The observed biological differences in safety and efficacy of intravenous (IV) iron formulations are attributable to physicochemical differences. In addition to differences in carbohydrate shell, polarographic signatures due to ferric iron [Fe(III)] and ferrous iron [Fe(II)] differ among IV iron formulations. Intravenous iron contains Fe(II) and releases labile iron in the circulation. Fe(II) generates toxic free radicals and reactive oxygen species and binds to bacterial siderophores and other in vivo sequestering agents. To evaluate whether differences in Fe(II) content may account for some observed biological differences between IV iron formulations, samples from multiple lots of various IV iron formulations were dissolved in 12 M concentrated HCl to dissociate and release all iron and then diluted with water to achieve 0.1 M HCl concentration. Fe(II) was then directly measured using ferrozine reagent and ultraviolet spectroscopy at 562 nm. Total iron content was measured by adding an excess of ascorbic acid to reduce Fe(III) to Fe(II), and Fe(II) was then measured by ferrozine assay. The Fe(II) concentration as a proportion of total iron content [Fe(III) + Fe(II)] in different lots of IV iron formulations was as follows: iron gluconate, 1.4 and 1.8 %; ferumoxytol, 0.26 %; ferric carboxymaltose, 1.4 %; iron dextran, 0.8 %; and iron sucrose, 10.2, 15.5, and 11.0 % (average, 12.2 %). The average Fe(II) content in iron sucrose was, therefore, ≥7.5-fold higher than in the other IV iron formulations. Further studies are needed to investigate the relationship between Fe(II) content and increased risk of oxidative stress and infections with iron sucrose.

Many microbes acquire environmental Fe by secreting organic chelators, siderophores, which possess the characteristics of a high and specific binding affinity for iron(iii) that results in the formation of thermodynamically stable, and kinetically inert iron(iii) complexes. Mechanisms to overcome the kinetic inertness include the labilization of iron(iii) by means of ternary complex formation with small chelators. This study describes a kinetic investigation of the labilization of iron(iii) between two stable binding sites, the prototypical siderophore ferrioxamine B and EDTA, by the bidentate siderophore mimic, 1,2-dimethyl-3-hydroxy-4-pyridinone (L1, H(DMHP)). The proposed mechanism is substantiated by investigating the iron(iii) exchange reaction between ferrioxamine B and H(DMHP) to form Fe(DMHP)3, as well as the iron(iii) exchange from Fe(DMHP)3 to EDTA. It is also shown that H(DMHP) is a more effective catalyst for the iron(iii) exchange reaction than bidentate hydroxamate chelators reported previously, supporting the hypothesis that chelator structure and iron(iii) affinity influence low denticity ligand facilitated catalysis of iron(iii) exchange reactions. The results are also discussed in the context of the design and use of combination chelator therapies in the treatment of Fe overload in humans.

Previous work suggested that hemoglobin (Hb) tetramer formation slows autoxidation and hemin loss and that the naturally occurring mutant, Hb Providence (HbProv; βK82D), is much more resistant to degradation by H2O2. We have examined systematically the effects of genetic cross-linking of Hb tetramers with and without the HbProv mutation on autoxidation, hemin loss, and reactions with H2O2, using native HbA and various wild-type recombinant Hbs as controls. Genetically cross-linked Hb Presbyterian (βN108K) was also examined as an example of a low oxygen affinity tetramer. Our conclusions are: (a) at low concentrations, all the cross-linked tetramers show smaller rates of autoxidation and hemin loss than HbA, which can dissociate into much less stable dimers and (b) the HbProv βK82D mutation confers more resistance to degradation by H2O2, by markedly inhibiting oxidation of the β93 cysteine side chain, particularly in cross-linked tetramers and even in the presence of the destabilizing Hb Presbyterian mutation. These results show that cross-linking and the βK82D mutation do enhance the resistance of Hb to oxidative degradation, a critical element in the design of a safe and effective oxygen therapeutic.

Bordetella pertussis is the causative agent of whooping cough. This pathogenic bacterium can obtain the essential nutrient iron using its native alcaligin siderophore and by utilizing xeno-siderophores such as desferrioxamine B, ferrichrome, and enterobactin. Previous genome-wide expression profiling identified an iron repressible B. pertussis gene encoding a periplasmic protein (FbpABp). A previously reported crystal structure shows significant similarity between FbpABp and previously characterized bacterial iron binding proteins, and established its iron-binding ability. Bordetella growth studies determined that FbpABp was required for utilization of not only unchelated iron, but also utilization of iron bound to both native and xeno-siderophores. In this in vitro solution study, we quantified the binding of unchelated ferric iron to FbpABp in the presence of various anions and importantly, we demonstrated that FbpABp binds all the ferric siderophores tested (native and xeno) with μM affinity. In silico modeling augmented solution data. FbpABp was incapable of iron removal from ferric xeno-siderophores in vitro. However, when FbpABp was reacted with native ferric-alcaligin, it elicited a pronounced change in the iron coordination environment, which may signify an early step in FbpABp-mediated iron removal from the native siderophore. To our knowledge, this is the first time the periplasmic component of an iron uptake system has been shown to bind iron directly as Fe(3+) and indirectly as a ferric siderophore complex.

