Abstract Background Iron deficiency is prevalent in patients with chronic kidney disease (CKD), even in those without anemia. However, the effects of iron deficiency on CKD progression and all-cause mortality in non-dialysis-dependent CKD (NDD-CKD) patients without anemia remain incompletely understood. Methods This multicenter retrospective nationwide cohort study included adult patients with non-anemia NDD-CKD from 24 hospitals across China. The study investigated the associations between serum ferritin or transferrin saturation (TSAT) levels and the risks of CKD progression and all-cause mortality. Results Among 18,878 patients with NDD-CKD, 9,989 patients were included in the kidney outcome analysis, and 18,481 patients in the all-cause mortality analysis. Of the patients with the measurement, 2,450 (27.2%) had ferritin levels ≤ 100ng/mL and 2,440 (13.1%) had a TSAT level ≤ 20%. Compared with patients with TSAT level of > 20%, those with TSAT level of ≤ 20% had significantly higher risks of CKD progression (adjusted hazard ratio [aHR]: 1.66, 95% confidence intervals [CI]: 1.16–2.37; P = 0.005) and all-cause mortality (aHR: 2.21, 95% CI: 1.36–3.57; P = 0.001). The robustness of results was supported by subgroup analyses. However, there was no significant association found between ferritin levels and the risk of CKD progression or all-cause mortality (P > 0.05). Conclusion Iron deficiency was prevalent in NDD-CKD patients without anemia, and TSAT could be a modifiable risk factor of CKD progression and all-cause mortality. The screening of iron biomarkers, especially TSAT, in the early stage of NDD-CKD is important to assess and improve prognosis.

Abstract Background The effects of acute kidney injury (AKI) on liver-related outcomes in patients with hepatitis B virus (HBV) infection remain unclear. The study aimed to evaluate the association between AKI with liver-related mortality and complications in patients with HBV infection. Methods The multicenter, retrospective cohort study included Chinese adults with HBV infection from 24 regional central hospitals between January 2000 and December 2022. AKI was defined as a ≥ 26.5 μmol/l increase in serum creatinine concentration within 48 h, or a ≥ 50% increase over the baseline within 7 days. The primary outcome was post-discharge liver-related mortality, while the secondary outcome was a composite of new-onset liver cirrhosis and hepatocellular carcinoma. Cox proportional hazard model was employed for analyses. Results Of the 86,204 inpatients with HBV infection and without liver cancer or cirrhosis at baseline, 4407(5.1%) patients experienced AKI. During a mean follow-up of 4.6 ± 2.4 years, 334 (0.4%) patients died of liver-related events. After adjustment, AKI during hospitalization was significantly associated with a higher risk of liver-related mortality after discharge (adjusted hazard ratio (HR), 1.78; 95% confidence intervals (CI), 1.26–2.51, P = 0.001), especially in those with severe AKI. Similarly, AKI was associated with a higher risk of cirrhosis or new-onset hepatocellular carcinoma (adjusted HR, 1.33; 95%CI, 1.10–1.60, P = 0.004). The association between AKI and liver-related outcomes remained consistent across different subgroups. Conclusions AKI during hospitalization was associated with substantial increased risk of liver-related mortality and incident liver-related complication. Our findings highlight the importance of monitoring AKI in patients with HBV infection for tailoring personalized treatments.

This study aimed to investigate the differences in volatile composition and aroma of different fractions of Osmanthus fragrans absolute purified by molecular distillation. Gas chromatography-mass spectrometry (GC-MS) combined with orthogonal partial least-squares discrimination analysis (OPLS-DA), P-value, variable importance in the projection (VIP), and relative odor activity value (ROAV) were used for analysis. The light, heavy, and residue fractions of Osmanthus fragrans absolute were analyzed. A total of 73 volatile components belonging to 8 major categories were detected in the three fractions, including 22 alcohols, 5 aldehydes, 10 acids, 3 phenols, 5 ketones, 17 esters, 5 alkanes, and 6 olefins. Alcohols were the main contributors to the volatile components in each fraction, and the types and relative contents of volatile components significantly differed among the fractions (P1) of the three fractions, the key aroma contributing components (ROAV>1) of the 14 light fractions, 9 recombinant fractions, and 11 residue fractions were analyzed by the aroma profile analysis. Light fractions exhibited more intense floral, sweet, and pine woody aromas compared to recombinant and residue fractions. The light fractions had the highest types and relative contents of volatile components and showcased the most intense Osmanthus fragrans floral flavor. This study provided a theoretical basis for the comprehensive utilization of different fractions of Osmanthus fragrans absolute through molecular distillation using GC-MS combined with chemometrics methods.

