The aim of this thesis was the identification and analysis of candidate genes for stress response in silver fir (Abies alba Mill.). This ecologically and economically important forest tree species is native to many mountainous regions of Europe but little is known about its ecological characteristics. Silver fir populations were heavily transformed by human activity, which results in a mismatch between past and current distribution. Recent studies suggest that silver fir can occupy warmer and dryer climates than it currently does. However, the species also suffered considerably during the 1970s and 1980s, including foliar damage, radial growth depression and local diebacks in Germany. This is attributed mainly to the peak in air pollution during this period, especially sulfur dioxide (SO2), which seems to heavily increase drought sensitivity in silver fir. The combination of both stressors, SO2 and drought events, negatively affected silver fir even in regions where drought is usually not a problem. In the context of anthropogenic global climate change that will very likely lead to an increase in temperature in Europe and to more extreme events such as severe drought periods, the question arises, how silver fir will cope with these environmental changes. Given the speed of the predicted changes and the increasing landscape fragmentation, silver fir might not be able to evade it via seed dispersal. As a sessile organism, the only other option is adaptation, which will likely draw from standing genetic variation. To successfully predict the fate of silver fir, especially in the face of global climate change, and to potentially manage populations based on such predictions, the genetic architecture of silver fir in the context of such important stressors as drought and air pollution has to be understood. There exist, however, little genomic resources for silver fir and conifers in general. This is due to their large and complex genomes and the long generational cycle, which makes conifers typical nonmodel species. As such, methods for the identification of the genetic basis of stress response are effectively limited to a candidate gene approach. The candidate gene approach includes the identification of functional candidate genes by measuring differential gene expression between a stressed and a control group. In the context of this thesis, the water content of silver fir seedlings was monitored in a laboratory using a novel terahertz spectroscopy setup. One group of seedlings was regularly irrigated while the other group was drought stressed. Continually measuring the water content allowed to harvest needles from both groups at a time when the water status was comparable between the individuals within each group. A differential expression analysis between the needles from both groups then revealed 296 genes that were significantly up- or down-regulated in response to drought stress. Of those genes, approximately 45% have not been previously described in any organism and are potentially unique to silver fir or conifers in general. However, since only needles of seedlings were analyzed at a specific level of drought stress, the results are limited in scope to the source material and stress intensity and cannot be directly applied to silver fir or drought stress in general. Also, this approach implies a cause-effect relationship between gene expression and a specific level of drought stress. Thus, it is very important that confounding factors are excluded from the experiment. Chlorophyll content in the needles, for example, might change over the course of the monitoring period due to the drought treatment. To test if the chlorophyll content could potentially influence the terahertz signal, chlorophyll was extracted from silver fir needles, in the course of this thesis, and different concentrations were measured using terahertz spectroscopy, showing that chlorophyll content does not influence terahertz monitoring. Another aspect of the candidate gene approach involves the variation within a polymorphic gene and its potential association with the variation in a phenotypic trait. Since the growth depression period of silver fir in the 1970s and 1980s was mostly influenced by the combination of air pollution and drought, in the context of this thesis, genetic variation, in the form of single nucleotide polymorphims (SNPs) in pre-selected genes, was associated with tree-ring derived phenotypes for individual trees in the Bavarian Forest National Park. These so called ’dendrophenotypes’ were measures for resistance, resilience and recovery during the depression period, as well as the drought year 1976. Using general linear models and feature selection techniques based on the machine learning algorithm random forest, 15 out of 103 polymorphic candidate genes for trait variation could be identified. Since the associated dendrophenotyes are potentially adaptively relevant, the variation in this candidate genes could influence the stress coping capability of individual trees. However, this approach is of an observational nature and thus, cause-effect relationships cannot be derived from this type of experiment. The identified SNPs might be the causal variant or physically close to the true causal variant or it might just be a spurious correlation. Further, reliance on advanced statistical techniques can be troublesome, as could be demonstrated in the course of this thesis for a random forest based feature selection technique, developed for genetic association studies in conifers. Replicating this study and evaluating the algorithm, non-uniqueness of the results could be demonstrated, which not only hinders biological interpretation but can severely negatively influence downstream analyses, such as tests for interaction between SNPs. In conclusion, this thesis presents new techniques to add to the current methodology for candidate gene selection and analysis in the stress response of the non-model organism silver fir and other conifer species. Both approaches should be combined, for example by drawing polymorphic candidate genes for trait variation from the pool of functional candidate genes to ensure the involvement of the studied genes in the variation of the trait of interest. Further, the results of this thesis add to the growing molecular resources in silver fir and thereby, hopefully, contribute to the successful prediction and management of this important forest tree species in the face of rapidly changing environmental conditions., Das Ziel dieser Dissertation war die Identifizierung und Analyse von Kandidatengenen f¨ur Stressreaktion in der Weißtanne (Abies alba Mill.). Diese ¨okologisch und ¨okonomisch wichtige Waldbaumart ist nat¨urlich beheimatet in bergigen Regionen Europas aber es ist wenig bekannt ¨uber ihre ¨okologische Charakterisierung. Weißtannenpopulationen wurden durch menschlichen Einfluss deutlich transformiert, was zu einer Diskrepanz zwischen dem fr¨uheren und dem heutigen Verbreitungsgebiet gef¨uhrt hat. Aktuelle Studien legen nahe, dass die Weißtanne in w¨armeren und trockeneren Klimaten ans¨assig sein kann, als sie es derzeit ist. Die Art hat jedoch in den 1970ern und 1980ern deutlich gelitten und zeigte in Deutschland Blattsch¨aden, eine Abnahme im Dickenwachstum und ¨ortliches Waldsterben. Als Grund hierf¨ur wird meist der H¨ochststand der Luftverschmutzung w¨ahrend dieses Zeitraums genannt, insbesondere von Schwefeldioxid (SO2), das offenbar die Empfindlichkeit der Weißtanne gegen¨uber Trockenstress deutlich erh¨oht. Die Kombination dieser beiden Stressoren, SO2 und Trockenperioden, hat die Weißtanne sogar in Regionen in denen Trockenstress ¨ublicherweise kein Problem ist negativ beeinflusst. Im Kontext des anthropogenen globalen Klimawandels, der sehr wahrscheinlich zu einem Ansteigen der Temperatur in Europa und zu mehr Extremereignissen, wie heftigen Trockenperioden, f¨uhren wird, stellt sich die Frage, wie die Weißtanne mit diesen Umweltver¨anderungen umgehen wird. Bedenkt man die Geschwindigkeit der vorhergesagten Ver¨anderungen und die zunehmende Fragmentierung der Landschaft, besteht die M¨oglichkeit, dass die Weißtanne diesem nicht durch Samenausbreitung entgehen kann. Als sessiler Organismus bleibt als die einzig andere Option nur Adaptation, die sich wahrscheinlich aus der stehenden genetischen Variation speisen wird. Um das Schicksal der Weißtanne, insbesondere im Kontext des globalen Klimawandels, erfolgreich abzusch¨atzen und Populationen m¨oglicherweise aufgrund solcher Vorhersagen zu managen, bedarf es der Kenntnis der genetischen Architektur im Kontext solcher bedeutender Stressoren wie Trockenperioden und Luftverschmutzung. Es gibt jedoch sehr wenige genomische Ressourcen f¨ur die Weißtanne und Koniferen im Allgemeinen. Das liegt maßgeblich an der Gr¨oße und Komplexit¨at der Genome und am langen Generationszyklus, was Koniferen zu typischen nicht-Modell Organismen macht. Aus