Abstract Latest solutions in biotechnologies and biosensing targeted cold-active extremozymes. Analysis of acetaldehyde as a relevant quality indicator of wine is one example of application that could benefit from using low-temperatures operating catalysts. In search of novel aldehyde dehydrogenases (ALDH) with high stability and activity at low temperatures, the recombinant S2-ALDH from the Antarctic Flavobacterium PL002 was obtained by cloning and expression in Escherichia coli BL21(DE3). Structural and phylogenetic analyses revealed strong protein similarities (95%) with psychrophilic homologs, conserved active residues and structural elements conferring enzyme flexibility. Arrhenius plot revealed a conformational shift at 30 °C, favoring catalysis (low activation energy) at lower temperatures. In addition to a broad substrate specificity with preference for acetaldehyde (Km = 1.88 mM), this enzyme showed a high tolerance for ethanol (15%) and several salts and chelators (an advantage for wine analysis), while being sensitive to mercury (I50 = 1.21 µM). The neutral optimal pH (7.5) and the stability up to 40 °C and after lyophilization represent major assets for developing S2-ALDH-based sensors. An enzymatic electrochemical assay was developed for acetaldehyde detection in wines with proven accuracy in comparison with the reference spectrophotometric method, thus evidencing the potential of S2-ALDH as effective biocatalyst for industry and biosensing.

Objectives. The gastrointestinal neuroendocrin tumors are rare events with clinical presentation widely variable and surgical management that is often challenging. Material and methods. We performed a retrospective study in the First Surgical Clinic, St Spiridon University Hospital, “Grigore T. Popa” University of Medicine and Pharmacy, Iaşi, Romania, in the 2005-2019 period, which included all the patients diagnosed with gastrointestinal neuroendocrin tumors by immunehistochemistry. Results. There were 37 cases diagnosed with gastrointestinal neuroendocrin tumors. The ratio male/female was 15/22 and mean age was de 42±4.365 years old (range 27-79 years). The gastrointestinal neuroendocrin tumors were: 13 – gastric, one – duodenal, 10 cases – small intestine, 10 cases – appendicular, 7 cases – large intestine and hepatic metastases – 4 cases. The carcinoid syndrome was present in 7 cases. The biological diagnosis included biological markers (e.g. serotonine, 5-HIAA). Diagnosis of the tumor site and dimension was done by ultrasound exam, Computed Tomography scan, Positron Emission Tomography scan, Octreoscan and intraoperative ultrasonography. Surgical procedures for gastric neuroendocrin tumors were: wedge tumor resection – one case; subtotal gastrectomy – one case, total gastrectomies – 3 cases. For neuroendocrin tumors of small bowel we performed 6 enterectomies and 4 ileocolectomies with lymphadenectomy. We also performed 7 appendectomies and 3 right colectomies for appendicular carcinoids. We performed 4 right colectomies, 2 left colectomies and one low anterior resection of the rectum for colorectal neuroendocrin tumors. For neuroendocrin tumors with hepatic metastases disease we performed one hepatectomy and 3 termoablations. Conclusions. The gastrointestinal neuroendocrin tumors are rare tumors, and their management is always challenging. Immunohistochemistry is mandatory for confirmation, appreciation of the proliferation and biological behavior, and permissible to use specific therapy. Aggressive surgical treatment is indicated, even in advanced stages. The treatment in patients with advanced gastrointestinal neuroendocrin tumors with metastatic disease include chemotherapy, biological therapies, and peptide receptor radionuclide therapy.

Establishing a system that allows the transition from environmental and traffic monitoring to environmental management at the personal level has become a necessity. Such a system will improve the life quality, reduce health costs and increase the support of vulnerable groups (i.e., the elderly, children). ESTABLISH project aims to advance an innovative platform which allows converting environmental (sensor) data into actionable information for users to provide a healthier and safer environment thereby improving the quality of life. Smart adaptive services providing real-time feedback tailored to specific user and application needs will be developed by combining networked sensors and other data sources with adaptive models. This paper will present the on-going research within the ESTABLISH project related to the development of a decision support component which assesses the air quality. Based on real-time monitoring and quantification methods, and certain types of notifications for risk groups and general populations, the proposed system links pollutant concentrations to individual health risks. The proposed decision support for air quality assessment is structured on two components. The first component is represented by the comparison with the limit values provided by the legislation, and the second is the forecast of near-real-time air pollution episodes (based on trigger values). Thus, for each pollutant considered (PM2.5, PM10, SO2, NO2, O3, and CO) the frequency of pollutants concentrations measurement, the averaging periods according to the legislation, the averaging period and the limit values used for the notification component were established.

