Abstract Aims Clinical trials in heart failure with mildly reduced or preserved ejection fraction (HFmrEF/HFpEF) commonly have detailed eligibility criteria. This may contribute to challenges with efficient enrolment and questions regarding the generalizability of trial findings. Methods and results Patients with HFmrEF/HFpEF from a large US healthcare system were identified through a computable phenotype applied in linked imaging and electronic health record databases. We evaluated shared eligibility criteria from five recent/ongoing HFmrEF/HFpEF trials (PARAGON‐HF, EMPEROR‐Preserved, DELIVER, FINE‐ARTS, and SPIRRIT‐HFpEF) and compared clinical and echocardiographic features as well as outcomes between trial‐eligible and trial‐ineligible patients. Among 5552 patients with HFpEF/HFmrEF, 792 (14%) were eligible for trial consideration, having met all criteria assessed. Causes of ineligibility included lack of recent loop diuretics (37%), significant pulmonary disease (24%), reduced estimated glomerular filtration rate (17%), recent stroke/transient ischaemic attack (13%), or low natriuretic peptides (12%); 53% of ineligible patients had >1 reason for exclusion. Abstract Clinical evidence generation from and for representative populations can be improved through increased research access and ease of trial participation. To improve access and participation, a modern trial infrastructure is needed that broadens research into more routine practice. This commentary highlights current barriers, areas of advancement, and actions needed to enable continued transformation toward a modern trial infrastructure for an improved evidence generation system. The focus of this commentary is on the development of medical products (e.g., drugs, devices, biologics) and infrastructure issues within the United States, with the aim to have broader, multi-national applicability.

The COVID-19 pandemic and the subsequent measures taken to control its spread had deep impacts on transportation behavior. This paper presents a methodology for evaluating the changes in bike-share use and focuses on the identification of extraordinary trip-making activity seen in Chicago’s Divvy bike-share program prior to and over the course of the COVID-19 pandemic. An analysis period from January 2018 to April 2021 was subdivided into three time periods: pre-pandemic, citywide lockdown, and post-lockdown for all census tracts within the city of Chicago. Over the analysis window defined, an anomaly detection algorithm was used to characterize the nature of bike-share usage where anomalous trips were those that fell outside of the forecasted ranges. The changes in census tract-level bike-share usage were used to interpret how static census tract-level attributes—land use, transit connectivity, demographics, and bike infrastructure—may have contributed to varying responses across the city’s bike-share system. Principal component analysis was used to interpret the relationship between the dynamic trip-making characterizations and the static census tract-level attributes. Census tracts that transitioned from exhibiting below average to above average bike-share trip-making tended to have highly correlated bike infrastructure metrics. On the other hand, census tracts with consistently above average trip-making had highly correlated demographic characteristics, the values of which were consistent with their standing as key communities for service and essential workers. Overall, results indicate that post-lockdown, bike-sharing activity had grown significantly across the entire city, supporting the claim that the pandemic worked to accelerate bicycling uptake in urban areas.

Abstract Background We examined multi-dimensional clinical and laboratory data in participants with normoglycemia, prediabetes, and diabetes to identify characteristics of prediabetes and predictors of progression from prediabetes to diabetes or reversion to no diabetes. Methods The Project Baseline Health Study (PBHS) is a multi-site prospective cohort study of 2502 adults that conducted deep clinical phenotyping through imaging, laboratory tests, clinical assessments, medical history, personal devices, and surveys. Participants were classified by diabetes status (diabetes [DM], prediabetes [preDM], or no diabetes [noDM]) at each visit based on glucose, HbA1c, medications, and self-report. Principal component analysis (PCA) was performed to create factors that were compared across groups cross-sectionally using linear models. Logistic regression was used to identify factors associated with progression from preDM to DM and for reversion from preDM to noDM. Results At enrollment, 1605 participants had noDM; 544 had preDM; and 352 had DM. Over 4 years of follow-up, 52 participants with preDM developed DM and 153 participants reverted to noDM. PCA identified 33 factors composed of clusters of clinical variables; these were tested along with eight individual variables identified a priori as being of interest. Abstract Nanobiotechnology is one of the leading research areas in biomedical science, developing rapidly worldwide. Among various types of nanoparticles, carbon nanomaterials (CNMs) have attracted a great deal of attention from the scientific community, especially with respect to their prospective application in the field of disease diagnosis and therapy. The unique features of these nanomaterials, including favorable size, high surface area, and electrical, structural, optical, and chemical properties, have provided an excellent opportunity for their utilization in theranostic systems. Carbon nanotubes, carbon quantum dots, graphene, and fullerene are the most employed CNMs in biomedical fields. They have been considered safe and efficient for non‐invasive diagnostic techniques such as fluorescence imaging, magnetic resonance imaging, and biosensors. Various functionalized CNMs exhibit a great capacity to improve cell targeting of anticancer drugs. Due to their thermal properties, they have been extensively used in cancer photothermal and photodynamic therapy assisted by laser irradiation and CNMs. CNMs also can cross the blood‐brain barrier and have the potential to treat various brain disorders, for instance, neurodegenerative diseases, by removing amyloid fibrils. This review has summarized and emphasized on biomedical application of CNMs and their recent advances in diagnosis and therapy.

Abstract The Project Baseline Health Study (PBHS) was launched to map human health through a comprehensive understanding of both the health of an individual and how it relates to the broader population. The study will contribute to the creation of a biomedical information system that accounts for the highly complex interplay of biological, behavioral, environmental, and social systems. The PBHS is a prospective, multicenter, longitudinal cohort study that aims to enroll thousands of participants with diverse backgrounds who are representative of the entire health spectrum. Enrolled participants will be evaluated serially using clinical, molecular, imaging, sensor, self-reported, behavioral, psychological, environmental, and other health-related measurements. An initial deeply phenotyped cohort will inform the development of a large, expanded virtual cohort. The PBHS will contribute to precision health and medicine by integrating state of the art testing, longitudinal monitoring and participant engagement, and by contributing to the development of an improved platform for data sharing and analysis.