diesem Grund sind Methoden zur Identifizierung der genetischen Basis von Stressantwort effektiv auf einen Kandidatengenansatz beschr¨ankt. Der Kandidatengenansatz beinhaltet die Identifikation von funktionellen Kandidatengenen, indem die differentielle Genexpression zwischen einer gestressten und einer Kontrollgruppe gemessen wird. Im Kontext dieser Dissertation wurde der Wassergehalt von Weißtannens¨amlingen mit einem neuartigen Terahertz-Spektroskopie-Aufbau in einem Labor ¨uberwacht. Eine Gruppe von S¨amlingen wurde regelm¨aßig gegossen, w¨ahrend eine andere Gruppe Trockenstress ausgesetzt war. Durch die kontinuierliche Messung des Wassergehalts konnten Nadeln der S¨amlingen aus beiden Gruppen zu einem Zeitpunkt geerntet werden, zu dem der Wassergehalt zwischen den Individuen einer Gruppe jeweils vergleichbar war. Eine differentielle Expressionsanalyse zwischen den Nadeln der beiden Gruppen resultierte dann in 296 Genen die als Reaktion auf Trockenstress signifikant hochoder herunter-reguliert waren. Ungef¨ahr 45% dieser Gene sind zuvor noch nicht in anderen Organismen beschrieben worden und sind potentiell spezifisch f¨ur die Weißtanne oder Koniferen im Allgemeinen. Da jedoch nur Nadeln von S¨amlingen bei einem bestimmten Trockenstressniveau analysiert wurden, sind die Ergebnisse in ihrem Umfang auf das Ausgangsmaterial und das spez-ifische Stresslevel reduziert und k¨onnen nicht direkt auf die Weißtanne oder Trockenstress allgemein ¨ubertragen werden. Weiterhin impliziert dieser Ansatz einen Kausalzusammenhang zwischen Genexpression und einem spezifischen Maß an Trockenstress. Daher ist es sehr wichtig st¨orende Faktoren vom Experiment auszuschließen. So kann der Chlorophyllgehalt der Nadeln sich beispielsweise w¨ahrend des Messzeitraumes in Folge der Trockenstressbehandlung ver¨andern. Um zu testen ob der Chlorophyllgehalt m¨oglicherweise einen Einfluss auf das Terahertz-Signal hat, wurde im Rahmen dieser Dissertation Chlorophyll aus Weißtannennadeln extrahiert und unterschiedliche Konzentrationen mittels Terahertz-Spektroskopie gemessen. Dabei konnte gezeigt werden, dass der Chlorophyllgehalt keinen Einfluss auf das Terahertz-Monitoring hat. Ein anderer Aspekt des Kandidatengenansatzes beinhaltet die Variation innerhalb eines polymorphen Gens und die m¨ogliche Assoziation mit der Variation in einem ph¨anotypischen Merkmal. Da die Periode der Wachstumsdepression von Weißtannen in den 1970ern und 1980ern maßgeblich durch eine Kombination von Luftverschmutzung und Trockenperioden verursacht war, wurde im Rahmen dieser Dissertation genetische Variation, in Form von Einzelnukleotid-Polymorphismen (engl. single nucleotide polymorphisms (SNPs)) in vorausgew¨ahlten Genen, mit Ph¨anotypen assoziiert, die aus Jahresringen f¨ur individuelle B¨aume im Nationalpark Bayerischer Wald abgeleitet wurden. Diese so genannten ’Dendroph¨anotypen’ waren Maße f¨ur die Resistenz, Resilienz und Erholung in der Depressionsperiode und dem Trockenjahr 1976. Basierend auf allgemeinen linearen Modellen und Feature Selection Techniken, die auf dem maschinellen Lernalgorithmus Random Forest beruhen, konnten 15 aus insgesamt 103 polymorphen Kandidatengenen f¨ur Merkmalsvariationen identifiziert werden. Da die assoziierten Dendroph¨anotypen potentiell adaptiv relevant sind, k¨onnte die Variation in diesen Kandidatengenen die F¨ahigkeit der Stressbew¨altigung individueller B¨aume beeinflussen. Dieser Ansatz ist jedoch grunds¨atzlich beobachtender Natur und diese Art von Experiment erlaubt daher keine Ableitung von Kausalzusammenh¨angen. Die identifizierten SNPs k¨onnen die urs¨achliche Variation sein, sie k¨onnen aber auch physikalisch nah an der tats¨achlich urs¨achlichen Variation sein oder es kann sich lediglich um einen Scheinzusammenhang handeln. Weiterhin kann das Vertrauen in fortgeschrittene statistische Verfahren problematisch sein, was im Rahmen dieser Dissertation f¨ur eine auf Random Forest basierende Feature Selection Methode gezeigt werden konnte, die f¨ur genetische Assoziationsanalysen in Koniferen entwickelt wurde. Durch die Replikation dieser Studie und die Evaluierung des Algorithmus konnte die Multiplizit¨at der Ergebnisse demonstriert werden, die nicht nur die biologische Interpretation behindert, sondern auch nachgelagerte Analysen, wie Tests auf Interaktion zwischen SNPs, negativ beeinflusst. Schlussfolgernd beschreibt diese Dissertation neue Techniken der Auswahl und Analyse von Kandidatengenen f¨ur die Stressreaktion im nicht-Modell-Organismus Weißtanne und anderen Koniferenarten die der g¨angigen Methodik hinzuzuf¨ugen sind. Beide Ans¨atze sollten kombiniert werden, beispielsweise indem polymorphe Kandidatengene f¨ur Merkmalsvariation aus dem Pool von funktionellen Kandidatengenen gezogen werden um die Beteiligung der untersuchten Gene an der Variation des zu untersuchenden Merkmals sicher zu stellen. Weiterhin tragen die Ergebnisse dieser Dissertation zu den wachsenden molekularen Ressourcen f¨ur die Weißtanne bei und haben dadurch, hoffentlich, einen Anteil an der erfolgreichen Vorhersage und am Management dieser wichtigen Baumart im Kontext rasanter Umweltver¨anderungen.

Coniferous trees are of major economic and ecologic importance. Yet, they are genomically poorly studied, because their long generation times, their high levels of heterozygosity and their huge genomes impede the access to conifer genomic resources. The European silver fir (Abies alba MILL) is widely distributed across diverse habitats in mountainous areas of temperate Europe. The species is of high value for forestry due to its high quality timber and its ecosystem functions such as erosion control and landslide prevention. Using next-generation sequencing the first silver fir transcriptome was sequenced and annotated. The annotation protocol lead to the identification of 460 candidate genes that are putatively involved in environmental stress resistance. Additionally, the transcriptomes of twelve more individuals were sequenced. Subsequent SNP detection produced on average 24580 SNPs per individual. For the SNPs that lie on the annotated candidate genes, a KASP genotyping assay was designed that allows for the fast and cost efficient genotyping of many A. alba trees. In the Mediterranean Basin, A. alba occurs in isolated and comparatively small populations. Recently, strong die-off events were observed in these populations, likely as a result of severe droughts. In the course of climate change, droughts are predicted to increase further in frequency and severity endangering the persistence of Mediterranean A. alba populations. Particularly these populations may, however, be of special value for the adaptive potential of the whole species since they may harbor genetic resources preadapted to relatively xeric environments. There is an ongoing debate about assisted migration, i.e. the transplantation of Mediterranean A. alba populations to more northern population in order to use them as donors of drought adapted alleles. To elucidate the question whether the idea of assisted migration is practical, knowledge about the state of local adaptation, its molecular basis and its environmental drivers is needed. Here, 376 individuals from high and low elevational populations which are located along a 170 km eastwest transect across four mountains in the south-western Mediterranean pre-Alps of France were investigated at 267 SNP loci distributed across 175 candidate genes. The methodological framework of landscape genetics was applied. Results of Bayesian cluster analysis showed that the dominant genetic pattern coincides with an east-west isolation-by-distance genetic structure, likely a result of post-glacial recolonization movement. Fst-outlier tests revealed 16 SNPs under divergent selection. Significant associations of allele frequencies of eight SNPs and winter drought were detected suggesting that this variable is a strong driver of local adaptation. Correlations tests of genetic distance of a subset of outlier loci and environmental distance demonstrated that isolation-by-environment occurs along altitude. Reanalysis of published common garden phenotypic data using Qst-Fst comparisons revealed adaptive divergence of bud flush and growth rate at the coldest and most humid site. The thesis illustrates a complex pattern of genetic structure in ecologically marginal populations that is shaped by demography and natural selection. It highlights that local adaptation is site specific in terms of both the genes under selection and the environmental drivers. Hence, whether a population is suitable to be the subject of assisted migration would have to be answered individually. Since this is highly impractical, the thesis states that in-situ conservation of marginal populations is the best option to ensure the capacity of evolutionary change in A. alba and to support the species’ ability to cope with the challenges of a rapidly changing climate.

