Background: High intakes of trans-fatty acids (TFAs), especially industrially produced TFAs, can lead to unfavorable lipid and lipoprotein concentrations and an increased risk of cardiovascular disease. It is unknown how this relation might change in a population after significant reductions in TFA intake.Objective: This study, which used a new analytical method for measuring plasma TFA concentrations, clarified the association between plasma TFA and serum lipid and lipoprotein concentrations before and after the US FDA enacted TFA food-labeling regulations in 2006.Methods: Data were selected from the NHANES of 1999-2000 and 2009-2010. Findings on 1383 and 2155 adults, respectively, aged ≥20 y, were evaluated. Multivariable linear regressions were used to examine the associations between plasma TFA concentration and lipid and lipoprotein concentrations. The outcome measures were serum concentrations of total cholesterol (TC), LDL cholesterol, HDL cholesterol, and triglycerides and the ratio of TC to HDL cholesterol.Results: The median plasma TFA concentration decreased from 80.6 μmol/L in 1999-2000 to 37.0 μmol/L in 2009-2010. Plasma TFA concentration continued to be associated with serum lipid and lipoprotein concentrations after significant reductions in TFA intake in the population. For example, by comparing the lowest with the highest quintiles of TFA concentration in 1999-2000, adjusted mean (95% CI) LDL-cholesterol concentrations increased from 118 mg/dL (112, 123 mg/dL) to 135 mg/dL (130, 141 mg/dL) (P-trend < 0.001). The corresponding values for 2009-2010 were 102 mg/dL (97.4, 107 mg/dL) and 129 mg/dL (125, 133 mg/dL) for LDL cholesterol (P-trend < 0.001). Differences between the highest and lowest quintiles were consistent across age groups, sexes, races/ethnicities, and other covariates.Conclusions: Despite a 54% reduction in plasma TFA concentrations in US adults from 1999-2000 to 2009-2010, concentrations remained significantly associated with serum lipid and lipoprotein concentrations. Long chain (LC) polyunsaturated fatty acids (PUFA) are major components of cell membrane phospholipids (PL) and serve as precursors for numerous bioactive lipid derivatives. Fatty acids (FA) are routinely analyzed in biological samples to assess composition of tissues, cells, and lipid fractions. In human studies, serum or plasma is often used because of their easy procurement. However, the lipid pool in serum and plasma is a mixture of triacylglycerol (TG), PL, cholesterol and its esters, and other components. Herein, we report findings from a serum FA analysis after fractionation of polar and neutral lipids by solid phase extraction in a large cohort of 400 hemodialysis patients. LC PUFA were found concentrated in the polar fraction compared to the total or the neutral lipid fraction. When correlated with clinical markers of disease, a greater number of significant correlations were found for PUFA in polar compared to total or the neutral fraction. We also observed that polar lipids are a reliable reflection of LC PUFA status compared to the total or neutral fractions because the latter are diluted by non-essential FA. The relative amounts of LC PUFA in the total and neutral fractions reflect the contribution of TG in blood that varies with diet, age, and physiologic state. Our data indicate that LC PUFA in the polar fraction are superior indicators of bioactive FA-status than in the total or the neutral fraction and should be used to establish important links between PUFA status, their bioactive substrates in hemodialysis patients. Lung cancer is the leading cause of cancer deaths worldwide. In the United States, only one in six lung cancer patients survives five years after diagnosis. These statistics may improve if new therapeutic targets are identified. We previously reported that an enzyme of fatty acid metabolism, very long-chain acyl-CoA synthetase 3 (ACSVL3), is overexpressed in malignant glioma, and that depleting glioblastoma cells of ACSVL3 diminishes their malignant properties. To determine whether ACSVL3 expression was also increased in lung cancer, we studied tumor histologic sections and lung cancer cell lines. Immunohistochemical analysis of normal human lung showed moderate ACSVL3 expression only in bronchial epithelial cells. In contrast, all of 69 different lung tumors tested, including adeno-, squamous cell, large cell, and small cell carcinomas, had robustly elevated ACSVL3 levels. Western blot analysis of lung cancer cell lines derived from these tumor types also had significantly increased ACSVL3 protein compared to normal bronchial epithelial cells. Decreasing the growth rate of lung cancer cell lines did not change ACSVL3 expression. However, knocking down ACSVL3 expression by RNA interference reduced cell growth rates in culture by 65–76%, and the ability of tumor cells to form colonies in soft agar suspension by 65–80%. We also conducted studies to gain a better understanding of the biochemical properties of human ACSVL3. ACSVL3 mRNA was detected in many human tissues, but the expression pattern differed somewhat from that of the mouse. The enzyme activated long- and very long-chain saturated fatty acid substrates, as well as long-chain mono- and polyunsaturated fatty acids to their respective coenzyme A derivatives. Endogenous human ACSVL3 protein was found in a punctate subcellular compartment that partially colocalized with mitochondria as determined by immunofluorescence microscopy and subcellular fractionation. Abstract: Ester bound, long chain fatty acids (e-LFAs), hydroxy fatty acids (FAs) and di-acids are key components of the hydrolysable aliphatic lipids in soil organic matter (SOM). Cutin and suberin are important sources of these SOM components, which may contribute to the recalcitrant soil carbon pool. However, their carbon isotopic composition and turnover rates are largely unknown. This study represents the first compound specific isotopic analysis (CSIA) of major hydrolysable lipids in the surface mineral soil horizons from the Duke Forest, North Carolina, USA. The investigated lipids include hydroxy FAs and di-acids that only originate from cutin and suberin (i.e., cutin- and suberin-derived compounds) and e-LFAs that originate from plant waxes as well as cutin and suberin. These compounds were 2–7‰ more depleted in 13C as compared to the bulk organic matter in plant tissues as well as in soils. Furthermore, they were more depleted in 13C in plant tissues produced under the elevated versus ambient CO2 concentration in the free air CO2 enrichment (FACE) experiment. Under a steady-state condition, this isotopic signature was incorporated into the hydrolysable lipids in the surface soil at a rate of 0.029–0.031year−1. This turnover rate corresponds to a mean residence time (MRT) of 32–34year, which is longer than that of bulk SOM but much shorter than that expected for recalcitrant SOM components. We suggest that a significant fraction of cutin and suberin is transformed into non-hydrolysable SOM, which may not be accounted for in the hydrolysable fraction of SOM. [Copyright &y& Elsevier]

