Leaf and cambium phenologies are both important aspects of tree environmental adaptation in temperate areas. Temperate tree species with non-porous, diffuse-porous and ring-porous woods diverge substantially in the strategy of coping with freezing-induced hydraulic dysfunction, which can be closely associated with the timing of both leaf phenology and xylogenesis. Nevertheless, we still know little about the potential differences in the intra-annual process of xylogenesis among species of the three functional groups as well as its association with leaf phenology. Here, we monitored leaf phenology and xylogenesis in a non-porous (Pinus), a diffuse-porous (Populus), and a ring-porous (Ulmus) temperate tree species in a common garden. The results showed clear divergences in leaf and cambium phenologies and their chronological orders among the three species. The two hardwood species exhibited earlier bud burst and leaf unfolding than the conifer. The cambial activity of the ring-porous species began earlier than the diffuse-porous species, although the leaf phenology of the diffuse-porous species was earlier. The conifer species showed the latest bud break but the initiation of cambium activity was the earliest, which can be attributed to its strong resistance to freezing-induced embolism in the tracheid-based xylem. The leaf phenology preceded the onset of cambial activity in the Populus species, which was permitted by the ability of diffuse-porous species in largely retaining the stem hydraulic function over the winter. In contrast, the Ulmus species with ring-porous wood had to restore its severely hampered stem hydraulic function by winter embolism before leaf flush. The results revealed that leaf and cambium phenologies are closely interconnected due to the coordination between xylem water transport and leaf water demand. These findings contribute to a better understanding of the divergent adaptive strategies of temperate trees with different wood types.

Abstract Background In agricultural production, fungal diseases significantly impact the yield and quality of cotton (Gossypium spp.) with Verticillium wilt posing a particularly severe threat. Results This study is focused on investigating the effectiveness of endophytic microbial communities present in the seeds of disease-resistant cotton genotypes in the control of cotton Verticillium wilt. The technique of 16S ribosomal RNA (16S rRNA) amplicon sequencing identified a significant enrichment of the Bacillus genus in the resistant genotype Xinluzao 78, which differed from the endophytic bacterial community structure in the susceptible genotype Xinluzao 63. Specific enriched strains were isolated and screened from the seeds of Xinluzao 78 to further explore the biological functions of seed endophytes. A synthetic microbial community (SynCom) was constructed using the broken-rod model, and seeds of the susceptible genotype Xinluzao 63 in this community that had been soaked with the SynCom were found to significantly control the occurrence of Verticillium wilt and regulate the growth of cotton plants. Antibiotic screening techniques were used to preliminarily identify the colonization of strains in the community. These techniques revealed that the strains can colonize plant tissues and occupy ecological niches in cotton tissues through a priority effect, which prevents infection by pathogens. Conclusion This study highlights the key role of seed endophytes in driving plant disease defense and provides a theoretical basis for the future application of SynComs in agriculture.

Hydraulic functionality is crucial for tree productivity and stress tolerance. According to the theory of the fast–slow economics spectrum, the adaptive strategies of different tree species diverge along a spectrum defined by coordination and trade-offs of a suite of functional traits. The fast- and slow-growing species are expected to differ in hydraulic efficiency and safety; however, there is still a lack of investigation on the mechanistic association between tree growth rate and tree hydraulic functionality. Here, in a common garden condition, we measured radial growth rate and hydraulic traits in a fast-growing (Populus alba L. × P. berolinensis Dippel) and a slow-growing tree species (Acer truncatum Bunge), which are both important tree species for afforestation in northern China. In line with the contrasts in radial growth rate and wood anatomical traits at both the tissue and pit levels between the two species, stem hydraulic conductivity of the Populus species was significantly higher than that of the Acer species, but the resistance to drought-induced xylem cavitation was the opposite. A trade-off between hydraulic efficiency and safety was observed across the sampled trees of the two species. Higher water-transport efficiency supports the greater leaf net photosynthetic carbon assimilation capacity of the Populus species and hence facilitates fast growth, while the conservative hydraulic traits of the Acer species result in a slower growth rate but enhanced drought tolerance.