The purpose of this study was to investigate the regulatory mechanism of Bacillus tequilensis (GCB-3) in improving the growth performance of grass carp by microbiome and targeted metabolomics analysis. Fish were fed with different amounts of GCB-3 including B0 (0 CFU/g), B6 (1×106 CFU/g), B7 (1×107 CFU/g), B8 (1×108 CFU/g), and B9 (1×109 CFU/g) for 8 weeks. The results indicate that with the increase of dietary GCB-3 concentration, grass carp showed a significant linear trend in growth performance, and appetite factors showed a significant linear and quadratic trend, the growth performance and appetite-promoting factors in the B8 group reach their maximum value in the B8 group. Microbiome analysis found that Proteobacteria and Actinobacteria were the most abundant phylum in the gut microbiota of grass carp. The functional potential predicted that GCB-3 affected the synthesis and degradation of carbohydrates, amino acids, fatty acids and lipids. It found that GCB-3 also affected the tricarboxylic acid (TCA) cycle VII (acetate-producers) pathway. The results of targeted metabolomics showed a significant linear and quadratic increase in intestinal acetate with GCB-3 concentration and a maximum in the B8 group. The results of the correlation analysis showed a significant positive correlation between Actinobacteria abundance and acetic acid content, and also between TCA cycle VII (acetate-producers) pathway abundance and Actinobacteria abundance and acetic acid content. In B8 group, grass carp showed a faster regulation speed of blood glucose levels after glucose injection, and the GLP-1 content remained stable. The activities of glycolytic enzymes, lipolytic enzymes, protein metabolism enzymes, and their related mRNA expression levels in grass carp showed a linear or quadratic increasing trend. However, the expression levels of gluconeogenic enzymes and fatty acid synthesis enzyme related mRNA showed an opposite trend. In summary, the addition of 1×108 CFU/g GCB-3 in grass carp feed can improve the abundance of dominant bacterial phyla in the intestine, especially Actinobacteria, increase the concentration of intestinal acetate, provide substrate for the TCA cycle VII (acetate-producers) pathway, thereby regulating the nutrient metabolism of grass carp and ultimately promoting growth.

Summary: Streptococcus agalactiae (S. agalactiae) is a contagious obligate parasite of the udder in dairy cows. Here, we examined S. agalactiae-host interactions in bovine mammary epithelial cells (BMECs) in vitro. We found that S. agalactiae infected BMECs through laminin β2 and integrin. Crk, Vps25, and RhoA were differentially expressed in S. agalactiae-infected cells. S. agalactiae infection activated FAK and Crk. FAK deficiency decreased the number of intracellular S. agalactiae and Crk activation. Knockdown of Crk or Vps25 increased the level of intracellular S. agalactiae, whereas its overexpression had the opposite effect. RhoA expression and actin cytoskeleton were altered in S. agalactiae-infected BMECs. Crk and Vps25 interact in cells, and invaded S. agalactiae also activates Crk, allowing it to cooperate with Vps25 to defend against intracellular infection by S. agalactiae. This study provides insights into the mechanism by which intracellular infection by S. agalactiae is regulated in BMECs.

With the advent of big data science, the field of geoscience has undergone a paradigm shift toward data-driven scientific discovery. However, the abundance of geoscience data distributed across multiple sources poses significant challenges to researchers in terms of data compilation, which includes data collection, collation, and database construction. To streamline the data compilation process, we present GeoKnowledgeFusion, a publicly accessible platform for the fusion of text, visual, and tabular knowledge extracted from the geoscience literature. GeoKnowledgeFusion leverages a powerful network of models that provide a joint multimodal understanding of text, image, and tabular data, enabling researchers to efficiently curate and continuously update their databases. To demonstrate the practical applications of GeoKnowledgeFusion, we present two scenarios: the compilation of Sm-Nd isotope data for constructing a domain-specific database and geographic analysis, and the data extraction process for debris flow disasters. The data compilation process for these use cases encompasses various tasks, including PDF pre-processing, target element recognition, human-in-the-loop annotation, and joint multimodal knowledge understanding. The findings consistently reveal patterns that align with manually compiled data, thus affirming the credibility and dependability of our automated data processing tool. To date, GeoKnowledgeFusion has supported forty geoscience research teams within the program by processing over 40,000 documents uploaded by geoscientists.

Generating SPARQL queries from natural language questions is challenging in Knowledge Base Question Answering (KBQA) systems. The current state-of-the-art models heavily rely on fine-tuning pretrained models such as T5. However, these methods still encounter critical issues such as triple-flip errors (e.g., (subject, relation, object) is predicted as (object, relation, subject)). To address this limitation, we introduce TSET (Triplet Structure Enhanced T5), a model with a novel pretraining stage positioned between the initial T5 pretraining and the fine-tuning for the Text-to-SPARQL task. In this intermediary stage, we introduce a new objective called Triplet Structure Correction (TSC) to train the model on a SPARQL corpus derived from Wikidata. This objective aims to deepen the model’s understanding of the order of triplets. After this specialized pretraining, the model undergoes fine-tuning for SPARQL query generation, augmenting its query-generation capabilities. We also propose a method named “semantic transformation” to fortify the model’s grasp of SPARQL syntax and semantics without compromising the pre-trained weights of T5. Experimental results demonstrate that our proposed TSET outperforms existing methods on three well-established KBQA datasets: LC-QuAD 2.0, QALD-9 plus, and QALD-10, establishing a new state-of-the-art performance (95.0% F1 and 93.1% QM on LC-QuAD 2.0, 75.85% F1 and 61.76% QM on QALD-9 plus, 51.37% F1 and 40.05% QM on QALD-10).