It is documented that maternal diseases or treatments influence a newborn's clinical status at birth. If a prenatal medical history is not available, or if signs or symptoms of a mother's disease are revealed for the first time during pregnancy or postpartum, their effects on the newborn may be misattributed. Objective: The objective of this study is to emphasize the paramount importance of prenatal care, for both mothers and newborns, as a lack of maternal signs and symptoms during pregnancy does not exclude a potential severe neonatal condition. Materials and methods: We present a series of three cases of pregnant women who gave birth to very sick preterm newborns that required admission to the Neonatal Intensive Care Unit (NICU). The mothers were asymptomatic during pregnancy and unaware of their subclinical disease. The newborns' complications, considered initially as consequences of prematurity or infection, subsequently revealed transient autoimmune disease in two of the cases (myasthenia gravis and hyperthyroidism) and a severe form of thrombophilia in the third case. Results: The newborns' diagnosis preceded maternal diagnosis and contributed to the identification of the maternal pathology; adequate treatment was prescribed, with favorable short- and long-term outcomes. Conclusions: Prenatal exams and investigations throughout pregnancy are a good opportunity to detect subclinical diseases or predispositions. As newborns usually develop non-specific signs, one should have experience and pay attention to differentiating among etiologies. Our paper takes a reversed approach to the usual medical diagnosis pathway: from infant to mother instead of from mother to infant, proving that inter-specialty collaboration can work bi-directionally. The relatively high vulnerability of young Europeans in the labour market compared to other age groups has led many to seek alternative employment solutions, such as entrepreneurship. While not a comprehensive solution, entrepreneurial initiatives among youth can offer a valuable opportunity for their integration into stable and decent work. This research uses Flash Eurobarometer 513—Social Entrepreneurship and Youth to explore entrepreneurial intentions among European Union youth. The study's novelty consists of the division of the target population into four distinct categories based on educational and employment status. The results indicate that EU youth are not a homogeneous group. While the motivation to create their own jobs—is a common feature of all young individuals with entrepreneurial intentions, suggesting a necessity-driven entrepreneurial intention, significant differences emerge among sub-groups. Young people who are only studying exhibit an interest in exploiting business opportunities, highlighting the role of education in fostering opportunity-driven entrepreneurship, while NEETs and employed youth are primarily motivated by better income prospects in the intention to start a business. Additionally, youth engaged in education and those integrated into the labour market are better informed about financing and training opportunities Perceived barriers include a lack of education for NEETs and financial risks for others. Funding preferences also differ: NEETs would rely on personal savings, while those balancing study and work would explore diverse sources, including venture capital and incubator support for starting businesses. Our study enhances the understanding of entrepreneurial intentions among EU young people, offering insights that can guide more effective support strategies for fostering entrepreneurial success. As national air quality networks and international research infrastructures, such as ACTRIS (EU) and ASCENT (US), continue to expand, the deployment of online aerosol chemistry measurements increases worldwide. These research infrastructures are focused on the ability to compare atmospheric properties from one region to another, making it crucial to understand instrument operation in various settings. This paper is part of a series of publications dedicated to better understanding the operation of these instruments using a series of laboratory tests. A particular focus was made on evaluating the organic aerosol (OA) measurement performance of six Aerosol Chemical Speciation Monitors (ACSMs) when sampling known mixtures of organic and inorganic aerosols and in ambient air. The study focuses on assessing the impact of instrument-to-instrument variability on ACSM data processing as well as identifying and quantifying the previously identified m/z44/NO3 artifacts that can affect the accuracy of the measurements. A high degree of variability was observed in instrument measurements of the m/z44/NO3 artifact when compared to results obtained two years earlier (e.g., an increase from ∼0 in 2016 to 0.16 in 2018 or a decrease from 0.12 in 2016 to 0.05 in 2018), confirming the need for frequent evaluation and quantification during calibration. This study underlines that the product between organic aerosol relative ionization efficiency and the instrument collection efficiency value is instrument dependent and that the variability in these values (1.78 ± 0.35) should be considered when estimating the measurement uncertainties. Using a range of specific compounds, an average RIEOA for levoglucosan (1.29 ± 0.23) close to the default value commonly used in ACSM was determined, obtaining a value more specific