Extreme droughts are anticipated to have detrimental impacts on forest ecosystems, especially in water-limited regions, due to the influence of climate change. However, considerable uncertainty remains regarding the patterns in species-specific responses to extreme droughts. Here, we conducted a study integrating dendrochronology and remote sensing methods to investigate the mosaic-distributed maple-oak (native) natural forests and poplar plantations (introduced) in the Horqin Sandy Land, Northeast China. We assessed the impacts of extreme droughts on tree performances by measuring interannual variations in radial growth and vegetation index. The results showed that precipitation and self-calibrated palmer drought severity index (scPDSI) are the major factors influencing tree-ring width index (RWI) and normalized difference vegetation index (NDVI). The severe droughts between 2000 and 2004 resulted in reduced RWI in the three studied tree species as well as led to NDVI reductions in both the maple-oak natural forests and the poplar plantations. The RWI reached the nadir during the 2000–2004 severe droughts and remained at low levels two years after the severe drought, creating a legacy effect. In contrast to the lack of significant correlation between RWI and scPDSI, NDVI exhibited a significant positive correlation with scPDSI indicating the greater sensitivity of canopy performance to droughts than radial growth. Furthermore, interspecific differences in RWI and NDVI responses were observed, with the fast-growing poplar species experiencing a more significant RWI decrease and more negative NDVI anomaly during severe droughts than native species, highlighting the species-specific trade-offs between drought resilience and growth rate. This study emphasizes the importance of combining tree-level radial growth with landscape-scale canopy remote sensing to understand forest resilience and response. Our study improves our understanding of forest responses to extreme drought and highlights species differences in climate responses, offering crucial insights for optimizing species selection in sustainable afforestation and forest management in water-limited regions under the influence of climate change.

Tomato (Solanumlycopersicum) is a model plant for research on fruit development and stress response, in which gene expression analysis is frequently conducted. Quantitative PCR (qPCR) is a widely used technique for gene expression analysis, and the selection of reference genes may affect the accuracy of results and even conclusions. Although there have been some frequently used reference genes in tomato, it has been shown that the expressions of some of these genes are not constant in different tissues and environmental conditions. Moreover, little information on genomic identification of reference genes is available in tomato. Here, we mined the publicly available transcriptional sequencing data and screened out fifteen candidate reference genes, and the expression stability of these candidate genes and seven traditionally used ones were evaluated under stress and hormone treatment. The results showed that over half of the selected candidate references were housekeeping genes in tomato cells. Among the candidate reference genes and the traditionally used ones, the most stably expressed genes varied under different treatments, and most of these genes were recommended as preferred reference genes at least once except Solyc04g009030 and Solyc07g066610, two traditionally used reference genes. This study provides some novel reference genes in tomato, and the preferred reference genes under different environmental stimuli, which may be useful for future research. Our study suggests that excavating stably expressed genes from transcriptome sequencing data is a reliable approach to screening reference genes for qPCR analysis.

Fructose-1,6-biphosphate aldolase (FBA) is a multifunctional enzyme in plants, which participates in the process of Calvin-Benson cycle, glycolysis and gluconeogenesis. Despite the importance of FBA genes in regulating plant growth, development and abiotic stress responses, little is known about their roles in cotton. In the present study, we performed a genome-wide identification and characterization of FBAs in Gossypium hirsutum. Totally seventeen GhFBA genes were identified. According to the analysis of functional domain, phylogenetic relationship, and gene structure, GhFBA genes were classified into two subgroups. Furthermore, nine GhFBAs were predicted to be in chloroplast and eight were located in cytoplasm. Moreover, the promoter prediction showed a variety of abiotic stresses and phytohormone related cis-acting elements exist in the 2k up-stream region of GhFBA. And the evolutionary characteristics of cotton FBA genes were clearly presented by synteny analysis. Moreover, the results of transcriptome and qRT-PCR analysis showed that the expression of GhFBAs were related to the tissue distribution, and further analysis suggested that GhFBAs could respond to various abiotic stress and phytohormonal treatments. Overall, our systematic analysis of GhFBA genes would not only provide a basis for the understanding of the evolution of GhFBAs, but also found a foundation for the further function analysis of GhFBAs to improve cotton yield and environmental adaptability.

