Abstract

The rapid development of quantum dot (QD) devices for quantum computing has necessitated more efficient and automated methods for device characterization and tuning. This work demonstrates the feasibility and advantages of applying explainable machine learning techniques to the analysis of quantum dot measurements, paving the way for further advances in automated and transparent QD device tuning. Many of the measurements acquired during the tuning process come in the form of images that need to be properly analyzed to guide the subsequent tuning steps. By design, features present in such images capture certain behaviors or states of the measured QD devices. When considered carefully, such features can aid the control and calibration of QD devices. An important example of such images are so-called $\textit{triangle plots}$, which visually represent current flow and reveal characteristics important for QD device calibration. While image-based classification tools, such as convolutional neural networks (CNNs), can be used to verify whether a given measurement is $\textit{good}$ and thus warrants the initiation of the next phase of tuning, they do not provide any insights into how the device should be adjusted in the case of $\textit{bad}$ images. This is because CNNs sacrifice prediction and model intelligibility for high accuracy. To ameliorate this trade-off, a recent study introduced an image vectorization approach that relies on the Gabor wavelet transform (Schug $\textit{et al.}$ 2024 $\textit{Proc. XAI4Sci: Explainable Machine Learning for Sciences Workshop (AAAI 2024) (Vancouver, Canada)}$ pp 1-6). Here we propose an alternative vectorization method that involves mathematical modeling of synthetic triangles to mimic the experimental data. Abstract

This article argues that the immediate critical responses to the blockbuster film Barbie (Greta Gerwig, 2023), which diverged along ideological lines, fail to account for the extent to which the film undercuts the very ideological divisions that sustain them. Rather than or in addition to presenting a left-wing or right-wing critique of contemporary gender roles, the film positions this contest within the vexed relationship between the ideal and the real. This metaphysical quandary is what propels the protagonists on a Buddhist-inspired quest for authentic selfhood, a selfhood characterized by both the effacement of the discrete self (the Buddhist concept of anatman) and the eclipse of monumentalized cultural and commercial idols, like Barbie, that organize and disseminate ideology.

Abstract We perform readout of a quantum-dot hybrid qubit coupled to a superconducting resonator through a parametric, longitudinal interaction mechanism. Our experiments are performed with the qubit and resonator frequencies detuned by ~10 GHz, demonstrating that longitudinal coupling can facilitate semiconductor qubit operation in the ‘ultra-dispersive’ regime of circuit quantum electrodynamics.

21. Scaffolding Critical Reading

Abstract

Teachers in any discipline where reading matters should practice a robust scaffolding pedagogy to teach critical reading, in contrast to the more common but less direct approaches that often leave students to learn or not learn these skills themselves. In this essay, I describe how to adapt established methods for teaching writing (including templates) to teaching reading. To answer critics who might find the approach too "reductive," I turn to scaffolding theory, which calls for purposefully--but temporarily--reductive teaching. Finally, I present qualitative and quantitative evidence from three years of an American literature course to show how a scaffolding approach can help students read critically.

Abstract

Chronic osteomyelitis impedes bone union, requiring immediate management, such as removing the infected bone to aid regeneration. In this case, a 22-year-old woman with a 12 cm bone defect from chronic osteomyelitis had unsuccessful bone healing after three internal fixation surgeries due to persistent infection. To prevent complications, distraction osteogenesis with external fixation was chosen. This method involves applying mechanical strain to stimulate the biological response of damaged tissue, enhancing osteoconductivity and prompting new bone formation with rich blood supply. Despite the scarcity of reports on the effectiveness of this approach for long bone defects caused by osteomyelitis, This case study indicates that distraction osteogenesis using external fixation can be a viable treatment option for bone defects resulting from chronic osteomyelitis.

