The oriental fruit fly, Bactrocera dorsalis is one of the most destructive pests of horticultural crops in tropical and subtropical Asia. The insect relies heavily on its olfactory system to select suitable hosts for development and reproduction. To understand the neural basis of its odor-driven behaviors, it is fundamental to characterize the anatomy of its olfactory system. In this study, we investigated the anatomical organization of the antennal lobe (AL), the primary olfactory center, in B. dorsalis, and constructed a 3D glomerular atlas of the AL based on synaptic antibody staining combined with computerized 3D reconstruction. To facilitate identification of individual glomeruli, we also applied mass staining of olfactory sensory neurons (OSNs) and projection neurons (PNs). In total, 64 or 65 glomeruli are identifiable in both sexes based on their shape, size, and relative spatial relationship. The overall glomerular volume of two sexes is not statistically different. However, eight glomeruli are sexually dimorphic: four (named AM2, C1, L2, and L3) are larger in males, and four are larger in females (A3, AD1, DM3, and M1). The results from anterograde staining, obtained by applying dye in the antennal lobe, show that three typical medial, media lateral, and lateral antennal-lobe tracts form parallel connections between the antennal lobe and protocerebrum. In addition to these three tracts, we also found a transverse antennal-lobe tract. Based on the retrograde staining of the calyx in the mushroom body, we also characterize the arrangement of roots and cell body clusters linked to the medial antennal-lobe tracts. These data provide a foundation for future studies on the olfactory processing of host odors in B. dorsalis.

In this study, we designed, simulated, and optimized proton exchanged integrated Mach-Zehnder modulators in a 0.5-μm-thick x-cut lithium niobate thin film. The single-mode conditions, the mode distributions, and the optical power distribution of the lithium niobate channel waveguides are discussed and compared in this study. The design parameters of the Y-branch and the separation distances between the electrodes were optimized. The relationship between the half-wave voltage length production of the electro-optic modulators and the thickness of the proton exchanged region was studied.

Sex pheromones are important for agricultural pest control. The main sex pheromone components of Spodoptera litura are (Z,E)-9,11- and (Z,E)-9,12-tetradecadienyl acetate (Z9,E11-14:Ac; Z9,E12-14:Ac). In this study, we investigated the optimal conditions for encapsulation of S. litura sex pheromonesin micelles via the self-assembly method using monomethoxy poly (ethylene glycol)-poly (ε-caprolactone) (MPEG-PCL) as a biodegradable wall-forming material with low toxicity. In the L9(3(4) orthogonal experiment, 3 amphiphilic block copolymers, with different hydrophilicity to hydrophobicity ratios, were examined. Optimal encapsulation conditions included stirring of MPEG5000-PCL2000 at 1000 rpm at 30°C with 2.5:1 wall-forming: core material mass ratio. S. litura sex pheromone-loaded MPEG5000-PCL2000 micelles presented a homogeneous spherical morphology with apparent core-shell structure. The release kinetics of optimized MPEG5000-PCL2000 micelles was best explained by a first-order model. Encapsulated Z9,E11-14:Ac and Z9,E12-14:Ac were released slowly, not suddenly. Methyl oleate (MO) was used as an agent to control micellar release performance. When MO content equalled block content, micelle half-life could be prolonged, thereby controlling the release speed. Overall, our results showed MPEG-PCL as a promising controlled-release substrate for sex pheromones.

SETD2 (SET domain containing protein 2) acts as a histone H3 lysine 36 (H3K36)-specific methyltransferase and may play important roles in active gene transcription in human cells. However, its expression and role in porcine oocytes and preimplantation embryos are not well understood. Here, we used immunofluorescence and laser scanning confocal microscopy to examine SETD2 expression in porcine fetal fibroblasts, oocytes, and preimplantation embryos derived from in vitro fertilization (IVF), parthenogenetic activation (PA), and somatic cell nuclear transfer (SCNT). In porcine fetal fibroblasts, SETD2 expression was detected in interphase cells, but not in M (mitotic)-phase cells. The SETD2 signal was observed in non-surrounded nucleolus (NSN)-stage oocytes, but not in surrounded nucleolus (SN)-, metaphase I (MI)-, or metaphase II (MII)-stage oocytes. The SETD2 signal was detectable in sperm, and undetectable immediately after fertilization, detectable at the 2-cell stage, and peaked at the 4-cell stage of IVF embryos in which porcine embryonic genome is activated. Similar to the pattern found in IVF embryos, the SETD2 signal was absent from PA embryos at the 1-cell stage, but it was detected at the 2-cell stage and thereafter maintained to the blastocyst stage. Interestingly, unlike the IVF and PA embryos, the SETD2 signal was detected throughout the development of SCNT embryos, including at the 1-cell stage. These data suggest that SETD2 may be functional for embryonic gene transcription in porcine preimplantation embryos. It is further speculated that the aberrant expression of SETD2 at the 1-cell stage of porcine SCNT embryos may be a factor in the low efficiency of cloning in pig.

