Introduction:Feronia limonia (synonyms: Feronia elephantum, Hesperethusa crenulata, Limonia acidissima Linn, Schinus limonia) is a member of Rutaceae family. F. limonia is an aesthetically pleasing plant with various medicinal and nutraceutical properties.

Creating salt-tolerant genotypes is crucial for maximizing the productivity under salinized land. To evaluate genetic diversity for salt tolerance, 35 diverse cotton accessions were screened under 17 dS m−1 salt stress conditions using 20 agronomic and physiological traits relevant to salt tolerance. The general linear model analysis indicated significant salinity impacts across the studied accessions, and genotype × treatment effects were also significant for all parameters examined. Among all 35 studied accessions; the genotypes CCB-1, CCB-2, CCB-28, CCB-3, CCB-4, Ghauri-1, JSQ-70, and JSQ-71 showed considerably higher performance for plant height, boll per plant, boll weight, lint percentage, seed cotton yield and fiber quality traits under salt stress. Physiological traits, chlorophyll and carotenoid contents, total soluble proteins, K+, and K+/Na+ were reduced under saline conditions, while biochemical traits such as catalase, superoxide dismutase, peroxidase, H2O2 and MDA level increased. The genotypes CCB-17, CCB-18, CCB-19, CCB-20, CCB-21, CCB-22, Hatf-3, Badar-1, Eagle-2, Eagle-4, CCB-23, CCB-24, CCB-25, CCB-26 and CCB-28 exhibited lower values for agro-physiological and fiber quality character respectively, signifying their sensitivity to salt stress. Under salinity, these genotypes showed reduced antioxidant levels and increased values for K+/Na+, Na+, H2O2, and MDA contents. Whereas the genotypes CCB-5, CCB-6, CCB-7, CCB-8, CCB-9, CCB-10, CCB-11, CCB-12, CCB-13, CCB-14, CCB-15, and CCB-16 demonstrated moderate performance for these traits under salt stress conditions respectively. Utilizing multivariate analysis techniques (cluster and PCA), 35 genotypes have been categorized into three groups based on studied traits: tolerant (cluster-1), moderately tolerant (cluster-2), and susceptible (cluster-3) under saline conditions. Liver cancer is the sixth most frequent malignancy and the fourth major cause of deaths worldwide. The current treatments are only effective in early stages of cancer. To overcome the therapeutic challenges and exploration of immunotherapeutic options, broad spectral therapeutic vaccines could have significant impact. Based on immunoinformatic and integrated machine learning tools, we predicted the potential therapeutic vaccine candidates of liver cancer. In this study, machine learning and MD simulation-based approach are effectively used to design T-cell epitopes that aid the immune system against liver cancer. Antigenicity, molecular weight, subcellular localization and expression site predictions were used to shortlist liver cancer associated proteins including AMBP, CFB, CDHR5, VTN, APOBR, AFP, SERPINA1 and APOE. We predicted CD8+ T-cell epitopes of these proteins containing LGEGATEAE, LLYIGKDRK, EDIGTEADV, QVDAAMAGR, HLEARKKSK, HLCIRHEMT, LKLSKAVHK, EQGRVRAAT and CD4+ T-cell epitopes of VLGEGATEA, WVTKQLNEI, VEEDTKVNS, FTRINCQGK, WGILGREEA, LQDGEKIMS, VKFNKPFVF, VRAATVGSL. We observed the substantial physicochemical properties of these epitopes with a significant binding affinity with MHC molecules. A polyvalent construct of these epitopes was designed using suitable linkers and adjuvant indicated significant binding energy (>-10.5 kcal/mol) with MHC class-I and II molecule. Based on in silico cloning, we found the considerable compatibility of this polyvalent construct with the E. coli expression system and the efficiency of its translation in host. The system-level and machine learning based cross validations showed the possible effect of these T-cell epitopes as potential vaccine candidates for the treatment of liver cancer. This study focuses on the cowpea weevil, Callosobruchus maculatus, a globally distributed grain pest that affects cereals and pulses. Using chemicals to store grains can harm pest control and pose risks to consumers and the environment. The facultative intracellular symbiont bacteria Wolbachia can affect