Haptoglobin (Hp) is an abundant and conserved plasma glycoprotein, which binds acellular adult hemoglobin (Hb) dimers with high affinity and facilitates their rapid clearance from circulation after hemolysis. Humans possess three main phenotypes of Hp, designated Hp 1-1, Hp 2-1, and Hp 2-2. These variants exhibit diverse structural configurations and have been reported to be functionally nonequivalent. We have investigated the functional and redox properties of Hb-Hp complexes prepared using commercially fractionated Hp and found that all forms exhibit similar behavior. The rate of Hb dimer binding to Hp occurs with bimolecular rate constants of ~0.9 μM(-1) s(-1), irrespective of the type of Hp assayed. Although Hp binding does accelerate the observed rate of HbO2 autoxidation by dissociating Hb tetramers into dimers, the rate observed for these bound dimers is three- to fourfold slower than that of Hb dimers free in solution. Co-incubation of ferric Hb with any form of Hp inhibits heme loss to below detectable levels. Intrinsic redox potentials (E1/2) of the ferric/ferrous pair of each Hb-Hp complex are similar, varying from +54 to +59 mV (vs NHE), and are essentially the same as reported by us previously for Hb-Hp complexes prepared from unfractionated Hp. All Hb-Hp complexes generate similar high amounts of ferryl Hb after exposure to hydrogen peroxide. Electron paramagnetic resonance data indicate that the yields of protein-based radicals during this process are approximately 4 to 5% and are unaffected by the variant of Hp assayed. These data indicate that the Hp fractions examined are equivalent to one another with respect to Hb binding and associated stability and redox properties and that this result should be taken into account in the design of phenotype-specific Hp therapeutics aimed at countering Hb-mediated vascular disease.

We compared oxygenation and anaerobic oxidation reactions of a purified complex of human hemoglobin (Hb) and haptoglobin (Hb-Hp) to those of uncomplexed Hb. Under equilibrium conditions, Hb-Hp exhibited active-site heterogeneity and noncooperative, high-affinity O(2) binding (n(1/2)=0.88, P(1/2)=0.33 mm Hg in inorganic phosphate buffer at pH 7 and 25 °C). Rapid-reaction kinetics also exhibited active-site heterogeneity, with a slower process of O(2) dissociation and a faster process of CO binding relative to uncomplexed Hb. Deoxygenated Hb-Hp had significantly reduced absorption at the λ(max) of 430 nm relative to uncomplexed Hb, as occurs for isolated Hb subunits that lack T-state stabilization. Under comparable experimental conditions, the redox potential (E(1/2)) of Hb-Hp was found to be +54 mV, showing that it is much more easily oxidized than uncomplexed Hb (E(1/2)=+125 mV). The Nernst plots for Hb-Hp oxidation showed no cooperativity and slopes less than unity indicated active-site heterogeneity. The redox potential of Hb-Hp was unchanged by pH over the range of 6.4-8.3. Exposure of Hb-Hp to excess hydrogen peroxide (H(2)O(2)) produced ferryl heme, which was found to be more kinetically inert in the Hb-Hp complex than in uncomplexed Hb. The negative shift in the redox potential of Hb-Hp and its stabilized ferryl state may be central elements in the protection against Hb-induced oxidative damage afforded by formation of the Hb-Hp complex.

Described here is a mass spectrometry-based protocol to study the thermodynamic stability of proteins and protein-ligand complexes using the chemical denaturant dependence of the slow H/D exchange reaction of the imidazole C(2) proton in histidine side chains. The protocol is developed using several model protein systems including: ribonuclease (Rnase) A, myoglobin, bovine carbonic anhydrase (BCA) II, hemoglobin (Hb), and the hemoglobin-haptoglobin (Hb-Hp) protein complex. Folding free energies consistent with those previously determined by other more conventional techniques were obtained for the two-state folding proteins, Rnase A and myoglobin. The protocol successfully detected a previously observed partially unfolded intermediate stabilized in the BCA II folding/unfolding reaction, and it could be used to generate a K(d) value of 0.24 nM for the Hb-Hp complex. The compatibility of the protocol with conventional mass spectrometry-based proteomic sample preparation and analysis methods was also demonstrated in an experiment in which the protocol was used to detect the binding of zinc to superoxide dismutase in the yeast cell lysate sample. The yeast cell sample analyses also helped define the scope of the technique, which requires the presence of globally protected histidine residues in a protein's three-dimensional structure for successful application.

The structural basis of the extreme pH dependence of oxygen binding to Root effect Hbs is a long-standing puzzle in the field of protein chemistry. A previously unappreciated role of steric factors in the Root effect was revealed by a comparison of pH effects on oxygenation and oxidation processes in human Hb relative to Spot ( Leiostomus xanthurus ) and Carp ( Cyprinodon carpio ) Hbs. The Root effect confers five-fold increased pH sensitivity to oxygenation of Spot and Carp Hbs relative to Hb A 0 in the absence of anionic effectors, and even larger relative elevations of pH sensitivity of oxygenation in the presence of 0.2 M phosphate. Remarkably, the Root effect was not evident in the oxidation of the Root effect Hbs. This finding rules out pH-dependent alterations in the thermodynamic properties of the heme iron, measured in the anaerobic oxidation reaction, as the basis of the Root effect. The alternative explanation supported by these results is that the elevated pH sensitivity of oxygenation of Root effect Hbs is attributable to globin-dependent steric effects that alter oxygen affinity by constraining conformational fluidity, but which have little influence on electron exchange via the heme edge. This elegant mode of allosteric control can regulate oxygen affinity within a given quaternary state, in addition to modifying the T–R equilibrium. Evolution of Hb sequences that result in proton-linked steric barriers to heme oxygenation could provide a general mechanism to account for the appearance of the Root effect in the structurally diverse Hbs of many species.

