The recentwidespread mortality and tissue loss in Florida from stony coral tissue loss disease (SCTLD) has propelled the need for assisted reproduction to restore reefs, especially for the ESA listed species Orbicella faveolata. In situ gamete collection can be challenging due to the weather and resources (boats and divers) required during the expected spawning window. In the northern portion of the Florida coral reef tract, coral spawn collection has been even more difficult due to historical inconsistency in annual spawning times and the potential for "zombie" corals, i.e. large but reproductively senescent individuals. Sexual reproduction is a crucial process for reef building coral populations to maximize genetic diversity and recover from large scale disturbances. Mass spawning events by Acropora species represent critical opportunities for populations to persist, and a process that is increasingly exploited to actively restore degraded reefs. However, the timing and predictive capacity of coral spawning throughout the broad thermal and environmental regime of the Red Sea – a region also undergoing significant development and active reef restoration – remains patchy. We, therefore, conducted three parallel reef surveys in the central Red Sea (Al-Fahal Reef, Thuwal - Saudi Arabia) and the eastern (Shushah Island - Saudi Arabia) and western (Hurghada – Egypt) coast of the northern Red Sea. Surveys assessed the gravidity of gonads, spawning timing, alignment with the lunar cycle of 21 Acropora spp. (total n= 572 colonies) around the full moons of April and May 2023. Consistent with past observations, synchronous spawning was observed for Acropora spp. in both the central and northern Red Sea during April and May, respectively. Interestingly, corals spawned on the full moon in both Shushah and Thuwal sites. In contrast, corals in Hurghada were independent of the lunar cycle and spawned 7-9 nights before the full moon in May. By integrating our 2023 observations with the historical spawning events in Hurghada and Thuwal (2002-2022), we found that the deviation of spawning timing from the full moon day was correlated with absolute Sea Surface Temperature (SST) (earlier spawning before the full moon day, lower SST) and warming rate (earlier spawning, more rapid warming) in 6- weeks prior to spawning. As such, temperature pattern is likely one of the primary factors governing gamete release, among other factors, that likely influence spawning day within the lunar month. These correlations between SST metrics and spawning timing suggest a potential framework to predict future Acropora spp. spawning dates. Our observations demonstrate the importance of parallel efforts across borders to collect critical data needed to inform management strategies aimed at conserving and restoring coral reefs in this ecologically diverse region. Most stony corals liberate their gametes into the water column via broadcast spawning, where fertilization hinges upon the activation of directional sperm motility. Sperm from gonochoric and hermaphroditic corals display distinct morphological and molecular phenotypes, yet it is unknown whether the signalling pathways controlling sperm motility are also distinct between these sexual systems. Here, we addressed this knowledge gap using the gonochoric, broadcast spawning coral Astrangia poculata. We found that cytosolic alkalinization of sperm activates the pH-sensing enzyme soluble adenylyl cyclase (sAC), which is required for motility. Additionally, we demonstrate for the first time in any cnidarian that sAC activity leads to protein kinase A (PKA) activation, and that PKA activity contributes to sperm motility activation. Ultrastructures of A. poculata sperm displayed morphological homology with other gonochoric cnidarians, and sAC exhibited broad structural and functional conservation across this phylum. These results indicate a conserved role for pH-dependent sAC-cAMP-PKA signalling in sperm motility across coral sexual systems, and suggest that the role of this pathway in sperm motility may be ancestral in metazoans. Finally, the dynamics of this pH-sensitive pathway may play a critical role in determining the sensitivity of marine invertebrate reproduction to anthropogenic ocean acidification. Acropora palmata is a foundational yet endangered Caribbean reef-building coral species. The lack of recovery after a disease outbreak and low recruitment has led to widespread use of fragmentation to restore populations. Another option is the production of sexual recruits (settlers) via assisted reproduction to improve the genetic diversity of depleted populations; however, the viability of this approach has not been tested over the long term. In 2011 and 2012, A. palmata larvae were cultured, settled, and the sexual recruits raised in an ex-situ nursery. Survival and growth were monitored over time. In 2014, these two F1 cohorts were moved to an in-situ nursery and after one year, a subset (29 colonies) was outplanted onto Cuevones Reef in the Mexican Caribbean. Growth and survival of these colonies were monitored periodically and compared to colonies that remained in the in-situ nursery. In 2019, samples were collected and analyzed for fertility and fecundity. 53% of the colonies were gravid and fecundity was 5.61 ± 1.91 oocytes and 3.04 ± 0.26 spermaries per polyp. A further 14 colonies from these two cohorts were outplanted in 2020 onto Picudas Reef and monitored during the subsequent spawning seasons. Two years after outplanting onto Picudas Reef, all colonies were alive and spawning of three of these colonies was recorded in 2022 in synchrony with the wild population. Gametes were collected from two colonies and crossed, with 15% fertilization success. Spermatozoa from wild colonies were then added and fertilization success increased to 95%. The resultant larvae followed normal development and symbiont uptake was visible within two weeks. The F2 generation was settled, maintained in an ex-situ nursery, and monitored for survival and growth. Both F1 and F2 generations followed a Type III survival curve with high initial mortality while in the ex-situ nursery and low later-stage mortality. The growth rates of these colonies increased three-fold after outplanting when compared to their growth rates in the ex-situ and in-situ nurseries. All colonies survived while in the in-situ nursery and for an additional nine years after outplanting onto Cuevones Reef. Overall, our results show that colonies produced by assisted breeding, once outplanted, may contribute to the genetic diversity and establishment of self-sustaining sexually-reproducing populations, which is an overarching goal of coral restoration programs. The continuing impact of local and global stressors on coral reefs worldwide is prompting the exploration of novel approaches aimed at mitigating and improving the bleak future projections for corals. Ex situ aquaculture has the potential to provide a standardized supply of source corals to support active intervention-based research endeavours as well as complementary reef restoration initiatives. To effectively achieve this supply of coral, however, ex