Abstract

Abstract Severe Typhoon Koinu passed south of Hong Kong on 8 and 9 October 2023, triggering the issuance of the Increasing Gale or Storm Signal No. 9, the second highest tropical cyclone (TC) warning signal in Hong Kong. Koinu was a difficult case for TC warning service due to its compact size and rather erratic movement over the coastal waters of Guangdong. To monitor Koinu's movement and wind structure, the Hong Kong Observatory utilized various observational platforms, including meteorological aircraft, ocean radar, and synthetic aperture radar on polar orbiting satellites. The paper presents major observations derived from these measurements. The aircraft probe and dropsonde data suggested boundary layer inflow, warm core structure, eyewall updraft, and high turbulence in the eyewall of the typhoon. The weather radar observations indicated the occurrence of a waterspout in the vicinity of the typhoon. Additionally, the study highlights the forecasting performance of the AI‐based Pangu‐Weather model, which could outperform the conventional global numerical weather prediction models in forecasting TC track in the region. The documentation of these observations aims to provide valuable references for weather forecasters and stimulate further research on forecasting this type of tropical cyclone.

2. Propagation rules of shock waves in confined space under different initial pressure environments

Abstract

Abstract In this paper, an initial pressure adjustable explosion vessel was developed, and the effect of negative pressure, positive pressure (0.2–1.8 atm) different initial ambient pressure on the explosive shock wave generated by the explosion of explosives was studied. The relationships between the specific impulse, shock wave velocity, the amount of explosive gas products and the ambient pressure were analyzed for different initial pressure environments. It was found that: the overpressure of the blast shock wave decreases with the initial ambient pressure of the explosion, and there exists a negative pressure environment with a dramatic pressure decrease near 0.6 atm, defined as the super-sensitive negative pressure Pcr. The propagation velocity of an explosive wave increases with a decrease in the ambient pressure, and the propagation velocity at a pressure of 1.8 atm is four times less than the velocity at a pressure of 0.2 atm. The production of explosive gas products did not change. The greater the initial pressure of the environment where the explosive is located, the smaller the ratio of the gas generated by the explosion to the initial force gas in the explosion vessel is, and the greater the impact on the propagation of shock waves is. The maximum attenuation of the first specific impulse i 1 is 72.97% and the maximum attenuation of the second specific impulse i 2 is 72.39%. The experiments provide reference data for high-altitude military confrontation, high-altitude weapons and ammunition development, and deep-earth protection engineering.

Abstract

An interstitial solute strengthened high entropy alloy (iHEA), Fe49.5Mn30Co10Cr10C0.5 (at.%), was successfully additively manufactured via selective laser melting. The as-built iHEA exhibits a hierarchically heterogeneous microstructure with length scales across several orders of magnitude, which engenders an enhanced strength–ductility combination relative to those fabricated by conventional processing routes. The high yield strength mainly stemmed from the dislocation strengthening besides the friction stress and grain boundary strengthening. The joint activation of multiple deformation mechanisms involving dislocation slip, deformation twinning and phase transformation can maintain the steady work-hardening behavior at high stress levels, leading to a high ductility.

Abstract

Abstract Atom probe tomography (APT) coupling high-resolution transmission electron microscopy (HRTEM) was used to analyze the fraction and compositions of different nanoparticles in two-stage double peaks aging process of Al−Zn−Mg alloy. Al content is found to be closely related to the size of nanoparticles and it can be greater than ~ 50.0 at. % in the nanoparticle with the equivalent radius under ~ 3.0 nm. Correspondingly, Al content of the nanoparticle, with the equivalent radius over ~ 5.0 nm, is measured under ~ 40.0 at. %. Evolution from Guinier–Preston (G.P.) zone to η phase is a growing process where Mg and Zn atoms enter the nanoparticle, therefore rejecting Al atoms. G.P. zones can take up a number fraction of ~ 85.0 and ~ 22.7% of nanoparticles in the first and second peak-aged samples, respectively, and even in the over-aged (T73) sample, they can still be found. As aging time increases, fraction of η′ phases monotonically rises to the peak value (~ 54.5%) in the second peak-aged state and then drops, which is significant for the second hardness peak and directly proves their function as the transitional medium. In T73 state, ~ 63.3% nanoparticles compose of η phases, which were measured to still contain ~ 10.2 to ~ 36.4 at. % Al atoms.