Two dimensional-fluorescence difference gel electrophoresis (2D DIGE) is an emerging technique for comparative proteomics, which improves the reproducibility and reliability of differential protein expression analysis between samples. The purpose of this study was to investigate bovine pregnancy-specific proteins in the proteome between bovine pregnant and non-pregnant serum using DIGE technique. Serums of 2 pregnant Holstein dairy cattle at day 21 after artificial insemination and those of 2 non-pregnant were used in this study. The pre-electrophoretic labeling of pregnant and non-pregnant serum proteins were mixed with Cy3 and Cy5 fluorescent dyes, respectively, and an internal standard was labeled with Cy2. Labeled proteins with Cy2, Cy3, and Cy5 were separated together in a single gel, and then were detected by fluorescence image analyzer. The 2D DIGE method using fluorescence CyDye DIGE flour had higher sensitivity than conventional 2D gel electrophoresis, and showed reproducible results. Approximately 1,500 protein spots were detected by 2D DIGE. Several proteins showed a more than 1.5-fold up and down regulation between non-pregnant and pregnant serum proteins. The differentially expressed proteins were identified by MALDI-TOF mass spectrometer. A total 16 protein spots were detected to regulate differentially in the pregnant serum, among which 7 spots were up-regulated proteins such as conglutinin precursor, modified bovine fibrinogen and IgG1, and 6 spots were down-regulated proteins such as hemoglobin, complement component 3, bovine fibrinogen and IgG2a three spots were not identified. The identified proteins demonstrate that early pregnant bovine serum may have several pregnancy-specific proteins, and these could be a valuable information for the development of pregnancy-diagnostic markers in early pregnancy bovine serum.

The humeral heads of rabbits were replaced with individualized C/C composite implants coated with hydroxyapatite (HA) via rotation plasma spraying. The effect of the implants was evaluated after 5 and 150 days by using histology, scanning electron microscopy, X-ray diffraction, and so on. Results showed that the coating interface of C/C composites facilitated phosphorite deposition and osseous tissue growth. Furthermore, the humeral bone and remaining bone tissues were morphologically similar and possessed enhanced bonding strength. These results indicated that individualized C/C composite implants with HA coatings enabled the humeral head to adapt to dynamic requirements and accelerate healing after bone transplantation.

Borrelia burgdorferi, the causative agent of Lyme disease in humans, is exposed to reactive oxygen and nitrogen species (ROS and RNS) in both the tick vector and vertebrate reservoir hosts. B. burgdorferi contains a limited repertoire of canonical oxidative stress response genes, suggesting that novel gene functions may be important for protection of B. burgdorferi against ROS or RNS exposure. Here, we use transposon insertion sequencing (Tn-seq) to conduct an unbiased search for genes involved in resistance to nitric oxide, hydrogen peroxide, and tertiary-butyl hydroperoxide in vitro. The screens identified 66 genes whose disruption resulted in increased susceptibility to at least one of the stressors. These genes include previously characterized mediators of ROS and RNS resistance (including components of the nucleotide excision repair pathway and a subunit of a riboflavin transporter), as well as novel putative resistance candidates. DNA repair mutants were among the most sensitive to RNS in the Tn-seq screen, and survival assays with individual Tn mutants confirmed that the putative ribonuclease BB0839 is involved in resistance to nitric oxide. In contrast, mutants lacking predicted inner membrane proteins or transporters were among the most sensitive to ROS, and the contribution of three such membrane proteins (BB0017, BB0164, and BB0202) to ROS sensitivity was confirmed using individual Tn mutants and complemented strains. Further analysis showed that levels of intracellular manganese are significantly reduced in the Tn::bb0164 mutant, identifying a novel role for BB0164 in B. burgdorferi manganese homeostasis. Infection of C57BL/6 and gp91phox-/- mice with a mini-library of 39 Tn mutants showed that many of the genes identified in the in vitro screens are required for infectivity in mice. Collectively, our data provide insight into how B. burgdorferi responds to ROS and RNS and suggests that this response is relevant to the in vivo success of the organism.

Traditional saliency detection algorithms lack object semantic character, and the segmentation algorithms cannot highlight the saliency of the segmentation regions. In order to compensate for the defects of these two algorithms, the salient object segmentation model, which is a novel combination of two algorithms, is established in this paper. With the help of a priori knowledge of image boundary background traits, the K-means++ algorithm is used to cluster the pixels for each region; in line with the sensitivity of the human eye to color and with its attention mechanism, the joint probability distribution of the regional contrast ratio and spatial saliency is established. The selection of the salient area is based on the probabilities, for which the region boundary is taken as the initial curve, and the level-set algorithm is used to perform the salient object segmentation of the image. The curve convergence condition is established according to the confidence level for the segmented region, thus avoiding over-convergence of the segmentation curve. With this method, the salient region boundary is adjacent to the object contour, so the curve evolution time is shorter, and compared with the traditional Li algorithm, the proposed algorithm has higher segmentation evaluation scores, with the additional benefit of emphasizing the importance of the object.