situ aquaculture systems need to be able to efficiently maintain reproducing colonies, and have the capacity to support each coral life stage. We monitored the monthly reproduction of the brooding coral, Pocillopora acuta, for one year in two distinct aquaculture systems: a natural seawater-supplied flow-through system (FTS) and an artificial seawater-supplied recirculating aquaculture system (RAS). The coral colonies in both aquaculture systems showed relatively consistent reproduction over time (on average > 70% of all colonies reproducing each month) and maintained natural diel reproduction patterns even after 1 year of ex situ culture. Coral colony reproductive output and timing (i.e., lunar day of release) differed between aquaculture systems in some, but not all, seasons. Planulae released from colonies settled and grew to reproductive size after ~16 months of culture, resulting in the ex situ production of an F2 generation of a brooding coral within two distinct aquaculture systems. This demonstrated that the use of ex situ aquaculture could be directly applied to research, restoration, and conservation aimed at mitigating coral decline in a future marred by climate change and local impacts. As assisted sexual reproduction interventions continue to become embedded within coral reef restoration initiatives, it is important to understand the potential for trade-offs between key traits like reproductive output and disease resistance. Oocyte size and fecundity, quantitative measures of reproductive output and important life-history traits, can be used as proxies for coral reproductive success and health. Sexual reproduction, particularly gametogenesis, is an energetically costly process and at the physiological level, trade-offs are caused by competitive allocation of limited resources to various functions. However, resource allocation trade-offs may also have a genetic basis, and thus, different genets may differ in these aspects. Therefore, the purpose of this study was to assess the reproductive output of A. cervicornis genets with known white-band disease resistance or susceptibility by quantifying the number and size of oocytes within colonies maintained within Mote Marine Laboratory’s offshore coral spawning nursery in the Lower Florida Keys, USA. We also quantified the number of eggs and sperm packaged within gamete bundles that were collected during the August 2020 spawning event. Consistent with previous studies, we found a positive correlation between colony size and fecundity. Interestingly though, we found no evidence for a trade-off between disease resistance and reproductive output and instead found a negative correlation between disease susceptibility and oocyte size. These data are relevant for population management interventions and for managing broodstock used for active restoration where a suite of corals with different genotypes and phenotypes are continuously propagated and outplanted. Having a more comprehensive understanding of the fitness differences among candidates can help guide such efforts and ensure that a diversity of fit genets is used for restoration, which should ultimately support greater adaptive potential and population resilience.

Many species across a wide range of taxa and habitats display phenological shifts and differences in response to both environmental gradients and climate change. Moreover, the wide‐scale decline of numerous ecosystems is leading to increasing efforts to identify zones that might serve as natural refuges from various disturbances, including ocean warming. One such refuge was suggested to be that of the deep coral reefs, but whether depth can provide coral populations with a viable and reproductive refuge remains unclear. Given the global coral‐reef degradation and the key role that corals play as ecosystem engineers, their reproductive ecology has been widely studied. A particular knowledge gap nonetheless exists regarding coral reproductive phenology along a depth gradient. Filling in this gap may uncover the environmental cues that regulate coral reproduction, leading to better predictions of population connectivity, and their possible responses to climate change and other environmental changes. Here, using long‐term in situ observations of the soft coral Rhytisma fulvum's reproductive activity along its entire depth range (0–45 m), we examined the relationship among several environmental factors and the coral's reproductive phenology and activity over five successive annual breeding seasons. Compared with the shallow depths, a lower number of reproducing colonies was found in habitats deeper than 30 m, highlighting possible constraints on coral reproduction at the deeper end of their range. Our results further revealed that an increase in seawater temperature over 1–2‐day intervals during the breeding season correlated with the onset of reproductive activity along the depth gradient, leading to different reproductive periodicities in different depths. These differences suggest that differential temperature regimes and reproductive timing across depth may create intraspecific temporal reproductive segregation, possibly reducing connectivity among populations along a depth gradient. Moreover, we found high variability among years in both the timing of breeding activities and in the level of reproductive synchrony among corals from different depths. Midway Atoll in the Northwestern Hawaiian Islands is home to ground nesting birds that are threatened by invasive mice. Planned rodent eradication efforts for the island involve aerial application of cereal bait pellets containing the chemical rodenticide brodifacoum. Given the nature of the application method, drift of cereal bait pellets into the coastal waters surrounding Midway Atoll is unavoidable. To understand whether cereal bait pellets impact marine invertebrates, gametes and larvae of the reef-building coral Montipora capitata were exposed to brodifacoum, cereal bait pellets containing brodifacoum, and inert cereal bait pellets without the rodenticide. Fertilization success and larval survival were assessed at nominal brodifacoum concentrations of 1, 10, and 100 ppb. Fertilization success decreased by 15% after exposure to 100 ppb brodifacoum solutions. Larval survival was not reduced by exposure to brodifacoum solutions. Cereal bait pellets containing brodifacoum reduced fertilization success at 10 ppb brodifacoum in 0.4 g per L pellet solutions by 34.84%, and inhibited fertilization at 100 ppb brodifacoum in 4 g of pellet per L solution. Inert cereal bait pellets had similar effects, reducing fertilization success at 0.4 g of pellet per L by 40.50%, and inhibiting fertilization at 4 g per L pellet solutions. Larval survival was reduced by >43% after prolonged exposure to 4 g per L pellet solutions. The highest concentration used in this study was meant to represent an extreme and unlikely condition resulting from an accidental spill. Our findings indicate large amounts of cereal bait pellets entering the coastal environment of Midway Atoll, if occurring during a coral spawning event, would reduce coral reproduction by decreasing fertilization