ObjectiveTo explore biomarkers of diabetic nephropathy (DN) and predict upstream miRNAs.MethodsThe data sets GSE142025 and GSE96804 were obtained from Gene Expression Omnibus database. Subsequently, common differentially expressed genes (DEGs) of renal tissue in DN and control group were identified and protein-protein interaction network (PPI) was constructed. Hub genes were screened from in DEGs and made an investigation on functional enrichment and pathway research. Finally, the target gene was selected for further study. The receiver operating characteristic (ROC) curve was used to evaluate the diagnostic efficiency of target gene and predicted its upstream miRNAs.Results130 common DEGs were obtained through analysis, and 10 Hub genes were further identified. The function of Hub genes was mainly related to extracellular matrix (ECM), collagen fibrous tissue, transforming growth factor (TGF) -β, advanced glycosylation end product (AGE) -receptor (RAGE) and so on. ABSTRACT: Plants demonstrate dynamic changes in molecular structures under fluctuating light conditions. Accumulating evidence suggests that light plays a vital role in plant growth and morphogenesis. In particular, light has a role in the absorption and utilization of nutrients in plants. Despite significant progress in understanding the mechanism of nutrient acquisition and assimilation, how light affects and regulates ion uptake remains a question. Studies in model plants, Arabidopsis thaliana, suggest that light affects the nutrient utilization in roots through a complex regulatory network; nonetheless, the molecular mechanisms underlying the various effects of light on these processes in crop plants remain fragmentary. In this review, we discuss the light effects (light quality, light intensity, and photoperiod) on nutrient uptake and utilization in horticultural crops for optimizing crop productivity and increasing fertilizer use efficiency.

IntroductionFamily history is a high-risk factor for colorectal cancer (CRC). The risk comes not only from known germline mutations but also from the other family-related mechanisms. Uncovering them would be an important step to improve the diagnosis and treatment of these patients.MethodSamples from 168 patients with advanced CRC were collected and applied to next-generation sequencing of 624 pan-cancer genes. Genomic mutations and significantly mutated genes were identified. Significantly mutated genes and co-mutated genes were used to cluster patients. For each cluster of patients, mutational signatures were extracted. The identified mutational signatures were further validated in the other independent cohort.ResultSignificantly mutated genes including TP53, APC, KRAS, and SMAD4 were found associated with tumor mutational burden and microsatellite instability. LRP1, ACVR2A, and SETBP1 were found co-mutated. Patients with mutations in LRP1, ACVR2A, and SETBP1 tend to have a family history of cancer. Those patients tended to have right-sided tumors with high tumor mutational burden and microsatellite instability. Among them, signature analysis identified two possible etiologies, SBS10a (defective polymerase epsilon exonuclease domain) and SBS6 (defective DNA mismatch repair and microsatellite unstable tumors). These signatures were also found in another independent cohort.ConclusionThe gene cluster (LRP1, ACVR2A, and SETBP1) could be a good biomarker of these patients with a family risk, which was characterized by right-sidedness, high tumor mutational burden, and high microsatellite instability.

Abstract Background Environmental lodging stress, which is a result of numerous factors, is characterized by uncertainty. However, several studies related to lodging in cereal crops have reported that lodging in the Hippeastrum rutilum environment is very rare. Hippeastrum rutilum is a garden flower with high ornamental value and abundant germplasm resources. Under past cultivation practices, it was found that the plant types of ‘Red Lion’, with red flowers, and ‘Apple Blossom’, with pink flowers, are quite different. The leaves of ‘Red Lion’ are upright, while the leaves of ‘Apple Blossom’ show lodging, which seriously affects its ornamental value. The aims of this study were to compare the differences between the two varieties with leaf lodging and upright leaves according to morphological and physiological attributes. In this study, karyotype analysis and phenotypic morphological and physiological characteristics were compared to explore the differences between the two plant types. Results The karyotype analysis of the two cultivars showed that their chromosome types were both tetraploid plants. The results showed that the lignin content in the leaves of ‘Red Lion’ was high, the cross-sectional structure of the leaf vascular bundle was more stable, and the chlorophyll content was high. In addition, significantly less energy was transferred to the electron transport chain (ETR) during the photoreaction. Similarly, the results regarding the maximum photosynthetic rate (Fv/Fm), nonphotochemical quenching (NPQ) and effective quantum yield of photosystem II photochemistry (△F/Fm′) all indicated that the photosynthetic capacity of “Red Lion” was greater than that of “Apple Blossom”, which was affected by leaf lodging. The size of the leaves was significantly smaller, and the leaf sag angle, leaf width, and leaf tip angle presented significantly lower values in ‘Red Lion’ than in ‘Apple Blossom’, which exhibits leaf sag. The difference in these factors may be the reason for the different phenotypes of the two cultivars. Conclusion The results of this study proved that lodging affects the photosynthetic capacity of Hippeastrum rutilum and revealed some indexes that might be related to leaf lodging, laying a theoretical foundation for cultivating and improving new varieties.

