Protoplast fusion technology originated from cell-cell fusion technology in the 1950s and was initially applied to animal cells. The advantage of applying it in the field of edible fungi breeding is that compared with conventional natural selection and hybrid breeding methods, it could more effectively overcome certain incompatibility obstacles to achieve distant hybridization, and more purposefully select parents to obtain new varieties with outstanding characteristics. For these reasons protoplast fusion technology is becoming of great significance for the improvement of edible fungi strains, the breeding of new varieties, and the taming of difficult-to-cultivate species. At present, relatively stable and capable-of-fruiting fusants are limited to several genus and species, and using protoplast fusion for distant hybridization requires new breakthroughs overcoming vegetative incompatibilities. Technically, the acquisition and identification of fusants could also be improved to a more controllable approach. This article focuses on the longitudinal discussion of the role of protoplast fusion technology in the genetic breeding of edible fungi. It explains: i) the preparation and regeneration of protoplasts in the process, ii) the specific methods of fusion, iii) the identification, mutagenesis, and rejuvenation of fusants, and iv) possible impacts of two incompatible systems. Certain application practice cases are also introduced to summarize its promotion prospects.

Abstract Background Plastic is one of the most widely used materials worldwide in various fields, including packaging and agriculture. Its large quantities require proper disposal and for this reason more and more attention is paid to the issue of degrading plastic. Thanks to the production of non-specific enzymes, fungi are able to attack complex and recalcitrant xenobiotics such as plastics. In recent years, several spectroscopic methods were used to study the plastic degradation ability of different fungal species. Among these, Fourier transform infrared (FT-IR) and FT-Raman spectroscopy techniques are the most used. Surface-enhanced Raman scattering (SERS) spectroscopy is a powerful technique which uses metal nanoparticles (NPs) to enhance the Raman signal of molecules adsorbed on the NPs surface. In this work, the isolation of different fungi from field-collected plastic debris and the ability of these isolates to growth and colonizing the low-density polyethylene (LDPE) were explored by using scanning electron microscope (SEM), attenuated total reflectance-Fourier transform infrared (ATR-FTIR) and SERS spectroscopies. Results Forty-seven fungal isolates belonging to 10 genera were obtained; among them only 11 were able to grow and colonize the LDPE film. However, after 90 days trial, only one isolate of Cladosporium cladosporioides (Clc/1) was able to carry out the initial degradation of the LDPE film. In particular, based on SEM observations, small cavities and depressed areas of circular shape were visible in the treated samples. Additionally, ATR-FTIR, normal Raman and SERS analyses supported the structural changes observed via SEM. Notably, ATR-FTIR and normal Raman spectra showed a significant decrease in the relative intensity of the methylene group bands. Similarly, the SERS spectra of LDPE after the fungal attack, confirmed the decrease of methylene groups bands and the appearance of other bands referring to LDPE polyphenolic admixtures. Conclusions These results suggest that Cladosporium cladosporioides Clc/1 is able to carry out an initial degradation of LDPE. Moreover, combining ATR-FTIR, Raman and SERS spectroscopies with SEM observations, the early stages of LDPE degradation can be explored without any sample pretreatment. Graphical Abstract

A recurrent task in coordinated systems is managing (estimating, predicting, or controlling) signals that vary in space, such as distributed sensed data or computation outcomes. Especially in large-scale settings, the problem can be addressed through decentralised and situated computing systems: nodes can locally sense, process, and act upon signals, and coordinate with neighbours to implement collective strategies. Accordingly, in this work we devise distributed coordination strategies for the estimation of a spatial phenomenon through collaborative adaptive sampling. Our design is based on the idea of dynamically partitioning space into regions that compete and grow/shrink to provide accurate aggregate sampling. Such regions hence define a sort of virtualised space that is "fluid", since its structure adapts in response to pressure forces exerted by the underlying phenomenon. We provide an adaptive sampling algorithm in the field-based coordination framework, and prove it is self-stabilising and locally optimal. Finally, we verify by simulation that the proposed algorithm effectively carries out a spatially adaptive sampling while maintaining a tuneable trade-off between accuracy and efficiency.

The ascomycetes in the genus Morchella (commonly called morels) have a considerable economic and biotechnological value for their culinary and medicinal proprieties. However, their biotechnological interest is not limited to the fruiting body cultivation, but also in mycelial production to obtain bioactive compounds and other biotech products. In order to better exploit the biotechnological potentialities of morels it is necessary to improve the knowledge on their biology and mycelial characteristics. In this paper morphological and growth characteristics of mycelia, as well as biomass formation of Italian collections of five edible medicinal Morchella species [M. esculenta (L.) Pers., M. dunalii Boud., M. importuna M. Kuo, O'Donnell & T.J. Volk, M. disparilis Loizides & P.-A. Moreau and M. purpurascens (Krombh. ex Boud.) Jacquet.] are presented.