• Premise of the study: We present a protocol for the annotation of transcriptome sequence data and the identification of candidate genes therein using the example of the nonmodel conifer Abies alba. • Methods and Results: A normalized cDNA library was built from an A. alba seedling. The sequencing on a 454 platform yielded more than 1.5 million reads that were de novo assembled into 25149 contigs. Two complementary approaches were applied to annotate gene fragments that code for (1) well-known proteins and (2) proteins that are potentially adaptively relevant. Primer development and testing yielded 88 amplicons that could successfully be resequenced from genomic DNA. • Conclusions: The annotation workflow offers an efficient way to identify potential adaptively relevant genes from the large quantity of transcriptome sequence data. The primer set presented should be prioritized for single-nucleotide polymorphism detection in adaptively relevant genes in A. alba.

The EC-supported Network of Excellence Evoltree (http://www.evoltree.eu) formed a group of scientists involved in and actively contributing to Jointly Executed Research Activities on community genetics in forest ecosystems. In addition to the main authors, the group includes S. Augu-stin, M. Brandle, C. Burban, J. Burczyk, S. Cavers, I. Chybicki, C. Conord, E. Cremer, J. DeWoody, K. Donges, B. Fady,L. Karlinski, C. Kerdelhue, B. Kieliszewska-Rokicka, G. Kost, M. Kulczyk-Skrzeszewska, F. Lakatos, F. Lefevre, S.Liepelt, S. Oddou-Muratorio, K.-H. Rexer, M. Rudawska, M.Schadler, G. Taylor, K. Tuba, M. Viger, F. Villani, M. Villar.; Understanding the interactions of co-occurring species within and across trophic levels provides key information needed for understanding the ecological and evolutionary processes that underlie biological diversity. As genetics has only recently been integrated into the study of community-level interactions, the time is right for a critical evaluation of potential new, gene-based approaches to studying communities. Next-generation molecular techniques, used in parallel with field-based observations and manipulative experiments across spatio-temporal gradients, are key to expanding our understanding of community-level processes. Here, we introduce a variety of ‘-omics’ tools, with recent studies of plant–insect herbivores and of ectomycorrhizal systems providing detailed examples of how next-generation approaches can revolutionize our understanding of interspecific interactions. We suggest ways that novel technologies may convert community genetics from a field that relies on correlative inference to one that reveals causal mechanisms of genetic co-variation and adaptations within communities.

Today human impact is the main driver of global environmental change. One of the most severe anthropogenic changes is land transformation, which has altered 40- 50% of Earth’s surface into urban and agricultural systems. Orchids are particularly vulnerable to environmental changes, because with germination and pollination two important life cycle stages directly depend on symbiotic interactions. Therefore a thorough understanding of orchid biology and the factors that determine habitat suitability and orchid distribution is essential for an effective conservation of orchid diversity. Using Gymnadenia conopsea s.l. as study system, a common orchid that occurs over a wide ecological amplitude, this thesis addresses aspects of orchid biology that might be critical for orchid recruitment and the long-term persistence of populations in the wild, namely the specificity of the mycorrhizal symbiosis, population genetic diversity and genetic differentiation. The availability of suitable fungal partners is a precondition for orchid germination in the wild as they provide carbohydrates for initial growth. In one study the fungal communities of six G. conopsea s.str. populations in two study regions in East and North Germany were analysed. The 28 identified taxa revealed a high diversity of fungi associated with G. conopsea. This high diversity of associated fungi indicates that this orchid shows only little specificity to certain fungal clades, which is likely to contribute to its ability to grow in very different habitat types with the respective fungal communities. However, the taxon composition showed a clear spatial structure and only little overlap between regions, suggesting that factors at the local scale may strongly affect local species composition and hence diversity at the regional level. In all populations taxa of the known OM genera Tulasnella, Ceratobasidium, Thanatephorus and Sebacina were detected, suggesting that G. conopsea utilizes typical OM fungi as mycorrhizas. However, also ascomycetous taxa from the Pezizales and Helotiales were identified. Their continuous detection indicates that this orchid is also able to utilize ascomycetous ectomycorrhizal taxa, which could be another aspect contributing to its ability to grow in a variety of different habitats. Many orchids are well known as primary colonisers and are often found in anthropogenic habitats. In order to assess whether founder effects during colonization threaten the long-term survival of recently established populations, another study compared the genetic diversity and fitness of populations from anthropogenically disturbed and surrounding natural habitats in two German regions (East Germany: lignite post-mining area; North Germany: quarries). The results revealed a reduced genetic diversity and lowered fruit set for the populations in the lignite post-mining area, whereas no such effects were found for the quarry populations. However, the general plant performance was similar for all investigated populations. This indicates that during the colonization of post-mining areas that are often distant from potential source populations, founder effects may occur. In contrast, this problem seems negligible for G. conopsea populations in the quarries, which were spatially intermixed with source populations. Gymnadenia conopsea (L.) R.BR. s.l. is a controversial taxon. The two most commonly distinguished taxa are G. conopsea (L.) R.BR. ssp. conopsea and G. conopsea ssp. densiflora (WAHLENB.) K. RICHT., for which also a species status has been suggested. Due to a high morphological overlap between the taxa, identification in the field is difficult. Hence, for taxon assignment all investigated populations were genetically and morphologically analyzed. The analysis of the ITS region revealed a 2% nucleotide divergence, similar to the divergence between other Gymnadenia species. This, together with largely non-overlapping sets of microsatellite alleles supports the view that Gymnadenia conopsea (L.) R.BR. s.str. and Gymnadenia densiflora (WAHLENB.) DIETRICH deserve species status. G. conopsea s.str. and G. densiflora are not even sister species as the sequences of G. densiflora form a well supported monophyletic group, sharing a most recent common ancestor with G. nigra and G. austriaca. G. conopsea s.str. was either diploid or tetraploid, while G. densiflora was diploid throughout. As the microsatellite patterns of the two ploidy levels of G. conopsea s.str. were hardly differentiated and the most frequent ITS haplotypes occurred in both of them, an autopolyploid origin of tetraploid from diploid G. conopsea s.str. is likely. However, morphological differentiation was less clear. Although some traits (e.g. flower number and density) allow a fairly good distinction, due to a considerable variation an unequivocal identification will remain difficult.

