In line with the post-nonclassical educational paradigm, humanitarian practices, including support, are becoming necessary. Understanding the personal self-determination support as mediation implies the search for a “toolbox” that is not alienated from the personality. An individual development trajectory integrating the intra- and inter-subjective aspects of self-determination is considered to be the main support. The need to objectify and operationalize the supporting process actualizes the problem of theoretical justification and design for supporting procedure. We consider the supporting procedure as a well-founded procedure for the subjects’ actions. The procedure is provided by the identification of the supporting stages, a description of the subjects’ actions and their results. Supporting stages are isomorphic to the basic stages of self-determination and, accordingly, include: support at the stage of “self-determination itself”, support at the stage of self-realization and support at the stage of personal self-actualization. The teacher’s “functionality” includes actions aimed at activating the processes of the personality’s self-determination in its inter- and inter-subjective aspects: personality “immersion” in the activities using a wide arsenal of cultural mediators (word, image, action) and methods such as analysis of problem situations, game imitation actions, social communication and professional tests; facilitation of analysis, reflection and practical student’s actions. The student activities include: analysis of problem situations, “trial” activity of the person in modelled situations, activity in professional and living contexts; reflex of his actions. The coordinated nature of subjects’ activities is ensured not only by the use of appropriate toolkit, but also by facilitating character of their interaction.

Presentamos un análisis de la Sentencia T-719/2003 de la Corte Constitu- cional Colombiana (Corte) que con ponencia del magistrado Manuel José Cepeda tutela el derecho a la libertad y seguridad personal de la viuda y el hijo menor de edad de un reinsertado de la guerrilla de las autodenomina- das Fuerzas Armadas Revolucionarias de Colombia (FARC). El análisis pone en evidencia el alcance de este derecho en el Estado colombiano mientras el conflicto armado interno, abierto desde hace más de setenta años, provoca su constante vulneración; y describe la intervención de la Corte frente a la ineficacia de los programas de reinserción por la dilación burocrática guber- namental, en el marco de la cual desarrolla precedentes para la protección del derecho en mención, bajo un paradigma de interpretación evolutiva y creativa de la Constitución, afirmando los postulados del garantismo judi- cial. Consideramos que lo aquí planteado puede servir de referencia para la adopción de medidas concretas a favor de desmovilizados de la guerra ante un eventual éxito de las negociaciones de paz que actualmente lidera el go- bierno de Juan Manuel Santos con las FARC.

The mariner-like transposon Mos1 is used for insertional mutagenesis and transgenesis in different animals (insects, nematodes), but has never been used in plants. In this paper, the transposition activity of Mos1 was tested in Nicotiana tabacum, but no transposition event was detected. In an attempt to understand the absence of in planta transposition, Mos1 transposase (MOS1) was produced and purified from transgenic tobacco (HM NtMOS1). HM NtMOS1 was able to perform all transposition reaction steps in vitro: binding to ITR, excision and integration of the same pseudo-transposon used in in planta transposition assays. The in vitro transposition reaction was not inhibited by tobacco nuclear proteins, and did not depend on the temperature used for plant growth. Several hypotheses are proposed that could explain the inhibition of HM NtMOS1 activity in planta. [ABSTRACT FROM AUTHOR]

Genetic dissection of signaling pathways in mammalian cells involves screening or selecting phenotypic mutants obtained by a variety of techniques. Limitations in current methods include inadequate genome coverage and difficulty in validating the link between mutation and phenotype. We describe an improved method for insertional mutagenesis with retroviral vectors and show that the ability to induce mutations increases greatly if a randomly inserted promoter directs transcription into the host DNA. The mutant phenotype is due to the expression of a hybrid transcript derived from the vector and the insertion site. Because other alleles of the affected gene remain intact, the phenotype is dominant, but is reversible by inactivating the promoter. for example, by site-specific recombination. Importantly, in mutant clones with multiple inserts, limited excision yields progeny with different patterns of inserts remaining. Characterizing these progeny allows the mutant phenotype to be associated with a specific target gene. Relative simplicity and robust target validation make the method suitable for a broad range of applications. We have used this technique to search for proteins that regulate NE-κB- dependent signaling in human cells. Two validated targets are the relA gene, which codes for the NF-κR p65 subunit, and the NF-κB regulator act1. Overexpression of the corresponding proteins, caused by insertion of a promoter into the first intron of each gene, leads to NF-κB-dependent secretion of factors that activate NF-κB through cell-surface receptors, establishing an autocrine loop.
