Loop-mediated isothermal amplification or LAMP has been identified to be an efficient technology for point-of-care diagnostics. Paper-based LAMP technique has tremendous potential in replacing the existing tube-based technology as the manufacturing cost of a paper-based device is comparatively lower and easy-to-use. LAMP-based paper diagnostic device for Mycobacterium tuberculosis (MTB) detection is of extreme importance as it will help in early and rapid diagnosis of the affected patients. The fabrication of these devices requires assessment of design parameters on the extent of LAMP amplification reaction. Hence, CFD studies would be extremely beneficial from the design perspective. The current work presents an insight into the CFD simulations for LAMP amplification reaction on a porous paper membrane (nitrocellulose membrane). The convection-diffusion-reaction model is solved on a COMSOL Multiphysics 5.0 platform. Studies on effect of pore size, aspect ratio and initial DNA concentration on the extent of DNA amplification reaction have been carried out. The current paper-based technique is effective in detecting a minimum of 5 copies of DNA contrasting the previous semi-quantitative technique which demonstrated the detection of minimum 98 copies. Overall, the simulation results displayed almost 96% enhancement in the DNA amplification rate on paper membrane. In cooperation with the world’s second largest manufacturer of beverage cartons (SIG Combibloc®) for liquid foodstuffs an innovative off-take for neutralisation of reactive gas in a paper laminating machine was constructed. A great challenge during engineering work was ensuring a high concentration of the reactive gas where needed and at the same time minimising work place impact in a machine basically without housing. Preliminary 2D-models of the machine geometry proved to be insufficient in describing all the governing flow phenomena. A simplified 3D-geometry containing all important parts of the complex machinery was necessary for accurate predictions. It was found that the driving force of air movement and transport of reactive gas (which acts as an adhesive agent) from the reaction zone in the interior of the laminating machine to the outside is a boundary flow caused by the rapid movement of carton material and rotating cylinders. A physically correct simulation result of the boundary flow is a premise for correct prediction of air flow in and around the machinery. Lacking experimental data (due to an inaccessible geometry) a worst case scenario was constructed by generating a grid and using turbulence models that maximised mass transport in the boundary layer region and thus emission of (tracer)gas from the machine. CFD simulations were done using the geometry preprocessor Gambit ™ , and the finite volume solver Fluent ™ . The results of the analysis of the emission paths from the machine were surprising and led to the construction of an effective off-take relatively far away from the emission source. The chosen position ensures low disturbance of highly sensitive flow patterns inside the machine and diffusive mixing, dilution and contamination of the surroundings. The effect of the new off-take is an immediate and significant rise in air quality in the vicinity of the laminating machine and ensures maximum allowed concentration in the plant area. The product quality furthermore is uncompromised by the working off-take which was another important goal of this work.

A good understanding of the hydraulic performance of aerated lagoons is required for their design and operation. A comprehensive numerical procedure has been developed for the three-dimensional computational modelling of the flow in large lagoons including high-speed floating mechanical surface aerators. This paper describes the procedure that consists of separate aerator modelling, then applying the obtained results as boundary data for a full lagoon model. A model application to an industrial aerated lagoon serves as an example of flow analysis. Post processing of the results by calculating the local average residence time (age of fluid) provides a powerful and intuitive technique to visualize and analyse the lagoon performance. The model has been verified by comparing the local average residence time predictions with measurements from a dye study. It is shown that the numerical modelling proposed is feasible and constitutes an effective new tool in improving the performance and design of industrial lagoons.

The absence of a systematic scale-up approach for biological conversion of cellulosic biomass to commodity products is a significant bottleneck to realizing the potential benefits offered by such conversion. Motivated by this, we undertook to develop a scale-up approach for conversion of waste paper sludge to ethanol. Physical properties of the system were measured and correlations were developed for their dependence upon cellulose conversion. Just-suspension of solid particles was identified as the scale up criterion based on experiments at lab scale. The impeller speed for just solids suspension at large scale was predicted using computational fluid dynamics simulations. The scale-up strategy was validated by analyzing mixing requirements such as solid-liquid mass transfer under the predicted level of agitation at large scale. The scale-up approach enhances the prediction of reactor performance and helps provide guidelines for the analysis and design of large scale bioreactors based on bench scale experimentation.