Pedodiversity is generally neglected in studies concerning soil organic carbon (SOC). Therefore, this investigation aimed to explore the effect of soil types on the following: (1) soil processes related to organic matter (OM) dynamics along the profile; and (2) the microbial community and functionality within the uppermost horizon. Humic Dystrudepts (HD), Typic Dystrudepts (TD), and Humic Lithic Dystrudepts (HLD) were selected in beech forests of the Apennine ridge in the Emilia-Romagna Region (Italy). Soils were sampled by horizons until parent material, and physico-chemical and functional analyses were performed. The results showed that both HD and HLD soils had a higher SOC accumulation than TD, particularly within the deeper horizons. Such accumulation might be due to the lower turnover rate of soil OM forms, namely fulvic acid-like substances, humic acid-like substances, and non-extractable OM. Noteworthy, the A horizons showed slight differences in SOC among the soil types, suggesting similar SOC decomposition processes. This fact was confirmed by the lack of differences in microbial DNA-based diversity and functionality. This study highlighted the importance of combining pedodiversity and microbial diversity for a wider perspective on SOC dynamics.

Tuber magnatum Picco is the most expensive of the truffles and a great deal of research has been carried out in an attempt to solve the mysteries of its ecology and biology. However, considerable work remains to be done particularly on those secrets of its life cycle that remain a mystery. It is known that T. magnatum is heterothallic, but it has yet to be determined how fertilization occurs between the two strains of different mating types. It is also known that the white truffle is an ectomycorrhizal fungus, and its mycorrhizas can be produced in greenhouses, but then they seem to disappear in the field. The role of other soil microorganisms, fungi and bacteria, on its soil mycelial development and fructification is intriguing but is far from being completely understood. All these uncertainties have made the cultivation of T. magnatum extremely difficult and only recently have we had the scientific proofs that it is possible. Even so, many questions remain unanswered and the management practices of T. magnatum plantations are still to be better defined to also enable the taming of this truffle.

Abstract Background Mushrooms have always been considered an important source of food and biologically active compounds with several medicinal properties. In recent years, different methods were used to study the quality and chemical composition of mushrooms. Among these, Fourier transform infrared (FT-IR) and FT-Raman spectroscopy techniques have been successfully applied to identify different mushroom species. However, the structural biomolecule components existing in the mycelium or in the fruiting bodies may produce strong fluorescence emission that overlaps the Raman radiation, thus avoiding their analyses by Raman. SERS spectroscopy is a powerful technique which uses metal nanoparticles (NPs) to enhance the Raman signal of molecules adsorbed on the NPs surface. In addition, SERS is able to quench the macromolecule florescence. In this work, we have employed silver nanoparticles in order to get mushroom fingerprints based on SERS as quick procedure to analyze and identify different chemical compounds from the fruiting bodies of six edible and/or medicinal mushrooms: Lentinula edodes, Ganoderma lucidum, Pleurotus cornucopiae, Pleurotus ostreatus, Tuber aestivum and Tuber magnatum. Results SERS analyses performed directly on fruiting body fragments produced characteristic spectra for each species. One group of mushrooms (L. edodes, G. lucidum, T. aestivum and T. magnatum) was dominated by the bands of nucleic acids; and the other one (P. cornucopiae and P. ostreatus), by the bands of pigments such as melanins; carotenoids; azafilones; polyketides; and flavonoids located in the cell wall. Additionally, bands corresponding to cell wall polysaccharides, particularly chitosan and 1,3-β D-glucan, were identified in the extracts of P. cornucopiae, P. ostreatus and L. edodes. No signal of cell wall polysaccharides was found in G. lucidum extract. Raman mapping of the analyzed samples was useful in tracking the spatial distribution of the marker bands. Moreover, the principal component analysis (PCA) carried out on the acquired SERS spectra, allows to discriminate the analyzed mushroom species. Conclusions The SERS technique has the ability to generate a strong Raman signal from mushroom fruiting bodies using Ag-NPs deposited directly on intact, untreated mushroom tissues. Using this methodology, commonly applied laboratory time-consuming methods can be avoided or bypassed as well as analysis time can be reduced. Graphical Abstract

Tuber borchii is a precious truffle, which forms ectomycorrhizal associations with several coniferous and broadleaf trees, arbutoid mycorrhizas with the strawberry tree and recently it was found also to form orchid mycorrhizas with Epipactis atrorubens. In this work, we investigated the formation of T. borchii mycorrhizas in other orchid species (Anacamptis morio, Cephalanthera longifolia, Limodorum abortivum, Ophrys sphegodes, Orchis purpurea). To this aim the orchids growing in three T. borchii production areas located in Emilia Romagna region (Italy) were collected and the presence of T. borchii mycorrhizas was investigated by morphological and molecular approaches. For T. borchii molecular identification, specific primers were used by a nested PCR approach. Tuber borchii was identified in 2 out of 6 samples of A. morio, 3 out of 4 samples of C. longifolia, and 2 out 6 samples of O. sphegodes. The ecological and biological roles of the orchid association in the truffle life cycle are discussed.