Newer data platforms offer increased opportunity to share multidimensional health data with research participants, but the preferences of participants for which data to receive and how is evolving. Our objective is to describe the preferences and expectations of participants for the return of individual research results within Project Baseline Health Study (PBHS). The PBHS is an ongoing, multicenter, longitudinal cohort study with data from four initial enrollment sites. PBHS participants are recruited from the general population along with groups enriched for heart disease and cancer disease risk. Cross-sectional data on return of results were collected in 2017-2018 from an (1) in-person enrollment survey (n = 1,890), (2) benchmark online survey (n = 1,059), and (3) participant interviews (n = 21). The main outcomes included (1) preferences for type of information to be added next to returned results, (2) participant plans for sharing returned results with a non-study clinician, and (3) choice to opt-out of receiving genetic results. Results were compared by sociodemographic characteristics. Enrollment and benchmark survey respondents were 57.1% and 53.5% female, and 60.0% and 66.2% white, respectively. Participants preferred the following data types be added to returned results in the future: genetics (29.9%), heart imaging, (16.4%), study watch (15.8%), and microbiome (13.3%). Older adults (OR 0.60, 95% CI: 0.41-0.87) were less likely to want their genetic results returned next. Forty percent of participants reported that they would not share all returned results with their non-study clinicians. Black (OR 0.64, 95% CI 0.43-0.95) and Asian (OR 0.47, 95% CI 0.30-0.73) participants were less likely, and older participants more likely (OR 1.45-1.61), to plan to share all results with their clinician than their counterparts. At enrollment, 5.8% of participants opted out of receiving their genetics results. The study showed that substantial heterogeneity existed in participant's preferences and expectations for return of results, and variations were related to sociodemographic characteristics.

Background There are conflicting data regarding the benefit of primary prevention implantable cardioverter‐defibrillators (ICDs) in patients with diabetes mellitus and heart failure (HF) with reduced ejection fraction. We aimed to assess the comparative effectiveness of ICD placement in patients with diabetes mellitus and HF with reduced ejection fraction. Methods and Results Data were obtained from the Get With the Guidelines–Health Failure registry, linked with claims from the Centers for Medicare & Medicaid Services. We used a Cox proportional hazards model censored at 5 years with propensity score matching. Of the 17 186 patients with HF with reduced ejection fraction from the Centers for Medicare & Medicaid Services claims database (6540 with diabetes mellitus; 38%), 1677 (646 with diabetes mellitus; 39%) received an ICD during their index HF hospitalization or were prescribed an ICD at discharge. Patients with diabetes mellitus and an ICD (n=646), as compared with those without an ICD (n=1031), were more likely to be younger (74 versus 78 years of age) and have coronary artery disease (68% versus 60%). After propensity matching, ICD use among patients with diabetes mellitus, as compared with those without an ICD, was associated with a reduced risk of all‐cause mortality at 5 years after HF discharge (54% versus 59%; multivariable hazard ratio, 0.73; 95% CI, 0.64–0.82; P

Background We aimed to determine if there is an association between hospital quality and the likelihood of a given hospital being a preferred transfer destination for stroke patients. Methods and Results Data from Medicare claims identified acute ischemic stroke transferred between 394 northeast US hospitals from 2007 to 2011. Hospitals were categorized as transferring (n=136), retaining (n=241), or receiving (n=17) hospitals based on the proportion of acute ischemic stroke encounters transferred or received. We identified all 6409 potential dyads of sending and receiving hospitals, and categorized dyads as connected if ≥5 patients were transferred between the hospitals annually (n=82). We used logistic regression to identify hospital characteristics associated with establishing a connected dyad, exploring the effect of adjusting for different quality measures and outcomes. We also adjusted for driving distance between hospitals, receiving hospital stroke volume, and the number of hospitals in the receiving hospital referral region. The odds of establishing a transfer connection increased when rate of alteplase administration increased at the receiving hospital or decreased at the sending hospital, however this finding did not hold after applying a potential strategy to adjust for clustering. Receiving hospital performance on 90‐day home time was not associated with likelihood of transfer connection. Conclusions Among northeast US hospitals, we found that differences in hospital quality, specifically higher levels of alteplase administration, may be associated with increased likelihood of being a transfer destination. Further research is needed to better understand acute ischemic stroke transfer patterns to optimize stroke transfer systems.

Glioblastoma (GBM) is the most common primary malignant brain tumor in adults with an extremely poor prognosis. There is a dire need to develop effective therapeutics to overcome the intrinsic and acquired resistance of GBM to current therapies. The process of developing novel anti-neoplastic drugs from bench to bedside can incur significant time and cost implications. Drug repurposing may help overcome that obstacle. A wide range of drugs that are already approved for clinical use for the treatment of other diseases have been found to target GBM-associated signaling pathways and are being repurposed for the treatment of GBM. While many of these drugs are undergoing pre-clinical testing, others are in the clinical trial phase. Since GBM stem cells (GSCs) have been found to be a main source of tumor recurrence after surgery, recent studies have also investigated whether repurposed drugs that target these pathways can be used to counteract tumor recurrence. While several repurposed drugs have shown significant efficacy against GBM cell lines, the blood–brain barrier (BBB) can limit the ability of many of these drugs to reach intratumoral therapeutic concentrations. Localized intracranial delivery may help to achieve therapeutic drug concentration at the site of tumor resection while simultaneously minimizing toxicity and side effects. These strategies can be considered while repurposing drugs for GBM.

