With the global demand for lipid compounds on the rise, concerns are growing about the environmental and economic impact of traditional lipid sources. This concern is exacerbated by the ever‐increasing demand for plant‐based lipids, which is contributing to unsustainable production practices and competition for land and food. Considering this challenge, this work aimed at exploring the potential of black soldier fly larvae (BSFL) as a low‐cost and ecofriendly source of lipid classes. A fractionation scheme consisting of a mixture of polar and nonpolar solvents at different ratios was employed to isolate the lipid classes from BSFL oil using silica gel column chromatography, which is a conventional method of chromatography. The fraction's separation efficiency was validated using thin‐layer chromatography and characterized with Fourier‐transformed infrared spectroscopy and gas chromatography flame ionization (GC‐FID). Triglycerides (30%) were found to be the most abundant component while cholesterols (0.6%) were the least abundant lipid fractions in the lipid mixture. GC‐FID in the various lipid fractions analyzed, lauric acid exhibited the highest percentage among the triglycerides (16.64%), diglycerides (19.10%), monoglycerides (22.70%), and free fatty acids (27.65%) fractions. We synthesized a novel, structured lipid containing caprylic acid at its sn‐1,3 position and docosahexaenoic acid at its sn‐2 position (1,3C‐2D‐TAG) by using a two‐step enzymatic method and then investigated the relationship between the types of fatty acids in the structured lipid and their physiological functions. Furthermore, we compared the effects of similarly structured lipids on postprandial lipid metabolism and obesity protection. The results revealed that the novel structured lipid synthesized using the enzymatic method not only exhibited better physical properties than traditional oils but also had high oxidation stability and crystallization degree. In terms of physiological function, the low‐dose 1,3C‐2D‐TAG group exhibited higher cholesterol and triglyceride levels, lower kidney weight or body weight, and higher serum aspartate aminotransferase and blood urea nitrogen levels than control group, whereas the high‐dose 1,3C‐2D‐TAG group exhibited higher triglyceride levels. Moreover, the medium‐dose 1,3C‐2D‐TAG group had remarkably higher high‐density lipoprotein cholesterol levels and lower low‐density lipoprotein cholesterol levels than the high‐lipid, control, and 1,2,3C‐TAG groups; however, the cholesterol and triglyceride levels and kidney and liver functions did not differ significantly among these groups. The study results suggest that 1,3C‐2D‐TAG can not only facilitate the effective utilization of the energy supplied by medium‐chain fatty acids but also help overcome difficulties in the absorption of long‐chain polyunsaturated fatty acids. In recent decades, plants have strengthened their relevance as sources of molecules potentially beneficial for health. This underpinning effect also arises from the extensive research that has been conducted on plants that are typically undervalued, besides being scarcely used. This is the case with Camellia japonica in Galicia (NW Spain), where, despite its abundance, it is exclusively used for ornamental purposes and has been studied only for its proximate composition. Thus, the present study was conducted on several additional parameters in the flowers of eight C. japonica varieties. Our results show that camellia has a high nutritional value, with carbohydrates as the most abundant macronutrients followed by a moderate protein content (4.4–6.3 g/100 g dry weight) and high levels of polyunsaturated fatty acids (especially ω-3 fatty acids, which represent 12.9–22.7% of the total fatty acids), raising its potential for use for nutritional purposes. According to the thermochemical characterization and elemental composition of camellia, the raw material has poor mineralization and low nitrogen content, but high percentages of volatile matter and high carbon-fixation rates, making it a promising alternative for biofuel production. Furthermore, preliminary analysis reveals a high concentration of different bioactive compounds. Lipid injectable emulsions have been in clinical use for over 60 years. The first product launched was Intralipid, which consisted of an emulsion of soybean oil in water for intravenous administration. It was a key source of essential fatty acids and an alternative source of energy for patients with gastrointestinal dysfunction requiring long‐term parenteral nutrition. With clinical experience, a condition known as parenteral nutrition–associated liver disease (PNALD), or intestinal failure–associated liver disease (IFALD), was observed, with a focus on carbohydrate and fat energy. Modifying the daily doses and infusion rates had some salutary effects, but PNALD persisted. Subsequently, on closer inspection of the fatty acids profile and phytosterol concentrations, degradation products arising from chemical and physical stability issues of the available lipid injectable emulsions were implicated. Recently, the US Food and Drug Administration convened an online workshop entitled "The Role of Phytosterols in PNALD/IFALD," with an emphasis on (1) the multifactorial pathophysiology of PNALD/IFALD, (2) risk associated with phytosterols, and (3) regulatory history. The scope of this review includes the multifactorial pathophysiology of PNALD/IFALD as it relates to the pharmaceutical aspects of the various lipid injectable emulsions on the market, with respect to potential proinflammatory components, as well as physical and chemical stability issues that may also affect products' safe intravenous administration to patients. There is a growing interest in using by-products rich in polyphenols, such as carob pulp (Cp, Ceratonia siliqua L.), as a dietary source of antioxidants for animals. This study assesses the effects of including Cp in lambs' diet and meat display time (0, 7, 9, and 11 days) in modified atmosphere packaging on meat colour, fatty acid (FA) composition, tocopherol levels, and lipid oxidation values in the Semimembranosus muscle of 40 light lambs. The lambs were fed with concentrates supplemented with increasing Cp levels (0, 150, and 300 g/kg) for 45 days before slaughter. Metmyoglobin (MMb) and malondialdehyde (MDA) contents increased linearly with display time (p < 0.05), regardless of diet (p > 0.05). At 11 days of display, MMb (28 ± 0.8%) and MDA (0.6 ± 0.1 mg MDA/kg of meat) contents remained within the acceptable limits. The α-tocopherol content was lower in the 30% Cp group and meat (p < 0.05). Total saturated and monounsaturated FA contents (934 ± 64 and 823 ± 65 mg/100 g of meat, respectively) did not differ significantly among the groups. However, the meat from lambs fed with 30% Cp showed reduced levels of branched-chain FAs, while polyunsaturated FAs increased (p < 0.05) compared to the control lambs. The inclusion of Cp in the lamb's diet, up to 30%, did not lead to meat deterioration and improved certain quality parameters, including a healthier FA profile. The aim of this study was to determine the effect of olive oil addition on the production, chemical composition, and fatty acid profile of sheep's milk. Twenty-four lactating ewes with a live weight of 34.6 ± 4.61 kg were used. The animals were randomly distributed into four treatments (n = 6) with dietary addition of 0%, 2%, 4%, and 6% (dry matter basis) olive oil for 45 days. Milk samples were taken every 7 days for fatty acid (FA) and chemical analyses. A decrease (p < 0.05) in dry matter and crude protein intake was observed with 4% oil inclusion. Milk production and milk components were similar between treatments. The kilograms of meat from weaned lambs linearly increased as the oil inclusion increased. Milk C4:0 to C17:0 decreased with 2% olive oil. The monounsaturated and polyunsaturated FA content in the milk increased with the oil inclusion. There was an increase in the milk's linoleic acid, linolenic acid, and eicosapentaenoic acid content with 2% olive oil. Overall, the addition of 2% extra virgin olive oil is recommended to improve milk's FA profile without negative effects on animal performance. [ABSTRACT FROM AUTHOR]