success. It is difficult to know the ecologically relevant concentrations of cereal bait pellets in coastal environments due to unavoidable bait drift after land applications, but results indicate small amounts of pellet drifting into coastal environments would not severely reduce coral reproductive capacity. Best management practices should consider known coral reproductive periods when scheduling applications of pellets on tropical islands to reduce the risk of negative impacts of large-scale accidents on corals. The restoration of reefs damaged by global and local pressures remains constrained by the scale of intervention currently feasible. Traditional methods for ex situ sexual propagation of corals produce limited materials, typically of limited genetic diversity and only sufficient for small field trials. The development and validation of new technologies to upscale and automate coral propagation is required to achieve logistically and financially feasible reef restoration at ecologically relevant scales. To address the need for upscaled production of genetically diverse material for use in reef restoration we designed an automated system (the AutoSpawner) for harvesting, fertilising and washing gametes from tropical broadcast-spawning corals. The system includes a novel high density dynamic fertilisation process, which enables the production of large numbers of fertilised coral eggs (>7 million per night for highly fecund species) without any downstream negative effects on larval quality. The functionality of the system and the quality of the produced larvae was assessed using multiple species from two coral families (Acroporidae and Merulinidae) across a range of spawning and gamete characteristics. We present the schematics and protocols required for automated sexual propagation of high-quality coral larvae using this novel system; and demonstrate that the time demands, and labour costs, associated with traditional manual-based sexual propagation of corals can be reduced by up to 113-fold using the AutoSpawner. • An automated system for harvest & fertilisation of coral spawn was developed & tested. • The system was successfully used for multiple species of broadcast spawning corals. • Larvae produced were of equal quality to larvae produced using traditional methods. • Time and labour required for larval production can be reduced up to 113-fold. • The AutoSpawner resolves some key barriers to upscaling of reef restoration efforts. Reef development occurs commonly under oligotrophic conditions that favor corals over competitors. In the Eastern Tropical Pacific (ETP), coral communities develop under highly dynamic environmental conditions that may benefit organisms involved in reef bioerosion. To understand how coral performance and maintenance change with increasing urban pressure in the ETP, we evaluated coral reproductive activity, calcification, and bioerosion in two representative coral genera in three Central Mexican Pacific sites. We hypothesize that stressors associated to urban development will negatively affect coral performance while enhancing bioerosion. Despite moderate to high levels of nitrite, nitrate, and phosphate and highly variable sedimentation rates, coral performance was not hampered in any site. Coral growth in Pocillopora and Pavona corals exhibited typical growth rates (~ 6.6 gCaCO3 cm−2 year−1 and ~ 1.1 gCaCO3 cm−2 year−1, respectively) and colonies were reproductively active from March to November. However, 76.5% of all coral colonies were invaded by excavating sponges. In Pavona colonies, ~ 24.37% of total CaCO3 volume was removed by bioeroders. While coral performance is not hampered by the current nutrient levels and sedimentation regimes, these conditions may increase the abundance of bioeroders such as sponges, leading to higher bioerosion rates that compromise reef integrity and functionality. The continuing impact of local and global stressors on coral reefs worldwide is prompting the exploration of novel approaches aimed at mitigating and improving the bleak future projections for corals. Ex situ aquaculture has the potential to provide a standardized supply of source corals to support active intervention-based research endeavours as well as complementary reef restoration initiatives. To effectively achieve this supply of coral, however, ex situ aquaculture systems need to be able to efficiently maintain reproducing colonies, and have the capacity to support each coral life stage. We monitored the monthly reproduction of the brooding coral, Pocillopora acuta, for one year in two distinct aquaculture systems: a natural seawater-supplied flow-through system (FTS) and an artificial seawater-supplied recirculating aquaculture system (RAS). The coral colonies in both aquaculture systems showed relatively consistent reproduction over time (on average > 70% of all colonies reproducing each month) and maintained natural diel reproduction patterns even after 1 year of ex situ culture. Coral colony reproductive output and timing (i.e., lunar day of release) differed between aquaculture systems in some, but not all, seasons. Planulae released from colonies settled and grew to reproductive size after ~16 months of culture, resulting in the ex situ production of an F2 generation of a brooding coral within two distinct aquaculture systems. This demonstrated that the use of ex situ aquaculture could be directly applied to research, restoration, and conservation aimed at mitigating coral decline in a future marred by climate change and local impacts.

The survival of coral reefs largely depends among other factors on the recruitment of a new generation of coral individuals that are more adapted to a rapidly changing climate and other anthropogenic stressors (e.g., pollution, sedimentation). Therefore, a better understanding of the coral settlement process, the molecules involved as well as crucial environmental drivers that control settlement success are needed. In this study, we identified a novel settlement inducer for the brooding scleractinian coral Leptastrea purpurea and highlight the importance of light for the settlement process. Crude extract of the red-pigmented bacterium Pseudoalteromonas rubra reliably triggered attachment and metamorphosis in L. purpurea larvae in less than 24 h. Prodigiosin (II) and the two derivatives, cycloprodigiosin (I) and 2-methyl-3-hexyl prodiginine (III) were isolated and structurally elucidated from the crude extract of P. rubra. We demonstrated that the photosensitive pigment cycloprodigiosin (I) was the responsible compound for attachment and metamorphosis in L. purpurea larvae. Under the tested light regimes (i.e., darkness, constant light and a dark-light alternation), cycloprodigiosin (I) triggered approximately 90% settlement at a concentration of 0.2 μg mL–1 under a 12 h alternating dark-light cycle, mimicking the light-flooded coral reef environment. Our findings enable for the first time a mechanistic understanding of the light-dependent larva to polyp transformation by discovering the novel bacterial settlement cue cycloprodigiosin and its photosensitivity as a determining factor for coral settlement.

