Abstract Pseudomonas aeruginosa (P. aeruginosa) is a major pathogen that causes nosocomial infections and often exhibits antibiotic resistance. Therefore, the development of an accurate method for detecting P. aeruginosa is required to control P. aeruginosa-related outbreaks. In this study, we established an enzyme-linked immunosorbent assay method for the sensitive detection of three P. aeruginosa strains, UCBPP PA14, ATCC 27853, and multidrug-resistant ATCC BAA-2108. We produced a recombinant antibody (rAb) against P. aeruginosa V‐antigen (PcrV), which is a needle tip protein of the type III secretion system of P. aeruginosa using mammalian cells with high yield and purity, and confirmed its P. aeruginosa binding efficiency. The rAb was paired with commercial anti-P. aeruginosa Ab for a sandwich ELISA, resulting in an antigen-concentration-dependent response with a limit of detection value of 230 CFU/mL. These results suggest that the rAb produced herein can be used for the sensitive detection of P. aeruginosa with a wide range of applications in clinical diagnosis and point-of-care testing.

Abstract Artificial olfactory sensors that recognize patterns transmitted by olfactory receptors are emerging as a technology for monitoring volatile organic compounds. Advances in statistical processing methods and data processing technology have made it possible to classify patterns in sensor arrays. Moreover, biomimetic olfactory recognition sensors in the form of pattern recognition have been developed. Deep learning and artificial intelligence technologies have enabled the classification of pattern data from more sensor arrays, and improved artificial olfactory sensor technology is being developed with the introduction of artificial neural networks. An example of an artificial olfactory sensor is the electronic nose. It is an array of various types of sensors, such as metal oxides, electrochemical sensors, surface acoustic waves, quartz crystal microbalances, organic dyes, colorimetric sensors, conductive polymers, and mass spectrometers. It can be tailored depending on the operating environment and the performance requirements of the artificial olfactory sensor. This review compiles artificial olfactory sensor technology based on olfactory mechanisms. We introduce the mechanisms of artificial olfactory sensors and examples used in food quality and stability assessment, environmental monitoring, and diagnostics. Although current artificial olfactory sensor technology has several limitations and there is limited commercialization owing to reliability and standardization issues, there is considerable potential for developing this technology. Artificial olfactory sensors are expected to be widely used in advanced pattern recognition and learning technologies, along with advanced sensor technology in the future.

Hydrogel capsules synthesized by conventional water-in-oil emulsion systems are the less preferred choice for biomedical applications due to the use of oils and surfactants. An aqueous two-phase system (ATPS), which allows the formation of water-in-water emulsion, is considered a green alternative and therefore has been explored a lot for its application in the biomedical field. Herein, we present the synthesis of hydrogel capsules using a set-up consisting of a pneumatic valve integrated with the ATPS microfluidic system. In this arrangement, at first, a pneumatic valve facilitates the generation of the droplets of one aqueous system i.e. sodium alginate (SA) containing dextran solution into another aqueous phase comprising polyethylene glycol solution. The present approach allows good control over droplet generation by tuning the pressure of the pneumatic valve and the flow rates of the core and middle phases. The synthesis of hybrid capsules within the microfluidic device is carried out mainly by using the interfacial complexation of oppositely charged polyelectrolytes, chitosan with SA via electrostatic interactions. The interfacial complexed SA and chitosan hydrogel capsules were collected via the settling collection method, which ensures the retaining of the shape of the hybrid capsules. The morphological properties of as-synthesized droplets and hybrid capsules were examined via optical microscopy. The hydrogel capsules show good encapsulation capability for the magnetic particles. Even though this study mainly focuses on the synthesis part, we anticipate that the proposed approach will enable the encapsulation of cells within the hybrid capsules as well as enhance the cell adhesion on the surface of the hydrogel capsules hence, these hydrogel capsules can find the potent application in the biomedical engineering.

Nitroreductase (NTR) has the ability to activate nitro group-containing prodrugs and decompose explosives; thus, the evaluation of NTR activity is specifically important in pharmaceutical and environmental areas. Numerous studies have verified effective fluorescent methods to detect and image NTR activity; however, near-infrared (NIR) fluorescence probes for biological applications are lacking. Thus, in this study, we synthesized novel NIR probes (NIR-HCy-NO2 1–3) by introducing a nitro group to the hemicyanine skeleton to obtain fluorescence images of NTR activity. Additionally, this study was also designed to propose a different water solubility and investigate the catalytic efficiency of NTR. NIR-HCy-NO2 inherently exhibited a low fluorescence background due to the interference of intramolecular charge transfer (ICT) by the nitro group. The conversion from the nitro to amine group by NTR induced a change in the absorbance spectra and lead to the intense enhancement of the fluorescence spectra. When assessing the catalytic efficiency and the limit of detection (LOD), including NTR activity imaging, it was demonstrated that NIR-HCy-NO2 1 was superior to the other two probes. Moreover, we found that NIR-HCy-NO2 1 reacted with type I mitochondrial NTR in live cell imaging. Conclusively, NIR-HCy-NO2 demonstrated a great potential for application in various NTR-related fields, including NTR activity for cell imaging in vivo.