Puffed grains made from whole wheat, corn or rice grains are ready-to-eat breakfast cereals that do not require additional preparation. Two recipes for puffed grain coatings were developed in the previous studies. Sunflower oil was used in the coating, which may affect storage stability. Due to lipid hydrolysis, free fatty acids may be released, causing rancid off-flavour. The aim of this study was to determine the changes in the acid value of lipids and sensory attributes of sweet-coated puffed wheat grains ‘Badyrak vanilla’ (BV) and ‘Badyrak with whey’ (MC) depending on temperature during their eight-month storage. The products were packaged in metallised polypropylene pouches, 30 g each. The acid value was studied monthly at different storage temperatures (20 ± 2 °С and 4 ± 2 °С). In puffed wheat grain products, 65% of fatty acids are polyunsaturated, 25% monounsaturated, and 10% saturated. The high proportion of unsaturated fatty acids over saturated makes oil more susceptible to oxidation. Literature studies and sensory evaluation of puffed wheat grains allowed setting a limiting acid value at 4.0 mg KOH g−1 lipids. The increase in acid value was storage temperature dependent. Thus, the acid value exceeded the established limit in the fifth month for MC and the sixth month for BV when stored at 20 ± 2 °С. At the same time, noticeable changes in puffed wheat grain flavour were observed. The temperature coefficient (Q10) for MC was 2.0, whereas for BV it was 1.5, indicating that the acid value increase in MC was more affected by temperature than that in BV. [ABSTRACT FROM AUTHOR]

Approximately 25% of psoriasis patients have an inflammatory arthritis termed psoriatic arthritis (PsA). There is strong interest in identifying and validating biomarkers that can accurately and reliably predict conversion from psoriasis to PsA using novel technologies such as metabolomics. Lipids, in particular, are of key interest in psoriatic disease. We sought to develop a liquid chromatography-mass spectrometry (LC-MS) method to be used in conjunction with solid-phase microextraction (SPME) for analyzing fatty acids and similar molecules. A total of 25 chromatographic methods based on published lipid studies were tested on two LC columns. As a proof of concept, serum samples from psoriatic disease patients (n = 27 psoriasis and n = 26 PsA) were processed using SPME and run on the selected LC-MS method. The method that was best for analyzing fatty acids and fatty acid-like molecules was optimized and applied to serum samples. A total of 18 tentatively annotated features classified as fatty acids and other lipid compounds were statistically significant between psoriasis and PsA groups using both multivariate and univariate approaches. The SPME-LC-MS method developed and optimized was capable of detecting fatty acids and similar lipids that may aid in differentiating psoriasis and PsA patients. [ABSTRACT FROM AUTHOR]

Investigation on the physiochemical nature involved in the production of fatty acid catalyzed by the vesicles is of importance to understand the digestion of lipid. In this paper, the effects of crowding degree, which was constructed by polyethylene glycol (PEG), on the autocatalytic production of fatty acid with different chain lengths was studied. The results showed that the higher crowding degree led to the slower production rate of decanoic acid but the faster rate of oleic acid. The reason lies in that the presence of macromolecules resulted in the increased sizes of decanoic acid vesicles, but decreased sizes of oleic acid vesicles. Meanwhile, decanoic acid vesicles in more crowded medium exhibited viscous behavior, whereas oleic acid displayed elastic behavior. This research provides useful information for understanding the unusual autocatalyzed production of fatty acid in macromolecular crowding and may also draw an attention to the physiologically relevant lipid digestion. Background: The role of fatty acids in coronary heart disease (CHD) remains uncertain. There is little evidence from large-scale epidemiological studies on the relevance of circulating fatty acids levels to CHD risk. This study aims to examine the independent associations of the major circulating types of fatty acids with CHD risk. Methods: UK Biobank is a prospective study of adults aged 40–69 in 2006–2010; in 2012–2013, a subset of the participants were resurveyed. Analyses were restricted to 89,242 participants with baseline plasma fatty acids (measured using nuclear magnetic resonance spectroscopy) and without prior CHD. Cox proportional hazards models were used to estimate hazard ratios (HRs) for the associations with incidence CHD, defined as the first-ever myocardial infarction, unstable angina pectoris, coronary-related death, or relevant procedure. And the major types of fatty acids were mutually adjusted to examine the independent associations. Hazard ratios were corrected for regression dilution using the correlation of baseline and resurvey fatty acids measures. Results: During a median follow-up of 11.8 years, 3,815 incident cases of CHD occurred. Independently of other fatty acids, CHD risk was positively associated with saturated fatty acids (SFA) and monounsaturated fatty acids (MUFA), inversely associated with omega-3 polyunsaturated fatty acids (PUFA), but there was no strong evidence of an association with omega-6 PUFA: HR per standard deviation higher were 1.14 (95% CI, 1.09–1.20), 1.15 (1.10–1.21), 0.91 (0.87–0.94), and 1.04 (0.99–1.09) respectively. Independently of triglycerides and cholesterol, the inverse association with omega-3 PUFA was not materially changed, but the positive associations with SFA and MUFA attenuated to null after adjusting for triglycerides levels. Conclusions: This large-scale study has quantitated the independent associations of circulating fatty acids with CHD risk. Omega-3 PUFA was inversely related to CHD risk, independently of other fatty acids and major lipid fractions. By contrast, independently of other fatty acids, the positive associations of circulating SFA and MUFA with CHD risk were mostly attributed to their relationship with triglycerides. The experiment was to investigate the effect of adding bile acid to high-lipid diet on growth performance, muscle nutrition and fatty acid composition of gibel carp. A total of 450 healthy and initial body weight (32.37±0.13) g juvenile fish were randomly divided into five groups, with three replicates in each group and 30 fish in each replicate. Five kinds of iso-nitrogen and iso-lipid feed were prepared by adding 0 (BA0), 200 (BA200), 400 (BA400), 600 (BA600) and 800 mg/kg (BA800) bile acids to basic feed containing 12% soybean oil, respectively. The experiment lasted for 8 w. The results showed that compared with BA0, FBW of BA400 and BA600 gibel carp was significantly increased (P<0.05), WGR of BA600 was significantly increased (P<0.05), VSI was significantly decreased (P<0.05). The FCR of gibel carp in BA200, BA400 and BA600 was significantly decreased (P<0.05). HSI of all experimental groups was significantly decreased (P<0.05). CF of BA800 was significantly increased (P<0.05). Compared with BA0, muscle crude fat content of experimental groups was significantly decreased (P<0.05), and MUFA content was significantly increased (P<0.05). The crude protein content of gibel carp in BA400 and BA600 were significantly increased (P<0.05). The crude ash content of gibel carp in BA600 and BA800 were significantly decreased (P<0.05). The study indicates that adding 400~600 mg/kg bile acid to high-lipid diet can promote growth of gibel carp, increase the utilization rate of feed, promote fat digestion, absorption and decomposition for energy supply, improve muscle fatty acid composition. The study focused on the extraction of valuable lipid compounds from squid (Doryteuthis gahi) waste by a low-toxicity solvent mixture (ethanol/acetone, 50:50, v/v). The effect of the waste weight/solvent volume (WW/SV, g·mL−1) ratio and the number of extractions (NoE) on the total lipid (TL), phospholipid (PL), and