Assisted gene flow (AGF) is a conservation intervention to accelerate species adaptation to climate change by importing genetic diversity into at-risk populations. Corals exemplify both the need for AGF and its technical challenges; corals have declined in abundance, suffered pervasive reproductive failures, and struggled to adapt to climate change, yet mature corals cannot be easily moved for breeding, and coral gametes lose viability within hours. Here, we report the successful demonstration of AGF in corals using cryopreserved sperm that was frozen for 2 to 10 y. We fertilized Acropora palmata eggs from the western Caribbean (Curaçao) with cryopreserved sperm from genetically distinct populations in the eastern and central Caribbean (Florida and Puerto Rico, respectively). We then confirmed interpopulation parentage in the Curaçao-Florida offspring using 19,696 single-nucleotide polymorphism markers. Thus, we provide evidence of reproductive compatibility of a Caribbean coral across a recognized barrier to gene flow. The 6-mo survival of AGF offspring was 42%, the highest ever achieved in this species, yielding the largest wildlife population ever raised from cryopreserved material. By breeding a critically endangered coral across its range without moving adults, we show that AGF using cryopreservation is a viable conservation tool to increase genetic diversity in threatened marine populations. Reports of synchronous spawning in corals of Lakshadweep reefs, India have been purely anecdotal. To understand factors that underlie coral spawning in Lakshadweep, we studied four Acropora species (A. muricata, A. cytherea, A. hemprichii and A. nobilis) in Kavaratti atoll from February to March 2018. These species were observed in situ for oocyte maturation, and the larvae of species A. muricata and A. cytherea were assessed for survival and settlement rates. Observations on oocyte maturation indicated possible spawning closer to the immediate full moon of species in which 7-28% of the colonies had mature oocytes. Thus, A. muricata, A. cytherea and A. hemprichii spawned on 25 and 26 February 2018, demonstrating multi-specific synchronous spawning, whereas A. nobilis spawned on 21 March 2018. Larval survival rates revealed a steep die-off beginning on day 12 postspawning and maximum survival up to 24-27 days. Settlement occurred between 12 and 20 days for A. muricata and between 12 and 22 days for A. cytherea. The comparatively low survival duration and fewer larvae surviving for settlement show that maximum settlement could be heavily compromised. However, the study reveals that the comparatively late onset of settlement (from day-12, post-spawning) and the broader settlement window of 8-10 days could allow room for long-distance dispersal of larvae. Members of the family Meandrinidae are highly susceptible to stony coral tissue loss disease, resulting in population reductions up to 88% in both Dendrogyra cylindrus and Meandrina meandrites along the Florida Reef Tract. Reductions in abundance on this scale leave these species susceptible to limitations in sexual reproduction and natural recovery without intervention. In response to the ongoing outbreak of the disease across the Caribbean, a variety of genetic rescue projects have been implemented to bring disease susceptible species into ex situ culture and preserve living genetic diversity. In this study, corals being held in a long-term ex situ genetic bank were maintained using artificial lighting and temperature cues programmed to mimic natural cycles in Key Largo, FL, United States. Synchronized broadcast spawning events in both species were documented in aquaria over two annual spawning cycles in 2019 and 2020. Timing of gamete release relative to the perceived date and sunset was highly synchronized with wild observations. Up to 21 unique D. cylindrus genotypes collected from reef locations spanning over 230 km contributed gametes to the larval pool. The majority of these parental colonies are no longer alive in the wild. Repeatable and predictable ex situ spawning events such as these will become an essential tool for managed breeding and assisted fertilization in species suffering from severe population declines. These annual events have the potential to produce thousands of genetically diverse offspring for restoration efforts and offer future hope for the long-term survival of these threatened species.

The effects of the xenoestrogen 4-nonylphenol (4NP) on endocrine and metabolic homeostasis in the reef building coral, Pocillopora damicornis were investigated. The aim was to understand if ubiquitous nonylphenol ethoxylate contaminants in the marine environment result in altered homeostatic function. Coral colonies were chronically exposed (6 weeks) to a sublethal concentration (1 ppb) of 4NP and sampled over the coral’s lunar reproductive cycle. Although activity of steroidogenic enzymes [cytochrome P450 (CYP) 17, CYP 19, and 3-β-Hydroxysteroid dehydrogenase] and the conjugation enzyme glutathione-S-transferase was not altered, significant increases in the activity of the steroid clearing enzyme UDP-glycosyltransferase (UGT) were observed. The natural fluctuation of UGT activity with the lunar cycle was replaced with consistently high UGT activity throughout the reproductive cycle during 4NP exposure. No effect of 4NP on the reverse reaction, mediated by β-glucuronidase, was observed. Thus, 4NP shifts the UGT:β-glucuronidase ratio toward greater clearance at points in the lunar cycle where retention of compounds is typically favored. Additionally, 4NP reduced activity of the steroid regeneration enzyme steroid sulfatase, further shifting the system toward clearance rather than regeneration. These data imply that environmentally relevant levels of 4NP may be impacting the reproductive health of corals and threatening the persistence of coral reefs.