Non-obstructive azoospermia is the most challenging type of male infertility. Stem cell based therapy provides the potential to enhance the recovery of spermatogenesis following cancer therapy. Bone marrow-derived mesenchymal stem cells (BMSCs) possess the potential to differentiate or trans-differentiate into multi-lineage cells, secrete paracrine factors to recruit the resident stem cells to participate in tissue regeneration, or fuse with the local cells in the affected region. In this study, we tested whether spermatogenically-induced BMSCs can restore spermatogenesis after administration of an anticancer drug. Allogeneic BMSCs were co-cultured in conditioned media derived from cultured testicular Sertoli cells in vitro, and then induced stem cells were transplanted into the seminiferous tubules of a busulfan-induced azoospermatic rat model for 8 weeks. The in vitro induced BMSCs exhibited specific spermatogonic gene and protein markers, and after implantation the donor cells survived and located at the basement membranes of the recipient seminiferous tubules, in accordance with what are considered the unique biological characteristics of spermatogenic stem cells. Molecular markers of spermatogonial stem cells and spermatogonia (Vasa, Stella, SMAD1, Dazl, GCNF, HSP90α, integrinβ1, and c-kit) were expressed in the recipient testis tissue. No tumor mass, immune response, or inflammatory reaction developed. In conclusion, BMSCs might provide the potential to trans-differentiate into spermatogenic-like-cells, enhancing endogenous fertility recovery. The present study indicates that BMSCs might offer alternative treatment for the patients with azoospermatic infertility after cancer chemotherapy.

The objective of this work was to investigate the possibility of using Na-bentonite as a raw material to produce aluminum salt-modified bentonite (AlMB) for desilication of green liquor. Batch experiments were carried out by varying the experimental parameters to find the optimum conditions. The optimum adsorbent dose was 20 g/L. The maximum adsorptions of 96.37% and 87.22% took place at pH 7.4 when the initial concentrations of silicate in green liquor were 4 and 6 g/L, respectively. The kinetic models showed that the process included physical adsorption and chemical adsorption. Finally, this study showed that the extent of desorption was 56.78% for silicate after desorption of AlMB in 2 M NaOH solution. The AlMB is an efficient and novel adsorbent for desilicating that can provide a good reference for the “silicon influence” in a papermaking mill.

ABSTRACT Yokota Fumiko’s novel, Hakujitsu No Sho, was a candidate for the third Akutagawa Prize in the first half of 1936. Two years later in 1938, it was published in the opening issue of the Japanese literary and art magazine Manchuria Romantic, which was influential during the Manchukuo period. However, in China this novel has not been adequately researched in academic circles. Centered on the relationship between Hakujitsu No Sho and Manchuria Romantic, this paper explains the essence of the novel and the literary attitude of the author by analyzing the text in detail. Through this lesbian narrative, the social anxiety of the 1930s in Japan is symbolically represented and, at the same time, Yokota Fumiko hinted at her veiled rebellion against society and the times. Progressing from a young Leftist literary girl to a colleague of Japanese Romanticism after eight years in “Manchukuo”, although it was hard for Yokota Fumiko to avoid following the trend, there is a clear difference between the “romance” in her works and the “grand romance” advocated by Kitamura Kennjirou, the editor of Manchuria Romantic. The “romance” in her works has always maintained a certain distance from the center of literary world and embodied her rebellion against the times.

Leptospirosis, caused by Leptospira, is one of the most important of neglected emerging zoonotic diseases that has important impacts on public health worldwide. Polyclonal antibody (pcAb) therapy is a potential method to process a series of pathogens for which there are limited determination of treatment, such as leptospirosis. First, we evaluated the efficacy of pcAb, derived from the sera of rabbits inoculated with Leptospira, against homotype (Leptospira interrogans serovar Lai) or heterotype (Leptospira interrogans serovar Autumnalis) Leptospira infection in a lethal hamster model. The pcAb treatment improved survival compared to the controls. The histopathology's of the infected kidney, liver and lung were also examined by hematoxylin and eosin staining. Using real-time quantitative PCR, we determined that most of the leptospires in the primary organs were almost completely removed by pcAb. In the second experiment, treatments, including antibiotic, pcAb, and combination, were started immediately after occurrence of the first serious sickness mouse in any group. No significant difference in survival rate between pcAb group and antibiotic group was found, but the combination therapy group significantly improved survival rate compared to the others (P