Ausbreitung ist ein entscheidender Prozess beim Überleben von Pflanzen, denn er ermöglicht Pflanzen als sessilen Organismen sich fortzubewegen. Ausbreitung ist sowohl zur Vermeidung von schlechten Bedingungen als auch zur Neubesiedlung von Flächen notwendig. Außerdem reduziert ein genetischer Austausch zwischen Populationen mögliche genetische Bedrohungen wie z.B. Inzuchtdepression. Die häufigsten Ausbreitungsvektoren bei Pflanzen sind Tiere und Wind. In vom Menschen dominierten Landschaften jedoch können Menschen die tierische Verbreitung ersetzen. Um die Bedeutung des Menschen auf die Pflanzenverbreitung zu untersuchen wurde ein Simulationsmodell erstellt. Der Effekt der menschlichen Bewegung und Gartenstile wurde für verschiedene ruderale Pflanzen getestet. Ich fand einen starken Einfluss des menschlichen Bewegungsverhaltens besonders bei nicht-einheimischen Arten heraus. Innerhalb von 20 Jahren vergrößerten diese Arten ihre Verbreitung, wobei sie von der ansteigenden Mobilität der Menschen profitierten, die in soziologischen Untersuchungen nachgewiesen worden war. Im Gegensatz dazu schienen einheimische Arten stärker auf den Wechsel der bäuerlichen Landschaft und von einem ländlichem zu einem städtischen Lebensstil zu reagieren. Allerdings konnte die Veränderung in der Artverbreitung in allen Fällen besser mittels einer menschlichen Ausbreitung basierend auf menschlichen Bewegungsverhalten simuliert werden als durch ein distanzabhängiges Diffusionsmodell. Eine weitere Herangehensweise zur Nutzung von Simulationsmodellen bei Ausbreitungsprozessen wurde anhand der windbestäubten, in der Weichholzaue von Flüssen vorkommenden Art Populus nigra in Mitteldeutschland durchgeführt. Das Ziel der Untersuchung war es, das Ausmaß von innerartlichem Pollenvermittelten Genfluß durch den Schwarzpappelhybriden P. x canadensis abzuschätzen. Daher war die Abschätzung eines passenden Ausbreitungskernel entscheidend für die Modellierung der Ausbreitung. Genetische Vaterschaftsanalysen ermöglichen die Berechnung von Ausbreitungsdistanzen des Pollens zwischen Vater- und Mutterbäumen. Ich habe Punktmusteranalysen benutzt um Ausbreitungskernel anzupassen und deren statistische Unsicherheit zu testen. Es gab nur bis zu einer Distanz von 300m eine signifikante Abweichung vom Paarungs-Nullmodell, obwohl es sich um eine windbestäubte Art handelt und die Analysen auf einem sehr großen Datensatz von mehr als 1.500 Samen beruhten. Ein Zwei-Komponenten-Ausbreitungskernel aus einer exponential-power Funktion und einer eingeschränkten Gleichverteilungsfunktion bildete das am besten angepasste Model zu den Daten. Der angepasste Kernel konnte die Anzahl von Samen, die von einem männlichen Baum bestäubt wurden, abschätzen und lieferte vergleichbare Ergebnisse wie veröffentlichte räumlich statistische Modelle. Unsere Ergebnisse zeigen deutlich, dass Ausbreitungskernel, die auf einer direkten Anpassung an die beobachteten Paarungsdistanzen beruhen, stark von Ergebnissen abweichen, die durch Methoden erzielt wurden, die die räumliche Struktur von Altbäumen berücksichtigen. Die Unsicherheit in der Abschätzung solcher direkt angepasster Ausbreitungskernel scheint unterschätzt zu werden. Mit dem angepassten Ausbreitungskernel wurden Simulationen durchgeführt, um die Diskrepanz zwischen geringer Einkreuzung durch den Hybriden in der natürlichen Schwarzpappelpopulation und der überwältigenden Anzahl dieser Hybride in der direkten Nachbarschaft zu untersuchten. Durch die Verwendung von verschiedenen standardmäßigen Ausbreitungskernel und des angepassten Zwei-Komponenten-Ausbreitungskernel fanden wir heraus, dass die Wahrscheinlichkeit einer Befruchtung durch Hybridpollen 10^-1 bis 10^-2 mal geringer als bei der Schwarzpappel war. Dieser Werteraum konnte über Ergebnisse bestätigt werden, die in Handbestäubungsexperimenten entstanden. Dies ist das erste Mal, dass die reproduktive Barriere zwischen P. nigra und P. x canadensis in frei bestäubten Bäumen in einem natürlichen Bestand gezeigt werden konnte. Die Verwendung von Simulationsmodellen zur Untersuchung von unterschiedlichen Ausbreitungsmechanismen stellte sich als viel versprechend heraus, nicht nur zur Beschreibung des Ausbreitungsprozesses selbst, sondern auch hinsichtlich der Kombination von Expertenwissen aus den verschiedensten Fachrichtungen in einem ökologischen Modell., Dispersal is an essential process in plant survival because it allows plants as static organism to move. Plant dispersal is necessary to avoid unsuitable conditions and to colonise new sites. Additionally gene flow between populations reduces possible genetic threads as inbreeding depression. Genetic exchange is not limited to the dispersal of seeds, but is also possible by pollen dispersal. The most common dispersal vectors for plants are animals and wind. However, in human dominated landscapes animals could be replaced by humans. To assess the importance of humans on plant distribution a simulation model was created. The effect of human movement and gardening styles were tested for different ruderal plant species. I found strong influence of human movement behaviour especially on alien species. During 20 years these species extended their distribution range taking benefit from increased human mobility as revealed by sociological studies. In contrast native species tended to be more affected by the change of the agricultural landscape and of rural to urban life styles. Nevertheless, the change in species distribution could in all cases be better explained by simulating human transport based on human movement behaviour than by a distance dependent diffusion model. Another approach using simulation models for dispersal has been done in the wind pollinated floodplain species Populus nigra in Central Germany. The aim of the study was to assess the extent of intraspecies respectively introgressive pollen mediated gene flow by the hybrid P. x canadensis. Therefore the estimation of a pollen dispersal kernel was essential to model the dispersal processes. Genetic paternity analyses allow the calculation of pollen dispersal distances between father and mother trees. I used marked point pattern analyses to estimate and assess the uncertainty in fitted pollen dispersal kernels. There was only a significant departure from the null random mating model up to a distance of approximately 300m, although we were dealing with a wind pollinated species and the analyses were based on a huge dataset of more than 1,500 seeds. A two-component pollen dispersal kernel comprising an exponential power function and a truncated uniform function was the most parsimonious model to fit the data. The fitted kernel was able to predict the number of seeds fathered by a male tree and provided comparable results with published spatial statistic models. Our results strongly suggest that kernel estimates based on the direct fit of the observed mating distances differed considerably from fitted kernels that account for the spatial structure of adult trees. The uncertainty in the estimation of those directly fitted kernel may be underestimated. With a suitable pollen dispersal kernel at hand, simulation studies were conducted to understand the discrepancy between low introgression rates in the natural population of P. nigra and the overwhelming number of hybrids in the close vicinity. Using different standard dispersal kernels and our fitted two-component exponential power function, we found that the probability of fertilisation was 10-1 to 10-2 times lower in hybrid than in P. nigra pollen. This range could be confirmed with published results from hand pollination experiments and is the first time that the reproductive barrier between P. nigra and P. x canadensis has been revealed in open pollinated trees in a natural population. Using simulation models to understand different kinds of dispersal mechanisms turned out to be a promising way not only to describe the process of dispersal itself but also to combine expert knowledge of various disciplines in an ecological simulation model.