The discovery of multi-species synchronous spawning of scleractinian corals on the Great Barrier Reef in the 1980s stimulated an extraordinary effort to document spawning times in other parts of the globe. Unfortunately, most of these data remain unpublished which limits our understanding of regional and global reproductive patterns. The Coral Spawning Database (CSD) collates much of these disparate data into a single place. The CSD includes 6178 observations (3085 of which were unpublished) of the time or day of spawning for over 300 scleractinian species in 61 genera from 101 sites in the Indo-Pacific. The goal of the CSD is to provide open access to coral spawning data to accelerate our understanding of coral reproductive biology and to provide a baseline against which to evaluate any future changes in reproductive phenology. The declining reproductive viability of corals threatens their ability to adapt to changing ocean conditions. It is vital that we monitor this viability quantitatively and comparatively. Computer-assisted sperm analysis (CASA) systems offer in-depth analysis used regularly for domestic and wildlife species, but not yet for coral. This study proposes quality control procedures and CASA settings that are effective for coral sperm analysis. To resolve disparities between CASA measurements and evaluations by eye, two negative effects on motility had to be resolved, slide adhesion (procedural) and sperm dilution (biological). We showed that the addition of bovine serum albumin, or caffeine, or both to fresh sperm reduced adhesion in the CASA cassettes, improved motility and motile sperm concentration (P < 0.0001), yet these additions did not affect measurements of total sperm concentration. Diluting coral sperm reduced sperm motility (P = 0.039), especially from heat-stressed corals. We found CASA concentration counts comparable to haemocytometer and flow cytometer measures (P = 0.54). We also found that motile sperm per egg is a useful predictor of fertilisation success, using cryopreserved sperm. Standard measurements of coral reproductive characteristics inform our understanding of the impacts of climate change on reef populations; this study provides a benchmark to begin this comparative work. As reef waters warm due to climate change, episodic largescale tropical storms are becoming more frequent, drastically altering the near shore water quality for short periods of time. Therefore, it is critical that we understand the effects warming waters, fresh water input, and run-off have on sexual reproduction of coral. To better understand the effects of these near shore stressors on Hawaiian coral, laboratory experiments were conducted at the Institute of Marine Biology to determine the independent effects of suspended sediment concentrations (100 mg l−1 and 200 mg l−1), lowered salinity (28‰), and elevated temperature (31 °C) on the successful fertilization, larval survival, and settlement of the scleractinian coral Montipora capitata. In the present study, early developmental stages of coral were exposed to one of three near shore stressors for a period of 24 h and the immediate (fertilization) and latent effects (larval survival and settlement) were observed and measured. Fertilization success and settlement were not affected by any of the treatments; however, larval survival was negatively affected by all of the treatments by 50% or greater (p > 0.05). These data show that early life stages of M. capitata may be impacted by near shore stressors associated with warming and more frequent storm events.

The extent to which populations persist under environmental stress depends on the reproductive output of individuals that survive the stress. In coral systems, corals bleach in response to stress from elevated water temperature. However, little is known of the extent to which thermal stress impairs the reproductive capacity of the survivors over the following years, limiting the capacity to predict how populations will persist in the Anthropocene.Using histology to quantify the abundance and size of oocytes and spermaries per polyp, we tested how bleaching impairs the reproductive response of the coral Pocillopora meandrina over two reproductive seasons following the 2015 mass bleaching event in the Hawaiian Islands.We found that smaller colonies not only had a greater probability of bleaching but also suffered greater reproductive impacts over a longer time. In contrast, larger colonies generated comparable reproductive output regardless of bleaching severity, although bleached colonies generated smaller oocytes the year after bleaching.These results show that reproductive impacts of bleaching are more complex and size‐specific than commonly assumed. Therefore, estimates of bleaching mortality may underestimate the true impact of thermal stress on populations, especially as populations lose larger individuals from repeated and co‐occurring stressors. Research with coral embryos and larvae often requires laborious manual counting and sorting of individual specimens, usually via microscopy. Because many coral species spawn only once per year during a narrow temporal window, sample processing is a time-limiting step for research on the early life-history stages of corals. Flow cytometry, an automated technique for measuring and sorting particles, cells, and cell-clusters, is a potential solution to this bottleneck. Yet most flow cytometers do not accommodate live organisms of the size of most coral embryos (> 250 µm), and sample processing is often destructive. Here we tested the ability of a large-particle flow cytometer with a gentle pneumatic sorting mechanism to process and spectrally sort live and preserved Montipora capitata coral embryos and larvae. Average survival rates of mechanically-sorted larvae were over 90% and were comparable to those achieved by careful hand-sorting. Preserved eggs and embryos remained intact throughout the sorting process and were successfully sorted based on real-time size and fluorescence detection. In-line bright-field microscopy images were captured for each sample object as it passed through the flow-cell, enabling the identification of early-stage embryos (2-cell to morula stage). Samples were counted and sorted at an average rate of 4 s larva−1 and as high as 0.2 s larva−1 for high-density samples. Results presented here suggest that large-particle flow cytometry has the potential to significantly increase efficiency and accuracy of data collection and sample processing during time-limited coral spawning events, facilitating larger-scale and higher-replication studies with an expanded number of species. As coral reefs continue to degrade globally due to climate change, considerable effort and investment is being put into coral restoration. The production of coral offspring via asexual and sexual reproduction are some of the proposed tools for restoring coral populations and will need to be delivered at scale. Simple, inexpensive, and high-throughput methods are therefore needed for rapid analysis of thousands of coral offspring. Here we develop a machine learning pipeline to rapidly and accurately measure three key indicators of coral juvenile fitness: survival, size, and color. Using machine learning, we classify pixels through an open-source, user-friendly interface to quickly identify and measure coral juveniles on two substrates (field deployed terracotta tiles and experimental, laboratory PVC plastic slides). The method’s ease of use and ability to be trained quickly and accurately using small training sets make it suitable for application with images of species of sexually produced corals without existing datasets. Our results show higher accuracy of survival for slides (94.6% accuracy with five training images) compared to field tiles measured over multiple months (March: 77.5%, June: 91.3%, October: 97.9% accuracy with 100 training images). When using fewer training images, accuracy of area measurements was also higher on slides (7.7% average size difference) compared to tiles (24.2% average size difference for October images). The pipeline was 36× faster than manual measurements. The slide images required fewer training images compared to tiles and we provided cut-off guidelines for training for both substrates. These results highlight the importance and power of incorporating high-throughput methods, substrate choice, image quality, and number of training images for measurement accuracy. This study demonstrates the utility of machine learning tools for scalable ecological studies and conservation practices to facilitate rapid management decisions for reef protection.