Cordyceps militaris is widely cultivated as a functional food and traditional medicine in China and Southeast Asia. In this study, C. militaris hybrid strains were isolated from different parts of hybrid fruiting body cultivated through mating-based sexual reproduction of ascospores. We investigated the effect of strains, solid-state substrates and light condition on production of yield and contents of bioactive compounds (cordycepin and adenosine) using HPLC in fruiting bodies. The results indicated that yield and production of bioactive compounds exist distinct differences on strain separated part, solid-substrates and light conditions. The middle strain obtained the highest yield on wheat substrate. The yield was higher on wheat substrate, the concentration of cordycepin and adenosine in fruiting bodies were higher on rice substrate. The red light is beneficial to high yield, and white light is conducive to the accumulation of cordycepin in fruiting bodies, the light condition has little effect on the adenosine content.

The far infrared spectrum of gaseous 2-fluoropropenoyl fluoride CH2CFCFO, has been recorded at a resolution of 0.10 cm-1 in the 350–35 cm-1 region. The asymmetric torsional fundamental of the more stable s-trans (two double bonds oriented trans to one another) and the high energy s-cis conformations have been observed at 84.1 and 68.5 cm-1, respectively, each with excited states falling to lower frequencies. From these data, the asymmetric torsional potential function governing internal rotation about the C–C bond has been determined. The potential coefficients are V1=-201±4, V2=2106±16, V3=382±6, V4=-41±5, and V5=-74±3 cm-1. The s-trans to s-cis and s-cis to s-trans barriers have been determined to be 2232 and 2124 cm-1, respectively, with an enthalpy difference between the conformations of 108±26 cm-1 (309±74 cal/mol). From variable temperature studies of the Raman spectrum, the conformational enthalpy difference has been determined to be 187±39 cm-1 (535±112 cal/mol) and 370±82 cm-1 (1058±234 cal/mol) for the gas and liquid, respectively. A complete assignment of the vibrational fundamentals observed from the infrared (3500–50 cm-1) spectra of the gas and solid and the Raman (3200–10 cm-1) spectra of all three physical states is proposed. All of these data are compared to the corresponding quantities obtained from ab initio Hartree–Fock gradient calculations employing both the 3-21G and 6-31G* basis sets. Additionally, complete equilibrium geometries have been determined for both rotamers. The results are discussed and compared with the corresponding quantities obtained for some similar molecules. Elevated tropospheric ozone (O3) concentration has been shown to affect many aspects of plant performance including detrimental effects on leaf photosynthesis and plant growth. However, it is not known whether such changes are accompanied by concomitant responses in plant hydraulic architecture and water relations, which would have great implications for plant growth and survival in face of unfavorable water conditions. A soybean (Glycine max (L.) Merr.) cultivar commonly used in Northeast China was exposed to non‐filtered air (NF, averaged 24.0 nl l−1) and elevated O3 concentrations (eO3, 40 nl l−1 supplied with NF air) in six open‐top chambers for 50 days. The eO3 treatment resulted in a significant decrease in whole‐plant hydraulic conductance that is mainly attributable to the reduced hydraulic conductance of the root system and the leaflets, while stem and leaf petiole hydraulic conductance showed no significant response to eO3. Stomatal conductance of plants grown under eO3 was lower during mid‐morning but significantly higher at midday, which resulted in substantially more negative daily minimum water potentials. Moreover, excised leaves from the eO3 treated plants showed significantly higher rates of water loss, suggesting a lower ability to withhold water when water supply is impeded. Our results indicate that, besides the direct detrimental effects of eO3 on photosynthetic carbon assimilation, its influences on hydraulic architecture and water relations may also negatively affect O3‐sensitive crops by deteriorating the detrimental effects of unfavorable water conditions. Mistletoes absorb water from the vascular system of their hosts and thus the water use of mistletoes can be influenced by the water status of their hosts besides abiotic environmental conditions; however, there is a lack of studies on the dynamics of mistletoe water utilization in relation to both types of controlling factors. By building a canopy platform at 20 m above the ground, we monitored the dynamic changes of sap flow of Viscum coloratum (Kom.) Nakai (Loranthaceae) in combination with continuous measurements of microclimatic variables and volumetric water content (VWC) of its host tree branch xylem. We found that the host tree VWC exhibited substantial fluctuations during sunny days but lower VWC of the host did not negatively affect the sap