The properties of flotation foam within a flotation system play a crucial role in its interaction with mineral particles, directly impacting the efficiency and selectivity of the entire flotation process. Currently, most collectors exhibit favorable foaming properties, and the formation and characteristics of the foam in the collector system significantly affect the regulation of the mineral flotation process. Given the poor selectivity of conventional anionic collectors in magnesite flotation, developing new collectors has become a hot topic in mineral processing research. Investigating the relationship between foam properties and flotation outcomes is significant for optimizing the mineral flotation process and enhancing the utilization rate of mineral resources. Previous research has revealed that potassium cetyl phosphate can selectively adsorb on magnesite surfaces, enabling effective flotation separation of magnesite from calcium-bearing gangue minerals. This suggests significant application prospects for potassium cetyl phosphate as a new anionic collector in froth flotation purification of magnesite ores. However, to our knowledge, the foam properties of this collector have not been extensively studied. This study employs the DFA 100 Dynamic Foam Analyzer to systematically investigate the foam properties of two anionic magnesite collectors. Key performance differences, including foaming characteristics, foam stability, and structural integrity under various pH conditions and different Mg2+ concentrations resulting from mineral dissolution, were examined. A comparative analysis relative to the conventional sodium oleate collector systems was also conducted. The results indicate that, under identical mass concentration conditions, the surface tension of the cetyl phosphate ester collector varies more gently. As a novel anionic magnesite collector, CP demonstrates significantly superior foam adaptability under changing pH values and Mg2+ concentrations compared to the traditional sodium oleate (NaOl) collector. Specifically, within the studied ranges of pH values and Mg2+ concentrations, cetyl phosphate exhibits superior foaming propensity and more pronounced foam stability. Notably, the foam structure generated by cetyl phosphate is more stable, with inhibited foam coalescence observed at elevated pH values. By contrast, the NaOl collector loses its foaming ability in solution environments with pH values below 8.0 and Mg2+ concentrations exceeding 20 mg·L−1. Single-mineral flotation tests further confirm the consistency between the flotation rate of magnesite and the impact of the solution environment on the foam properties of both collectors. Under conditions where the pH is below 6.0 or the Mg2+ concentration exceeds 30 mg·L−1, traditional collector NaOl struggles to capture mineral particles during magnesite flotation. By contrast, the flotation rate of magnesite with the cetyl phosphate collector remains above 60%. The experiments also revealed that the microstructure of foam generated by the cetyl phosphate collector has a more uniform size distribution and a more compact foam layer structure. These characteristics are crucial for improving the collection efficiency of mineral particles and enhancing the selectivity of the flotation process. This study provides a solid theoretical foundation for the application of phosphate ester collectors in magnesite flotation through an in-depth analysis of foam microstructure and a comprehensive comparative assessment of foam properties between phosphate ester and oleate collectors under varying solution conditions. Furthermore, this study offers valuable practical insights for efficient collector selection and flotation process optimization in industrial applications. These insights contribute to advancing magnesite flotation upgrading technology and processes. The results presented in this paper also serve as an important reference for optimizing the theory and practice of calcium and magnesium salt minerals. This is significant for the development of flotation technology.

Abstract Background Pseudorabies virus (PRV), porcine parvovirus (PPV) and porcine circovirus 3 (PCV3) are common in swine farms in China. Single infection or co-infection with PRV, PPV and/or PCV3 was difficult to distinguish between their clinical symptoms and pathological changes. Therefore, a quick and accurate detection method is needed for epidemiological surveillance, disease management, import and export control. Methods In the present study, we established a multiplex real-time PCR assay based on SYBR Green I for the simultaneous detection of PRV, PPV and PCV3 genomes. Results PRV, PPV and PCV3 were distinguished in the same sample by their different melting temperatures (Tm), with melting peaks at 90 °C for PRV, 84 °C for PPV and 80 °C for PCV3, respectively, and other non-targeted swine pathogens did not exhibit specific melting peaks. The assay showed a high degree of linearity (R2≧0.995), and the detection limits were 4.76 copies/μL for PRV, 3.67 copies/μL for PPV, 3.07 copies/μL for PCV3 and 1.87 × 102 copies/μL for the three mixed plasmids, respectively. In this research, 81 clinical samples from pig farms in nine different regions of Guangdong Province were used to evaluate this new method. The detection rate of the multiplex real-time PCR assay was higher than that of the conventional PCR assay. Conclusions This multiplex real-time PCR assay could be used as a diagnostic tool that is rapid, sensitive and reliable for the detection of co-infection of PRV, PPV and PCV3 as well as for molecular epidemiological surveillance.

Susceptibility to pathogens in the elderly is heightened with age, largely because of immunosenescence. As an immune regulatory organ, bone marrow creates immune cells that move to other organs and tissues through the blood. Despite the significance of this process of this organ, there is limited research on changes in immune cell generation in the bone marrow and their effects on immunosenescence. In this study, the compositions of immune cells in bone marrow from young (three months) and old (24+ months) mice were compared by means of mass cytometry, with further validation obtained through the reanalysis of single-cell RNA sequencing data and cell sorting via flow cytometry. The effects of differential immune cells on immunosenescence in old mice were evaluated using the Clostridium difficile (C. difficile) infection model. Our results showed that aged mice presented with a reduction in bone trabeculae structure, which was accompanied by a notable increase in polymorphonuclear (PMN)-myeloid-derived suppressor cell (MDSC) abundance. Through bulk-seq and reverse transcription quantitative polymerase chain reaction (RT-qPCR) analysis, we identified differential genes associated with the immune response—specifically, the Th17 cell differentiation pathway. Furthermore, the increase in exported PMN-MDSCs to the large intestine resulted in increased gut permeability and inflammatory damage to the colon following C. difficile infection. After clearing the PMN-MDSCs in old mice using the anti-Gr-1 antibody, the symptoms induced by C. difficile were significantly relieved, as evidenced by an inhibited IL-17 pathway in the colon and reduced gut permeability. In conclusion, aging increases the number of PMN-MDSCs in both the generated bone marrow and the outputted intestine, which contributes to susceptibility to C. difficile infection. This study provides a novel target for anti-aging therapy for immunosenescence, which is beneficial for improving immune function in elders.

Abstract Background China is ranked third globally in terms of burden and has a moderately high to high prevalence of tuberculosis (TB). This study meticulously investigated the notification rates of TB and assessed the epidemic in China from 2000 to 2021. The aim of the study was to provide robust supporting data that is crucial for enhancing TB prevention and control strategies. Methods Extensive data regarding TB notification rates in China between 2000 and 2021 was collected. The joinpoint regression model was subsequently utilized to assess the temporal trends in the notification rates of TB, which were analyzed through the annual percentage change (APC) and the average annual percentage change (AAPC). Abstract Objective Inadequate tuberculosis (TB) knowledge and awareness of proper TB control practices among health care workers (HCWs) may increase the risk of nosocomial TB transmission. This study aimed to assess HCWs’ TB-related knowledge and control practices to guide the development of more effective targeted TB health education and training programs. Methods In January 2023 a cross-sectional survey was administered to 323 HCWs employed by five primary health care centers and three secondary comprehensive medical institutions in Beijing, China. Survey data were collected using a standard questionnaire. Results Analysis of survey responses revealed TB knowledge and practices awareness rates of 60.4% and 90.6%, respectively. The overall average awareness rate across all 19 TB knowledge- and practice-related questions was 70.0%. Intermediate- and senior-level HCW’s average TB knowledge score was respectively 2.225 and 8.175 times higher than that of primary-level HCWs, while the average TB knowledge score of HCWs in secondary comprehensive medical institutions was 3.052 times higher than that of HCWs in primary health care centers. Higher average TB knowledge score correlated with higher-level professional titles and higher level work units, but higher average TB control practices score correlated with employment at primary health care center rather than secondary comprehensive medical institution. Notably, 13.6% of HCWs had not received TB training during the past three years, while 86.1% expressed willingness to undergo online TB training. Conclusion These findings highlight inadequate TB knowledge and awareness of proper TB control practices among HCWs in primary health care centers and secondary comprehensive medical institutions in Beijing, underscoring the urgent need for targeted educational and training initiatives to improve TB awareness and control efforts.