Sexually produced larvae are used in various fields of coral research. Because the vast majority of scleractinians reproduces only on one or few occasions per year through simultaneous release of gametes, and because an ex situ spawning induction is still very hard to achieve, great efforts are required to obtain planula larvae. Brooding corals have been used to harvest planulae although their larvae often differ in various traits from most spawning corals, e.g., in settlement behavior. Other cnidarians, such as Aiptasia spp., have been substituting for scleractinians in many aspects of coral research. However, organisms such as Aiptasia differ strongly from scleractinians, thus limiting the transferability of obtained results. This study examines the potential of Leptastrea purpurea as a reliable source of larvae for coral research. Larval output as well as settlement behavior of planulae was investigated. Our results show that colonies of L. purpurea released a daily average of 3.7 (±0.2) larvae during a period of 65 days, thus allowing continual access to planula larvae. We collected a total of 58127 larvae from our broodstock of 243 colonies. Larval settlement is induced by the same and/or similar cues as in many spawning species which increases the transferability of conclusions. We discuss the utility of L. purpurea for research on scleractinian physiology, ecology and larval settlement and conclude that L. purpurea is a well-suited organism to accelerate progress in many fields of coral research.

Abstract: Mesophotic coral ecosystems (i.e., deep coral reefs at 30–120 m depth) appear to be thriving while many shallow reefs in the world are declining. Amid efforts to understand and manage their decline, it was suggested that mesophotic reefs might serve as natural refuges and a possible source of propagules for the shallow reefs. However, our knowledge of how reproductive performance of corals alters with depth is sparse. Here, we present a comprehensive study of the reproductive phenology, fecundity, and abundance of seven reef‐building conspecific corals in shallow and mesophotic habitats. Significant differences were found in the synchrony and timing of gametogenesis and spawning between shallow and mesophotic coral populations. Thus, mesophotic populations exhibited delayed or protracted spawning events, which led to spawning of the mesophotic colonies in large proportions at times where the shallow ones had long been depleted of reproductive material. All species investigated demonstrated a substantial reduction in fecundity and/or oocyte sizes at mesophotic depths (40–60 m). Two species (Seriatopora hystrix and Galaxea fascicularis) displayed a reduction in both fecundity and oocyte size at mesophotic depths. Turbinaria reniformis had only reduced fecundity and Acropora squarrosa and Acropora valida only reduced oocyte size. In Montipora verrucosa, reduced fecundity was found during one annual reproductive season while, in the following year, only reduced oocyte size was found. In contrast, reduced oocyte size in mesophotic populations of Acropora squarrosa was consistent along three studied years. One species, Acropora pharaonis, was found to be infertile at mesophotic depths along two studied years. This indicates that reproductive performance decreases with depth; and that although some species are capable of reproducing at mesophotic depths, their contribution to the replenishment of shallow reefs may be inconsequential. Reduced reproductive performance with depth, combined with the possible narrower tolerance to environmental factors, further suggests that mesophotic corals may in fact be more vulnerable than previously conceived. Furthermore, we posit that the observed temporal segregation in reproduction could lead to assortative mating, and this, in turn, may facilitate adaptive divergence across depth. The future of coral reefs under increasing CO2 depends on their capacity to recover from disturbances. To predict the recovery potential of coral communities that are fully acclimatized to elevated CO2, we compared the relative success of coral recruitment and later life stages at two volcanic CO2 seeps and adjacent control sites in Papua New Guinea. Our field experiments showed that the effects of ocean acidification (OA) on coral recruitment rates were up to an order of magnitude greater than the effects on the survival and growth of established corals. Settlement rates, recruit and juvenile densities were best predicted by the presence of crustose coralline algae, as opposed to the direct effects of seawater CO2. Offspring from high CO2 acclimatized parents had similarly impaired settlement rates as offspring from control parents. For most coral taxa, field data showed no evidence of cumulative and compounding detrimental effects of high CO2 on successive life stages, and three taxa showed improved adult performance at high CO2 that compensated for their low recruitment rates. Our data suggest that severely declining capacity for reefs to recover, due to altered settlement substrata and reduced coral recruitment, is likely to become a dominant mechanism of how OA will alter coral reefs. Global environmental change, in marine ecosystems, is associated with concurrent shifts in water temperature, circulation, stratification, and nutrient input, with potentially wide-ranging biological effects. Variations in seawater temperature might alter physiological functioning, reproductive efficiency, and demographic traits of marine organisms, leading to shifts in population size and abundance. Differences in temperature tolerances between organisms can identify individual and ecological characteristics, which make corals able to persist and adapt in a climate change context. Here we investigated the possible effect of temperature on the reproductive output of the solitary non-zooxanthellate temperate coral Leptopsammia pruvoti, along an 8° latitudinal gradient. Samples have been collected in six populations along the gradient and each polyp was examined using histological and cyto-histometric analyses. We coupled our results with previous studies on the growth, demography, and calcification of L. pruvoti along the same temperature gradient, and compared them with those of another sympatric zooxanthellate coral Balanophyllia europaea to understand which trophic strategy makes the coral more tolerant to increasing temperature. The non-zooxanthellate species seemed to be quite tolerant to temperature increases, probably due to the lack of the symbiosis with zooxanthellae. To our knowledge, this is the first field investigation of the relationship between reproductive output and temperature increase of a temperate asymbiotic coral, providing novel insights into the poorly studied non-zooxanthellate scleractinians. The sexual pattern, reproductive mode, and timing of reproduction of Isophyllia sinuosa and Isophyllia rigida, two Caribbean Mussids, were assessed by histological analysis of specimens collected monthly during 2000–2001. Both species are simultaneous hermaphroditic brooders characterized by a single annual gametogenetic cycle. Spermatocytes and oocytes of different stages were found to develop within the same mesentery indicating sequential maturation for extended planulation. Oogenesis took place during May through April in I. sinuosa and from August through June in I. rigida. Oocytes began development 7–8 months prior to spermaries but both sexes matured simultaneously. Zooxanthellate planulae were observed in I. sinuosa during April and in I. rigida from June through September. Higher polyp and mesenterial fecundity were found in I. rigida compared to I. sinuosa. Larger oocyte sizes were found in I. sinuosa than in I. rigida, however larger planula sizes were found in I. rigida. Hermaphroditism is the exclusive sexual pattern within the Mussidae while brooding has been documented within the related genera Mussa, Scolymia and Mycetophyllia. This study represents the first description of the sexual characteristics of I. rigida and provides an updated description of I. sinuosa.