Smart mobility is a central issue in the recent discourse about urban development policy towards smart cities. The design of innovative and sustainable mobility infrastructures as well as public policies require cooperation and innovations between various stakeholders—businesses as well as policy makers—of the business ecosystems that emerge around smart city initiatives. This poses a challenge for deploying instruments and approaches for the proactive management of such business ecosystems. In this article, we report on findings from a smart city initiative we have used as a case study to inform the development, implementation, and prototypical deployment of a visual analytic system (VAS). As results of our design science research we present an agile framework to collaboratively collect, aggregate and map data about the ecosystem. The VAS and the agile framework are intended to inform and stimulate knowledge flows between ecosystem stakeholders in order to reflect on viable business and policy strategies. Agile processes and roles to collaboratively manage and adapt business ecosystem models and visualizations are defined. We further introduce basic categories for identifying, assessing and selecting Internet data sources that provide the data for ecosystem models and we detail the ecosystem data and view models developed in our case study. Our model represents a first explication of categories for visualizing business ecosystem models in a smart city mobility context.

Lactobacillus reuteri and Bifidobacterium bifidum were inoculated (2.8·10^8 and 4.7·10^8 CFU/mL, respectively) into reconstituted whey containing sucrose and pectin in order to prepare a fermented probiotic product. Inoculation levels were: 0.5, 1 or 2 % for Lactobacillus reuteri and 0.5 or 1 % for Bifidobacterium bifidum. The treatment with the highest bacterial counts and sensory scores was selected and stored at 4 °C for 30 days. Microbial counts, changes in pH values, titratable acidity and both triangle test and sensory attributes were monitored on the stored product. The beverage fermented for approx. 11 h and prepared with 2 % Lactobacillus reuteri and 0.5 % Bifidobacterium bifidum met the probiotic criterion by maintaining both bacterial populations at counts greater than 10^6 CFU/mL for the whole storage period. Titratable acidity and pH values as well as sensory properties did not change appreciably during the first 14 days of storage. At the end of the storage period (30 days), slight acidification was detected, although the beverage still retained an acceptable flavour.

Mucus hypersecretion by airway epithelium is a hallmark of inflammation in allergic asthma and results in airway narrowing and obstruction. Others have shown that administration a TH2 cytokine, IL-13 is sufficient to cause mucus hypersecretion in vivo and in vitro. Asthma therapy often utilizes β2-adrenoceptor (β2AR) agonists, which are effective acutely as bronchodilators, however chronic use may lead to a worsening of asthma symptoms. In this study, we asked whether β2AR signaling in normal human airway epithelial (NHBE) cells affected mucin production in response to IL-13. This cytokine markedly increased mucin production, but only in the presence of epinephrine. Mucin production was blocked by ICI-118,551, a preferential β2AR antagonist, but not by CGP-20712A, a preferential β1AR antagonist. Constitutive β2AR activity was not sufficient for IL-13 induced mucin production and β-agonist-induced signaling is required. A clinically important long-acting β-agonist, formoterol, was as effective as epinephrine in potentiating IL-13 induced MUC5AC transcription. IL-13 induced mucin production in the presence of epinephrine was significantly reduced by treatment with selective inhibitors of ERK1/2 (FR180204), p38 (SB203580) and JNK (SP600125). Replacement of epinephrine with forskolin + IBMX resulted in a marked increase in mucin production in NHBE cells in response to IL-13, and treatment with the inhibitory cAMP analogue Rp-cAMPS decreased mucin levels induced by epinephrine + IL-13. Our findings suggest that β2AR signaling is required for mucin production in response to IL-13, and that mitogen activated protein kinases and cAMP are necessary for this effect. These data lend support to the notion that β2AR-agonists may contribute to asthma exacerbations by increasing mucin production via activation of β2ARs on epithelial cells.

Recent projections suggest that food security may face threats due to climate change and population growth. Global warming is altering ocean temperatures, impacting productivity and aquatic ecosystems. This review delves into the impact of heat stress on fish aquaculture, focusing on the neuroendocrine system, controlled by the hypothalamic-pituitary-interrenal (HPI) axis. This axis regulates stress responses via cortisol and catecholamines, influencing energy metabolism and stress adaptation. The review discusses molecular adaptations in skeletal muscle and liver, affecting gene expression related to growth and energy metabolism. On the other hand, heat stress provokes notable molecular responses, altering the expression of genes involved in glycolysis and oxidative phosphorylation, and hormonal pathways like growth hormone and insulin growth factors, which are vital for metabolic regulation and muscle growth. In summary, this review provides insights into the neuroendocrine and molecular mechanisms underlying fish responses to thermal stress, emphasizing the importance of skeletal muscle and liver tissues in mediating physiological adaptations. Furthermore, it analyzes the implications for protein synthesis, cellular signaling pathways, oxidative stress, and enzymatic responses in aquatic organisms, highlighting the importance of understanding these molecular adaptations to develop effective mitigation strategies that ensure the sustainability of aquaculture systems amidst changing conditions.

