Wildfires occur frequently in various regions of the world, causing serious damage to natural and human resources. Traditional wildfire prevention and management methods are often hampered by monitoring challenges and low efficiency. Digital twin technology, as a highly integrated virtual simulation model, shows great potential in wildfire management and prevention. At the same time, the virtual–reality combination of digital twin technology can provide new solutions for wildfire management. This paper summarizes the key technologies required to establish a wildfire digital twin system, focusing on the technical requirements and research progress in fire detection, simulation, and prediction. This paper also proposes the wildfire digital twin (WFDT) model, which integrates real-time data and computational simulations to replicate and predict wildfire behavior. The synthesis of these techniques within the framework of a digital twin offers a comprehensive approach to wildfire management, providing critical insights for decision-makers to mitigate risks and improve emergency response strategies. Protein-protein interactions (PPI) play a key role in many biological systems. Over the past few years, an explosion in availability of functional biological data obtained from high-throughput technologies to infer PPI has been observed. However, results obtained from such experiments show high rates of false positives and false negatives predictions as well as systematic predictive bias. Recent research has revealed that several machine and statistical learning methods applied to integrate relatively weak, diverse sources of large-scale functional data may provide improved predictive accuracy and coverage of PPI. In this paper we describe the effects of applying different computational, integrative methods to predict PPI in Saccharomyces cerevisiae. We investigated the predictive ability of combining different sets of relatively strong and weak predictive datasets. We analysed several genomic datasets ranging from mRNA co-expression to marginal essentiality. Moreover, we expanded an existing multi-source dataset from S. cerevisiae by constructing a new set of putative interactions extracted from Gene Ontology (GO)- driven annotations in the Saccharomyces Genome Database. Different classification techniques: Simple Naive Bayesian (SNB), Multilayer Perceptron (MLP) and K-Nearest Neighbors (KNN) were evaluated. Relatively simple classification methods (i.e. less computing intensive and mathematically complex), such as SNB, have been proven to be proficient at predicting PPI. SNB produced the “highest” predictive quality obtaining an area under Receiver Operating Characteristic (ROC) curve (AUC) value of 0.99. The lowest AUC value of 0.90 was obtained by the KNN classifier. This assessment also demonstrates the strong predictive power of GO-driven models, which offered predictive performance above 0.90 using the different machine learning and statistical techniques. As the predictive power of single-source datasets became weaker MLP and SNB performed better than KNN. Moreover, predictive performance saturation may be reached independently of the classification models applied, which may be explained by predictive bias and incompleteness of existing “Gold Standards”. More comprehensive and accurate PPI maps will be produced for S. cerevisiae and beyond with the emergence of largescale datasets of better predictive quality and the integration of intelligent classification methods.

Inertial sensors are widely used in human motion monitoring. Orientation and position are the two most widely used measurements for motion monitoring. Tracking with the use of multiple inertial sensors is based on kinematic modelling which achieves a good level of accuracy when biomechanical constraints are applied. More recently, there is growing interest in tracking motion with a single inertial sensor to simplify the measurement system. The dead reckoning method is commonly used for estimating position from inertial sensors. However, significant errors are generated after applying the dead reckoning method because of the presence of sensor offsets and drift. These errors limit the feasibility of monitoring upper limb motion via a single inertial sensing system. In this paper, error correction methods are evaluated to investigate the feasibility of using a single sensor to track the movement of one upper limb segment. These include zero velocity update, wavelet analysis and high-pass filtering. The experiments were carried out using the nine-hole peg test. The results show that zero velocity update is the most effective method to correct the drift from the dead reckoning-based position tracking. If this method is used, then the use of a single inertial sensor to track the movement of a single limb segment is feasible. [ABSTRACT FROM AUTHOR]

Protein-protein interactions (PPIs) play crucial roles in virtually every aspect of cellular function within an organism. One important objective of modern biology is the extraction of functional modules, such as protein complexes from global protein interaction networks. This paper describes how seven genomic features and four experimental interaction data sets were combined using a Bayesian-networks-based data integration approach to infer PPI networks in yeast. Greater coverage and higher accuracy were achieved than in previous high-throughput studies of PPI networks in yeast. A Markov clustering algorithm was then used to extract protein complexes from the inferred protein interaction networks. The quality of the computed complexes was evaluated using the hand-curated complexes from the Munich Information Center for Protein Sequences database and gene-ontology-driven semantic similarity. The results indicated that, by integrating multiple genomic information sources, a better clustering result was obtained in terms of both statistical measures and biological relevance. Human Activity Recognition (HAR) is increasingly used in a variety of applications, including health care, fitness tracking, and rehabilitation. To reduce the impact on the user's daily activities, wearable technologies have been advanced throughout the years. In this study, an improved smart insole-based HAR system is proposed. The impact of data segmentation, sensors used, and feature selection on HAR was fully investigated. The Support Vector Machine (SVM), a supervised learning algorithm, has been used to recognise six ambulation activities: downstairs, sit to stand, sitting, standing, upstairs, and walking. Considering the impact that data segmentation can have on the classification, the sliding window size was optimised, identifying the length of 10 s with 50% of overlap as the best performing. The inertial sensors and pressure sensors embedded into the smart insoles have been assessed to determine the importance that each one has in the classification. A feature selection technique has been applied to reduce the number of features from 272 to 227 to improve the robustness of the proposed system and to investigate the importance of features in the dataset. According to the findings, the inertial sensors are reliable for the recognition of dynamic activities, while pressure sensors are reliable for stationary activities; however, the highest accuracy ( 94.66 % ) was achieved by combining both types of sensors. Abstract Serial analysis of gene expression (SAGE) is one of the most powerful tools for global gene expression profiling. It has led to several biological discoveries and biomedical applications, such as the prediction of new gene functions and the identification of biomarkers in human cancer research. Clustering techniques have become fundamental approaches in these applications. This paper reviews relevant clustering techniques specifically designed for this type of data. It places an emphasis on current limitations and opportunities in this area for supporting biologically-meaningful data mining and visualisation.