A genome-wide association study (GWAS) was performed as a preliminary step to identify regions potentially related to ham quality traits. In this research, genomic information was obtained from 238 commercial hybrid pigs utilising the GeneSeek® Genomic Profiler genome-wide porcine genotyping array. Carcasses were tested for hot weight, the thickness of backfat and loin, and lean meat percentage. The corresponding fresh hams were assayed for weight and ultimate pH; the activities of Cathepsin B and Ferrochelatase of Semimembranosus muscle were determined through fluorimetric methods. The lean meat percentage of fresh ham (LMPH), salt absorbed after first (SALT1) and overall salting stages (SALT) were estimated online by the Ham Inspector™ apparatus. Hams were processed in compliance with the procedures established for Protected Designation of Origin Parma ham, and ham weight losses were measured at the main processing stages. Hot carcass weights showed a significant negative correlation with their lean meat percentage and LMPH, while LMPH was correlated positively with carcass lean meat, SALT1, SALT, and weight losses. The GWAS detected genome-wide association for 12 single nucleotide polymorphisms with Ferrochelatase activity. The results obtained in this preliminary study were achieved by combining innovative and non-destructive technologies for screening hams under processing, measures of enzymatic muscle properties relevant to dry-cured ham quality, and genomic information obtained through a GWAS. Additional studies carried out in a larger number of pigs have been planned to investigate the effect of gene variants of Ferrochelatase activity in the dry-cured ham’s quality with main reference to colour development and to confirm the GWAS results obtained in this study.

Tuber melanosporum is an ascomycete that forms ectomycorrhizal (ECM) symbioses with a wide range of host plants, producing edible fruiting bodies with high economic value. The quality of seedlings in the early symbiotic stage is important for successful truffle cultivation. Numerous bacterial species have been reported to take part in the truffle biological cycle and influence the establishment of roots symbiosis in plant hosts and the development of the carpophore. In this work, three different bacteria formulations were co-inoculated in Quercus ilex L. seedlings two months after T. melanosporum inoculation. At four months of bacterial application, the T. melanosporum ECM root tip rate of colonization and bacterial presence were assessed using both morphological and molecular techniques. A 2.5-fold increase in ECM colonization rate was found in the presence of Pseudomonas sp. compared to the seedlings inoculated only with T. melanosporum. The same treatment caused reduced plant growth either for the aerial and root part. Meanwhile, the ECM colonization combined with Bradyrhizobium sp. and Pseudomonas sp. + Bradyrhizobium sp. reduced the relative density of fibrous roots (nutrient absorption). Our work suggests that the role of bacteria in the early symbiotic stages of ECM colonization involves both the mycorrhizal symbiosis rate and plant root development processes, both essential for improve the quality of truffle-inoculated seedlings produced in commercial nurseries.

Tuberaceae is one of the most diverse lineages of symbiotic truffle-forming fungi. To understand the molecular underpinning of the ectomycorrhizal truffle lifestyle, we compared the genomes of Piedmont white truffle (Tuber magnatum), Périgord black truffle (Tuber melanosporum), Burgundy truffle (Tuber aestivum), pig truffle (Choiromyces venosus) and desert truffle (Terfezia boudieri) to saprotrophic Pezizomycetes. Reconstructed gene duplication/loss histories along a time-calibrated phylogeny of Ascomycetes revealed that Tuberaceae-specific traits may be related to a higher gene diversification rate. Genomic features in Tuber species appear to be very similar, with high transposon content, few genes coding lignocellulose-degrading enzymes, a substantial set of lineage-specific fruiting-body-upregulated genes and high expression of genes involved in volatile organic compound metabolism. Developmental and metabolic pathways expressed in ectomycorrhizae and fruiting bodies of T. magnatum and T. melanosporum are unexpectedly very similar, owing to the fact that they diverged ~100 Ma. Volatile organic compounds from pungent truffle odours are not the products of Tuber-specific gene innovations, but rely on the differential expression of an existing gene repertoire. These genomic resources will help to address fundamental questions in the evolution of the truffle lifestyle and the ecology of fungi that have been praised as food delicacies for centuries.