tocopherol yields and on the fatty acid (FA) profile (eicosapentaenoic and docosahexaenoic acid contents; polyunsaturated FAs/saturated FAs and ω3/ω6 ratios) was investigated. As a result, an increased NoE led to an increased (p < 0.05) TL yield but a decreased (p < 0.05) proportion of PLs in the lipid extract. Additionally, a lower (p < 0.05) polyunsaturated FA/saturated FA ratio was detected by increasing the NoE. Some differences (p < 0.05) could be outlined as a result of increasing the WW/SV ratio; however, a definite trend for this extraction condition could not be concluded for any of the lipid parameters measured. Yield results were compared to those obtained by the conventional chloroform/methanol procedure. In order to attain an increased yield, the NoE required would depend on the polarity degree of the lipid molecule concerned. All ethanol/acetone extracting conditions tested led to remarkable yields for lipid compounds (PLs, α-tocopherol, ω3 FAs) and FA ratios with healthy, nutritional, and preserving properties. Food waste can be converted into insectile fatty acids (FAs) by the larvae of black soldier fly (BSFL), Hermetia illucens , for use in the feed sector or as a source of biodiesel. However, waste oil was less decomposed than carbohydrate or protein in frass due to the limitation of larval lipid metabolism. In this study, 10 yeast strains were screened, corresponding to six species, to examine their capacity of improving lipid transformation performance by BSFL. The species of Candida lipolytica was superior to the other five species, which exhibited significantly higher lipid reduction rate (95.0–97.1 %) than the control (88.7 %), and the larval FA yields achieved 82.3–115.5 % of the food waste FA matters, suggesting that BSFL not only transformed waste oil but also biosynthesized FAs from waste carbohydrate and other substances. Further, the CL2 strain of Candida lipolytica was examined for treating food waste containing high lipid content (16–32 %). The lipid removal rate was found improved from 21.4 to 42.3 % (control) to 80.5–93.3% in the waste containing 20–32 % lipid. The upper limit of lipid content that could be endured by BSFL was ≈16 %, and the CL2-enrichment elevated the upper limit to ≈24 %. Fungal community analysis indicated that Candida spp. accounted for the lipid removal improvement. The Candida spp. CL2 strain may facilitate the lipid reduction and transformation by BSFL through microbial catabolizing and assimilation of waste FAs. Altogether, this study suggests that yeast enrichment is feasible in improving lipid transformation by BSFL especially for food waste exhibiting high lipid content. [ABSTRACT FROM AUTHOR]

It is hypothesized that plasma proportion of selected fatty acids (FAs) and dietary habits are linked with the risk of cardiovascular disease (CVD) in postmenopausal women. Therefore, this study was designed to determine the association of plasma FA composition and markers of dietary habits with an atherogenic index of plasma (AIP), a predictor of CVD risk in postmenopausal women. In total, 87 postmenopausal women with an average age of 57 ± 7 years were recruited and their dietary intake, anthropometric and biochemical parameters, and FA status in total plasma lipid proportions were determined, showing that 65.5% of the participants had a high risk of CVD according to their AIP value. After adjusting for some confounders (age, body mass index, and physical activity level), the risk of CVD was only positively associated with the frequency of consumption of animal fat spreads (butter and lard) of terrestrial origin. Regarding the FA profile, CVD risk was positively associated with the percentages of vaccenic acid, dihomo-γ-linolenic acid, and monounsaturated fatty acids (MUFA; mainly n-7) in total FA, as well as the MUFA/SFA ratio in total plasma and stearoyl-CoA desaturase-16 activity (16:1/16:0 ratio). In contrast, the risk of CVD was negatively associated with percentages of α-linolenic acid, total polyunsaturated fatty acids (PUFA), and PUFA/MUFA ratio in total plasma lipid, and the estimated activity of Δ5-desaturase (20:4/20:3 n-6 ratio). These results support the current recommendations to decrease the frequency of animal fat spread intake because it is associated with a reduced CVD risk based on AIP in postmenopausal women. In accordance with these plasma percentages of ALA, vaccenic acid, dihomo-γ-linolenic acid, PUFA, PUFA/MUFA ratio, and 16:1/16:0 ratio may be important parameters in CVD risk assessment. Intramuscular fat content and fatty acid composition are important factors contributing to the nutritional value of meat. Beef meat is characterized by a high content of saturated fatty acids (SFA) which can contribute to elevated serum cholesterol levels and coronary heart disease in humans. Consumer concerns regarding the association between beef consumption and chronic cardiovascular diseases have motivated increased interest in developing strategies for reducing the lipid content and improving the fatty acid (FA) composition of beef. Although beef fat quality is determined by many factors, such as breed, gender, age, live weight, fatness degree, but the bovine diet has a key role in enhancing the nutritional quality of beef lipids. Nutritional strategies to improve the FA composition of beef focussed mainly on reducing the SFA and the n-6:n-3 poly-unsaturated fatty acid (PUFA) ratio, simultaneously increasing the PUFA of beef fat. Milk fat depression (MFD) syndrome has been associated with the antilipogenic effects of trans‐10 fatty acids (FA), such as t10, c12‐CLA (CLA) and t10‐18:1 (T10). However, these FA alone cannot completely explain the changes in milk fat in small ruminants. Thus, the aim of this study was to use multiple regression analysis to evaluate other FA that may be related to shifts in milk fat, as well as to improve model accuracy when different milk FA are used as covariates in the models. Previously published data were used in multiple regression analysis for goats (n = 106) and ewes (n = 68). Body weight (BW), vaccenic acid (t11‐18:1), both trans‐10 FA and the major milk FA were tested as covariates to model four response variables associated with MFD: fat concentration (FC), percentage change in milk fat concentration (CFC; %), fat yield (FY; g/d) and percentage change in milk fat yield (CFY; %). All four multiple regression models were significant for both species. When compared with simple regression models, all multiple regression models improved accuracy when estimating MFD. The improvements in model accuracy (lower RMSE) for FC, CFC, FY and CFY were 60.6%, 43.3%, 35.6% and 44.4% for ewes, and 52.1%, 60.1%, 33.6% and 14.9% for goats respectively. Linolenic acid and t11‐18:1 were covariates in all models for goats, and palmitic acid and CLA were covariates in all ewe models. These FA should be investigated regarding their direct effect on gene expression associated with milk fat metabolism in the mammary gland of small ruminants. Mammalian arachidonic acid lipoxygenases (ALOXs) have been implicated in the pathogenesis of inflammatory diseases, and its pro- and anti-inflammatory effects have been reported for different ALOX-isoforms. Human ALOX15B oxygenates arachidonic acid to its 15-hydroperoxy derivative, whereas the corresponding 8-hydroperoxide is formed by mouse Alox15b (Alox8). This functional difference impacts the biosynthetic capacity of the two enzymes for creating pro- and anti-inflammatory eicosanoids. To explore the functional consequences of the humanization of the reaction specificity of mouse Alox15b in vivo, we tested Alox15b knock-in mice that express the arachidonic acid 15-lipoxygenating Tyr603Asp and His604Val double mutant of Alox15b, instead of the arachidonic acid 8-lipoxygenating wildtype enzyme, in two different animal inflammation models. In the dextran sodium sulfate-induced colitis model, female Alox15b-KI mice lost significantly more bodyweight during the acute phase of inflammation and recovered less rapidly during the resolution phase. Although we observed significant differences in the colonic levels of selected pro- and anti-inflammatory