Abstract

ABSTRACT: The objective of this study was to examine the effect of stage of lactation (early, mid, and late) and proportion of pasture in the cow's diet (high: GRS, medium: PMR, and no: TMR) on the composition and quality of Cheddar cheese. Triplicate trials were carried out in each stage of lactation, and milk protein and fat contents were standardized for Cheddar cheese manufacture at pilot scale. Because cheese milks were standardized for milk fat and protein contents, gross composition did not differ as a result of diet. Fatty acid profiles of GRS cheese were significantly different from TMR, while PMR profiles were less distinct and more similar to both GRS and TMR profiles, as illustrated by partial least squares discriminatory analysis. Fatty acids including CLA C18:2 cis-9,trans-11, C22:1 n-9, and C18:3 n-3 were most influential in this separation of profiles. Fatty acid profiling revealed that GRS-derived cheese contained higher proportions of nutrients considered beneficial for human health including higher proportions of unsaturated fatty acids and omega-3 fatty acids. A biomarker model utilizing the proportions of 5 fatty acids was constructed and was effective at distinguishing between cheese of GRS, TMR, and PMR feeding systems. Proportions of ρ-κ-CN, αs2-CN and αs1-CN in cheese also differed among diets while proportions of ρ-κ-CN, αs1-CN, and β-CN were lowest in late lactation cheese. The effect of diet was less influential compared with that of stage of lactation on the ripening characteristics of cheese. An index of primary proteolysis was highest in late lactation cheese. The peptides derived from the proteolysis of κ-CN and β-CN and levels of secondary proteolysis, in particular, the proportions of 12 free AA were most influenced by stage of lactation. Overall this study demonstrated the effects of increasing pasture allowance and wstage of lactation on the nutritional quality and ripening properties of Cheddar cheese.

27. High School Enrollment Choices -- Understanding the STEM Gender Gap

Abstract

Students' high school decisions will always impact efforts to achieve gender parity in STEM at the university level and beyond. Without a comprehensive understanding of gendered disparities in high school course selection, it will be impossible to close completely the gender gap in many STEM disciplines. This study examines eleven years of detailed administrative data to determine gendered enrolment trends in university-stream secondary school STEM courses. Male and female enrolments for all publicly funded secondary schools across the province of Ontario (N = 844) were tracked from the 2007/08 academic year to 2017/18. The data reveal a clear trend of growing enrolment in STEM disciplines, with the increase in female students continuing their STEM education significantly outpacing males in almost all courses. However, these results also demonstrate the disparities that persist across STEM disciplines. The existing gender gap in physics remains large - in 2018, the median grade 12 physics class was only $36.5\pm0.05%$ female - with virtually no progress having been made to close this gap. By tracking individual student cohorts, we also demonstrate a newly discovered result showing the continuation rate of male students in biology stream courses has experienced a precipitous drop-off. The proportion of male students continuing from grade 10 science to grade 12 biology two years later has seen an average yearly decline of $-0.44\pm0.08$ percentage points, potentially foreshadowing the emergence of another significant gender gap in STEM. We suggest that researchers and educators cease treating STEM as a monolith when addressing gender disparities, as doing so obscures significant differences between disciplines. Abstract

Building a fault-tolerant quantum computer will require vast numbers of physical qubits. For qubit technologies based on solid state electronic devices, integrating millions of qubits in a single processor will require device fabrication to reach a scale comparable to that of the modern CMOS industry. Equally importantly, the scale of cryogenic device testing must keep pace to enable efficient device screening and to improve statistical metrics like qubit yield and voltage variation. Spin qubits based on electrons in Si have shown impressive control fidelities but have historically been challenged by yield and process variation. Here we present a testing process using a cryogenic 300 mm wafer prober to collect high-volume data on the performance of hundreds of industry-manufactured spin qubit devices at 1.6 K. This testing method provides fast feedback to enable optimization of the CMOS-compatible fabrication process, leading to high yield and low process variation. Using this system, we automate measurements of the operating point of spin qubits and probe the transitions of single electrons across full wafers. We analyze the random variation in single-electron operating voltages and find that the optimized fabrication process leads to low levels of disorder at the 300 mm scale. Together these results demonstrate the advances that can be achieved through the application of CMOS industry techniques to the fabrication and measurement of spin qubit devices., Comment: 33 pages, 4 figures, 10 extended data figures. Abstract Testicular cancer is a rare solid organ tumour associated with high cure rates and young age at diagnosis, hence it has a sizeable cohort of survivors worldwide. As it is one of the earliest tumours to be cured, a lot of studies have highlighted the late side effects of cancer and its different treatment modalities including surgery, radiotherapy and chemotherapy. While we are trying to identify the population at higher risk of platinum based chemotherapy and reduce its exposure, cisplatin based regimes remain an important tool to cure testicular cancer. The list of late side effects include a number of fatal and morbid conditions including but not limited to the second malignant neoplasms, cardiovascular disease, hypogonadism, infertility, metabolic syndrome, chronic respiratory disease, renal insufficiency, hearing loss, peripheral neuropathy, infertility and psychological illnesses like stress and anxiety. These complications eventually result in compromised social and economic health as well as lower life expectancy compared to the normal population. This article provides a comprehensive review of the latest data regarding the late side effects in testicular cancer survivors. A review of these conditions can help us develop recommendations and guidelines to improve the morbidity and mortality in survivors of testicular cancer.