Donkeys (Equus asinus) have been used extensively in agriculture and transportations since their domestication, ca. 5000–7000 years ago, but the increased mechanization of the last century has largely spoiled their role as burden animals, particularly in developed countries. Consequently, donkey breeds and population sizes have been declining for decades, and the diversity contributed by autochthonous gene pools has been eroded. Here, we examined coding-region data extracted from 164 complete mitogenomes and 1392 donkey mitochondrial DNA (mtDNA) control-region sequences to (i) assess worldwide diversity, (ii) evaluate geographical patterns of variation, and (iii) provide a new nomenclature of mtDNA haplogroups. The topology of the Maximum Parsimony tree confirmed the two previously identified major clades, i.e. Clades 1 and 2, but also highlighted the occurrence of a deep-diverging lineage within Clade 2 that left a marginal trace in modern donkeys. Thanks to the identification of stable and highly diagnostic coding-region mutational motifs, the two lineages were renamed as haplogroup A and haplogroup B, respectively, to harmonize clade nomenclature with the standard currently adopted for other livestock species. Control-region diversity and population expansion metrics varied considerably between geographical areas but confirmed North-eastern Africa as the likely domestication center. The patterns of geographical distribution of variation analyzed through phylogenetic networks and AMOVA confirmed the co-occurrence of both haplogroups in all sampled populations, while differences at the regional level point to the joint effects of demography, past human migrations and trade following the spread of donkeys out of the domestication center. Despite the strong decline that donkey populations have undergone for decades in many areas of the world, the sizeable mtDNA variability we scored, and the possible identification of a new early radiating lineage further stress the need for an extensive and large-scale characterization of donkey nuclear genome diversity to identify hotspots of variation and aid the conservation of local breeds worldwide. Management of competencies is a crucial concern for both learners and workers as well as for training institutions and companies. For the former, it allows users to track and certify the acquired skills to apply for positions; for the latter, it enables better organisation of business processes. However, currently, most software systems for competency management adopted by the industry are either organisation-centric or centralised: that is, they either lock-in students and employees wishing to export their competencies elsewhere, or they require users’ trust and for users to give up privacy (to store their personal data) while being prone to faults. In this paper, we propose a user-centric, fully decentralised competency management system enabling verifiable, secure, and robust management of competencies digitalised as Open Badges via notarization on a public blockchain. This way, whoever acquires the competence or achievement retains full control over it and can disclose his/her own digital certifications only when needed and to the extent required, migrate them across storage platforms, and let anyone verify the integrity and validity of such certifications independently of any centralised organisation. The proposed solution is based on C-Box®, an existing application for the management of digital competencies that has been improved to fully support models, standards, and technologies of the so-called Web 3.0 vision—a global effort by major web organisations to “give the web back to the people”, pushing for maximum decentralisation of control and user-centric data ownership.

The ascomycete Tuber borchii (Pezizomycetes) is a whitish edible truffle that establishes ectomycorrhizal symbiosis with trees and shrubs. This fungus is ubiquitous in Europe and is also cultivated outside Europe. Here, we present the draft genome sequence of T. borchii strain Tbo3840 (97.18 Mb in 969 scaffolds, with 12,346 predicted protein-coding genes).

Truffles are ascomycete hypogeous fungi belonging to the Tuberaceae family of the Pezizales order that grow in ectomycorrhizal symbiosis with tree roots, and they are known for their peculiar aromas and flavors. The axenic culture of truffle mycelium is problematic because it is not possible in many cases, and the growth rate is meager when it is possible. This limitation has prompted searching and characterizing new strains that can be handled in laboratory conditions for basic and applied studies. In this work, a new strain of Tuber borchii (strain SP1) was isolated and cultured, and its transcriptome was analyzed under different in vitro culture conditions. The results showed that the highest growth of T. borchii SP1 was obtained using maltose-enriched cultures made with soft-agar and in static submerged cultures made at 22 °C. We analyzed the transcriptome of this strain cultured in different media to establish a framework for future comparative studies, paying particular attention to the central metabolic pathways, principal secondary metabolite gene clusters, and the genes involved in producing volatile aromatic compounds (VOCs). The results showed a transcription signal for around 80% of the annotated genes. In contrast, most of the transcription effort was concentrated on a limited number of genes (20% of genes account for 80% of the transcription), and the transcription profile of the central metabolism genes was similar in the different conditions analyzed. The gene expression profile suggests that T. borchii uses fermentative rather than respiratory metabolism in these cultures, even in aerobic conditions. Finally, there was a reduced expression of genes belonging to secondary metabolite clusters, whereas there was a significative transcription of those involved in producing volatile aromatic compounds.