eicosanoids during the time-course of inflammation, there were no differences between the two genotypes at any time-point of the disease. In Freund's complete adjuvant-induced paw edema model, Alox15b-KI mice were less susceptible than outbred wildtype controls, though we did not observe significant differences in pain perception (Hargreaves-test, von Frey-test) when the two genotypes were compared. our data indicate that humanization of the reaction specificity of mouse Alox15b (Alox8) sensitizes mice for dextran sodium sulfate-induced experimental colitis, but partly protects the animals in the complete Freund's adjuvant-induced paw edema model. Specimens of Garra rufa were collected from a warm river and a cool stream in the Bingöl Province, Turkey, once a month over a period of one year. The effects of month, season, gender and location on the fatty acid composition in the muscle and the lipid content were investigated and dietary marker fatty acids were used to obtain dietary preferences in different locations (Ilıcalar, Garip) and periods. Total lipid change was seasonally significant (ANOSIM-R=0.49) at both locations and 18:1ω9, 20:5ω3 and 20:6ω3 were the most abundant dietary fatty acids. Although G. rufa are predominantly herbivores, they can also feed omnivorously on mixed diets depending on the presence and absence of their primary diet. The effect of season was significant on fatty acid composition, regardless of the location (Pperm=0.001). Significant seasonal changes in all the fatty acid compositions could be attributed to seasonal changes in the abundance and diversity of dietary sources in the environment due to the effect of temperature. Chemosensory communication in lizards may be based on multiple compounds coming from multiple sources. Both secretions from external epidermal glands, and internal cloacal glands and feces are known to convey information (sex, familiarity, body size, etc.) for conspecifics. However, although some compounds in femoral gland secretions have been characterized and their function examined, there is very little information on potential semiochemicals in cloacal products and feces. More importantly, it is not well-known whether the compounds and information from multiple sources are redundant, complementary, or different. We analyzed the lipids in femoral gland secretions and feces of male Carpetan rock lizards (Iberolacerta cyreni) and examined which compounds might predict body size. We found many compounds in feces, mainly steroids, alkanes, and branched alkanes, while the main compounds in femoral secretions were steroids and fatty acids. The body size of males was related to the proportions of some alkanes, alkenes, and terpenoids in feces, while it is related to the proportions of some steroids and fatty acids in femoral secretions. There were also differences in the chemical profiles of feces of males and females, which may allow sex recognition. Using emulsifiers in poultry diets may increase digestibility of alternative lipid sources and enrich poultry egg yolks altering their fatty acid composition. This study aimed to evaluate performance, nutrient digestibility, egg quality, egg quality according to their storage period, and the fatty acid profile of Japanese quail egg yolk (Coturnix coturnix japônica). The design was completely randomized, in which 270 female quails were allocated in a 3x2 factorial diet arrangement: three lipid sources (soybean oil, poultry fat, and beef tallow), supplemented or not with an emulsifier, following two nutritional strategies – a diet formulated to meet the nutritional requirements proposed by INRA and a diet formulated with a reduction of 96 Kcal/kg of feed and added emulsifier. The effects of interactions between the lipid sources and the emulsifier were studied, their isolated effects when interactions were absent and the effect of storage time, and their interactions with the factors evaluated for egg quality variables. Feed intake was greater when beef tallow was added to the diets, while feed conversion was worse for birds fed diets with soybean oil and the emulsifier. Quails fed the diet with the emulsifier and soybean oil produced better quality eggs represented by the higher Haugh unit. Diets with beef tallow and poultry fat provided a higher percentage of palmitic and unsaturated fatty acids in the quail egg yolk. Alternative lipid sources such as beef tallow and poultry fat can be used as a substitute for soybean oil with added emulsifiers in diets for egg-laying quails without impairing performance and egg quality. The use of emulsifiers with alternative lipid sources to soybean oil can be considered a nutritional strategy in laying quail diets, but the reduction of energy in the diet must be adequate for the species. We are only beginning to understand how changes in skin microbiome composition are causes for, or consequences of, inflammatory skin diseases. The lamellar membranes in the stratum corneum represent a barrier for loss of internal, or intrusion of external, molecules. At the same time, it is a source of lipid-derived nutrients and antimicrobials with crucial roles in microbiome–host interaction. The full range of lipid-hydrolytic enzymes and of fatty acid resistance mechanisms awaits further analysis. Microbe-associated molecular patterns (MAMPs) are retained in skin lamellar membranes. How they are degraded there remains to be elucidated. Defects in epidermal lipid barrier integrity are probably the reason for altered skin microbiome composition and increased penetration of MAMPs towards responsive keratinocytes and leukocytes in deeper skin layers, resulting in chronic inflammatory disorders. The corneocyte layers forming the upper surface of mammalian skin are embedded in a lamellar-membrane matrix which repels harmful molecules while retaining solutes from subcutaneous tissues. Only certain bacterial and fungal taxa colonize skin surfaces. They have ways to use epidermal lipids as nutrients while resisting antimicrobial fatty acids. Skin microorganisms release lipophilic microbe-associated molecular pattern (MAMP) molecules which are largely retained by the epidermal lipid barrier. Skin barrier defects, as in atopic dermatitis, impair lamellar-membrane integrity, resulting in altered skin microbiomes, which then include the pathogen Staphylococcus aureus. The resulting increased penetration of MAMPs and toxins promotes skin inflammation. This paper investigates the fatty acid profiles of fish oil extracted from by-products of cultured Black Sea salmon, Oncorhynchus mykiss, using conventional [conventional fish oil (CFO)] and dry freezing oil [dry freezer oil (DFO)] techniques. In the CFO and DFO groups, monounsaturated fatty acid + polyunsaturated fatty acid (PUFA) comprised 74.00% and 72.68% of the total fatty acids, respectively. The highest PUFA was linoleic acid (CFO=14.22%, DFO=13.15%) with Docosahexaenoic acid (DHA, C22:6n3) being the second most concentrated fatty acid for PUFA in the CFO (8.12%) and DFO (8.02%) groups, followed by eicosapentaenoic acid (EPA, C20:5n3) (CFO=4.39%, DFO=2.87%). Similarly, the difference between groups in omega-3 was statistically significant (p<0.05) and the CFO ratio was higher in the DFO. The PI, AI, TI, h/H, and UI percentages in the CFO group were 0.99, 0.37, 0.26, 2.98, and 1.73%, respectively, while in the DFO group they were 0.80, 0.35, 0.31, 2.83, and 1.61%, respectively. The content of fatty acids in the lipids of mesenchymal stem cells of dog adipose tissue culture was studied. Mesenchymal stem cells of dog adipose tissue culture were obtained by culturing the primary material in a CO2 incubator with a content of 5 % CO2, at a temperature of 37 °C in DMEM medium with the addition of 10–15 % fetal bovine serum and 1 % antibiotic-antimycotic. When the confluency of the monolayer reached 70–80 %, the cells were transferred to a suspension and subcultivated in order to reduce the heterogeneity of the culture and obtain a sufficient amount of biological material. The lipids of the obtained stem cells were analyzed for the content of fatty acids by the method of thin-layer gasliquid