Abstract Background Pyroptosis is a recently identified pathway of caspase-mediated cell death in response to microbes, lipopolysaccharide, or chemotherapy in certain types of cells. However, the mechanism of how pyroptosis is regulated is not well-established. Methods Herein, the intracellular bacteria were detected by staining and laser confocal microscopy and TEM. Live/dead cell imaging assay was used to examine macrophage death. Western blot and immunohistochemical staining were used to examine the protein changes. IFA was used to identify typical budding vesicles of pyroptosis and the STAT3 nuclear localization. SEM was used to observe the morphological characteristics of pyroptosis. ELISA was used to detect the level of inflammatory cytokines. Pyroptosis was filmed in macrophages by LSCM. Results S. aureus was internalized by human macrophages. Intracellular S. aureus induced macrophage death. S. aureus invasion increased the expression of NLRP3, Caspase1 (Casp-1 p20) and the accumulation of GSDMD-NT, GSDMD-NT pore structures, and the release of IL-1β and IL-18 in macrophages. Macrophages pyroptosis induced by S. aureus can be abrogated by blockage of S. aureus phagocytosis. The pyroptosic effect by S. aureus infection was promoted by either rapamycin or Stattic, a specific inhibitor for mTORC1 or STAT3. Inhibition of mTORC1 or STAT3 induced pyroptosis. mTORC1 regulated the pyroptosic gene expression through governing the nuclear localization of STAT3. mTORC1/STAT3 axis may play a regulatory role in pyroptosis within macrophages. Conclusions S. aureus infection induces human macrophage pyroptosis, inhibition of mTORC1/STAT3 axis facilitates S. aureus-induced pyroptosis. mTORC1 and STAT3 are associated with pyroptosis. Our findings demonstrate a regulatory function of the mTORC1/STAT3 axis in macrophage pyroptosis, constituting a novel mechanism by which pyroptosis is regulated in macrophages. Artemisia selengensis Turcz. is a valuable edible and medicinal vegetable crop widely cultivated in Northeast China. Powdery mildew (PM) disease occurs during field and greenhouse cultivation, resulting in production losses and quality deterioration. The pathogen in A. selengensis was Golovinomyces artemisiae identified using optical microscopic and scanning electron microscopic observations, morphological identification, and molecular biological analyses. Parameters of chlorophyll fluorescence (ChlF) and antioxidant system responses as well as callose and lignin contents in A. selengensis were analyzed with inoculating G. artemisiae. Obvious of PM-infected leaves were confirmed with significantly lower values in electron transport rate (ETR), non-photochemical quenching (NPQ), photochemical quenching (qP), and actual photochemical efficiency [Y(II)], but higher values in non-adjusting energy dissipation yield [Y(NO)], supposed that maximal photosystem II quantum yield (Fv/Fm) value and images could be used to monitor PM degree on infectedA. selengensis. In addition, malondialdehyde (MDA), superoxide anion (O2–), callose, lignin contents, and peroxidase (POD) activity increased, while superoxide dismutase (SOD) activity, catalase (CAT) activity, and ascorbic acid (AsA) content decreased significantly in infected leaves compared to mock-inoculated leaves, indicated that lignin and protective enzymes are the key indicators for detecting PM resistant in A. selengensis. These results suggest that PM caused by G. artemisiae disrupted the photosynthetic capacity and induced imbalance of antioxidant system inA. selengensis. The findings were of great significance for designing a feasible approach to effectively prevent and control the PM disease in A. selengensis as well as in other vegetable crops.

Kentucky bluegrass (Poa pratensis L.) is a cool-season turfgrass species that responds strongly to nitrogen (N), but the metabolomic responses of this grass species to N supply is unknown. The N-tolerant cultivar Bluemoon and N-sensitive cultivar Balin were exposed to normal N (15 mM) and low N (0.5 mM) for 21 days for identification of differentially expressed metabolites (DEMs) between normal N and low N treatments. Balin had more reductions of chlorophyll and total soluble protein concentrations and a higher accumulation of superoxide radicals under low N stress. A total of 99 known DEMs were identified in either cultivar or both including 22 amino acids and derivatives, 16 carbohydrates, 29 organic acids, and 32 other metabolites. In Bluemoon, β-alanine metabolism was most enriched, followed by alanine, aspartate, and glutamate metabolism, biosynthesis of valine, leucine, and isoleucine biosynthesis, and glycine, serine, and threonine metabolism. In Balin, alanine, aspartate, and glutamate metabolism were most enriched, followed by the tricarboxylic acid (TCA), glyoxylate and decarbohydrate metabolism, and carbon fixation. Bluemoon generally maintained higher TCA cycle capacity and had more downregulated amino acids, while changes in more organic acids occurred in Balin under low N stress. Some metabolite changes by low-N stress were cultivar-specific. The results suggested that regulation of metabolites related to energy production or energy saving could contribute to low N tolerance in Kentucky bluegrass.