The reduction potentials for two Wurster’s crowns, aza crown ethers which incorporate the redox active N , N , N ′, N ′-tetraalkyl-1,4-phenylenediamine into the structure of 18-crown-6, were studied in the presence of the siderophore ferrioxamine B, FeHDFB + . Addition of FeHDFB + resulted in a positive shift in the host reduction potential for both aza crown ethers studied. This shift is explained in terms of host-guest supramolecular assembly formation, which was independently verified by FAB-MS. An enhanced affinity for host-guest formation of the reduced aza crown ether was calculated for each aza crown ether-siderophore assembly using a thermochemical cycle. These differences in host binding affinity as a function of redox state can be harnessed for use in specific metal ion compartmentalization with application, for example, to environmental remediation.

Ferric binding protein A (FbpA) plays a central role in the iron acquisition processes of pathogenic Neisseria gonorrheae, Neisseria meningitidis, and Haemophilus influenzae. FbpA functions as an iron shuttle within the periplasmic space of these Gram-negative human pathogens. Iron is picked up by FbpA at the periplasmic aspect of the outer membrane with concomitant acquisition of a synergistic anion. Here we report the kinetics and mechanisms involved with loading of iron(III) into iron-free FbpA using iron(III) citrate as an iron source in the presence of excess citrate or phosphate (physiologically available anions) at pH 6.5. In the presence of excess phosphate, iron(III) citrate loads into apo-FbpA in three kinetically distinguishable steps, while in the presence of excess citrate, only two steps are discernible. A stable intermediate containing iron(III) citrate-bound FbpA is observed in each case. The observation of an additional kinetic step and moderate increase in apparent rate constants suggests an active role for phosphate in the iron insertion process. To further elucidate a mechanism for iron loading, we report on the sequestration kinetics of iron(III) citrate in the presence of phosphate with binding site mutant apo-FbpAs, H9E, E57D, E57Q, Q58A, Y195F, and Y196H. Tyrosine mutations drastically alter the kinetics and hamper iron sequestration ability. H9E, E57D, and E57Q have near native iron sequestration behavior; however, iron binding rates are altered, enabling assignment of sequential side chain interactions. Additionally, this investigation elaborates on the function of FbpA as a carrier for iron chelates as well as "naked" or free iron as originally proposed.

An ESI-MS method for the determination of host–guest association constants and binding selectivities for systems of binary and complex mixtures, respectively, is presented in an effort to quantitatively study the strength of host–guest interactions, while accounting for the different ESI-MS spray efficiencies of each component. Association constants were obtained for host–guest complexes formed between a series of crown ether and linear ionophore hosts and various guests, including protonated amines, and an apo- and ferri-siderophore, ferrioxamine B.

Studies of structure-function relationships in the respiratory proteins of marine mammals revealed unexpected variations in the number and types of hemoglobins (Hbs) present in coastal bottlenose dolphins, Tursiops truncatus. We obtained blood samples from free-ranging coastal bottlenose dolphins as a component of capture-release studies. We found that the oxygen-binding functions of bottlenose dolphin blood are poised between effector-saturated and unsaturated levels, enabling exercise-dependent shifts in oxygen transfer functions. Isolated bottlenose dolphin Hbs showed elevated pH sensitivities (Bohr effects) and appreciably lower oxygen affinities than adult human Hb in the absence of allosteric effectors. These properties may be an adaptive modification that enhances oxygen delivery during diving episodes when oxygen tensions and effector levels are low. The Hbs of individual dolphins showed similar oxygen affinities, responses to effectors, and expression of heme-heme interaction in oxygen binding, but differed in their redox potentials and rates of autoxidation. The heterogeneity suggested by these functional variations in Hbs of individual dolphins was born out by variations in the molecular weights and numbers of their alpha and beta globin chains. Although coastal bottlenose dolphins were expected to have a single type of Hb, the mass differences observed revealed considerable genetic diversity. There were multiple Hb forms in some individuals and differences in Hb patterns among individuals within the same community.

While the naturally occurring reducing agents glutathione (GSH) and ascorbate (H2A) alone are ineffective at reducing iron(III) sequestered by the siderophore ferrioxamine B, the addition of an iron(II) chelator, sulfonated bathophenanthroline (BPDS), facilitates reduction by either reducing agent. A mechanism is described in which a ternary complex is formed between ferrioxamine B and BPDS in a rapidly established pre-equilibrium step, which is followed by rate limiting reduction of the ternary complex by glutathione or ascorbate. Spectral, thermodynamic, and kinetic evidence are given for ternary complex formation. Ascorbate was found to be slightly more efficient at reducing the ternary complex than glutathione (k4=2.1 x 10(-3) M(-1) s(-1) and k4=6.3 x 10(-4) M(-1) s(-1), respectively) at pH 7. Reduction is followed by a rapid ligand exchange step where iron is released from ferrioxamine B to form tris-(BPDS)iron(II). The implications of these results for siderophore mediated iron transport and release are discussed.

The B3LYP/cc-pvtz approach is used to obtain Kohn–Sham wavefunctions for NO2 and N2O4 and several related compounds. ELF and AIM analyses on these systems lead to a better understanding of the unusual character of the NN bond in N2O4 in which competing Lewis structures with one or two electrons on the nitrogen atom in NO2 lead to a very weak sigma covalent bond that receives essentially no bonding contribution from the pπ part of the molecule.