There is a growing interest in performing time-resolved cryo-EM studies. Here, the authors present a time-resolved sample preparation method for cryo-EM called trEM, which uses a microfluidic device to initiate the biochemical reaction by rapid mixing of the components and then spraying the sample onto a cryo-EM grid to snap-freeze it in a blot-free, automated manner within several milliseconds.

Poly(dimethylsiloxane) (PDMS) is widely used as a microfluidics platform material; however, it absorbs various molecules, perturbing specific chemical concentrations in microfluidic channels. We present a simple solution to prevent adsorption into a PDMS microfluidic device. We used a vapor-phase-deposited nanoadhesive layer to seal PDMS microfluidic channels. Absorption of fluorescent molecules into PDMS was efficiently prevented in the nanolayer-treated PDMS device. Importantly, when cultured in a nanolayer-treated PDMS device, yeast cells exhibited the expected concentration-dependent response to a mating pheromone, including mating-specific morphological and gene expression changes, while yeast cultured in an untreated PDMS device did not properly respond to the pheromone. Our method greatly expands microfluidic applications that require precise control of molecule concentrations.

Photonic fibers are important raw materials for structurally colored fabrics. In particular, the mechanochromic fibers potentially provide color‐tunable clothes, like chameleon skins. Herein, microfluidic jetting to continuously produce mechanochromic fibers in a controlled manner is used. The jet is produced by flow‐focusing the photocurable dispersions in a surfactant‐laden carrier fluid using microfluidic devices. The dispersions contain monodispersed silica particles in an elastomer‐forming resin. As the silica particles have repulsive interparticle potential in the resin, they spontaneously organize into a nonclose‐packed regular array, developing structural colors. The jet of the dispersions is photocured in situ by irradiation of ultraviolet at the exit of the microfluidic channel, continuously producing the photonic fibers. As the interparticle separation among silica particles is shortened along the radial direction by stretching the elastic fibers, the structural colors are dynamically blue shifted. Also, the original colors are reversibly recovered by relaxing the fibers as the deformation is fully elastic. The nonclose‐packed array of inelastic silica particles provides a wide range of color tuning and high reversibility. Moreover, microfluidic jetting enables the production of Janus fibers composed of two distinct color domains, which enriches the available structural colors through color mixing.

Loop-mediated isothermal amplification (LAMP) is a molecular diagnosis technology with the advantages of rapid results, isothermal reaction conditions, and high sensitivity. However, this diagnostic system often produces false positive results due to a high rate of non-specific reactions caused by formation of hairpin structures, self-dimers, and mismatched hybridization. The non-specific signals can be due to primers used in the methods because the utilization of multiple LAMP primers increases the possibility of self-annealing of primers or mismatches between primers and templates. In this study, we report a nanomaterial-assisted LAMP method that uses a graphene oxide–gold nanoparticles (AuNPs@GO) nanocomposite to enable the detection of foot-and-mouth disease virus (FMDV) with high sensitivity and specificity. Foot-and-mouth disease (FMD) is a highly contagious and deadly disease in cloven-hoofed animals; hence, a rapid, sensitive, and specific detection method is necessary. The proposed approach exhibited high sensitivity and successful reduction of non-specific signals compared to the traditionally established LAMP assays. Additionally, a mechanism study revealed that these results arose from the adsorption of single-stranded DNA on AuNPs@GO nanocomposite. Thus, AuNPs@GO nanocomposite is demonstrated to be a promising additive in the LAMP system to achieve highly sensitive and specific detection of diverse diseases, including FMD.

Foot-and-mouth disease virus (FMDV) is a highly contagious disease that affects cloven-hoofed animals. The traditional diagnostic methods for FMDV have several drawbacks such as cross-reactivity, low sensitivity, and low selectivity. To overcome these drawbacks, we present an optical and electrochemical dual-modal approach for the specific detection of FMDV serotypes O and A by utilizing a magnetic nanoparticle labeling technique with resorufin β-d-glucopyranoside (res-β-glc) and β-glucosidase (β-glc), without the use of typical lateral flow assay or polymerase chain reaction. FMDV serotypes O and A were reacted with pan-FMDV antibodies that recognize all seven FMDV serotypes (O, A, C, Asia 1, SAT 1, SAT 2, and SAT 3). The antigen–antibody complex was then immobilized on magnetic nanoparticles and reacted with β-glc-conjugated FMDV type O or type A antibodies. Subsequently, the addition of res-β-glc resulted in the release of fluorescent resorufin and glucose owing to catalytic hydrolysis by β-glc. The detection limit of fluorescent signals using a fluorescence spectrophotometer was estimated to be log(6.7) and log(5.9) copies/mL for FMDV type O and A, respectively, while that of electrochemical signals using a glucometer was estimated to be log(6.9) and log(6.1) copies/mL for FMDV type O and A, respectively. Compared with a commercially available lateral flow assay diagnostic kit for immunochromatographic detection of FMDV type O and A, this dual-modal detection platform offers approximately four-fold greater sensitivity. This highly sensitive and accurate dual-modal detection method can be used for effective disease diagnosis and treatment, and will find application in the early-stage diagnosis of viral diseases and next-generation diagnostic platforms.