chromatography. Determination of the content of lipids of fatty acids in FSK of a cat was carried out by the Folch method. A mixture of fatty acid methyl esters was analyzed on a Trace GC Ultra gas chromatograph with a flame ionization detector on a capillary column SPTM –2560, 100 m x 0.25 mm ID, 0.20 μm film (Supelco). Identification of fatty acids was carried out using a standard sample of Supelco 37 Сomponent FAME Mix. Quantitative assessment of the LC spectrum was carried out by the method of normalization of the peak planes of methylated LC derivatives and their content was determined as a percentage of the total content of all LC. The conducted study of the content of fatty acids in lipids made it possible to reveal certain features of the lipid metabolism of mesenchymal stem cells cultured in dog adipose tissue. A high content of oleic acid, characteristic of cells resistant to apoptosis and with high proliferative potential, was determined; a high ratio of unsaturated linoleic to saturated stearic acid (С18:1/С18.0), which reflects the high activity of the stearoyl-coenzyme-desaturase enzyme and, indirectly, the active state of the Wnt/β-catenin signaling pathway; inability to lengthen the chain of saturated fatty acids; lack or low activity of de novo synthesis of omega-6 polyunsaturated fatty acids. 18 fatty acids were found in the composition of lipids of fetal stem cells of a cat, of the saturated ones - the most palmitic acid (33.70 ± 0.02 %), of the monounsaturated ones – oleic acid (21.63 ± 0.03 %), of the polyunsaturated ones - linoleic acid (6.45 ± 0.07 %). The least amount of cis-,11,14-eicosadienoic acid (0.04 ± 0.01 %) was found in the composition of cell lipids. The total amount of saturated fatty acids in dog mesenchymal stem cell lipids was 65.65 ± 0.02 %), unsaturated fatty acids – 34.35 ± 0.02 %. Monoene fatty acids were determined in the amount of 24.46 ± 0.02 %, and polyene – 9.89 ± 0.02 %. The ratio index of polyunsaturated fatty acids ω 3 to ω 6 is 0.40. Lipids of mesenchymal stem cells of adipose tissue culture were characterized by a lower content of monoene unsaturated fatty acids 24.46 ± 0.02; (P < 0.05), with a higher content of ω3 fatty acids 3.04 ± 0.02 %; (P < 0.05), with a lower content of ω6 fatty acids 6.86 ± 0.02 %; (P < 0.05) in contrast to lipids of red bone marrow stem cells. Measuring acyl lipid content and fatty acid composition is important in plant lipid research, and direct transmethylation followed by analysis with gas chromatography‐flame ionization detection has been broadly used in various studies. However, many existing protocols are time‐consuming and labor‐intensive, and the long reaction time under high temperature may artificially change fatty acid compositions of the samples. In this study, we optimized the direct transmethylation method for seed and leaf samples with a wide range of seed and tissue sizes, oil contents, and fatty acid profiles, which can be completed in 2 h. We also demonstrated that commercial disposable pipet tips, as well as polypropylene tubes under certain conditions, could be reliably used to replace glass tubes, Pasteur pipettes, and rubber bulbs to improve efficiency. Taken together, this optimized and generalized method could be used as a reliable and less time‐ and labor‐intensive approach to analyze acyl lipid content and fatty acid profile of plant samples. Purpose: Fatty acid metabolism plays an important role in the survival and pathogenesis of Mycobacterium tuberculosis. During dormancy, lipids are considered to be the main source of energy. A previous study found that Rv0494 is a starvation-inducible, lipid-responsive transcriptional regulator. However, the role of Rv0494 in bacterial persister survival has not been studied.Methods: We constructed a Rv0494 deletion mutant strain of Mycobacterium tuberculosis H37Rv and evaluated the susceptibility of the mutant strain to antibiotics using a persistence test.Results: We found that mutations in Rv0494 lead to survival defects of persisters, which reflected in increased sensitivity to isoniazid.Conclusion: We conclude that Rv0494 is important for persister survival and may serve as a good target for developing new antibiotics that kill persister bacteria for improved treatment of persistent bacterial infections. Ultra-long-chain fatty acids (ULCFAs) are biosynthesized in the restricted tissues such as retina, testis, and skin. The conformation of a single ULCFA, in which the sn-1 unsaturated chain has 32 carbons, in three types of phospholipid bilayers is studied by molecular dynamics simulations. It is found that the ultra-long tail of the ULCFA flips between two leaflets and fluctuates among an elongation into the opposite leaflet, lies between two leaflets, and turns back. As the number ratio of lipids in the opposite leaflet increases, the ratio of the elongated shape linearly decreases in all three cases. Microalgae can convert carbon dioxide into organic matter through photosynthesis. Thus, they are considered as an environment-friendly and efficient cell chassis for biologically active metabolites. Microalgal lipids are a class of organic compounds that can be used as raw materials for food, feed, cosmetics, healthcare products, bioenergy, etc., with tremendous potential for commercialization. In this review, we summarized the commercial lipid products from eukaryotic microalgae, and updated the mechanisms of lipid synthesis in microalgae. Moreover, we reviewed the enhancement of lipids, triglycerides, polyunsaturated fatty acids, pigments, and terpenes in microalgae via environmental induction and/or metabolic engineering in the past five years. Collectively, we provided a comprehensive overview of the products, biosynthesis, induced strategies and genetic engineering in microalgal lipids. Meanwhile, the outlook has been presented for the development of microalgal lipids industries, emphasizing the significance of the accurate analysis of lipid bioactivity, as well as the high-throughput screening of microalgae with specific lipids.

Lipids are a large group of natural compounds, together with proteins and carbohydrates, and are essential for various processes in the body. After death, the organism’s tissues undergo a series of reactions that generate changes in some molecules, including lipids. This means that determining the lipid change profile can be beneficial in estimating the postmortem interval (PMI). These changes can also help determine burial sites and advance the localization of graves. The aim was to explore and analyze the decomposition process of corpses, focusing on the transformation of lipids, especially triglycerides (TGs) and fatty acids (FAs), and the possible application of these compounds as markers to estimate PMI and detect burial sites. A systematic review of 24 scientific articles from the last 23 years (2000–2023) was conducted. The results show that membrane glycerophospholipids (such as phosphatidylcholine and phosphatidylglycerol, among others) are the most studied, and the most promising results are obtained, with decreasing patterns as PMI varies. Fatty acids (FAs) are also identified as potential biomarkers owing to the variations in their postmortem concentration. An increase in saturated fatty acids (SFAs), such as stearic acid and palmitic acid, and a decrease in unsaturated fatty acids (UFAs), such as oleic acid and linoleic acid, were observed. The importance of intrinsic and extrinsic factors in decomposition is also observed. Finally, as for the burial sites, the presence of fatty acids and some sterols in burial areas of animal and human remains can be verified. In conclusion, glycerophospholipids and fatty acids are good markers for estimating PMI. It has been observed that there are still no equations for estimating the PMI that can be applied to forensic practice, as intrinsic and extrinsic factors are seen to play a vital role in the decomposition process. As for determining burial sites, the importance of soil and textile samples has been demonstrated, showing a direct relationship between saturated fatty acids, hydroxy fatty acids, and some sterols with decomposing remains.