In this mini-review we present an environmental iron mobility/transport scheme consisting of inter-related controls, whereby the first coordination shell of iron modulates the iron redox potential (E1/2), and the oxidation state of iron controls the chemistry of the first coordination sphere and therefore the immediate chemical environment of the iron. Siderophores (microbially generated iron specific chelators) may be viewed as iron redox mediators. Siderophore chelation of environmental iron in a reduced (Fe(II)) oxidation state results in facile air oxidation of iron due to the negative redox potentials observed for Fe-siderophore complexes. This solubilizes the iron and locks it into a specific coordination environment, thereby preventing hydrolysis and precipitation. The high-spin Fe3+ → Fe+ electron transfer process may be viewed as a switch that controls the thermodynamic stability and kinetic lability of the first coordination shell. Nitric oxide (NO) is an important signaling molecule. Relatively long-lived NO adducts at the heme and SH groups of hemoglobin (Hb) could enable NO to carry out long-range signaling functions. In spite of significant advances, there remain as yet unresolved issues regarding the possible role of Hb in moderating NO-signaling events that affect blood pressure regulation. In this review, we summarize recent reports concerning the redox and allosteric aspects of NO/Hb interactions that have advanced our understanding of the physiological significance of NO binding to heme groups (forming NO-Hb) and of reactions promoting formation of S-nitrosated Hb (SNO-Hb). Allosteric mechanisms modify the bioactivity of NO/Hb complexes by altering the lifetime of NO-Hb and the properties of SNO-Hb. Redox reactions are significant because of the complex chemistry possible for NO and its oxidation products. Reactions at ferrous and ferric heme sites have differing consequences and affinities for interactions with NO. Moreover, redox changes at heme groups affect reactivity of SH groups and vice versa. In spite of low levels of NO-Hb and SNO-Hb found in vivo, recent findings do not rule out participation of NO-Hb or SNO-Hb in NO-dependent signaling reactions.

A long-standing puzzle with regard to protein structure/function relationships is the proton-dependent modification of haemoglobin (Hb) structure that causes oxygen to be unloaded from Root effect Hbs into the swim bladders and eyes of fish even against high oxygen pressure gradients. Although oxygen unloading in Root effect Hbs has generally been attributed to proton-dependent stabilization of the T-state, protonation of Root effect Hbs can alter their ligand affinities in both R- and T-state conformations and either stabilize the T-state or destabilize the R-state. The C-terminal residues that are so important in the Bohr effect of human Hb appear to be involved in the Root effects of some fish Hbs and not in others, indicating that several evolutionary pathways have resulted in expression of highly pH-dependent Hbs. New data are presented that show surprising similarities in the pH- and anion-dependence of sulfhydryl group reactivity and anaerobic oxidation of human and fish Hbs. The available evidence supports the concept that in both Bohr effect and Root effect Hbs a large steric component acts in addition to quaternary shifts between R and T conformations to regulate ligand affinity. Allosteric effectors moderate these steric effects within both R- and T-state conformations and allow for an elegant match between Hb function and the wide-ranging physiological needs of diverse organisms.

The coordination chemistry of an extracellular siderophore produced by Mycobacterium smegmatis, exochelin MS (ExoMS), is reported along with its pKa values, Fe(III) and Fe(II) chelation constants, and aqueous solution speciation as determined by spectrophotometric and potentiometric titrations. Exochelin MS has three hydroxamic acid groups for Fe(III) chelation and has four additional acidic protons from a carboxylic acid group and three primary amine groups, on the backbone of the molecule. The pK a values for the three hydroxamic acid moieties, the carboxylic acid group and the alkylammonium groups on ExoMS, correspond well with the literature values for these moieties. Equilibrium constants for proton-dependent Fe(III)-ExoMS equilibria were determined using a model involving the sequential protonation of the Fe(III)-ExoMS complexes at the first and second coordination shells. The equilibrium constants (β) for the overall formation of Fe(III)ExoMS(H 3 ) 2 + and Fe(II)ExoMS(H 3 ) + from Fe ( a q ) 3 + or Fe ( a q ) 2 + and the deprotonated hydroxamate coordinating group form of the siderophore, ExoMS(H 3 ) - , are calculated as log β I I I = 28.9 and log β I I = 10.1. A calculated pFe value of 25.0 is very similar to that of other linear trihydroxamic acid siderophores, and indicates that ExoMS is thermodynamically capable of removing Fe(III) from transferrin. The E 1 / 2 for the Fe(lll)-ExoMS/Fe(II)-ExoMS couple was determined from quasi reversible cyclic voltammograms at pH = 6.5 and found to be -380 mV.

Carrier-facilitated bulk liquid membrane (BLM) transport from an aqueous source phase through a chloroform membrane phase to an aqueous receiving phase was studied for various hydrophilic synthetic and naturally occurring Fe(III)-siderophore complexes using first coordination sphere recognition. Iron transport systems were designed such that two cis coordination sites on a hydrophilic Fe(III) complex are occupied by labile aquo ligands, while the other four coordination sites are blocked by strong tetradentate ligands (siderophores). The labile aquo coordination sites can be "recognized" by a liquid membrane-bound hydrophobic bidentate ligand, which carries the hydrophilic Fe(III)-siderophore complex across the hydrophobic membrane to an aqueous receiving phase. The system is further designed for uphill transport of Fe(III) against a concentration gradient, driven by anti-port H(+) transport. Three tetradentate siderophore and siderophore mimic ligands were investigated: rhodotorulic acid (H(2)L(RA)), alcaligin (H(2)L(AG)), and N,N'-dihydroxy-N,N'-dimethyldecanediamide (H(2)L(8)). Flux values for the transport of Fe(L(x))(OH(2))(2)(+) (x = RA, AG, 8) facilitated by the hydrophobic lauroyl hydroxamic acid (HLHA) membrane carrier were the highest when x = 8, which is attributed to substrate lipophilicity. Ferrioxamine B (FeHDFB(+)) was also selectively transported through a BLM by HLHA. The process involves partial dechelation of ferrioxamine B to produce the tetradentate form of the complex (Fe(H(2)DFB)(OH(2))(2)(2+)), followed by ternary complex formation with HLHA (Fe(H(2)DFB)(LHA)(+)) and transport across the membrane into the receiving phase. Uphill transport of ferrioxamine B was confirmed by increased flux as [H(+)](source phase) < [H(+)](receiving phase). The membrane flux of ferrioxamine B occurs near neutral pH, as evidence that ternary complex formation and ligand exchange are viable processes at the membrane/receptor surface of microbial cells.