Bone homeostasis plays a major role in supporting and protecting various organs as well as a body structure by maintaining the balance of activities of the osteoblasts and osteoclasts. Unbalanced differentiation and functions of these cells result in various skeletal diseases, such as osteoporosis, osteopetrosis, and Paget’s disease. Although various synthetic nanomaterials have been developed for bone imaging and therapy through the chemical conjugation, they are associated with serious drawbacks, including heterogeneity and random orientation, in turn resulting in low efficiency. Here, we report the synthesis of bone-targeting ferritin nanoparticles for bone imaging. Ferritin, which is a globular protein composed of 24 subunits, was employed as a carrier molecule. Bone-targeting peptides that have been reported to specifically bind to osteoblast and hydroxyapatite were genetically fused to the N-terminus of the heavy subunit of human ferritin in such a way that the peptides faced outwards. Ferritin nanoparticles with fused bone-targeting peptides were also conjugated with fluorescent dyes to assess their binding ability using osteoblast imaging and a hydroxyapatite binding assay; the results showed their specific binding with osteoblasts and hydroxyapatite. Using in vivo analysis, a specific fluorescent signal from the lower limb was observed, demonstrating a highly selective affinity of the modified nanoparticles for the bone tissue. These promising results indicate a specific binding ability of the nanoscale targeting system to the bone tissue, which might potentially be used for bone disease therapy in future clinical applications.

The calcium-binding protein S100A9 regulates inflammatory processes and the immune response. It is overexpressed in a variety of inflammatory and oncologic conditions. In this study, we produced a recombinant human S100A9 (hS100A9) antigen with high yield and purity and used it to generate a hybridoma cell culture-based monoclonal anti-hS100A9 antibody. We selected five anti-hS100A9 antibodies from cell supernatants that showed high antigen binding efficiency and identified the nucleotide sequences of three antibodies: two with high effective concentration values and one with the lowest value. The antigen and antibody development procedures described herein are useful for producing large amounts of monoclonal antibodies against hS100A9 and other antigens of interest. The nucleotide sequences of the anti-hS100A9 monoclonal antibody revealed herein will be helpful in the generation of recombinant antibodies or antibody fragments against hS100A9.

The polymerase chain reaction (PCR) has become a powerful molecular diagnostic technique over the past few decades, but remains somewhat impaired due to low specificity, poor sensitivity, and false positive results. Metal and carbon nanomaterials, quantum dots, and metal oxides, can improve the quality and productivity of PCR assays. Here, we describe the ability of PCR assisted with nanomaterials (nano-PCR) comprising a nanocomposite of graphene oxide (GO) and gold nanoparticles (AuNPs) for sensitive detection of the foot-and-mouth disease virus (FMDV). Graphene oxide and AuNPs have been widely applied as biomedical materials for diagnosis, therapy, and drug delivery due to their unique chemical and physical properties. Foot-and-mouth disease (FMD) is highly contagious and fatal for cloven-hoofed animals including pigs, and it can thus seriously damage the swine industry. Therefore, a highly sensitive, specific, and practical method is needed to detect FMDV. The detection limit of real-time PCR improved by ~1000 fold when assisted by GO-AuNPs. We also designed a system of detecting serotypes in a single assay based on melting temperatures. Our sensitive and specific nano-PCR system can be applied to diagnose early FMDV infection, and thus may prove to be useful for clinical and biomedical applications.

Next-generation sequencing (NGS) of the Flammulina rossica (wood-rotting basidiomycete) genome was performed to identify its carbohydrate-active enzymes (CAZymes). De novo genome assembly (31 kmer) revealed a total length of 35,646,506 bp (49.79% GC content). In total, 12,588 gene models of F. rossica were predicted using an ab initio gene prediction tool (AUGUSTUS). Orthologous analysis with other fungal species revealed that 7433 groups contained at least one F. rossica gene. Additionally, 12,033 (95.6%) of 12,588 genes for F. rossica proteins had orthologs among the Dikarya, and F. rossica contained 12 species-specific genes. CAZyme annotation in the F. rossica genome revealed 511 genes predicted to encode CAZymes including 102 auxiliary activities, 236 glycoside hydrolases, 94 glycosyltransferases, 19 polysaccharide lyases, 56 carbohydrate esterases, and 21 carbohydrate binding-modules. Among the 511 genes, several genes were predicted to simultaneously encode two different CAZymes such as glycoside hydrolases (GH) as well as carbohydrate-binding module (CBM). The genome information of F. rossica offers opportunities to understand the wood-degrading machinery of this fungus and will be useful for biotechnological and industrial applications.