Abstract The alteration of structure and function of gut microbiota (dysbiosis) appears to be a major factor associated with metabolic disorders such as dyslipidemia and subsequent development of cardiovascular diseases. However, the consumption of fermented milks is a promising strategy to enhance health and restore the function of gut microbiota; specifically, in individuals with intestinal dysbiosis and hypercholesterolemia. Therefore, the aim of the present study was to evaluate the potential association between gut microbiota and the hypocholesterolemic effect of fermented milks with Lactococcus lactis NRRL B-571 (FM-571), NRRL B-572 (FM-572) and NRRL B-600 (FM-600) in Sprague–Dawley rats. Fermented milks were administered to hypercholesterolemic Sprague–Dawley rats during seven weeks. At the end of the experimental period, fecal and colonic microbiota were characterized using 16S RNA gene sequencing. Also, the short chain fatty acids (SCFAs) content was quantified in feces. Results showed that a high-cholesterol diet (HCD) altered the bacterial community in both fecal and mucosal samples. The consumption of fermented milks, specifically FM-572 promoted changes in the structure (beta diversity) in fecal, but not in mucosal microbiota. To transform Mexico's electric load infrastructure, accurate electric load forecasts are required that are crucial to efficiently allocate resources, maintain system stability, and manage energy. The purpose of this study is to use the Quantile Transformer Network (QTN) as a novel approach for a deep learning framework for load forecasting, emphasizing its potential and practical consequences in enhancing the accuracy of load forecasting in real-world energy systems. Moreover, it is shown that QTN efficiently captures complex patterns, temporal relationships and interconnections among factors that influence electric load. The dataset utilized consists of past records of energy consumption in the Baja California System in Mexico. It includes several factors such as electricity demand, marginal prices, temporal characteristics, temperature-related variables, seasonal patterns, and holidays. Additionally, QTN is combined with the Rainbow Technique (RT) to manage categorical variables, resulting in the creation of a unified feature called category. In this case, RT examines the connections between descriptive phrases that reflect distinct combinations of categorical factors depending on the load values. Moreover, this research shows that the common Quantile Regression (QR) outperforms QTN in capturing dependencies in sequential data. Finally, several recommendations for promoting Mexico's Electric Load Infrastructure are provided.

This research develops a new electric charge prediction method by using Convolutional Neural Networks with Quantile Regression (CNN-QR) combined with the Rainbow Technique for Categorical Features (RTCF) and using Deep Learning to create layers for the architecture of the neural network. This combination captures both local and global interdependencies within the load data. In particular, RTCF employs advanced natural language processing (NLP) techniques to convert several important categorical features into a single feature called “category,” which is tailored to the various attributes of the Baja California Sur system, in Mexico, taking into consideration climatic conditions, local circumstances and a significant increase in energy consumption. Furthermore, this research compares CNN-QR with classical quantile regression and shows that CNN-QR works better at capturing sophisticated load patterns and producing probabilistic estimates. The above methodology uses hourly data from January 2019 to October 2020. The results obtained provide valuable information for policy formulation in the energy sector, specifically in the areas of load forecasting and expansion of renewable energy and electricity consumption. Finally, it is worth mentioning that the utilization of CNN-QR with RTCF not only improves the accuracy of load forecasting, but also provides a strategic framework for energy management and resource planning in dynamic energy systems, which demonstrates its substantial importance for market participants and authorities, as well as regulators.

Abstract Predators in food webs are valuable sentinel species for zoonotic and multi-host pathogens such as Toxoplasma gondii. This protozoan parasite is ubiquitous in warm-blooded vertebrates, and can have serious adverse effects in immunocompromised hosts and foetuses. In northern ecosystems, T. gondii is disproportionately prevalent in Inuit people and wildlife, in part due to multiple routes of transmission. We combined data on T. gondii infection in foxes from Nunavik (northern Québec, Canada) with stable isotope data tracking trophic relationships between foxes and several of their main prey species. Red (Vulpes vulpes) and Arctic fox (Vulpes lagopus) carcasses were collected by local trappers from 2015 to 2019. We used magnetic capture PCR to detect DNA of T. gondii in heart and brain tissues, and enzyme-linked immunosorbent assay to detect antibodies in blood. By linking infection status with diet composition, we showed that infected foxes had a higher probability of consuming aquatic prey and migratory geese, suggesting that these may be important sources of T. gondii transmission in the Arctic. This use of stable isotopes to reveal parasite transmission pathways can be applied more broadly to other foodborne pathogens, and provides evidence to assess and mitigate potential human and animal health risks associated with T. gondii in northern ecosystems.

La gentrificación es un proceso de revitalización, elitización y blanqueamiento de algunos barrios en las ciudades. En este fenómeno urbano las contrasexualidades han encontrado una oportunidad —mediante la capacidad de gasto y el consumo— para habitarlos y constituir otros lugares de enunciación. De esta manera, la presencia de las contrasexualidades en barrios gentrificados inicia un proceso de queerificación, es decir, se asimilan y se integran estilos de vida, familias e identidades LGBTIQ+ que terminan por cuestionar y abrir grietas en el sistema cisheteropatriarcal que por mucho tiempo han imperado. En este contexto, se analiza el punto de conexión entre queerificación y gentrificación en el barrio Santa María la Ribera en Ciudad de México. Para ello se apuntan las prácticas del consumo de espacios que ofertan alimentos, bebidas, cultura, arte y militancia. La impronta está colocada en el consumo cultural LGBTIQ+ como el dinamizador de cuerpos y habitares que incentivan el proceso de gentrificación y al mismo tiempo la deconstrucción del heteropatriarcado.

Anti-soybean agglutinin (SBA) IgY was produced, and its potential to neutralize the haemagglutinating activity of SBA in vitro was tested. Thirty-five-week-old hens [treatment (n = 5) and control (n = 5)] were immunized with SBA or injected with saline 4 times every 15 days. Eggs were collected after the last immunization, and IgY was extracted using the polyethylene glycol (PEG) method. Serum anti-SBA IgY titres in immunized hens increased after the first immunization and reached a plateau between days 45 and 60. In contrast, specific IgY titres in the control group remained at basal levels throughout the evaluation. Average IgY titres were significantly higher in the treatment group on days 15, 30, 45, and 60. Total IgY content in the egg yolk extract was 38.7 ± 1.6 and 37.7 ± 1.5 mg/ml for the treatment and control groups, respectively. The specific anti-SBA IgY titer detected in the egg yolk extract was significantly higher (p < 0.001) for hens in the treatment group compared to the control group, with OD450nm values of 0.98 ± 0.05 and 0.058 ± 0.02, respectively. The specificity of anti-SBA IgY was confirmed by the Western blotting, and the inhibition of SBA-induced haemagglutination in vitro was compared with D-galactose, a known molecule that binds to SBA and blocks its binding to erythrocytes. The inhibition of SBA-induced haemagglutination by the anti-SBA IgY reached 512 units of haemagglutination inhibition (UHI), compared to 8 or 256 UHI, respectively, when IgY from control chickens or D-galactose was used. Thus, anti-SBA IgY antibodies were efficiently produced in large quantities and effectively inhibited SBA-induced haemagglutination in vitro.