In this mini-review we describe selected aspects of the coordination chemistry relevant to siderophore mediated iron transport and bioavailability. Specific emphasis is placed on a discussion of in vitro kinetic and thermodynamic data that are relevant to elucidating possible in vivo mechanisms for environmental iron acquisition by microbial cells.

Linear synthetic dihydroxamic acids ([CH3N(OH)C=O)]2(CH2)n; H2Ln) with short (n = 2) and long (n = 8) hydrocarbon-connecting chains form mono- and dinuclear complexes with Fe(III) in aqueous solution. At conditions where the formation of Fe2(Ln)3 is favored, complexes with each of the two ligand systems undergo [H+]-induced ligand dissociation processes via multiple sequential and parallel paths, some of which are common and some of which are different for the two ligands. The pH jump induced ligand dissociation proceeds in two major stages (I and II) where each stage is shown to be comprised of multiple components (Ix, where x = 1-3 for L2 and L8, and IIy, where y = 1-3 for L2 and y = 1-4 for L8). A reaction scheme consistent with kinetic and independent ESI-MS data is proposed that includes the tris-chelated complexes (coordinated H2O omitted for clarity) (Fe2(Ln)3, Fe2(L2)2(L2H)2, Fe(LnH)3, Fe(L8)(L8H)), bis-chelated complexes (Fe2(Ln)2(2+), Fe(LnH)2+, Fe(L8)+), and monochelated complexes (Fe(LnH)2+). Analysis of kinetic data for ligand dissociation from Fe2(Ln)(LnH)3+ (n = 2, 4, 6, 8) allows us to estimate the dielectric constant at the reactive dinuclear Fe(III) site. The existence of multiple ligand dissociation paths for the dihydroxamic acid complexes of Fe(III) is a feature that distinguishes these systems from their bidentate monohydroxamic acid and hexadentate trihydroxamic acid counterparts and may be a reason for the biosynthesis of dihydroxamic acid siderophores, despite higher environmental molar concentrations necessary to completely chelate Fe(III).

Bulk liquid membrane (BLM) transport of ferrioxamine B (FeHDFB+) across an artificial hydrophobic membrane facilitated by neutral and ionizable carriers was studied. Under controlled conditions, ferrioxamine B transport by neutral ionophores follows a simple theoretical model for a diffusion controlled process which predicts a linear dependence of flux upon aqueous-to-chloroform extraction constants. Unsubstituted 18C6 crown ether exhibits the best transport properties among the neutral carriers investigated with a flux of 1.4 × 10–10 mol s–1 cm–2. When transport of FeHDFB+, metal-free desferrioxamine B, H4DFB+, and AlHDFB+ mediated by DC18C6 crown ether was followed, a selectivity was observed with a ratio of measured relative fluxes of 5∶3∶1, respectively. An ionizable carrier lasalocid {6-[7-[5-ethyl-5-(5-ethyltetrahydro-5-hydroxy-6-methyl-2H-pyran-2-yl)tetrahydro-3-methyl-2-furanyl]-4-hydroxy-3,5-dimethyl-6-oxonoyl]-2-hydroxy-3-methylbenzoic acid}, negatively charged at neutral pH, outperformed the crown ethers as a ferrioxamine B transporting agent. However, transport by ionizable carriers follows a different mechanism and occurs only when an electrolyte is present in the receiving phase. This requirement was exploited by designing a BLM experiment in which H4DFB+ acts as a counter cation at the receiving interface, whereby FeHDFB+ and H4DFB+ were transported in opposite directions in a synergistic fashion. When the ionizable salicylic acid moiety of lasalocid is transformed into an ester, lasalocid almost entirely loses its transporting properties. This suggests a co-operative mode of binding in which electrostatic recognition of the FeHDFB+ cation by lasalocid anion is a prerequisite for a subsequent encapsulation of the amine site in a host–guest fashion.

While there has been extensive interest in the use of boron isotope ratios as a surrogate of pH in paleoclimate studies in the context of climate change-related questions, the high (0.4 mM) concentration and the depth-independent (conservative or non-nutrient-like) concentration profile of this element have led to boron being neglected as a potentially biologically relevant element in the modern ocean. Here we report that boron affects the expression of a number of protein and genes in the “algal-associated” Gram-negative marine bacterium Marinobacter algicola DG893. Most intriguingly, a number of these proteins and genes are related to iron uptake. In a recent separate publication we have shown that boron regulates one such iron transport related protein, i.e. the periplasmic iron binding protein FbpA via a direct interaction of the metalloid with this protein. Here we show that a number of other iron uptake related genes are also affected by boron but in the opposite way i.e. they are up-regulated. We propose that the differential effect of boron on FbpA expression relative to other iron transport related genes is a result of an interaction between boron and the global iron regulatory protein Fur.