Paper-based analytical devices are fluidic chips fabricated with extremely inexpensive materials, namely paper, thereby allowing their use as a zero-cost analytical device in third-world countries that lack access to expensive diagnostic infrastructures. The aim of this review is to discuss: (1) microfluidic paper-based analytical devices (µPADs) for quantitative analysis, (2) fabrication of two- or three-dimensional µPADs, (3) analytical methods of µPADs, and (4) our opinions regarding the future applications of µPADs for quantitative urinalysis.

Bioactive microcapsules containing Bacillus thuringiensis (BT) spores were generated by a combination of a hydro gel, microfluidic device and chemical polymerization method. As a proof-of-principle, we used BT spores displaying enhanced green fluorescent protein (EGFP) on the spore surface to spatially direct the EGFP-presenting spores within microcapsules. BT spore-encapsulated microdroplets of uniform size and shape are prepared through a flow-focusing method in a microfluidic device and converted into microcapsules through hydrogel polymerization. The size of microdroplets can be controlled by changing both the dispersion and continuous flow rate. Poly(N-isoproplyacrylamide) (PNIPAM), known as a hydrogel material, was employed as a biocompatible material for the encapsulation of BT spores and long-term storage and outstanding stability. Due to these unique properties of PNIPAM, the nutrients from Luria-Bertani complex medium diffused into the microcapsules and the microencapsulated spores germinated into vegetative cells under adequate environmental conditions. These results suggest that there is no limitation of transferring low-molecular-weight-substrates through the PNIPAM structures, and the viability of microencapsulated spores was confirmed by the culture of vegetative cells after the germinations. This microfluidic-based microencapsulation methodology provides a unique way of synthesizing bioactive microcapsules in a one-step process. This microfluidic-based strategy would be potentially suitable to produce microcapsules of various microbial spores for on-site biosensor analysis.

We have developed a simple electrochemical biosensing strategy for the label-free diagnosis of hepatitis B virus (HBV) on a gold electrode surface. Gold-binding polypeptide (GBP) fused with single-chain antibody (ScFv) against HBV surface antigen (HBsAg), in forms of genetically engineered protein, was utilized. This GBP-ScFv fusion protein can directly bind onto the gold substrate with the strong binding affinity between the GBP and the gold surface, while the recognition site orients toward the sample for target binding at the same time. Furthermore, this one-step immobilization strategy greatly simplifies a fabrication process without any chemical modification as well as maintaining activity of biological recognition elements. This system allows specific immobilization of proteins and sensitive detection of targets, which were verified by surface plasmon resonance analysis and successfully applied to electrochemical cyclic voltammetry and impedance spectroscopy upto 0.14 ng/mL HBsAg.

In this paper, we estimate the cost of the GHG abatement target by Korean government using a trade-based model, GTAP-E, after updating all the database of industrial and trade structures as well as carbon dioxide emissions of most of regions of the world. Major findings of this paper are as follows. First, estimates of the costs, GDP and welfare costs as well as abatement cost, of the GHG abatement target differ substantially by two things: (1) assumption on carbon reductions in other countries (the reduction only in Korea or commitments with Annex I countries), (2) assumption on international emission trading. Second, governmental policy to set sectoral abatement target would increase the costs than otherwise in spite of the same level of the carbon reduction. But in the case of mild adjustments of sectoral targets by the government, the policy with more reductions in industrial sectors than private consumptions are absolutely better than the other (reducing more in private consumptions).

In this study, analysis of sensing signal profiles was conducted focusing on the close relationship between electrical conductivity and signal intensity in surface treated poly(dimethylsiloxane)/carbon nanotube (PDMS/CNT) composite patches for the purpose of their practical application as flexible chemical sensors. The flexible PDMS/CNT composite patches were prepared from a PDMS/CNT mixture with a two-roll apparatus. It was found that the PDMS/CNT pads showed a high electrical conductivity (10−1 S/m) even at low CNT loading (0.6 wt %) and a contact angle range of 105–118°. The surface of the obtained PDMS/CNT composite patches was treated using a simple bio-conjugation method to incorporate beta-cyclodextrin (beta-CD) molecules onto the surface as a sensing medium, in order to detect a model compound (Methyl Paraben, MePRB). FT-IR spectra indicated that beta-cyclodextrin molecules were effectively introduced on the surface of the PDMS/CNT patches. It was shown that the sensor signal intensity was substantially dependent on the base current value, which increased with increasing CNT loading. Accordingly, the base current value was intimately associated with the electrical conductivity of the composite patches. On the other hand, the increase in current over the base current (ΔI/I0) obtained after the addition of the model compound was inversely proportional to the CNT content. In this way, analysis on the sensing signal profiles of the flexible chemical sensor system was conducted to determine a process window. This study is a very useful springboard for future research activities, as more profound studies are necessary to fully understand sensing signal profiles.