host's reproductive capacities in a variety of ways, which makes it useful in the management of pests such as C. maculatus. The main goal of the study was to identify Wolbachia diversity in the C. maculatus population. Phylogenetic analysis utilized mitochondrial COI and 12S rRNA genes to identify the host C. maculatus, while screening for Wolbachia was conducted using genes (wsp, coxA, and ftsZ) genes. Molecular phylogenetic analysis of the Wolbachia genes resulted in one new Wolbachia strain (wCmac1) in C. maculatus populations and contrasting already published data of other Callosobruchus strains. The study discussed the detection of Wolbachia and its phylogenetic comparison with other C. maculatus and Coleopteran populations. Simple Summary: The global supply of the primary component of poultry feed, i.e., soybean meal, has dwindled due to an imbalance between demand and supply. Hence, there is an urgent need for exploring some cost-effective and environmentally friendly alternative protein sources to ensure sustainable poultry production. The outcomes of the present study exhibited that replacing 12% of soybean meal with either Periplaneta americana or Hermetia illucens improves the growth, blood hematology, gut morphometry, and meat quality of broilers. Consequently, these insects can serve as a viable alternative to traditional protein sources in poultry nutrition, offering a promising solution to meet the rising demand for animal protein, while mitigating the environmental impacts associated with conventional feed ingredients. Insects are becoming increasingly popular as a sustainable and nutritious alternative protein source in poultry feeds, due to their high protein content, low environmental impact, and efficient feed conversion rates. Using insect-based feeds can reduce the need for traditional protein sources like soybean meal (SBM), which often contribute to environmental issues such as deforestation and high water consumption. The current experiments were devised to assess the impacts of the partial replacement of SBM with the American cockroach Periplaneta americana and black soldier fly Hermetia illucens on the performances, hematology, gut morphometry, and meat quality of male broilers (Ross 308). A total of 350, 1-day-old chicks weighing 40.05 ± 0.27 g were divided into 7 dietary treatments (5 pens/treatment and 10 birds/pen) at random, i.e., a 4, 8, or 12% SMB replacement with P. americana and H. illucens. Soybean meal was used as a basal diet and taken as a control. The results indicated that broilers fed on 12% P. americana or H. illucens showed significant improvements (p < 0.05) in feed conversion ratio, live weight, and daily weight gain. Hematological traits significantly improved (p < 0.05). A gut histology showed increased villus height, villus width, crypt depth, and villus height/crypt depth ratios, indicating improved nutrient absorption. Broiler meat fed on 12% P. americana meal had significantly higher redness and yellowness (p < 0.05). Substituting soybean meal with up to 12% P. americana or H. illucens meal in poultry feed can enhance the broilers' growth performance, hematology, gut morphometry, and meat quality. Hence, these findings imply that P. americana or H. illucens meal are viable and constructive alternative protein sources in poultry nutrition, offering a sustainable approach to meet the increasing demand for animal protein across the world. To investigate in vitro anticancer, antimicrobial, antioxidant and in vivo hypoglycaemic effects of crude methanolic extracts (CMEs) of Berberis baluchistanica and Daphne oleoides. MTT assay for cytotoxicity on HeLa and NIH cells, disc diffusion protocols for antimicrobial and DPPH assay for antioxidant potential were applied. In vivo hypoglycaemic effect was investigated on Alloxan-induced diabetic rabbits. D. oleoides CME exhibited moderate cytotoxic behaviour against HeLa cells (IC50 77.87µg/mL) whereas B. baluchistanica CME was found deficient (IC50 170.02µg/mL). P. aeruginosa was susceptible to both CMEs. M. luteus and B. subtilis was prone to the bactericidal effects of D. oleoides and B. baluchistanica CMEs respectively. D. oleoides