary lighting at night to induce flowering. To further explore the molecular regulating mechanisms of flower induction in pitaya plants exposed to supplementary lighting, we used de novo RNA sequencing-based transcriptomic analysis for four stages of pitaya plants subjected to light induction. Results We assembled 68,113 unigenes in total, comprising 29,782 unigenes with functional annotations in the NR database, 20,716 annotations in SwissProt, 18,088 annotations in KOG, and 11,059 annotations in KEGG. Comparisons between different samples revealed different numbers of significantly differentially expressed genes (DEGs). A number of DEGs involved in energy metabolism-related processes and plant hormone signaling were detected. Moreover, we identified many CONSTANS-LIKE, FLOWERING LOCUS T, and other DEGs involved in the direct regulation of flowering including CDF and TCP, which function as typical transcription factor genes in the flowering process. At the transcriptomic level, we verified 13 DEGs with different functions in the time-course response to light-induced flowering by quantitative reverse-transcription PCR analysis. Conclusions The identified DEGs may include some key genes controlling the pitaya floral-induction network, the flower induction and development is very complicated, and it involves photoperiod perception and different phytohormone signaling. These findings will increase our understanding to the molecular mechanism of floral regulation of long-day pitaya plants in short-day winter season induced by supplementary lighting.

Summary: The functional inactivation of tumor suppressor microRNA (miRNA) is closely related to the tumorigenesis of cancer. There are instances where the miRNA and the corresponding target both exist in a cell, but the target gene silencing do not occur as expected. Herein, we found that both miR-506 and its target CDK6 are highly co-expressed in lung cancer cells. Sequence analyses suggested that a miR-506 binding site (1648–1654) and a cis-element (1785–1795) for binding by heterogeneous nuclear ribonucleoprotein A2/B1 (HNRNPA2B1) are evolutionarily conserved and forms a stem structure in the 3′ untranslated region (3′UTR) of CDK6. Furthermore, HNRNPA2B1 can bind to the stem structure to denature it and recruit the RNA helicase DExH-box helicase 9 (DHX9) to the 3′UTR, which ultimately facilitates miRNAs-mediated CDK6 silencing. These results indicate that the cis-element of the 3′UTR of CDK6, where HNRNPA2B1 binds, serves as an RNA switch to regulate miRNAs’ function in cancer cells.

This study aimed to identify the genes related to the body size of pigs by conducting genome-wide selection analysis (GWSA). We performed a GWSA scan on 50 pigs belonging to four small-bodied pig populations (Diannan small-eared pig, Bama Xiang pig, Wuzhishan pig, and Jeju black pig from South Korea) and 124 large-bodied pigs. We used the genetic parameters of the pairwise fixation index (FST) and π ratio (case/control) to screen candidate genome regions and genes related to body size. The results revealed 47,339,509 high-quality SNPs obtained from 174 individuals, while 280 interacting candidate regions were obtained from the top 1% signal windows of both parameters, along with 187 genes (e.g., ADCK4, AMDHD2, ASPN, ASS1, and ATP6V0C). The results of the candidate gene (CG) annotation showed that a series of CGs (e.g., MSTN, LTBP4, PDPK1, PKMYT1, ASS1, and STAT6) was enriched into the gene ontology terms. Moreover, molecular pathways, such as the PI3K-Akt, HIF-1, and AMPK signaling pathways, were verified to be related to body development. Overall, we identified a series of key genes that may be closely related to the body size of pigs, further elucidating the heredity basis of body shape determination in pigs and providing a theoretical reference for molecular breeding.

Boer goats, as kemp in meat-type goats, are selected and bred from African indigenous goats under a long period of artificial selection. Their advantages in multiple economic traits, particularly their plump growth, have attracted worldwide attention. The current study displayed the genome-wide selection signature analyses of South African indigenous goat (AF), African Boer (BH), and Australian Boer (AS) to investigate the hereditary basis of artificial selection in different stages. Four methods (principal component analysis, nucleotide diversity, linkage disequilibrium decay, and neighbor-joining tree) implied the genomic diversity changes with different artificial selection intensities in Boer goats. In addition, the θπ, FST, and XP-CLR methods were used to search for the candidate signatures of positive selection in Boer goats. Consequently, 339 (BH vs. AF) and 295 (AS vs. BH) candidate genes were obtained from SNP data. Especially, 10 genes (e.g., BMPR1B, DNER, ITGAL, and KIT) under selection in both groups were identified. Functional annotation analysis revealed that these genes are potentially responsible for reproduction, metabolism, growth, and development. This study used genome-wide sequencing data to identify inheritance by artificial selection. The results of the current study are valuable for future molecular-assisted breeding and genetic improvement of goats.

