TP53, functioning as the keeper of the genome, assumes a pivotal function in the inhibition of tumorigenesis. Recent studies have revealed that p53 regulates ferroptosis pathways within tumor cells and is closely related to tumorigenesis. Therefore, we summarize the pathways and mechanisms by which p53 regulates ferroptosis and identify a series of upstream and downstream molecules involved in this process. Furthermore, we construct a p53-ferroptosis network centered on p53. Finally, we present the progress of drugs to prevent wild-type p53 (wtp53) degeneration and restore wtp53, highlighting the deficiencies of drug development and the prospects for p53 in cancer treatment. These findings provide novel strategies and directions for future cancer therapy.

Radiotherapy (RT) has been widely used in the clinical treatment of cancers, but radiotherapy resistance (RR) leads to RT failure, tumor recurrence and metastasis. Many studies have been performed on the potential mechanisms behind RR, and a strong link has been found between RR and DNA damage. RT-induced DNA damage triggers a protective mechanism called the DNA damage response (DDR). DDR consists of several aspects, including the detection of DNA damage and induction of cell cycle checkpoint, DNA repair, and eventual induction of cell death. A large number of studies have shown that DDR inhibition leads to significantly enhanced sensitivity of cancer cells to RT. DDR may be an effective target for radio- and chemo-sensitization during cancer treatment. Therefore, many inhibitors of important enzymes involved in the DDR have been developed, such as PARP inhibitors, DNA-PK inhibitors, and ATM/ATR inhibitors. In addition, DNA damage also triggers the cGAS-STING signaling pathway and the ATM/ATR (CHK)/STAT pathway to induce immune infiltration and T-cell activation. This review discusses the effects of DDR pathway dysregulation on the tumor response to RT and the strategies for targeting these pathways to increase tumor susceptibility to RT. Finally, the potential for the combination treatment of radiation, DDR inhibition, and immunotherapy is described.

Positioning technology is the basis for the application of Internet of Vehicles (IoV). The positioning accuracy is generally very low in infrastructure limited scenarios based on satellites or base stations. In this paper, a vehicle positioning algorithm is proposed that combines the information of vehicle historical trajectory and inertial navigation technology. The simulation results show that the proposed algorithm can significantly improve the positioning accuracy in infrastructure limited scenarios.

Wake-up radio is a promising approach to mitigate the problem of idle listening, which incurs additional power consumption for the Internet of Things (IoT) wireless transmission. Radio frequency (RF) energy harvesting technique allows the wake-up radio to remain in a deep sleep and only become active after receiving an external RF signal to ‘wake-up’ the radio, thus eliminating necessary hardware and signal processing to perform idle listening, resulting in higher energy efficiency. This review paper focuses on cross-layer; physical and media access control (PHY and MAC) approaches on passive wake-up radio based on the previous works from the literature. First, an explanation of the circuit design and system architecture of the passive wake-up radios is presented. Afterward, the previous works on RF energy harvesting techniques and the existing passive wake-up radio hardware architectures available in the literature are surveyed and classified. An evaluation of the various MAC protocols utilized for the novel passive wake-up radio technologies is presented. Finally, the paper highlights the potential research opportunities and practical challenges related to the practical implementation of wake-up technology for future IoT applications.

The spectrum allocation for cognitive radio networks (CRNs) has received considerable studies under the assumption that the bandwidth of spectrum holes is static. However, in practice, the bandwidth of spectrum holes is time-varied due to primary user/secondary user (PU/SU) activity and mobility, which result in non-determinacy. This paper studies the spectrum allocation for CRNs with non-deterministic bandwidth of spectrum holes. We present a novel probability density function (PDF) model through order statistic to describe the non-deterministic bandwidth of spectrum holes and provide a bound to approximate it. After that, a statistical spectrum allocation model based on stochastic multiple knapsack problem (MKP) is established for spectrum allocation with non-deterministic bandwidth of spectrum holes. To reduce the computational complexity, we transform this stochastic programming probleminto a constant MKP though exploiting the properties of cumulative distribution function (CDF), which can be solved via MTHG algorithm by using auxiliary variable. Simulation results illustrate that the proposed statistical spectrum allocation algorithm can achieve better performances compared to the existing algorithms when the bandwidth of spectrum holes istime-varied.
