Abstract

In recent years, quinoid redox mediators (QRMs) have attracted increasing attention because of their key role in wastewater biotreatment. Previous studies have shown that the anaerobic respiration of many bacteria could be coupled to the reduction and reoxidation of quinone groups. Thus, QRMs are widely involved in the microbial transformation of various organic and inorganic substances. To date, few reviews have focused on the involvement of quinoid compounds in environmental pollution biotreatment processes. In this paper, we review the different types of QRMs that interact closely with microorganisms, the characteristics of those QRMs, the involvement of QRMs during the biotransformation of recalcitrant organic pollutants, heavy metal ions and metallic oxides, and their enhancement on microbial fuel cells. Finally, the future research focus and application prospects with regard to different types of QRMs are proposed. This study can improve our understanding of QRM-mediated environmental pollution biotreatment processes and provide fundamental guidance on what kinds of QRMs are practical for engineering applications. 31. Picosecond dissipative soliton generation from an ytterbium-doped fiber laser based on a BP/SnSe2-PVA mixture saturable absorber.

Abstract

Stable picosecond dissipative soliton pulses were observed in an ytterbium-doped fiber laser employing a high-quality mixture of BP/SnSe2-PVA saturable absorber (SA). The modulation depth, saturation intensity, and non-saturable loss of the mixture of BP/SnSe2-PVA SA were measured with values of 5.98%, 18.37 MW/cm2, and 33%, respectively. Within the pump power range of 150–270 mW, stable dissipative soliton pulses were obtained with an output power of 1.68–4 mW. When the minimum pulse duration is 1.28 ps, a repetition rate of 0.903 MHz, center wavelength of 1064.38 nm and 3 dB bandwidth of 2 nm were obtained. The maximum pulse energy of 4.43 nJ and the signal-to-noise ratio up to 72 dB were achieved at pump power of 270 mW. The results suggest that the BP/SnSe2-PVA mixture SA has outstanding nonlinear saturable absorption characteristics and broad ultrafast laser applications. [ABSTRACT FROM AUTHOR]

Abstract

Background A wide variety of bacterial adaptative responses to environmental conditions are mediated by signal transduction pathways. Two-component signal transduction systems are one of the predominant means used by bacteria to sense the signals of the host plant and adjust their interaction behaviour. A total of seven open reading frames have been identified as putative two-component response regulators in the gram-negative nitrogen-fixing bacteria Azorhizobium caulinodans ORS571. However, the biological functions of these response regulators in the symbiotic interactions between A. caulinodans ORS571 and the host plant Sesbania rostrata have not been elucidated to date. Results In this study, we identified and investigated a two-component response regulator, AcfR, with a phosphorylatable N-terminal REC (receiver) domain and a C-terminal HTH (helix-turn-helix) LuxR DNA-binding domain in A. caulinodans ORS571. Phylogenetic analysis showed that AcfR possessed close evolutionary relationships with NarL/FixJ family regulators. In addition, six histidine kinases containing HATPase_c and HisKA domains were predicted to interact with AcfR. Furthermore, the biological function of AcfR in free-living and symbiotic conditions was elucidated by comparing the wild-type strain and the ΔacfR mutant strain. In the free-living state, the cell motility behaviour and exopolysaccharide production of the ΔacfR mutant were significantly reduced compared to those of the wild-type strain. In the symbiotic state, the ΔacfR mutant showed a competitive nodule defect on the stems and roots of the host plant, suggesting that AcfR can provide A. caulinodans with an effective competitive ability for symbiotic nodulation. Conclusions Our results showed that AcfR, as a response regulator, regulates numerous phenotypes of A. caulinodans under the free-living conditions and in symbiosis with the host plant. The results of this study help to elucidate the involvement of a REC + HTH_LuxR two-component response regulator in the Rhizobium-host plant interaction.

37. A General Carboxylate-Assisted Approach to Boost the ORR Performance of ZIF-Derived Fe/N/C Catalysts for Proton Exchange Membrane Fuel Cells

Abstract

An Fe/N/C catalyst derived from the pyrolysis of metal-organic frameworks, for example, a zeolitic-imidazolate-framework-8 (ZIF-8), has been regarded as one of the most promising non-precious metal catalysts toward oxygen reduction reaction (ORR) in proton exchange membrane fuel cells (PEMFCs). However, its ORR mass activity is still much inferior to that of Pt, partly because of the lack of general and efficient synthetic strategies. Herein, a general carboxylate-assisted strategy that dramatically enhances the ORR mass activity of ZIF-derived Fe/N/C catalysts is reported. The carboxylate is found to promote the formation of Fe/N/C catalysts with denser accessible active sites and entangled carbon nanotubes, as well as a higher mesoporosity. These structural advantages make the carboxylate-assisted Fe/N/C catalysts show a 2-10 fold higher ORR mass activity than the common carboxylate-free one in various cases. When applied in H-2-O-2 PEMFCs, the active acetate-assisted Fe/N/C catalyst generates a peak power density of 1.33 W cm(-2), a new record of peak power density for a H-2-O-2 PEMFC with non-Pt ORR catalysts.