Tuber borchii is an edible ectomycorrhizal mushroom of considerable economic value. Its cultivation has become popular in recent years, but there are few studies on the factors affecting its productivity. In this work, the ascoma production and the ectomycorrhizal (ECM) community of a T. borchii plantation, established in an intensive farming area where this truffle is not naturally present, were studied. Tuber borchii production drastically declined from 2016 to 2021, and ascomata of other Tuber species (T. maculatum and T. rufum) were found from 2017. Molecular characterization of ectomycorrhizae carried out in 2016 identified 21 ECM fungal species, of which T. maculatum (22%) and Tomentella coerulea (19%) were the most abundant. Tuber borchii ectomycorrizae (16%) were almost entirely confined to the fruiting points. The diversity and structure of the ECM community on Pinus pinea were significantly different from those observed on hardwood trees. The obtained results suggest that T. maculatum (a native of the study site) tends to replace T. borchii through a mechanism of competitive exclusion. Although T. borchii cultivation is possible in suboptimal environments, particular care should be taken to limit competition with ECM fungi more suitable for local conditions.

From the early 1800s science virtually ignored the cultivation of edible mycorrhizal mushrooms other than the true truffles. The drought was finally broken by Nicole Poitou when she cultivated Lactarius deliciosus and Suillus granulatus in the Institut National de la Recherche Agronomique’s laboratories at Pont-de-la-Maye, France, in the mid-1970s. However, another 20 years were to pass before Yun Wang working at Invermay Agricultural Centre near Dunedin, New Zealand, was able to begin the routine cultivation of L. deliciosus. The purpose of this paper is to document the history of the cultivation of L. deliciosus in New Zealand since its introduction in the late 1990s, its spread back to Europe and its subsequent introduction into Yunnan, Tibet and Sichuan starting about 2014.

Inside commercial batches of T. aestivum imported from Iran to Italy the ascomata of six Tuber spp. (Tuber borchii, Tuber brumale, Tuber macrosporum, Tuber rufum f. lucidum, Tuber excavatum and Tuber fulgens) were found and described. Sixteen specimens were analyzed; most of them were completely immature probably because truffles in Iran are harvested by digging all the soil forest without using trained dogs. The morphology of the Iranian ascomata and their spore dimensions are similar to those of the European ascomata and phylogenetic analyses based on their ITS sequences placed them inside the European clades of Tuber. In Iran, truffles are found in the northern part of the country with the highest production in the Hyrcanian region (between Guilan and Golestan provinces). To date, Iran seems to be the most Eastern location where European species of Tuber are found.

In the last decades, the number of studies about the valorization of agricultural by-products has strongly increased due to the growing focus on the circular economy. In this context, mushroom cultivation is raising attention for its nutritional properties and adaptability to different growing conditions. The goal of this project is, therefore, to create a link between two production chains, valorizing the solid anaerobic digestate fraction from the biogas plant as a potential substrate for Pleurotus ostreatus full-scale cultivation. A preliminary in vitro trial shows that the mycelia can colonize at the same rate in both the conventional substrate, which was wheat straw added with sunflower cake and the experimental ones supplemented with corn digestate. The most important and innovative part of the work was full-scale cultivation. Quantitative analyses demonstrate that P. ostreatus is capable of growing on the conventional substrate as well as on the substrate containing 15% of digestate without reducing the mushroom harvest. Even better, digestate seems to stimulate a precocious and more homogeneous production. Chemical and spectroscopic analyses on the fruiting bodies confirm the results; in fact, mushrooms show a comparable structural composition, highlighted with FTIR and SERS spectroscopic evaluations. Moreover, the nutrient content did not change due to the addition of digestate. These promising results demonstrate that anaerobic digestate can become a resource for P. ostreatus cultivation without production losses and maintaining the same qualitative characteristics.

The addition of n-3 polyunsaturated fatty acids (n-3 PUFAs) to the swine diet increases their content in muscle cells, and the additional supplementation of antioxidants promotes their oxidative stability. However, to date, the functionality of these components within muscle tissue is not well understood. Using a published RNA-seq dataset and a selective workflow, the study aimed to find the differences in gene expression and investigate how differentially expressed genes (DEGs) were implicated in the cellular composition and metabolism of muscle tissue of 48 Italian Large White pigs under different dietary conditions. A functional enrichment analysis of DEGs, using Cytoscape, revealed that the diet enriched with extruded linseed and supplemented with vitamin E and selenium promoted a more rapid and massive immune system response because the overall function of muscle tissue was improved, while those enriched with extruded linseed and supplemented with grape skin and oregano extracts promoted the presence and oxidative stability of n-3 PUFAs, increasing the anti-inflammatory potential of the muscular tissue.