Abstract

Evaluating the credibility of online information, a key component of digital literacy, is challenging for secondary students because they often rely on superficial strategies that do little to help them differentiate between information and disinformation. For example, our research has shown that students are prone to believe sites that appear professional but are in fact of a more nefarious nature (e.g., fake news sites designed to intentionally deceive). For this Description of Classroom Practices report, we describe a series of six lessons that we designed with secondary EAL teachers that help students think critically about online information while supporting their growth as additional language learners. We found that the lessons helped students evaluate the credibility of online sources holistically by evaluating within and across content, source, and context. After reporting the students’ overall positive comments about the materials, we describe their favourite activity in more detail with scaffolding suggestions.

31. Prioritizing Equitable Social Outcomes with and for Diverse Readers: A Conceptual Framework for the Development and Use of Justice-Based Reading Assessment

Abstract

Scholarship on the science of reading (SoR) has, in some instances, taken up more narrow views of reading in discussions and instantiations of reading assessment that do not center equity and justice, especially in schools. This can lead to less valid and even harmful reading assessment, especially for students from historically marginalized communities with diverse language, cultural, and neurological differences. Here, we draw on critically-minded reading research, as well as on work in equity-oriented educational assessment, to inform a justice-based reading assessment framework that can guide research, theory, policy, and practice. Using an equity-oriented and justice-based lens, the framework outlines three interwoven components: (1) relational and humanizing assessment practices; (2) justice-based products and outcomes; and, (3) a critical construct of reading. The framework compels designers, developers, and users to center the needs of rights-holders, and especially those from historically marginalized communities, throughout the assessment process. To do so, the framework outlines five principles that include orienting to equity and justice; prioritizing humanizing and critical assessment practices; grounding assessment in a complex, dynamic, and critical construct of reading for diverse populations; designing for justice-based social consequences, and engaging in critical debrief throughout. These principles guide eight phases of assessment, which we outline in detail. Finally, we discuss conceptual contributions as well as practical implications.

32. Human Rights 'Barbie': Using Critical Media Literacy to Teach about Feminism

Abstract

This article uses critical media literacy to approach themes of sexism and gender inequality depicted in the 2023 film "Barbie." Teachers cognizant of the need to address women's rights issues in American history often struggle to find resources that are accessible and meaningful for their students. Furthermore, the use of film in social studies instruction can be an engaging and culturally relevant way for students to grapple with contentious issues in social studies education. Additionally, by using human rights concepts to frame inequality and inspire advocacy, teachers can address dimensions of oppression and inequality with a more 'neutral' or objective framing. Given the current political climate that instills fear in educators who teach about issues of gender, strategic approaches can empower social studies teachers to engage students in examinations of this contentious topic in American schools. Lastly, this article introduces perspectives of Black feminists and international activists to provide intersectional analyses of women's rights issues to curb the common emphasis on White feminism. The lesson plan offered in this article includes a film viewing guide, enrichment opportunities, and an activity that connects themes in "Barbie" with women's rights movements.