flow of V. coloratum. Hourly and daily mean transpiration rates (Esap) of V. coloratum calculated from sap flow measurements showed strong positive correlations with photosynthetic photon flux density (PPFD) and vapor pressure deficit (VPD) measured in close vicinity to the point of mistletoe attachment. The mean Esap of V. coloratum was substantially higher than that of their host during clear days (4.55 ± 0.54 vs 2.01 ± 0.15 kgm-2 day-1). Moreover, the mistletoe-to-host transpiration ratio was not constant but became increasingly larger with the increase of PPFD or VPD on both hourly and daily bases, suggesting a weaker control of water loss in the mistletoe in comparison to its host species. The strong dependence of mistletoe Esap on micrometeorological variables and its decoupling from the host tree xylem water status suggests that the development of dense tree canopy functions as a potential mechanism for the host trees in reducing the competitive water use of mistletoes. Nitrogen (N) deposition is expected to have great impact on forest ecosystems by affecting many aspects of plant-environmental interactions, one of which involves its influences on plant water relations through modifications of plant hydraulic architecture. However, there is a surprising lack of integrative study on tree hydraulic architecture responses to N deposition, especially at the whole-plant level. In the present study, we used a 5-year N addition experiment to simulate the effects of six different levels of N deposition (20-120 kg ha-1 year-1) on growth and whole-plant hydraulic conductance of a dominant tree species (Fraxinus mandshurica Rupr.) from the typical temperate forest of NE China. The results showed that alleviation of N limitation by moderate concentrations of fertilization (20-80 kg ha-1 year-1) promoted plant growth, but further N additions on top of the threshold level showed negative effects on plant growth. Growth responses of F. mandshurica seedlings to N addition of different concentrations were accompanied by corresponding changes in whole-plant hydraulic conductance; higher growth rate was accompanied by reduced whole-plant hydraulic conductance (Kplant) and higher leaf water-use efficiency. A detailed analysis on hydraulic conductance of different components of the whole-plant water transport pathway revealed that changes in root and leaf hydraulic conductance, rather than that of the stem, were responsible for Kplant responses to N fertilization. Both plant growth and hydraulic architecture responses to increasing levels of N addition were not linear, i.e., the correlation between measured parameters and N availability exhibited bell-shaped curves with peak values observed at medium levels of N fertilization. Changes in hydraulic architecture in response to fertilization found in the present study may represent an important underlying mechanism for the commonly observed changes in water-related tree performances in response to N deposition. The brown planthopper (BPH), Nilaparvata lugens , is a destructive insect pest of rice throughout Asia. Different from brown-eye color wild type, BPH also has red-eye color mutation phenotype. As a visible genetic marker, the red-eye mutant in BPH is a valuable material. To reveal the eye color mutation mechanism, a karmoisin homologue gene (named as Nlka ) was cloned from BPH. And karmoisin is always deemd as a xanthommatin-related gene in other insects, encoding phenoxazinone synthetase (PHS). Nlka is consisted of 7 exons and encodes a protein with 502 amino acids (NlKA). NlKA showed high amino acid identities with its insect homologues (48.8%–51.8%). Nlka transcripts can be detected at all the developmental stages and in all tissues tested, including egg, nymph, adult, body wall, ovary, fat body, midgut and Malpighian tubule. However, no constant In/Del or non-synonymous mutation was observed between the mutant and the wild type strains. Quantitative real-time PCR experiment also showed that Nlka transcript level had no significant differences between them. These results indicated that Nlka is not the target gene causing the red-eye color mutation phenotype of BPH. Through the second structure and motif analysis, the present study also showed that all the proteins deduced from the karmoisin genes in insects may be members of monocarboxylate transporters (MCTs) rather than PHSs. The epiphytic growth habit in many Ficus species during their juvenile stages has commonly been hypothesized to be an adaptation for avoiding deep shade in the forest understory, but this has never been tested experimentally. We examined growth and ecophysiology in seedlings of three hemiepiphytic (Hs) and three non-hemiepiphytic ( NHs) Ficus species grown under different irradiance levels. Both Hs and NHs exhibited characteristics of high light requiring species, such as high plasticity to growth irradiance and relatively high maximum photosynthetic assimilation rates. Diurnal measurements of leaf gas exchange showed that Hs