Because of the eco-systematic significance of forests and their close link to the climatic history a large part of paleo-ecological research has dealt with the history of forests during the Pleistocene climatic oscillations. On the basis of these efforts the global patterns of vegetation and forest histories could largely be reconstructed especially by means of pollen analysis, macrofossils and finally phylogeography. However, in recent years an increasing number of studies have shown that microrefugia in temperate and boreal areas, often called cryptic refugia, had been overlooked by traditional methods. As a consequence the importance of these microrefugia for the conservation of genetic diversity, as well as their role in postglacial re-expansions has not been adequately acclaimed. Furthermore it became obvious in recent years that the early anthropogenic influence on the forest histories was not sufficiently reflected in the paleo-ecological sciences. This potentially led and still leads to a considerable bias in climatic parameters derived from paleo-ecological reconstructions. In this context the cumulative dissertation here presented deals with the forest history of the Tibetan Plateau. It postulates a special significance of high-mountain areas for the preservation of microrefugia. In the included four articles both classical as well as molecular genetic methods were used to reconstruct the forest history of the Southern Tibetan Plateau as exemplified by the Tibetan juniper forests. In paper I current forest remains were mapped and characterized. According to these findings forest and woodland relics were found in large parts of the southern Tibetan Plateau. Natural reproduction on many of these sites, as well as reforestation trials showed that the area is potentially forested under current climatic conditions. In paper II charcoal analytical investigations showed that up to the late Holocene the Tibetan juniper forests had a broader distribution range. The reduction to the current extent was interpreted as a consequence of anthropo-zoogenic pressure in combination with dessiccation. However, to support these findings high resolution charcoal data bases will be needed in the future. The necessary anthracological potential in the geo-archives of the southern Tibetan Plateau could be demonstrated through this work. By means of molecular genetic analysis paper III could show that unlike former assumption the juniper forests of the southern Tibetan Plateau survived in microrefugia on the plateau platform itself throughout today’s range. Furthermore it could be shown that the Himalayan juniper populations did not contribute significantly to the postglacial expansion on the plateau. The observed patterns along with the known mutation rates in the used chloroplast markers indicate a late tertiary origin of the spatial genetic patterns. In addition, the numerous decentralized microrefugia as well as the high number of private haplotypes therein demonstrate the potential importance of high mountain areas with their heterogenic topographies and microclimates for the conservation of genetic resources. Finally paper IV provided technical resources for subsequent analysis of genetic resources within the Tibetan juniper forests. By means of next generation sequencing a set of microsatellite marker were developed that will enable us to analyze gene flow, as well as nuclear genetic diversity. Furthermore these markers are a foundation to realize the research agenda proposed in the outlook of the dissertation., Aufgrund der ökosystemaren Bedeutung von Wäldern und ihrer engen Kopplung an das Klima setzen sich weite Teile der Paläoökologie mit der Geschichte der Wälder im Zuge pleistozäner Klimaoszillationen auseinander. Weltweit konnten so die Grundzüge der Ve-getations- und Waldgeschichte mit Hilfe der Pollenanalyse, Analyse von Makrofossilien und zuletzt der Phylogeographie erkundet werden. Allerdings kamen in den letzten Jahren vermehrt Hinweise auf, die zeigen, dass Mikrorefugien und ihre Bedeutung für die Konservierung von genetischem Material, sowie ihrer Bedeutung bei der postglazialen Wiederbesiedlung lange Zeit übersehen wurden. Diese übersehenen Refugien werden Kryptische Refugien genannt. Ausserdem wurde in den letzten Jahren deutlich, dass der z.T. sehr gravierende und vor allen Dingen frühe menschliche Einfluss auf Vegetations- und insbesondere Waldentwicklung in der Paläoökologie keine angemessene Berücksichtigung fand. Daher kam und kommt es zu Mißinterpretationen bei der Ableitung von Klimaparametern und der Prognose zukünftiger Vegetationsentwicklung. Die hier vorliegende kumulative Dissertation bearbeitet die Waldgeschichte des Tibetischen Plateaus vor diesem Hintergrund und postuliert eine besondere Bedeutung von Hochgebirgslandschaften für den Erhalt von Mikrorefugien. In insgesamt 4 Artikeln werden sowohl klassiche paläoökologische Methoden sowie molekulargenetische Methoden verwendet, um die Waldgeschichte Südtibets am Beispiel der Wacholderwälder zu rekonstruieren. Da das Tibetische Plateau heute in weiten Teilen durch alpine Steppen, Hochgebirgshalbwüsten und Weideländern geprägt ist und neben Auwäldern nur vereinzelt Waldinseln und Einzelbäume auf Normalstandorten auftreten, wurde die Waldgeschichte des Plateaus lange Zeit übersehen. Im ersten Teil dieser Arbeit (Paper I) wurden die aktuell bzw. historisch bis in die 1960er Jahre hinein existierenden Wacholderwälder kartiert und charakterisiert. Demnach sind in weiten Teilen des südlichen Plateaus Waldrelikte vorhanden. Die natürliche Verjüngung an vielen dieser Standorte, sowie Versuche in Baumschulen zeigen, dass das Waldpotential auch unter derzeitigen Klimabedingungen gegeben ist. Ungeklärt bleibt hingegen die Ursache für die extreme Diskrepanz zwischen der hohen Anzahl an Beerenzapfen und dem geringen Anteil an vitalen Embryonen. Die Tatsache, dass dieses Phänomen in unterschiedlichen Wacholderarten unter unterschiedlichen klimatischen Bedingungen weltweit beobachtet wurde macht einen Zusammenhang mit klimatischen Stress allerdings unwahrscheinlich. In Paper II wird mit Hilfe von Holzkohleanalysen gezeigt, dass bis ins späte Holozän die Wacholderwälder Südtibets weiter verbreitet waren. Ihre Reduzierung auf das heutige Maß wird mit wachsendem menschlichen Einfluss einschliesslich der Yakviehhaltung im Zusammenspiel mit klimatischer Austrocknung gedeutet. Um diese Befunde zu bekräftigen ist allerdings in Zukunft die Erarbeitung hochauflösender Holzkohledatenbanken notwendig. Das dazu nötige anthrakologische Potential in den Geoarchiven Südtibets konnte mit dieser Arbeit aufgezeigt werden. Im Dritten Artikel (Paper III) wird mit Hilfe molekulargenetischer Methoden gezeigt, dass entgegen bisheriger Annahmen die Wacholderwälder Südtibets im Letzten Glazialen Maximum auf dem Tibetischen Plateau in Mikrorefugien überdauerten. Auch konnte gezeigt werden, dass die himalayischen Wacholderbestände nicht wesentlich zu einer postglazialen Wiederbesiedlung des Plateaus beigetragen haben. Die hohe Anzahl privater Haplotypen auf dem Plateau sowie die gewöhnlich geringen Mutationsraten im Chloroplastengenom machen sogar eine Entstehung der beobachteten genetischen Muster im späten Tertiär plausibel. Darüber hinaus demonstrieren diese zahlreichen dezentralen Mikrorefugien, sowie die hohe Anzahl endemischer Haplotypen das Potential von Hochgebirgslandschaften mit ihren heterogenen Topographien für den Erhalt genetischen Ressourcen. Das rein methodische Paper IV liefert Grundlagen für die weitere Erforschung dieser genetischen Ressourcen und ihrer Verteilung in der Landschaft. Mit Hilfe von Hoch-Durchsatz-Sequenziermethoden wurden Mikrosatelliten entwickelt, die bei der Analyse von aktuellen Genflüssen, sowie nukleärer Diversität zum Einsatz kommen sollen. Darüber hinaus sind sie die Grundlage um die in Paper V und im Kapitel Outlook skizzierten Forschungsvorhaben zu realisieren.