33. The Taboo of Mental Health Problems, Stigma and Fear of Disclosure among Asian International Students: Implications for Help-Seeking, Guidance and Support

The current study explored the experiences of Asian International Students (AISs) in terms of mental health, disclosure and help-seeking within Higher Education in Scotland, UK. A qualitative study using individual semi-structured interviews with AISs (n = 20) was used and an inductive thematic approach to analysis was conducted. Three major themes were developed: (1) Negative beliefs, stigma and fear of judgment, (2) Adaptation and acculturation difficulties and (3) Barriers in communication, social disconnection and loneliness. Supporting AISs involves challenging negative judgements surrounding mental health, increasing mental health literacy and addressing barriers that may inhibit disclosure and help-seeking behaviour. The need for culturally sensitive mental health practitioners and awareness of diverse understandings of mental health issues is essential to improving support for AISs.

35. Cost-effective Amperometric Immunosensor for cardiac troponin I as a step towards affordable point-of-care diagnosis of acute myocardial infarction

Abstract

Early detection of cardiac troponin I in blood is vital for acute myocardial infarction diagnosis. A low-cost thin film gold electrode array was used with affordable ELISA antibodies and reagents to fabricate two cardiac troponin I amperometric immunosensors. The HRP-labelled sandwich immunocomplex was constructed on the gold electrode surface, and chronoamperometry was used to quantify cTnI indirectly by measuring the amount of TMB+ produced at the electrode surface. First, the system was evaluated using a physisorption approach to immobilise the capture antibody to the electrode with a 309 pg/mL LOD observed. Subsequently, a second sensor variant was produced using sulfo-LC-SPDP as a crosslinker to control antibody immobilisation, which resulted in an improved sensitivity with an LOD of 109 pg/mL. The chemisorption sensor outperformed the working range of the commercially available ELISA kit used (8000–125 pg/mL), demonstrating the power of enhanced antibody immobilisation and electrochemical detection for clinically relevant levels of cardiac troponin I. Amperometric immunosensors offer vital advantages including being cost-effective, simple to use, and compatible with commercially available reagents. These features make the sensor accessible to users and easy to manufacture. With further improvements to sensitivity and performance in complex samples, the sensor could be deployed to streamline acute myocardial infarction diagnosis and reduce the burden of chest pain patients on the healthcare system.

Abstract

Purpose: To evaluate the efficacy of prominent machine learning algorithms in predicting normal tissue complication probability using clinical data obtained from 2 distinct disease sites and to create a software tool that facilitates the automatic determination of the optimal algorithm to model any given labeled data set. Methods and Materials: We obtained 3 sets of radiation toxicity data (478 patients) from our clinic: gastrointestinal toxicity, radiation pneumonitis, and radiation esophagitis. These data comprised clinicopathological and dosimetric information for patients diagnosed with non-small cell lung cancer and anal squamous cell carcinoma. Each data set was modeled using 11 commonly employed machine learning algorithms (elastic net, least absolute shrinkage and selection operator [LASSO], random forest, random forest regression, support vector machine, extreme gradient boosting, light gradient boosting machine, k-nearest neighbors, neural network, Bayesian-LASSO, and Bayesian neural network) by randomly dividing the data set into a training and test set. The training set was used to create and tune the model, and the test set served to assess it by calculating performance metrics. This process was repeated 100 times by each algorithm for each data set. Figures were generated to visually compare the performance of the algorithms. A graphical user interface was developed to automate this whole process. Results: LASSO achieved the highest area under the precision-recall curve (0.807 ± 0.067) for radiation esophagitis, random forest for gastrointestinal toxicity (0.726 ± 0.096), and the neural network for radiation pneumonitis (0.878 ± 0.060). The area under the curve was 0.754 ± 0.069, 0.889 ± 0.043, and 0.905 ± 0.045, respectively. The graphical user interface was used to compare all algorithms for each data set automatically. When averaging the area under the precision-recall curve across all toxicities, Bayesian-LASSO was the best model. Conclusions: Our results show that there is no best algorithm for all data sets. Therefore, it is important to compare multiple algorithms when training an outcome prediction model on a new data set. The graphical user interface created for this study automatically compares the performance of these 11 algorithms for any data set.