Fatty acids (FA) in follicular fluid (FF) are present in an esterified form [triglycerides, cholesterol esters and phospholipids] or as non-esterified FA, which partly originate from blood. However, a comprehensive comparison of blood vs. FF FA in various lipid classes is missing. The aim of this study was to determine the distribution of the FA composition in each lipid class of serum and FF, and to investigate their mutual correlations. A total of 74 patients undergoing assisted reproductive technology treatment were involved in the study. Both in serum as well as FF, saturated FA and mono-unsaturated FA were predominant in non-esterified FA and triglycerides fractions while poly-unsaturated FA were mainly present in phospholipids and cholesterol esters fractions, although phospholipids also contained high proportions of saturated FA. Irrespective of the lipid class, the FA proportions differed between serum and FF (P < 0.05). Despite these differences, most of the FA in triglycerides, phospholipids and cholesterol esters of FF were well correlated with their proportions in serum. Nevertheless, only weak to moderate associations (r < 0.60) were observed for the majority of the FA in the non-esterified FA fraction. Differences in FA product/precursor-ratios were found between serum and FF, such as higher C20:4n-6 to C18:2n-6 and C20:5n-3 to C18:3n-3 in FF. FA metabolism (e.g. desaturation and elongation) takes place in cells of the intrafollicular micro-environment. The arachidonic acid lipoxygenase 15B (ALOX15B) orthologs of men and mice form different reaction products when arachidonic acid is used as the substrate. Tyr603Asp+His604Val double mutation in mouse arachidonic acid lipoxygenase 15b humanized the product pattern and an inverse mutagenesis strategy murinized the specificity of the human enzyme. As the mechanistic basis for these functional differences, an inverse substrate binding at the active site of the enzymes has been suggested, but experimental proof for this hypothesis is still pending. Here we expressed wildtype mouse and human arachidonic acid lipoxygenase 15B orthologs as well as their humanized and murinized double mutants as recombinant proteins and analyzed the product patterns of these enzymes with different polyenoic fatty acids. In addition, in silico substrate docking studies and molecular dynamics simulation were performed to explore the mechanistic basis for the distinct reaction specificities of the different enzyme variants. Wildtype human arachidonic acid lipoxygenase 15B converted arachidonic acid and eicosapentaenoic acid to their 15-hydroperoxy derivatives but the Asp602Tyr+Val603His exchange murinized the product pattern. The inverse mutagenesis strategy in mouse arachidonic acid lipoxygenase 15b (Tyr603Asp+His604Val exchange) humanized the product pattern with these substrates, but the situation was different with docosahexaenoic acid. Here, Tyr603Asp+His604Val substitution in mouse arachidonic acid lipoxygenase 15b also humanized the specificity but the inverse mutagenesis (Asp602Tyr+Val603His) did not murinize the human enzyme. With linoleic acid Tyr603Asp+His604Val substitution in mouse arachidonic acid lipoxygenase 15b humanized the product pattern but the inverse mutagenesis in human arachidonic acid lipoxygenase 15B induced racemic product formation. Amino acid exchanges at critical positions of human and mouse arachidonic acid lipoxygenase 15B orthologs humanized/murinized the product pattern with C20 fatty acids, but this was not the case with fatty acid substrates of different chain lengths. Asp602Tyr+Val603His exchange murinized the product pattern of human arachidonic acid lipoxygenase 15B with arachidonic acid, eicosapentaenoic acid, and docosahexaenoic acid. An inverse mutagenesis strategy on mouse arachidonic acid lipoxygenase 15b (Tyr603Asp+His604Val exchange) did humanize the reaction products with arachidonic acid and eicosapentaenoic acid, but not with docosahexaenoic acid. The systemic RNAi-defective (SID) transmembrane family member 2 (SIDT2) is a putative nucleic acid channel or transporter that plays essential roles in nucleic acid transport and lipid metabolism. Here, we report the cryo-electron microscopy (EM) structures of human SIDT2, which forms a tightly packed dimer with extensive interactions mediated by two previously uncharacterized extracellular/luminal β-strand-rich domains and the unique transmembrane domain (TMD). The TMD of each SIDT2 protomer contains eleven transmembrane helices (TMs), and no discernible nucleic acid conduction pathway has been identified within the TMD, suggesting that it may act as a transporter. Intriguingly, TM3-6 and TM9-11 form a large cavity with a putative catalytic zinc atom coordinated by three conserved histidine residues and one aspartate residue lying approximately 6 Å from the extracellular/luminal surface of the membrane. Notably, SIDT2 can hydrolyze C18 ceramide into sphingosine and fatty acid with a slow rate. The information presented advances the understanding of the structure-function relationships in the SID1 family proteins. The SID-1 family is a putative channel/transporter for transporting nucleic acids. Aim: The aim of the study was to be thoroughly examined the chemical and lipid composition of three species of wild edible mushrooms (Boletus pinophilus, Cantharellus cibarius and Craterellus cornucopioides) grown in Bulgaria as well as performing a principal component analysis in order to clarify specific relationships between the species and their nutritional compositions. Methods: Standard methods following the ISO procedures were used for determination of the components. Results: Moisture, crude fat, proteins, total carbohydrates, crude fibre and ash were determined in the examined mushroom species. Potassium and phosphorus were the essential elements in the mushrooms. Unsaturated fatty acids predominated in the lipid fraction of triacylglycerols, and oleic acid was the main one. Ergosterol was the major sterol in the lipid fraction of all mushrooms (42.4 – 72.8%). Phospholipid composition of the mushrooms differed, and phosphatidic acids were the main class in B. pinophilus and C. cibarius, while in C. cornucopioides all phospholipid classes were present in similar quantities. Fatty acid composition of the main phospholipid classes of the mushrooms was determined for the first time and was observed that saturated fatty acids (51.2 – 73.3%) predominated in all main phospholipids except in phosphatidic acids from C. cornucopioides, where unsaturated fatty acids prevailed (54.9%). Conclusion: Some key differences were observed in the chemical and lipid composition of the examined mushroom species but all of them were abundant in valuable fat-soluble biologically active components which made them suitable for possible application as food additives in many products and functional foods. A pollen grain is a unique haploid organism characterized by a special composition and structure. The pollen of angiosperms and gymnosperms germinate in fundamentally similar ways, but the latter also have important features, including slow growth rates and lower dependence on female tissues. These features are, to some extent, due to the properties of pollen lipids, which perform a number of functions during germination. Here, we compared the absolute content and the fatty acid (FA) composition of pollen lipids of two species of flowering plants and spruce using GC-MS. The FA composition of spruce pollen differed significantly, including the predominance of saturated and monoene FAs, and a high proportion of very-long-chain FAs (VLCFAs). Significant differences between FAs from integumentary lipids (pollen coat (PC)) and lipids of gametophyte cells were found for lily and tobacco, including a very low unsaturation index of the PC. The proportion of VLCFAs in the integument was several times higher than in gametophyte cells. We found that the absolute content of lipids in lily pollen is almost three times higher than in tobacco and spruce pollen. For the first time, changes in the FA composition were analyzed during pollen germination in gymnosperms and angiosperms. The stimulating effect of H2O2 on spruce germination also led to noticeable changes in the FA content and composition of growing pollen. Trypanosomatids are a diverse group of uniflagellate protozoan parasites that include globally relevant pathogens such as Trypanosoma cruzi, the causative agent of Chagas disease. Trypanosomes lack the fatty acid synthase system typically used for de novo fatty acid (FA) synthesis in other eukaryotes. Instead, these microbes have evolved a modular FA elongase (ELO) system comprised of individual ELO enzymes (ELO1-4) that can operate processively to generate long chain- and very long chain-FAs. The importance of ELO's for maintaining lipid homeostasis in trypanosomatids is currently unclear, given their ability to take up and utilize exogenous FAs for lipid synthesis. To assess ELO function in T. cruzi, we generated individual KO lines, Δelo1, Δelo2, and Δelo3, in which the genes encoding ELO1-3 were functionally disrupted in