Benzodiazepines are frequently encountered in crime scenes, often mixed with adulterants and diluents, complicating their analysis. This study investigates the interactions between two benzodiazepines, lorazepam (LOR) and alprazolam (ALP), with common adulterants/diluents (paracetamol, caffeine, glucose, and lactose) using infrared (IR) spectroscopy and quantum chemical methods. The crystallographic structures of LOR and ALP were optimized using several functionals (B3LYP, B3LYP-D3BJ, B3PW91, CAM-B3LYP, M05-2X, and M06-2X) combined with the 6-311++G(d,p) basis set. M05-2X was the most accurate when comparing experimental and theoretical bond lengths and angles. Vibrational and 13C NMR spectra were calculated to validate the functional's applicability. The differences between LOR's experimental and theoretical IR spectra were attributed to intramolecular interactions between LOR monomers, examined through density functional theory (DFT) optimization and quantum theory of atoms in molecules (QTAIM) analysis. Molecular dynamics simulations modeled benzodiazepine–adulterant/diluent systems, predicting the most stable structures, which were further analyzed using QTAIM. The strongest interactions and their effects on IR spectra were identified. Comparisons between experimental and theoretical spectra confirmed spectral changes due to interactions. This study demonstrates the potential of quantum chemical methods in analyzing complex mixtures, elucidating spectral changes, and assessing the structural stability of benzodiazepines in forensic samples. Milk is an essential part of the human diet and is a nutrient‐rich food that improves nutrition and food security. The aim of this study was to determine the presence and concentration of aflatoxin M1 (AFM1), adulterants, microbial loads, and physicochemical properties of raw cow's milk (CM) in Nekemte City, Ethiopia. A total of 12 samples of fresh CM were purposefully collected from four kebeles in the city (Bake Jama, Burka Jato, Cheleleki, and Bakanisa Kese) based on the potential of each milk production and distributor site. The AFM1 concentration was determined by high‐performance liquid chromatography (HPLC) with a Sigma‐Aldrich standard (St. Louis, MO, USA). The concentrations of AFM1 in Bake Jama, Burka Jato, Cheleleki, and Bakanisa Kese were found to be 0.01–0.03 g/L, 0.31–0.35 g/L, 0.19–0.21 g/L, and 0.04–0.07 g/L, respectively. The concentrations of AFM1 in the present study varied significantly (p < 0.05) and ranged from 0.01 g/L to 0.35 g/L. These results show that of the 12 samples tested, all were positive for AFM1 and contaminated to varying degrees. The results of this study also revealed that the concentration of AFM1 in 7 (58%) of the 12 milk samples was above the European Union's (EU) maximum tolerance limit (0.05 g/L). The present study also revealed that of the investigated adulterants, only the addition of water had positive effects on three milk samples, while the remaining adulterants were not detected in any of the milk samples. The total bacterial count (TBC) and total coliform count (TCC) were significantly (p < 0.05) different and ranged from 5.53 to 6.82 log10cfumL−1 and from 4.21 to 4.74 log10cfumL−1, respectively. The physicochemical properties of the milk samples in the present study were significantly (p < 0.05) different and ranged from 2.8% to 5.75% fat, 7.03% to 9.75% solid‐not‐fat (SNF), 2.35% to 3.61% protein, 3.33% to 5.15% lactose, 11.54% to 13.69% total solid, 0.16% to 0.18% titratable acid, 26.7 to 32.1°C, 6.35 to 6.55 pH, and 1.027 to 1.030 specific gravity. The physicochemical parameters of the raw milk in the study area met the required quality standards. Hence, further studies are required to determine the extent of the problem and the factors associated with high levels of AFM1 in raw milk in the study areas, including the detection of aflatoxin B1 (AFB1) in animal feed. This manuscript proposes a new extraction method for analyzing natural compounds in herbal supplements for weight loss and determining adulterants by a comprehensive two-dimensional gas chromatography system coupled to a quadrupole time-of-flight mass spectrometer (GCxGC/Q-TOFMS). In this new approach, a hydrophilic microporous cartridge (HMCart) was developed to enclose the samples, protecting the SPME (solid-phase microextraction) fibers. The cartridge is loaded with herbal supplement samples during direct immersion of SPME fiber in the aqueous phase to allow the analytes to be trapped. The new sample preparation method using chromatographic analyses enabled the detection of 650 compounds, including natural compounds, such as terpenes, aldehydes, ketones, and fatty acids. Adulterants found in other studies in the literature, such as sibutramine and ephedrine. The fluoxetine and caffeine were detected and quantified. The new method has achieved the goal of simple, effective, solvent-free, and versatile sample preparation that can be applied in future studies, including determining other compound classes in various matrix samples. Milk has been an indispensable part of our diet because of its rich content of vital nutrients. The demand and supply gap in milk production along with its perishable nature have resulted in the increased menace of milk adulteration. Various adulterants such as detergents, urea, ammonium salts, sodium chloride, nitrates, sulphates, salicylic acid, benzoic acid, hydrogen peroxide, formaldehyde, boric acid, neutralisers and melamine present in the milk give rise to serious health problems such as nausea, gastroenteritis, hepatitis, renal failure, heart problems, asthma, pneumonia, cancer and allergic reactions. This paper describes the adverse health effects of consuming adulterated milk.

