Abstract Understanding the long‐term effects of population and GDP changes requires a multisectoral and regional understanding of the coupled human‐Earth system, as the long‐term evolution of this coupled system is influenced by human decisions and the Earth system. This study investigates the impact of compounding economic and population growth uncertainties on long‐term multisectoral outcomes. We use the Global Change Analysis Model (GCAM) to explore the influence of compounding and feedback between future GDP and population growth on four key sectors: final energy consumption, water withdrawal, staple food prices, and CO2 emissions. The results show that uncertainties in GDP and population compound, resulting in a magnification of tail risks for outcomes across sectors and regions. Compounding uncertainties significantly impact metrics such as CO2 emissions and final energy consumption, particularly at the upper tail at both global and regional levels. However, the impact of staple food prices and water withdrawal depends on regional factors. Additionally, an alternative low‐carbon transition scenario could compound uncertainties and increase tail risk, particularly in staple food prices, highlighting the influence of emergent constraints on land availability and food‐energy competition for land use. The findings underscore the importance of considering and adequately accounting for compounding uncertainties in key drivers of multisectoral systems to enhance our comprehensive understanding of the complex nature of multisectoral systems. The paper provides valuable insights into the potential implications of compounding uncertainties.

Abstract Deep human‐Earth system uncertainties and strong multi‐sector dynamics make it difficult to anticipate which conditions are most likely to lead to higher or lower adoption of renewable energy, and models project a broad range of future solar and wind energy shares across future scenarios. To elucidate these dynamics, we explore a large data set of scenarios simulated from the Global Change Analysis Model (GCAM), and use scenario discovery to identify the most significant factors affecting solar and wind adoption by mid‐century. We generated a data set of over 4,000 scenarios from GCAM by varying 12 different socioeconomic factors at high and low levels, including assumptions about future energy demand, resource costs, and fossil fuel emissions paths, as well as specific technology assumptions including wind and solar backup requirements and storage costs. Using scenario discovery, we assess the most important factors globally and regionally in creating high fractions of solar and wind energy and explore interconnected effects on other systems including water and non‐CO2 emissions. Globally and regionally, we found that solar and wind‐related technology costs were the primary drivers of high wind and solar energy adoption, though a few regions depend heavily on other parameters like carbon capture and storage costs, population and gross domestic product trajectories, and fossil fuel costs. We also identify four key paths to high solar and wind energy by mid‐century and discuss their tradeoffs in terms of other outcomes.

Studies have found that understanding forest management is critical in understanding the interaction between the carbon cycle and the integrated human-Earth system. This makes effectively representing forest management decisions such as planting and harvesting important. Here, we implement a novel dynamic forest harvest model in a global state of the art multi-sector dynamics model, namely the Global Change Analysis Model (GCAM). We implement an approach that explicitly tracks forest age and generates rotation ages for forest harvest that are responsive to changes in wood prices, changes in forest age and regional preferences for forest rotation. Furthermore, the forest sector in GCAM competes for investment with other land use types in the future years based on expected profit. Our baseline scenario results indicate that with the new forest harvest model, the current global wood product demand in GCAM can be met with minimal loss of old growth forest through the age-based harvest decisions. We find that economic pressure for deforestation and consequent loss of forest carbon is a bigger driver of global forest change than wood harvests, especially in developing regions. Under alternative scenarios where an economic value is placed on carbon across the terrestrial and energy systems, while there is an increase in forest plantations, there can be corresponding decreases in forest cover in some regions as forest land competes with land for bio-energy crops. When the carbon in forests is assigned a price, we find that the average rotation age for wood harvests can be reduced across regions to harvest forests in a more carbon efficient manner.

There is significant uncertainty in how global water supply will evolve in the future, due to uncertain climate, socioeconomic, and land use change drivers and variability of hydrologic processes. It is critical to characterize the potential impacts of uncertainty in future water supply given its importance for food and energy production. In this work, we introduce a framework that integrates stochastic hydrology and human-environmental systems to characterize uncertainty in future water supply and its multisector impacts. We develop a global stochastic watershed model and demonstrate that this model can generate a large ensemble of realizations of basin-scale runoff with global coverage that preserves the mean, variance, and spatial correlation of a historical benchmark. We couple this model with a well-known human-environmental systems model to explore the impacts of runoff variability on the water and agricultural sectors across spatial scales. We find that the impacts of future hydrologic variability vary across sectors and regions. Impacts are felt most strongly in the water and agricultural sectors for basins that are expected to have unsustainable water use in the future, such as the Indus River basin. For this basin, we find that the variability in future irrigation water withdrawals and irrigated cropland increase over time due to uncertainty in renewable water supply. We also use the Indus basin to show how our stochastic ensemble can be leveraged to explore the global multisector consequences of local extreme runoff conditions. This work introduces a novel technique to explore the propagation of future hydrologic variability across human and natural systems and spatial scales.

