3. Nematicidal Efficacy of Cymbopogon nardus, Pelargonium graveolens Essential Oils and Annona squamosa Extract against Meloidogyne incognita and M. graminicola

A crude hexane extract of seed kernel was processed into fatty acid methyl esters (FAME) using methanolic-sulfuric acid. Herbal mixtures (seed extracts of A. squamosa and essential oils of Cymbopogon nardus (citronella) and Pelargonium graveolens (geranium)) were evaluated against root knot nematode (Meloidogyne spp). The mortality of J2s of M. incognita in hexane and methanol extracts (AHE+AME) of A. squamosa with geranium oil (GO) ranged from 50.75-89.75% to 71.50-99.00% at 31.25 to 1000 μgmL-1 in 24 to 96 hours. The number of galls after CO+AHE+AME treatment was 4.0 ± 0.0 and 5.67 ± 0.58 galls/seedling and were recorded at 1000 and 500 μgmL-1 treatments on 30 days after inoculation, compared to carbofuran (3.0 ± 0.0 and 4.3 ± 0.6 galls/seedling) at 1000 and 500 μgmL-1 and velum prime (3.6 ± 0.6 and 4.6 ± 0.6 galls/seedling) at 1000 and 500 μgmL-1. The maximum shoot length of brinjal seedlings was recorded in CO+AHE+AME treatment compared to carbofuran (47.3 ± 0.6cm) and velum prime (48.7 ± 0.6 cm) at 1000 μg mL-1 in soil drenching application. Methanol and hexane extracts (AHE+AME) obtained from Annona squamosa seeds were investigated for their nematicidal properties. M. graminicola in direct-seeded rice at the nursery level and M. incognita in brinjal (eggplant) were controlled for up to sixty days when treated with these mixtures.

4. Aphidicidal activity of nano-emulsions of spearmint oil and carvone against Rhopalosiphum maidis and Sitobion avenae

Abstract Different species of aphids, responsible for severe yield losses of cereal crops including wheat, (Triticum aestivum L.) are managed by insecticides, which are harmful to organisms and the environment under field conditions. Therefore, an environment friendly aphidicidal product of plant origin is required. Mentha spicata oil was found to be rich in carvone (81.88%), but the use of its oil and carvone in crop protection is lacking due to their volatility, poor solubility, and stability. A nanoformulaton not only solves these problems but also improve the efficacy and dose of the bioactive compounds. Thus, nano-emulsions of the oil and carvone prepared were characterized, and evaluated against Rhopalosiphum maidis (corn aphid) and Sitobion avenae (wheat aphid) The average droplet size of nano-emulsions of the oil and carvone was found to be 22.1 and 41.21 nm. Nano-emulsion of carvone exhibited higher aphid mortality (LC50 = 0.87–1.94 mg/mL) at 24 h and acetylcholinesterase inhibitory activity (IC50 = 0.07–3.83 mg/mL) compared to the nano-emulsion of the oil (LC50 = 2.87–2.81 mg/mL; IC50 = 1.66–5.34 mg/mL). The repellence index (RI) in nano-emulsion of essential oil was found to be higher (84.73 and 81.72%) at the highest concentration (0.05 µL/cm2) than that of carvone (77.59 and 80.98%) for R. maidis and S. avenae. Further, in silico studies also revealed the favourable binding energy (− 6.6 to − 8.5 kcal/mol) of the main compounds in the oil with acetylcholinesterase, facilitated by hydrophobic interactions and hydrogen bonding. This study suggests that the nano-emulsions of the essential oil and carvone can be explored under field conditions to establish efficacy for their utilization as aphidicidal and repellent products against aphids. In the present study, aphidicial and repellent activities of its essential oil and carvone were reported for the first time against R.maidis and S.avenae.