A holistic view of wellbeing, which encompasses both physical and mental health, underscores their interconnected nature. This integrated approach reduces the artificial separation of wellbeing facets, and highlights how mental states influence not only individuals, but also their interactions with animals, the environment, and others in the workplace. Wellbeing challenges in veterinary medicine may contribute to negative impacts in animal, human, and environmental health. Veterinary education institutions and systems are also experiencing complex challenges as they adapt to rapidly changing societal, workforce, and professional wellbeing related pressures. This review paper explores the field of positive psychology and its application in educational contexts, commonly known as positive education. A thorough exploration of the systems-informed positive education approach and ways in which it can proactively enhance veterinary professional wellbeing from within the veterinary education ecosystem are presented. It is important to recognize that individual self-care, while valuable, cannot compensate for systemic dysfunctions such as poor team dynamics, ineffective leadership, or organizational culture issues. Addressing these systemic factors is critical for creating environments that support sustained flourishing. Positive psychology interventions delivered through the pathways of individuals, groups, and organizations specifically within a veterinary education context are discussed. Limitations, considerations, and proposed measurement strategies are reviewed. The implications of implementing a systems-informed positive psychology approach to enhance wellbeing in veterinary education include creating curriculum and cultures that enable flourishing within veterinary education institutions. Strengthening the individual and collective wellbeing of veterinary professionals has the potential to enhance the quality of care provided to animals, which has myriad positive implications for animal caregivers, their communities, the environment, and society.

Abstract

Summary: Neuronal populations expand their information-encoding capacity using mixed selective neurons. This is particularly prominent in association areas such as the lateral prefrontal cortex (LPFC), which integrate information from multiple sensory systems. However, during conditions that approximate natural behaviors, it is unclear how LPFC neuronal ensembles process space- and time-varying information about task features. Here, we show that, during a virtual reality task with naturalistic elements that requires associative memory, individual neurons and neuronal ensembles in the primate LPFC dynamically mix unconstrained features of the task, such as eye movements, with task-related visual features. Neurons in dorsal regions show more selectivity for space and eye movements, while ventral regions show more selectivity for visual features, representing them in a separate subspace. In summary, LPFC neurons exhibit dynamic and mixed selectivity for unconstrained and constrained task elements, and neural ensembles can separate task features in different subspaces.

Abstract

Summary: Cortical neurons in brain slices display intrinsic spike frequency adaptation (I-SFA) to constant current inputs, while extracellular recordings show extrinsic SFA (E-SFA) during sustained visual stimulation. Inferring how I-SFA contributes to E-SFA during behavior is challenging due to the isolated nature of slice recordings. To address this, we recorded macaque lateral prefrontal cortex (LPFC) neurons in vivo during a visually guided saccade task and in vitro in brain slices. Broad-spiking (BS) putative pyramidal cells and narrow-spiking (NS) putative inhibitory interneurons exhibit both E-SFA and I-SFA. Developing a data-driven hybrid circuit model comprising NS model neurons receiving BS input reveals that NS model neurons exhibit longer SFA than observed in vivo; however, adding feedforward inhibition corrects this in a manner dependent on I-SFA. Identification of this circuit motif shaping E-SFA in LPFC highlights the roles of both intrinsic and network mechanisms in neural activity underlying behavior.