Abstract Human autoimmune diseases are complex and highly diverse conditions that can be of localised or systemic nature. Understanding the basic biology of autoimmune diseases goes hand in hand with providing the clinics with valuable biomarkers for managing these diseases. The focus of this review is paid to localised scleroderma, an autoimmune disease affecting skin and subcutaneous tissue. Localised scleroderma has very few serological biomarkers for clinical analyses distinguishing it from main differentials, and yet noneffective prognostic biomarkers. With this regard, the review covers well‐established and new biomarkers such as cell surface proteins, autoantibodies and cytokines. In recent few years, several new biomarkers have been suggested, many provided with modern genomic studies. This includes epigenetic regulation of DNA, RNA transcriptomics and regulatory RNA such as microRNA and long non‐coding RNA. These findings can for the first time shed light on the genetic mechanisms behind the disease and contribute to improved diagnosis and treatment.

Caribou are keystone species important for human harvest and of conservation concern; even so, much is unknown about the impact of parasites on caribou health and ecology. The aim of this study was to determine the seroprevalence, tissue prevalence, and diversity of tissue-dwelling coccidian parasites (including Toxoplasma gondii, Neospora caninum and Sarcocystis spp.) in 88 migratory caribou (Rangifer tarandus) harvested for human consumption in two communities in Nunavik, Québec, Canada. Both T. gondii and N. caninum have potential to cause abortions and neurological disease in caribou. Seroprevalence for antibodies to T. gondii using ELISA on fluid from thawed hearts was 18% overall, and no DNA of T. gondii was detected in tissues, which has positive implications for food safety since this parasite is zoonotic. Seroprevalence for antibodies to N. caninum using competitive ELISA was 5%, and DNA of N. caninum was detected in only one heart sample. DNA of Sarcocystis, a non-zoonotic, related coccidian, was detected in tissue samples from 85% of caribou, with higher prevalence in heart (82%) than skeletal muscle (47%). This is the first time that Sarcocystis spp. from caribou in Canada have been identified to species level, many of which have been described in reindeer from Fennoscandia. The high prevalence and diversity of Sarcocystis spp. suggests intact trophic relationships between canids and caribou in Nunavik. Besnoitia spp. was serendipitously detected in three muscle samples, a parasite previously associated with skin lesions in caribou in Nunavik. Community-level differences in T. gondii exposure and prevalence of Sarcocystis spp. in skeletal muscle tissues may reflect differences in hunter selection of individual animals and muscles, or possibly regional differences in the ecology of carnivore definitive hosts for these parasites. Further work is needed to explore effects of tissue coccidians in caribou, their taxonomic classifications, and community level differences in parasite prevalence and diversity.

La altura dominante de los árboles de un sitio es un indicador indirecto de la productividad de los suelos forestales y se le denomina calidad de estación, la cual se mide a través del índice de sitio. El objetivo fue ajustar modelos de altura dominante en función de la edad con enfoque de modelos de efectos mixtos para generar curvas de índice de sitio en plantaciones forestales de Pinus oocarpaShiede en Ario de Rosales, Michoacán, México. Con 1 773 pares de datos altura dominante-edad de plantaciones forestales de entre dos a 20 años y relictos de 56 años, se ajustaron cinco modelos de crecimiento para elegir uno base. El modelo de Weibull se ajustó por modelos de efectos mixtos con la parcela como covariable aditiva en el parámetro relacionado al sitio. La curva guía en altura dominante y las curvas polimórficas cubren adecuadamente la variabilidad muestral de la información y representan de forma confiable la productividad del sitio donde crecen estas plantaciones. El sesgo y la validación de los resultados mostró confiabilidad en las estimaciones. El turno técnico en altura dominante está entre cinco y 15 años de acuerdo con su calidad de estación. Con estos resultados es posible realizar la clasificación de las plantaciones forestales de Pinus oocarpa de manera confiable y acorde con el nivel de productividad de cada sitio de establecimiento.

Electronic flexible devices are prone to degrade their electrical performance or lose functionality when subjected to deformations. Brittle fracture is a common damaging effect observed in devices composed of low-thickness layered materials stacked onto a flexible substrate by dissimilar mechanical properties interaction. This work studies the mechanical behavior of Organic Flexible Solar Cells (OFSC) with a heterostructure PET/ITO/P3HT:PCBM/Ag subjected to uniaxial displacements through an experimental and numeric point of view. Experimental showed that damage proceeds in two ways. First, the formation of a grid crack pattern begins at the ITO layer, and second, the delamination in the ITO/P3HT:PCBM interface. The numerical model analyzed the force and displacements and the absorption/dissipation of strain energy on layers and interfaces of the device. The comparison of the global Young's module for experimental and numeric studies validated the numeric analysis, with results of 4.16 ± 0.05 GPa for experimental and 4.36 ± 0.15 GPa for numeric. Additionally, the model associates the ITO layer with the highest strain energy dissipation or the most prone to failure, which agrees with the experiments. Then, the model successfully predicts the mechanical behavior of OFSC and represents a valuable tool for studying flexible devices and predicting the appearance of mechanical damage when subjected to uniaxial deformations, even being able to avoid potential damage changing parameters such as the thickness of the layers.