Recently, there has been increased interest in self-healing membranes containing functional microcapsules in relation to challenges involving water treatment membranes. In this study, a self-healing membrane has been prepared by incorporating microcapsules with a polyurethane (PU) shell and a diisocyanate core in a poly(ether sulfone) (PES) membrane. Depending on the characteristics of the microcapsule, to precisely quantify the self-healing behavior and performance of the produced microcapsule embedded membranes, it is important to understand the effect of a used surfactant on microcapsule synthesis. It is noteworthy that mixed surfactants have been employed to control and tailor the size and morphology of microcapsules during the synthetic process, and the surfactant system employed was one of the most dominant parameters for affecting the healing capability of microcapsule embedded membranes. Various techniques including microscopy (optical and electron), thermal analyses (DSC and TGA), and water flux measurements have been employed. This article provides essential and important information for future research into the subtle relation between microcapsule properties with varied synthetic parameters and the self-healing behavior of membrane.

Polydimethyl-siloxane (PDMS) is often applied to fabricate cell chips. In this study, we fabricated an adipocyte microcell pattern chips using PDMS to analyze the inhibition activity of lipid droplets in mouse embryo fibroblast cells (3T3-L1) with anti-obesity agents. To form the PDMS based micropattern, we applied the micro-contact printing technique using PDMS micro-stamps that had been fabricated by conventional soft lithography. This PDMS micro-pattern enabled the selective growth of 3T3-L1 cells onto the specific region by preventing cell adhesion on the PDMS region. It then allowed growth of the 3T3-L1 cells in the chip for 10 days and confirmed that lipid droplets were formed in the 3T3-L1 cells. After treatment of orlistat and quercetin were treated in an adipocyte micro-cell pattern chip with 3T3-L1 cells for six days, we found that orlistat and quercetin exhibited fat inhibition capacities of 19.3% and 24.4% from 0.2 μM of lipid droplets in 3T3-L1 cells. In addition, we conducted a direct quantitative analysis of 3T3-L1 cell differentiation using Oil Red O staining. In conclusion, PDMS-based adipocyte micro-cell pattern chips may contribute to the development of novel bioactive compounds.

We report on the development of an enzyme-amplified sandwich-type immunosensor based on a thin gold film sputtered on an anodic nanoporous niobium oxide (Au@Nb2O5) electrode. The electrocatalytic activity of enzymatically amplified electroactive species and a stable electrode consisting of Au@Nb2O5 were used to obtain a powerful signal amplification of the electrochemical immunobiosensor. The method using this electrochemical biosensor based on an Au@Nb2O5 electrode provides a much better performance than those based on conventional bulk gold or niobium oxide electrodes. Our novel approach does not require any time-consuming cleaning steps to yield reproducible electrochemical signals. In addition, the strong adhesion of gold films on the niobium oxide electrodes offers a very stable substrate during electrochemical biosensing. Cyclic voltammetry measurements indicate that non-specific binding of proteins to the modified Au@Nb2O5 surface is sufficiently low to be ignored in the case of our novel system. Finally, we demonstrated the ability of the biosensor based on an Au@Nb2O5 offering the enhanced performance with a high resolution and sensitivity. Therefore, it is expected that the biosensor based on an Au@Nb2O5 has great potential for highly efficient biological devices.

In recent years there has been great progress in applying FET-type biosensors for highly sensitive biological detection. Among them, the ISFET (ion-sensitive field-effect transistor) is one of the most intriguing approaches in electrical biosensing technology. Here, we review some of the main advances in this field over the past few years, explore its application prospects, and discuss the main issues, approaches, and challenges, with the aim of stimulating a broader interest in developing ISFET-based biosensors and extending their applications for reliable and sensitive analysis of various biomolecules such as DNA, proteins, enzymes, and cells.