The Lyme disease spirochete Borrelia burgdorferi encounters a wide range of environmental conditions as it cycles between ticks of the genus Ixodes and its various mammalian hosts. Reactive oxygen species (ROS) and reactive nitrogen species (RNS) are potent antimicrobial molecules generated during the innate immune response to infection, however, it is unclear whether ROS and RNS pose a significant challenge to B. burgdorferi in vivo. In this study, we screened a library of B. burgdorferi strains with mutations in DNA repair genes for increased susceptibility to ROS or RNS in vitro. Strains with mutations in the methyl-directed mismatch repair (MMR) gene mutS1 are hypersensitive to killing by ROS, while strains lacking the nucleotide excision repair (NER) gene uvrB show increased susceptibility to both ROS and RNS. Therefore, mutS1-deficient and uvrB-deficient strains were compared for their ability to complete their infectious cycle in Swiss Webster mice and I. scapularis ticks to help identify sites of oxidative and nitrosative stresses encountered by B. burgdorferi in vivo. Both mutS1¬ and uvrB were dispensable for infection of mice, while uvrB promoted the survival of spirochetes in I. scapularis ticks. The decreased survival of uvrB-deficient B. burgdorferi was associated with the generation of RNS in I. scapularis midguts and salivary glands during feeding. Collectively, these data suggest that B. burgdorferi must withstand cytotoxic levels of RNS produced during infection of I. scapularis ticks.

The smart district heating system (SDHS) is an important element of the construction of smart cities in Northern China; it plays a significant role in meeting heating requirements and green energy saving in winter. Various Internet of Things (IoT) sensors and wireless transmission technologies are applied to monitor data in real-time and to form a historical database. The accurate prediction of heating loads based on massive historical datasets is the necessary condition and key basis for formulating an optimal heating control strategy in the SDHS, which contributes to the reduction in the consumption of energy and the improvement in the energy dispatching efficiency and accuracy. In order to achieve the high prediction accuracy of SDHS and to improve the representation ability of multi-time-scale features, a novel short-term heating load prediction algorithm based on a feature fusion long short-term memory (LSTM) model (FFLSTM) is proposed. Three characteristics, namely proximity, periodicity, and trend, are found after analyzing the heating load data from the aspect of the hourly time dimension. In order to comprehensively utilize the data’s intrinsic characteristics, three LSTM models are employed to make separate predictions, and, then, the prediction results based on internal features and other external features at the corresponding moments are imported into the high-level LSTM model for fusion processing, which brings a more accurate prediction result of the heating load. Detailed comparisons between the proposed FFLSTM algorithm and the-state-of-art algorithms are conducted in this paper. The experimental results show that the proposed FFLSTM algorithm outperforms others and can obtain a higher prediction accuracy. Furthermore, the impact of selecting different parameters of the FFLSTM model is also studied thoroughly.

Spatial information can be integrated into almost all fields of industrial ecology. Many researchers have shown that spatial proximity affects a variety of behaviors and interactions, and thus matters for materials stocks and flows analysis. However, normal tools or models in industrial ecology based on temporal dependence cannot be simply applied to the case of spatial dependence. This paper proposes a framework integrating material stocks and flows analysis with spatial analysis. We argue that spatial analysis can help data management and visualization, determine spatio-temporal patterns-processes-drivers, and finally develop dynamic and spatially explicit models, to improve the performance of simulating and assessing stocks and flows of materials. Scaling in spatial, temporal, and organizational dimensions and other current limitations are also discussed. Combined with spatial analysis, industrial ecology can really be more powerful in achieving its origin and destination—sustainability.

Sampling for DUS test of flower colors should be fixed at the stages and sites that petals are fully colored, and besides, flower colorations are uniform among individuals and stable for a period of time to allow testers to get consistent results. It remains a problem since spatial and temporal flower colorations are reported a lot but their change traits are little discussed. In this study, expression state, uniformity and stability of color phenotypes, anthocyanin contents, and gene expression levels were taken into account based on measurements at 12 development stages and three layers (inner, middle, and outer petals) of two varieties of Ranunculus asiaticus L. to get their best sampling. Our results showed that, outer petals of L9⁻L10 (stage 9⁻stage 10 of variety ‘Jiaoyan zhuanhong’) and C5⁻C6 (stage 5⁻stage 6 of variety ‘Jiaoyan yanghong’) were the best sampling, respectively. For DUS test, it is suggested to track flower colorations continuously to get the best sampling as well as representative colors since different cultivars had different change traits, and moreover, full expression of color phenotypes came later and lasted for a shorter duration than those of anthocyanin contents and gene expressions. Our innovation exists in following two points. Firstly, a model of change dynamic was introduced to illustrate the change traits of flower colorations, anthocyanin contents, and gene expressions. Secondly, genes used for expression analysis were screened on account of tentative anthocyanins, which were identified based on comparison between liquid chromatography⁻mass spectrometry (LC⁻MS) results and molecular mass and mass fragment pattern (M2) of each putative anthocyanin and their fragments deduced in our previous study. Gene screening in this regard may also be interest for other non-model plant genera with little molecular background.