Abstract In advanced hepatocellular carcinoma (HCC), RNA helicase DDX5 regulates the Wnt/β-catenin-ferroptosis axis, influencing the efficacy of the multi-tyrosine kinase inhibitor (mTKI) sorafenib. DDX5 inhibits Wnt/β-catenin signaling, preventing sorafenib-induced ferroptosis escape. Sorafenib/mTKIs reduce DDX5 expression, correlating with poor patient survival post-sorafenib treatment. Notably, DDX5-knockout in HCC cells activates Wnt/β-catenin signaling persistently. Herein, we investigate the mechanistic impact of Wnt/β-catenin activation resulting from DDX5 downregulation in the progression and treatment of HCC. RNAseq analyses identified shared genes repressed by DDX5 and upregulated by sorafenib, including Wnt signaling genes, NF-κB-inducing kinase (NIK) essential for non-canonical NF-κB (p52/RelB) activation, and cytoprotective transcription factor NRF2. We demonstrate, Wnt/β-catenin activation induced NIK transcription, leading to non-canonical NF-κB activation, which subsequently mediated NRF2 transcription. Additionally, DDX5 deficiency extended NRF2 protein half-life by inactivating KEAP1 through p62/SQSTM1 stabilization. In a preclinical HCC mouse model, NRF2 knockdown or DDX5 overexpression restricted tumor growth upon sorafenib treatment, via induction of ferroptosis. Importantly, DDX5-knockout HCC cells exhibited elevated expression of Wnt signaling genes, NIK, p52/RelB, and NRF2-regulated genes, regardless of sorafenib treatment. Transcriptomic analyses of HCCs from TCGA and the Stelic Animal Model (STAM) of non-alcoholic steatohepatitis revealed elevated expression of these interconnected pathways in the context of DDX5 downregulation. In conclusion, DDX5 deficiency triggers Wnt/β-catenin signaling, promoting p52/RelB and NRF2 activation, thereby enabling ferroptosis evasion upon sorafenib treatment. Similarly, independent of sorafenib, DDX5 deficiency in liver tumors enhances activation and gene expression of these interconnected pathways, underscoring the clinical relevance of DDX5 deficiency in HCC progression and therapeutic response.

In order to explore a new way for the application of phosphogypsum in road materials, phosphogypsum powder and asphalt mortar were mixed to prepare phosphogypsum modified asphalt. The effects of mixing temperature, powder-binder ratio and polyphosphoric acid content on the three major indicators of phosphogypsum modified asphalt were studied. The research results show that the softening point increases as the mixing temperature increases, and the ductility increased first and then kept constant. Considering energy efficiency and aging temperature, the processing temperature of asphalt should be in the range 135 °C and 145 °C. increasing the powder gum ratio can increase the softening point of the modified asphalt and decrease the ductility of the asphalt. Therefore, the ratio of filler bitumen should not be greater than 0.9 using phosphogypsum as the modifier. The addition of polyphosphoric acid greatly increases the softening point of modified asphalt, but reduces its ductility and penetration. The optimal amount of polyphosphoric acid should be around 1.5%. Infrared spectroscopy (FTIR) and thermogravimetric analysis (TG-DSC) showed that phosphogypsum physically functioned with Asphalt. When polyphosphoric acid was mixed into asphalt, it would react chemically with asphalt. The research results could provide references for the engineering application and theoretical research of phosphogypsum in road materials.

Feed efficiency and growth performance are economically important traits in pigs. Precious studies have been revealed that both genetics and gut microbes could influence host phenotypes, however, the mechanisms by which they affect pig growth and feed efficiency remain poorly understood. In this study, 361 crossbred Duroc × (Landrace × Yorkshire) commercial pigs were genotyped using GeneSeek Porcine SNP50K BeadChip, and the microbiotas from fecal samples were acquired using microbial 16S rRNA gene sequencing technology to investigate the impact of host genetics and gut microorganisms on growth and feed efficiency. The results showed that the heritability and enterobacterial force ranged from 0.27 to 0.46 and 0 to 0.03, respectively. Genome-wide association studies (GWAS) identified seven significant SNPs to be associated with growth and feed efficiency, and several genes, including AIF1L, ASS1, and QRFP were highlighted as candidates for the analyzed traits. Additionally, microbiome-genome-wide association studies GWAS revealed potential links between CCAR2, EGR3, GSTM3, and GPR61 genes and the abundance of microorganisms, such as Trueperella, Victivallis, and Erysipelatoclostridium. In addition, six microbial genera linked to growth and feed efficiency were identified as follows Lachnospiraceae_UCG-005, Prevotellaceae_UCG-003, Prevotellaceae_NK3B31_group, Prevotella_1, Prevotella_9, and Veillonella. Our findings provide novel insights into the factors influencing host phenotypic complexity and identify potential microbial targets for enhancing pig feed efficiency through selective breeding. This could aid in the development of strategies to manipulate the gut microbiota to optimize growth rates and feed efficiency in pig breeding.

Thus, it is a promising green adsorbent. Due to its small size and large specific surface area, nano-hydroxyapatite (nHAP) exposes more adsorption sites, showing supreme adsorption capacity. It is a tendency to produce nHAP composites of strong adsorption capacity, easy for solid-liquid separation and regeneration, and low-cost. This article reviews the recent research progress of nano-HAP and its composites as adsorbents for wastewater treatment, and prospects the application of them as adsorbents.

Abstract Background A better diagnostic marker is in need to distinguish breast cancer from suspicious breast lesions. The abnormal glycosylation of haptoglobin has been documented to assist cancer diagnosis. This study aims to evaluate disease-specific haptoglobin (DSHp)-β N-glycosylation as a potential biomarker for breast cancer diagnosis. Methods DSHp-β chains of 497 patients with suspicious breast lesions who underwent breast surgery were separated from serum immunoinflammatory-related protein complexes. DSHp-β N-glycosylation was quantified by mass spectrometric analysis. After missing data imputation and propensity score matching, patients were randomly assigned to the training set (n = 269) and validation set (n = 113). Logistic regression analysis was employed in model and nomogram construction. The diagnostic performance was analyzed with receiver operating characteristic and calibration curves. Results 95 N-glycopeptides at glycosylation sites N207/N211, N241, and N184 were identified in 235 patients with benign breast diseases and 262 patients with breast cancer. Systematic process mineralogy study on phosphorus tailings was carried out by using the advanced mineral identification and characterization system (AMICS). The chemical compositions, mineral components, embedded characteristics of minerals, particle size, and the characteristics of liberation degree and the other characteristics were mainly studied. The results show that the gangue minerals are mainly dolomite and quartz. The phosphate mineral is mainly collophane which exists in the form of poor intergrowth. The phosphate mineral is extremely finely disseminated and closely associated with the gangue minerals. This study can provide some guidance for the optimization of phosphate ore dressing and the comprehensive utilization of tailings resources.

ABSTRACT: This study aims to identify candidate genes related to ovarian development after ovarian tissue transplantation through transcriptome sequencing (RNA-seq) and expression network analyses, as well as to provide a reference for determining the molecular mechanism of improving ovarian development following ovarian tissue transplantation. We collected ovarian tissues from 15 thirty-day-old ducks and split each ovary into 4 equal portions of comparable sizes before orthotopically transplanting them into 2-day-old ducks. Samples were collected on days 0 (untransplanted), 3, 6, and 9. The samples were paraffin sectioned and then subjected to Hematoxylin-Eosin (HE) staining and follicular counting. We extracted RNA from ovarian samples via the Trizol method to construct a transcriptome library, which was then sequenced by the Illumina Novaseq 6000 sequencing platform. The sequencing results were examined for differentially expressed genes (DEG) through gene ontology (GO) function and the Kyoto encyclopedia of genes and genomes (KEGG) pathway analyses, gene set enrichment analysis (GSEA), weighted correlation network analysis (WGCNA), and protein-protein interaction (PPI) networks. Some of the candidate genes were selected for verification using real-time fluorescence quantitative PCR (qRT-PCR). Histological analysis revealed a significant reduction in the number of morphologically normal follicles at 3, 6, and 9 d after ovarian transplantation, along with significantly higher abnormality rates (P < 0.05). The transcriptome analysis results revealed 2,114, 2,224, and 2,257 upregulated DEGs and 2,647, 2,883, and 2,665 downregulated DEGs at 3, 6, and 9 d after ovarian transplantation, respectively. Enrichment analysis revealed the involvement multiple pathways in inflammatory signaling, signal transduction, and cellular processes. Furthermore, WGCNA yielded 13 modules, with 10, 4, and 6 candidate genes mined at 3, 6 and 9 d after ovarian transplantation, respectively. Transcription factor (TF) prediction showed that STAT1 was the most important TF. Finally, the qRT-PCR verification results revealed that 12 candidate genes exhibited an expression trend consistent with sequencing data. In summary, significant differences were observed in the number of follicles in duck ovaries following ovarian transplantation. Candidate genes involved in ovarian vascular remodeling and proliferation were screened using RNA-Seq and WGCNA.