In recent years, the quest to advance fuel cell technologies has intensified, driven by the imperative to reduce reliance on hydrocarbon-derived fuels and mitigate pollutant emissions. Proton exchange membranes are a critical material of fuel cell technologies. The potential of ionic liquid-based polymer inclusion membranes or ionogels for proton exchange membrane fuel cells (PEMFCs) has recently appeared. Thermal stability, SEM-EDX characterization, NMR and IR characterization, thermogravimetric analysis, ion exchange capacity, and water uptake are key properties of these membranes which need to be investigated. In this work, ionogel based on quaternary ammonium salts, such as [N8,8,8,1+][Cl−], [N8,8,8,1+][Br−], and [N8-10,8-10,8-10,1+][Cl−] in various compositions with poly(vinyl chloride) are extensively studied and characterized based on those key properties. The best properties were obtained when a quaternary ammonium cation was combined with a bromide anion. Finally, ionogels are tested in microbial fuel cells. Microbial fuel cells based on the ionogel reach a maximum of 147 mW/m2, which represents 55% of the reference membrane (Nafion 212). These results indicate that we still have the possibility of improvement through the appropriate selection of the cation and anion of the ionic liquid. Overall, the promise of ionogel membranes as a viable alternative in fuel cell applications has been demonstrated.

En este trabajo mostramos las tensiones que se generan en el paisaje alimentario de la demarcación de Cholula (México). En ésta existen dos municipios entre los que se genera una frontera culinaria. A partir de una aproximación cualitativa por medio de entrevistas semiestructuradas, observación participante y análisis de contenido en medios digitales, queremos demostrar que la existencia de la frontera gastronómica parece habilitar dos paisajes culinarios diferenciados que obedecen a distintos intereses. De esta manera, podríamos afirmar que la frontera gastronómica habilita y dinamiza fenómenos opuestos que favorecen el contraste en el ensamblaje gastronómico. Por un lado, la intención de la estatización del paisaje que obedece a lógica de la turistificación que apela a la recuperación de la tradicionalidad y la historia para favorecer los discursos en torno al turismo y al patrimonio. Por otro, el cosmopolitismo culinario que incentiva procesos de gentrificación alimentaria que evidencian los particularismos propios de la modernidad alimentaria.

Toxoplasma gondii is a zoonotic parasite globally infecting a wide range of species, including humans. Felids are the only known hosts that can excrete environmentally resistant oocysts into ecosystems. In boreal regions, Canada lynx (Lynx canadensis) are sought by hunters primarily for their fur, and they are occasionally eaten. We examined carcasses salvaged from trappers from boreal regions of eastern (n = 97) and western (n = 357) Canada. We detected T. gondii antibodies in fluid from thawed heart tissue using an enzyme-linked immunosorbent assay, DNA in brain and heart via magnetic capture and real-time PCR assay, and presence of DNA in feces using a real-time PCR with melt curve analysis. We detected antibodies against T. gondii and DNA in tissues in 24% and 19% of lynx, respectively. One lynx was positive for DNA of T. gondii in feces, which could indicate intestinal infection and potential for shedding oocysts. Our results indicate that lynx may be a useful sentinel species for monitoring environmental circulation of T. gondii in northern boreal regions and may pose a risk for transmission to other wildlife and to people handling or consuming lynx.

Northern Canada is warming at 3 times the global rate. Thus, changing diversity and distribution of vectors and pathogens is an increasing health concern. California serogroup (CSG) viruses are mosquitoborne arboviruses; wildlife reservoirs in northern ecosystems have not been identified. We detected CSG virus antibodies in 63% (95% CI 58%–67%) of caribou (n = 517), 4% (95% CI 2%–7%) of Arctic foxes (n = 297), 12% (95% CI 6%–21%) of red foxes (n = 77), and 28% (95% CI 24%–33%) of polar bears (n = 377). Sex, age, and summer temperatures were positively associated with polar bear exposure; location, year, and ecotype were associated with caribou exposure. Exposure was highest in boreal caribou and increased from baseline in polar bears after warmer summers. CSG virus exposure of wildlife is linked to climate change in northern Canada and sustained surveillance could be used to measure human health risks.

Atlantic salmon (Salmo salar) fed a carbohydrate-rich diet exhibit suboptimal growth performance, along with other metabolic disturbances. It is well known that gut microbes play a pivotal role in influencing metabolism of the host, and these microbes can be modified by the diet. The main goal of the present study was to determine the effect of feeding graded levels of digestible carbohydrates to Atlantic salmon on the distal intestine digesta microbiota at 3 sampling times (i.e., weeks 4, 8 and 12), during a 12-week trial. A low carbohydrate-to-high protein diet (LC/HP, 0% wheat starch), a medium carbohydrate-to-medium protein diet (MC/MP, 15% wheat starch) or a high carbohydrate-to-low protein diet (HC/LP, 30% wheat starch) was fed to triplicate fish tanks (27 to 28 fish per tank). We performed an in-depth characterization of the distal intestine digesta microbiota. Further, growth parameters, liver histology and the expression of genes involved in hepatic neolipogenesis in fish were measured. Fish fed a HC/LP diet showed greater hepatosomatic and viscerosomatic indexes (P = 0.026 and P = 0.018, respectively), lower final weight (P = 0.005), weight gain (P = 0.003), feed efficiency (P = 0.033) and growth rate (P = 0.003) compared with fish fed the LC/HP diet. Further, feeding salmon a high digestible carbohydrate diet caused greater lipid vacuolization, steatosis index (P = 0.007) and expression of fatty acid synthase (fas) and delta-6 fatty acyl desaturase (d6fad) (P = 0.001 and P = 0.001, respectively) in the liver compared with fish fed the LC/HP diet. Although, the major impact of feeding a carbohydrate-rich diet to Atlantic salmon in beta diversity of distal intestine digesta microbiota was observed at week 4 (HC/LP vs MC/MP and HC/LP vs LC/HP; P = 0.007 and P = 0.008, respectively) and week 8 (HC/LP vs MC/MP; P = 0.04), no differences between experimental groups were detected after 12 weeks of feeding. Finally, at the end of the trial, there was a negative correlation between lactic acid bacteria (LAB) members, including Leuconostoc and Lactobacillus, with hepatic steatosis level, the hepatosomatic and viscerosomatic indexes as well as the expression of fas and d6fad. Weissella showed negative correlation with hepatic steatosis level and the hepatosomatic index. Finally, further research to explore the potential use of LAB as probiotics to improve liver health in carnivorous fish fed fatty liver-induced diet is warranted.