Abstract Background Retinal photoreceptors are highly specialised cells, which detect light and are central to mammalian vision. Many retinal diseases occur as a result of inherited dysfunction of the rod and cone photoreceptor cells. Development and maintenance of photoreceptors requires appropriate regulation of the many genes specifically or highly expressed in these cells. Over the last decades, different experimental approaches have been developed to identify photoreceptor enriched genes. Recent progress in RNA analysis technology has generated large amounts of gene expression data relevant to retinal development. This paper assesses a machine learning methodology for supporting the identification of photoreceptor enriched genes based on expression data. Results Based on the analysis of publicly-available gene expression data from the developing mouse retina generated by serial analysis of gene expression (SAGE), this paper presents a predictive methodology comprising several in silico models for detecting key complex features and relationships encoded in the data, which may be useful to distinguish genes in terms of their functional roles. In order to understand temporal patterns of photoreceptor gene expression during retinal development, a two-way cluster analysis was firstly performed. By clustering SAGE libraries, a hierarchical tree reflecting relationships between developmental stages was obtained. By clustering SAGE tags, a more comprehensive expression profile for photoreceptor cells was revealed. To demonstrate the usefulness of machine learning-based models in predicting functional associations from the SAGE data, three supervised classification models were compared. The results indicated that a relatively simple instance-based model (KStar model) performed significantly better than relatively more complex algorithms, e.g. neural networks. To deal with the problem of functional class imbalance occurring in the dataset, two data re-sampling techniques were studied. A random over-sampling method supported the implementation of the most powerful prediction models. The KStar model was also able to achieve higher predictive sensitivities and specificities using random over-sampling techniques. Conclusion The approaches assessed in this paper represent an efficient and relatively inexpensive in silico methodology for supporting large-scale analysis of photoreceptor gene expression by SAGE. They may be applied as complementary methodologies to support functional predictions before implementing more comprehensive, experimental prediction and validation methods. They may also be combined with other large-scale, data-driven methods to facilitate the inference of transcriptional regulatory networks in the developing retina. Furthermore, the methodology assessed may be applied to other data domains.

Systems medicine (SM) has emerged as a powerful tool for studying the human body at the systems level with the aim of improving our understanding, prevention and treatment of complex diseases. Being able to automatically extract relevant features needed for a given task from high-dimensional, heterogeneous data, deep learning (DL) holds great promise in this endeavour. This review paper addresses the main developments of DL algorithms and a set of general topics where DL is decisive, namely, within the SM landscape. It discusses how DL can be applied to SM with an emphasis on the applications to predictive, preventive and precision medicine. Several key challenges have been highlighted including delivering clinical impact and improving interpretability. We used some prototypical examples to highlight the relevance and significance of the adoption of DL in SM, one of them is involving the creation of a model for personalized Parkinson's disease. Systems medicine holds many promises, but has so far provided only a limited number of proofs of principle. To address this road block, possible barriers and challenges of translating systems medicine into clinical practice need to be identified and addressed. The members of the European Cooperation in Science and Technology (COST) Action CA15120 Open Multiscale Systems Medicine (OpenMultiMed) wish to engage the scientific community of systems medicine and multiscale modelling, data science and computing, to provide their feedback in a structured manner. Background Research into the use of digital technology for weight loss maintenance (intentionally losing at least 10% of initial body weight and actively maintaining it) is limited. The aim of this article was to systematically review randomized controlled trials (RCTs) reporting on the use of digital technologies for communicating on weight loss maintenance to determine its' effectiveness, and identify gaps and areas for further research. Methods A systematic literature review was conducted by searching electronic databases to locate publications dated between 2006 and February 2018. Criteria were applied, and RCTs using digital technologies for weight loss maintenance were selected. Results Seven RCTs were selected from a total of 6541 hits after de-duplication and criteria applied. Three trials used text messaging, one used e-mail, one used a web-based system and two compared such a system with face-to-face contact. From the seven RCTs, one included children (n = 141) and reported no difference in BMI Standard Deviation between groups. From the seven trials, four reported that technology is effective for significantly aiding weight loss maintenance compared with control (no contact) or face-to face-contact in the short term (between 3 and 24 months). Conclusions It was concluded that digital technologies have the potential to be effective communication tools for significantly aiding weight loss maintenance, especially in the short term (from 3 to 24 months).