Semen quality and persistence are critical for evaluating the usability of individual boars in AI, a standard practice in pig breeding. We conducted GWASs on various semen traits of Duroc boars, including MOT, DEN, ABN, MMP, AIR, and ROS levels. These traits were assessed using FCM and CASA. A total of 1183 Duroc boars were genotyped using the GeneSeek GGP Porcine 50 K SNP BeadChip. The GWAS was performed using three different models: GLM, MLM, and FarmCPU. Additionally, trait heritability was estimated using single- and multiple-trait PBLUP models, yielding 0.19, 0.29, 0.13, 0.18, 0.11, and 0.14 heritability for MOT, DEN, ABN, MMP, AIR, and ROS, respectively. All semen traits exhibited low heritability except ABN, which demonstrated medium heritability. Nine candidate genes (GPX5, AWN, PSP-II, CCDC62, TMEM65, SLC8B1, TRPV4, UBE3B, and SIRT5) were potentially associated with semen traits. These genes are associated with antioxidant and mitochondrial functions in porcine sperm. Our findings provide insight into the genetic architecture of semen traits in Duroc boars, and the identified SNPs and candidate genes may enhance economic outcomes in the pig breeding industry while improving sperm quality through targeted breeding strategies.

Blastocystis spp. and Enterocytozoon bieneusi are common intestinal pathogens capable of infecting both humans and animals, which lead to severe diarrhea and other intestinal diseases, posing a threat to public health. The Bamaxiang pig, a specialty of Bama Yao Autonomous County in Guangxi Province, China, is an important local breed in the regional pork market and an excellent model animal for biomedical research. Currently, no data is available on the prevalence or genotype distribution of these pathogens in Bamaxiang pigs. This study aimed to determine the prevalence and genetic characteristics of Blastocystis spp. and E. bieneusi in three Bamaxiang pig farms located in Bama Yao Autonomous County, using molecular techniques based on the small subunit ribosomal RNA (SSU rRNA) gene fragment of Blastocystis spp. and the internal transcribed spacer (ITS) region of E. bieneusi. All positive PCR products from the 311 fecal samples were sequenced to identify the species and genotypes of these organisms. The overall infection rates of Blastocystis spp. and E. bieneusi were 34.08% (106/311) and 18.32% (57/311), respectively. Three subtypes of Blastocystis spp. were detected: ST1 (n = 8), ST3 (n = 3), and ST5 (n = 95). Among them, zoonotic ST5 was the dominant genotype, accounting for 89.62% (95/106) of strains, followed by the genotypes ST1 (7.54%, 8/106) and ST3 (2.83%, 3/106). Two genotypes of E. bieneusi were detected: EbpC (n = 52) and CHG23 (n = 5), with EbpC being the dominant genotype. The human-pathogenic subtypes (ST1, ST3, and ST5) and genotypes (EbpC, CHG23) that were observed in this study indicate a potential threat to public health. Our findings offer basic information for preventing and controlling these zoonotic pathogens in the study area. Additional investigations are necessary to better understand their genetic characteristics and zoonotic potential within Guangxi Province.

Abstract Reduced expression of the RNA helicase DDX5 associated with increased hepatocellular carcinoma (HCC) tumor grade and poor patient survival following treatment with sorafenib. While immunotherapy is the first-line treatment for HCC, sorafenib and other multi-tyrosine kinase inhibitors (mTKIs) are widely used when immunotherapy is contra-indicated or fails. Herein, we elucidate the role of DDX5 in sensitizing HCC to sorafenib, offering new therapeutic strategies. Treatment of various human HCC cell lines with sorafenib/mTKIs downregulated DDX5 in vitro and in preclinical HCC models. Conversely, DDX5 overexpression reduced the viability of sorafenib-treated cells via ferroptosis, suggesting a role for DDX5 in sorafenib sensitivity. RNAseq of wild-type vs. DDX5-knockdown cells treated with or without sorafenib identified a set of common genes repressed by DDX5 and upregulated by sorafenib. This set significantly overlaps with Wnt signaling genes, including Disheveled-1 (DVL1), an indispensable Wnt activator and prognostic indicator of poor survival for sorafenib-treated patients. DDX5-knockout (DDX5KO) HCC cells exhibited DVL1 induction, Wnt/β-catenin pathway activation, and ferroptosis upon inhibition of canonical Wnt signaling. Consistently, xenograft HCC tumors exhibited reduced growth by inhibition of Wnt/β-catenin signaling via induction of ferroptosis. Significantly, overexpression of DDX5 in HCC xenografts repressed DVL1 expression and increased ferroptosis, resulting in reduced tumor growth by sorafenib. We conclude that DDX5 downregulation by sorafenib mediates adaptive resistance by activating Wnt/β-catenin signaling, leading to ferroptosis escape. Conversely, overexpression of DDX5 in vivo enhances the anti-tumor efficacy of sorafenib by suppressing Wnt/β-catenin activation and induction of ferroptosis. Thus, DDX5 overexpression in combination with mTKIs is a promising therapeutic strategy for HCC.

Porcine epidemic diarrhea (PED) is a highly contagious intestinal infection primarily affecting pigs. It is caused by the porcine epidemic diarrhea virus (PEDV). PEDV targets the villus tissue cells in the small intestine and mesenteric lymph nodes, resulting in shortened intestinal villi and, in extreme cases, causing necrosis of the intestinal lining. Moreover, PEDV infection can disrupt the balance of the intestinal microflora, leading to an overgrowth of harmful bacteria like Escherichia coli. Exosomes, tiny membrane vesicles ranging from 30 to 150 nm in size, contain a complex mixture of RNA and proteins. MicroRNA (miRNA) regulates various cell signaling, development, and disease progression processes. This study extracted exosomes from both groups and performed high-throughput miRNA sequencing and bioinformatics techniques to investigate differences in miRNA expression within exosomes isolated from PEDV-infected porcine small intestine tissue compared to healthy controls. Notably, two miRNA types displayed upregulation in infected exosomes, while 12 exhibited downregulation. These findings unveil abnormal miRNA regulation patterns in PEDV-infected intestinal exosomes, shedding light on the intricate interplay between PEDV and its host. This will enable further exploration of the relationship between these miRNA changes and signaling pathways, enlightening PEDV pathogenesis and potential therapeutic targets.

Cement-sand reinforced soft clay (C-SRSC) is a complex multiphase geomaterial. Its strength is determined by the physical properties of the internal multiphase substances and the coupling mechanical response between various phases of substances. By considering the effect of the particle size and content of sand particles on the unconfined compressive strength (UCS) and failure mechanism of C-SRSC, the C-SRSC is divided into two phases of the cement soil matrix and sand particles to construct a micro cell model of C-SRSC. Based on the strain gradient theory, the theoretical model of the UCS of C-SRSC based on the physical mechanism at the microscale is derived. Forty five groups of UCS tests were conducted to analyze the effect of sand particle size and content on the UCS of C-SRSC, and to calculate the theoretical model parameters. The results show that the UCS of C-SRSC increases with increasing curing age, cement content, and sand particle content, and decreases with the increasing sand particle size. The theoretical model of the UCS of C-SRSC based on physical mechanism initially verified the consistency of the experimental and theoretical results.