To investigate the heavy metal contamination of soil in a uranium mining area in northern Guangdong, a physicochemical evaluation method was used to evaluate the contaminated soil near the pit and tailings pond of the uranium mining area, determine its heavy metal content and evaluate its ecological risk using the Nemerow integrated contamination index, ground accumulation index and potential ecological risk index. The results show that the average content of nine heavy metal elements in the soil of the uranium mining area exceeds the background value of red soil in Guangdong Province. Three pollution evaluation indices all indicate that Cd, As and U have serious pollution and high ecological risk, while the remaining elements are weakly polluted and the potential ecological risk of the six sampling sites all show very strong risk. On this basis, soil ecotoxicity was evaluated using ostracods (Cypridopsis vidua and Heterocypris sp.), Vibrio fischeri and Vicia faba L. Higher concentrations of heavy metals at individual sites (T1, T2, P2) resulted in higher mortality of ostracods, higher inhibition of Vibrio fischeri luminescence and a significant reduction in germination and pigmentation of broad beans. The results of the biotoxicity evaluation were consistent with the results of the physicochemical evaluation, allowing for a more direct and comprehensive evaluation of the ecotoxic effects of uranium and heavy metals in the mine soils.

Rapid rise in temperature in summer causes severe injury to cool-season turfgrass of both native species and introduced ones in Heilongjiang of Northeast China. The objectives of this study were to compare physiological responses to seasonal heat stresses and turf performances between native and introduced commercial Poa accessions. Three Chinese native Poa species (i.e., P. pratensis, P. sibirica and P. sphondylodes) and three USA Kentucky bluegrass cultivars (ie. ‘Midnight’, ‘Moonlight’ and ‘BlueChip’) were evaluated under field conditions in 2017 and 2018. All accessions showed unique characteristics and considerable seasonal differences in response to temperatures. However, performances over all accessions were largely similar in early spring and autumn. In summer, native P. pratensis performed similar to ‘Midnight’, ‘Moonlight’ or ‘BlueChip’, with respect to such traits or parameters as quality, coverage, color intensity, growth rate, osmolytes, ROS and anti-oxidant production. Native P. pratensis could be used as a new turf resource for further improvement and application under the specific climatic conditions in Heilongjiang; native P. sphondylodes may be used in repairing damaged environments or for alternative seasonal greenness.

Phenanthrene (PHE), a typical organic pollutant, has drawn attention in recent years due to its toxicity to plants and human health. Gamma-aminobutyric acid (GABA) induce plant tolerance to diverse stresses. However, the role and regulatory mechanisms of GABA in PHE stress responses in plants remains largely uncharacterized. Here, we showed that GABA content increased by 44.5%, 89.2%, 160% and 39.2% under 50, 100, 200 and 300 µM PHE treatment, respectively compared with mock. GABA treatment alleviated PHE-induced growth inhibition in a dose-dependent manner, with the most effective concentration of 50 mM GABA. Although exogenous GABA could not influence the accumulation of PHE in cucumber, it significantly mitigated photosynthetic inhibition and enhanced the transcripts and activities of the antioxidant enzymes such as ascorbate peroxidase (APX), catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD), resulting in less accumulation of hydrogen peroxide (H2O2) and superoxide (O2.–). Importantly, timecourse analyses of glutathione (GSH) homeostasis showed that GABA markedly increased GSH content and GR activity as well as the transcripts of GSH biosynthesis-related genes GSH1, GSH2 and GR during PHE stress. Conversely, pretreatment with GSH biosynthesis inhibitor buthionine sulfoximine (BSO) abolished the GABA-induced changes in PHE stress. Together, these results suggest that GABA enhances tolerance to PHE stress via a GSH-dependent system of antioxidant defense in cucumber.