Detection of food quality by identifying different adulterants in food products is a grave requirement for the food industry. Various methods of food analysis have been evolved to identify the quality and quantity of impurities but those methods are time-consuming, expensive and laborious. Our research aims at reducing the challenges experienced in these existing methods. Our procedure is to detect food adulterants using a microwave signal and observe the change in reflection coefficient and resonant frequency of the reflected microwave signal. The property of modification of reflection coefficient and resonant frequency of reflected microwave signal from pure rice grains in the presence of adulterants like small stones/pebbles and broken rice is exploited. In this work, the detection and proportion of adulterants, such as stones and low-quality broken rice in fresh rice in varying proportions, have been determined. The present study investigates the chemical composition variances among Pinelliae Rhizoma, a widely used Chinese herbal medicine, and its common adulterants including Typhonium flagelliforme, Arisaema erubescens, and Pinellia pedatisecta. Utilizing the non-targeted metabolomics technique of employing UHPLC-Q-Orbitrap HRMS, this research aims to comprehensively delineate the metabolic profiles of Pinelliae Rhizoma and its adulterants. Multivariate statistical methods including PCA and OPLS-DA are employed for the identification of differential metabolites. Volcano plot analysis is utilized to discern upregulated and downregulated compounds. KEGG pathway analysis is conducted to elucidate the differences in metabolic pathways associated with these compounds, and significant pathway enrichment analysis is performed. A total of 769 compounds are identified through metabolomics analysis, with alkaloids being predominant, followed by lipids and lipid molecules. Significant differential metabolites were screened out based on VIP > 1 and p-value < 0.05 criteria, followed by KEGG enrichment analysis of these differential metabolites. Differential metabolites between Pinelliae Rhizoma and Typhonium flagelliforme, as well as between Pinelliae Rhizoma and Pinellia pedatisecta, are significantly enriched in the biosynthesis of amino acids and protein digestion and absorption pathways. Differential metabolites between Pinelliae Rhizoma and Arisaema erubescens are mainly enriched in tyrosine metabolism and phenylalanine metabolism pathways. These findings aim to provide valuable data support and theoretical references for further research on the pharmacological substances, resource development and utilization, and quality control of Pinelliae Rhizoma. Introduction: Ecstasy (3,4‐methylenedioxymethamphetamine [MDMA]) is a drug commonly used by people who attend electronic dance music (EDM) events at nightclubs and dance festivals. Drug checking has gained popularity in recent years to test for adulterants, but epidemiology studies are needed to estimate potential shifts in prevalence of drug checking to further inform harm reduction efforts. Methods: Adults entering randomly selected EDM events in New York City were surveyed in 2017 and 2022. Those reporting past‐year ecstasy use were asked if they tested their ecstasy in the past year using a drug testing kit and whether they found out or suspected their ecstasy contained other drugs. We compared estimates between 2017 and 2022. Results: In 2017, an estimated 23.1% had tested their ecstasy, and this estimate increased to 43.1% in 2022 (86.6% increase, p = 0.006). Among those who tested their ecstasy, in 2017, 31.2% always tested their drug, and this increased to 60.6% in 2022 (94.2% increase, p = 0.026). In 2017, 59.6% of those who tested their ecstasy reported finding out or suspecting their drug was adulterated, which decreased to 18.4% in 2022 (69.1% decrease, p < 0.001). Suspected methamphetamine adulteration in particular decreased, from 21.9% in 2017 to 3.6% in 2022 (83.6% decrease, p = 0.007). Discussion and Conclusions: The use of drug testing kits has increased among EDM event attendees who use ecstasy and, at the same time, among those who had tested their ecstasy, suspected adulteration has decreased. Sorbitol has been the new and emerging adulterant in dairy industry. The main aim of the study was to develop a method to detect sorbitol in milk, which is not affected by other sugars, polyols and formalin. Hence, a thin layer chromatographic (TLC) method was standardized to detect the sorbitol in milk. In the study 90 s duration for the impregnation of Silica gel 60F TLC plates with Cu- ions was found suitable to resolve sorbitol as a distinct spot. The standardized conditions were (1) developing solvent system consisting of n-propanol: ethyl acetate: water (7:1:2), (2) 0.5% of potassium permanganate in 0.1 M NaOH as color developing reagent. (3) Drying temperature (65°C/ 10 min.) after spraying the color developing reagent. The limit of detection was 0.2% of added sorbitol in milk. The standardized method could also detect the sorbitol in the presence of sucrose, glucose and polyols like mannitol and maltitol. In both cow and buffalo milk samples the standardized methodology performed well in detection of sorbitol. The method also performed well in sorbitol spiked formalin preserved milk samples. This method can be an alternative to the other methods involving costly equipment in detecting adulteration of milk with sorbitol. [ABSTRACT FROM AUTHOR]

Abstract Introduction Unregulated supply of fentanyl and adulterants continues to drive the overdose crisis. Mobile Overdose Response Services (MORS) are novel technologies that offer virtual supervised consumption to minimize the risk of fatal overdose for those who are unable to access other forms of harm reduction. However, as newly implemented services, they are also faced with numerous limitations. The aim of this study was to examine the facilitators and barriers to the adoption of MORS in Canada. Methods A total of 64 semi-structured interviews were conducted between November 2021 and April 2022. Participants consisted of people who use substances (PWUS), family members of PWUS, health care professionals, harm reduction workers, MORS operators, and members of the general public. Inductive thematic analysis was used to identify the major themes and subthemes. Results Respondents revealed that MORS facilitated a safe, anonymous, and nonjudgmental environment for PWUS to seek harm reduction and other necessary support. It also created a new sense of purpose for operators to positively contribute to the community. Further advertising and promotional efforts were deemed important to increase its awareness. However, barriers to MORS implementation included concerns regarding privacy/confidentiality, uncertainty of funding, and compassion fatigue among the operators. Conclusion Although MORS were generally viewed as a useful addition to the currently existing harm reduction services, it’s important to monitor and tackle these barriers by engaging the perspectives of key interest groups.