ABSTRACT: Since 2021, a novel strain of goose reovirus (GRV) has emerged within the goose farming industry in Guangdong province, China. This particular viral variant is distinguished by the presence of white necrotic foci primarily localized in the liver and spleen, leading to substantial economic losses for the poultry industry. However, the etiology, prevalence and genomic characteristics of the causative agent have not been thoroughly investigated. In this study, we conducted an epidemiological inquiry employing suspected GRV samples collected from May 2021 to September 2022. The macroscopic pathological and histopathological lesions associated with GRV-infected clinical specimens were examined. Moreover, we successfully isolated the GRV strain and elucidated the complete genome sequence of the isolate GD21/88. Through phylogenetic and recombination analysis, we unveiled that the GRV strains represent a novel variant resulting from multiple reassortment events. Specifically, the μNS, λC, and σNS genes of GRV were found to have originated from chicken reovirus, while the σA gene of GRV exhibited a higher degree of similarity with a novel duck reovirus. The remaining genes of GRV were traced back to Muscovy duck reovirus. Collectively, our findings underscore the significance of GRV as a pathogenic agent impacting the goose farming industry. The insights gleaned from this study contribute to a more comprehensive understanding of the epidemiology of GRV in Southern China and shed light on the genetic reassortment events exhibited by the virus.

ABSTRACT: Avian influenza virus (AIV) poses a significant threat to the poultry industry and public health. Among the diverse AIV subtypes, H3, H4, and H5 are frequently detected in waterfowl and live poultry markets (LPM). The expeditious and precise identification of these subtypes is imperative in impeding the dissemination of the disease. In this study, we have developed a triplex real-time PCR assay endowed with the capacity to simultaneously discriminate AIV subtypes H3, H4, and H5. This method showcases remarkable specificity, selectively amplifying H3, H4, and H5 AIV subtypes sans any cross-reactivity with other subtypes or common avian pathogens. Furthermore, this method exhibits high sensitivity, with a detection threshold of 2.1 × 102 copies/μL for H3, H4, and H5 AIV subtypes. Additionally, the assay demonstrates reproducibility, as evidenced by intra- and interassay variability, with a coefficient of variation below 1.5%. A total of 338 cloacal swabs were collected from LPM to evaluate the performance of our assay. The obtained results evinced a high level of concordance with the sequencing data. In summary, our study has developed a triplex real-time PCR method that can be employed in laboratory-based testing and surveillance of AIV. This assay holds promise in augmenting our ability to detect and monitor AIV subtypes, thereby facilitating timely interventions and safeguarding both the poultry industry and public health.

Abstract The exact pathophysiology of the spaceflight-associated neuro-ocular syndrome (SANS) has so far not been completely elucidated. In this study we assessed the effect of acute head-down tilt position on the mean flow of the intra- and extracranial vessels. Our results suggest a shift from the external to the internal system that might play an important role in the pathomechanism of SANS.

AbstractBackground Glioma is a common primary central nervous system tumor with complex pathogenesis. DNA damage and repair (DDR) is widely involved in regulating cell proliferation and tumorigenesis by correcting and repairing DNA damage mechanisms. Recent studies have reported the following properties in cancer cells in glioma, increased DNA damage and reduced DNA repair capacity. However, the relationship between glioma and DDR-related genes was unclear.Methods DDR-related risk score model was built. The validity of this model was validated in detail through the Kaplan-Meier survival analysis, tumor mutational burden (TMB) analysis, immune cell infiltration, sensitivity to treatment regimens. Moreover, the model’s adaptability was validated in different glioma data cohorts and different glioma subgroups. To further investigate the molecular mechanism of one of DDR-related gene (NUDT1) in glioma, U251 cell was used for the knockdown experiment, followed by MTT, wound healing and transwell analysis.Results Ten prognostic-related DDR-related signature genes were obtained, including EID3, MGMT, YWHAG, PMS1, SHPRH, HUS1, NUDT1, GADD45G, APEX1 and FAM175A. The RT-qPCR results suggested that the latter five genes were highly expressed in glioma patients. Interestingly, high TMB score had longer survival. In high-risk score groups, reduced immune cell infiltration in the tumor microenvironment lead to poorer patient outcomes. Sensitivity to treatment regimens analysis indicated that low-risk score groups were more sensitive to chemotherapeutics. Moreover, the risk score model had a good prediction effect on different glioma datasets and different glioma subgroups. In vitro mechanism study showed that knockdown of NUDT1 reduced tumorigenesis. Furthermore, knockdown of NUDT1 remarkably reduced the expression level of HIF-1α.Conclusion DDR-related risk score model built-in this work has good predictive performance for glioma.Key messagesTen prognostic-related DDR-related signature genes were obtained, including EID3, MGMT, YWHAG, PMS1, SHPRH, HUS1, NUDT1, GADD45G, APEX1 and FAM175A.In high-risk score groups, reduced immune cell infiltration in the tumor microenvironment leads to poorer patient outcomes.The risk score model had a good prediction effect on different glioma datasets and different glioma subgroups.Knockdown of NUDT1 reduced tumorigenesis of glioma and remarkably reduced the expression level of HIF-1α.

ABSTRACT: The purpose of this study was to investigate the effects of melittin on production performance, antioxidant function, immune function, heat shock protein, intestinal morphology, and cecal microbiota of heat-stressed quails. A total of 120 (30-day-old) male quails were randomly divided into 3 groups. Each group consisted of 4 replicates with 10 birds per replicate. The ambient temperature of the control group (group W) was 24°C ± 2°C. The heat stress group (group WH) and the heat stress + melittin group (group WHA2) were subjected to heat stress for 4 h from 12:00 to 16:00 every day, and the temperature was 36°C ± 2°C for 10 d. The results showed that compared with the group W, heat stress significantly decreased growth performance, serum and liver antioxidative function, immune function, intestinal villus height (VH) and villus height-to-crypt depth ratio (VH/CD), and cecal microbiota Chao and ACE index (P < 0.05). The crypt depth (CD) in the small intestine, and HSP70 and HSP90 mRNA levels in the heart, liver, spleen, and kidney were significantly increased (P < 0.05). Dietary melittin significantly increased growth performance, serum and liver antioxidative function, immune function, intestinal VH and VH/CD, and cecal microbiota Shannon index in heat-stressed quails (P < 0.05). Melittin significantly decreased small intestinal CD, and HSP70 and HSP90 mRNA levels in the viscera (P < 0.05). Furthermore, dietary melittin could have balanced the disorder of cecal microbiota caused by heat stress and increased the abundance and diversity of beneficial microbiota (e.g., Firmicutes were significantly increased). PICRUSt2 functional prediction revealed that most of the KEGG pathways with differential abundance caused by high temperature were related to metabolism, and melittin could have restored them close to normal levels. Spearman correlation analysis showed that the beneficial intestinal bacteria Anaerotruncus, Bacteroidales_S24-7_group_norank, Lachnospiraceae_unclassified, Shuttleworthia, and Ruminococcaceae_UCG-014 increased by melittin were positively correlated with average daily feed intake, the average daily gain, serum and liver superoxide dismutase, IgG, IgA, bursa of Fabricius index, and ileum VH and VH/CD. In sum, our results demonstrate for the first time that dietary melittin could improve the adverse effects of heat stress on antioxidant function, immune function, heat shock protein, intestinal morphology, and cecal microbiota in quails, consequently improving their production performance under heat stress.

Artificial insemination plays a crucial role in pig production, particularly in enhancing the genetic potential of elite boars. To accelerate genetic progress for semen traits in pigs, it is vital to understand and identify the underlying genetic markers associated with desirable traits. Herein, we genotyped 1238 Landrace boars with GeneSeek Porcine SNP50 K Bead chip and conducted genome-wide association studies to identify genetic regions and candidate genes associated with 12 semen traits. Our study identified 38 SNPs associated with the analyzed 12 semen traits. Furthermore, we identified several promising candidate genes, including HIBADH, DLG1, MED1, APAF1, MGST3, MTG2, and ZP4. These candidate genes have the potential function to facilitate the breeding of boars with improved semen traits. By further investigating and understanding the roles of these genes, we can develop more effective breeding strategies that contribute to the overall enhancement of pig production. The results of our study provide valuable insights for the pig-breeding industry and support ongoing research efforts to optimize genetic selection for superior semen traits.