Abstract

Summary: Background: Post-traumatic stress disorder (PTSD) and depression are common after mild traumatic brain injury (mTBI), but their biological drivers are uncertain. We therefore explored whether polygenic risk scores (PRS) derived for PTSD and major depressive disorder (MDD) are associated with the development of cognate TBI-related phenotypes. Methods: Meta-analyses were conducted using data from two multicenter, prospective observational cohort studies of patients with mTBI: the CENTER-TBI study (ClinicalTrials.gov ID NCT02210221) in Europe (December 2014–December 2017) and the TRACK-TBI study in the US (March 2014–July 2018). In both cohorts, the most common causes of injury were road traffic accidents and falls. Primary outcomes, specifically probable PTSD and depression, were defined at 6 months post-injury using scores ≥33 on the PTSD Checklist-5 and ≥15 on the Patient Health Questionnaire-9, respectively. We calculated PTSD-PRS and MDD-PRS for patients aged ≥17 years who had a Glasgow Coma Scale score of 13–15 upon hospital arrival and assessed their association with PTSD and depression following TBI. We also evaluated the transferability of the findings in a cohort of African Americans. Findings: Overall, 11.8% (219/1869) and 6.7% (124/1869) patients were classified as having probable PTSD and depression, respectively. Abstract

We hypothesized that pre-consult patient education videos can improve patient understanding about their diagnosis, lead to high satisfaction and low distress. In this pilot study, we developed a patient education video curriculum for patients with newly-diagnosed anal cancer. Comprehension of key content was evaluated by comparing pre- and post-test scores. Patient satisfaction scores were collected. Patient distress scores (0–10) were collected at the beginning of their consult visit prior to seeing the physician. We found that patient education videos prior to consult improved patient understanding, resulted in high patient satisfaction, and low patient distress at the time of consult.

Abstract

Purpose: Definitive intent radiation therapy (RT) for early-stage mucosa-associated lymphoid tissue (MALT) lymphoma typically includes a dose of 24 to 30 Gy. While modest, these doses may have associated toxicity. For patients with indolent B-cell lymphoma, there is increasing support for the use of ultra–low-dose RT (ULDRT) using 4 Gy in 2 fractions as part of a response-adapted approach, as high rates of complete response have been documented. This paradigm has been prospectively evaluated in the management of orbital and gastric indolent B-cell lymphomas; however, there is limited data guiding the use of ULDRT for lung MALT. Methods: We conducted a retrospective review of 20 patients at our institution with lung MALT treated with ULDRT as part of a response-adapted approach. Clinical variables including prior systemic therapy and symptoms were abstracted from the electronic health record. Responses were assessed using the revised Lugano criteria. Results: At a median follow up of 17 months following 4 Gy (IQR, 8-37 months), we observed 100% local control. Nineteen patients (95%) experienced a complete response. No patients with stage IE disease at RT (17/20; 85%) experienced distant progression. Nine patients (45%) were symptomatic prior to RT, with improvement or resolution of symptoms in 7 (7/9; 78%). One patient developed grade 2 pleuritic pain following RT, which resolved with a brief course of steroids. No other toxicities were noted. Conclusions: ULDRT, given in a response-adapted approach, is effective and well tolerated by patients with lung MALT.

Abstract

For United States of America (USA) and other developed countries, science achievement gaps begin to emerge in elementary and primary school. Such gaps between USA student groups typically are connected to socio-economic status (SES) and issues such as students still learning the English language. Through an experimental design, this National Science Foundation funded study explores how integrating the arts into science, technology, engineering, and mathematics (STEM) curriculum and leading with a more STEAMfirst approach (e.g., curriculum which integrates science, technology, engineering, arts, and mathematics) might provide more equitable science learning opportunities for elementary or primary grade level students. More specifically, the project's research efforts seek to also examine how integrating the arts into science instruction might help emerging bilingual (EB) students who are simultaneously learning the English language and science. Although results provide somewhat conflicting findings of statistical significance with small to moderate effect sizes, outcomes provide initial evidence that leading with STEAM science instruction before STEM efforts can be beneficial to early readers, and for EB students this benefit is magnified. As the title of this study suggest, sometimes finding nothing is something.