The analysis of heroin samples, before use in the protected environment of user centra, could be a supplementary service in the context of harm reduction. Infrared spectroscopy hyphenated with multivariate calibration could be a valuable asset in this context, and therefore 125 heroin samples were collected directly from users and analysed with classical chromatographic techniques. Further, Mid-Infrared spectra were collected for all samples, to be used in Partial Least Squares (PLS) modelling, in order to obtain qualitative and quantitative models based on real live samples. The approach showed that it was possible to identify and quantify heroin in the samples based on the collected spectral data and PLS modelling. These models were able to identify heroin correctly for 96% of the samples of the external test set with precision, specificity and sensitivity values of 100.0, 75.0 and 95.5%, respectively. For regression, a root mean squared error of prediction (RMSEP) of 0.04 was obtained, pointing at good predictive properties. Furthermore, during mass spectrometric screening, 10 different adulterants and impurities were encountered. Using the spectral data to model the presence of each of these resulted in performant models for seven of them. All models showed promising correct-classification rates (between 92 and 96%) and good values for sensitivity, specificity and precision. For codeine and morphine, the models were not satisfactory, probably due to the low concentration of these impurities as a consequence of acetylation. For methacetin, the approach failed. The aim of this study was to identify drug composition in an electronic music outdoor festival nearby Mexico City. Methods The participants completed a questionnaire with demographic data, harm reduction strategies, drug-use patterns, history, and the drug they expected to find. We took a small sample of each substance and prepared it for drug checking. A two-section drug testing station was placed within the grounds of the festival. Interaction with participants occurred at the front part. Drug checking was conducted at the rear part. The service was free of charge, voluntary and confidential. Forty persons aged 22 to 48 years participated (mode = 28), of which 92.5% were male, most (82.5%) were single. Through the Substance Analysis Program of “ReverdeSer Collective,” we conducted the testing with the attendants that provided 51 drug samples, following ethical and biosafety protocols. We used colorimetry, Fourier Transform Infrared Spectroscopy, and fentanyl immunoassay strips for sample analysis. Results Substances of choice among attendants were psychostimulants (MDMA and other amphetamine-like drugs) and hallucinogens. Most samples contained what the users expected plus adulterants. Main adulterants were methylene-dioxy-ethyl-amphetamine, methylene-dioxy-propyl-amphetamine, hydroxyamphetamine, and the selective serotonin reuptake inhibitor venlafaxine. Fentanyl was present in 2 out of 4 cocaine samples and in 14 of the 22 confirmed MDMA samples. Conclusions Some of the adulterants found pose serious health risks, especially fentanyl, amphetamine-like substances, and venlafaxine. Therefore, it is urgent to monitor these adulterants at electronic music festivals and to implement prevention, treatment, and harm reduction public policies. Naloxone distribution and drug-assisted therapies should be part of government programs in Mexico.

Nanocomposites are emerging as a new generation of materials that can be used to combat water pollution. Zeolite-based nanocomposites consisting of combinations of metals, metal oxides, carbon materials, and polymers are particularly effective for separating and adsorbing multiple contaminants from water. This review presents the potential of zeolite-based nanocomposites for eliminating a range of toxic organic and inorganic substances, dyes, heavy metals, microplastics, and ammonia from water. The review emphasizes that nanocomposites offer enhanced mechanical, catalytic, adsorptive, and porosity properties necessary for sustainable water purification techniques compared to individual composite materials. The adsorption potential of several zeolite-metal/metal oxide/polymer-based composites for heavy metals, anionic/cationic dyes, microplastics, ammonia, and other organic contaminants ranges between approximately 81 and over 99%. However, zeolite substrates or zeolite-amended soil have limited benefits for hyperaccumulators, which have been utilized for phytoremediation. Further research is needed to evaluate the potential of zeolite-based composites for phytoremediation. Cosmetics still have a glitzy attraction, but the public is increasingly concerned about their toxicity. Since 2500 BC, one way of enhancing the beauty of lips has been using lipstick compositions. Lip care cosmetic product usually contains emollients, waxes, texturing agents, preservatives, and oils. Women apply lipstick two to several times per day, according to the researchers. In terms of substance exposure, that translates as consuming or holding onto approximately 87 mg of lipstick every day. It has been shown to contain trace amounts of heavy metals such as Cd, Cr, Cu, Co, Ni and Pb. Cosmetics either contain trace levels of these harmful metals on purpose or as impurities in the raw materials. Long-term exposure to these chemicals can pose serious risks to human health, most notably skin cancer. Mineral pigments, which are frequently used to make lipstick, cause products to get contaminated with heavy metals. So, there is necessity for stringent regulation of cosmetics in a harmonized way. It is crucial to regulate this business to safeguard consumers and society. Abstract Monteverdia ilicifolia (Mart. ex Reissek) Biral (basionym: Maytenus ilicifolia Mart. ex Reissek) (Celastraceae) is popularly known as holy thorn or “espinheira-santa” in Brazil. This herb is traditionally used for gastric and digestive problems. However, similar species are often used as adulterants. Considering the pharmacological interest and the existence of adulterants, this study was devoted to characterizing M. ilicifolia samples by Fourier-transform infrared (FTIR) spectroscopy coupled with principal component analysis (PCA) and photoacoustic spectroscopy (PAS) by using Citronella gongonha (Mart.) R. A. Howard as adulterant sample. Six commercial samples were purchased, and seven collected samples were obtained in the Campos Gerais region. The leaves were then dehydrated and converted to powder. The FTIR spectroscopic assay was carried out and consisted of ten different measurements for each sample with a time interval of seven days between them. The PCA was performed from these spectra. The characterization by FTIR demonstrated the presence of tannins and flavonoids in the studied samples. FTIR spectroscopy coupled with PCA was able to discriminate the commercial and the collected samples of M. ilicifolia using three principal components. The photoacoustic spectroscopy resulted in absorption bands centered at ~280, ~380, ~480, and ~630 nm. These bands presented a higher spectral resolution for the collected samples. The commercial samples showed broadening bands, which allowed the differentiation between these two sets of samples. FTIR spectroscopy coupled with PCA, and photoacoustic spectroscopy are alternative tools for the differentiation of M. ilicifolia samples.

