The mesoporous silica particles were prepared by the sol–gel method in one-step synthesis, in acidic conditions, from tetraethoxysilane (TEOS) and methyltriethoxysilane (MTES), varying the mole ratio of the silica precursors. Nitric acid was used as catalyst at room temperature and hexadecyltrimethyl ammonium bromide (CTAB) as structure directing agent. Optical properties, porosity and microstructure of the materials in function of the MTES/TEOS ratio were evaluated using infrared spectroscopy, nitrogen adsorption and small angle X-ray scattering. All materials showed the ordered pore structure and the high specific surfaces, making them suitable as the drug delivery systems. Drug loading and release tests using ketoprofen were performed to assess their performance for drug delivery applications. The amount of the methylated precursor used in the synthesis had little effect on the drug loading capacity, but had a strong influence on the initial rate of the drug release.

The thermodynamic and structural behaviors of lamellar dimyristoylphosphatidylcholine-alkanol (abbreviation DMPC–CnOH, n = 8–18 is the even number of carbons in the alkyl chain) systems were studied by using DSC and SAXD/WAXD methods at a 0–0.8 CnOH : DMPC molar ratio range. Up to n ≤ 10 a significant biphasic effect depending on the main transition temperature tm on the CnOH concentration was observed. Two breakpoints were revealed: turning point (TP), corresponding to the minimum, and threshold concentration (cT), corresponding to the end of the biphasic tendency. These breakpoints were also observed in the alkanol concentration dependent change in the enthalpy of the main transition ΔHm. In the case of CnOHs with n > 10 we propose a marked shift of TP and cT to very low concentrations; consequently, only increase of tm is observed. A partial phase diagram was constructed for a pseudo-binary DMPC–C12OH system. We suggest a fluid–fluid immiscibility of the DMPC–C12OH system above cT with a consequent formation of domains with different C12OH contents. At a constant CnOH concentration, the effects of CnOHs on ΔHm and bilayer repeat distance were found to depend predominantly on the mismatch between CnOH and lipid chain lengths. Here it is demonstrated how some anionic food additives commonly used in our diet, such as tartrazine (TZ), bind to DHVAR4, an antimicrobial peptide (AMP) derived from oral host defense peptides, resulting in significantly fostered toxic activity against both Gram-positive and Gram-negative bacteria, but not against mammalian cells. Biophysical studies on the DHVAR4–TZ interaction indicate that initially large, positively charged aggregates are formed, but in the presence of lipid bilayers, they rather associate with the membrane surface. In contrast to synergistic effects observed for mixed antibacterial compounds, this is a principally different mechanism, where TZ directly acts on the membrane-associated AMP promoting its biologically active helical conformation. Model vesicle studies show that compared to dye-free DHVAR4, peptide–TZ complexes are more prone to form H-bonds with the phosphate ester moiety of the bilayer head-group region resulting in more controlled bilayer fusion mechanism and concerted severe cell damage. AMPs are considered as promising compounds to combat formidable antibiotic-resistant bacterial infections; however, we know very little on their in vivo actions, especially on how they interact with other chemical agents. The current example illustrates how food dyes can modulate AMP activity, which is hoped to inspire improved therapies against microbial infections in the alimentary tract. Results also imply that the structure and function of natural AMPs could be manipulated by small compounds, which may also offer a new strategic concept for the future design of peptide-based antimicrobials. A reverse Monte Carlo-type simulation method was developed for the evaluation of anomalous small-angle x-ray scattering data of a Raney-type Ni catalyst. Based on other experimental data the catalytic Ni particles were modeled as small crystalline cylinders dispersed in the matrix. The average size of the Ni particles and their pair-correlation function were determined. Despite the unknown density of the catalyst, it is shown that each particle has about 2 neighbors in the first neighboring shell independent of the modeling density, and the position of the first peak of the pair-correlation function does not depend on the modeling density. A method was elaborated to get reasonable performance of the Reverse Monte Carlo-type simulation. The