With the rapid expansion of foundation pit excavation areas due to the burgeoning underground transportation networks in large cities, spatial combined double-row retaining structures have become indispensable for ensuring stability and minimizing deformation. Integrating piers into these structures enhances stiffness; however, the research on this topic is limited. This study investigates the Chisha Metro project, which utilizes a double-row support structure with added support piers to improve overall stiffness. Numerical simulations are employed to model the foundation pit and analyze the stress distribution and deformation of double-row retaining systems with T-shaped, cross-shaped, or square-shaped piers. The results demonstrate that the retaining system, after the introduction of piers, exhibits reduced maximum horizontal displacement and surface settlement of soil, and indicates spatial characteristics and the transformation from a planar problem to a spatial problem. Furthermore, in specific geological conditions, the T-shaped support pier proves to be more effective in connecting the front and rear support structures compared to the cross-shaped and square-shaped support piers, which mainly reinforce the soil without connecting the support structures. This research provides valuable insights for improving design and construction practices in foundation pit engineering.

Microelectromechanical systems (MEMS) gas sensors have numerous advantages such as compact size, low power consumption, ease of integration, etc., while encountering challenges in sensitivity and high resistance because of their low sintering temperature. This work utilizes the in situ growth of Zeolitic Imidazolate Framework-8 (ZIF-8) followed by its conversion to N-doped ZnO. The results obtained from scanning electron microscopy (SEM) and transmission electron microscopy (TEM) indicate that the in situ derivation of ZIF-8 facilitates the adhesion of ZnO particles, forming an island-like structure and significantly reducing the interfaces between these particles. Furthermore, powder X-ray diffraction (XRD) analysis, elemental mapping, and X-ray photoelectron spectroscopy (XPS) analysis confirm the conversion of ZIF-8 to ZnO, the successful incorporation of N atoms into the ZnO lattice, and the creation of more oxygen vacancies. The ZIF-8-derived N-doped ZnO/MEMS sensor (ZIF (3)-ZnO/MEMS) exhibits remarkable gas sensitivity for ethanol detection. At an operating temperature of 290 °C, it delivers a substantial response value of 80 towards 25 ppm ethanol, a 13-fold enhancement compared with pristine ZnO/MEMS sensors. The sensor also exhibits an ultra-low theoretical detection limit of 11.5 ppb to ethanol, showcasing its excellent selectivity. The enhanced performance is attributed to the incorporation of N-doped ZnO, which generates abundant oxygen vacancies on the sensor’s surface, leading to enhanced interaction with ethanol molecules. Additionally, a substantial two-order-of-magnitude decrease in the resistance of the gas-sensitive film is observed. Overall, this study provides valuable insights into the design and fabrication strategies applicable to high-performance MEMS gas sensors in a broader range of gas sensing.

Shrub encroachment via anthropogenic introduction has been used for ecological restoration in degraded desert grassland. However, there have been debates concerning whether this activity can be effective, as contrasting results were frequently reported. The objectives of this study were to address if various Caragana korshinskii planting conditions (planting year and density) had varying influences on soil organic carbon (SOC), soil total nitrogen (STN), and their spatial heterogeneities along the transition from desert grassland to shrubland. We investigated SOC and STN at the community and landscape scales, with the former containing eight land types of enclosed grassland, grazed grassland, and shrubland with varying levels of planting years (3, 12, or 22 years) and planting intervals (2, 6, or 40 m), and the latter consisting of 43 grassland-shrubland mosaic sites (desert grassland, grassland-shrubland edge, and shrubland) covering approximately 2,976 km2 area. At the community scale, the SOC and STN contents from 0 to 100 cm soil depth in shrubland, regardless of varying planting years and densities, were higher than those observed in grazed grassland. Though it appeared that short-term of shrub establishment (3 and 12 years) in shrubland outscored enclosed grassland in SOC accumulation, it was still questionable if this can be consistent for a longer time period as no significant difference was seen for SOC contents between enclosed grassland and shrubland after 22 years of shrub establishment. At the landscape scale, the SOC contents were higher in shrubland than those in desert grassland. However, no significant difference was observed for STN among shrubland and desert grassland. Regarding their spatial heterogeneities, SOC exhibited stronger spatial autocorrelation in grassland-shrubland edge than in desert grassland and shrubland, while STN did not evidently differ in spatial autocorrelation among these transition states. In brief, we have observed differences in soil carbon and nitrogen at community scales, but when crossing over to broad scales do not show the kind of significant spatial heterogeneity that we thought would be present in shrub introductions over the past 30 years. There may be a certain lag in the evolution of soil carbon and nitrogen responses from the introduction of shrubs from the community scale to the landscape scale.

Patients with advanced breast cancer are difficult to treat and have poor prognosis. At present, the commonly used methods to monitor the disease progression of breast cancer are imaging examinations such as breast ultrasound, mammography and peripheral blood tumor markers such as carcinoembryonic antigen (CEA) and carbohydrate antigen 15-3 (CA15-3). However, none of them can detect tumor progression at an early stage. Serum immunoinflammation-related protein complexes (IIRPCs) showed potential to indicate cancer progression. Therefore, we attempted to monitor the level of IIRPCs in peripheral blood of patients with metastatic breast cancer and compare it with patients’ treatment and disease progression, and here we performed case reports of two of them.

The intensities and phase profiles of the elliptic optics vortex beam both are determined by the topological charge (TC) and ellipticity. This paper presents an efficient and simple method for measuring the TC and ellipticity of elliptic vortex. By observing the diffraction patterns, the TC modulus and ellipticity can be probed with the suitable rectangular aperture, and the TC sign can be detected with the suitable right triangular aperture. The scheme works well even for high-order elliptic vortex with topological charge value as high as ± 18, and ellipticity range from 0.5 to 2 at least. Obviously, the measurement of TC and ellipticity have important research value for promoting the application of elliptical vortex.

Subclinical mastitis (SCM) is a very common disease of dairy cows. Currently, somatic cell count (SCC) is used for SCM diagnoses. There are no prognostic tests to detect which cows may develop SCM during the dry-off period. Therefore, the objectives of this study were to identify metabolic alterations in the serum of pre-SCM cows during the dry-off period, at −8 and −4 weeks before calving, through a targeted mass spectrometry (MS) assay. Fifteen cows, free of any disease, and 10 cows affected only by SCM postpartum served as controls (CON) and the SCM group, respectively. Results showed 59 and 47 metabolites that differentiated (p ≤ 0.05) CON and pre-SCM cows at –8 and −4 weeks prior to the expected date of parturition, respectively. Regression analysis indicated that a panel of four serum metabolites (AUC = 0.92, p < 0.001) at −8 weeks and another four metabolites (AUC = 0.92, p < 0.01) at −4 weeks prior to parturition might serve as predictive biomarkers for SCM. Early identification of susceptible cows can enable development of better preventive measurements ahead of disease occurrence.

Cycloid gear tooth profile modification is very important to the performance of RV reducer，in order to select the shape modification method and parameter size reasonably and improve the stress state of the cycloid tooth surface，an equidistant & radical-moving modification method based on particle swarm optimization algorithm to optimize bearing capacity is proposed. Based on the RV-60E reducer as an optimization example，the mechanical analysis of the cycloid needle wheel transmission part is carried out first，and the mathematical model of the cycloid wheel shape is established with the goal of reducing the maximum bearing capacity of the needle teeth. The required shape modification parameter values and tooth profile curve are obtained by using the proposed optimization design method. Compared with equidistant trimming，radical-moving and angle fitting modification methods，it is found that the maximum contact force of cycloidal gear profile modification method based on particle swarm optimization algorithm is reduced by 15.9%，16.0% and 11.8%，respectively，thus improving the bearing capacity and life of RV reducer.