Terrestrial plants sequester carbon through photosynthesis, and that carbon is eventually returned to the atmosphere through respiration by plants and soil microbes. Here the authors show a large, unexpected gap in estimations of these two carbon fluxes.

Abstract Land scarcity is increasing over time, driven by complex multisector dynamics. The impacts of land scarcity on the economy and environment are multi‐faceted and regional, so any action to convert land will contain inherent tradeoffs. These impacts are complicated by the deeply uncertain evolution of the various sectors influencing land scarcity. A need therefore exists to provide multi‐metric and multi‐sector assessments that are robust to myriad uncertainties. Land conservation effectively limits the supply of productive land, while biofuel consumption increases the demand and competition for that land, and how these dynamics individually and jointly propagate to economic and environmental impacts is an important open question. To address this, we adopt the Global Change Analysis Model (GCAM) that has representations of various important systems including the climate, macroeconomic, energy, agriculture and land, and water resources systems. Various scenarios of increased land demand (from biofuels) and decreased land supply (from conservation) under various socioeconomic scenarios drawn from the SSPs were simulated using GCAM. We find that while biofuel consumption and land conservation reduce carbon emissions, this comes at the cost of higher food prices, reduced crop production, and increased water withdrawals. Additionally, some regions experience these tradeoffs more severely than others and are more heavily impacted from the same biofuel mandate or by an additional percent of protected land. These and other findings highlight the importance of multisector modeling frameworks that capture many cross‐sector linkages, and acknowledge the important uncertainties confronting the human‐Earth system when making any analysis of land scarcity impacts.

Abstract This study seeks to understand how Argentina's energy, water, and land (EWL) systems will co‐evolve under a representative array of human and earth system influences, including socioeconomic change, climate change, and climate policy. To capture Argentina's sub‐national EWL dynamics in the context of global change, we couple the Global Change Analysis Model with a suite of consistent, gridded sectoral downscaling models to explore multiple stakeholder‐engaged scenarios. Across scenarios, Argentina has the economic opportunity to use its vast land resources to satisfy growing domestic and international demand for crops, such as oil (e.g., soy) and biomass. The human (rather than earth) system produces the most dominant changes in mid‐century EWL resource use. A Reference scenario characterized by modest socioeconomic growth projects a 40% increase in Argentina's agricultural production by 2050 (relative to 2020) by using 50,000 km2 of additional cropland and 40% more water. A Climate Policy scenario designed to achieve net‐zero carbon emissions globally shortly after mid‐century projects that Argentina could use 100,000 km2 of additional land (and 65% more water) to grow biomass and other crops. The burden of navigating these national opportunities and challenges could fall disproportionately on a subset of Argentina's river basins. The Colorado and Negro basins could experience moderate‐to‐severe water scarcity as they simultaneously navigate substantial irrigated crop demand growth and climate‐induced declines in natural water availability. Argentina serves as a generalizable testbed to demonstrate that multi‐scale EWL planning challenges can be identified and managed more effectively via integrated analysis of coupled human‐earth systems.

Abstract Simple climate models (SCMs) are computationally efficient and capable of emulating global mean output of more complex Earth system models (ESMs). In doing so, SCMs can play a critical role in climate research as stand‐ins for the computationally more expensive models, especially in studies involving low, spatial, and/or temporal resolution, providing more computationally efficient sources of climate data. Here we use Hector v2.5.0 to emulate the multiforcing historical and RCP scenario output for 31 concentration and seven emission‐driven ESMs. When calibrating Hector, sufficient calibration data must be used to constrain the model; otherwise, climate and/or carbon parameters affecting physical processes may be able to trade off with one another, allowing for solutions to use physically unreasonable fitted parameter values as well as limiting the application of the SCM as an emulator. We also present a novel methodology that uses the ESM range as a calibration data, which can be adopted when faced with missing variable output from a specific model.