Abstract: Stalk juice from sweet sorghum grown in Southern Illinois, USA, was examined for lipid production through microalgal fermentation. Juice concentrations at 100%, 75%, 50%, and 25% led to different biomass, lipid, and docosahexaenoic acid (DHA) production by Schizochytrium limacinum SR21. Biomass dry weight as 9.4g/l at 50% juice concentration was similar to that from pure glucose (10.9g/l). But with a 73.4% lipid content, this dose resulted in higher lipid and DHA production than those from pure glucose. Major fatty acids in cells grown on juice were identical to those fed by other substrates. Among the three sugars – glucose, fructose, and sucrose in sorghum juice, only glucose was utilized for growth. Spent medium after algal removal may be further processed for white sugar production in a traditional way since sucrose content remained the same throughout the algal fermentation process. Algal cells or lipids harvested can be utilized as fish meal, human nutrition supplements, or for biodiesel purpose. [Copyright &y& Elsevier]

Abstract: Recently, the United States Pharmacopeia (USP) has established Chapter 〈729〉 with GSD limits for all lipid emulsions where the mean droplet size (MDS) must be <500nm and the percent of fat larger than 5μm (PFAT5) must be <0.05%, irrespective of the final lipid concentration. As well, the European Pharmacopeia (EP) Monograph no. 1352 specifies n3-fatty acid (FA) limits (EPA+DHA≥45%; total n3 or T-n3≥60%) for fish oil. We assessed compliance with USP physical and EP chemical limits of two fish oil-containing lipid emulsion mixtures. All lipid emulsions passed USP 〈729〉 limits. No samples tested had an MDS >302nm or a PFAT5 value >0.011%. Only one product met EP limits while the other failed. All emulsions tested were extremely fine dispersions and easily met USP 〈729〉 GSD limits. The n3-FAs profiles were lower in one, despite being labeled to contain 50% more fish oil than the other product. This latter finding suggests the n3-FA content of the fish oil source and/or the applied manufacturing processes in these products is different. [Copyright &y& Elsevier]

Four different catfish oil extraction processes were used to extract oil from catfish viscera: process CF1 involved a mixture of ground catfish viscera and water, no heat treatment, and centrifugation; process CF2 involved ground catfish viscera (no added water), heat treatment, and centrifugation; process CF3 involved a mixture of ground catfish viscera and water, heat treatment, and centrifugation; process CF4 involved ground catfish viscera, enzymatic hydrolysis, and centrifugation. Chemical and physical properties of the resulting of catfish oils were evaluated. The CF4 process recovered significantly higher amounts of crude oil from catfish viscera than the other 3 extraction methods. The CF4 oil contained a higher percent of free fatty acid and peroxide values than CF1, CF2, and CF3 oils. Oleic acid in catfish oil was the predominant fatty acid accounting for about 50% of total fatty acids. Weight loss of oils increased with increasing temperatures between 250 and 500 °C. All the catfish oil samples melted around −32 °C regardless of the extraction methods. The flow behavior index of all the oil samples was less than 1, which indicated that the catfish oils exhibited non-Newtonian fluid behavior. The apparent viscosity at −5 and 0 °C was significantly higher ( P < 0.05) than those at 5, 10, 15, 20, 25, and 30 °C. The average magnitude of activation energy for apparent viscosity of the oil was higher for CF2 than CF1, CF3, and CF4. [ABSTRACT FROM AUTHOR]

The lipemic response to a high-fat meal is greater in men than in women. However, sex-related differences in the metabolic fate of ingested fat are not well understood. The purpose of this study was to measure the recovery of ingested fat in plasma fractions of chylomicrons (CHYLO), VLDL, and plasma fatty acids, as well as in expired breath (i.e., oxidation) in men and women. Nonobese subjects (n = 10; 5 men, 5 women) consumed 0.7 g fat/kg body weight containing 7 mg/kg of [1,1,1-13C]-trioleate the morning after an overnight fast. Plasma total triglyceride (TG) concentration and 13C recovery in the CHYLO, VLDL, and plasma fatty acid fractions, as well as expired breath samples, were measured over the 11-h period after the meal. Plasma total TG excursion was greater (P < 0.05) in men than in women during the 11-h period after the meal. Similarly, the recovery of the ingested tracer-labeled fat in the CHYLO fraction was greater in men than in women (main effect for sex; P < 0.05). Recovery of ingested tracer-labeled fat in VLDL, the plasma fatty acid fraction, and expired breath did not differ in men and women. Therefore, the elevated postprandial lipemia found in men compared with women was due to a prolonged availability of the lipid in chylomicrons, but was not related to differences in oxidation rates or incorporation of the ingested lipid into VLDL by the liver.