The novel duck reovirus (NDRV) emerged in southeast China in 2005. The virus causes severe liver and spleen hemorrhage and necrosis in various duck species, bringing serious harm to waterfowl farming. In this study, three strains of NDRV designated as NDRV-ZSS-FJ20, NDRV-LRS-GD20, and NDRV-FJ19 were isolated from diseased Muscovy ducks in Guangdong and Fujian provinces. Pairwise sequence comparisons revealed that the three strains were closely related to NDRV, with nucleotide sequence identities for 10 genomic fragments ranging between 84.8 and 99.8%. In contrast, the nucleotide sequences of the three strains were only 38.9–80.9% similar to the chicken-origin reovirus and only 37.6–98.9% similar to the classical waterfowl-origin reovirus. Similarly, phylogenetic analysis revealed that the three strains clustered together with NDRV and were significantly different from classical waterfowl-origin reovirus and chicken-origin reovirus. In addition, the analyses showed that the L1 segment of the NDRV-FJ19 strain was a recombinant of 03G and J18 strains. Experimental reproduction of the disease showed that the NDRV-FJ19 strain was pathogenic to both ducks and chickens and could lead to symptoms of hemorrhage and necrosis in the liver and spleen. This was somewhat different from previous reports that NDRV is less pathogenic to chickens. In conclusion, we speculated that the NDRV-FJ19 causing duck liver and spleen necrosis is a new variant of a duck orthoreovirus that is significantly different in pathogenicity from any previously reported waterfowl-origin orthoreovirus.

BackgroundThere are no universally acknowledged standardized treatment strategies for blood blister-like aneurysms (BBAs). This study compared the prognosis of patients with BBA who underwent craniotomy microsurgery vs. endovascular intervention.MethodsThis retrospective cohort study included patients with BBA treated between September 2009 and August 2020 at Sichuan Provincial People's Hospital affiliated to the Sichuan Academy of Medical Science. Patients were divided into the microsurgery and endovascular groups. The preoperative Hunt-Hess grade and modified Fisher grade were collected. The intraoperative and postoperative complications (including intraoperative aneurysm rupture and hemorrhage, postoperative cerebral hemorrhage, and BBA recurrence) were recorded.ResultsSeventy-two patients were included: 28 and 44 in the microsurgery and endovascular groups, respectively. Only the preoperative Fisher grade was different between the two groups (P = 0.041). The proportion of patients with good outcomes was lower in the microsurgery group (28.6%) than in the endovascular group (72.7%), and the mortality rate was higher in the microsurgery group (32.1%) than in the endovascular group (11.4%) (P < 0.05). After adjustment for the modified Fisher grade, the multivariable analysis showed that compared with craniotomy microsurgery, an endovascular intervention was associated with the prognosis of patients with BBA (OR = 0.128, 95%CI: 0.040–0.415, P < 0.001). The rate of complications (intraoperative hemorrhage, cerebral infarction, and recurrence) was higher in the microsurgery group than in the endovascular group.ConclusionIn patients with BBA, an endovascular intervention appears to be associated with a better prognosis compared with craniotomy microsurgery.

ABSTRACT: To study the effects of melittin on egg-laying performance and intestinal barrier of quails, 240 quails (aged 70 d) were randomly divided into 4 groups with 6 replicates (10 quails per replicate). They were fed with basal diet (group B), basal diet + 0.08 g/kg melittin (group BA1), basal diet + 0.12 g/kg melittin (group BA2) and basal diet + 0.16 g/kg melittin (group BA3). The experiment lasted for 21 days. The eggs were collected every day. At the end of the experiment, duodenal, jejunal, and ileal tissues were collected, and the cecal contents were sampled. Intestinal antioxidant index, barrier function, and intestinal flora were analyzed. The results showed that the addition of melittin significantly increased the laying rate and average egg weight. Addition of melittin significantly increased the antioxidant function, mechanical barrier, immune barrier, and the villus height to crypt depth ratio of small intestine. Addition of melittin had no significant effect on the α and β diversity of cecal flora, but significantly increased the abundance of Bacteroidales at family level and genus level. Bioinformatics analysis of cecal content showed significant increase in COG functional category of cytoskeleton, and significant decrease in RNA processing and modification in group BA2. KEGG functional analysis showed significant decrease in steroid biosynthesis, caffeine metabolism, and cytochrome P450 pathways in group BA2. In conclusion, addition of 0.12 g/kg melittin to feed improved the laying performance and the intestinal antioxidant capacity and barrier function of quails but had no significant effect on the composition and structure of cecal microbial community. This study provides experimental data and theoretical basis for the application of melittin as a new quail feed additive.

Liquiritin is a natural flavone with a variety of pharmacological effects derived from the medicinal food homology plant Glycyrrhiza uralensis Fisch. As a kind of lethal allergic reactions, pseudo-allergic reactions (PARs) arise from the Mas-related G protein coupled receptor X2 (MRGPRX2)-triggered fast degranulation of mast cells (MCs). In the current work, the anti-pseudo-allergy action and potential mechanisms of liquiritin were explored in vivo and in vitro. Liquiritin suppressed the calcium influx and degranulation elicited by Compound 48/80 (C48/80) in mouse peritoneal mast cells (MPMCs). In mice, liquiritin also inhibited the C48/80-elicited hind paw extravasation, as well as the elevations in TNF-α and histamine levels. Molecular docking combined with detection of HEK293T cells expressing human MRGPRX2 showed that liquiritin was a potential MRGPRX2 antagonist and inhibited PARs through the PI3K/AKT and PLCγ signaling pathways downstream of MRGPRX2. The present work opens a new avenue for the PARs management.

Nitrogen is the most important driving factor in primary production and decomposition in arid and semi-arid ecosystems. The effects of shrub encroachment on nitrogen cycling have been investigated at the site scale but seldomly conducted at the landscape scale. Here, we selected 43 shrubland sites distributing across 3000 km2 area in temperate desert grassland in eastern Yanchi County of Ningxia Hui Autonomous. We investigated the spatial heterogeneity and driving factors of soil total nitrogen (STN) at the landscape scale by using geostatistical analysis and the geographical detector method. Our results showed that the average soil total nitrogen decreased in the order of 0–5 cm (0.21 g kg−1) > 5–15 cm (0.19 g kg−1) > 15–40 cm (0.18 g kg−1). Geostatistical analysis showed that soil total nitrogen exhibited the strong spatial autocorrelation in the 0–5 and 5–15 cm soil layers and the moderate spatial autocorrelation in the 15–40 cm soil layer. Furthermore, the geographic detector method indicated that soil physicochemical properties exhibited the stronger effects than these of topographic and vegetation biomass in determining the spatial distribution of soil total nitrogen. Specifically, soil water content in the 0–20 cm soil layer explained 35% of variation in soil total nitrogen spatial pattern in the 0–5 cm soil layer, while soil organic carbon content in the 15–40 cm soil layer explained 64% and 45% of variation in soil total nitrogen spatial patterns in the 5–15 cm and 15–40 cm soil layers, respectively. It was concluded that soil water content and organic carbon content primarily drove the formation of soil total nitrogen spatial heterogeneity in shrubland at the landscape scale, indicating that anthropogenic shrub encroachment evidently affected soil water content and redistribution in dryland.