Agricultural crop yields are susceptible to changes in future temperature, precipitation, and other Earth system factors. Future changes to these physical Earth system attributes and their effects on agricultural crop yields are highly uncertain. United States agricultural producers will be affected by such changes whether they occur domestically or internationally through international commodity markets. Here we present a replication study of previous investigations (with different models) showing that potential direct domestic climate effects on crop yields in the U.S. have financial consequences for U.S. producers on the same order of magnitude but opposite in sign to indirect financial impacts on U.S. producers from climate effects on crop yields elsewhere in the world. We conclude that the analysis of country-specific financial climate impacts cannot ignore indirect effects arising through international markets. We find our results to be robust across a wide range of potential future crop yield impacts analyzed in the multi-sector dynamic global model GCAM.

The increasing data requirements of complex models demand robust, reproducible, and transparent systems to track and prepare models’ inputs. Here we describe version 1.0 of the gcamdata R package that processes raw inputs to produce the hundreds of XML files needed by the GCAM integrated human-earth systems model. It features extensive functional and unit testing, data tracing and visualization, and enforces metadata, documentation, and flexibility in its component data-processing subunits. Although this package is specific to GCAM, many of its structural pieces and approaches should be broadly applicable to, and reusable by, other complex model/data systems aiming to improve transparency, reproducibility, and flexibility. Earth System Models (ESMs) are excellent tools for quantifying many aspects of future climate dynamics but are too computationally expensive to produce large collections of scenarios for downstream users of ESM data. In particular, many researchers focused on the impacts of climate change require large collections of ESM runs to rigorously study the impacts to both human and natural systems of low-frequency high-importance events, such as multi-year droughts. Climate model emulators provide an effective mechanism for filling this gap, reproducing many aspects of ESMs rapidly but with lower precision. The fldgen v1.0 R package quickly generates thousands of realizations of gridded temperature fields by randomizing the residuals of pattern scaling temperature output from any single ESM, retaining the spatial and temporal variance and covariance structures of the input data at a low computational cost. The fldgen v2.0 R package described here extends this capability to produce joint realizations of multiple variables, with a focus on temperature and precipitation in an open source software package available for community use (https://github.com/jgcri/fldgen). This substantially improves the fldgen package by removing the requirement that the ESM variables be normally distributed, and will enable researchers to quickly generate covarying temperature and precipitation data that are synthetic but faithful to the characteristics of the original ESM.

Agricultural yields are highly susceptible to changes in weather system patterns, including annual and sub-annual changes in temperature and precipitation. The impacts of future meteorological variable changes on crop yields have been widely studied in both empirical and process-based models. These changes in future yields can be used in economic models to adjust future crop yields or production functions to reflect the effects of changing weather conditions. This work presents an econometric approach that combines historical weather data with the biophysical growth cycles of maize, winter wheat, and soybean to predict the year-to-year weather-driven yield shocks for rainfed crops. Temperature and modeled soil moisture are taken as predictors, allowing testing of the fitted rainfed model’s ability to predict shocks to irrigated yields by assuming irrigation produces the yield-maximizing level of soil moisture. This approach enables prediction of the potential impacts of changing weather patterns on irrigated crops in areas that are currently primarily rainfed. We present the results of the empirical model, fitted with rainfed data; out-of-sample validation on irrigated crops; and projections of yield shocks under multiple future climate scenarios. Under a bias-corrected GFDL RCP8.5 scenario, this approach predicts the average of annual weather-induced yield shocks, relative to the average of 2006–2020 annual yield shocks, across U.S. counties in the 2040–2060 period of −17% and −13% for rainfed and irrigated maize, −19% and −18% for rainfed and irrigated soybean, and −4% and −2% for rainfed and irrigated winter wheat. Predicted changes in the 2070–2090 period are −30% and −29% for rainfed and irrigated maize, −33% for both rainfed and irrigated soybean, and −7% and −5%for rainfed and irrigated winter wheat. The annual yield shocks presented here will enable modeling of the economic consequences of extreme weather events and potential for irrigation to mitigate such events.