Fungal diseases cause substantial loss to agricultural crops, affecting both quantities and quality. Although several methods are used for preventing disease incidence, fungicides remain crucial for higher yields and better quality. But in the past, the efficacy of several fungicides has decreased due to increased cases of fungicide resistant. In our pursuit of new effective fungicides, we synthesised a series of twenty 2-Indazol-1-yl-chromen-4-one derivatives (6a- 6t). The characterization of synthesized compounds was performed by several spectroscopic methods including Infrared, Nuclear Magnetic Resonance (1H and 13C) and HRMS. Out of 20 synthesised compounds, 19 (6b- 6t) were found to be novel. All synthesised indazolylchromones showed very good antifungal activity against Sclerotium rolfsii and Fusarium oxysporum. Among the tested compounds, 6t and 6f exhibited very good fungicidal activity against S. rolfsii with an ED50 of 10.10 mg L-1 and 16.18 mg L-1, respectively. In case of Fusarium oxysporum compound 6f displayed good’ activity with an ED50 value of 27.82 mg L-1. Molecular docking study was done to predict the binding sites of most active compounds, 6t and 6f with Cytochrome P450 14alpha -sterol demethylase (CYP51) enzyme using molsoft software. The acute toxicity predictions the of synthesized compounds for fish (LC50,96 Hr), daphnid (LC50, 48 Hr) and green algae (EC50, 96Hr) and the chronic toxicity predictions (ChV) were assessed using Ecological Structure Activity Relationship (ECOSAR) model. As per ECOSAR prediction, all the chemicals are inside AD and not missing predictions.

6. Green synthesis, structure–activity relationships, in silico molecular docking, and antifungal activities of novel prenylated chalcones

A series of 16 novel prenylated chalcones (5A-5P) was synthesized by microwave-assisted green synthesis using 5-prenyloxy-2-hydroxyacetophenone and different benzaldehydes. Comparisons were also performed between the microwave and conventional methods in terms of the reaction times and yields of all compounds, where the reaction times in the microwave and conventional methods were 1–4 min and 12–48 h, respectively. The synthesized compounds were characterized using different spectroscopic techniques, including IR, 1H-NMR, 13C-NMR, and LC-HRMS. The antifungal activities of all compounds were evaluated against Sclerotium rolfsii and Fusarium oxysporum under in vitro conditions and were additionally supported by structure–activity relationship (SAR) and molecular docking studies. Out of the 16 compounds screened, 2’-hydroxy-4-benzyloxy-5’-O-prenylchalcone (5P) showed the highest activity against both S. rolfsii and F. oxysporum, with ED50 of 25.02 and 31.87 mg/L, respectively. The molecular docking studies of the prenylated chalcones within the active sites of the EF1α and RPB2 gene sequences and FoCut5a sequence as the respective receptors for S. rolfsii and F. oxysporum revealed the importance of the compounds, where the binding energies of the docked molecules ranged from −38.3538 to −26.6837 kcal/mol for S. rolfsii and −43.400 to −23.839 kcal/mol for F. oxysporum. Additional docking parameters showed that these compounds formed stable complexes with the protein molecules.

8. Whole-exome sequencing and variant spectrum in children with suspected inherited renal tubular disorder: the East India Tubulopathy Gene Study

Background Inherited tubulopathies are a heterogeneous group of genetic disorders making whole-exome sequencing (WES) the preferred diagnostic methodology. Methods This was a multicenter descriptive study wherein children (< 18 years) with clinically suspected tubular disorders were recruited for molecular testing through WES. Multiplex ligation-dependent probe amplification (MLPA) and Sanger sequencing were done when required. Variants were classified as per American College of Medical Genetics 2015 guidelines and pathogenic (P)/likely pathogenic (LP) variants were considered causative. Results There were 77 index cases (male =73%). Median age at diagnosis was 48 months (IQR 18.5 to 108 months). At recruitment, the number of children in each clinical group was as follows: distal renal tubular acidosis (dRTA) = 25; Bartter syndrome = 18; isolated hypophosphatemic rickets (HP) = 6; proximal tubular dysfunction (pTD) = 12; nephrogenic diabetes insipidus (NDI) = 6; kidney stone/nephrocalcinosis (NC) = 6; others = 4. We detected 55 (24 novel) P/LP variants, providing genetic diagnoses in 54 children (70%). The diagnostic yield of WES was highest for NDI (100%), followed by HP (83%; all X-linked HP), Bartter syndrome (78%), pTD (75%), dRTA (64%), and NC (33%). Molecular testing had a definite impact on clinical management in 24 (31%) children. This included revising clinical diagnosis among 14 children (26% of those with a confirmed genetic diagnosis and 18% of the overall cohort), detection of previously unrecognized co-morbidities among 8 children (sensorineural deafness n = 5, hemolytic anemia n = 2, and dental changes n = 1) and facilitating specific medical treatment for 7 children (primary hyperoxaluria n = 1, cystinosis n = 4, tyrosinemia n = 2). Conclusion WES is a powerful tool in the diagnosis and management of children with inherited tubulopathies in the Indian population. 9. Composition and nematicidal activity of the essential oil from Piper longum against root knot nematode