Desde hace algunos años la gentrificación irrumpió en los estudios urbanos castellano hablantes como un concepto potente y polémico que revitalizó los debates sobre la ciudad. En ese contexto, el objetivo del presente trabajo consiste en analizar la manera en que se han desarrollado las investigaciones sobre gentrificación en México y España, a partir de un ejercicio comparativo que toma como referencia cuatro ejes de análisis: la localización central, la patrimonialización y el turismo, el desplazamiento y las resistencias. La metodología incluyó una amplia revisión bibliográfica de investigaciones académicas que tuvieron como argumento central la gentrificación. Entre los hallazgos de este documento se pueden mencionar convergencias ubicadas en la localización central de la gentrificación, así como en su relación con fenómenos de patrimonialización. Mientras que hubo divergencias en la manera en que se expresa el desplazamiento y las resistencias al proceso por parte de movimientos sociales.

Abstract Background In changing northern ecosystems, understanding the mechanisms of transmission of zoonotic pathogens, including the coccidian parasite Toxoplasma gondii, is essential to protect the health of vulnerable animals and humans. As high-level predators and scavengers, foxes represent a potentially sensitive indicator of the circulation of T. gondii in environments where humans co-exist. The objectives of our research were to compare serological and molecular assays to detect T. gondii, generate baseline data on T. gondii antibody and tissue prevalence in foxes in northern Canada, and compare regional seroprevalence in foxes with that in people from recently published surveys across northern Canada. Methods Fox carcasses (Vulpes vulpes/Vulpes lagopus, n = 749) were collected by local trappers from the eastern (Labrador and Québec) and western Canadian Arctic (northern Manitoba, Nunavut, and the Northwest Territories) during the winters of 2015–2019. Antibodies in heart fluid were detected using a commercial enzyme-linked immunosorbent assay. Toxoplasma gondii DNA was detected in hearts and brains using a magnetic capture DNA extraction and real-time PCR assay. Results Antibodies against T. gondii and DNA were detected in 36% and 27% of foxes, respectively. Detection of antibodies was higher in older (64%) compared to younger foxes (22%). More males (36%) than females (31%) were positive for antibodies to T. gondii. Tissue prevalence in foxes from western Nunavik (51%) was higher than in eastern Nunavik (19%). At the Canadian scale, T. gondii exposure was lower in western Inuit regions (13%) compared to eastern Inuit regions (39%), possibly because of regional differences in fox diet and/or environment. Exposure to T. gondii decreased at higher latitude and in foxes having moderate to little fat. Higher mean infection intensity was observed in Arctic foxes compared to red foxes. Fox and human seroprevalence showed similar trends across Inuit regions of Canada, but were less correlated in the eastern sub-Arctic, which may reflect regional differences in human dietary preferences. Conclusions Our study sheds new light on the current status of T. gondii in foxes in northern Canada and shows that foxes serve as a good sentinel species for environmental circulation and, in some regions, human exposure to this parasite in the Arctic. El propósito de esta investigación consistió en conocer los significados que los profesores de escuelas primarias atribuyeron a la docencia ejercida en tiempos de pandemia, en Puebla (México). El estudio tuvo un posicionamiento epistémico de corte fenomenológico-hermenéutico, metodología cualitativa y diseño centrado en el interaccionismo simbólico. La técnica de asociación libre fue utilizada para la recolección de la información. En los resultados los profesores relacionaron a la pandemia con las categorías: problema de salud pública, finitud humana y confinamiento como forma de vida. La docencia fue definida con las categorías: impacto laboral y personal, perfil laboral, formas de praxis docente y formas de educación a distancia. Se concluye que los profesores construyeron significados a su función con base en la definición de nuevas rutinas docentes derivadas de la diversificación de sus roles y de su adaptación a las demandas tecnológicas y pedagógicas, lo que hicieron estando inmersos en un marco de tensión, priorizando el valor de la vida, la salud, el compromiso ético-profesional con sus estudiantes y la trascendencia de la dimensión ontológica como el mayor baluarte social.

The most widely prescribed antidepressant, fluoxetine (FLX), is known for its antioxidant and anti-inflammatory effects when administered post-stress. Few studies have evaluated the effects of FLX treatment when chronic stress has induced deleterious effects in patients. Our objective was to evaluate FLX treatment (20 mg/kg/day, i.v.) once these effects are manifested, and the drug's relation to extracellular circulating microRNAs associated with inflammation, a hedonic response (sucrose intake), the forced swim test (FST), and corticosterone levels (CORT) and monoamine concentrations in limbic areas. A group of Wistar rats was divided into groups: Control; FLX; CUMS (for six weeks of exposure to chronic, unpredictable mild stress); and CUMS + FLX, a mixed group. After CUMS, the rats performed the FST, and serum levels of CORT and six microRNAs (miR-16, -21, -144, -155, -146a, -223) were analyzed, as were levels of dopamine, noradrenaline, and serotonin in the prefrontal cortex, hippocampus, and hypothalamus. CUMS reduced body weight, sucrose intake, and hippocampal noradrenaline levels, but increased CORT, immobility behavior on the FST, dopamine concentrations in the prefrontal cortex, and all miRNAs except miR-146a expression. Administering FLX during CUMS reduced CORT levels and immobility behavior on the FST and increased the expression of miR-16, -21, -146a, -223, and dopamine. FLX protects against the deleterious effects of stress by reducing CORT and has an antidepressant effect on the FST, with minimally-modified neurotransmitter levels. FLX increased the expression of miRNAs as part of the antidepressant effect. It also regulates both neuroinflammation and serotoninergic neurotransmission through miRNAs, such as the miR-16.