the parasite insect stage (epimastigote). Using unbiased lipidomic and metabolomic analyses, in combination with metabolic tracing and biochemical approaches, we demonstrate that ELO2 and ELO3 are required for global lipid homeostasis, whereas ELO1 is dispensable for this function. Instead, ELO1 activity is needed to sustain mitochondrial activity and normal growth in T. cruzi epimastigotes. The cross-talk between microsomal ELO1 and the mitochondrion is a novel finding that, we propose, merits further examination of the trypanosomatid ELO pathway as critical for central metabolism. Lipid and fatty acid modifications induced by the effects of various salinity patterns on the juvenile pink salmon Oncorhynchus gorbuscha (the Olkhovka River and the White Sea) within the experiment have been revealed. Concentrations of steroids, saturated fatty acids, and signaling molecules such as phospholipids (phosphatidylserine and phosphatidylinositol) and arachidonic acid under the hyperosmotic stress-related effects (keeping the fish for 1 h in the seawater after the time of transfer from the freswater) tend to increase. Decreases in phosphatidylcholine and n-6 saturated, monounsaturated, and polyunsaturated fatty acids are recorded in the juvenile pink salmon fish kept for 24 h in the seawater after the time of transfer them from the freswater, while the levels of phosphatidylethanolamine and n-3 polyunsaturated fatty acids (especially eicosapentaenoic and docosahexaenoic acids), on the contrary, tend to rise significantly. Lipid composition modifications in the juvenile pink salmon fish kept under the hypoosmotic stress conditions (24 h in freshwater after 24 h in seawater) induce stabilization of functioning the cell membrane structure, since the levels of bioeffectors including phosphatidylserine, phosphatidylinositol, and arachidonic, eicosapentaenoic, and docosahexaenoic acids tend to decrease. A reduced amount of lipids (triacylglycerols) stored as an energy reserve is shown for all the experimental fish groups. The outcomes indicate the high levels of adaptive potential of the juvenile pink salmon fish kept under the effects of abrupt salinity change in the water environments and their readiness for downstream migration not long before leaving the nests. Nutrient deficiency induces a variety of cellular responses, including an increase in lipid accumulation in microalgae. Nitrogen starvation is the most studied deprivation. Here, we determine the effects of phosphorus and sulfur limitation on lipid accumulation in Chlorella vulgaris. A set of 9 experiments were performed, varying the initial concentration of these nutrients (set to 0, 50, and 100% of their original composition in Bold's basal medium). According to our results, the variation of P and S modified the specific growth rate, lag phase, and cell generation time. The ratio of 50%P and 0%S significantly increased the total lipid concentration. The fatty acid profile was dominated by C16:0, C18:0, and C18:1; a considerable increase in C20:5 was observed with 0%P and 50%S and 0%P and 100%S. Regarding neutral lipids, the response surface methodology (RSM) indicates that the maximum was observed when S was between 40 and 60% and P was between 95 and 100%. The fruit of the oil palm (Elaeis guineensis Jacq.) has fleshy mesocarpic tissue rich in lipids. This edible vegetable oil is economically and nutritionally significant across the world. The core concepts of oil biosynthesis in oil palms remain to be researched as the knowledge of oil biosynthesis in plants improves. In this study, we utilized a metabolite approach and mass spectral analysis to characterize metabolite changes and identify the sequences of protein accumulation during the physiological processes that regulate oil synthesis during oil palm fruit ripening. Here, we performed a comprehensive lipidomic data analysis in order to understand the role of lipid metabolism in oil biosynthesis mechanisms. The experimental materials were collected from the mesocarp of oil palm (Tenera) at 95 days (early accumulation of fatty acid, first stage), 125 days (rapid growth of fatty acid accumulation, second stage), and 185 days (stable period of fatty acid accumulation, third stage) after pollination. To gain a clear understanding of the lipid changes that occurred during the growth of the oil palm, the metabolome data were found using principal component analysis (PCA). Furthermore, the accumulations of diacylglycerols, ceramides, phosphatidylethanolamine, and phosphatidic acid varied between the developmental stages. Differentially expressed lipids were successfully identified and functionally classified using KEGG analysis. Proteins related to the metabolic pathway, glycerolipid metabolism, and glycerphospholipid metabolism were the most significantly changed proteins during fruit development. Background: Rice (Oryza sativa) straw is a common waste product that represents a considerable amount of bound energy. This energy can be used for biogas production, but the rate and level of methane produced from rice straw is still low. To investigate the potential for an increased biogas production from rice straw, we have here utilized WRINKLED1 (WRI1), a plant AP2/ERF transcription factor, to increase triacylglycerol (TAG) biosynthesis in rice plants. Two forms of Arabidopsis thaliana WRI1 were evaluated by transient expression and stable transformation of rice plants, and transgenic plants were analyzed both for TAG levels and biogas production from straw. Results: Both full-length AtWRI1, and a truncated form lacking the initial 141 amino acids (including the N-terminal AP2 domain), increased fatty acid and TAG levels in vegetative and reproductive tissues of Indica rice. The stimulatory effect of the truncated AtWRI1 was significantly lower than that of the full-length protein, suggesting a role for the deleted AP2 domain in WRI1 activity. Full-length AtWRI1 increased TAG levels also in Japonica rice, indicating a conserved effect of WRI1 in rice lipid biosynthesis. The bio-methane production from rice straw was 20% higher in transformants than in the wild type. Moreover, a higher producing rate and final yield of methane was obtained for rice straw compared with rice husks, suggesting positive links between methane production and a high amount of fatty acids. Conclusions: Our results suggest that heterologous WRI1 expression in transgenic plants can be used to improve the metabolic potential for bioenergy purposes, in particular methane production. The role of recovery after drought has been proposed to play a more prominent role during the whole drought-adaption process than previously thought. Two maize hybrids with comparable growth but contrasting physiological responses were investigated using physiological, metabolic, and lipidomic tools to understand the plants' strategies of lipid remodeling in response to repeated drought stimuli. Profound differences in adaptation between hybrids were discovered during the recovery phase, which likely gave rise to different degrees of lipid adaptability to the subsequent drought event. These differences in adaptability are visible in galactolipid metabolism and fatty acid saturation patterns during recovery and may lead to a membrane dysregulation in the sensitive maize hybrid. Moreover, the more drought-tolerant hybrid displays more changes of metabolite and lipid abundance with a higher number of differences within individual lipids, despite a lower physiological response, while the responses in the sensitive hybrid are higher in magnitude but lower in significance on the level of individual lipids and metabolites. Cotton (Gossypium spp.) is the fifth largest oil crop in the world, and cottonseed provides abundant vegetable oil resources and industrial bioenergy fuels for people; therefore, it is of practical significance to increase the oil content of cotton seeds for improving the oil yield and economic benefits of planting cotton. Long-chain acyl-coenzyme A (CoA) synthetase (LACS) capable of catalyzing the formation of acyl-CoAs from free fatty acids has been proven to significantly participate in lipid metabolism, of which whole-genome identification and functional characterization of the gene family have not yet been comprehensively analyzed in cotton. In this study, a total of sixty-five LACS genes were confirmed in two diploid and two tetraploid Gossypium species, which were divided into six subgroups based on phylogenetic relationships with twenty-one other plants. An analysis of protein motif and genomic organizations displayed structural and functional conservation within the same group but diverged among the different group. Gene duplication relationship analysis illustrates the LACS gene family in large scale expansion through WGDs/segmental duplications. The overall Ka/Ks ratio indicated the intense purifying selection of LACS genes in four cotton species during evolution. The LACS genes promoter elements contain numerous light response cis-elements associated with fatty acids synthesis and catabolism. In addition, the expression of almost all GhLACS genes in high