Im Zuge des Klimawandels ist von einer zunehmenden Häufigkeit extremer Wetterlagen auszugehen, die die Waldökosysteme beeinflussen. In den letzten Jahrzehnten haben zahlreiche Sturmereignisse mitteleuropäische Waldbestände großflächig geworfen. Die entstandenen Windwurfflächen müssen nun wiederverjüngt werden. Lebensgemeinschaften von Bäumen mit Ektomykorrhizapilzen (ECM) spielen hierbei eine entscheidende Rolle für die Stabilität und Vitalität von Waldökosystemen. Da die Weißtanne (Abies alba Mill.) eine starke Reduktion in ihrem natürlichen Verbreitungsgebiet in Deutschland erfahren hat, ist eine Erhöhung ihres Anteils aufgrund der stabilisierenden Eigenschaften waldbaulich vorgesehen. Angesichts der Zunahme von Sturmereignissen wurden drei Tannenbestände im Nordschwarzwald genetisch und mykologisch untersucht, um das Wiederbesiedlungspotential der Tanne abzuschätzen. Zu diesem Zweck wurden in einem ersten Schritt Kern-Mikrosatellitenmarker (nSSR) für die Tanne entwickelt. Anschließend wurden Tannenkollektive verschiedener Altersklassen (Altbäume, Jungbäume, Sämlinge) an sechs nSSR-Orten genotypisiert sowie hinsichtlich Vorkommen und Diversität der mit ihr vergesellschafteten ECM untersucht. Die Ergebnisse zeigen, dass die Weißtannen über ein ausreichendes genetisches Potential mit einer hohen Diversität innerhalb und einer geringen Differenzierung zwischen den Beständen verfügen. Die Naturverjüngung der Tanne auf den angrenzenden Freiflächen wies keine reduzierte genetische Diversität auf im Vergleich zu den ungleichaltrigen Beständen. Die Untersuchungen zum Genfluss bei der Tanne lassen zudem auf eine ausreichende Samen- und Pollenausbreitung schließen, zumindest im Abstand von wenigen hundert Metern vom Mutterbaum. Eine Vielzahl von Mutterbäumen trug zur Samenverbreitung bei und führte zu einem vielfältigen Sameneintrag sowohl innerhalb der Bestände als auch in die Windwurfflächen hinein. Die Analyse der ECM zeigte im Altbestand und auf der Freifläche ein vergleichbares Spektrum hinsichtlich Artenzahl und Häufigkeit. Es gab keine Anhaltspunkte dafür, dass das Alter der Bäume in einer Population die Artzusammensetzung der ECM beeinflusst. Auf der Ebene des Einzelbaumes zeigten jedoch ältere Tannen eine höhere Anzahl verschiedener ECM als jüngere. Die auf den Windwurfflächen bereits vorhandene Verjüngung zeichnet sich durch ein ausgewogenes ECM-Profil aus. Sie besitzt damit eine ‚Reservoir-Funktion’ für die später ankommende Tannen-Verjüngung. Darüber hinaus wurde geprüft, ob das Tannen-Genom, d.h. die nSSR-Genotypen, einen Einfluss auf die Besiedlung mit ECM hat. Es zeigte sich, dass bestimmte nSSR-Genotypen mit der ECM-Artzusammensetzung signifikant korreliert sind. Daher kann die ECM-Gesellschaft als so genannter ‚erweiterter Phänotyp’ (extended phenotype) der Tannen betrachtet werden. Der Schutz und die Förderung der Weißtanne als Maßnahme im Rahmen der forstlichen Generhaltung bezieht daher nicht nur die Baumart selbst ein, sondern ebenfalls die mit ihr assoziierten ECM-Gesellschaften als Teil des Ökosystems. Aus genetischer, verbreitungsbiologischer und mykologischer Sicht bieten Weißtannenpopulationen im Nordschwarzwald geeignete Voraussetzungen für eine natürliche Verjüngung sowohl im Bestand als auch zur Wiederbesiedlung von Freiflächen., Facing climate change, we expect an increasing frequency of extreme weather events such as storms that affect forest ecosystems. In the last decades, several storm events in Central Europe have damaged huge areas of forest stands that have to be recolonized. Symbiotic systems between trees and ectomycorrhizal (ECM) fungi play a decisive role for the stability and the vitality of trees. In the context of promoting the rare, but ‘stabilizing’ tree species silver fir in mountain forests and facing the recolonization of windthrow areas, three fir populations were genetically investigated in the Northern Black Forest, Germany. For this purpose, in a first step nuclear microsatellite (nSSR) markers were developed for silver fir. Fir trees of different ontogenetic stages (adults, saplings, seedlings) were genotyped at six nSSR loci and analysed in terms of diversity and abundance of the associated ECM fungi. The results demonstrate that silver fir populations in the Black Forest maintain a suitable genetic potential with high diversity within and less differentiation among populations. The remaining natural fir regeneration on the windthrow area did not show a reduced genetic diversity in comparison to the adjacent forest stands which include different generations. In addition, dispersal characteristics (gene flow) of firs revealed a sufficient seed and pollen flow of at least a few hundred meters from the mother trees. A high number of mother trees contributed to the seed dispersal and led to a multifaceted seed entry, even into the windthrow areas. Beyond, the analysis of the associated ECM fungi exhibited an identical spectrum of ECM fungi on the windthrow area and in the forest stand. We did not find evidence that the age of the trees can be regarded as driving factor for associated ECM communities on the population level. Based on the individual tree, adults host a higher number of ECM fungi than juveniles. Since the pre-windthrow offspring exhibited a well-balanced ECM profile they serve as ‘reservoir hosts’ for post-windthrow offspring promoting their vitality. Finally, we examined the data with respect to a possible correlation between host genotypes and associated ECM fungi. It became evident that the genomic background of silver fir as represented by single-locus variation has an effect on the composition of the associated ECM community. Consequently, ECM communities may be considered as extended phenotypes of the host populations. Protecting silver fir as a means of forest gene conservation therefore implies not only the tree species, but as well the interacting ECM community as part of the ecosystem. Based on the overall findings including tree genetic, dispersal and mycological aspects, silver fir populations in the Black Forest provide an appropriate basis for natural regeneration processes within the forest stand as well as for the recolonization of windthrow areas. Natural regeneration is an appropriate method for the reintroduction of larger proportions of silver fir in the Black Forest.