scattered intensity was calculated on the basis of probe scattering atoms put inside the cylinders. The effects of the omission of the real number of the atoms, the unknown density, the lack of normalization and the uncertainties in the cross sections were unified in two constants that were determined during the simulation. The method can be used for nanoparticles with other shape, where analytic form factors are complicated, and it may be powerful in the investigation of the usually neglected or simplified inter-particle structure of these systems. © 2004 American Institute of Physics. Graphical abstract Abstract Stable unilamellar dipalmitoylphosphatidylcholine vesicles were produced by using oligo(malic acid) and cholesterol. Detailed physico-chemical characterization prove that by using oligo(malic acid) the substitution of PEGylated lipids for sterically stabilization comes possible. The polymer molecules cover the outer surface of spherical-shaped vesicles, and an asymmetrical composition occurs in the two leaflets of the phospholipid bilayer. The oligo(malic-acid) and cholesterol are enriched in the outer side assuring the stabilization of vesicles. Cholesterol plays an important role in the self-assembly of components as it makes the entering of oligomers possible deep into the polar head-region of lipids. The presence of oligo(malic acid) molecules does not induce degradation by hydrolysis of lipid molecules but the vesicle system turns into a sensitive form giving a possibility for pH sensitive targeting. Preliminary investigation on the investigated oligo(malic acid)-stabilized vesicles do not show any toxic effect promising their applicability in the field of liposomal drug delivery. Protease encapsulation and its targeted release in thrombi may contribute to the reduction of haemorrhagic complications of thrombolysis. We aimed to prepare sterically stabilized trypsin-loaded liposomes (SSLT) and characterize their structure and fibrinolytic efficiency. Hydrogenated soybean phosphatidylcholine-based SSLT were prepared and their structure was studied by transmission electron microscopy combined with freeze fracture (FF-TEM), Fourier transform infrared spectroscopy (FT-IR), and small-angle X-ray scattering (SAXS). Fibrinolytic activity was examined at 45, 37, or 24°C on fibrin or plasma clots with turbidimetric and permeation-driven lysis assays. Trypsin was shown to be attached to the inner surface of vesicles (SAXS and FF-TEM) close to the lipid hydrophilic/hydrophobic interface (FT-IR). The thermosensitivity of SSLT was evidenced by enhanced fibrinolysis at 45°C: time to reduce the maximal turbidity to 20% decreased by 8.6% compared to 37°C and fibrin degradation product concentration in the permeation lysis assay was 2-fold to 5-fold higher than that at 24°C. SSLT exerted its fibrinolytic action on fibrin clots under both static and dynamic conditions, whereas plasma clot dissolution was observed only in the permeation-driven assay. The improved fibrinolytic efficiency of SSLT under dynamic conditions suggests that they may serve as a novel therapeutic candidate for dissolution of intravascular thrombi, which are typically exposed to permeation forces. Application of nanoscale materials (NMs) displays a rapidly increasing trend in electronics, optics, chemical catalysis, biotechnology, and medicine due to versatile nature of NMs and easily adjustable physical, physicochemical, and chemical properties. However, the increasing abundance of NMs also poses significant new and emerging health and environmental risks. Despite growing efforts, understanding toxicity of NMs does not seem to cope with the demand, because NMs usually act entirely different from those of conventional small molecule drugs. Currently, large-scale application of available safety assessment protocols, as well as their furthering through case-by-case practice, is advisable. We define a standard work-scheme for nanotoxicity evaluation of NMs, comprising thorough characterization of structural, physical, physicochemical, and chemical traits, followed by measuring biodistribution in live tissue and blood combined with investigation of organ-specific effects especially regarding the function of the brain and the liver. We propose a range of biochemical, cellular, and immunological processes to be explored in order to provide information on the early effects of NMs on some basic physiological functions and chemical defense mechanisms. Novel, unique amphiphilic pentablock terpolymers consisting of the highly hydrophobic polyisobutylene (PIB) mid-segment attached