With the introduction of polythioester, PMPs demonstrate remarkably increased capture efficiency, leading to the dramatically improved sensitivity of mass spectrometry detection.Results: The analysis method with PMPs treatment has been applied in rapid detection of more than 100 amines in lung adenocarcinoma cell lines, mouse organ tissues, and 103 human serum samples with high-throughput and high-coverage. Statistical analysis shows that arginine biosynthesis differed between lung adenocarcinoma cell lines.Discussion: Phenylalanine, tyrosine and tryptophan biosynthesis differed between tissues. The combination indicators demonstrate a great diagnostic accuracy for distinguishing between health and lung disease subjects as well as differentiating the patients with benign lung disease and lung cancer. With powerful capture ability, low-cost preparation, and convenient separation, the PMPs demonstrate promising application in the intensive study of metabolic pathways and early diagnosis of disease.high-throughput and high-coverage. The Liang guang Small-spotted pig is a well-known Chinese indigenous pig that is valued for its exceptional meat quality. However, the Liang guang Small-spotted pig has a lower semen storage capacity, shorter storage time and worse semen quality compared to Duroc. Pig sperm used for artificial insemination (AI) loses part of vitality and quality when being stored in commercial solutions. Serious vitality losses and short shelf life of the semen are particularly prominent in Liang guang Small-spotted pig. In this study, the metabolites in seminal plasma and spermatozoa of Duroc and Liang guang Small-spotted pigs were identified using UHPLC–Q-TOF/MS technology. The findings indicated forty distinct metabolites concentrating on energy metabolic substrates and antioxidant capacity in Liang guang Small-spotted pig and Duroc seminal plasma, including D-Fructose, succinate, 2-dehydro-3-deoxy-d-gluconate, alanine betaine, citrate, carnitine, acetylcarnitine and so on. Seventeen different metabolites were explored, with a focus on glycerophospholipid metabolism in Liang guang Small-spotted pig and Duroc spermatozoa, primarily including glycerol 3-phosphate, acetylcarnitine, phosphatidylcholine (PC) 16:0/16:0, palmitoyl sphingomyelin, acetylcholine, choline, glycerophosphocholine, betaine, L-carnitine, creatinine and others. This study reveals the metabolite profile of spermatozoa and seminal plasma among different pig breeds and might be valuable for understanding the mechanisms that lead to sperm storage capacity. Metabolites involved in energy metabolism, antioxidant capacity and glycerophospholipid metabolism might be key to the poor sperm storage capacity in Liang guang Small-spotted pig.

Countermeasures in weightlessness are a predominant technique for long-term residency in space. Head-down bed rest (HDBR) has long been used to simulate the physiological effects of spaceflight and test the efficacy of countermeasures. In 2019, the Astronaut Center of China (ACC) has held a 90-d HDBR on the effects of exercises’ intervention. Thirty-six subjects were recruited and randomized into 5 intervention groups: (a) control, (b) aerobic exercises, (c) resistance exercises, (d) concurrent aerobic and lower load resistance exercises, and (e) concurrent aerobic and high-load resistance exercises. Moreover, exercise interventions were performed in supine or 6° head-down tilt. They were provided with plain medical, psychological, and nutritional supports during the experiment. In addition, cardiovascular and pulmonary functions, muscle strength, bone density, and neuro–immune–endocrine functions were assessed over time. Subjects were monitored by clinical evaluations, psychological assessments, and nutritional status evaluations. All subjects spent 138 d in the bed rest study laboratory and received 2 follow-up tests per 3 months in the home recovery period. It was found that exercises in the 4 groups were effective interventions for aerobic capacity compared with control. The resistance exercises or concurrent aerobic and high-load resistance exercises improved isometric maximal voluntary contraction. However, the protection against bone loss still needs further exploration. Appropriate psychosocial and nutrition support techniques are worth to be reinforced.

Using inertial measurement units (IMUs) to estimate lower limb joint kinematics and kinetics can provide valuable information for disease diagnosis and rehabilitation assessment. To estimate gait parameters using IMUs, model-based filtering approaches have been proposed, such as the Kalman filter and complementary filter. However, these methods require special calibration and alignment of IMUs. The development of deep learning algorithms has facilitated the application of IMUs in biomechanics as it does not require particular calibration and alignment procedures of IMUs in use. To estimate hip/knee/ankle joint angles and moments in the sagittal plane, a subject-independent temporal convolutional neural network-bidirectional long short-term memory network (TCN-BiLSTM) model was proposed using three IMUs. A public benchmark dataset containing the most representative locomotive activities in daily life was used to train and evaluate the TCN-BiLSTM model. The mean Pearson correlation coefficient of joint angles and moments estimated by the proposed model reached 0.92 and 0.87, respectively. This indicates that the TCN-BiLSTM model can effectively estimate joint angles and moments in multiple scenarios, demonstrating its potential for application in clinical and daily life scenarios.

Porcine circovirus disease poses a significant threat to the pig farming industry. Globally, four genotypes of porcine circovirus are circulating, with porcine circovirus type 2 and 3 (PCV2 and PCV3) being most strongly associated with clinical manifestations. The recently discovered porcine circovirus type 4 (PCV4) exhibits clinical symptoms resembling porcine dermatitis nephropathy syndrome. This study aimed to assess the prevalence and genetic characteristics of PCVs in Guangdong province, China. A comprehensive analysis was conducted on 193 samples collected from 83 distinct pig farms during the period of 2017–2023. A conventional PCR was employed to investigate the presence of PCV2, PCV3, and PCV4. Among the samples, 56.48%, 8.81%, and 8.81% tested positive for PCV2, PCV3, and PCV2/3 co-infection, respectively. Interestingly, PCV4 was not detected. Whole-genome sequencing was performed on 80 PCV2 isolates and 7 PCV3 isolates. A phylo-genetic analysis revealed that 12 strains belonged to PCV2a, 8 strains belonged to PCV2b, and 60 strains belonged to PCV2d, indicating the prevailing presence of PCV2d in Guangdong province, China. Furthermore, two PCV3 isolates were classified as PCV3a and five strains as PCV3b. Notably, an in-depth analysis of the Cap protein sequence of the PCV2 and PCV3 isolates identified high-frequency mutation sites located in predicted epitope regions. Overall, this study provides valuable insights into the prevalence and evolution of PCV2 and PCV3 during the period of 2017–2023 in Guangdong province, China, thereby contributing to the development of effective prevention and control measures.

Axial bearing performance and mechanism of continuous flight auger (CFA) pile in weathered granitic soils, i.e., a widespread special soil in South China, were investigated by field test in this study. Load–settlement responses of four CFA piles were examined, and evolutions of shaft/base resistances were captured by ultra-weak fiber Bragg gratings (UWFBG) with a reflectivity ≤−40 dB. Performances of CFA piles were compared with those of a slurry displacement (SD) pile at the same site, thirteen pretensioned spun high-strength concrete (PHC) piles in the literature and empirical data in design code. Test results show that the ultimate bearing capacity of the CFA pile is highest among different pile types, and typically is twice that of the SD pile. Again, CFA pile produces the highest shaft resistances at 140 kPa and 153 kPa in two weathered granitic soils, while the base resistance of 3080 kPa is between those of the SD pile and the PHC pile. By field excavation, the superior mechanism of the CFA pile is suggested to avoid the formation of in-between bentonite layers and prevent preferential baseflow along fissures, both of which can weaken the soil–pile interface. Overall, this study provides fundamental data through UWFBG and explanations based on field observations which underpin the need for developing a design code specified for CFA piles in South China.

The effect of salinity on biological nitrogen and denitrifying phosphorus removal was investigated in a Modified University of Cape Town (MUCT) system. Removal rates of COD, NH4+-N, NO3−-N, NO2−-N, phosphorus and the sludge characteristics at salt concentrations (0.0, 3.2, 6.4, 11.2 and 16.0 g L−1) were analyzed. With the salt concentration increasing, all the COD, NH4+-N, TN and TP removal rates exhibited a trend of decline, and exhibited an initial reduction and subsequent increase at every stage of salt concentration. NH4+-N, TN and TP removal rates were 92.7%, 51.5% and 67.2% in 16 g L−1 salt concentration, respectively. And they were outperformed the literature reported and acceptable in practical applications. When the salinity of wastewater changed from 0.0 to 16.0 g L−1, the biomass yield coefficients increased from 0.0794 to 0.126 g VSS/g COD. Increased salinity had a detrimental effect on phosphorus-accumulating organisms (PAOs) and denitrifying PAOs (DPAOs) (especially DPAOs). Therefore, phosphorus removal gradually depended on PAO. The simultaneous nitrification and denitrification (SND) rate and nitrogen removal rate (including nitrification rate, denitrification rate, and total nitrogen removal rate) gradually decreased with the increased salinity.