Zusammenfassung Eingeschränkter Genfluss in Populationen führt zur räumlichen Klumpung von verwandten Individuen. Dies hat Inzucht und einen Verlust der genetischen Diversität zur Folge. Die theoretischen Folgen dieser Prozesse sind grundsätzlich nachvollziehbar. Die konkreten Folgen für reale Organismen in einer realen, vom Menschen veränderten Landschaft sind jedoch nicht sofort offenkundig. Zusätzlich ergibt sich aus dem anthropogenen Einbringen von exotischen Arten oder gezüchteten Kulturpflanzen in die Landschaft die Möglichkeit von Introgression durch Genfluss zu ihren einheimischen nahen Verwandten. Die beiden Taxa Populus nigra (Europäische Schwarzpappel) und P. x canadensis (,Kanadapappel’, Hybrid aus P. deltoides und P. nigra) wurden als Modellsystem benutzt, um Kenntnis über quantitative Ausbreitungsmuster innerhalb des Mosaiks von verbliebenen natürlichen Vorkommen von P. nigra und Plantagen von P. x canadensis zu gewinnen. Hierfür wurden genetische Untersuchungen mit Hilfe von nSSR-Makern (nuclear simple sequence repeats, Mikrosatelliten-Orte des Zellkerns) durchgeführt. Dabei konnten Eigenschaften wie ein hoher Polymorphiegrad, kodominante Vererbung und das Vorhandensein von artspezifischen Allelen dazu verwendet werden, eine einfach zu handhabende Methode zur Gewinnung von genetischen Fingerabdrücken der Individuen zu erstellen. Es wurden öffentlich erhältliche allelische Leitern für jeden Locus hergestellt. Auf diese Weise kann das Markersystem dazu genutzt werden, verlässliche und eindeutige Ergebnisse für genetische Studien an Pappel zu erzeugen, die zwischen unterschiedlichen Genotypisierungssystemen übertragbar sind. Auf diese Weise ist es möglich, Klonvarietäten zu zertifizieren und molekulare Datenbanken zu vergrößern. Dies ist insbesondere für die Forstwirtschaft von großem Nutzen. In dieser Studie wurde das Markersystem außerdem dazu verwendet, die genetische Diversität und die räumlich-genetische Struktur einer natürlichen Schwarzpappelopulation zu bestimmen. Im Zusammenhang mit Plantagen in der Umgebung wurde die Hybridisierung zwischen P. nigra und P. x canadensis untersucht. Elternschaftsanalysen von Sämlingen sowie von Jungwuchs aus räumlich begrenzter Naturverjüngung wurden dazu genutzt, den pollen- und samenvermittelten Genfluss zu quantifizieren. Die räumlich-genetische Struktur der Schwarzpappel-Altbaumpopulation deutet darauf hin, dass Genfluss über kurze Entfernungen vorherrscht, dessen Hauptanteil von 70 % innerhalb von 1 km Umkreis um die Quelle stattfindet. Anhand dieser Ergebnisse lässt sich die verminderte genetische Diversität des untersuchten Jungwuchses erklären. Genfluss zwischen P. nigra und P. x canadensis wurde in beide Richtungen gefunden. Die jeweiligen Raten schwankten allerdings stark, was auf stochastische Umweltbedingungen sowie vor allem auf die räumliche Verteilung der Altbäume zurückgeführt wurde. Grundsätzlich begünstigt war die Hybridisierung von weiblichen P. x canadensis- und männlichen P. nigra- Bäumen. Zusätzlich konnte die Etablierung von Nachkommen mit Hybridisierungshintergrund nachgewiesen werden. Bezüglich dieser Ergebnisse werden praktische Konsequenzen sowohl für zukünftige Studien als auch für effektive Schutzmaßnahmen der Europäischen Schwarzpappel vorgeschlagen. Die Ergebnisse werden außerdem im Bezug auf Hybridisierungsprozesse und im Hinblick auf die Risikoeinschätzung für die Ausbreitung von Transgenen durch Genfluss von gentechnisch veränderten Kultursorten diskutiert. Die Ergebnisse dieser Studie können außerdem auf das Management von Pflanzenarten übertragen werden, die der Pappel ähnliche Ausbreitungsmechanismen aufweisen., English summary Limitations of gene flow in populations lead to spatial aggregation of related individuals, which implicates inbreeding and a loss of genetic diversity. The theoretical consequences of these processes are generally understood. However, consequences for real populations, distributed across man made landscapes are not immediately apparent. The anthropogenic introduction of exotic species or domesticated cultivars into landscapes has increased the possibility of introgressive gene flow to their wild relatives. The two taxa Populus nigra and P. x canadensis were used as a model system to obtain knowledge on the quantitative patterns of dispersal within and between the mosaic of remnant native individuals of P. nigra and plantations of P. x canadensis. For this purpose, key features of nSSR markers (nuclear simple sequence repeats), namely high polymorphism, codominant inheritance and availability of species-specific alleles were utilised to create an easy-to-handle tool for obtaining multilocus genetic fingerprints. As publicly available allelic ladders of each locus have been created, the presented marker system enables the generation of transferable genetic data on poplar in different laboratory settings. By this means, the marker assay can help to enlarge present clonal molecular data bases. It is also essential for certification purposes in commercial forestry. Using the marker assay, a natural population of P. nigra was analysed for genetic diversity and spatial genetic structure. Parentage analyses of seedlings as well as juveniles from a restricted area of natural regeneration enabled the quantification of pollen and seed-mediated gene flow, respectively. Consequences for genetic diversity could be concluded and consequences for the management of natural recruitment could be deduced. Spatial genetic patterns of the P. nigra adult tree population suggest prevailing short-distance gene flow, the major part of which (i. e.70 %) takes place within distances of less than 1 km. This helps to explain the reduced diversity in investigated juveniles. In the context of surrounding plantations, introgressive gene flow between P. nigra and the bred taxon P. x canadensis was studied. For this purpose, progeny of both taxa was analysed. Introgressive gene flow was found in both directions. Particular rates varied greatly and were probably due to stochastic environmental conditions and the spatial distribution of trees. However, preferential hybridisation was found between female P. x canadensis and male P. nigra. Moreover, introgressed individuals could be found in natural recruitment. Practical consequences for both upcoming studies and the conservation of natural P. nigra populations are implicated. Results of this research are discussed with respect to hybridisation processes and concerning the risk assessment of transgene flow from genetically modified taxa. Findings may also be transferred to management plans of plant species exhibiting similar dispersal mechanisms as poplar.