Piper longum L. (Piperaceae family), commonly known as long pepper, is used in traditional medicine and in recognized system of medicine in India for the treatment of inflammation, pain, epilepsy, arthritis, cancer, ulcer, and asthma in addition to pest control activity. Its fruits are rich in essential oil and possess different biological activities, but oil is not much explored against the root-knot nematodes. Thus, the essential oil from the fruits of P. longum was isolated by hydrodistillation method and analyzed by Gas chromatography-Flame ionisation detector and Mass spectrometry. Analysis showed that the oil is a complex mixture of volatile compounds. (Z)-β-Farnesene (25.08%), β-Caryophyllene (13.57%), α-Humulene (13.37%), 8-Heptadecene (9.28%), Heptadecane (7.07%), α-Patchoulene (5.44%), 3-Heptadecene (5.09%), γ-Elemene (4.37%), β-Humulene (3.69%) and Terpinen-4-ol (1.74%) were identified as main compounds in its oil. The essential oil was evaluated for its nematicidal activity against root-knot nematode at 62.5–1000 ppm concentrations in vitro conditions during 2020–21 at ICAR-Indian Agricultural Research Institute, New Delhi. Results showed promising nematicidal activity of the oil and have caused mortality of juvenile (J2), ranging from 2.33–88.0% after 24–96 hours of treatment compared to untreated control. The oil was further evaluated under the pot conditions in tomato (Solanum lycopercicum L.) (var. Pusa Ruby) at 3000 and 2000 ppm. Results showed a significant reduction in the number of galls/root of tomato with better plant growth in the essential oil based treatment as compared to the untreated control. These results suggest that the oil can be used for the development of nematicidal product for the protection of crops from the infestation of M. incognita.

Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) is an invasive pest native to the American continent. The present study focused on bio-intensive tactics like intercropping, using natural enemies, botanical insecticides and biopesticides for managing S. frugiperda for the organic production of maize in Indian conditions. A total of eight different parasitoids attacking the different stages of S. frugiperda viz., eggs and larvae were found in the study area. The total parasitism rate due to all the parasitoids ranged from 28.37 to 42.44%. The egg-larval parasitoid, Chelonus formosanus Sonan (Hymenoptera: Braconidae) was the dominant parasitoid (12.55%), followed by Chelonus nr. blackburni (Hymenoptera: Braconidae) (10.98%) and Coccygydium sp. (4.85%). About 36.58 percent of the egg masses collected was parasitized by egg parasitoids, among which Telenomus remus (Nixon) (Hymenoptera: Scelionidae) was the dominant parasitoid. The botanicals insecticides such as citronella and annona extract were most effective, resulting in 100% mortality of FAW larvae (168 h after treatment). The essential oil of garlic (100%) was found highly effective in inhibiting egg hatching, followed by geraniol (90.76%). The maize intercropped with lady’s finger (okra) recorded significantly the lowest pest infestation and recorded higher grain yield (6.17 q/ha) than other intercropping systems and control (5.10 q/ha). The overall bioefficacy of commercial biopesticides against the larvae of S. frugiperda was in the following order azadirachtin > Metarhizium anisopliae (Metch.) Sorokin (Hypocreales: Clavicipitaceae) > Beauveria bassiana (Balsamo) Vuillemin (Hypocreales: Clavicipitaceae) at 168 h after treatment.