Background: Opioid overdose deaths continue to present major public health challenges in the U.S. Harm reduction agencies have begun using drug checking technologies to identify adulterants in the local drug supply and reduce overdose risk among people who use drugs (PWUD). Through qualitative and ethnographic methods, we assess the use of portable mass spectrometers at a harm reduction agency in a Northeastern U.S. city. Methods: We conducted participant observation, and on-the-spot qualitative interviews with harm-reduction staff members (n = 10) and their clientele (n = 17) between May 2019 and December 2020. Interviews explored emic views on drug checking (process, logistics, technology), and perceived benefits and challenges. We used thematic content analysis techniques to code and analyze interview transcriptions. Results: Implementation and use of drug checking devices were not free of challenges and malfunctions, often delaying drug checking opportunities and increasing suspicions and distrust among clients. Yet, staff members perceived that when working properly, or in conjunction with an additional device, they offered information about purchased drugs that could empower clients and potentially lead to positive behavior change. Use of these devices also enhanced engagement between harm reduction staff and PWUD, facilitating meaningful conversations around self-advocacy and harm reduction engagement. Conclusion: We report qualitative findings on the experiences and perceptions of drug checking devices among harm reduction staff and PWUD. Our findings indicate that use of this technology has the potential to decrease risk behaviors, expand health promotion services, and help reduce high rates of fentanyl-related overdose. There is a necessity to protect the quality and authenticity of herbs and spices because of the increase in the fraud and adulteration incidence during the last 30 years. There are several aspects that make herbs and spices quite vulnerable to fraud and adulteration, including their positive and desirable sensorial and health-related properties, the form in which they are sold, which is mostly powdered, and their economic relevance around the world, even in developing countries. For these reasons, sensitive, rapid, and reliable techniques are needed to verify the authenticity of these agri-food products and implement effective adulteration prevention measures. This review highlights why spices and herbs are highly valued ingredients, their economic importance, and the official quality schemes to protect their quality and authenticity. In addition to this, the type of frauds that can take place with spices and herbs have been disclosed, and the fraud incidence and an overview of scientific articles related to fraud and adulteration based on the Rapid Alert System Feed and Food (RASFF) and the Web of Science databases, respectively, during the last 30 years, is carried out here. Next, the methods used to detect adulterants in spices and herbs are reviewed, with DNA-based techniques and mainly spectroscopy and image analysis methods being the most recommended. Urea is naturally present in milk, yet urea is added intentionally to increase milk's nitrogen content and shelf life. In this study, a total of 50 Ultra heat treatment (UHT) milk samples were spiked with known urea concentrations (0–5 w/v%). Attenuated total reflectance–Fourier transform infrared (ATR-FTIR) spectroscopy with principal component analysis (PCA), discriminant analysis (DA), and multiple linear regression (MLR) were used for the discrimination and quantification of urea. The PCA was built using 387 variables with higher FL > 0.75 from the first PCA with cumulative variability (90.036%). Subsequently, the DA model was built using the same variables from PCA and demonstrated the good distinction between unadulterated and adulterated milk, with a correct classification rate of 98% for cross-validation. The MLR model used 48 variables with p-value < 0.05 from the DA model and gave R2 values greater than 0.90, with RMSE and MSE below 1 for cross-validation and prediction. The DA and MLR models were then validated externally using a test dataset, which shows 100% correct classification, and the t-test result (p > 0.05) indicated that the MLR could determine the percentage of urea in UHT milk within the permission limit (70 mg/mL). In short, the wavenumbers 1626.63, 1601.98, and 1585.5534 cm−1 are suitable as fingerprint regions for detecting urea in UHT milk. [ABSTRACT FROM AUTHOR]

The analysis of heroin samples, before use in the protected environment of user centra, could be a supplementary service in the context of harm reduction. Infrared spectroscopy hyphenated with multivariate calibration could be a valuable asset in this context, and therefore 125 heroin samples were collected directly from users and analysed with classical chromatographic techniques. Further, Mid-Infrared spectra were collected for all samples, to be used in Partial Least Squares (PLS) modelling, in order to obtain qualitative and quantitative models based on real live samples. The approach showed that it was possible to identify and quantify heroin in the samples based on the collected spectral data and PLS modelling. These models were able to identify heroin correctly for 96% of the samples of the external test set with precision, specificity and sensitivity values of 100.0, 75.0 and 95.5%, respectively. For regression, a root mean squared error of prediction (RMSEP) of 0.04 was obtained, pointing at good predictive properties. Furthermore, during mass spectrometric screening, 10 different adulterants and impurities were encountered. Using the spectral data to model the presence of each of these resulted in performant models for seven of them. All models showed promising correct-classification rates (between 92 and 96%) and good values for sensitivity, specificity and precision. For codeine and morphine, the models were not satisfactory, probably due to the low concentration of these impurities as a consequence of acetylation. For methacetin, the approach failed.

The diagnosis and management of poisoning is essential in critical care medicine. Traditionally, these conditions fall under the category of toxidromes that are the signs and symptoms associated with a particular class of poisons. However, there has been a steady increase in designer drugs and contaminants of recreational drugs themselves. Examples of adulterants in cocaine include the local anesthetic benzocaine and the anti-parasitic levamisole. This paper presents the clinical signs, laboratory findings, and treatment of patients who have been exposed to these substances.

Dairy products are widely consumed in the world due to their nutritional and functional characteristics. This group of food products are consumed by all age groups due to their health-giving properties. One of these products is cheese which has a high price compared to other dairy products. Because of this, it can be prone to fraud all over the world. Fraud in food products threatens the world's food safety and can cause serious damage to human health. There are many concerns among food authorities in the world about the fraud of food products. FDA, WHO, and the European Commission provide different legislations and definitions for fraud. The purpose of this review is to identify the most susceptible cheese type for fraud and effective methods for evaluating fraud in all types of cheeses. For this, we examined the Web of Science, Scopus, PubMed, and ScienceDirect databases. Mozzarella cheese had the largest share among all cheeses in terms of adulteration due to its many uses. Also, the methods used to evaluate different types of cheese frauds were PCR, Spectrometry, stable isotope, image analysis, electrophoretic, ELISA, sensors, sensory analysis, near-infrared and NMR. The methods that were most used in detecting fraud were PCR and spectrometry methods. Also, the least used method was sensory evaluation.