BackgroundBased on 122 cases reported in China, data mining indicated that Sini Powder (SNP) and the Zuojin Pill (ZJP) are both widely used as the basic recipe for treating Gastroesophageal Reflux Disease (GERD).ObjectivesTo evaluate the intervention effects of Sini Zuojin Decoction (SNZJD) in patients with GERD.MethodsA comprehensive collection of randomized controlled trials (RCTs) using SNZJD in patients with GERD that were published in domestic and foreign journals was made by computer retrieval. RevMan 5.3 software was used for meta-analysis and bias risk assessment, Stata 14.0 software was used for sensitivity analysis, GRADE profiler 3.6 was used to evaluate the level of evidence, and trial sequential analysis (TSA), employed to control for random errors, was performed to assess the main outcomes. Network pharmacology analysis was applied to preliminarily study the mechanisms of action of SNZJD on GERD.ResultsThirteen articles were eventually included, covering a total of 966 patients. Meta-analysis indicated that: ① the SNZJD plus traditional stomach medicines (SPTSM) group was more effective than the traditional stomach medicines (TSM) group (RR = 1.16, 95% CI [1.04, 1.29], P = 0.009); ② the experimental group with SNZJD was significantly better than TSM controls in improving heartburn, substernal chest pain, acid regurgitation, and food regurgitation symptoms (P < 0.0001); ③ SPTSM could significantly decrease total symptom scores with substantial effectiveness (P < 0.00001). The recurrence rate and adverse effects of SNZJD treatment were significantly reduced (P < 0.05). TSA showed that the effective rate of meta-analysis might be reliable, but the recurrence and safety results were still uncertain. According to the evaluation by the GRADE method, the quality of evidence was low. Besides, SNZJD might treat GERD by acting on related targets and pathways such as inflammation, hormone regulation, and so on.ConclusionsSNZJD might be useful in the treatment of GERD, but its long-term effects and specific clinical mechanisms are unclear. Due to the poor quality of the evidence, more samples and high-quality clinical studies should be tested and verified in the future.

The follicle development (FD) is an important factor determining litter size in animals. Recent studies have found that noncoding RNAs (ncRNAs) play an important role in FD. In particular, the role of the regulatory mechanism of competing endogenous RNAs (ceRNAs) that drive FD has attracted increasing attention. Therefore, this study explored the genetic basis of goat FD by obtaining the complete follicular transcriptome of Dazu black goats at different developmental stages. Results revealed that 128 messenger RNAs (mRNAs), 4 long noncoding RNAs (lncRNAs), 49 microRNAs (miRNAs), and 290 circular RNAs (circRNAs) were significantly differentially expressed (DE) between large and small follicles. Moreover, DEmRNAs were enriched in many signaling pathways related to FD, as well as GO terms related to molecular binding and enzyme activity. Based on the analysis of the ceRNA network (CRN), 34 nodes (1 DElncRNAs, 10 DEcircRNAs, 14 DEmiRNAs, and 9 DEmRNAs) and 35 interactions (17 DEcircRNAs–DEmRNAs, 2 DElncRNAs–DEmiRNAs, and 16 DEmRNA–DEmiRNAs) implied that the CRN could be involved in the FD of goats. In conclusion, we described gene regulation by DERNAs and lncRNA/circRNA–miRNA–mRNA CRNs in the FD of goats. This study provided insights into the genetic basis of FD in precise transcriptional regulation.