Mammalian oocytes and early embryos derived from in vitro production are highly susceptible to a variety of cellular stresses. During oocyte maturation and preimplantation embryo development, functional proteins must be folded properly in the endoplasmic reticulum (ER) to maintain oocyte and embryo development. However, some adverse factors negatively impact ER functions and protein synthesis, resulting in the activation of ER stress and unfolded protein response (UPR) signaling pathways. ER stress and UPR signaling have been identified in mammalian oocytes and embryos produced in vitro, suggesting that modulation of ER stress and UPR signaling play very important roles in oocyte maturation and the development of preimplantation embryos. In this review, we briefly describe the current state of knowledge regarding ER stress, UPR signaling pathways, and their roles and mechanisms in mammalian (excluding human) oocyte maturation and preimplantation embryo development.

Fine-scale, accurate intra-urban functional zones (urban land use) are important for applications that rely on exploring urban dynamic and complexity. However, current methods of mapping functional zones in built-up areas with high spatial resolution remote sensing images are incomplete due to a lack of social attributes. To address this issue, this paper explores a novel approach to mapping urban functional zones by integrating points of interest (POIs) with social properties and very high spatial resolution remote sensing imagery with natural attributes, and classifying urban function as residence zones, transportation zones, convenience shops, shopping centers, factory zones, companies, and public service zones. First, non-built and built-up areas were classified using high spatial resolution remote sensing images. Second, the built-up areas were segmented using an object-based approach by utilizing building rooftop characteristics (reflectance and shapes). At the same time, the functional POIs of the segments were identified to determine the functional attributes of the segmented polygon. Third, the functional values—the mean priority of the functions in a road-based parcel—were calculated by functional segments and segmental weight coefficients. This method was demonstrated on Xiamen Island, China with an overall accuracy of 78.47% and with a kappa coefficient of 74.52%. The proposed approach could be easily applied in other parts of the world where social data and high spatial resolution imagery are available and improve accuracy when automatically mapping urban functional zones using remote sensing imagery. It will also potentially provide large-scale land-use information.

ABSTRACT In many bacteria, the FtsH protease and its modulators, HflK and HflC, form a large protein complex that contributes to both membrane protein quality control and regulation of the cellular response to environmental stress. Both activities are crucial to the Lyme disease pathogen Borrelia burgdorferi, which depends on membrane functions, such as motility, protein transport, and cell signaling, to respond to rapid changes in its environment. Using an inducible system, we demonstrate that FtsH production is essential for both mouse and tick infectivity and for in vitro growth of B. burgdorferi. FtsH depletion in B. burgdorferi cells resulted in membrane deformation and cell death. Overproduction of the protease did not have any detectable adverse effects on B. burgdorferi growth in vitro, suggesting that excess FtsH does not proteolytically overwhelm its substrates. In contrast, we did not observe any phenotype for cells lacking the protease modulators HflK and HflC (ΔHflK/C), although we examined morphology, growth rate, growth under stress conditions, and the complete mouse-tick infectious cycle. Our results demonstrate that FtsH provides an essential function in the life cycle of the obligate pathogen B. burgdorferi but that HflK and HflC do not detectably affect FtsH function. IMPORTANCE Lyme disease is caused by Borrelia burgdorferi, which is maintained in nature in an infectious cycle alternating between small mammals and Ixodes ticks. B. burgdorferi produces specific membrane proteins to successfully infect and persist in these diverse organisms. We hypothesized that B. burgdorferi has a specific mechanism to ensure that membrane proteins are properly folded and biologically active when needed and removed if improperly folded or dysfunctional. Our experiments demonstrate that FtsH, a protease that fulfills this role in other microorganisms, is essential to B. burgdorferi viability. Cells depleted of FtsH do not survive in laboratory culture medium and cannot colonize mice or ticks, revealing an absolute requirement for this protease. However, the loss of two potential modulators of FtsH activity, HflK and HflC, does not detectably affect B. burgdorferi physiology. Our results provide the groundwork for the identification of FtsH substrates that are critical for the bacterium’s viability.

Positive transcription elongation factor b (P-TEFb) is a RNA polymerase II carboxyl-terminal domain (Pol II CTD) kinase that phosphorylates Ser2 of the CTD and promotes the elongation phase of transcription. Despite the fact that P-TEFb has role in many cellular processes, the role of this kinase complex remains to be understood in mammalian early developmental events. In this study, using immunocytochemical analyses, we found that the P-TEFb components, CDK9, Cyclin T1 and Cyclin T2 were localized to nuclear speckles, as well as in nucleolar-like bodies in pig germinal vesicle oocytes. Using nascent RNA labeling and small molecule inhibitors, we showed that inhibition of CDK9 activity abolished the transcription of GV oocytes globally. Moreover, using fluorescence in situ hybridization, in absence of CDK9 kinase activity the production of ribosomal RNAs was impaired. We also presented the evidences indicating that P-TEFb kinase activity is essential for resumption of oocyte meiosis and embryo development. Treatment with CDK9 inhibitors resulted in germinal vesicle arrest in maturing oocytes in vitro. Inhibition of CDK9 kinase activity did not interfere with in vitro fertilization and pronuclear formation. However, when in vitro produced zygotes were treated with CDK9 inhibitors, their development beyond the 4-cell stage was impaired. In these embryos, inhibition of CDK9 abrogated global transcriptional activity and rRNA production. Collectively, our data suggested that P-TEFb kinase activity is crucial for oocyte maturation, embryo development and regulation of RNA transcription in pig.