This article focuses on the emergence of cooperation in societies of self-interested agents. In particular, it introduces a mechanism based on indirect—stigmergic—interactions between agents moving in an environment, to express the likeliness of finding cooperative partners. On the one hand, agents that find themselves cooperating with others emit pheromones in their current location, expressing the presence of agents willing to cooperate. On the other hand, agents that sense pheromones tend to move towards regions with a higher pheromone concentration. Results show that this mechanism leads to the emergence of spatial regions where cooperation can be effectively sustained, and in which agents can overall get better payoffs than those agents not taking into account pheromones in their choices. Tuber magnatum is the most expensive truffle, but its large‐scale cultivation is still a challenge compared to other valuable Tuber species. T. magnatum mycelium has never been grown profitably until now, which has led to difficulties to studying it in vitro. This study describes beneficial interactions between T. magnatum mycelium and never before described bradyrhizobia, which allows the in vitro growth of T. magnatum mycelium. Three T. magnatum strains were co‐isolated on modified Woody Plant Medium (mWPM) with aerobic bacteria and characterised through microscopic observations. The difficulties of growing alone both partners, bacteria and T. magnatum mycelium, on mWPM demonstrated the reciprocal dependency. Three bacterial isolates for each T. magnatum strain were obtained and molecularly characterised by sequencing the 16S rRNA, glnII, recA and nifH genes. Phylogenetic analyses showed that all nine bacterial strains were distributed among five subclades included in a new monophyletic lineage belonging to the Bradyrhizobium genus within the Bradyrhizobium jicamae supergroup. The nifH genes were detected in all bacterial isolates, suggesting nitrogen‐fixing capacities. This is the first report of consistent T. magnatum mycelium growth in vitro conditions. It has important implications for the development of new technologies in white truffle cultivation and for further studies on T. magnatum biology and genetics. [ABSTRACT FROM AUTHOR]

Sensor-based human activity recognition (HAR) is having a significant impact in a wide range of applications in smart city, smart home, and personal healthcare. Such wide deployment of HAR systems often faces the annotation-scarcity challenge; that is, most of the HAR techniques, especially the deep learning techniques, require a large number of training data while annotating sensor data is very time- and effort-consuming. Unsupervised domain adaptation has been successfully applied to tackle this challenge, where the activity knowledge from a well-annotated domain can be transferred to a new, unlabelled domain. However, these existing techniques do not perform well on highly heterogeneous domains. This article proposes shift-GAN that integrate bidirectional generative adversarial networks (Bi-GAN) and kernel mean matching (KMM) in an innovative way to learn intrinsic, robust feature transfer between two heterogeneous domains. Bi-GAN consists of two GANs that are bound by a cyclic constraint, which enables more effective feature transfer than a classic, single GAN model. KMM is a powerful non-parametric technique to correct covariate shift, which further improves feature space alignment. Through a series of comprehensive, empirical evaluations, shift-GAN has not only achieved its superior performance over 10 state-of-the-art domain adaptation techniques but also demonstrated its effectiveness in learning activity-independent, intrinsic feature mappings between two domains, robustness to sensor noise, and less sensitivity to training data.

In this work we used an inductive and deductive approach to produce the map of suitability for the spontaneous growth of Tuber magnatum (white truffle) (SSGT) in Emilia-Romagna region (northern Italy). This map was produced in order to identify the environments where appropriate actions should be applied to protect this threatened truffle species.The steps used to define the map were: 1) surveying and mapping the actual productive areas (APAs) in two provinces of Emilia-Romagna (Bologna and Modena); 2) identification of some morphological, climatic, vegetational and pedological properties related to T. magnatum development; 3) production of digital maps representing the value (classified) that each property assumes in discrete land portions; 4) overlapping the map of APAs with each of the thematic maps and selection of the properties showing the strongest relationships with the presence of T. magnatum; 5) creating the map of SSGT, covering the whole hilly area of Emilia Romagna region; 6) assessing the reliability of the SSGT map, by overlapping the APAs map and the areas of the provinces of Parma and Piacenza where the mycelium of T. magnatum was found using species-specific primers.The relationships found by means of the inductive approach (comparison between selected properties and observed frequency of truffle) demonstrated its effectiveness in predicting deductively the areas with distinct suitability for truffle.