On 23 June 2020, a major thrust faulting earthquake ruptured the megathrust near a sharp transition of the plate boundary beneath the Mexico subduction zone, overlapping the rupture area of the 1965 Mw 7.5 (Ms 7.6) event. A joint inversion of geodetic and seismic observations and prediction of coastal uplifts indicate a compact rupture with a peak slip of ∼3.6 m. The cumulative seismic moment is 1.41 × 1020 N · m within 20 s of coseismic rupture, giving a moment magnitude of 7.4. The rupture propagated with a relatively high rupture speed (∼3.5 km/s) downdip at depths from 20 to 30 km, terminating at the edge of a slow slip region. The energy-related static stress drop is ∼8.1 MPa. The aftershock distribution and the region where slow slip events occurred show a clear complementary relationship with the coseismic rupture zone of the 2020 Oaxaca event. The high-stress drop and associated deep compact rupture pattern suggest a homogeneous asperity with high strength beneath the Oaxaca region. Comparisons of rupture areas, mechanisms, and seismic waveforms show that the 2020 Mw 7.4 Oaxaca earthquake is a quasi-repeat of the 1965 earthquake. The inferred recurrence interval is ∼55 yr. Static stress transfer from mainshock may trigger near trench aftershocks but has small loading on the megathrust interface in the Tehuantepec seismic gap. [ABSTRACT FROM AUTHOR]

The largest earthquake since 1954 to strike the state of Nevada, United States, ruptured on 15 May 2020 along the Monte Cristo range of west-central Nevada. The Mw 6.5 event involved predominantly left-lateral strike-slip faulting with minor normal components on three aligned east-west-trending faults that vary in strike by 23°. The kinematic rupture process is determined by joint inversion of Global Navigation Satellite Systems displacements, Interferometric Synthetic Aperture Radar (InSAR) data, regional strong motions, and teleseismic P and SH waves, with the three-fault geometry being constrained by InSAR surface deformation observations, surface ruptures, and relocated aftershock distributions. The average rupture velocity is 1.5 km/s, with a peak slip of ~1.6 m and a ~20s rupture duration. The seismic moment is 6.9×1018 N·m. Complex surface deformation is observed near the fault junction, with a deep near-vertical fault and a southeast-dipping fault at shallow depth on the western segment, along which normal-faulting aftershocks are observed. There is a shallow slip deficit in the Nevada ruptures, probably due to the immature fault system. The causative faults had not been previously identified and are located near the transition from the Walker Lane belt to the Basin and Range province. The aim of this study is to evaluate the effects of different mulching treatments, and to serve as a theoretical foundation for mulching practices in pitaya orchards of Hainan, China. In this study, the physicochemical properties, enzyme activity and also the structure and abundance of the soil microbial community were examined after a year's treatment. The results show that black fabric mulch and nature grass mulch significantly improved the soil water content, whereas its pH value was significantly reduced by purslane (Portulaca oleracea L.) mulch and black fabric mulch as compared to the control. Nature grass mulch increased soil organic carbon to a greater extent than control, coconut chaff mulch and also nature grass mulch significantly increased the content of alkaline hydrolysis nitrogen, nature grass mulch and black fabric mulch increased the content of available potassium more significantly than purslane (Portulaca oleracea L.) mulch and control. Treatments of nature grass mulch and purslane (Portulaca oleracea L.) mulch increased the activities of soil enzymes in the soil. The abundance of bacteria was highest in nature grass mulch and purslane (Portulaca oleracea L.) mulch, and the four mulch treatments helped to increase the richness of soil fungi more than control. The results show that all these types of mulch improved the characteristics of the soil compared with control, and nature grass mulch provided the best edaphic environment for pitaya orchards. Italy has a historical earthquake record that is complete for events with a magnitude above 5.8 since A.D. 1349, making it possible to study Coulomb failure stress changes (ΔCFS) over a long period. In this study, we investigated the interactions between moderate-to-large earthquakes through ΔCFS over 100 yr in central Italy. This region is characterized by intense seismicity with predominantly extensional components. Hence, earthquake hazard assessment is of great public concern. Besides, earthquake interactions on normal faults are relatively less studied compared to reverse and strike-slip faults. ΔCFS calculations in this study incorporated both coseismic stress transfer and postseismic viscoelastic relaxation, and found the epicenters of 13 out of 15 events located in positively stressed