The cardioprotective effects of KR-31761, a newly synthesized K+ATP opener, were evaluated in rat models of ischemia/reperfusion (I/R) heart injury. In isolated rat hearts subjected to 30-min global ischemia/30-min reperfusion, KR-31761 perfused prior to ischemia significantly increased both the left ventricular developed pressure (% of predrug LVDP: 17.8, 45.1, 54.2, and 62.6 for the control, 1 μM, 3 μM, and 10 μM, respectively) and double product (DP: heart rate × LVDP; % of predrug DP: 17.5, 44.9, 56.2, and 64.5 for the control, 1 μM, 3 μM, and 10 μM, respectively) at 30-min reperfusion while decreasing the left ventricular end-diastolic pressure (LVEDP). KR-31761 (10 μM) significantly increased the time to contracture during the ischemic period, whereas it concentration-dependently decreased the lactate dehydrogenase release during reperfusion. All these parameters were significantly reversed by 5-hydroxydecanoate (5-HD, 100 μM) and glyburide (1 μM), selective and nonselective blockers of the mitochondrial K+ATP (mitoK+ATP) channel and K+ATP channel, respectively. In anesthetized rats subjected to 30-min occlusion of left anterior descending coronary artery/2.5-h reperfusion, KR-31761 administered 15 min before the onset of ischemia significantly decreased the infarct size (72.2%, 55.1%, and 47.1% for the control, 0.3 mg/kg, i.v., and 1.0 mg/kg, i.v., respectively); and these effects were completely and almost completely abolished by 5-HD (10 mg/kg, i.v.) and HMR-1098, a selective blocker of sarcolemmal K+ATP (sarcK+ATP) channel (6 mg/kg, i.v.) administered 5 min prior to KR-31761 (72.3% and 67.9%, respectively). KR-31761 only slightly relaxed methoxamine-precontracted rat aorta (IC50: >30.0 μM). These results suggest that KR-31761 exerts potent cardioprotective effects through the opening of both mitoK+ATP and sarcK+ATP channels in rat hearts with a minimal vasorelaxant effect. This paper estimates the economic impacts of the various liberalization scenarios of Russia (accession to the WTO, Russia-EU FTA, and Korea-Russia FTA) using GTAP recursive dynamic and capital accumulation models. To compare liberalization gains from goods liberalization with those from goods-and-services liberalization, the original GTAP database is adjusted by inputting Australian sectoral indices as barriers in the service trade. This paper is differentiated from other studies in that the errors in measurement arising from changes in databases resulting from China's WTO accession are controlled. The major findings and implications of this paper are as follows. First, CGE analyses ignoring the impacts of China's accession to the WTO overestimate China's economic benefits (China-ASEAN: 61%; China-Hong Kong: 112%; China-Korea: 42%; China-Japan-Korea: 25%). Second, CGE analyses ignoring the impacts of China's accession to the WTO overestimate economic benefits for partner countries (China-ASEAN: 59%; China-Hong Kong: 119%), whereas they are underestimated in the case of the China-Korea FTA (42%) and the China-Japan-Korea FTA (25%). The reason for underestimation can be explained by the production network based on vertical division of labor of production processes (fragmentation) in the East Asia. Third, the impacts of China's WTO accession on the Chinese economy are far greater than those of China's FTAs with considering countries. Fourth, the gains created from China's FTA policies are very small or almost negligible. Combining the third and fourth implications, it can be said that China continues to enjoy economic gains from free trade governed by multilateral liberalization of institutions and market forces. This also implies that China is likely to use FTA policy as a strategic tool to thwart possible changes in international order away from the present free-trade and multilateral system, which prevent China from sustainable realization of economic gains from trade.

Flammulina velutipes is a fungus with health and medicinal benefits that has been used for consumption and cultivation in East Asia. F. velutipes is also known to degrade lignocellulose and produce ethanol. The overlapping interests of mushroom production and wood bioconversion make F. velutipes an attractive new model for fungal wood related studies. Here, we present the complete sequence of the F. velutipes genome. This is the first sequenced genome for a commercially produced edible mushroom that also degrades wood. The 35.6-Mb genome contained 12,218 predicted protein-encoding genes and 287 tRNA genes assembled into 11 scaffolds corresponding with the 11 chromosomes of strain KACC42780. The 88.4-kb mitochondrial genome contained 35 genes. Well-developed wood degrading machinery with strong potential for lignin degradation (69 auxiliary activities, formerly FOLymes) and carbohydrate degradation (392 CAZymes), along with 58 alcohol dehydrogenase genes were highly expressed in the mycelium, demonstrating the potential application of this organism to bioethanol production. Thus, the newly uncovered wood degrading capacity and sequential nature of this process in F. velutipes, offer interesting possibilities for more detailed studies on either lignin or (hemi-) cellulose degradation in complex wood substrates. The mutual interest in wood degradation by the mushroom industry and (ligno-)cellulose biomass related industries further increase the significance of F. velutipes as a new model.

Ganoderma lucidum, a species of the Basidiomycetes class, has been attracting international attention owing to its wide variety of biological activities and great potential as an ingredient in skin care cosmetics including “skin-whitening” products. However, there is little information available on its inhibitory effect against tyrosinase activity. Therefore, the objectives of this study were to investigate the chemical composition of G. lucidum and its inhibitory effects on melanogenesis. We isolated the active compound from G. lucidum using ethanol extraction and ethyl acetate fractionation. In addition, we assayed its inhibitory effects on tyrosinase activity and melanin biosynthesis in B16F10 melanoma cells. In this study, we identified a bioactive compound, ganodermanondiol, which inhibits the activity and expression of cellular tyrosinase and the expression of tyrosinase-related protein-1 (TRP-1), TRP-2, and microphthalmia-associated transcription factor (MITF), thereby decreasing melanin production. Furthermore, ganodermanondiol also affected the mitogen-activated protein kinase (MAPK) cascade and cyclic adenosine monophosphate (cAMP)-dependent signaling pathway, which are involved in the melanogenesis of B16F10 melanoma cells. The finding that ganodermanondiol from G. lucidum exerts an inhibitory effect on tyrosinase will contribute to the use of this mushroom in the preparation of skin care products in the future.