This study was designed to find out an optimised planting system of reducing non-point (source) pollution by analyzing the reasons and the factors of influence non-point pollution in farmland of Erhai Lake basin. The results showed that incomes, residual nitrogen in soil, and the loss of nitrogen in surface water in rice-garlic system were higher than those in rice-fava bean system. There were positive correlations between the nitrogen loss of farmland, nitrogen inputs, residual nitrogen in soil, and incomes of farmland. Economic benefits and environment benefits are both appropriate, if the area of rice-garlic system would be reduced to 53% and the area of rice-fava bean system increased to 36% of total cropping area in the investigated watershed. Adjustment of planting structure and introduction of reasonable rotation systems is considered an effective measure of controlling agricultural non-point pollution in watersheds of Erhai Lake.

With the current state-of-the-art computer aided manufacturing tools, the spatial resolution of hyperspectral sensors is becoming increasingly higher thus making it easy to obtain much more detailed information of the scene captured. However, the improvement of the spatial resolution also brings new challenging problems to address with signal dependent photon noise being one of them. Unlike the signal independent thermal noise, the variance of photon noise is dependent on the signal, therefore many denoising methods developed for the stationary noise cannot be applied directly to the photon noise. To make things worse, both photon and thermal noise coexist in the captured hyperspectral image (HSI), thus making it more difficult to whiten noise. In this paper, we propose a new denoising framework to cope with signal dependent nonwhite noise (SDNW), Pre-estimate—Whitening—Post-estimate (PWP) loop, to reduce both photon and thermal noise in HSI. Previously, we proposed a method based on multidimensional wavelet packet transform and multi-way Wiener filter which performs both white noise and spectral dimensionality reduction, referred to as MWPT-MWF, which was restricted to white noise. We get inspired from this MWPT-MWF to develop a new iterative method for reducing photon and thermal noise. Firstly, the hyperspectral noise parameters estimation (HYNPE) algorithm is used to estimate the noise parameters, the SD noise is converted to an additive white Gaussian noise by pre-whitening procedure and then the whitened HSI is denoised by the proposed method SDNW-MWPT-MWF. As comparative experiments, the Multiple Linear Regression (MLR) based denoising method and tensor-based Multiway Wiener Filter (MWF) are also used in the denoising framework. An HSI captured by Reflective Optics System Imaging Spectrometer (ROSIS) is used in the experiments and the denoising performances are assessed from various aspects: the noise whitening performance, the Signal-to-Noise Ratio (SNR), and the classification performance. The results on the real-world airborne hyperspectral image HYDICE (Hyperspectral Digital Imagery Collection Experiment) are also presented and analyzed. These experiments show that it is worth taking into account noise signal-dependency hypothesis for processing HYDICE and ROSIS HSIs.

This paper introduces a novel double-differential vector phase-locked loop (DD-VPLL) for Global Navigation Satellite Systems (GNSS) that leverages carrier phase position solutions as well as base station measurements in the estimation of rover tracking loop parameters. The use of double differencing alleviates the need for estimating receiver clock dynamics and atmospheric delays; therefore, the navigation filter consists of the baseline dynamic states only. It is shown that using vector processing for carrier phase tracking leads to a significant enhancement in the receiver sensitivity compared to using the conventional scalar-based tracking loop (STL) and vector frequency locked loop (VFLL). The sensitivity improvement of 8 to 10 dB compared to STL, and 7 to 8 dB compared to VFLL, is obtained based on the test cases reported in the paper. Also, an increased probability of ambiguity resolution in the proposed method results in better availability for real time kinematic (RTK) applications.

Active power steady-state security regions (APSSRs), which can provide guidance for prevention and control through security checks, is of great importance for the safe operation of power systems in which more and more sustainable energy power generation is integrated. As a mature flexible AC transmission system (FACTS) device, thyristor-controlled series compensators (TCSCs) can carry out series compensation for the transmission line by controlling its equivalent reactance. With the change of the equivalent reactance parameter of a TCSC, the nodal admittance matrix and power flow distribution of the power system also changes. Inevitably, the APSSR will be different. Therefore, it is necessary and important to further incorporate the equivalent reactance parameters of TCSCs in the APSSR expression, which is generally established in the space of node active power injections. In this paper, a rapid construction method of APSSRs incorporating the equivalent reactances of TCSCs is proposed. Firstly, applicability and efficiency of the conventional APSSR construction method for power systems with TCSCs are analyzed. Further, with equivalent disconnection of TCSC branches, the effect of TCSC equivalent reactances on the distribution of active power flow through changing the structure parameters is treated as modifying node active power injections. On this basis, explicit expressions of APSSRs with a single TCSC equivalent reactance parameter and double TCSC equivalent reactance parameters are derived, respectively. Moreover, by deducing the general formula of APSSRs with multiple TCSC equivalent reactance parameters, the feasibility of the proposed method for power systems with multiple TCSCs is analyzed. Eventually, via benchmarks with different scales and a different number of TCSCs, validity and superiorities of the proposed method in computational efficiency are demonstrated.