lobes induced by previous earthquakes, confirming a correlation between the ΔCFS pattern and locations of moderate-to-large earthquakes. Next, we estimated the current distribution of ΔCFS on active faults, and after a comprehensive analysis of ΔCFS accumulation, slip rates, historical seismicity, and locations of populated cities, we identified three regions of potential seismic hazards in this region. At GMT time 13:19, August 8, 2017, an Ms7.0 earthquake struck the Jiuzhaigou region in Sichuan Province, China, causing severe damages and casualties. To investigate the source properties, seismogenic structures, and seismic hazards, we systematically analyzed the tectonic environment, crustal velocity structure in the source region, source parameters and rupture process, Coulomb failure stress changes, and 3-D features of the rupture plane of the Jiuzhaigou earthquake. Our results indicate the following: (1) The Jiuzhaigou earthquake occurred on an unmarked fault belonging to the transition zone of the east Kunlun fault system and is located northwest of the Huya fault. (2) Both the mainshock and aftershock rupture zones are located in a region where crustal seismic velocity changes dramatically. Southeast to the source region, shear wave velocity at the middle to lower crust is significantly low, but it rapidly increases northeastward and lies close to the background velocity across the rupture fault. (3) The aftershock zone is narrow and distributes along the northwest-southeast trend, and most aftershocks occur within a depth range of 5-20 km. (4) The focal mechanism of the Jiuzhaigou earthquake indicates a left-lateral strike-slip fault, with strike, dip, and rake angles of 152°, 74° and 8°, respectively. The hypocenter depth measures 20 km, whereas the centroid depth is about 6 km. The co-seismic rupture mainly concentrates at depths of 3-13 km, with a moment magnitude (Mw) of 6.5. (5) The co-seismic rupture also strengthens the Coulomb failure stress at the two ends of the rupture fault and the east segment of the Tazang fault. Aftershocks relocation results together with geological surveys indicate that the causative fault is a near vertical fault with notable spatial variations: dip angle varies within 66°-89° from northwest to southeast and the average dip angle measures ~84°. The results of this work are of fundamental importance for further studies on the source characteristics, tectonic environment, and seismic hazard evaluation of the Jiuzhaigou earthquake. On 2012 September 5, the anticipated interplate thrust earthquake ruptured beneath the Nicoya peninsula in northwestern Costa Rica close to the Middle America trench, with a magnitude Mw 7.6. Extensive co-seismic observations were provided by dense near-field strong ground motion, Global Positioning Systems (GPS) networks and teleseismic recordings from global seismic networks. The wealthy data sets available for the 2012 Mw 7.6 Nicoya earthquake provide a unique opportunity to investigate the details of the rupture process of this earthquake. By implementing a non-linear joint inversion with high-rate GPS waveform, more static GPS offsets, strong-motion data and teleseismic body waveform, we obtained a robust and accurate rupture model of the 2012 Mw 7.6 Nicoya earthquake. The earthquake is dominantly a pure thrust component with a maximum slip of 3.5 m, and the main large slip patch is located below the hypocentre, spanning ~50 km along dip and ~110 km along strike. The static stress drop is about 3.4MPa. The total seismic moment of our preferred model is 3.46 × 1020 Nm, which gives Mw = 7.6. Due to the fast rupture velocity, most of the seismic moment was released within 70 s. The largest slip patch directly overlaps the interseismic locked region identified by geodetic observations and extends downdip to the intersection with the upper plate Moho. We also find that there is a complementary pattern between the distribution of aftershocks and the co-seismic rupture; most aftershocks locate in the crust of the upper plate and are possibly induced by the stress change caused by the large slip patch. On 2014 April 1, a magnitude Mw 8.1 interplate thrust earthquake ruptured a densely instrumented region of Iquique seismic gap in northern Chile. The abundant data sets near and around the rupture zone provide a unique opportunity to study the detailed source process of this megathrust earthquake. We retrieved the spatial and temporal distributions of slip during the main shock and one strong aftershock through a joint inversion of teleseismic records, GPS offsets and strong motion data. The main shock rupture initiated at a focal depth of about 25 km and propagated around the hypocentre. The peak slip amplitude in the model is ~6.5 m, located in the southeast of the hypocentre. The major slip patch is located around the hypocentre, spanning ~150 km along dip and ~160 km along strike. The associated static stress drop is ~3 MPa. Most