seed oil were higher compared to those in low seed oil. We proposed LACS gene models and shed light on their functional roles in lipid metabolism, demonstrating their engineering potential for modulating TAG synthesis in cotton, and the genetic engineering of cottonseed oil provides a theoretical basis. This study was conducted to evaluate the effects of a low-protein (LP) diet supplemented with sodium butyrate (SB), medium-chain fatty acids (MCFAs) and n-3 polyunsaturated fatty acids (PUFAs) on nutrient utilization and lipid and amino acid metabolism in weaned pigs. A total of 120 Duroc × Landrace × Yorkshire pigs (initial body weight: 7.93 ± 0.65 kg) were randomly assigned to five dietary treatments, including the control diet (CON), LP diet, LP + 0.2% SB diet (LP + SB), LP + 0.2% MCFA diet (LP + MCFA) and LP + 0.2% n-3 PUFA diet (LP + PUFA). The results show that the LP + MCFA diet increased (p < 0.05) the digestibility of dry matter and total P in pigs compared with the CON and LP diets. In the liver of the pigs, the metabolites involved in sugar metabolism and oxidative phosphorylation significantly changed with the LP diet compared with the CON diet. Compared with the LP diet, the altered metabolites in the liver of the pigs fed with the LP + SB diet were mainly associated with sugar metabolism and pyrimidine metabolism; the altered metabolites in the liver of pigs fed with the LP + MCFA and LP + PUFA diets were mainly associated with lipid metabolism and amino acid metabolism. In addition, the LP + PUFA diet increased (p < 0.05) the concentration of glutamate dehydrogenase in the liver of pigs compared with the LP diet. Furthermore, the LP + MCFA and LP + PUFA diets increased (p < 0.05) the mRNA abundance of sterol regulatory element-binding protein 1 and acetyl-CoA carboxylase in the liver compared with the CON diet. The LP + PUFA diet increased (p < 0.05) mRNA abundances of fatty acid synthase in the liver compared with the CON and LP diets. Collectively, the LP diet supplemented with MCFAs improved nutrient digestibility, and the LP diet supplemented with MCFAs and n-3 PUFAs promoted lipid and amino acid metabolisms. Background: Breast cancer is associated with alterations in lipid metabolism. The treatment of breast cancer can also affect serum lipid composition. The purpose of this study was the examination of serum fatty acids (FAs) profiles in breast cancer survivors to assess if the FA levels normalize. Methods: Serum levels of FAs were determined by gas chromatography–mass spectrometry in a group of breast cancer patients at baseline (before treatment, n = 28), at two follow-up visits at 12 months (n = 27) and 24 months (n = 19) after the breast cancer resection, and in the group of healthy controls (n = 25). Multivariate analysis was performed to assess how FA serum profile changes following treatment. Results: Breast cancer patients' serum FA profiles at follow-ups did not normalize to the levels of control group. The greatest differences were found for levels of branched-chain (BCFA), odd-chain (OCFA) and polyunsaturated (PUFAs) FAs, all of which were significantly increased 12 months after the surgery. Conclusions: After treatment for breast cancer, the patients' serum FA profile differs from the profile before treatment and from controls, especially 12 months after treatment. Some changes may be beneficial – increased BCFA and OCFA levels, and improved n-6/n-3 PUFA ratio. Wild fish attracted to salmon farms feed on waste feed that presently contain high levels of fatty acids of terrestrial origin. This study examines whether mature Atlantic cod (Gadus morhua) caught at spawning grounds has eaten salmon waste feed. Cod were caught at four spawning grounds around Smøla (Norway), an area with multiple salmon farms, during the spawning season in 2018 (n = 327) and 2019 (n = 488). The fatty acid (FA) profile of their livers, gonads (ovary and testis), and muscles (2019) were determined. Multivariate k-mean cluster analysis of liver FA profiles revealed three main clusters, which could be allocated to trophic niches using known fatty acid trophic markers (FATMs). Of the sampled cod in 2018 and 2019, 13 % and 20 % respectively had high liver concentrations of terrestrial FATMs (18:1n-9, 18:2n-6, and 18:3n-3), indicating waste feed feeding. The remaining cod could be assigned to either the pelagic or benthic food chain. The cod identified as feeding on waste feed had large, fatty livers. The terrestrial FAs were also transferred to the muscle and gonad lipids. Simple Summary: The digestibility of ingredients in fish diets must be known to use the least-cost feed formulation. However, there is very little published information on lipid (fat) digestibility in fish. With this information, commercial feed producers can choose lipids based on high utilization by the fish, as well as cost. We conducted a feeding trial with catfish fed diets with different lipids: soybean oil, soybean oil with conjugated linoleic acids, catfish offal oil, flaxseed oil, menhaden fish oil and poultry fat. After feeding, fish feces were collected for nutrient analysis and compared to the lipid content of the diets. Lipid and fatty acid digestibility were high overall for all of the lipids tested. However, the digestibility of certain fatty acids was different from overall lipid digestibility. This information can be used to choose the best lipids to meet catfish needs, enhance healthy fats in the fish for human consumers, and produce a cost-effective feed. Lipid and fatty acid digestibility is presumably high in Channel Catfish, but data is lacking. We determined the lipid and fatty acid digestibility of traditional and alternative dietary lipids in Channel Catfish to inform lipid choice for commercial diets. Six diets contained 4% of different lipids: soybean oil (SBO), soybean oil containing conjugated linoleic acids (CLA-SBO), catfish offal oil (COO), flaxseed oil (FXO), menhaden fish oil (MFO) and poultry fat (PF). Diets were fed to Channel Catfish (150–200 g) maintained at 26.5 °C in each of six 110 L aquaria. Six hours post-prandial, feces were collected for analysis. Total lipid, crude protein and fatty acids of lyophilized feces were analyzed, and apparent digestibility coefficients (ADCs) were calculated. ADCs of lipid, saturated and monounsaturated fatty acids, linoleic acid and protein digestibility were similar among diets. CLA isomers (cis-9, trans-11 (84.1%) and trans-10, cis-12 (90%)) in the CLA-SBO diet were highly digestible. Oleic acid digestibility was highest in the PF diet. ADC was high for α-linolenic acid in the FXO diet, and for arachidonic acid and n-3 LC-PUFA in the MFO diet. Objective. To assess the fatty acids composition (FA) in Wagyu beef and its crosses with Angus, Beefmaster, Brangus and Hereford, and to analyze its relationship with genetic markers related to lipid metabolism. Materials and methods. 111 Longissimus dorsi samples were collected and grouped by genetic group. FA were extracted and quantified in a gas-liquid chromatography and DNA markers theoretically associated to FA were typed. Hardy-Weinberg equilibrium and linkage disequilibrium were examined, the effect of crosses and the effect of genotypes were estimated. Results. The crosses did not show substantial differences in FA composition. Nine SNPs showed association with FA composition, and a significant effect was found in the SLC2A4 marker ss62538460 which influenced SFA, MUFA and MUFA/SFA; PLTP ss77832104 and IGF2R ss77831885 markers, influenced C16:0, MYOZ1 ss77832104 on C17:1 and PPARGC1A c.1892+19 on C18:2. In addition, previously described effects of MEF2C ss38329156 and SCD c.878 were supported. Conclusions. These results are first evidence on FA deposition in Wagyu cattle and their crosses, and proposes some loci in candidate genes with the possibility of implementation in assisted selection strategies. Background: The success of larval rearing is greatly influenced by first feeding regimes and the nutritional quality of weaning diets, with dietary lipids being recognized as one of the most important nutritional factors that affect larval growth and survival. Reports are scanty on milkfish larval nutrition and growth and survival unlike other marine species. Methods: In this investigation during 2020, five larval diets were prepared with 40 g kg-1 of entirely fish oil (F4), corn oil (C4) or fish oil and corn oil in 3:1 (F3C1), 2:2 (F2C2) and 1:3 (F1C3) ratios. Each diet was fed to triplicate groups of milkfish larvae (45 mg) in a flow-through rearing system for 42 days. Result: A growth indices were highest in the F3C1 group, followed by F2C2, F4 and other dietary treatments. The whole-body fatty acid profile was found to change significantly with increasing fish oil replacement with corn oil, i.e., the n-3 polyunsaturated and saturated fatty acid proportions decreased linearly, while the n-6 polyunsaturated fatty acids content increased. Overall, this study reveals that growth and survival of milkfish is dependent on dietary lipid source or combinations that meet the essential fatty acid requirements during the early life stages.