Background: Milk is important part of diet due to its nutritive values. It is recommended for population of every age group.Milk is required for the growth and maintenance of health. When consumers buy milk they have a right toassume that milk will be pure and unadulterated. It is the duty of the milk vendors to provide unadulterated milk to the consumers. Adulteration is the act of addition of substances to a product that makes it unfit for human consumption. These additives are used to make the product cheaper and increase its amount.1 Material and Method: A prospective study was conducted by Central Research Laboratory of Muzaffarnagar Medical College, to find out the adulteration of raw buffalo milk sampleobtained from rural areas around Muzaffarnagar Medical College,by color test methodology. The Institutional ethical committee permission was taken.100 samples were collected from 30 different villages around Muzaffarnagar Medical College. The raw milk sample was analyzed for physical appearance, pH, quality,dilution and presence of adulterant by color test. Result: It was observed that out of 100 samples, 85% samples were white and 15% samples were slightly yellow. The pH of the raw milk samples ranged from 6.5-7.0. The Methylene Blue Reduction Test (MBRT) showed 10samples of raw milk were of poor quality, 15 raw milk samples were fair quality, 45 raw milk samples were good quality and 30 raw milk samples were of very good quality. Result: In our study water and table sugar were the common adulterants and Benzoic acid was present in traces. So it was concluded that the raw buffalo milk obtained from the near by rural area around Muzaffarnagar Medical College was comparatively of good quality. The common adulterants obtained from raw milk of buffalo were water and table sugar. These malpractices by local dairy owner should be checked to ensure good quality of milk to the consumers. This study proposed a high-performance thin-layer chromatography (HPTLC) screening method to detect phosphodiesterase 5 (PDE-5) inhibitors as possible adulterant agents in various dietary supplements. Chromatographic analysis was performed on silica gel 60F254 plates using a mixture of ethyl acetate:toluene:methanol:ammonia in a volume ratio of 50:30:20:0.5 as a mobile phase. The system provided compact spots and symmetrical peaks of sildenafil and tadalafil with retardation factor values of 0.55 and 0.90, respectively. The analysis of products purchased from the internet or specialized stores demonstrated the presence of sildenafil, tadalafil, or both compounds in 73.3% of products, highlighting inadequacies and inconsistencies in the labeling, as all dietary supplements were declared to be natural. The results were confirmed using ultra-high-performance liquid chromatography coupled with a positive electrospray ionization high-resolution tandem mass spectrometry (UHPLC-HRMS-MS) method. Furthermore, in some samples, vardenafil and various analogs of PDE-5 inhibitors were detected using a non-target HRMS-MS approach. The results of the quantitative analysis revealed similar findings between the two methods, with adulterant quantities found to be similar to or higher than those in approved medicinal products. This study demonstrated that the HPTLC method is a suitable and economical method for screening PDE-5 inhibitors as adulterants in dietary supplements intended for sexual activity enhancement. Abstract Background Food adulteration is an increasingly recognized global public health problem. In low- and middle-income countries like Bangladesh, adulteration is difficult to detect and respond to. We explored customers’ perceptions on food adulteration, perception of risk and connections between information, participant characteristics and patterns of adulterated food concerns that impact risk perception in urban Bangladesh. Methods A formative study was conducted in Dhaka, between June and August 2015 at a supermarket and a wet market. We explored community awareness and response to chemical contaminants (adulterants) among participants from a range of socio-economic backgrounds. The team conducted 38 in-depth interviews with 12 customers and 4 staff from a supermarket, and 12 customers and 10 vendors from a wet market. Participants were selected purposively. Audio recorded data were coded based on thematic content and analyzed manually. Results We asked participants how common foods were likely adulterated, and most gave figures of 70% or more. They reported that foods were adulterated with chemicals or artificial colors, especially fish, milk, and vegetables. The supermarket more commonly sold packaged foods with nutritional and expiry information on the label; and offered convenience in terms of building size, layout, and cleanliness. All customers from the wet market thought that foods were cheaper and fresher than from supermarkets. Supermarket customers expressed greater concern about adulterated foods than wet market customers. Most participants from both markets reported that food adulteration is invisible, adulterated foods cannot be avoided, and have long-term negative health impacts including cancer, diabetes, paralysis, heart attack, and others. Nearly half of customers from both markets were concerned about the poor nutritional value of adulterated food. Participants from both settings expressed the need for access to credible information about adulteration to help choose safe foods. The majority expressed the need for government action against those who are responsible for adulteration. Conclusions Food adulteration was considered a major health threat. The government could act on food adulteration prevention if provided credible population-based data on disease burden, a model food sampling and testing protocol, a model for inspections, organizational strengthening and training, example social and behavioral change communications with estimated costs.