Abstract

RGS (regulator of G protein signaling) proteins containing the G protein [gamma]-like (GGL) domain (RGS6, RGS7, RGS9, and RGS11) interact with the fifth member of the G protein [beta]-subunit family, G[beta]5. This interaction is necessary for the stability of both the RGS protein and for G[beta]5. Consistent with this notion, we have found that elevation of RGS9-1 mRNA levels by transgene expression does not increase RGS9-1 protein level in the retina, suggesting that G[beta]5 levels may be limiting. To examine further the interactions of G[beta]5 and the GGL domain-containing RGS proteins, we inactivated the G[beta]5 gene. We found that the levels of GGL domain-containing RGS proteins in retinas and in striatum are eliminated or reduced drastically, whereas the levels of [G.sub.[gamma]2] and RGS4 proteins remain normal in the absence of G[beta]5. The homozygous G[beta]5 knockout (G[beta][5.sup.-/-]) mice derived from heterozygous knockout mating are runty and exhibit a high preweaning mortality rate. We concluded that complex formation between GGL domain-containing RGS proteins and the G[beta]5 protein is necessary to maintain their mutual stability in vivo. Furthermore, in the absence of G[beta]5 and all four RGS proteins that form protein complexes with G[beta]5, the animals that survive into adulthood are viable and have no gross defects in brain or retinal morphology.

41. Seagrass vegetation affect the vertical organization of microbial communities in sediment

Abstract

Seagrasses represent high primary productivity and provide important ecosystem services to the marine environment. Seagrass-associated microbial communities are playing essential ecological functional roles in biogeochemical cycles. However, little is known about the effect of seagrass vegetation on microbial communities in sediment. In the present study, the sediment cores of seagrass bed (dominated by Zostera japonica and Zostera marine) and degradation area in Swan Lake (China) were sampled; then, biogeochemical parameters were analyzed, and microbial community composition was investigated by using high-throughput sequencing of the 16S rRNA gene. The results showed that the presence of seagrass could lead to a decrease in the richness and diversity of the microbial community. In the vertical direction, a pronounced shift from Proteobacteria-dominated upper layers to Chloroflexi and Crenarchaeota-dominated deep layers in all sediment cores were observed. Besides, Bathyarchaeia is more abundant at degradation area, while Vibrionaceae, Sulfurovum and Lokiarchaeial overrepresent at the seagrass bed area. Vibrionaceae was abundant in the rhizosphere of Z. marina and Z. japonica, and the proportions reached 84.45% and 63.89%, respectively. This enrichment of Vibrio spp. may be caused by the macrobenthic species near the seagrass rhizosphere, and these Vibrio spp. reduced the diversity and stability of microbial community, which may lead to the degradation of seagrass. This study would provide clues for the distribution patterns and niche preferences of seagrass microbiome. The conservation strategy of seagrass would be further elucidated from the perspective of the microbiome.

Abstract

Arsenic exposure poses numerous threats to human health. Our previous work in mice has shown that arsenic causes anemia by inhibiting erythropoiesis. However, the impacts of arsenic exposure on human erythropoiesis remain largely unclear. We report here that low-dose arsenic exposure inhibits the erythroid differentiation of human hematopoietic progenitor cells (HPCs). The impacts of arsenic (in the form of arsenite; As3+) on red blood cell (RBC) development was evaluated using a long-term culture of normal human bone marrow CD34+-HPCs stimulated in vitro to undergo erythropoiesis. Over the time course studied, we analyzed the expression of the cell surface antigens CD34, CD71 and CD235a, which are markers commonly used to monitor the progression of HPCs through the stages of erythropoiesis. Simultaneously, we measured hemoglobin content, which is an important criterion used clinically for diagnosing anemia. As compared to control, low-dose As3+ exposure (100 nM and 500 nM) inhibited the expansion of CD34+-HPCs over the time course investigated; decreased the number of committed erythroid progenitors (BFU-E and CFU-E) and erythroblast differentiation in the subsequent stages; and caused a reduction of hemoglobin content. These findings demonstrate that low-dose arsenic exposure impairs human erythropoiesis, likely by combined effects on various stages of RBC formation. [ABSTRACT FROM AUTHOR]