Huntington's disease (HD) is a fatal autosomal dominant neurodegenerative disorder caused by an expansion of the polyglutamine (polyQ) repeat in exon-1 in the Huntingtin gene (HTT). This results in misfolding and accumulation of the huntingtin (htt) protein, forming nuclear and cytoplasmic inclusions. HD is associated with dysregulation of gene expression as well as mitochondrial dysfunction. We hypothesized that by improving transcriptional regulation of genes necessary for energy metabolism, the HD motor phenotype would also improve. We therefore examined the protective effects of nicotinamide (NAM), a well-characterized water-soluble B vitamin that is an inhibitor of sirtuin1/class III NAD+-dependent histone deacetylase (HDAC). In this study, both mini-osmotic pumps and drinking water deliveries were tested at 250 mg NAM/kg/day, using the B6.HDR6/1 transgenic mouse model. Results were similar for both modes of delivery, and there was no evidence of toxicity. We found that NAM treatment increased mRNA levels of brain-derived neurotrophic factor (BDNF), and Peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α), the master regulator of mitochondrial biogenesis. Protein levels of BDNF were also significantly increased. In addition, NAM treatment increased PGC-1α activation in HD mice, pointing to a possible mode of action as a therapeutic. Critically, NAM treatment was able to improve motor deficits associated with the HD phenotype, tested as time courses of open field, rotarod, and balance beam activities. These improvements were substantial, despite the fact that NAM did not appear to reduce htt aggregation, or to prevent late-stage weight loss. Our study therefore concludes that NAM or similar drugs may be beneficial in clinical treatment of the motor dysfunctions of HD, while additional therapeutic approaches must be added to combat the aggregation phenotype and overall physiological decline.

Integrated assessment models (IAMs) hold great potential to assess how future agricultural systems will be shaped by socioeconomic development, technological innovation, and changing climate conditions. By coupling with climate and crop model emulators, IAMs have the potential to resolve important agricultural feedback loops and identify unintended consequences of socioeconomic development for agricultural systems. Here we propose a framework to develop robust representation of agricultural system responses within IAMs, linking downstream applications with model development and the coordinated evaluation of key climate responses from local to global scales. We survey the strengths and weaknesses of protocol-based assessments linked to the Agricultural Model Intercomparison and Improvement Project (AgMIP), each utilizing multiple sites and models to evaluate crop response to core climate changes including shifts in carbon dioxide concentration, temperature, and water availability, with some studies further exploring how climate responses are affected by nitrogen levels and adaptation in farm systems. Site-based studies with carefully calibrated models encompass the largest number of activities; however they are limited in their ability to capture the full range of global agricultural system diversity. Representative site networks provide more targeted response information than broadly-sampled networks, with limitations stemming from difficulties in covering the diversity of farming systems. Global gridded crop models provide comprehensive coverage, although with large challenges for calibration and quality control of inputs. Diversity in climate responses underscores that crop model emulators must distinguish between regions and farming system while recognizing model uncertainty. Finally, to bridge the gap between bottom-up and top-down approaches we recommend the deployment of a hybrid climate response system employing a representative network of sites to bias-correct comprehensive gridded simulations, opening the door to accelerated development and a broad range of applications.

Polychlorinated biphenyls (PCBs) are persistent environmental contaminants that are highly toxic to the developing nervous system, particularly via their disruption of dopamine (DA) function. In order to characterize the effects of PCBs on the developing basal ganglia DA system, we utilized an organotypic coculture system of developing rat striatum and ventral mesencephalon (VM). Exposure of the cocultures to an environmentally relevant mixture of PCBs for 1, 3, 7, or 14 days reduced tissue DA concentrations and increased medium levels of DA, homovanillic acid, and 3,4-dihydroxyphenylacetic acid. PCB exposure also increased neuronal cell death in both the VM and the striatum and reduced the number of DA neurons in the VM. Decreases in both tyrosine hydroxylase and DA transporter protein expression were shown by Western blot analysis in PCB-exposed cocultures. There was also an increase in neuronal cell death, identified by Fluoro Jade B, prior to a reduction in the number of VM DA neurons; we hypothesize this increase to be partly due to a loss of gamma-aminobutyric acid (GABA) neurons. Indeed, Western blot analysis revealed up to a 50% reduction in both VM and striatal glutamic acid decarboxylase 65/67. Analysis of tissue PCB levels revealed that concentrations were at or below 10 ppm following all exposure paradigms. This coculture system provides an excellent model to examine the chronology of PCB-induced neurotoxic events in the developing basal ganglia. Our results suggest that PCB-induced neurotoxicity in the developing basal ganglia involves GABAergic neuronal dysfunction, in addition to PCBs better-recognized effects on DA function. These findings have important implications for disease states such as Parkinsons disease and for developmental deficits associated with exposure to PCBs and toxicologically similar environmental contaminants.