to the hydrophilic combshaped poly(poly(ethylene glycol) methacrylate) (PPEGMA) polymacromonomer chains, which are coupled to poly(methyl methacrylate) (PMMA) outer segments were synthesized by the combination of quasiliving carbocationic polymerization and atom transfer radical polymerization (ATRP). First, a bifunctional PIB macroinitiator was prepared by quasiliving carbocationic polymerization and subsequent quantitative chain end derivatizations. Quasiliving ATRP of PEGMAs with different molecular weights (Mn= 188, 300 and 475 g/mol) led to triblock copolymers which were further reacted with MMA under ATRP conditions to obtain PMMA-PPEGMA-PIB-PPEGMA-PMMA ABCBA-type pentablock copolymers. It was found that slow initiation takes place between the PIB macroinitiator and PEGMA macromonomers with higher molecular weights via ATRP. ATRP of MMA with the resulting block copolymers composed of PIB and PPEGMA chain segments led to the desired block copolymers with high initiating efficiency. Investigations of the resulting pentablock copolymers by DSC, SAXS and phase mode AFM revealed that nanophase separation occurs in these new macromolecular structures with average domain distances of 11-14 nm, and local lamellar self-assembly takes place in the pentablocks with PPEGMA polymacromonomer segments of PEGMAs with Mnof 118 g/mol and 300 g/mol, while disordered nanophases are observed in the block copolymer with PEGMA having molecular weight of 475 g/mol. Time-resolved synchrotron small-angle X-ray scattering (SAXS) was used to study the structural changes during the osmotic shrinkage of a pharmacologically relevant liposomal drug delivery system. Sterically stabilized liposomes (SSLs) with a diameter of 100 nm and composed of hydrogenated soy phosphocholine, cholesterol and distearoyl-phosphoethanolamine-PEG 2000 prepared in a salt-free buffer were mixed with a buffered 0.3 M NaCl solution using a stopped flow apparatus. The changes in the liposome size and the bilayer structure were followed by using SAXS with a time resolution of 20 ms. A linear decrease in liposome size is observed during the first ∼4 s of the osmotic shrinkage, which reveals a water permeability value of 0.215 (15) μm s-1. The change in the size of the liposomes upon the osmotic shrinkage is also confirmed by dynamic light scattering. After this initial step, broad correlation peaks appear on the SAXS curves in the q range of the bilayer form factor, which indicates the formation of bi- or oligolamellar structures. Freeze-fracture combined with transmission electron microscopy revealed that lens-shaped liposomes are formed during the shrinkage, which account for the appearance of the quasi-Bragg peaks superimposed on the bilayer form factor. On the basis of these observations, it is proposed that the osmotic shrinkage of SSLs is a twostep process: in the initial step, the liposome shrinks in size, while the area/lipid adapts to the decreased surface area, which is then followed by the deformation of the spherical liposomes into lens-shaped vesicles. The red-emitting fluorescent properties of bovine serum albumin (BSA)–gold conjugates are commonly attributed to gold nanoclusters formed by metallic and ionized gold atoms, stabilized by the protein. Others argue that red fluorescence originates from gold cation–protein complexes instead, not gold nanoclusters. Our fluorescence and infrared spectroscopy, neutron, and X-ray small-angle scattering measurements show that the fluorescence and structural behavior of BSA–Au conjugates are different in normal and heavy water, strengthening the argument for the existence of loose ionic gold–protein complexes. The quantum yield for red-emitting luminescence is higher in heavy water (3.5%) than normal water (2.4%), emphasizing the impact of hydration effects. Changes in red luminescence are associated with the perturbations of BSA conformations and alterations to interatomic gold–sulfur and gold–oxygen interactions. The relative alignment of domains I and II, II and III, III and IV of BSA, determined from small-angle scattering measurements, indicate a loose ("expanded-like") structure at pH 12 (pD ~12); by contrast, at pH 7 (pD ~7), a more regular formation appears with an increased distance between the I and II domains, suggesting the localization of gold atoms in these regions. A novel type of X-ray collimation system attached to commercial powder diffractometers makes the structural characterization of nanomaterials possible in a wide size range from <0.1 to 100 nm by combination of the small- and wide-angle X-ray scattering