Illicit opioids, consisting largely of fentanyl, novel synthetic opioids, and adulterants, are the primary cause of drug overdose fatality in the United States. Xylazine, an alpha-2 adrenergic agonist and veterinary tranquilizer, is being increasingly detected among decedents following illicit opioid overdose. Clinical outcomes in non-fatal overdose involving xylazine are unexplored. Therefore, among emergency department patients with illicit opioid overdose, we evaluated clinical outcome differences for patients with and without xylazine exposures. This multicenter, prospective cohort study enrolled adult patients with opioid overdose who presented to one of nine United States emergency departments between 21 September 2020, and 17 August 2021. Patients with opioid overdose were screened and included if they tested positive for an illicit opioid (heroin, fentanyl, fentanyl analog, or novel synthetic opioid) or xylazine. Patient serum was analyzed via liquid chromatography quadrupole time-of-flight mass spectroscopy to detect current illicit opioids, novel synthetic opioids, xylazine and adulterants. Overdose severity surrogate outcomes were: (a) cardiac arrest requiring cardiopulmonary resuscitation (primary); and (b) coma within 4 h of arrival (secondary). Three hundred and twenty-one patients met inclusion criteria: 90 tested positive for xylazine and 231 were negative. The primary outcome occurred in 37 patients, and the secondary outcome occurred in 111 patients. Using multivariable regression analysis, patients positive for xylazine had significantly lower adjusted odds of cardiac arrest (adjusted OR 0.30, 95% CI 0.10–0.92) and coma (adjusted OR 0.52, 95% CI 0.29–0.94). In this large multicenter cohort, cardiac arrest and coma in emergency department patients with illicit opioid overdose were significantly less severe in those testing positive for xylazine. As they become more health conscious, consumers are paying increasing attention to food quality and safety. In coffee production, fraudulent strategies to reduce costs and maximize profits include mixing beans from two species of different economic value, the addition of other substances and/or foods, and mislabeling. Therefore, testing for coffee authenticity and detecting adulterants is required for value assessment and consumer protection. Here we provide an overview of the chromatography, spectroscopy, and single-nucleotide polymorphism-based methods used to distinguish between the major coffee species Arabica and Robusta. This review also describes the techniques applied to trace the geographical origin of coffee, based mainly on the chemical composition of the beans, an approach that can discriminate between coffee-growing regions on a continental or more local level. Finally, the analytical techniques used to detect coffee adulteration with other foods and/or coffee by-products are discussed, with a look at the practice of adding pharmacologically active compounds to coffee, and their harmful effects on health. Milk is a substrate for adulteration, and it is crucial to identify adulterants, and their quantities in milk products. In order to safeguard consumers from fake products and health risks, authenticity of milk products is in high demand. Analytical tools that are comprehensive, quick, and sensitive are necessary to monitor milk quality and screen for any undesirable substances. The traditional methods for the detection of adulterants in milk products are experiencing more difficulties due to their poor selectivity, sensitivity, and unsuitability for complex matrices. The researchers are paying close attention to nanomaterial based sensing systems, which are seen to be among the most effective alternatives to the traditional approaches. This review focuses on optical and electrochemical nanosensors including mechanisms and performances, in relation to their benefits, drawbacks, and applicability to milk products. It also addresses new developments in the field of nanoemsembles platforms for the rapid detection of adulterants in dairy products. A section of particular interest is the detection of milk authenticity, as it is known that milk from different animal species are intermixed.

Milk is a nutritionally rich food for humans. However, fulfilling the quality of milk is a major concern for milk factories, nutrient requirements, and public health. The objective of this research was to assess the composition of raw and pasteurized milk and cheese, evaluate change in milk and cheese composition along the value chain, and identify adulteration of milk. A total of 160 composite samples were determined using lactoscan and conventional approved methods along value chain. Results indicate that there were significant (p 0.05) difference in cheese nutritional quality between farmers and retailers. The grand mean for moisture, protein, fat, total ash, Ca, P and pH values were 77.1%, 17.1%, 1.42%, 1.18%, 37.8 mg/100 g, 88.2 mg/100 g and 3.7 respectively. Comparison of liquid products with the Compulsory Ethiopian Standard (CES) indicates that 80.2% for fat, protein, and SNF in raw and pasteurized milk were below the CES. In conclusion, liquid milk had poor nutritional composition and varied along the value chain in the study regions. Moreover, there is milk fraud where all dairy value chain add water into milk and milk consumers are consuming lower nutrients and paying for substandard liquid milk. Therefore, training should be provided to all value chain to improve the quality of milk products and quantification of formalin and other adulterants need to be further studied.

In recent period of time, mankind has been giving more and more effort towards dietary supplements because now people are more concerned about their health. So, the availability and uses are also increased. For enhancing the frequency and effect of the dietary supplements, synthetic products like Phosphodiesterase Type 5 (PDE-5) Inhibitor, Cocaine, Fluoxetine, or some unwanted steroids are added to the supplements to enhance their market value. The consumption of these adulterated supplements has been linked to health problems and has had a negative impact on the quality and safety of dietary supplements as well as public health. To stop such dishonest practises, analytical techniques that enable quick and accurate testing of dietary supplements for the presence of synthetic drugs are required. For the determination of adulterated products in dietary supplements there are so many hyphenated techniques are available among all Gas Chromatography and Mass Spectrometry (GC-MS) is more specific and delicate because of its unique combination of Mass Spectrometry and Gas chromatography which helps in the field of Separation and structural identification of the adulterant. The role of GC-MS in the detection, quantification, conformation analysis of adulterants in dietary supplements is evaluated critically in the current review.

A novel vardenafil analogue was identified in dietary supplement as an adulterant in herbal formulations. The structure of this analogue was elucidated using HRMS, NMR after extraction from the pulverized powder. It was named morphardenafil as a morpholine ring has replaced the N-ethyl piperazine ring in vardenafil. A tablet of this dietary supplement contained about 50 mg of unspecified morphardenafil, which is 2.5 – 20-times the prescriptive dosage of Levetra, the commercial formulation of the vardenafil monohydrochloride salt in the market and probably places unwary consumers at risk for potentially serious adverse effects or drug-drug interaction (DDI).