to each instrument. This study provides valuable information for the calibration and operation of ACSM instruments, ensuring that future studies can build on this work to evaluate and improve instrument performance. Background/Objectives: Hashimoto's thyroiditis (HT) is an autoimmune disorder characterized by chronic inflammation of the thyroid gland. Recent evidence indicates that the inflammation may extend beyond the thyroid. The study aims to explore the potential of complete blood count (CBC)-derived indices as markers of systemic inflammation in HT. Materials and Methods: This cross-sectional retrospective study from 1 January 2015, to 31 December 2023 included 147 pediatric HT patients and 144 apparently healthy controls. Thyroid profiles, antibodies, CBC, and protein electrophoresis data were collected from patient records. CBC-derived indices were calculated and compared between the HT and control groups, as well as among HT subgroups. Results: The median age of HT patients was 13.6 years (range: 11.2–15.5 years), with 66% being girls. The control group had a similar age and gender distribution, with a median age of 13.7 years (range: 11–15.8 years) and 70.8% girls. Of the HT patients, 50% had subclinical HT, 15% were euthyroid, and 34% had overt thyroid dysfunction. HT patients showed significantly higher neutrophil and lymphocyte counts, as well as all evaluated CBC-derived indices than controls (p < 0.001)). These differences were not significant among HT subgroups. Logistic regression indicated a strong association between an elevated neutrophil-to-lymphocyte ratio (NLR) and HT diagnosis (p < 0.001), while ROC analysis confirmed NLR as the most accurate CBC-derived marker for distinguishing HT from controls. Conclusions: Elevated NLR levels in pediatric HT patients provide additional evidence that inflammation may extend beyond the thyroid gland. These results support the potential of NLR as a reliable and accessible biomarker for evaluating inflammation in Hashimoto's thyroiditis. [ABSTRACT FROM AUTHOR]

A new non-sparking metallic material, Cu-Ti, with applications in potentially explosive environments is proposed as an alternative to CuBe, to reduce the processing and toxic effects of Be. Using high-purity Cu and Ti materials, a Cu (~3–4 wt%) Ti alloy with good chemical and structural homogeneity was fabricated in an induction furnace under an Ar atmosphere. The hot-rolled material was tested in an explosive gas mixture (10% H2 or 6.5% CH4) under extremely severe wear tests for 15,000 cycles, and no hot sparks were produced to ignite the medium. The material was investigated as hot-rolled plates (600 s at 950 °C and 10% reduction). The microstructures and surface of the wear test samples were investigated by light optical microscopy (LOM) and scanning electron microscopy (SEM). The chemical compositions were determined by energy dispersive spectrometry (EDS). The corrosion behavior was studied using electrochemical techniques: open-circuit, linear, and cyclic potentiometry in saline electrolyte solutions. The mechanical properties, such as microhardness and friction coefficient, were determined using UMT equipment. The results showed that the alloy is suitable for applications requiring non-ignition properties, with good hot rolling deformability and chemical composition homogeneity. Regarding the corrosion analysis and mechanical properties of the experimental CuTi alloy, minor differences were observed between the cast- and hot-rolled material. [ABSTRACT FROM AUTHOR]

The increasing frequency, severity, and complexity of cyber-attacks at EU level highlights the significant importance of robust cyber security frameworks. This article examines the role of EU regulations, in particular the Directive on measures for a high common level of cybersecurity across the Union (NIS 2 Directive) and the Cyber Resilience Act (CRA), in strengthening cyber resilience in critical sectors. First, the review of EU regulations was carried out through a comparative analysis of the legislative documents to highlight objectives, targeted entities, security measures, and compliance and clarify how they are designed to improve cyber resilience in critical sectors. Furthermore, the methodology includes an applied case study designed to explore the practical implications of European regulations on cyber resilience through the examination of the hybrid threat - multiple cyber-attacks mixed with influence operations through the cyberspace targeting the civilian and defense infrastructures of an essential port. By analyzing the attacks through the lens of the NIS 2 Directive and CRA, this case study illustrates how EU regulations provide an efficient foundation for strengthening security and response measures. It also identifies operational challenges and demonstrates how adaptable frameworks aligned with EU regulations enable organizations to recover more effectively. These findings contribute to the broader discourse on securing critical infrastructures and advancing practical solutions to cyber resilience in an interconnected digital environment. To further improve understanding and preparedness, future research should focus on organizing tabletop exercises (TTX) that simulate scenarios similar to those analyzed in this case study. In general, bark serves a protective role for trees and is genetically determined. The quantification of bark based on biometric