have much shorter active photosynthetic periods than NHs; moreover, leaves of Hs have lower xylem hydraulic conductivity but stronger drought tolerance as indicated by much lower rates of leaf diebacks during the drought treatment. Seedlings of NHs had 3.3- and 13.3-fold greater height and biomass than those of Hs species after growing in the nursery for 5 months, indicating a trade-off between growth and drought tolerance due to the conflicting requirements for xylem conductivity and cavitation resistance. This study does not support the shade-avoidance hypothesis; rather, it suggests that the canopy regeneration in Hs is an adaptation to avoid alternative terrestrial growth-related risks imposed to tiny Ficus seedlings. The NHs with terrestrial regeneration reduce these risks by having an initial burst of growth to rapidly gain relatively large seedling sizes, while in Hs seedlings more conservative water use and greater drought tolerance for surviving the canopy environment are intrinsically associated with slow growth. Leaf structural and physiological traits are associated with growth form and habitat, but little is known of the specific traits associated with hemiepiphytes, which are an important component of many tropical forests. Given their life history that includes a drought prone epiphytic stage, hemiepiphytes should be expected to have more drought tolerance-related traits than co-occurring terrestrial species. 2. The genus Ficus includes woody hemiepiphytes distributed in tropical areas throughout the world. Traits related to the flux of water through the leaf and to drought adaptations were studied in five hemiephiphytic (H) and five non-hemiepiphytic (NH) Ficus tree species grown in a common garden to determine genetically based differences. 3. Leaves of H and NH species differed substantially in structure and physiology; on average, H species had smaller leaves with higher leaf mass per unit area, thicker epidermis, smaller vessel lumen diameters in petioles and lower petiole hydraulic conductivity. Leaf traits also indicated stronger drought tolerance in H species, including lower epidermal conductance and turgor loss point and earlier stomatal closure with desiccation than NH species. Across H and NH species, traits related to water flux capacity were negatively correlated with traits related to drought tolerance. 4. The divergences in hydraulics and water relations between growth forms for these closely related species reflected specialization according to contrasting habitat and life form. Conservative water use and increased ability of leaves to persist under severe drought would provide an advantage for H species, especially during the epiphytic phase, while the higher potential water use of NH species would be associated with higher assimilation rates and competitiveness under high water supply. 5. The results indicate a trade-off between leaf water flux capacity and leaf drought tolerance across these hemiephiphytic and non-hemiepiphytic species. To examine the effects of dietary lipids on the growth and feed utilization of jade perch juveniles, Scortum barcoo, diets containing 36.3% crude protein supplemented with increasing lipid levels (6, 9, 12, and 15% of the dry matter) were used to feed triplicate groups of 30 fish for 60 d. At the end of the experiment, more than 95% fish survived well from all diet groups ( P > 0.05). Measurements on the weight gains and the daily specific growth rates indicated that fish fed with diets of 12 and 15% lipids exhibited higher growth rates ( P < 0.05); evaluations for the feed conversion ratio and the protein efficiency ratio indicated that fish fed with 12 and 15% lipid diets used their feed and dietary proteins more efficiently ( P < 0.05). The muscle lipid and dry matter contents increased dramatically in fish fed with higher dietary lipid levels ( P < 0.05). The highest lipid contents were obtained from fish in the 15% lipid diet group and the highest amount of dry matters from the 12% lipid diet group. On the other hand, protein contents in fish muscles declined with increasing dietary lipid levels ( P < 0.05), and the lowest values were shown in the 15% lipid diet group. Ash contents showed no significant differences from muscles of fish fed with four different diets ( P > 0.05). Unusual stress behavior was observed in W-incorporated hydrogenated amorphous carbon films prepared by a hybrid process composed of ion-beam deposition and magnetron sputtering. As the tungsten concentration increased from 0 to 2.8 at.%, the residual compressive stress decreased by 50%, without significant deterioration in the mechanical properties. This was followed by a rapid increase and a gradual decrease in the residual stress with increasing W concentration. High-resolution transmission electron microscopy analysis and first-principle calculations show that the reduced directionality of the W–C bonds in the W-incorporated amorphous carbon matrix relaxes the stress caused by the distorted bonds.