Se evaluó el crecimiento y rendimiento cárnico en pollos (Gallus gallus domesticus) F1 (Plumaje Normal y Cuello Desnudo × Rhode Island Red) bajo condiciones de traspatio y trópico seco. Para ello, se utilizaron 17 especímenes machos cuello desnudo (MCD) y 17 machos plumaje normal (MPN) criados bajo condiciones de traspatio, en San Nicolás Arroyo Hondo, municipio de Huetamo, Michoacán, México., bajo el esquema tradicional de alimentación (alimento comercial y maíz). Las variables analizadas Fenotipo-1 fueron: peso corporal (PC); consumo de alimento (CA); conversión alimenticia (Cal) y ganancia de peso (GP); así como, consumo de alimento total (CAt), peso al sacrificio (PS) y en canal (Pcan) y peso de las piezas de mayor valor comercial (PPMVC). La información del PC se analizó mediante un modelo en parcelas divididas con mediciones repetidas en el tiempo, con un modelo mixto. Las otras variables fueron analizadas con un modelo a una vía de clasificación con fenotipos como fuente de variación. En ambos casos, las diferencias entre fenotipos se obtuvieron a través de pruebas de “t” entre las medias de mínimos cuadrados. A higher Th17-immune response characterises obesity and obesity-related asthma phenotype. Nevertheless, obesity-related asthma has a more significant Th17-immune response than obesity alone. Retinoid-related orphan receptor C (RORC) is the essential transcription factor for Th17 polarisation. Previous studies have found that adolescents with obesity-related asthma presented upregulation of RORC, IL17A, and TNFA. However, the mechanisms that cause these higher mRNA expression levels in this asthmatic phenotype are poorly understood. Methylation directly regulates gene expression by adding a methyl group to carbon 5 of dinucleotide CpG cytosine. Thus, we evaluated the relationship between RORC, IL17A, and TNFA methylation status and mRNA expression levels to investigate a possible epigenetic regulation. A total of 102 adolescents (11–18 years) were studied in the following four groups: 1) healthy participants (HP), 2) allergic asthmatic participants (AAP), 3) obese participants without asthma (OP), and 4) non-allergic obesity-related asthma participants (OAP). Real-time qPCR assessed the methylation status and gene expression levels in peripheral blood leukocytes. Remarkably, the OAP and AAP groups have lower promoter methylation patterns of RORC, IL17A, and TNFA than the HP group. Notably, the OAP group presents lower RORC promoter methylation status than the OP group. Interestingly, RORC promoter methylation status was moderately negatively associated with gene expression of RORC (rs = −0.39, p < 0.001) and IL17A (rs = −0.37, p < 0.01), respectively. Similarly, the promoter methylation pattern of IL17A was moderately negatively correlated with IL17A gene expression (rs = −0.3, p < 0.01). There is also a moderate inverse relationship between TNFA promoter methylation status and TNFA gene expression (rs = −0.3, p < 0.01). The present study suggests an association between lower RORC, IL17A, and TNFA gene promoter methylation status with obesity-related asthma and allergic asthma. RORC, IL17A, and TNFA gene promoter methylation patterns are moderately inversely correlated with their respective mRNA expression levels. Therefore, DNA methylation may regulate RORC, IL17A, and TNF gene expression in both asthmatic phenotypes.

The implementation of a microbial fuel cell for wastewater treatment and bioenergy production requires a cost reduction, especially when it comes to the ion exchange membrane part and the catalysts needed for this purpose. Ionic liquids in their immobilized phase in proton exchange membranes and non-noble catalysts, as alternatives to conventional systems, have been intensively investigated in recent years. In the present study, a new microbial fuel cell technology, based on an ionic liquid membrane assembly for CoCu mixed oxide catalysts, is proposed to treat animal slurry. The new low-cost membrane–cathode system is prepared in one single step, thus simplifying the manufacturing process of a membrane–cathode system. The novel MFCs based on the new low-cost membrane–cathode system achieved up to 51% of the power reached when platinum was used as a catalyst. Furthermore, the removal of organic matter in suspension after 12 days was higher than that achieved with a conventional system based on the use of platinum catalysts. Moreover, struvite, a precipitate consisting of ammonium, magnesium, and phosphate, which could be used as a fertilizer, was recovered using this membrane–cathode system.

In this work, poly(vinyl chloride)-based polymeric ionic liquid inclusion membranes were used in the selective separation of Fe(III), Zn(II), Cd(II), and Cu(II) from hydrochloride aqueous solutions. The ionic liquids under study were 1-octyl-3-methylimidazolium hexafluorophosphate, [omim+][PF6−] and methyl trioctyl ammonium chloride, [MTOA+][Cl−]. For this purpose, stability studies of different IL/base polymer compositions against aqueous phases were carried out. Among all polymer inclusion membranes studied, [omim+][PF6−]/PVC membranes at a ratio of 30/70 and [MTOA+][Cl−]/PVC membranes at a ratio of 70/30 were able to retain up to 82% and 48% of the weight of the initial ionic liquid, respectively, after being exposed to a solution of metal ions in 1 M HCl for 2048 h (85 days). It was found that polymer inclusion membranes based on the ionic liquid methyl trioctyl ammonium chloride allowed the selective separation of Zn(II)/Cu(II) and Zn(II)/Fe(III) mixtures with separation factors of 1996, 606 and, to a lesser extent but also satisfactorily, Cd(II)/Cu(II) mixtures, with a separation factor of 112. Therefore, selecting the appropriate ionic liquid/base polymer mixture makes it possible to create polymeric inclusion membranes capable of selectively separating target metal ions.