Der ehemals landschaftsbestimmende Wacholder (Juniperus communis L.) ist heute in vielen Ländern Europas nur noch in fragmentierten Populationen anzutreffen. Viele dieser Populationen weisen ein Naturverjüngungsdefizit und eine überalterte Vertikalstruktur auf. Um den Wacholder als einen Vertreter der europäischen Kulturlandschaft zu erhalten, erachte ich Restaurierungsmaßnahmen als unabdingbar. Hierin ist die Motivation der vorliegenden Doktorarbeit begründet. Zunächst nutzte ich unterschiedliche DNA-Marker-Systeme, um verschiedene Wacholderpopulationen auf unterschiedlichen räumlichen und zeitlichen Skalen auf genetische Effekte der Habitatfragmentierung hin zu überprüfen. Die gewonnenen Ergebnisse sollten dazu dienen, die auf regionaler Ebene untersuchten Populationen naturschutzfachlich zu bewerten und einen wissenschaftlich begründeten Managementplan für gefährdete Wacholderpopulationen zu erstellen. Dieser Plan sollte sich mit Nachbesserungen durch Ausbringen von Saat- und Pflanzgut beschäftigen. Auf europäischer Ebene setzte ich AFLP-Marker ein, um Aspekte der biogeographischen Geschichte des Wacholders zu rekonstruieren und mögliche genetische Abstammungslinien zu identifizieren. Es wird angenommen, dass derartige Linien geographische Areale beschreiben, innerhalb derer Saat- und Pflanzgut ausgetauscht werden kann. Die genetische Analyse ließ keine Abstammungslinien erkennen. Aufgrund der AFLP-Daten und im Zusammenhang mit Kenntnissen aus anderen wissenschaftlichen Disziplinen vermute ich, dass der Wacholder während der letzten Eiszeit in Zentraleuropa in zahlreichen kleinen Habitaten überdauerte. Möglicherweise waren diese diffus über Zentraleuropa verteilt und veränderten sich mehrmals in ihrer Größe und geographischen Lage. Für die Zeit zwischen der letzten maximalen Eisausdehnung bis heute sehe ich für Wacholder periodisch auftretende Fragmentierungs- und Gründerereignisse als wahrscheinlich an. Auf regionaler Ebene, im Rheinischen Schiefergebirge (RS) (Westdeutschland), setzte ich nSSR-Marker ein, um Einblicke in die genetische Struktur und Diversität von acht Wacholderpopulationen zu erhalten. Um die genetische Identität der Bestände durch Nachbesserungen mit Saat- und Pflanzgut nicht zu verändern und deren genetische Diversität zu erhalten, ist es notwendig, deren genetische Struktur und Diversität zu kennen. Gleichzeitig testete ich die Populationen auf mögliche genetische Effekte der derzeitigen Habitatfragmentierung. Die untersuchten Mikrosatellitenorte wurden im Rahmen dieser Arbeit charakterisiert, und es erfolgte eine detaillierte Validierung. In einer Fallstudie analysierte ich die genetische Diversität und Differenzierung unterschiedlicher Pollenwolken, die in der Folgegeneration (Embryonen) reproduktiv effektiv geworden sind. Hierfür wurde eigens ein neues Computerprogramm entwickelt. Darüber hinaus wurden durch die Analyse von in Moospolstern abgelagerten Pollenkörnern physikalische Pollenflugdistanzen für Wacholder bestimmt. Für die räumliche Organisation von Saat- und Pflanzgut könnten derartige Daten von gewisser Relevanz sein. Die beachtlich hohe genetische Diversität und das Fehlen genetischer Flaschenhalseffekte sowie das Fehlen eines „isolation-by-distance“-Effektes deuten darauf hin, dass sich die heutige Habitatfragmentierung noch nicht auf die genetische Struktur und Diversität der Bestände ausgewirkt hat. Stattdessen vermute ich, dass der Status der genetischen Diversität seit Beginn der Fragmentierung „festgefroren“ ist. Die genetische Diversität der Folgegenerationen ist im Vergleich zu den Altbeständen nicht reduziert, obwohl die palynologische Untersuchung auf stark lokal begrenzten Pollenflug hinweist. Auf Grundlage der genetischen Ergebnisse erfolgte eine naturschutzfachliche Bewertung der untersuchten Populationen im RS. Hierfür erstellte ich ein Leitbild, welches auf weit verbreiteten populationsökologischen und -genetischen Theorien basiert. Warum diese Bewertung nicht zufriedenstellend ausfiel, wird im Rahmen dieser Arbeit detailliert diskutiert. Weiterhin beleuchtete ich im Hinblick auf die „life-history traits“ und die hier postulierte biogeographische Geschichte des Wacholders die Kriterien und deren Qualitätsforderungen des aufgestellten Leitbildes kritisch. Aufgrund der vorliegenden genetischen Ergebnisse und des Naturverjüngungsdefizites in den Populationen bleibt es weiterhin ungewiss, ob sich die heutige Fragmentierung im RS in Zukunft negativ auswirken wird. Allerdings erscheint es als sehr wahrscheinlich, dass sich Populationen, die sich dauerhaft nicht verjüngen, durch sukzessives Sterben seneszenter Individuen verkleinern und in der Folge komplett aussterben werden. Deshalb entwickelte ich einen genetisch begründeten Managementplan zur Restaurierung sich nicht verjüngender Wacholderpopulationen. Er umfasst Leitlinien und Empfehlungen für die Sammelstrategie von Pflanzmaterial, für dessen Behandlung im Gewächshaus und für dessen Inverkehrbringen., The formerly continuous and widespread juniper (Juniperus communis L.) populations are currently divided into small and fragmented relics in numerous European countries. Additionally, many of these populations suffer from an absence of natural regeneration and consist predominantly of senescent individuals. In order to maintain juniper as a valuable element of the cultural landscape in Europe, I considered a restoration management to be indispensable. The goal of the present thesis is based on this consideration. Using different DNA marker systems, I firstly tested various juniper populations on different spatial and temporal scales for potential imprints of habitat fragmentation. Afterwards my intention was to evaluate the analysed populations on the regional scale in terms of nature conservation and to develop a scientifically based conservation management plan, which should focus on planting activities. In a Europe-wide study I used an AFLP marker approach to reconstruct aspects of the biogeographic history of juniper and to detect potential distinct genetic lineages. Those lineages are supposed to delineate geographic regions within which plant material can be interchanged. The genetic analysis revealed no distinct genetic lineages. Along with other scientific findings about juniper the results point to a glacial persistence of juniper in Central Europe. I suppose that during the last glacial period, this species managed to survive in several small and suitable habitats, which were probably diffusely scattered and permanently changing. Moreover, I hypothesise that recurrent fragmentation and founder events since the last glacial maximum (LGM) up until today are highly likely to have occurred in this species. On a regional scale, i.e. in the Rhenish Uplands (RU) (West Germany), I used nSSR markers to gain insights into the genetic structure and variation of eight relict juniper populations. Such knowledge is necessary for planting activities in order to prevent negative effects in the respective populations. At the same time, I tested these populations for genetic imprints of the recent habitat fragmentation. The investigated nSSR loci in juniper were characterised within the scope of this thesis. A detailed validation of the newly developed nSSR markers is presented. In addition, I performed a case study by investigating the genetic diversity and differentiation of different pollen clouds, which have become reproductively effective in the filial generations (embryos). For this purpose a specific computer software was developed. Next, a palynological study was conducted to determine physical pollen flow distances of juniper pollen grains. In terms of plantings I assume that such data is relevant for the spatial organisation of already existing juniper individuals and the respective plant material. Considerably high levels of genetic diversity and an absence of recent genetic bottlenecks in all populations as well as an absence of an isolation- by-distance effect led me to the assumption that the current habitat fragmentation has not yet affected the genetic diversity in the investigated juniper populations. Instead, I postulate that the genetic diversity and differentiation have been ‘frozen’ since the recent fragmentation started. The genetic diversity of the filial generation is not reduced in comparison to the adult generation, although the palynological study points to locally restricted pollen flow distances. After defining a ‘Leitbild’ for viable juniper populations based on widely accepted population ecological and genetic theories, I used the genetic results to evaluate the analysed populations of the RU in terms of nature conservation. The reasons why this evaluation was not satisfactory are discussed in detail. Further on, I commented critically on the evaluation criteria of the ‘Leitbild’ and their respective quality demands with regard to the life-history traits of juniper and its biogeographic history as presented within this thesis. Based on the presented genetic results and on the apparent absence of natural regeneration in all populations it remains uncertain whether the current habitat fragmentation will affect the genetic diversity and structure of the eight populations deleteriously in the future. However, if juniper will not start with natural regeneration again, this will certainly lead to an extinction of the respective populations because without substitution, senescent individuals will gradually die off. Thus, in the distant future juniper will probably become extinct in areas where it does not regenerate naturally. Therefore, I developed a sustainable, demographically and genetically substantiated restoration management plan as a final outcome of this thesis. It is based on the genetic analysis presented here and on expert knowledge, and it includes guidelines and recommendations concerning the collection of plant material, its treatment in the greenhouse and plantings in the field.