A simple, rapid, and convenient colorimetric chemosensor of a specific target toward the end user is still required for on-site detection and real-time monitoring applications. In this study, we developed a rapid in situ colorimetric assay for cobalt detection using the naked eye. Interestingly, a yellow to light orange visual color transition was observed within 3 s when a Chrysoidine G (CG) chemosensor was exposed to cobalt. Surprisingly, the CG chemosensor had great selectivity toward cobalt without any interference of other metal ions. Under optimized conditions, a lower detection limit of 0.1 ppm via a spectrophotometer and a visual detection limit of 2 ppm with a linear range from 0.4 to 1 ppm (R2 = 0.97) were determined. Moreover, the CG chemosensor is reversible and maintains its functionality after treatment with chelating agents. In conclusion, we show the superior capabilities of the CG chemosensor, which has the potential to provide extremely facile handling, high sensitivity, and a fast response time for applications of on-site detection to real-time cobalt monitoring for the general public.

Hydrogel capsules synthesized by conventional water-in-oil emulsion systems are the less preferred choice for biomedical applications due to the use of oils and surfactants. An aqueous two-phase system (ATPS), which allows the formation of water-in-water emulsion, is considered a green alternative and therefore has been explored a lot for its application in the biomedical field. Herein, we present the synthesis of hydrogel capsules using a set-up consisting of a pneumatic valve integrated with the ATPS microfluidic system. In this arrangement, at first, a pneumatic valve facilitates the generation of the droplets of one aqueous system i.e. sodium alginate (SA) containing dextran solution into another aqueous phase comprising polyethylene glycol solution. The present approach allows good control over droplet generation by tuning the pressure of the pneumatic valve and the flow rates of the core and middle phases. The synthesis of hybrid capsules within the microfluidic device is carried out mainly by using the interfacial complexation of oppositely charged polyelectrolytes, chitosan with SA via electrostatic interactions. The interfacial complexed SA and chitosan hydrogel capsules were collected via the settling collection method, which ensures the retaining of the shape of the hybrid capsules. The morphological properties of as-synthesized droplets and hybrid capsules were examined via optical microscopy. The hydrogel capsules show good encapsulation capability for the magnetic particles. Even though this study mainly focuses on the synthesis part, we anticipate that the proposed approach will enable the encapsulation of cells within the hybrid capsules as well as enhance the cell adhesion on the surface of the hydrogel capsules hence, these hydrogel capsules can find the potent application in the biomedical engineering. Toxicity assessment using microalgae adopted various endpoint measurements like mortality rate, photosynthetic activity, chlorophyll content, enzymatic activity, cell density, 14C assimilation, and phosphate uptake. These algal toxicity tests usually require 3-4 days of exposure time and laborious work to measure these endpoints parameters. In the present study, we described a simple and rapid toxicity assessment procedure using photosynthetic oxygen evolution as an endpoint measurement to determine heavy metal-induced toxicity. Oxygen evolution in gaseous phase was measured over a 12 h exposure time. The toxicity of six selected heavy metals was assessed. Concentrations of 1.02, 1.91, 0.46, 3.2, 7.5, and 65.6 mg/L were obtained as EC50 values for mercury, silver, cyanide, copper, cadmium, and hexavalent chromium, respectively, for microalgae (Chlorella vulgaris). Our results demonstrated that hexavalent chromium is less sensitive depending on the conditions (low exposure time, pH 6, and high initial biomass). Toxicities in decreasing order of strength were cyanide > mercury > silver > copper > cadmium > hexavalent chromium. Results obtained in the current and earlier studies demonstrate that photosynthetic oxygen evolution offers an alternative endpoint measurement to assess toxicity since it allows sensitive and rapid detection of heavy metal-induced toxicity in water. The differences in phenotype and antibiotic production between a deletion mutant and a wild-type strain (WT) were compared to demonstrate the function of an unknown regulator in Streptomyces. The method which allowed to newly characterize a member of the MarR-like regulator family, SCO5405 (AbsC) to control pyruvate dehydrogenase in Streptomyces coelicolor could also be used as high-throughput approach for the characterization of unknown regulators.