The widespread use of genome-wide association studies resulted in the discovery of genomic regions associated with fatty acid (FA) composition in different porcine tissues, but little information exists about the genes involved in FA composition of meat obtained from heavy pigs selected for the production of Italian dry-cured hams. To this objective, we genotyped with a single nucleotide polymorphism (SNP) panel 795 Italian Large White heavy pigs to identify the markers and genomic regions associated with Semimembranosus muscle FA profile. Heritability estimates for intramuscular fat FA profile were of low-to-moderate magnitude, suggesting that these traits may be improved with genomic selection. On the whole, 45 SNPs were significantly associated with 14 FAs, and 4 of them (ALGA008109, ALGA0081097, CASI0010164 and SIRI0000267) were associated with more than 1 FA. The palmitoleic : palmitic and oleic : stearic ratios displayed the highest number of significant markers and the most significant associations (Bonferroni adjusted P < 5.00E−07). Of particular interest, the palmitoleic : palmitic ratio was strongly associated with markers located at 111 to 114 Mb on chromosome 14, in the same chromosomal region where Stearoyl-CoA desaturase Δ9 (SCD) gene is located. Several significant chromosomal regions were found; some of them harbour key genes playing pivotal roles in FA desaturation and elongation, such as SCD and some members of the Elongation of Very Long-Chain FA (ELOVL) gene family. The results suggest that the identification of causal mutations in these regions may provide a set of markers useful for selection schemes aimed at improving FA composition in pork products.

Emerging application scenarios, such as cyber-physical systems (CPSs), the Internet of Things (IoT), and edge computing, call for coordination approaches addressing openness, self-adaptation, heterogeneity, and deployment agnosticism. Field-based coordination is one such approach, promoting the idea of programming system coordination declaratively from a global perspective, in terms of functional manipulation and evolution in "space and time" of distributed data structures called fields. More specifically regarding time, in field-based coordination (as in many other distributed approaches to coordination) it is assumed that local activities in each device are regulated by a fair and unsynchronised fixed clock working at the platform level. In this work, we challenge this assumption, and propose an alternative approach where scheduling is programmed in a natural way (along with usual field-based coordination) in terms of causality fields, each enacting a programmable distributed notion of a computation "cause" (why and when a field computation has to be locally computed) and how it should change across time and space. Starting from low-level platform triggers, such causality fields can be organised into multiple layers, up to high-level, collectively-computed time abstractions, to be used at the application level. This reinterpretation of time in terms of articulated causality relations allows us to express what we call "time-fluid" coordination, where scheduling can be finely tuned so as to select the triggers to react to, generally allowing to adaptively balance performance (system reactivity) and cost (resource usage) of computations. We formalise the proposed scheduling framework for field-based coordination in the context of the field calculus, discuss an implementation in the aggregate computing framework, and finally evaluate the approach via simulation on several case studies.

Hypervariable loops of HIV-1 Env protein gp120 are speculated to play roles in the conformational transition of Env to the receptor binding-induced metastable state. Structural analysis of full-length Env-based immunogens, containing the entire V2 loop, displayed tighter association between gp120 subunits, resulting in a smaller trimeric diameter than constructs lacking V2. A prominent basal quaternary location of V2 and V3' that challenges previous reports would facilitate gp41-independent gp120-gp120 interactions and suggests a quaternary mechanism of epitope occlusion facilitated by hypervariable loops. Deletion of V2 resulted in dramatic exposure of basal, membrane-proximal gp41 epitopes, consistent with its predicted basal location. The structural features of HIV-1 Env characterized here provide grounds for a paradigm shift in loop exposure and epitope occlusion, while providing substantive rationale for epitope display required for elicitation of broadly neutralizing antibodies, as well as substantiating previous pertinent literature disregarded in recent reports.