Introduction Body stores of vitamin D are measured as 'total' serum 25-hydroxy vitamin D (25(OH)D). Its largest component is protein bound and lost in urine in nephrotic syndrome (NS). Our study investigates whether 'free' 25(OH)D levels are a better guide to bone health and need for vitamin D supplementation in patients with steroid-sensitive NS (SSNS). Methods A cross-sectional study was performed in children with SSNS and healthy controls. Blood was tested for albumin, creatinine, calcium, phosphate, ALP, total and free (by direct ELISA) 25(OH)D, iPTH, and urine for protein-creatinine ratio. Results Seventy-nine NS patients (48 in relapse, 31 in remission) and 60 healthy controls were included. The levels of total 25(OH)D were significantly different (lowest in NS relapse and highest in controls) (p < 0.001). Corrected calcium and phosphate levels were normal, and there were no differences in free 25(OH)D, ALP, or iPTH levels between groups. Only total and not free 25(OH)D correlated significantly and negatively with urinary protein creatinine ratios (r.sub.s = - 0.42 vs. 0.04). Free 25(OH)D values of 3.75 and 2.85 pg/ml corresponded to total 25(OH)D levels of 20 and 12 ng/ml, respectively, in healthy controls. Conclusion These results confirm that total 25(OH)D levels are low in NS and related to degree of proteinuria. However levels of free 25(OH)D, ALP, and iPTH did not change in relapse or remission in comparison with healthy controls. Central Asia historically remained pivotal in hegemonic battles of great powers and now china's enhanced regional involvement through BRI and SCO is changing traditional regional paradigm. Historical and descriptive methodology with comparative analysis of qualitative data obtained from secondary sources is used to analyze geopolitical developments for predictions. Decade long journey of BRI has helped Asian GDP substantial growth as a sign of economic development and SCO nexus of Central Asian states under Chinese patronage has shown enhanced cooperation with accelerated connectivity. Geopolitical environment is tilted in favor of Chinese regional hegemony particularly in central Asian region only if china is able to maintain regional security. Global and regional stakeholders ranging from states, business, IGOs, and NGOs should reap the benefits of BRI materialization by venturing business deals with ongoing projects. Purpose of the Study: This paper aims to analyze the series of significant events which contributed to the deterioration of the Pakistan-US relations before November 26 2011, and the strategic, diplomatic, and financial implications of this event for the two countries. Methodology: This qualitative study is based on secondary sources such as journal articles, PhD thesis, newspapers, official reports, and websites. Principal Findings: On November 26 2011, an aerial attack of NATO forces on two of the Pakistani border Military Check posts leaving at least 24 soldiers dead outraged Pakistani people and government. Subsequently, already strained Pakistan-US relations touched the lowest ebb. NATO supplies to Afghanistan were immediately suspended by Pakistan, demanding a formal investigation and apology by the U.S. It is pertinent to note that the Salala incident was only the tip of the iceberg. There were other events before this that contributed to the growing mistrust between the two strategic partners. Application of this Study: This study can be helpful for the foreign policy decision-makers of the U.S. and Pakistan to maintain these bilateral relations on a smooth footing based on mutual trust and respect for each other's sovereignty and security. 21. Pakistan-China Economic Corridor: Security Implications for Indian Ocean Region.

This study aims to analyse the developing security environment of Indian Ocean Region (IOR), in the context of China Pakistan Economic Corridor (CPEC). The study also discusses the important stake holders and strategic and economic significance of IOR. CPEC is aimed to increase regional connectivity by overland routes from Gwadar to Kashgar which will trigger economic development in the region. However, CPEC has its own security implications for the IOR. This is a qualitative study based on the secondary sources. The study finds that the Indian Ocean already has naval presence of major powers and with the commencement of CPEC the Ocean is expected to be heavily militarised by two Asian powers, India and China. An investment of 72 billion US dollars by China on CPEC requires increased Chinese military presence which pushes India and the US to balance. Likewise, India's attempts to enhance its military power vis-à-vis China force Pakistan to balance against India. Thus the CPEC has increased the chances of initiating a deadly arms race in the IOR. - Abstract
Although Acute Kidney Injury (AKI) has been described among childhood diabetes ketocidosis (cDKA) there is scarcity of literature on the role of concomitant rhabdomyolysis. A retrospective chart review was undertaken (2014–2018) to identify cDKA who developed AKI and had evidence of rhabdomyolysis defined by serum creatine phosphokinase (CPK) > 5 times upper limit of normal. 46 cDKA were identified. Ten (22%) developed AKI with 6/10 reaching peak AKI Stage 3 and 8/10 had co-current rhabdomyolysis. In comparison to non rhabdomyolysis group, cDKA with rhabdomyolysis were at presentation significantly more likely to be hypotensive and have higher corrected sodium and calculated osmolality. Subsequently they were more likely to develop lower trough potassium levels during treatment. Five patients, all with rhabdomyolysis, needed dialysis: median duration 9 days (range 4–35). Three children in our cohort died, all from infection complications during treatment, one in AKI only group who did not receive dialysis and two in AKI with rhabdomyolysis on dialysis. Rhabdomyolysis was common among our cohort of cDKA with AKI and was associated with high morbidity and mortality. Rapid flux in electrolytes and osmolality may be important precipitating factors. Lemierre’s Syndrome (LS) is a rare syndrome most frequently due to an anaerobic organism, Fusobacterium Necrophorum. It is commonly a complication of an acute oropharyngeal infection, but there are exceptions to its presentations. In our case the cause of LS was otitis media caused by Streptococcus species. This is a rather unusual presentation of LS. LS is caused due to septic complications of oropharyngeal infections, which lead to thrombophlebitis of internal jugular vein leading to thrombosis formation. In this case report, we present a case of Lemierre’s syndrome in a seven-year-old male child. The patient presented with high grade fever spikes and earache, which were unresponsive to oral antibiotics. LS was diagnosed in this patient on the basis of clinical, microbiological and radiological findings. After the diagnosis, treatment involved using broad spectrum antibiotics and anticoagulants, followed by surgery. Though role of anticoagulants is controversial in LS, but there is no specific guideline contraindicating the use of anti-coagulants. In our case, timely diagnosis and management enabled us to discharge the patient without any symptoms. [ABSTRACT FROM AUTHOR]