Second-sphere co-ordination of ferrioxamine B [Fe(HL)]+ by dicyclohexano-18-crown-6 (eicosahydrodibenzo[b,k][1,4,7,10,13,16]hexaoxacyclooctadecine) occurs through host–guest complex formation with the protonated amine side chain (CH2)5NH3+. The influence of steric factors on the association constants (Ka) for both nitrate and perchlorate salts of ferrioxamine B with cis-syn-cis and cis-anti-cis isomers of dicyclohexano-18-crown-6 (CE) in CHCl3 has been investigated. For comparison purposes, association of the perchlorate salt of the indium(III) analogue of ferrioxamine B [In(HL)]+ and the picrate salts of NH4+ and alkali-metal cations are included in this study (picrate = 2,4,6-trinitrophenolate). Although the absolute values of Ka for [M(HL)]X association with CE are strongly dependent on the anion identity, the selectivity ratio expressed for the syn crown ether isomer is 3 : 1 and is independent of anion variations. Comparison with alkali-metal and ammonium cations shows that the discrimination between syn and anti isomers of cis-dicyclohexano-18-crown-6 is dependent upon cation size and is the least for K+ where the ionic radius best matches the cavity size. While syn isomer discrimination in CHCl3 is closely related to the ionic radius, in water it is related to the hydrated radius. The syn over anti isomer preference exhibited by alkali-metal and ammonium cations is much more pronounced in water than in chloroform, which is likely due to the steric hindrance imposed by the large effective radius of the cations due to their hydration shell. Discrimination between the syn and anti isomers by [M(HL)]+ in CHCl3 is greater than expected based on comparison with the parent NH4+ ion. This is likely due to the significant steric requirements of the bulky metal complex molecule. The selectivity of the syn over the anti isomer is dominated by the preference of the bulky [M(HL)]+ complex for the less hindered face of the syn isomer.

The association constant for the Fe(III) complex of the linear trihydroxamic acid siderophore ferrioxamine B (FeHDFB + ;HDFB 2 =H 3 N-[(CH 2 ) 5 -N(O)C(O)-(CH 2 ) 2 C(O)NH-] 2 (CH 2 ) 5 -N(O)C(O)-CH 3 binding sulfate (SDS) micelles was deterined by ultrafiltration and kinetics methods. Both methods are in agreement and yield an average value of the association constant K m =98 M -1 . Association constants for the Al(III) analogue of ferrioxamine B, AlHDFB + , and the metal-free ligand deferriferrioxamine B, H 4 DFB + , were also determined by ultrafiltration and found to be K m =90 and 362 M -1 , respectively

One method of mobilization of iron by mycorrhizal organisms is through the secretion of small organic chelators called siderophores. Hydroxamate donor chelators are a common type of siderophore that is frequently used by fungal organisms. The primary siderophore that is produced by fungi from the genera Ceratobasidium and Rhizoctonia is the tripodal trishydroxamate siderophore basidiochrome. To gain some insight into the iron uptake mechanisms of these symbiotic fungi, the iron binding characteristics of basidiochrome were determined. It was found that basidiochrome exhibits a log β 110 of 27.8 ± 0.1 and a pFe value of 25.0. These values are similar to those of another fungal trishydroxamate siderophore, ferrichrome. The similarity in iron affinity between the two siderophores suggests that the structure of the backbone has little influence in complex formation due to the length of the pendant arms, although the identity of the terminating groups of the pendant arms is likely related to complex stability. The role of basidiochrome in the biogeochemical cycling of iron is also discussed.

The electrochemistry of a series of phosphazene polymers {[Ph(Me)=N] x °Ph{R'RC(OH)CH 2 }P=N] y } n where R' is (η 5 -C 5 H 5 )Fe(η 5 -C 5 H 5 ) with varying degrees of ferrocene substitution (3, x=0.94, y=0.06, R=H; 4, x=0.80, y=0.20, R=H; 5, x=0.64, y=0.36, R=CH 3 ; 6, x=0.56, y=0.44, R=H) has been investigated in CH 2 Cl 2 and as films evaporatively deposited on the electrode surface. The E 1/2 of the ferrocene units (463±12 mV vs Ag/AgCl) for the polymer dissolved in CH 2 Cl 2 is essentially independent of the degree of substitution and background electrolyte

The plasma polymerization of triallylphosphine in order to form a crosslinked polymer thin film is described. The plasma polymer thin film has been characterized and shown to contain a high proporation of CC and P groups conserved from the monomer. The accessibilty of the phosphine groups has been demonstrated by reaction with O2, NO, Ni(CO)4, and Fe(CO)5. Reaction with O2, and NO results in a PO moiety formation. Reaction with the gaseous metal carbonyls results in fixation of the metal via complexation by the P groups as a result of exchange with CO ligand(s). This illustrates the utility of a ligand donor group containing plasma polymer as an avenue for the formation of a single phase transition metal containing plasma polymer.

This article describes one of the first examples of concurrent metal evaporation and substituted hydrocarbon plasma polymerization as a general method for preparing thin films containing a metal coordinated to a plasma polymer. In this report, the evaporation of Ni into a triallylphosphine (TAP) glow discharge is described Deposition of a plasma polymer with concurrent reaction of Ni atoms with phosphine groups forms nickel containing triallylphosphine plasma polymer, NiTAPPP. The chemical nature of the product is derived on the basis of infrared spectra taken of the film as formed and after reaction with air, CO, and NO. The extent of the reaction is deduced from gravimetric data obtained during reaction with CO and with NO; each case was followed by evacuation of the gas. The CO gravimetric data also suggest that uncoordinated Ni is predominantly formed at high (Ni evaporation rate)/(plasma polymer deposition rate) ratios; coordinated Ni predominates at low values of this parameter. The ESCA spectra are also obtained for this plasma polymer (NiTAPPP) as well as for that described in the previous paper, in which TAP is plasma polymerized (yielding TAPPP) and subsequently reacted with Ni(CO)4 (TAPPP + Ni(CO)4). In both cases, it was found that Ni is oxidized on contact with air. After such air exposure, ESCA spectra reveal the presence of residual NiP moieties on the surface of TAPPP + Ni(CO)4, but not of NiTAPPP. This is attributed to the greater reactivity of the coordinatively unsaturated Ni sites postulated for NiTAPPP.