techniques. There is no dead interval in the detection between the small- and wide-angle regimes. This device can be attached to any existing `θ/θ' powder diffractometer, providing a multi-functional small- and wide-angle X-ray scattering/diffraction (SWAXS) apparatus. Polyurethanes were prepared from 4,4′-methylenebis(phenyl isocyanate), 1,4-butanediol and poly(1,4-butanediol adipate) polyester polyol. The hydroxyl functional group ratio of polyol/total diol was kept constant at 0.4, while the ratio of the isocyanate and hydroxyl groups (NCO/OH ratio) changed between 0.90 and 1.15. The polymers were prepared by one-step bulk polymerization in an internal mixer. They were characterized using a number of methods including Fourier transform infrared spectroscopy, wide- and small-angle X-ray scattering, differential scanning calorimetry, dynamic mechanical analysis, light transmittance and tensile testing. Changing the stoichiometry modifies the molecular weight in accordance with the laws of stepwise polymerization, and the relative concentration of end groups also changes at the same time. Competitive interactions among various groups including chain-end functional groups lead to the formation of slightly ordered phases of sub-nanometre size at both ends of the composition range. These structures assemble into larger units at the 10 nm level, which associate further to even larger entities of micrometre size causing scattering of light and a decreased transparency of the samples. The order of the primary units, together with the number and size of assemblies at both higher levels, decrease as the composition approaches equimolar stoichiometry. The amount of less ordered amorphous phase has a maximum in this range. The stiffness of the polymers is determined by the amount of this phase, while ultimate properties are influenced also by molecular weight and the number of physical crosslinks formed. Abstract: Three series of activated carbon monoliths have been prepared from logs of different trees (spruce (Picea abies), beech (Fagus silvatica), oak (Quercus robur)) by pyrolization in nitrogen flow at 700°C and then by steam activation at 900°C for different duration of the conversion. The characterization of carbon skeleton in the samples was performed by small angle X-ray scattering (SAXS), atomic force microscopy (AFM) and scanning electronmicrography (SEM). The carbonized xylem retained the original tracheal structure of the wood. Biological diversity of wood precursors reflected in the nanometer range likewise than in micrometer range. The evolution of the microporosity has been followed by static gas adsorption (N2 at −196°C). The characterization of dynamic adsorption properties took advantage of the unique potentials of the frequency response (FR) technique (N2 or O2 at −78°C). The obtained three-dimensional carbon frames exhibit low transport diffusional resistance due to the wide straight transport channels of micron size. All of the three wood types displayed nearly identical dependence of burn-off upon activation time, which in view of their similar cellulose composition, is not entirely unexpected. The shape of the isotherms indicates the presence of both micro and mesopores and the history of the activation is crucial for the developing porosity. The changes in the adsorption capacities of micropores reflect the differences of precursors and length of activation time. Abstract: The effect of Salmonella minnesota R595 lipopolysaccharide (LPS) on model membrane consisting of a mixture of fully hydrated lipids (dipalmitoylphosphatidylethanolamine (DPPE) and dipalmitoylglycerol (DPG)) was investigated by differential scanning calorimetry (DSC), small-angle X-ray scattering (SAXS) and freeze–fracture methods. The DPPE–DPG/water system forms a multilamellar arrangement in the gel phase which transforms into a mixture of inverted hexagonal and cubic structures. By the presence of LPS the thermotropic behaviour of the system was affected significantly only at its high concentration (1:1mol/mol LPS/DPPE–DPG) in the gel phase, while above the chain melting transition the ratio of the inverted cubic and the hexagonal structures was changed and at the 1:1mol/mol LPS/DPPE–DPG ratio a complex and amorphous phase was formed. The structural parameters of the inverted hexagonal and cubic phases are modified by the temperature and also by the LPS concentration, as deduced from the characteristic SAXS curves. Summarizing the effects of the LPS molecules on the DPPE–DPG/water vesicle system a schematic phase diagram was constructed. [Copyright &y& Elsevier]