Background: There is scarcity of data on impact of rituximab on anthropometrical parameters (weight, height and body mass index i.e. BMI SD score (SDS)) among children with steroid-dependent nephrotic syndromes (SDNS).Methods: Multicentre retrospective review.Results: 102 children with SDNS (male: 63%; n=64), median age 7 (IQR: 4.3-9.6) years, received a total of 217 rituximab infusions (total 110 cycles). At median follow-up of 2.1 (IQR: 1.3-2.8) years, 58 (57%) children were off steroids and a significant fall in steroid threshold for relapse was noted (median 0.6; IQR 0.4-0.9 to median 0.3; IQR 0.12 - 0.5 mg/kg/alternate day, p=0.005). Anthropometric parameters (BMI SDS: 0.92±1.8 to 0.25±1.47, p=0.003; weight SDS: 0.20±1.6 to -0.11±1.3, p=0.01; and height SDS: -0.93±1.88 to -0.45±1.54, p=0.04) as well as obesity (38% to 20%, p=0.003) and short stature (11% to 3%, p=0.02) improved. Results remained significant even when analysis was restricted to children ≤12 years (n=88), (BMI SDS: 0.97±1.98 to 0.25±1.5, p=0.001; weight SDS: 0.33±1.6 to 0.02±1.2, p=0.01; and height SDS: -0.67±1.84 to -0.186±1.42, p=0.001).Conclusions: Use of rituximab resulted in significant steroid sparing effect with an improvement in both growth and obesity parameters. Employees from nearly every sector of work are under the influence of stress that ultimately influences negatively on their job satisfaction. This study aims to see the impact of employees'stress on their job satisfaction in private banks working in Punjab particularly in district Multan. The study was conducted using a structured questionnaire. A total of 185 questionnaires were distributed from which 162 questionnaires were responded back. Reliability test was applied to check the reliability of the research instrument. Data analyses were done using SPSS version 17. Correlation analysis and regression analysiswere used as statistical techniques for data analysis. The impact of work environment, monetary rewards, workload, decision making authority, and management's behaviour on job satisfaction was checked. It was concluded that there is a strong negative relationship of work environment, monetary rewards, decision making authority, and management's behaviour with the job satisfaction. Abstract: Economic dispatch (ED) generally formulated as convex problem using optimization techniques by approximating generator input/output characteristic curves of monotonically increasing nature results in an inaccurate dispatch. The genetic algorithm has previously been used for the solution of problem for economic dispatch but takes longer time to converge to near optimal results. The hybrid approach is one of the methodologies used to fine tune the near optimal results produced by GA. This paper proposes new hybrid approach to solve the ED problem by using the valve-point effect. The approach we propose combines the genetic algorithm (GA) with active power optimization (APO) based on the Newton''s second order approach (NSO). The genetic algorithm acts as a global optimizer giving near optimal generation schedule, which becomes the input for generation buses in APO algorithm. This algorithm acting as local search technique dispatching the generated active power of units for minimization of cost and gives optimum generation schedule. Three machines 6-bus, IEEE 5-machines 14-bus, and IEEE 6-mchines 30-bus systems have been tested for validation of our approach. Results of the proposed scheme compared with results obtained from GA alone give significant improvements in the generation cost showing the promise of the proposed approach. [Copyright &y& Elsevier]