Summary A review of the oxidative and nitrosative reactions of cell-free hemoglobin-based oxygen carriers (HBOCs) shows that these reactions are intimately linked and are subject to allosteric control. Cross-linking reactions used to produce HBOCs introduce conformational constraints and result in Hbs with reduced responses to heterotropic and homotropic allosteric effectors. The Nernst plots of heme oxidation of cross-linked HBOCs are shifted to higher potentials relative to unmodified Hb in the absence of allosteric effectors, in accord with their T-state stabilization and right-shifted Hill plots of O2 binding. They exhibit enhanced rates of autoxidation and nitrite-induced oxidation, features that appear due to their having more solvent-accessible heme pockets. The stability of their NO-Hb derivatives varies as a result of allosteric effects on the extent of formation of pentacoordinate NO-heme geometry by a chains and subsequent oxidation of partner b chains. The physiological implications of these findings on the safety, efficacy and design of second generation HBOCs are discussed in the framework of a reaction scheme showing linkages between Hbmediated redox reactions. These redox reactions can drive formation of SNO-Hb and other reactive species and are of significance for the use of cell-free Hbs in vivo. IUBMB Life, 59: 498–505, 2007

The probiotic Escherichia coli strain Nissle 1917 produces four siderophores: the catecholates enterobactin and salmochelin, the hydroxamate aerobactin, and the mixed-type siderophore yersiniabactin. We studied the influence of pH, temperature, and carbon source on the production of these four siderophores. Yersiniabactin and salmochelin were maximally produced under neutral to alkaline conditions (pH 7.0 and 7.6, respectively), whereas aerobactin was maximally produced at a more acidic pH (pH 5.6), which agrees with the slightly higher complex stability of hydroxamates at acidic pH values compared to the catecholates. Under nearly all conditions studied, catecholate siderophore production was higher with glycerol than with glucose as the carbon source. Yersiniabactin production was also higher with glycerol as the carbon source at pH 7.0. At 42 degrees C, strain Nissle 1917 grew poorly or not at all because of the iron-limiting conditions. In a competition experiment between wild-type strain Nissle 1917 and a mutant of this strain with a deletion in the yersiniabactin operon, the wild-type overgrew the mutant at pH 7.0 and 7.6 and not at pH 5.6. These results agree with yersiniabactin production being of greater advantage at neutral and slightly alkaline pH values. The production of four siderophores may help the probiotic E. coli Nissle 1917 to compete with other E. coli strains in the colon. The probiotic strain Nissle 1917 used in our experiments has many characteristics in common with uropathogenic E. coli and other pathogenic strains which also secrete these siderophores. Uropathogenic E. coli strains may need the multitude of siderophores to adapt to the pH of urine, which varies between pH 4.6 and 8.0.

In this mini-review we describe selected aspects of the coordination chemistry relevant to siderophore mediated iron transport and bioavailability. Specific emphasis is placed on a discussion of in vitro kinetic and thermodynamic data that are relevant to elucidating possible in vivo mechanisms for environmental iron acquisition by microbial cells.

pK(a) values for the hydroxamic acid, alpha-NH(3)(+), and epsilon-NH(3)(+) groups of L-lysinehydroxamic acid (LyHA, H(3)L(2+)) were found to be 6.87, 8.89, and 10.76, respectively, in aqueous solution (I = 0.1 M, NaClO(4)) at 25 degrees C. O,O coordination to Fe(III) by LyHA is supported by H(+) stoichiometry, UV-vis spectral shifts, and a shift in nu(CO) from 1648 to 1592 cm(-1) upon formation of mono(L-lysinehydroxamato)tetra(aquo)iron(III) (Fe(H(2)L)(H(2)O)(4)(4+)). The stepwise formation of tris(L-lysinehydroxamato)iron(III) from Fe(H(2)O)(6)(3+) and H(3)L(2+) was characterized by spectrophotometric titration, and the values for log beta(1), log beta(2), and log beta(3) are 6.80(9), 12.4(2), and 16.1(2), respectively, at 25 degrees C and I = 2.0 M (NaClO(4)). Stopped-flow spectrophotometry was used to study the proton-driven stepwise ligand dissociation kinetics of tris(L-lysinehydroxamato)iron(III) at 25 degrees C and I = 2.0 M (HClO(4)/NaClO(4)). Defining k(n) and k(-n) as the stepwise ligand dissociation and association rate constants and n as the number of bound LyHA ligands, k(3), k(-3), k(2), k(-2), k(1), and k(-1) are 3.0 x 10(4), 2.4 x 10(1), 3.9 x 10(2), 1.9 x 10(1), 1.4 x 10(-1), and 1.2 x 10(-1) M(-1) s(-1), respectively. These rate and equilibrium constants are compared with corresponding constants for Fe(III) complexes of acetohydroxamic acid (AHA) and N-methylacetohydroxamic acid (NMAHA) in the form of a linear free energy relationship. The role of electrostatics in these complexation reactions to form the highly charged Fe(LyHA)(3)(6+) species is discussed, and an interchange mechanism mediated by charge repulsion is presented. The reduction potential for tris(L-lysinehydroxamato)iron(III) is -214 mV (vs. NHE), and a comparison to other hydroxamic acid complexes of Fe(III) is made through a correlation between E(1/2) and pFe.

Inhaled silicate dusts may cause lung disease through their surface coordination of iron with subsequent oxidant generation via the Fenton reaction. Pneumoconiosis, irritant bronchitis, focal emphysema, and carcinoma may be produced by oxidants either directly through lipid peroxidation and protein inactivation, or indirectly by oxidant-mediated release of cytokines such as platelet-derived growth factor. The increased incidence of tuberculosis observed among silicate workers could be explained by accumulation of iron complexed by dust particles in the lung and made available to dormant mycobacteria as a virulence factor.