The S-CO2 Brayton cycle system has many characteristics such as low cost and compact structure, and is one of the hotspots in the fields of waste heat utilization, new energy and so on. In this paper, a mathematical model of the S-CO2 simple Brayton cycle is constructed, and the cyclic characteristics are analysed. The relationship between the component parameters of the system and the calculation parameters of the cycle such as the cycle efficiency and the output net work are obtained under the design conditions. Meanwhile, calculation models of turbine and compressor under off-design conditions are established, and the characteristics of the whole cycle are analyzed. Thus, the characteristics of off-design conditions of the simple cycle are obtained. This study provides some reference for the related research of the S-CO2 Brayton cycle and the practical engineering application.

Accurate detection and effective control strategy of commutation failure (CF) of high voltage direct current (HVDC) are of great significance for keeping the safe and stable operations of the hybrid power grid. At first, a novel detection method for consecutive CF is proposed. Concretely, the 2nd and higher orders’ derivative values of direct current are summarized as the core to judge CF by analyzing the physical characteristics of the direct current waveform of the converter station in CF. Then, the Daubechies wavelet coefficient that can represent the 2nd and higher order derivative values of direct current is derived. Once the wavelet coefficients of the sampling points are detected to exceed the threshold, the occurrence of CF is confirmed. Furthermore, by instantly increasing advanced firing angle β in the inverter side, an additional emergency control strategy to prevent subsequent CF is proposed. Eventually, with simulations of the benchmark model, the effectiveness and superiorities of the proposed detection method and additional control strategy in accuracy and rapidity are verified.

With the aid of power quality monitoring systems (PQMSs), accurate locating of voltage sag sources, which has important significance for guiding maintenance personnel in finding and repairing faults as well as improving power supply reliability, has been becoming a new research hotspot. However, existing methods have unsatisfactory locating accuracy due to the integration of distributed generators (DGs) and fault resistance. In this paper, a novel strategy for accurately locating voltage sag sources in smart distribution networks is proposed. Based on inverse theory, which is well applied in geophysics, the accurate location issue is treated as a two-step optimization model. It aims at making the distribution of voltage phasors and current phasors obtained by theoretical short-circuit calculation match those actually observed as closely as possible. To guarantee the feasibility of the strategy, the effect of inverter-interfaced DGs (IIDGs) which are the main form of DG is considered in the short-circuit calculations. To guarantee the location accuracy of the strategy, fault resistance is treated as an optimization variable in the two-step optimization model to eliminate estimation error of fault resistance. Via two modified IEEE benchmarks with different scales, the validity and the superiorities in applicability and accuracy of the proposed strategy are verified.

Harmonic resonance may cause abnormal operation and even damage of power facilities, further threatening normal and safe operation of power systems. For renewable energy generations, controlled loads and parallel reactive power compensating equipment, their operating statuses can vary frequently. Therefore, the parameters of equivalent fundamental and harmonic admittance/impedance of these components exist in uncertainty, which will change the elements and eigenvalues of harmonic network admittance matrix. Consequently, harmonic resonance in power grid is becoming increasingly more complex. Hence, intense research about prevention and suppression of harmonic resonance, particularly the parameter feasible domain (PFD) which can keep away from harmonic resonance, are needed. For rapid online evaluation of PFD, a novel method without time-consuming pointwise precise eigenvalue computations is proposed. By analyzing the singularity of harmonic network admittance matrix, the explicit sufficient condition that the matrix elements should meet to prevent harmonic resonance is derived by the extended Gersgorin theorem. Further, via the non-uniqueness of similar transformation matrix (STM), a strategy to determine the appropriate STM is proposed to minimize the conservation of the obtained PFD. Eventually, the availability and advantages in computation efficiency and conservation of the method, are demonstrated through four different scale benchmarks.