characteristics is linked to studies on the distribution of forest species across the globe and vegetation fires. In Romania, on the other hand, the improvement of the wood traceability system requires an increase in the accuracy of the estimation of the biometric characteristics of bark and, implicitly, of the volume of wood under the bark. The aim of this study was to develop more precise models for predicting bark thickness along the stem of three key Romanian species, taking into account a comprehensive range of models and stem sections, including those with a diameter over bark smaller than 8 cm, which have been excluded in previous studies. The study is based on two datasets, one containing the national measurements of three commercially valuable forest species, i.e., Norway spruce (Picea abies (L.) Karst), European beech (Fagus sylvatica L.), and pedunculate oak (Quercus robur L.) from 12,186 trees, and a second dataset containing the measurements from 61 logs of the same species at a specific forest site. A set of seven double bark thickness (DBT) estimation models with stem diameter over bark (DOB), DOB and total tree height (H), DOB and relative height along the stem (h/H), and diameter over bark at breast height (DBH) and DOB as predictors were used. The DBT models were evaluated using the coefficient of determination (R2), mean absolute error (MAE), root mean squared error (RMSE), the Akaike information criterion (AIC), and the Bayesian information criterion (BIC). This led to the selection of two more accurate models, Model 2 (based on a third-degree polynomial) and Model 3 (based on a logarithmic function), with DOB as the predictor. Relative double bark thickness (RDBT) and proportion of bark area (PBA) were also estimated using a sixth-degree polynomial and relative height as a predictor variable after stratifying the data by DBH classes to reduce variability. The results of this study indicate that there is a need to complete the database, for all three forest species of commercial value in Romania especially for large trees with DBH greater than 60–70 cm. The models obtained for PBA are of great use to the industry and the economy, in particular in the context of the traceability of wood. This is due to the fact that PBA can be equated with the proportion of bark volume (PBV), which describes the variation in the proportion of bark in the volume of the wood assortments along the stem. For a given DBH, PBA and PBV demonstrate minimal variability in sections from the tree's base to a relative height of 0.6; however, a pronounced increase is observed at crown level in sections above relative heights of 0.8. In recent years, EU member states have faced two significant crises: the COVID-19 pandemic and high inflation, exacerbated by geopolitical instability. These events have led to a decline in household purchasing power. Our study aims to quantify the impact of these crises on household income dynamics in Romania, using data from the EU Survey on Income and Living Conditions and the EUROMOD tax-benefit microsimulation model. We analyze income changes across demographic segments from 2019 to 2021 (COVID-19 period) and 2021 to 2023 (inflation crisis), focusing on household characteristics such as the presence and number of children, the gender of the household head, and the presence of elderly members. Our findings reveal that while disposable income increased from 2019 to 2021, the poorest 10% experienced the smallest gains. In 2022–2023, inflation eroded these increases, and the lower half of the income distribution struggled with reduced earnings and pensions. Despite this, the Gini coefficient for disposable income decreased from 2019 to 2023, indicating a reduced inequality. Our study highlighted that targeted public policy interventions are essential to support vulnerable households and mitigate the effects of economic shocks. [ABSTRACT FROM AUTHOR]

Background and Objectives: The role of surgery in pancreatic metastases of renal cell carcinoma (PM_RCC) is highly controversial, particularly in the context of modern systemic therapies and the conflicting results of studies published so far. This study aims to explore a single surgical center experience (including mainly pancreatic resections) regarding the indications, the type of pancreatectomies, and early and long-term outcomes for PM_RCC. Materials and Methods: The data of all patients with surgery for PM_RCC (from 1 January 2002 to 31 December 2023) were retrospectively assessed, and potential predictors of survival were explored. Results: 20 patients underwent surgery for PM_RCC (pancreatectomies—95%). Metachronous PM_RCC was 90%, with a median interval between the initial nephrectomy and PM_RCC occurrence of 104 months. For elective pancreatectomies, the overall and severe morbidity and mortality rates were 24%, 12%, and 0%, respectively; 32% of patients underwent non-standardized pancreatic resections. The median survival of patients with negative resection margins was 128 months after pancreatectomies, with an 82% 5-year survival rate. Left kidney RCC and the body/tail PM_RCC were favorable prognostic factors for the overall survival after pancreatectomies for PM_RCC. Body/tail, asymptomatic PM_RCC, and an interval after initial nephrectomy > 2 were favorable prognostic factors for the overall survival after initial nephrectomy for RCC. Conclusions: Pancreatectomies for PM_RCC can achieve long-term survival whenever complete resection is feasible, with acceptable complication rates. Patients with left kidney RCC, body/tail, and asymptomatic PM_RCC and an interval of more than 2 years after nephrectomy exhibit the best survival rates. [ABSTRACT FROM AUTHOR]