CME inhibited more than 80% S. cerevisiae and 60% C. glabrata colonies. B. baluchistanica CME showed significant antioxidant activity (IC50 52.86µg/ml) than D. oleoides CME (IC50 87.30µg/ml) and standard resveratrol (IC50 109.46µg/ml). B. baluchistanica CME showed superior antidiabetic effect (135.75 mg/dl ±0.53) as compared to D. oleoides CME (191.50 mg/dl ± 0.48) but less antidiabetic effect than metformin hydrochloride (standard). All the above potentials exhibited by D. oleoides and B. baluchistanica CMEs propose further investigations to isolate and purify responsible biologically active lead molecule(s) with diverse capabilities. Background: Degenerative kidney diseases are mostly associated with oxidative stress. Natural products are considered as the antioxidants enrich food that can restrict the progress of oxidative stress induced disorders. Therefore, the present study was aimed to evaluate the renal protective effect of Ajuga parviflora leaf extract in carbon tetrachloride intoxicated rats. Methods: The hydromethanolic extract of A. parviflora leaves was obtained by extracting twice in 60% methanol. The principal bioactive constituents were detected by LC/MS analysis. Toxicity of plant extract was assessed using brine shrimp lethal toxicity test and acute toxicity model on healthy Sprague-Dawley male rats. Nephroprotective effects of plant extract were also evaluated on rats by inducing CCl4 renal toxicity in comparison with positive control and naïve groups. The dose of A. parviflora administered to animal was 100, 200 and 300 mg/kg. All administrations were given orally on an alternate day basis for 30 days. Urine and serum biomarkers were analyzed, along with antioxidant enzymes. Finally, the DNA damages, lipid peroxides, hydrogen peroxides and nitrites were assessed in rat's renal tissue. The histopathology alterations in renal tissues were further studied for kidney damages. Results: The LC/MS analysis confirmed the presence of different important pharmacological compounds in A. parviflora methanolic leaf extract. The key bioactive compounds include pyocyanin, zonisamide, D Saccharic acid, altretamine, carbocyclic thromboxane A2, Sinapyl alcohol, and vitamin C. The important polypeptides identified include Lys-Tyr-Lys, His-His-Lys, Met-Asp-Arg, Phe-Val-Arg, and PyroGlu-Val-Arg. The LD50 of A. parviflora was found to be > 1000 μg/mL. A. parviflora administration significantly subsides CCl4 toxicity in rats, reduced the elevated level of RBCs, pus and epithelial cells. The abnormal elevated level of specific gravity, creatinine, urobilinogen, urea and albumin were also reduced to normal physiological level. The reduced urinary protein and pH were also normalized. The serum urobilinogen, urea and total bilirubin levels were also reversed to normal levels while the diminished albumin and total protein levels also came to normal. The important phase I and II enzyme levels were also reversed in A. parviflora administered rats. The H2O2, thiobarbituric acid reactive substance (TBARS) and nitrite levels were significantly decreased. Furthermore, the damaged DNA and histopathological changes in CCl4 exposed rats were also highly significantly reversed after the administration of A. parviflora. All effects were significant (P < 0.05) and highly significant (P < 0.005) at 100 and 300 mg/kg respectively. Conclusion: The restored urine and serum profile of various parameters to normal physiological levels suggests that the A. parviflora has potential antioxidant and repairing potential in renal disorders. The threat of multi-drug resistant bacterial pathogens evokes researchers to synthesized safe and effective chemotherapeutic agents for nano-drug delivery system. In current study, Schiff base of nicotinic hydrazide(NHD) and its silver nanoparticles(NHD-AgNPs) were synthesized and characterized. These compounds were investigated for cytotoxicity, antibacterial and AFM activity. The NHD showed LD50 at >1000μg/mL while NHD-AgNPs didn't exhibit toxicity at 1000μg/mL against 3T3 cell line. The NHD showed zone of inhibition against two strains of salmonella enteric (ATCC 14028 and 700408) 45.29±1.66 and 