ABSTRACT The Lyme disease spirochete Borrelia burgdorferi migrates to distant sites in the tick vectors and mammalian hosts through robust motility and chemotaxis activities. FliH and FliI are two cytoplasmic proteins that play important roles in the type III secretion system (T3SS)-mediated export and assembly of flagellar structural proteins. However, detailed analyses of the roles of FliH and FliI in B. burgdorferi have not been reported. In this study, fliH and fliI transposon mutants were utilized to dissect the mechanism of the Borrelia type III secretion system. The fliH and fliI mutants exhibited rod-shaped or string-like morphology, greatly reduced motility, division defects (resulting in elongated organisms with incomplete division points), and noninfectivity in mice by needle inoculation. Mutants in fliH and fliI were incapable of translational motion in 1% methylcellulose or soft agar. Inactivation of either fliH or fliI resulted in the loss of the FliH-FliI complex from otherwise intact flagellar motors, as determined by cryo-electron tomography (cryo-ET). Flagellar assemblies were still present in the mutant cells, albeit in lower numbers than in wild-type cells and with truncated flagella. Genetic complementation of fliH and fliI mutants in trans restored their wild-type morphology, motility, and flagellar motor structure; however, full-length flagella and infectivity were not recovered in these complemented mutants. Based on these results, disruption of either fliH or fliI in B. burgdorferi results in a severe defect in flagellar structure and function and cell division but does not completely block the export and assembly of flagellar hook and filament proteins. IMPORTANCE Many bacteria are able to rapidly transport themselves through their surroundings using specialized organelles called flagella. In spiral-shaped organisms called spirochetes, flagella act like inboard motors and give the bacteria the ability to bore their way through dense materials (such as human tissue) in a corkscrew manner. In this article, we studied how two proteins, called FliH and FliI, are important for the production of full-length flagella in the Lyme disease spirochete Borrelia burgdorferi. Mutants with defective production of FliH and FliI have reduced flagellar length and motility; this deficiency in turn affects many aspects of B. burgdorferi's biology, including the ability to undergo cell division and cause disease in mammals. Using a microscopic computed tomography (CT) scan approach called cryo-electron tomography, the structure that contains FliH and FliI was defined in the context of the flagellar motor, providing clues regarding how this amazing nanomachine is assembled and functions.

To improve the fuel consumption of a motor grader, a method for shifting the power curve of the grader engine is proposed. The working principle, calculation method, and design procedure of the novel method are depicted. Considering the nonlinear and time-varying uncertainty of the load of a motor grader, the fuzzy adaptive control is provided to shift the power curve. The shifting action is decided by the gears and the corresponding engine speed together with the displacements of both the hydraulic pump and motor. As a result, the power line adapts to the frequent changes in different external loads. A prototype motor grader is developed to test the above-mentioned energy conservation method in different working conditions. The results show that the fuel consumption of the prototype motor is reduced by 23%–34% and the skip ratio is decreased by 10% compared with the traditional grader with only one power curve. The proposed shifting power curve method can be widely applied to other heavy-load construction machinery also.

Discussion on the role of DEHP in the critical period of gonadal development in pregnant rats (F0), studied the evolution of F1-F4 generation of inter-generational inheritance of cryptorchidism and the alteration of DNA methylation levels in testis. Pregnant SD rats were randomly divided into two groups: normal control group and DEHP experimental group. From pregnancy 7 d to 19 d, experimental group was sustained to gavage DEHP 750 mg/kg bw/day, observed the incidence of cryptorchidism in offspring and examined the pregnancy rate of female rats through mating experiments. Continuous recording the rat's weight and AGD value, after maturation (PND80) recording testis and epididymis' size and weight, detected the sperm number and quality. Subsequently, we examined the evolution morphological changes of testicular tissue for 4 generation rats by HE staining and Western Blot. Completed the MeDIP-sequencing analysis of 6 samples (F1 generation, F4 generation and Control). DEHP successfully induced cryptorchidism occurrence in offspring during pregnancy. The incidence of cryptorchidism in F1 was 30%, in F2 was 12.5%, and there was no cryptorchidism coming up in F3 and F4. Mating experiment shows conception rate 50% in F1, F2 generation was 75%, the F3 and F4 generation were 100%. HE staining showed that the seminiferous epithelium of F1 generation was atrophy and with a few spermatogenic cell, F2 generation had improved, F3 and F4 generation were tend to be normal. The DNA methyltransferase expression was up-regulated with the increase of generations by Real Time-PCR, immunohistochemistry and Western Blot. MeDIP-seq Data Analysis Results show many differentially methylated DNA sequences between F1 and F4. DEHP damage male reproductive function in rats, affect expression of DNA methyltransferase enzyme, which in turn leads to genomic imprinting methylation pattern changes and passed on to the next generation, so that the offspring of male reproductive system critical role in the development of imprinted genes imbalances, and eventually lead to producing offspring cryptorchidism. This may be an important mechanism of reproductive system damage.

A novel chitosan hydrogel with high porosity was fabricated by a crosslinking method. Cartilage tissue engineering formed after mesenchymal stem cells was cultured on this hydrogel scaffold for 12 weeks. The immunohistochemistry tests demonstrated that the obtained cartilage had the specific histological properties of natural cartilage. And the qPCR tests also proved that the genes for type II collagen in the obtained cartilage were expressed the same as in the natural one.