The corrosion behavior of the TiTaNbZrAg alloys with different amounts of tantalum (0%, 10% and 20%) and with distinct surface topography (smooth and rough) was investigated in phosphate buffer solution (PBS) for long-time immersion (1000 h). By this approach, we expect to bring about new insights into the influence of both the amount of Ta in the alloy composition and the surface topography on the corrosion behavior of the Ti-based alloys. The corrosion resistance was studied by Open Circuit Potential (OCP), Potentiodynamic Polarization (PP), and Electrochemical Impedance Spectroscopy (EIS). From the potentiodynamic investigations, it was observed that all types of samples showed good corrosion resistance (i.e., Rcorr < 10 µm y−1) and may be used successfully for medical applications. However, the samples with smooth surfaces and with a certain amount of Ta (10% and 20%) exhibit the best corrosion performance (Rcorr < 1 µm y−1). As regards the samples with rough surfaces, the results evidenced that they showed lower corrosion resistance (1 < Rcorr < 3 µm y−1), suggesting that the Ta presence does not necessarily hinder the corrosion processes. Actually, the synergetic effect of both the presence of Ta and surface roughness plays an important role in corrosion resistance. [ABSTRACT FROM AUTHOR]

An ectopic thyroid (ET) involves numerous scenarios of detection and outcomes, while its current management is not standardised. A mediastinal ET (MET) represents a low index of suspicion. In this paper, we introduce a 47-year-old female who was accidentally identified with an MET, and a modern surgical approach was provided. An anterior mediastinal mass of 3.2 cm was found at CT upon a prior COVID-19 infection. Previous to the infection, she experienced non-specific complaints for a few months (intermittent night sweats, facial erythema, chest pressure, and dyspnoea). Also, CT identified a thymus-like mass and a left adrenal incidentaloma of 3 cm. The endocrine panel was normal, and the subject declined further investigations. She was re-admitted 12 months later: the MET had increased +1 cm (+45% volume) and was confirmed at a 99mTc pertechnetate scintigraphy. Noting the symptoms, mediastinal anatomy, and size change, the MET was removed via a minimally invasive trans-cervical approach (eutopic gland preservation) with the help of a Cooper thymectomy retractor (which also allowed for a synchronous thymus mass resection). No post-operatory complications were registered, the thyroid function remained normal, and the mentioned symptoms were remitted. A histological exam confirmed a benign MET and thymus hyperplasia, respectively. To conclude, this case pinpoints important aspects, such as the clinical picture became clear only upon thoracic surgery due to the complete remission of the complaints that initially seemed widely non-specific. The incidental MET finding was associated with a second (adrenal) incidentaloma, a scenario that might not be so rare, following multiple imaging scans amid the COVID-19 era (no common pathogenic traits have been identified so far). The co-presence of a thymus mass represented one more argument for surgery. Minimally invasive cervicotomy associated with eutopic gland conservation and the use of a Cooper thymectomy retractor highlight modern aspects in video-assisted thoracic surgery, which provided an excellent outcome, involving one of the lowest mediastinal thyroids to be removed by this specific procedure. Superior mesenteric artery aneurysms are a rare pathology, and rupture due to a pancreatic cyst in the context of alcohol-induced pancreatitis is an even rarer condition. The first line of treatment is usually an endovascular approach. We present the case of a 51-year-old male with alcohol-induced pancreatitis, diagnosed with a superior mesenteric artery aneurysm with active bleeding in close contact with a large pancreatic cyst. A covered stent was used to treat this condition. The patient developed hemorrhagic shock 12 h after the procedure and an urgent laparotomy was performed. A second rupture of the arterial wall at the distal end of the stent was observed and in order to obtain distal perfusion, first, an infrarenal aorta to superior mesenteric artery bypass distal to the rupture was performed in order to exclude the aneurysm. Secondly, a bypass originating from the distal end of the first bypass to the distal end of the superior mesenteric artery was performed. The patient had an uneventful recovery and was discharged after 10 days. We reviewed the literature regarding the incidence and the therapeutic management of superior mesenteric artery aneurysm complicated by pancreatic cyst. An advanced search on PubMed from 2004 to 2024 returned 194 results and after applying the inclusion–exclusion criteria, 11 publications were selected. Although the endovascular approach is usually the first line of treatment with obvious advantages for the patient, a patient-tailored approach should be made in such cases and surgery could be the first option, when considering that the mechanism of aneurysm rupture is due to erosion of the arterial wall by the pancreatic enzymes.