Highly porous zeolitic-imidazole frameworks (ZIFs) are synthesized to produce N-doped mesoporous carbon electrocatalysts via calcination. The N-doped carbon (m-NC) and carbon nanotubes (m-NCNT) are obtained from ZIF-8 and ZIF-67, while the core-shell structure of ZIF-8@ZIF-67 produced with ZIF-8 seeds (m-NC@NCNT) is prepared by hydrothermal method. Chemical and optical evaluations of the catalysts are characterized using BET, FT-IR, XPS, XRD, Raman spectroscopy and SEM/STEM and they are used as the catalysts for redox reactions of vanadium ions and redox flow battery (VRFB) performance. In the utilization, m-NC@NCNT and m-NCNT are effective for improving VO2+/VO2 + redox reaction, although m-NC does not influence that. Even in VRFB tests using the catalysts, charge/discharge potential and energy efficiency (EE) of m-NC@NCNT and m-NCNT are highest, not to mention excellent EE resilience after undergoing tougher cycling condition. These results are due to the large graphitic-N portion of the two catalysts. Namely, electrons produced by the graphitic-N are delocalized, forming pi-conjugated system and vanadium-nitrogen transition state. The microbial communities in recreational freshwaters play important roles in both environmental and public health perspectives. In this study, the bacterial community structure and its associations with freshwater environments were investigated by analyzing the summertime microbiomes of three beach waters in Ohio (East Fork, Delaware, and Madison lakes) together with environmental and microbial water quality parameters. From the swimming season of 2009, 21 water samples were collected from the three freshwater beaches. From the samples, 110,000 quality-checked bacterial 16S rRNA gene sequences were obtained and analyzed, resulting in an observation of 4500 bacterial operational taxonomic units (OTUs). The most abundant bacteria were Mycobacterium and Arthrobacter of the Actinobacteria (33.2%), Exiguobacterium and Paenisporosarcina of the Firmicutes (23.4%), Planktothrix and Synechococcus of the Cyanobacteria (20.8%), and Methylocystis and Polynucleobacter of the Proteobacteria (16.3%). Considerable spatial and temporal variations were observed in the bacterial community of Actinobacteria, Cyanobacteria, and Firmicutes, where the bacterial community structure was greatly influenced by hydrological and weather conditions. The most influential factors were (1) water inflow for Bacteroidia and Clostridia, (2) turbidity for Gammaproteobacteria, (3) precipitation for Bacilli, and (4) temperature and pH for Cyanobacteria. One noticeable microbial interaction in the bacterial community was a significant negative relationship between Cyanobacteria and Bacilli (P < 0.05). Concerning beach water quality, the level of the genetic markers for cyanobacterial toxin (mcyA) was linked to the abundance of Cyanobacteria. In addition, unique distributions of the genera Enterococcus, Staphylococcus, Streptococcus, Bacteroides, Clostridium, Finegoldia, Burkholderia, and Klebsiella, together with a high density of fecal indicator Escherichia coli, were markedly observed in the sample from Madison Lake on July 13, suggesting a distinctly different source of bacterial loading into the lake, possibly fecal contamination. In conclusion, deep sequencing-based microbial community analysis can provide detailed profiles of bacterial communities and information on potential public health risks at freshwater beaches. Canoderma lucidum has a long history of use as a traditional medicine in Asian countries. However, the taxonomy of Ganoderma species remains controversial, since they were initially classified on the basis of their morphological characteristics. Recently, it was proposed that C. lucidum from China be renamed as C. sichuanense or C. lingzhi. In the present study, phylogenetic analysis using the internal transcribed spacer region rDNA sequences of the Ganoderma species indicated that all strains of the Korean 'G. lucidum' clustered into one group together with G. sichuanense and G. lingzhi from China. However, strains from Europe and North American, which were regarded as true G. lucidum, were positioned in a clearly different group. In addition, the average size of the basidiospores from the Korean cultivated Yeongji strains was similar to that of G. lingzhi. Based on these results, we propose that the Korean cultivated Yeongji strains of 'G. lucidum' should be renamed as G. lingzhi. [ABSTRACT FROM AUTHOR]

The aim of this study was to identify and characterize new Flammulina velutipes laccases from its whole-genome sequence. Of the 15 putative laccase genes detected in the F. velutipes genome, four new laccase genes (fvLac-1, fvLac-2, fvLac3, and fvLac-4) were found to contain four complete copper-binding regions (ten histidine residues and one cysteine residue) and four cysteine residues involved in forming disulfide bridges, fvLac-1, fvLac-2, fvLac3, and fvLac-4, encoding proteins consisting of 516, 518, 515, and 533 amino acid residues, respectively. Potential N-glycosylation sites (Asn-Xaa-Ser/Thr) were identified in the cDNA sequence of fvLac-1 (Asn-454), fvLac-2 (Asn-437 and Asn-455), fvLac-3 (Asn-111 and Asn-237), and fvLac4 (Asn-402 and Asn-457). In addition, the first 19~20 amino acid residues of these proteins were predicted to comprise signal peptides. Laccase activity assays and reverse transcription polymerase chain reaction analyses clearly reveal that CuSO4 affects the induction and the transcription level of these laccase genes. We prepared pyrene-labeled peptide amphiphiles (PAs) consisting of a hydrophobic linear- or branchedalkyl chain (for 1 or 2, respectively) and a hydrophilic histidine-rich peptide of HGGGHGHGGGHG (HG12). Both peptides have a strong tendency to form nanofibrils (NFs) in aqueous media. The resulting histidine-coated NFs show a great binding affinity to Cu2+ as a fluorescence "light-off" sensor. Interestingly, the emission spectra of the pyrene probe show that the different supramolecular assemblies between 1 and 2 can significantly affect the binding affinity to specific metal ions. In particular, "light-up" fluorescent Ag+ detection of NFs of 2 through inhibition of photoinduced electron transfer (PET) was observed even at a low concentration of PA solution. As a means to determine the biological responsibility of 2 to Ag+, intracellular detection using the "turn-on" response was performed. A considerable enhancement of fluorescence in NF-loaded HeLa cells was observed. In addition, the NFs were used as a template scaffold for the production of Ag nanoparticles (AgNPs) with high monodispersity and stability. The NFs decorated with AgNPs are shown to possess highly effective and long-term antibacterial activity against both Gram-negative and -positive bacteria. [ABSTRACT FROM AUTHOR]