of the seismic moment was released within 150 s. The total seismic moment of our preferred model is 1.72 × 1021 Nm, equivalent to Mw 8.1. For the strong aftershock on 2014 April 3, the slip mainly occurred in a relatively compact area, and the major slip area surrounded the hypocentre with the peak amplitude of ~2.5m. There is a secondary slip patch located downdip from the hypocentre with the peak slip of ~2.1m. The total seismic moment is about 3.9 × 1020 Nm, equivalent to Mw 7.7. Between the rupture areas of the main shock and the 2007 November 14 Mw 7.7 Antofagasta, Chile earthquake, there is an earthquake vacant zone with a total length of about 150 km. Historically, if there is no big earthquake or obvious aseismic creep occurring in this area, it has a great potential of generating strong earthquakes with magnitude larger than Mw 7.0 in the future. Background: The prognosis for pancreatic cancer (PC) is very poor. The SnoN gene may have a role in cell proliferation and apoptosis in human cancer. However, the influence of SnoN on cell proliferation and apoptosis in human PC cells remains unknown. Methods: SnoN expression was assessed in SW1990 PC cell lines using real-time polymerase chain reaction (PCR). A luciferase reporter assay was used to confirm the target associations. The effect of SnoN on cell proliferation in vitro was confirmed using Cell Counting Kit-8. Apoptosis was confirmed using flow cytometry. Gene and protein expression were examined using real time PCR and Western blotting, respectively. Results: SnoN siRNA significantly inhibited the growth of SW1990 cells by decreasing cell proliferation (P < 0.05) and increasing cell apoptosis (P < 0.05), compared with the blank group and the negative control group. The highest inhibition of cell proliferation appeared at 3 days post-transfection. Cell apoptosis more obvious at 48 h after transfection. Conclusions: In summary, our results reveal that the RNAi-mediated downregulation of SnoN effectively inhibited the proliferation of PC cells. SnoN-siRNA also enhanced SW1990 PC cell apoptosis. These findings indicate that SnoN gene plays an important role in pancreatic cancer development, and might serve as a potential therapeutic target for pancreatic cancer. Background: It has been shown that heat shock-related 70-kDa protein 2 (HSPA2), a member of the HSP70 family of heat shock proteins, is important for cancer cell growth and metastasis. However, the status of HSPA2 expression and its prognostic significance in pancreatic cancer remain unknown. Methods: Quantitative reverse transcriptase ploymerase chain reaction (qRT-PCR) was applied to examine HSPA2 messenger RNA (mRNA) expression in 104 pairs of pancreatic cancer tissues and adjacent noncancerous tissues. Statistical analyses were applied to evaluate the diagnostic value and associations of HSPA2 expression with clinicopathological characteristics. Results: HSPA2 mRNA was significantly overexpressed in pancreatic cancer tissues (3.9 ± 0.8) than in adjacent normal tissues (1.1 ± 0.4) (P < 0.001). Clinicopathological analysis showed that HSPA2 expression was significantly correlated with tumor size (P = 0.024), histological differentiation (P = 0.012), TNM stage (P = 0.006), lymph node metastasis (P = 0.043) and serum CA19-9 level (P = 0.046). Moreover, patients with higher HSPA2 expression levels had shorter overall survival time than those with lower HSPA2 expression levels (P = 0.019). Furthermore, Cox regression analyses showed that HSPA2 expression was an independent predictor of overall survival (P = 0.011). Conclusions: Our results suggest that overexpression of HSPA2 in pancreatic cancer is associated with aggressive progression and poor prognosis and that HSPA2 may be served as a prognostic marker. Ephrin (EPH) receptors can be classified into EPHA and EPHB receptors and are important in diverse cellular processes. EPHA4, a member of the EPHA receptors, has been demonstrated to be elevated in various human cancers and involved in the tumor progression. However, the role of EPHA4 in pancreatic cancer cells remains unclear. Therefore, the present study transfected Panc-1 and BxPC-3 cells with small interfering RNA (siRNA) to knockdown the expression of EPHA4. Wound healing and invasion assays were then performed to assess the effect of EPHA4 knockdown on the motility and invasion of pancreatic cancer cells. The results demonstrated that the knockdown of EPHA4 by siRNA inhibits the motility and invasion of pancreatic cancer cells. Furthermore, gelatin zymography assay showed that EPHA4 may regulate the activity of matrix metalloproteinase (MMP)-2. In addition, the knockdown of EPHA4 increased the expression of epithelial (E)-cadherin, as well as decreased the expression of Snail. Overall, these results suggested that EPHA4 may promote the motility and invasion of pancreatic cancer cells via the upregulation of MMP-2 and Snail, as well as the downregulation of E-cadherin.