To gain insights into the roles of beneficial PGPR in controlling soil-borne disease, we adopted a metabolomics approach to investigate the beneficial impacts of P. polymyxa NSY50 on cucumber seedling roots under the pathogen of Fusarium oxysporum f. sp. cucumerinum (FOC). We found that NSY50 pretreatment (NSY50 + FOC) obviously reduced the production of reactive oxygen species (ROS). Untargeted metabolomic analysis revealed that 106 metabolites responded to NSY50 and/or FOC inoculation. Under FOC stress, the contents of root osmotic adjustment substances, such as proline and betaine were significantly increased, and dehydroascorbic acid and oxidized glutathione (GSH) considerably accumulated. Furthermore, the contents of free amino acids such as tryptophan, phenylalanine, and glutamic acid were also significantly accumulated under FOC stress. Similarly, FOC stress adversely affected glycolysis and the tricarboxylic acid cycles and transferred to the pentose phosphate pathway. Conversely, NSY50 + FOC better promoted the accumulation of α-ketoglutaric acid, ribulose-5-phosphate, and 7-phosphosodiheptanone compared to FOC alone. Furthermore, NSY50 + FOC activated GSH metabolism and increased GSH synthesis and metabolism-related enzyme activity and their encoding gene expressions, which may have improved redox homoeostasis, energy flow, and defense ability. Our results provide a novel perspective to understanding the function of P. polymyxa NSY50, accelerating the application of this beneficial PGPR in sustainable agricultural practices.

The development of uranium mines has been necessary to obtain abundant and scarce uranium resources, but they also bring inevitable radioactive contamination to the surrounding soil, rivers and lakes. This paper explores the sensitivity of Cypridopsis vidua to the radioactive element uranium and the heavy elements cadmium and copper with single and combined acute toxicity experiments and combined toxicity model predictions. The results from the single toxicity experiments showed that the degree of toxic effects was cadmium > copper > uranium. The combined toxicity experiments showed that the compound toxicity of U-Cd and U-Cu was higher than the weakest component and lower than the strongest component, whereas the compound toxicity of Cd-Cu was higher than either of its components. When the overall proportion of a more toxic metal was increased, its mixed toxicity also increased, and vice versa. Combined toxicity predictions showed that the U-Cd combination was best described by the concentration additive (CA) model, the independent action (IA) model was more applicable to the Cd-Cu combination, and the most applicable model for the U-Cu combination changed depending on the concentration gradient. The acute toxicity data from this study provide a reference for the development of wastewater discharge standards for uranium mines, enriches the data related to the toxicity of uranium for ostracods and deepens the understanding of the threat of uranium pollution to aquatic ecosystems.

Understanding the differences in cool-season turfgrass responses to shade is critical for future turfgrass management and breeding for improved shade tolerance. The purpose of this study was to evaluate the shade-tolerance mechanisms of two cool-season turfgrass species in terms of turf performance, growth, and physiological characteristics. Two turfgrass species, namely, ’SupraNova’ (Poa. supina Schrad.) and ‘Lark’ (Lolium perenne L.), were subjected to 0 (CK, unshaded), 35% (LS), 70% (MS), and 92% (HS) shade, respectively. Compared with ‘Lark’, ‘SupraNova’ showed better turf quality (TQ) and turf color intensity (TCI) under shade. The total length and surface area of the roots of ‘Lark’ gradually decreased as the shade increased, while those of ‘SupraNova’ increased and then decreased with increasing shade. The chlorophyll fluorescence photochemical quenching coefficient (qP), electron transport rate (ETR), and maximum quantum yield of primary photosystem II (PSII) photochemistry (Fv/Fm) decreased significantly under HS; however, these decreases were more significant in ‘Lark’ than in ‘SupraNova’. The leaf reflectance of the two turfgrasses under shade was lower than that under CK, but the leaf reflectance of ‘Lark’ was higher than that of ‘SupraNova’ in the visible light band. The normalized difference vegetation index (NDVI) of the two grasses first decreased and then increased. The NDVI of ‘Lark’ under shade was slightly higher than that under CK. ‘SupraNova’ showed strong tolerance on the basis of malondialdehyde (MDA), hydrogen peroxide (H2O2), superoxide anion (O2·−), and ascorbic acid (AsA) contents and superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) activity. The MDA, H2O2, O2·−, and AsA contents and SOD, POD, and CAT activity (which represent indicators) changed the most under MS. Taken together, the results indicated that the adaptability of ‘SupraNova’ to shade was better than that of ‘Lark’.

The excessive accumulation of copper (Cu2+) has become a threat to worldwide crop production. Recently, it was revealed that melatonin (MT) could play a crucial role against heavy metal (HM) stresses in plants. However, the underlying mechanism of MT function acted upon by Cu2+ stress (CS) has not been substantiated in tomatoes. In the present work, we produced MT-rich tomato plants by foliar usage of MT, and MT-deficient tomato plants by employing a virus-induced gene silencing methodology and exogenous foliar application of MT synthesis inhibitor para-chlorophenylalanine (pCPA). The obtained results indicate that exogenous MT meaningfully alleviated the dwarf phenotype and impeded the reduction in plant growth caused by excess Cu2+. Furthermore, MT effectively restricted the generation of reactive oxygen species (ROS) and habilitated cellular integrity by triggering antioxidant enzyme activities, especially via CAT and APX, but not SOD and POD. In addition, MT increased nonenzymatic antioxidant activity, including FRAP and the GSH/GSSG and ASA/DHA ratios. MT usage improved the expression of several defense genes (CAT, APX, GR and MDHAR) and MT biosynthesis-related genes (TDC, SNAT and COMT). Taken together, our results preliminarily reveal that MT alleviates Cu2+ toxicity via ROS scavenging, enhancing antioxidant capacity when subjected to excessive Cu2+. These results build a solid foundation for developing new insights to solve problems related to CS.