Tea established as a refreshing stimulant beverage rich in caffeine, sugars, lipids, and aromatic substances demonstrates a wide variation in the quality depending on the type of brand. In this study, five different black tea market brand samples collected from the local market were evaluated for adulterants, caffeine content, and antioxidant activity. Based on the laboratory tests, artificial colors, azo colours, and sand was detected in 80% of tea samples. Caffeine content was determined by the chloroform method. The tea samples exhibited a wide range of antioxidant activity (3.29±1.03 to 15.96±1.2) assayed by the DPPH free radical scavenging method. Adulteration is a well-known practice of drug manufacturers at different stages of drug production. The intentional addition of active ingredients to adulterate the primary drug may enhance or mask pharmacological effects or may produce more potent drugs to increase the number of available doses and the dealer's profit. Adulterants found in different drugs change over time in response to different factors. A systematic literature search in PubMed and Scopus databases and official international organizations' websites according to PRISMA guidelines was performed. A total of 724 studies were initially screened, with 145 articles from PubMed and 462 from Scopus excluded according to the criteria described in the Method Section. The remaining 117 records were further assessed for eligibility to exclude articles without sufficient data. Finally, 79 studies were classified as "non-biological" (n = 35) or "biological" (n = 35 case reports; n = 9 case series) according to the samples investigated. Although the seized samples analyses revealed the presence of well-established adulterants such as levamisole for cocaine or paracetamol/acetaminophen for heroin, the reported data disclosed new adulteration practices, such as the use of NPS as cutting agents for classic drugs of abuse and other NPS. For example, heroin adulterated with synthetic cannabinoids or cocaine adulterated with fentanyl/fentalogues raised particular concern. Notably, adulterants play a role in some adverse effects commonly associated with the primary drug, such as levamisole-adulterated cocaine that may induce vasculitis via an autoimmune process. Coconut (CO) and virgin coconut oil (VCO) are popular functional oils owing to their health benefits. The fraudsters intentionally blend cheap and poor-quality vegetable oils with CO and VCO for economic benefits. Adulteration causes safety and health issues for consumers. In this context, there is a need for rapid, accurate, and precise analytical techniques to detect adulteration in CO and VCO. Conventional analytical methods are arduous and lengthy processes using toxic chemicals and involve tedious sample preparation. Thus, food researchers are in search of precise, rapid, and green analytical techniques to detect and quantify the adulteration of CO and VCO. The present review focuses on summarizing all the analytical techniques along with chemometric tools developed in recent times. Various techniques, such as chromatographic, spectroscopic, and thermal techniques coupled with chemometrics, have been successfully applied in this field. The NIR-based spectroscopic technique is a very optimisable and practical method for adulteration detection in CO and VCO. Exposure to xylazine has been associated with wounds distinct from typical injection-related skin and soft tissue infections. We sought to understand drug use and wound care practices, and treatment experiences of people who use drugs (PWUD) in a high-prevalence area of xylazine adulteration. In August 2023, we surveyed adult PWUD reporting at least one past-year drug use-related wound across three Massachusetts syringe service programs. Using a representative illustration, participants indicated if they had experienced a xylazine wound in the past 90 days. We compared demographic, drug use factors, wound care, and medical treatment experiences among those with and without xylazine wounds. We also conducted additional content analysis of open-ended responses. Of the 171 respondents, 87 % (n=148) had a xylazine wound in the past 90 days. There were no statistically significant demographic differences between those with and without xylazine wounds. Among those primarily injecting (n=155), subcutaneous injection was nearly ten times more likely among people with xylazine wounds. For those with xylazine wounds (n=148), many engaged in heterogeneous wound self-treatment practices, and when seeking medical care, 74 % experienced healthcare stigma and 58 % had inadequate pain and withdrawal management. People with self-identified xylazine wounds were more likely to engage in subcutaneous injection and faced several barriers seeking medical wound treatment. Programs serving people exposed to xylazine should work to support safer injection practices, including alternatives to injecting and improving access to high-quality, effective wound care. Further study is warranted to understand the causes, promoters, and prevention of xylazine-related wounds. The P. heterophylla and its adulterants were identified by HPLC-CAD fingerprint of sucrose and oligosaccharides in P. heterophylla. The improved quantitative analysis of multi-components with a single marker (iQAMS) was further established for simultaneous determinations of sucrose and oligosaccharides in P. heterophylla. The HPLC-CAD fingerprint and similarity coefficients between P. heterophylla and its adulterants showed significant differences. The relative errors (REs) between iQAMS method and external standard method (ESM) were below 3.00%, but significant difference was shown between iQAMS (different marker for whole program with gradient elution) and QAMS (one marker for whole program with gradient elution), indicating that QAMS method should be improved, especially for gradient elution which influence the response of analytes. The accuracy, precision, reproducibility, and stability of this method were validated which exhibited satisfactory results, indicating that iQAMS method could be used for quantitative analysis of sucrose and oligosaccharides in P. heterophylla instead of ESM. Amomum tsaoko (AT) is commonly used in clinical practice to treat abdominal distension and pain. It is also a seasoning for cooking, with the functions of appetizing, invigorating the spleen, and being digestive-promoting. Amomum tsaoko (AT) has three adulterants, Amomum paratsaoko (AP), Amomum koenigii (AK), and Alpinia katsumadai Hayata, because of the confusion in historical classics regarding recorded sources as well as the near geographic distribution and fruit morphological similarities. In this study, we established a functional dyspepsia (FD) rat model and then treated it with the corresponding medicinal solutions AT, AP, AK, and AKH. The gastric emptying rate, intestinal propulsion rate, serum biochemical indicators, histopathological changes, and fecal metabolism were measured. The efficacy and mechanism of AT, AP, AK, and AKH in the treatment of FD were compared. Fecal metabolomics revealed that 20 potential biomarkers were involved in seven significant metabolic pathways in FD rats. These pathways include ubiquinone and other terpenoid-quinone biosynthesis, glycerophospholipid metabolism, tyrosine metabolism, primary bile acid biosynthesis, purine metabolism, folate biosynthesis, and amino sugar and nucleotide sugar metabolism. AP regulates 6 metabolic pathways, 5 metabolic pathways affected by AT, 4 metabolic pathways affected by AK, and 2 metabolic pathways affected by AKH.The above results suggest that the different effects of AT, AP, AK, and AKH on FD rats may be due to their different regulatory effects on the metabolome. Oat (Avena sativa L.) is recognized for its nutritional value and gluten-free status, however, ensuring its authenticity and purity is crucial, given the potential for contamination with gluten-containing grains. In this study, near-infrared spectroscopy coupled with chemometrics were employed to authenticate oat flour (from different forms of oat; oat groats, steel-cut and rolled oats) and distinguish it from common gluten-containing adulterants including wheat, farro, triticale, barley, rye, and ryegrass. Both unsupervised and supervised chemometric methodologies, encompassing PCA, SIMCA, and OPLS-DA, were applied. Both SIMCA and OPLS-DA models displayed 100% sensitivity, enabling reliable identification of oat flour and detection of potential adulteration with these gluten-containing grains with specificity of 97.78% and 100%, respectively. In the SIMCA model for oat groats and adulterated mixtures, samples with 1% and 2% adulteration were incorrectly classified as oat groats, yet successful discrimination from deliberately-adulterated mixtures was accomplished through the OPLS-DA model with 100% specificity. Moreover, PLS regression analysis was employed to precisely quantify the levels of adulterants in oat groats flour. The models exhibited reliable performance, as reflected in Root Mean Square Error of Calibration (RMSEC) values. Validation of these models using test samples provided further confirmation of their efficacy in detecting sample adulteration and ensuring product authenticity, all while maintaining a high sample throughput rate. There is a necessity to protect the quality and authenticity of herbs and spices because of the increase in the fraud and adulteration incidence during the last 30 years. There are several aspects that make herbs and spices quite vulnerable to fraud and adulteration, including their positive and desirable sensorial and health-related properties, the form in which they are sold, which is mostly powdered, and their economic relevance around the world, even in developing countries. For these reasons, sensitive, rapid, and reliable techniques are needed to verify the authenticity of these agri-food products and implement effective adulteration prevention measures. This review highlights why spices and herbs are highly valued ingredients, their economic importance, and the official quality schemes to protect their quality and authenticity. In addition to this, the type of frauds that can take place with spices and herbs have been disclosed, and the fraud incidence and an overview of scientific articles related to fraud and adulteration based on the Rapid Alert System Feed and Food (RASFF) and the Web of Science databases, respectively, during the last 30 years, is carried out here. Next, the methods used to detect adulterants in spices and herbs are reviewed, with DNA-based techniques and mainly spectroscopy and image analysis methods being the most recommended. Finally, the available adulteration prevention measurements for spices and herbs are presented, and future perspectives are also discussed.