Pedodiversity is generally neglected in studies concerning soil organic carbon (SOC). Therefore, this investigation aimed to explore the effect of soil types on the following: (1) soil processes related to organic matter (OM) dynamics along the profile; and (2) the microbial community and functionality within the uppermost horizon. Humic Dystrudepts (HD), Typic Dystrudepts (TD), and Humic Lithic Dystrudepts (HLD) were selected in beech forests of the Apennine ridge in the Emilia-Romagna Region (Italy). Soils were sampled by horizons until parent material, and physico-chemical and functional analyses were performed. The results showed that both HD and HLD soils had a higher SOC accumulation than TD, particularly within the deeper horizons. Such accumulation might be due to the lower turnover rate of soil OM forms, namely fulvic acid-like substances, humic acid-like substances, and non-extractable OM. Noteworthy, the A horizons showed slight differences in SOC among the soil types, suggesting similar SOC decomposition processes. This fact was confirmed by the lack of differences in microbial DNA-based diversity and functionality. Morchella species are known as famous and prized edible fungi due to their culinary flavor and medicinal properties. The asomata are collected throughout the temperate regions of the northern hemisphere. Morchella spp. taxonomy has long been debated as a result of the high phenotypic plasticity charaterizing the genus. Most morels are considered saprobic but some species has been reported to interact with roots of many plant species forming different types of associations. In Emilia-Romagna (Italy), morels became a part of the culinary tradition, especially for the populations of the coastal areas. This work aimed to describe and identify the ascomata collected on the white dune habitat as well as to verify the extent of the interaction with plant species growing in the same area. All ascomata collected since 2001 shared a similar morphology with a range of variability mainly due to the harvesting period. Ascomata collected in 2017 were grouped within the Mes-17 clade, in the Esculenta group, based on their ITS rDNA sequences. Nine different plant species were found to be connected with morel ascomata through characteristic sand-mycelial structures never described before. The amount of the ascomata collected on the sand dunes demonstrates that the study area is particularly suitable for morel fruiting. Therefore, the protection of the dune system is imperative for preserving Morchella genetic diversity within the local population.

The screening of morphological and growth characteristics of mycelial collections of medicinal bracket fungus Ganoderma adspersum (Agaricomycetes, Polyporales) of different geographical origins (Armenia, Georgia, Iran) on malt-extract agar (MEA) and potato-dextrose agar (PDA) media for 6 days, as well as in submerged culture (malt-extract, ME, 200 rt min) for 14 days at different temperature (25, 30, 35, 38 °C) was performed. Species-specific mycelial macro- and micromorphological characteristics, such as white, cottony-felt, later chamois, leathery, creamy-lemon-yellowish fast-growing colonies, round-shaped hyphal clamps, hyaline, round-shaped, smooth chlamydospore-like swellings and brownish cuticular cells, numerous tetrahedral crystals were described in agar and submerged cultures. The formation of small, dense and smooth pellets, as well as hyphal swellings during submerged growth was observed. The favorable growth temperature for studied collections of G. adspersum was 25-30 °C. The revealed cultural characteristics will be used for taxonomic identification and quality control of mycelial cultures during their biotechnological cultivation.

Subcutaneous fat thickness and fatty acid composition (FAC) play an important role on seasoning loss and organoleptic characteristics of seasoned hams. Dry-cured ham industry prefers meats with low contents of polyunsaturated fatty acids (PUFA) because these negatively affect fat firmness and ham quality, whereas consumers require higher contents in those fatty acids (FA) for their positive effect on human health. A population of 950 Italian Large White pigs from the Italian National Sib Test Selection Programme was investigated with the aim to estimate heritabilities, genetic and phenotypic correlations of backfat FAC, Semimembranosus muscle intramuscular fat (IMF) content and other carcass traits. The pigs were reared in controlled environmental condition at the same central testing station and were slaughtered at reaching 150 kg live weight. Backfat samples were collected to analyze FAC by gas chromatography. Carcass traits showed heritability levels from 0.087 for estimated carcass lean percentage to 0.361 for hot carcass weight. Heritability values of FA classes were low-to-moderate, all in the range 0.245 for n-3 PUFA to 0.264 for monounsaturated FA (MUFA). Polyunsaturated fatty acids showed a significant genetic correlation with loin thickness (0.128), backfat thickness (−0.124 for backfat measured by Fat-O-Meat’er and −0.175 for backfat measured by calibre) and IMF (−0.102). Obviously, C18:2(n-6) shows similar genetic correlations with the same traits (0.211 with loin thickness, −0.206 with backfat measured by Fat-O-Meat’er, −0.291 with backfat measured by calibre and −0.171 with IMF). True truffles in the genus Tuber are the most valuable ectomycorrhizal fungi and their cultivation has become widespread around the world. Competition with other ectomycorrhizal fungi and especially with undesired Tuber species, like T. brumale, can threaten the success of a truffle plantation. In this work, the competitiveness of T. brumale towards T. melanosporum and T. aestivum was assessed in a 14 year-old plantation carried out planting seedlings inoculated with these three truffle species in adjacent plots. Analyses of both truffle ectomycorrhizas and extra-radical mycelium were carried out in the transects separating the T. brumale plot from T. melanosporum and T. aestivum plots. The results confirm the competitiveness of T. brumale against T. aestivum and T. melanosporum due to its major ability to colonize the soil around its ectomycorrhizas. However, its competitiveness is limited to the transect areas and it was never found inside T. melanosporum plot. These results remark that, in presence of optimal conditions for T. melanosporum and T. aestivum, the greatest risk of contamination with T. brumale is due to wrong greenhouse activity.