In this paper, we propose a unified and flexible framework for general image fusion tasks, including multi-exposure image fusion, multi-focus image fusion, infrared/visible image fusion, and multi-modality medical image fusion. Unlike other deep learning-based image fusion methods applied to a fixed number of input sources (normally two inputs), the proposed framework can simultaneously handle an arbitrary number of inputs. Specifically, we use the symmetrical function (e.g., Max-pooling) to extract the most significant features from all the input images, which are then fused with the respective features from each input source. This symmetry function enables permutation-invariance of the network, which means the network can successfully extract and fuse the saliency features of each image without needing to remember the input order of the inputs. The property of permutation-invariance also brings convenience for the network during inference with unfixed inputs. To handle multiple image fusion tasks with one unified framework, we adopt continual learning based on Elastic Weight Consolidation (EWC) for different fusion tasks. Subjective and objective experiments on several public datasets demonstrate that the proposed method outperforms state-of-the-art methods on multiple image fusion tasks.

An extracellular signal-regulated kinase (ERK) is a mitogen-activated protein kinase (MAPK) in mammalian cells that interact with the mechanistic target of rapamycin (mTOR) signalling pathway. ERK signalling integrates signals from growth factors, nutrients, and mitogens to regulate cell proliferation, differentiation, and the cell cycle. The purpose of this study was to examine the inhibitory effects of U0126, a specific MEK/ERK inhibitor, on ERK and mTOR signalling, Lipin 1 protein level, PPARγ activation, and triglyceride (TAG) and palmitic acid (PA) accumulation in Cashmere goat foetal fibroblasts (GFbs). The proliferation of GFbs was significantly impeded by U0126, the IC50 of which was 1.54 µM. Image dehazing is a technique to enhance the images acquired in poor weather conditions, such as fog and haze. Existing image dehazing methods are mainly based on dark channel prior. Since the dark channel is not reasonable for sky regions, a sky segmentation and region wised medium transmission based image dehazing method is proposed in this paper. Firstly, sky regions are segmented by quad-tree splitting based feature pixels detection. Then, a medium transmission estimation method for sky regions is proposed based on color characteristic observation of sky regions. The medium transmission is then filtered by an edge preserving guided filter. Finally, based on the estimated medium transmission, the hazed images are restored. Experimental results demonstrate that the performance of the proposed method is better than that of existing methods. The restored image is more natural, especially in the sky regions.

Mammalian target of rapamycin complex 1 (mTORC1) is a central regulator of cell growth and metabolism and is sufficient to induce specific metabolic processes, including de novo lipid biosynthesis. Elongation of very-long-chain fatty acids 1 (ELOVL1) is a ubiquitously expressed gene and the product of which was thought to be associated with elongation of carbon (C) chain in fatty acids. In the present study, we examined the effects of rapamycin, a specific inhibitor of mTORC1, on ELOVL1 expression and docosahexaenoic acid (DHA, C22:6 n-3) synthesis in bovine mammary epithelial cells (BMECs). We found that rapamycin decreased the relative abundance of ELOVL1 mRNA, ELOVL1 expression and the level of DHA in a time-dependent manner. These data indicate that ELOVL1 expression and DHA synthesis are regulated by mTORC1 in BMECs.

Nitric oxide (NO), a well-known signaling molecule plays an important role in abiotic and biotic stress-induced production of plant secondary metabolites. In this study, roles of NO in water stress-induced tanshinone production in Salvia miltiorrhiza hairy roots were investigated. The results showed that accumulations of four tanshinone compounds in S. miltiorrhiza hairy roots were significantly stimulated by sodium nitroprusside (SNP, a NO donor) at 100 μM. Effects of SNP were just partially arrested by the mevalonate (MVA) pathway inhibitor (mevinolin), but were completely inhibited by the 2-C-methyl-d-erythritol-4-phosphate pathway (MEP) inhibitor (fosmidomycin). The increase of tanshinone accumulation and the up-regulation of HMGR and DXR expression by PEG and ABA treatments were partially inhibited by an inhibitor of NO biosynthesis (Nω-nitro-L-arginine methyl ester (L-NAME)) and a NO scavenger (2-(4-Carboxyphenyl)- 4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (c-PTIO)). Simultaneously, NO generation in the hairy roots was triggered by PEG and ABA, and the effects were also arrested by c-PTIO and L-NAME. These results indicated that NO signaling probably plays a central role in water stress-induced tanshinone production in S. miltiorrhiza hairy roots. SNP mainly stimulated the MEP pathway to increase tanshinone accumulation.