The gut microbial composition of the Luchuan (LC) piglet, one of China’s native breeds, has rarely been studied, especially when compared to other breeds. This study developed a porcine epidemic diarrhea virus (PEDV) infection model in LC and Largewhite (LW) piglets, and analyzed the patterns and differences of intestinal microbial communities and metabolites in piglets of these two breeds after infection. The diarrhea score, survival time, and distribution of viral antigens in the intestine of piglets infected with PEDV differed among breeds, with the jejunal immunohistochemistry score of LW piglets being significantly higher than that of LC piglets (P < 0.001). The results of 16S rRNA sequencing showed differences in microbial diversity and community composition in the intestine of piglets with different breeds between PEDV infection piglets and the healthy controls. There were differences in the species and number of dominant phyla and dominant genera in the same intestinal segment. The relative abundance of Shigella in the jejunum of LC piglets after PEDV infection was significantly lower than that of LW piglets (P < 0.05). The key microorganisms differed in the microbiota were Streptococcus alactolyticus, Roseburia faecis, Lactobacillus iners, Streptococcus equi, and Lactobacillus mucosae (P < 0.05). The non-targeted metabolite analysis revealed that intestinal metabolites showed great differences among the different breeds related to infection. Spearman correlation analysis was conducted to examine any links between the microbiota and metabolites. The metabolites in the intestine of different breeds related to infection were mainly involved in arginine biosynthesis, synaptic vesicle cycle, nicotinic acid and nicotinamide metabolism and mTOR signaling pathway, with significantly positive or negative correlations (P < 0.05) between the various microorganisms. This study provides a theoretical foundation for investigating the application of core microorganisms in the gut of piglets of different breeds in the digestive tracts of those infected with PEDV, and helps to tackle the antimicrobial resistance problem further.

Avian infectious bronchitis virus (IBV) is a prevalent RNA virus that causes respiratory distress, nephritis, salpingitis, and egg production decline in chickens, resulting in significant economic loss. IBV is composed of complex genotypes and serotypes, which poses a great challenge for disease control. The current study reports 2 IBV outbreaks which were characterized by respiratory symptoms in IBV vaccinated commercial broilers and layers in Guangdong, China, in 2021. Two IBV strains, ZH01 and HH09, were identified via a RT-PCR assay through targeting the N gene and further characterization through full-length spike (S) gene sequence analysis. Phylogenetic analysis of S1 gene revealed that both ZH01 and HH09 belonged to the GI-19 lineage but contained a certain genetic distance from the GI-19 strain. Of note, the ZH01 and HH09 strains share a low homology of 70 and 86%, respectively, with common vaccine strains (H120), resulting in low vaccine protection. Further recombination analysis based on the S1 sequence suggested the newly identified IBV strains emerged through an intragroup recombination events between CK/CH/SCDY2003-2 and I0305/19 from G1-19 lineage. In addition, a number of novel mutations such as T273I, T292A, and S331K were found in the emerging IBV strains. Taken together, this study reports the genetic characteristics of 2 recent IBV outbreaks in southern China and emphasizes the urgent need for enhanced surveillance and development of novel vaccines for the control of IBV.

We estimated the symptomatic, PCR-confirmed secondary attack rate (SAR) for 2,382 close contacts of 476 symptomatic persons with coronavirus disease in Yichang, Hubei Province, China, identified during January 23–February 25, 2020. The SAR among all close contacts was 6.5%; among close contacts who lived with an index case-patient, the SAR was 10.8%; among close-contact spouses of index case-patients, the SAR was 15.9%. The SAR varied by close contact age, from 3.0% for those 60 years of age. Multilevel logistic regression showed that factors significantly associated with increased SAR were living together, being a spouse, and being >60 years of age. Multilevel regression did not support SAR differing significantly by whether the most recent contact occurred before or after the index case-patient’s onset of illness (p = 0.66). The relatively high SAR for coronavirus disease suggests relatively high virus transmissibility.

The increased prevalence of pulmonary methicillin-resistant Staphylococcus aureus (MRSA) infection in patients living with cystic fibrosis (CF) is concerning due to a correlation with reduced life expectancy and lack of available treatment options. RV94 is a next generation lipoglycopeptide designed for pulmonary delivery that preclinically demonstrated high potency against MRSA in planktonic and protected colonies and improved pulmonary clearance relative to same class molecules. Here, RV94 was formulated into a dry powder for inhalation (DPI) to investigate the localized treatment of pulmonary MRSA presented in a potentially more convenient dosage form. RV94 DPI was generated using a spray-drying process with 12.5 wt% trileucine and demonstrated aerosol characteristics (2.0 μm MMAD and 73% FPF) predictive of efficient pulmonary deposition. In vivo PK from a single dose of RV94 DPI delivered by inhalation to rats yielded lung levels (127 μg/g) much greater than the MRSA minimum inhibitory concentration (0.063 μg/mL), low systemic levels (0.1 μg/mL), and a lung t1/2 equal to 3.5 days. In a rat acute pulmonary MRSA model, a single dose of RV94 DPI delivered by inhalation either up to seven days prior to or 24 h after infection resulted in a statistically significant reduction in lung MRSA titer.

Despite many studies on host or viral gene expression, how the cellular proteome responds to internal or external cues during the infection process remains unclear. In this study, we used a Hepatitis B Virus (HBV) replication model and performed proteomic analyses to understand how HBV evades innate immunity as a function of cell cycle progression. Specifically, we performed proteomic analyses of HBV-replicating cells in G1/S and G2/M phases, as a function of IFN-α treatment. We identified that the conserved LSm (Like-Sm1-8) proteins were differentially regulated in HBV replicating cells treated with IFN-α. Specifically, in G2/M phase, IFN-α increased protein level of LSm1, the unique subunit of cytoplasmic LSm1-7 complex involved in mRNA decay. By contrast, IFN-α decreased LSm8, the unique subunit of nuclear LSm2-8 complex, a chaperone of U6 spliceosomal RNA, suggesting the cytoplasmic LSm1-7 complex is antiviral, whereas the nuclear LSm2-8 complex is pro-viral. In HBV replication and infection models, siRNA-mediated knockdown of LSm1 increased all viral RNAs. Conversely, LSm8 knockdown reduced viral RNA levels, dependent on N6-adenosine methylation (m6A) of the epsilon stem-loop at the 5′ end of pre-Core/pregenomic (preC/pg) RNA. Methylated RNA immunoprecipitation (MeRIP) assays demonstrated reduced viral RNA methylation by LSm8 knockdown, dependent on the 5’ m6A modification, suggesting the LSm2-8 complex has a role in mediating this modification. Interestingly, splicing inhibitor Cp028 acting upstream of the LSm2-8 complex suppressed viral RNA levels without reducing the 5’ m6A modification. This observation suggests Cp028 has novel antiviral effects, likely potentiating IFN-α-mediated suppression of HBV biosynthesis.

Spaceflight is associated with enhanced inactivity, resulting in muscular and cardiovascular deconditioning. Although physical exercise is commonly used as a countermeasure, separate applications of running and resistive exercise modalities have never been directly compared during long-term bedrest. This study aimed to compare the effectiveness of two exercise countermeasure programs, running and resistance training, applied separately, for counteracting cardiovascular deconditioning induced by 90-day head-down bedrest (HDBR). Maximal oxygen uptake (V˙O2max), orthostatic tolerance, continuous ECG and blood pressure (BP), body composition, and leg circumferences were measured in the control group (CON: n = 8), running exercise group (RUN: n = 7), and resistive exercise group (RES: n = 7). After HDBR, the decrease in V˙O2max was prevented by RUN countermeasure and limited by RES countermeasure (−26% in CON p < 0.05, −15% in RES p < 0.05, and −4% in RUN ns). Subjects demonstrated surprisingly modest orthostatic tolerance decrease for different groups, including controls. Lean mass loss was limited by RES and RUN protocols (−10% in CON vs. −5% to 6% in RES and RUN). Both countermeasures prevented the loss in thigh circumference (−7% in CON p < 0.05, −2% in RES ns, and −0.6% in RUN ns) and limited loss in calf circumference (−10% in CON vs. −7% in RES vs. −5% in RUN). Day–night variations in systolic BP were preserved during HDBR. Decrease in V˙O2max positively correlated with decrease in thigh (r = 0.54 and p = 0.009) and calf (r = 0.52 and p = 0.012) circumferences. During this 90-day strict HDBR, running exercise successfully preserved V˙O2max, and resistance exercise limited its decline. Both countermeasures limited loss in global lean mass and leg circumferences. The V˙O2max reduction seems to be conditioned more by muscular than by cardiovascular parameters.