Abstract

Abstract A search for leptoquark pair production decaying into $$te^- \bar{t}e^+$$ t e - t ¯ e + or $$t\mu ^- \bar{t}\mu ^+$$ t μ - t ¯ μ + in final states with multiple leptons is presented. The search is based on a dataset of pp collisions at $$\sqrt{s}=13~\text {TeV} $$ s = 13 TeV recorded with the ATLAS detector during Run 2 of the Large Hadron Collider, corresponding to an integrated luminosity of 139 fb $$^{-1}$$ - 1 . Four signal regions, with the requirement of at least three light leptons (electron or muon) and at least two jets out of which at least one jet is identified as coming from a b-hadron, are considered based on the number of leptons of a given flavour. The main background processes are estimated using dedicated control regions in a simultaneous fit with the signal regions to data. No excess above the Standard Model background prediction is observed and 95% confidence level limits on the production cross section times branching ratio are derived as a function of the leptoquark mass. Under the assumption of exclusive decays into $$te^{-}$$ t e - ( $$t\mu ^{-}$$ t μ - ), the corresponding lower limit on the scalar mixed-generation leptoquark mass $$m_{\textrm{LQ}_{\textrm{mix}}^{\textrm{d}}}$$ m LQ mix d is at 1.58 (1.59) TeV and on the vector leptoquark mass $$m_{{\tilde{U}}_1}$$ m U ~ 1 at 1.67 (1.67) TeV in the minimal coupling scenario and at 1.95 (1.95) TeV in the Yang–Mills scenario.

Abstract

Culture-dependent and culture-independent microbiological methods are two approaches used to study microbial community composition. Culture-dependent methods have been the standard method used for many years but have limited utility with unculturable microorganisms. Culture-independent methods, including molecular techniques, enable direct analysis of microbial DNA without requiring cultivation. Both culture-dependent and -independent methods have roles in advancing our understanding of microbiology, and a combination of these approaches often yields a comprehensive depiction of the microbial diversity within a dynamic system. Bacterial activity reaction tests (BARTs) are a common culture-dependent test used to identify bacteria growing in industrial water samples. In this study, next-generation sequencing (NGS) was used to identify the taxa growing in BARTs and compared with the BART reaction patterns. Additionally, several water samples were analyzed by both BART and NGS analysis to determine whether the bacteria found in the water were also present in the BARTs. The results showed overall agreement between NGS and BARTs, though, in some cases, the most abundant taxa found in the water samples differed from those in the BARTs. This highlights the need for further study into the microbial community dynamics of culture-dependent tests to determine whether they are representative of the original sample.

Abstract

Both experiments and simulations with SOLPS-ITER and EDGE2D-EIRENE show that the onset of detachment for the low-field side (LFS) divertor – defined here as the line-averaged upstream density (〈ne〉edge) at which the plasma flux to the LFS target (ILFS−plate) starts to decrease with increasing 〈ne〉edge – is independent of the isotope mass. However, there are three major simulation-experiment discrepancies: (i) the absolute values of ILFS−plate and the electron density (ne) in the LFS divertor at the onset of detachment are significantly lower in simulations, i.e., approximately a factor of 2 for ILFS−plate and a factor of 3-4 for ne; (ii) the degree of detachment – defined here as the difference between ILFS−plate at the onset of detachment and at an 〈ne〉edge value close to the density limit – is smaller in simulations compared to experiments; and (iii) the experimentally observed larger degree of detachment for D and T plasmas compared to H plasmas cannot be clearly distinguished from the simulation results. There are strong indications that discrepancy (i) is to a large extent caused by neglecting Lyman-opacity effects in our simulations. The simulations predict a similar net volumetric recombination source for all isotopes due to the fact that molecule-activated recombination (MAR) compensates for the reduced electron–ion recombination (EIR) for H, whereas MAR is negligible for D and T. This similar net volumetric recombination source for all isotopes leads to an isotope-independent degree of detachment in simulations. An analysis of the Balmer-α and Balmer-γ emission confirms the underestimate of MAR in simulations (especially for D and T) for the JET metallic wall, which was previously observed for devices with a carbon wall. The underestimate of MAR is an important cause for discrepancy (ii) and the fact that there is a stronger underestimate of MAR for D and T than for H explains discrepancy (iii). Extending the plasma grid to the vessel wall increases ILFS−plate and ne at the onset of detachment by 25%, and the EIR source increases by 80% in detached conditions. Hence, while the extended grid results are closer to the experimental observations, the previously described qualitative discrepancies still persist.