To detect goat vascular endothelial growth factor (VEGF)-mediated regrowth of hair, full-length VEGF164 cDNA was cloned from Inner Mongolia cashmere goat (Capra hircus) into the pET-his prokaryotic expression vector, and the recombinant plasmid was transferred into E. coli BL21 cells. The expression of recombinant 6×his-gVEGF164 protein was induced by 0.5 mM isopropyl thio-β-D-galactoside at 32°C. Recombinant goat VEGF164 (rgVEGF164) was purified and identi ed by western blot using monoclonal anti-his and anti-VEGF antibodies. The rgVEGF164 was smeared onto the dorsal area of a shaved mouse, and we noted that hair regrowth in this area was faster than in the control group. Thus, rgVEGF164 increases hair growth in mice.

Better features have been driving the progress of pedestrian detection over the past years. However, as features become richer and higher dimensional, noise and redundancy in the feature sets become bigger problems. These problems slow down learning and can even reduce the performance of the learned model. Current solutions typically exploit dimension reduction techniques. In this paper, we propose a simple but effective feature selection framework for pedestrian detection. Moreover, we introduce occluded pedestrian samples into the training process and combine it with a new feature selection criterion, which enables improved performances for occlusion handling problems. Experimental results on the Caltech Pedestrian dataset demonstrate the efficiency of our method over the state-of-art methods, especially for the occluded pedestrians.

Salvia miltiorrhiza is one of the most important traditional Chinese medicinal plants because of its excellent performance in treating coronary heart disease. Phenolic acids mainly including caffeic acid, rosmarinic acid and salvianolic acid B are a group of active ingredients in S. miltiorrhiza. Abscisic acid (ABA), gibberellin (GA) and ethylene are three important phytohormones. In this study, effects of the three phytohormones and their interactions on phenolic production in S. miltiorrhiza hairy roots were investigated. The results showed that ABA, GA and ethylene were all effective to induce production of phenolic acids and increase activities of PAL and TAT in S. miltiorrhiza hairy roots. Effects of phytohormones were reversed by their biosynthetic inhibitors. Antagonistic actions between the three phytohormones played important roles in the biosynthesis of phenolic acids. GA signaling is necessary for ABA and ethylene-induced phenolic production. Yet, ABA and ethylene signaling is probably not necessary for GA3-induced phenolic production. The complex interactions of phytohormones help us reveal regulation mechanism of secondary metabolism and scale-up production of active ingredients in plants.

Sweroside is a natural monoterpene derived from Swertia pseudochinensis Hara. Recently, studies have shown that sweroside exhibits a variety of biological activities, such as anti-inflammatory, antioxidant, and hypoglycemic effects. However, its role and mechanisms in high glucose (HG)-induced renal injury remain unclear. Herein, we established a renal injury model in vitro by inducing human renal tubular epithelial cell (HK-2 cells) injury by HG. Then, the effects of sweroside on HK-2 cell activity, inflammation, reactive oxygen species (ROS) production, and epithelial mesenchymal transition (EMT) were observed. As a result, sweroside treatment ameliorated the viability, inhibited the secretion of inflammatory cytokines (TNF-α, IL-1β, and VCAM-1), reduced the generation of ROS, and inhibited EMT in HK-2 cells. Moreover, the protein expression of SIRT1 was increased and the acetylation of p65 NFkB was decreased in HK-2 cells with sweroside treatment. More importantly, EX527, an inhibitor of SIRT1, that inactivated SIRT1, abolished the improvement effects of sweroside on HK-2 cells. Our findings suggested that sweroside may mitigate HGcaused injury in HK-2 cells by promoting SIRT1-mediated deacetylation of p65 NF-kB. [ABSTRACT FROM AUTHOR]

Hereditary spastic paraplegia (HSP) is a severe neurodegenerative movement disorder, the underlying pathophysiology of which remains poorly understood. Mounting evidence has suggested that iron homeostasis dysregulation can lead to motor function impairment. However, whether deficits in iron homeostasis are involved in the pathophysiology of HSP remains unknown. To address this knowledge gap, we focused on parvalbumin-positive (PV+) interneurons, a large category of inhibitory neurons in the central nervous system, which play a critical role in motor regulation. The PV+ interneuron-specific deletion of the gene encoding transferrin receptor 1 (TFR1), a key component of the neuronal iron uptake machinery, induced severe progressive motor deficits in both male and female mice. In addition, we observed skeletal muscle atrophy, axon degeneration in the spinal cord dorsal column, and alterations in the expression of HSP-related proteins in male mice with Tfr1 deletion in the PV+ interneurons. These phenotypes were highly consistent with the core clinical features of HSP cases. Furthermore, the effects on motor function induced by Tfr1 ablation in PV+ interneurons were mostly concentrated in the dorsal spinal cord; however, iron repletion partly rescued the motor defects and axon loss seen in both sexes of conditional Tfr1 mutant mice. Our study describes a new mouse model for mechanistic and therapeutic studies relating to HSP and provides novel insights into iron metabolism in spinal cord PV+ interneurons and its role in the regulation of motor functions.