Ayurveda is a centuries old traditional medicine practiced in India even today. There are certain safe medicinal plants with well-established medicinal properties both in clinical practice as well as in modern scientific publications. Guduchi or Tinospora cordifolia (Willd.) Miers (Menispermaceae), is one such medicinal plant that has well known anti-inflammatory, immune-modulatory and other safe therapeutic applications including hepato-protection, because of which it was recommended by the Ministry of AYUSH, Government of India to be used in COVID-19 care. Therefore, Aabha Nagral's article “Herbal Immune Booster-Induced Liver Injury in the COVID-19 Pandemic-a Case Series,” published in 2021, was unanticipated. The article recounted histologically documented clinical cases of six patients who developed drug-induced autoimmune-like hepatitis after reported consumption of Guduchi or Guduchi containing formulations during the COVID-19 pandemic. Since the Ayurveda practitioners vouch by the safety of T. cordifolia (TC), it was felt that the story needed to be further scrutinized.This article reviews the botanical entities, the substitutes and adulterants of species used as Guduchi, their pharmacological and toxicological properties. While the authentic botanical entity of Guduchi is TC, Tinospora sinensis and Tinospora crispa are also commonly traded in the Indian subcontinent as Guduchi or Giloy. Among these species, T. crispa is known to induce heapto-toxicity. In Nagral's article, there were variations in the reported six cases in terms of patient history and TC/TC product consumption. More importantly, the botanical authenticity of the consumed products was not investigated.A review of published literature indicates that it is unlikely that the authentic TC could have induced autoimmune-like hepatitis of the patients. It is probable that a wrong species was self-administered by the patients. It is worth following up with the cases (patients), to investigate details of the products, so that other consumers do not suffer. Nagral's article however does highlight the serious issue of adulteration in herbal markets and the need for establishing a robust pharmacovigilant system in India.

The following paper presents a construct for a paper-based device which utilizes latex as the hydrophobic material for the fabrication of its hydrophobic barrier, which was deposited onto the cellulose surface either by free-hand or stenciled drawing. This method demands the least amount of expertise and time from its use, enabling a simple and rapid fabrication experience. Several properties of the hydrophobic material were characterized, such as the hydro head and penetration rate, with the aim of assessing its robustness and stability. The presented hydrophobic barriers fabricated using this approach have a barrier width of 4 mm, a coating thickness of 208 µm, and a hydrophilic resolution of 446.5 µm. This fabrication modality boasts an excellent solvent resistance with regard to the hydrophobic barrier. These devices were employed for on-the-spot detection of Metanil Yellow, a banned food adulterant often used in curcumin and pigeon peas, within successful limits of detection (LOD) of 0.5% (w/w) and 0.25% (w/w), respectively. These results indicate the great potential this fabricated hydrophobic device has in numerous paper-based applications and other closely related domains, such as diagnostics and sensing, signalling its capacity to become commonplace in both industrial and domestic settings. Infrared spectroscopy has been shown to be efficient in cheese authentication due to the advantages of high sensitivity and speed of analysis, especially when associated with chemometrics. This review discusses approaches on the authenticity, the principles of near‐ and middle‐infrared techniques and the importance of chemometrics for cheese authentication. The spectroscopic techniques proved to be promising for the cheese geographical origin identification, analysis of adulterants and monitoring of maturation stages. The application of principal component analysis, partial least squares and linear discriminant analysis associated with spectroscopy has provided powerful tools for the cheese authentication.