Larval production and recruitment underpin the maintenance of coral populations, but these early life history stages are vulnerable to extreme variation in physical conditions. Environmental managers aim to minimise human impacts during significant periods of larval production and recruitment on reefs, but doing so requires knowledge of the modes and timing of coral reproduction. Most corals are hermaphroditic or gonochoric, with a brooding or broadcast spawning mode of reproduction. Brooding corals are a significant component of some reefs and produce larvae over consecutive months. Broadcast spawning corals are more common and display considerable variation in their patterns of spawning among reefs. Highly synchronous spawning can occur on reefs around Australia, particularly on the Great Barrier Reef. On Australia’s remote north-west coast there have been fewer studies of coral reproduction. The recent industrial expansion into these regions has facilitated research, but the associated data are often contained within confidential reports. Here we combine information in this grey-literature with that available publicly to update our knowledge of coral reproduction in WA, for tens of thousands of corals and hundreds of species from over a dozen reefs spanning 20° of latitude. We identified broad patterns in coral reproduction, but more detailed insights were hindered by biased sampling; most studies focused on species of Acropora sampled over a few months at several reefs. Within the existing data, there was a latitudinal gradient in spawning activity among seasons, with mass spawning during autumn occurring on all reefs (but the temperate south-west). Participation in a smaller, multi-specific spawning during spring decreased from approximately one quarter of corals on the Kimberley Oceanic reefs to little participation at Ningaloo. Within these seasons, spawning was concentrated in March and/or April, and October and/or November, depending on the timing of the full moon. The timing of the full moon determined whether spawning was split over two months, which was common on tropical reefs. There were few data available for non-Acropora corals, which may have different patterns of reproduction. For example, the massive Porites seemed to spawn through spring to autumn on Kimberley Oceanic reefs and during summer in the Pilbara region, where other common corals (e.g. Turbinaria & Pavona) also displayed different patterns of reproduction to the Acropora. The brooding corals (Isopora & Seriatopora) on Kimberley Oceanic reefs appeared to planulate during many months, possibly with peaks from spring to autumn; a similar pattern is likely on other WA reefs. Gaps in knowledge were also due to the difficulty in identifying species and issues with methodology. We briefly discuss some of these issues and suggest an approach to quantifying variation in reproductive output throughout a year.

The direct impacts of coral diseases on coral populations have been assessed by quantifying coral tissue loss and colony mortality, but the determination of the indirect effects of diseases, such as disruptions in life history functions (e.g. reproduction, growth and maintenance), are more difficult to ascertain and have been scant. This study involved a comparison of various measures of reproductive output from histological slides of healthy tissue samples of Montastraea faveolata and tissue samples from colonies with white plague (WP) infections in Dominica (West Indies). Although the variability in the reproductive data was high, WP had significant negative impacts on the percentage of reproductive polyps per cm2, the percentage of reproductive mesenteries within a polyp, oocyte quantity per polyp, mean oocyte volume (mm3), and fecundity (oocyte volume per cm2 of tissue). However, these effects were only observed in the tissue directly impacted by the WP disease "band" and were not observed in tissue samples taken 20 cm away from the lesion. Summary Large environmental fluctuations often cause mass extinctions, extirpating species and transforming communities [ 1, 2 ]. While the effects on community structure are evident in the fossil record, demographic consequences for populations of individual species are harder to evaluate because fossils reveal relative, but not absolute, abundances. However, genomic analyses of living species that have survived a mass extinction event offer the potential for understanding the demographic effects of such environmental fluctuations on extant species. Here, we show how environmental variation since the Pliocene has shaped demographic changes in extant corals of the genus Orbicella , major extant reef builders in the Caribbean that today are endangered. We use genomic approaches to estimate previously unknown current and past population sizes over the last 3 million years. Populations of all three Orbicella declined around 2–1 million years ago, coincident with the extinction of at least 50% of Caribbean coral species. The estimated changes in population size are consistent across the three species despite their ecological differences. Subsequently, two shallow-water specialists expanded their population sizes at least 2-fold, over a time that overlaps with the disappearance of their sister competitor species O. nancyi (the organ-pipe Orbicella ). Our study suggests that populations of Orbicella species are capable of rebounding from reductions in population size under suitable conditions and that the effective population size of modern corals provides rich standing genetic variation for corals to adapt to climate change. Understanding species differences in demographic strategies is a fundamental goal of ecology. In scleractinian corals, colony morphology is tightly linked with many demographic traits, such as size-specific growth and morality. Here we test how well morphology predicts the colony size-fecundity relationship in eight species of broadcast-spawning corals. Variation in colony fecundity is greater among morphologies than between species with a similar morphology, demonstrating that colony morphology can be used as a quantitative proxy for demographic strategies. Additionally, we examine the relationship between size-specific colony fecundity and mechanical vulnerability (i.e., vulnerability to colony dislodgment). Interestingly, the relationship between size-specific fecundity and mechanical vulnerability varied among morphologies. For tabular species, the most fecund colonies are the most mechanically vulnerable, while the opposite is true for massive species. For corymbose and digitate colonies, mechanical vulnerability remains relatively constant as fecundity increases. These results reveal strong differences in the demographic tradeoffs among species of different morphologies. Using colony morphology as a quantitative proxy for demographic strategies can help predict coral community dynamics and responses to anthropogenic change. [ABSTRACT FROM AUTHOR]