48.01±1.43mm respectively at 160μg/mL (p<0.01) while NHDAgNPs exhibited 55.87±2.08 and 52.88±1.42 mm respectively at 130μg/mL (p<0.001) in disc diffusion method. NHD showed more than 70% growth inhibition for both strains at 85 and 125μg/ml (p<0.01) respectively, while NHD-AgNPs inhibit 80% and 75% respectively at 75 and 125 μg/ml (p<0.01, p<0.001) against Alamar blue antibacterial assay. For morphological changes in bacterial cell wall NHD and NHD-AgNPs treated bacterial cells were observed under atomic force microscope(AFM) and treated bacterial cells were severely damaged with leaked cytoplasmic contents as compare to untreated bacterial cell. These results validate that NHD-AgNPs were highly active as compared to NHD against both strains at their MIC concentrations. In future, comparative wound healing potential will be emphasized. Medicinal plants are proven to reveal vast promising potential providing novel drug candidates to combat health-related problems. The aim of current study is to discover new drug compounds with anti-anticancer, antioxidant, antibacterial and antifungal potential, to serve the purpose Saussurea heteromalla (Family: Asteraceae) indigenous to Pakistan was screened for the in vitro cytotoxicity against HeLa cells (Human cervical cancer cell line) compared to the NIH / 3T3 cells (mouse normal fibroblast cells) by performing the MTT colorimetric assay and antifungal, antibacterial and antioxidant potential by adopting standard protocols. S. heteromalla crude methanolic extract (CME) demonstrated strong cytotoxic potential against HeLa cells at 200µg/mL; (77.28 ±1.53% kill; IC50: 62.13µg/mL) compared to standard doxorubicin (95.90% kill; IC50: 0.2µg/mL). Inhibitory Zone of the extract at concentrations (30, 60, 90µg/mL) against Bacillus subtilis, Serratia marecescens, Staphylococcus aureus, Micrococcus luteus, Pseudomonas aeruginosa, Strptotropomonas maltophilia, Escherichia coli, Salmonella typhi, and Saccharomyces cerevisiae, Candida albicans, Candida glabrata was measured. Manifestation of intensified results against Gram-negative Serratia marecescens qualifies the S. heteromalla extract as a considerable source of narrow spectrum antibiotic. However, antifungal activity against C. albicans was found to be logical. Antioxidant potential was determined through DPPH assay which declared no notable antioxidant effects. Response of 49 accessions of cotton (Gossypium hirsutum L.) was examined at seedling stage in normal and limited water supply. The experiment was conducted in glasshouse conditions using three moisture levels i.e. normal water supply and two stress conditions. After fifteen days root length, shoot length, root weight, shoot weight, excised leaf water loss and relative water content were measured. Analysis of variance indicated significant differences among accessions for all the traits. Differences among cotton accessions were assessed using least significant difference test. Accession Shaheen showed appreciable response for the six traits, whilst Ali Akbar 802 showed maximum performance for the traits i.e. root length, shoot length, root weight, and shoot weight. IR-3701 performed better for relative water content and excised leaf water loss under normal conditions, whereas, TADLA-32 showed better performance for four traits namely, root length, shoot length, root weight, and shoot weight under stress condition. In this study Error Correction Model (ECM) and Johansen Cointegration Technique (JCT) are applied to explore short-run and long-run association among the chosen variables, i.e. Manufacturing Sector Output (MNF), Gross Fixed Capital Formation (GFCF), Liberalization of Trade (RTLIB) and GDP Deflator (DEF), to evaluate impact of Neo-liberalism on manufacturing sector and economic growth. The data of the variables is taken from 1973-2012. It is found that GFCF and RTLIB have a significant (at 5% level) and positive impact on MNF, but DEF has a significant (at 5% level) and negative impact on MNF. Further these results describe the fact, if Pakistan wants to make trade liberalization effectively then reform process to be made strengthen with appropriate institutional support.
