12. Response of Soil Bacterial Communities and Potato Productivity to Fertilizer Application in Farmlands in the Agropastoral Zone of Northern China.

Abstract

The characteristics and responses of soil bacterial communities and potato productivity to different fertilization treatments in farmlands in the agropastoral zone of Inner Mongolia were investigated. Moreover, the diversity and structure of soil bacterial communities and potato productivity under different fertilization treatments (no fertilization, CK; phosphorus-deficient treatment, NK; conventional fertilization, NPK; and organic–inorganic combination, NPKM) were assessed using Illumina high-throughput sequencing. The results revealed that soil pH, organic matter (SOM), total nitrogen (TN), and total phosphorus (TP) content, and potato productivity were significantly increased under fertilizer treatments (NK, NPK, and NPKM) compared with those under CK, with NPKM treatment having the best enhancement effect. The application of organic fertilizers significantly increased the Shannon, evenness, Chao1, and Ace indices of soil bacterial communities and reshaped the bacterial community structure. Random forest model analysis revealed that soil pH and TP significantly affected soil bacterial diversity, whereas soil pH, SOM, TP, and TN significantly affected soil bacterial community structure. Correlation and structural equation modeling analyses revealed that soil TP and SOM indirectly affected potato productivity by changing soil bacterial diversity and community composition. The results of this study provide a scientific basis for improving the quality and productivity of farmland soil to guide the rational fertilization of farmlands in the agropastoral zone of northern China. Abstract

The distribution characteristics of machining errors in high energy efficiency milling are an important index to evaluate the surface geometry parameters, cutting stability, and dynamic cutting efficiency. The existing methods for machining errors focus on the overall level of the parameters of a machined surface and the degree of deviation from the index and ignore the influence of the instantaneous cutting behavior of a milling cutter and its teeth on the dynamic formation process of a machined surface. The dynamic distribution of machining errors in high energy efficiency milling needs to be revealed. According to the dynamic characteristics of the machined surface formation process with the effects of the cutter tooth error and milling vibration, a method for solving relative position vector of each point on the machined surface was proposed, and the calculation model of the dynamic distribution of the machining errors was constructed to unveil its formation mechanism in high energy efficiency milling. Using the time-frequency analysis method of the milling vibration and machining errors, the dynamic distribution of the machining errors on the machined surface was characterized, and the variety of the geometric error variation of the machined surface was described. The effects of the milling cutter design pose, cutting parameters, cutter tooth error, and milling vibration on the dynamic distribution of the machining errors were revealed, with the proposed identification method of its influencing factors. The response of the dynamic distribution of machining errors was studied, and a method for its identification was proposed and verified with experiments. The results showed that there was a high similarity between the calculated and measured results of the dynamic distribution of the machining errors. The influence mechanism of the key process variables on the dynamic distribution of the machining errors could be identified using the above model and method.

25. Effects of long-term application of organic manure and chemical fertilizer on soil properties and microbial communities in the agro-pastoral ecotone of North China

Abstract

Long-term irrational fertilizer inputs affect soil nutrients conditions in the agro-pastoral ecotone of North China. However, the mechanisms by which biotic and abiotic factors are affected by different fertilizer types remain unclear. A 16-year, long-term fertilization experiment was conducted to explore how soil physicochemical properties and microbial communities respond to different fertilizer types at an experimental site in North China. The key environmental factors that drove changes in soil microbial communities were also determined. In September 2019, soils were collected from plots of four fertilizer treatments: 1) non-fertilization control (CK), 2) chemical fertilization only (CF), 3) organic manure fertilization only (M), and 4) chemical fertilization plus organic manure (CFM). Compared with CK, soil organic matter, total nitrogen, available nitrogen, available phosphorus, and available potassium contents were higher in M and CFM, whereas soil pH was significantly lower in CF. Abundances of dominant soil bacterial phyla Proteobacteria, Bacteroidetes, and Gemmatimonadetes were higher in M and CFM than CK. Abundances of dominant soil fungal phyla Ascomycota was lower in CFM than in other treatments. The pathogenic fungi Fusarium, Paramyrothecium, Cladosporium, and Alternaria had the highest abundances in CK and CF, whereas abundances of the beneficial fungi Mortierella were significantly higher in M and CFM than in CF and CK. According to partial least squares path modeling, differences in fertilizer types had direct positive effects on fungal communities but little effect on bacterial communities. Overall, CFM maintained higher soil fertility and a healthy ecosystem because it increased beneficial microorganisms and inhibited pathogenic microorganisms, whereas CF increased the risk of crop infection with soil-borne diseases. The study provided a better understanding of how long-term fertilization affects microbial community composition and their associated ecosystem functions.

Abstract

Common vetch (Vicia sativa L.) is a leguminous plant used as forage and cover crops. A protocol of ethyl methanesulfonate (EMS)‐induced mutations was established in the present study, based on analysis of seed germination and emergence rate, seedling growth and abnormal plant percentage as affected by combinations of EMS concentration and treatment time. Treatment of seeds with 0.1% EMS for 9 and 12 hr or 0.5% EMS for 6 and 9 hr led to 43%–66% normal plants, which was suggested to be used for EMS‐induced mutations in common vetch. The wild‐type common vetch showed red with accumulation of anthocyanins in winter, and two lines showing separation of anthocyanin accumulation (labeled as A+) and without anthocyanin accumulation (labeled as A–) plants were selected from M2 plants induced by EMS in the field. The A– plants showing green color had higher plant height and fresh shoot weight than the A+ plants during wintering, with lower ion leakage after freezing treatment, indicating that the green mutants (A–) had increased cold tolerance. Compared to A+ plants, the green mutants had significantly increased ascorbate (AsA) level in response to low temperature treatment. The results suggest that the increased AsA level under low temperature was associated with the increased cold tolerance in the green mutants. [ABSTRACT FROM AUTHOR]

47. Exosomes-based dual drug-loaded nanocarrier for targeted and multiple proliferative vitreoretinopathy therapy

Abstract

Proliferative vitreoretinopathy (PVR) is a common cause of vision loss after retinal reattachment surgery and ocular trauma. The key pathogenic mechanisms of PVR development include the proliferation, migration and epithelial-mesenchymal transition (EMT) of retinal pigment epithelial cells (RPEs) activated by the growth factors and cytokines after surgery. Although some drugs have been tried in PVR treatments as basic investigations, the limited efficacy remains an obstacle, which may be due to the single pharmacological action and lack of targeting. Herein, the anti-proliferative Daunorubicin and anti-inflammatory Dexamethasone were co-loaded in the RPEs-derived exosomes (Exos), obtaining an Exos-based dual drug-loaded nanocarrier (Exos@D-D), and used for multiple PVR therapy. Owing to the advantages of homologous Exos and the dual drug loading, Exos@D-D showed good RPEs targeting as well as improved uptake efficiency, and could inhibit the proliferation, migration, as well as EMT of RPEs effectively. The animal studies have also demonstrated that Exos@D-D effectively inhibits the production of proliferative membranes and prevents the further development of inflammation, shows significant therapeutic effects on PVR and good biocompatibility. Such Exos-based dual drug-loaded nanocarrier investigation not only provides a promising approach for multifunctional exosome drug delivery systems construction, but also has great potential in PVR clinical therapy application.