We have demonstrated the feasibility of quantitatively detecting selected toxic materials through their inhibitory effect on an enzyme electrode that utilizes colloidal gold-immobilized horseradish peroxidase and does not require a mediator. Quantitative detection of azide, cyanide, thiourea, sulfide, and dichromate is demonstrated. The sensitivity and inhibition kinetics for this immobilized enzyme electrode are found to be different from those observed previously for homogeneous horseradish peroxidase. Possible reasons for this difference are discussed. Due to the availability of a large number of enzymes and their toxic inhibitors, this work based on immobilized enzyme inhibition coupled to an electrode surface significantly broadens the possible applications of biosensors and offers alternative methods for toxic substance determination.

The effect of both a positive and a negative applied potential on the p-NPA hydrolysis activity of bovine carbonic anhydrase (BCA) immobilized on graphite rods has been investigated. Background experiments show that the pH-activity profile for BCA free in solution is not affected by either a negative or a positive potential applied to graphite rods placed in the same solution. However, the activity of BCA immobilized by covalent attachment to a graphite rod is influenced by a potential externally applied to the graphite rod. An overall increase in activity (as determined by the initial rate of the p-NPA hydrolysis reaction) is observed in the presence of a -0.2 V (Ag/AgCl) applied potential, while decreased activity is evident at +0.6 V (Ag/AgCl). This is indicative of an electrolyte anion effect rather than a local pH effect. In the presence of the specific anion inhibitors Cl(-) and SCN(-), the relative BCA activity increases at -0.2 V (Ag/AgCl) and decreases at +0.6 V (Ag/AgCl) are consistent with the different BCA inhibition constants for Cl(-) and SCN(-). Accelerated loss of immobilized BCA activity also accompanies the application of the external potentials, particularly at +0.6 V (Ag/AgCl). Results described here represent an early example of potentiostatic control of nonredox enzyme activity. Several possible mechanisms are discussed including specific anion inhibition, enzyme surface charge/charged support material interactions, and charged product inhibition. It is likely that a combination of such mechanisms is operational in this system. The implications of external potentials affecting the activity of immobilized enzymes in the design of stable immobilized enzyme electrodes are also discussed.

Techniques for the immobilization of bovine carbonic anhydrase (BCA) on porous silica beads and graphite are presented. Surface coverage on porous silica beads was found to be 1.5 x 10(-5) mmol BCA/m(2), and on graphite it was 1.7 x 10(-3) mmol BCA/m(2) nominal surface area. Greater than 97% (silica support) and 85% (graphite support) enzyme activity was maintained upon storage of the immobilized enzyme for 50 days in pH 8 buffer at 4 degrees C. After 500 days storage, the porous silica bead immobilized enzyme exhibited over 70% activity. Operational stability of the enzyme on silica at 23 degrees C and pH 8 was found to be 50% after 30 days. Catalytic activity expressed as an apparent second-order rate constant K'(Enz) for the hydrolysis of p-nitrophenyl acetate (p-NPA) catalyzed by BCA immobilized on silica beads and graphite at pH 8 and 25 degrees C is 2.6 x 10(2) and 5.6 x 10(2) M(-1)s(-1) respectively. The corresponding K(ENZ) value for the free enzyme is 9.1 x 10(2) M(-1)s(-1). Activity of the immobilized enzyme was found to vary with pH in such a manner that the active site pK, on the porous silica bead support is 6.75, and on graphite it is 7.41. Possible reasons for a microenvironmental influence on carbonic anhydrase pK(a), are discussed. Comparison with literature data shows that the enzyme surface coverage on silica beads reported here is superior to previously reported data on silica beads and polyacrylamide gels and is comparable to an organic matrix support. Shifts in BCA-active site pK(a) values with support material, a lack of pH dependent activity studies in the literature, and differing criteria for reporting enzyme activity complicate literature comparisons of activity; however, immobilized BCA reported here generally exhibits comparable or greater activity than previous reports for immobilized BCA.

The aqueous coordination chemistry of Al3+ and Fe3+ is compared. The differences in electronegativities (Xm ), ionic radii (ri ) and electron configuration (dn ) for the two ions are shown to influence trends in aquo ion structure and reactivity, hydrolysis, and metal complex stability. Similarities in coordination chemistry result in similarities in distribution and modes of transport for Al3+ and Fe3+ in a biological system.

The plasma polymerization of the C2 hydrocarbons in the presence of Fe(CO)5 vapor can lead to the formation of soluble cluster complexes (I) and/or high oxidation state iron both in the form of the oxide (II) and associated with the carboxylate or β‐diketonate ion (III). Analysis of the electron spectroscopy for chemical analysis (ESCA) spectra obtained yields information regarding the nature of I on the one hand, and regarding the prevalence of II relative to III, on the other. The relative yield of I with respect to (II+III) can be controlled, at will, by variation of W/F. This permits control of the coating’s morphology (continuity) as shown by the electrodeposition of Prussian blue on smooth surfaces coated with plasma polymer containing I+II+III.

Reactions of molecular oxygen with copper(I) complexes are of interest partly as a result of the observation that certain marine life utilize a copper based oxygen transport system, hemocyanin, and that copper has been proposed as an active site for the disproportionation of superoxide by the enzyme superoxide dismutase.2,3 Additional interest arises from a general study of the mechanism of autoxidation of transition metal complexes. A limited number of studies have been made on the kinetics of autoxidation of various cuprous complexes.4–13 We wish to report here on the influence of electronic effects on the autoxidation kinetics of a series of substituted phenanthroline complexes of copper(I).