During mass spawning of tropical corals, extremely dense accumulations of eggs develop throughout the water column, the fate of which depends on physical conditions, such as topography, currents, and prevailing winds. We measured egg transport [mainly acroporid corals and the palolo worm, Eunice viridis (Gray, 1847)] during a mass spawning at Ningaloo Reef, Western Australia, in April 2010. We tracked changes in egg and embryo abundance in the lagoon over five consecutive days with plankton tows at the sea surface immediately after spawning and four and 16 hours later, and characterized the flow regime with drifter deployments. Egg dispersal was rapid with a pronounced decrease in concentration within four hours of spawning, from approximately 106 to 101 and 104 n r3 for corals and E. viridis, respectively, on the peak date; few eggs or embryos remained within the lagoon after 16 hrs. Changes in concentration of eggs and embryos along transects inside the lagoon and perpendicular to the reef flat reflected the local hydrodynamics and high flushing rate, providing empirical support for a biophysical model from the same site. Global warming and ocean acidification affect coral calcification. Nevertheless, there is not enough information regarding the growth parameters of the main reef-building coral species in marginal growth areas such as the Pacific coast of southern Mexico. In order to fill this gap, coral growth parameters of 8 hermatypic coral species (massive species: Porites panamensis, Porites lobata, Pavona gigantea, and Pavona varians; branching species: Pocillopora meandrina, Pocillopora damicornis, Pocillopora verrucosa, and Pocillopora capitata) were estimated in 2 areas of the southern Mexican Pacific. Branching coral species had a higher calcification rate (2.99-5.23 g CaCO3 cm-2 yr-1) than massive species (0.34-1.13 g CaCO3 cm-2 yr-1). A significant relation between sea surface temperature (SST) and skeletal density was observed in all massive coral species. Also, 2 massive species (P. gigantea and P. varians) showed a significant relation between SST and calcification rate. Upwelling in the Gulf of Tehuantepec transports deep water with low pH and low aragonite saturation, and may be affecting the calcification rate of stony corals in the studied area. Coral reefs in the northern region of the Straits of Malacca have a diverse group of octocorals growing on its bed. The octocorals identified in this study are from islands along the Straits. In this study, 23 specimens were identified, belonging to 4 sub-orders, which have been subdivided into 8 families. From these 8 families, 15 different genera have been identified. The identification process for this research was conducted based on five important keys; the external form and colouration, polyps or colonial and fundamental structure of colonies, monomorphic or dimorphic, the arrangement of polyps, and the arrangement of sclerites. Coral spawning times have been linked to multiple environmental factors; however, to what extent these factors act as generalized cues across multiple species and large spatial scales is unknown. We used a unique dataset of coral spawning from 34 reefs in the Indian and Pacific Oceans to test if month of spawning and peak spawning month in assemblages of Acropora spp. can be predicted by sea surface temperature (SST), photosynthetically available radiation, wind speed, current speed, rainfall or sunset time. Contrary to the classic view that high mean SST initiates coral spawning, we found rapid increases in SST to be the best predictor in both cases (month of spawning: R2=0.73, peak: R2 = 0.62). Our findings suggest that a rapid increase in SST provides the dominant proximate cue for coral mass spawning over large geographical scales. Energetic responses of zooxanthellate reef corals along depth gradients have relevance to the refugia potential of mesophotic coral ecosystems (MCEs). Previous observations suggested that MCEs in the Caribbean are thermally buffered during the warmest parts of the year and occur within or just below the chlorophyll maximum, suggesting abundant trophic resources. However, it is not known if mesophotic corals can maintain constant energy needs throughout the year with changing environmental and biological conditions. The energetic content of tissues from the stony coral species Orbicella faveolata and Agaricia lamarcki was measured on the southern insular shelf of St. Thomas, US Virgin Islands (USVI), using micro-bomb calorimetry. Three sites for each species, at depths of 6m, 25m, 38m and 63m, were selected to capture energetic differences across the major vertical range extent of both species in the USVI—and sampled over five periods from April 2013 to April 2014. Mesophotic colonies of O. faveolata exhibited a significant reduction in energetic content during the month of September 2013 compared to mid-depth and shallow colonies (p = 0.032), whereas A. lamarcki experienced similar energetic variability, but with a significant reduction in energy content that occurred in July 2013 for colonies at sites deeper than 25m (p = 0.014). The results of calorimetric analyses indicate that O. faveolata may be at risk during late summer stress events, possibly due to the timing of reproductive activities. The low-point of A. lamarcki energy content, which may also coincide with reproduction, occurs prior to seasonal stress events, indicating contrasting, species-specific responses to environmental variability on MCEs. Caribbean coral reefs have experienced dramatic declines in live coral cover in recent decades. Primary branching framework Caribbean corals, Acropora cervicornis (Lamarck, 1816) and Acropora palmata (Lamarck, 1816), have suffered the greatest collapse. Coral Gardens, Belize, is one of few remaining, and perhaps the largest, refugia for abundant, healthy, but undocumented populations of both Acropora species in the Caribbean Sea. In the present study, GeoEye-1 multispectral satellite imagery of a 25 km2 reefal area near Ambergris Caye, Belize, was analyzed to identify live Acropora spp. cover. We used a supervised classification to predict occurrence of areas with live Acropora spp. and to separate them from other benthic cover types, such as sandy bottom, seagrass, and mixed massive coral species. We tested classification accuracy in the field, and new Acropora spp. patches were mapped using differential GPS. Of 11 predicted new areas of Acropora spp., eight were composed of healthy Acropora spp. An unsupervised classification of a red (Band 3):blue (Band 1) ratio calculation of the image successfully separated Acropora corals from other benthic cover, with an overall accuracy of 90%. Our study identified 7.58 ha of reef dominated by Acropora spp. at Coral Gardens, which is one of the largest populations in the Caribbean Sea. We suggest that Coral Gardens may be an important site for the study of modern Acropora spp. resilience.