5. Role of antioxidants in preventing testicular ischemia‑reperfusion injury: a narrative review

Abstract

Testicular ischemia-reperfusion injury (TIR) is a urological emergency common among male newborns, children, and adolescents. Testicular injury and its consequences, such as altered hormone production, subfertility, and infertility, are determined by the duration and degree of testicular torsion. Early diagnosis and treatment are crucial to preserving the testes and fertility. Previous studies suggest that reactive oxygen species contribute to the pathogenesis of TIR injury, but the underlying mechanism remains unclear. Several drugs/plants reportedly exhibit antioxidative activities to protect against TIR. This review summarizes current studies on the role of antioxidants in preventing experimental TIR injury and discusses the underlying pathophysiological mechanisms.

Abstract

This paper proposes an automatic approach for residential areas revision by means of analysing the correlation between the residential area and the topographic and geographical factors. The approach consists of four major steps: the extracting of missing residential area from the remote sensing images with high resolution; the statistic analysing on the size changes of missing residential area in each grade of the elevation, slope, distance from the road and other impact factors; modelling of residential area modification in the urban and rural region; testing the methods using 100 counties data which are located in the middle part of China North-South Seismic Belt and comparing the result to the Land Use in map scale 1:100000. The experimental results present the accuracy of urban residents by 70% increased to 89.4%, rural residents by 47% up to 81.9%, rural residents from 8% increased to 78.5%. Therefore, there is available risk exposure information in a sparsely populated area because the spatial grid distributions of population and buildings are based on the residential areas. The proposed approach in this paper will improve the accuracy of the seismic risk assessment if it is applied to the national or the whole world.

Abstract

Background Occult peritoneal metastasis (PM) in advanced gastric cancer (AGC) patients is highly possible to be missed on computed tomography (CT) images. Patients with occult PMs are subject to late detection or even improper surgical treatment. We therefore aimed to develop a radiomic nomogram to preoperatively identify occult PMs in AGC patients. Patients and methods A total of 554 AGC patients from 4 centers were divided into 1 training, 1 internal validation, and 2 external validation cohorts. All patients’ PM status was firstly diagnosed as negative by CT, but later confirmed by laparoscopy (PM-positive n = 122, PM-negative n = 432). Radiomic signatures reflecting phenotypes of the primary tumor (RS1) and peritoneum region (RS2) were built as predictors of PM from 266 quantitative image features. Individualized nomograms of PM status incorporating RS1, RS2, or clinical factors were developed and evaluated regarding prediction ability. Abstract

Inflammation and fibrosis progress of nucleus pulposus (NP) cells participate in the pathologic changes of intervertebral disc degeneration (IDD). ANGPTL2 is well known for its angiogenesis and proinflammatory properties and transforming growth factor β1 (TGF-β1) is also responsible for tissue fibrosis. However, the role of ANGPTL2 in IDD and whether it is related to TGF-β1 remains unclear. This study aims to explore the relation of TGF-β1 and ANGPTL2 in the degenerative process of NP cells.We isolated NP cells of NP tissues provided from the spine fracture patients. IL-1β was used to induce the NP cells degeneration. To determine the effect of TGF-β1 and ANGPTL2 on NP cell degeneration, we regulated the cellular TGF-β1 and ANGPTL2 expression by Recombinant human protein stimulation and siRNA transfection. Quantitative real-time polymerase chain reaction (qRT-PCR) or Western blot was employed to investigate the expression of TGF-β1, ANGPTL2, IL-6, TNF-α, collagen I, and collagen III.TGF-β1 overexpression aggravated the ANGPTL2, IL-6, TNF-α, collagen I, and collagen III expressions of NP cells that caused by IL-1β, which was rejected by ANGPTL2 gene silencing. Besides, the silencing of TGF-β1 weakened the ANGPTL2 expression. ANGPTL2 overexpression promoted the NP cells inflammation and fibrosis via increasing IL-6, TNF-α, collagen I, and collagen III expression, which was sharpened by a consequent increase of TGF-β1 expression.This study, for the first time, points that TGF-β1 aggravates degenerative NP cells inflammation and fibrosis via the mediation of ANGPTL2.

15. Changes in serum inflammatory factors, adiponectin, intestinal flora and immunity in patients with non-small cell lung cancer

Abstract

The aim of this study was to explore the changes in the body state of patients with non-small cell lung cancer (NSCLC), including intestinal flora, serum inflammatory factors, immunity and adiponectin.A total of 18 NSCLC patients (disease group) and 16 healthy people from the Medical Center (control group) were selected as research objects. The levels of immune molecules immunoglobulin A (IgA), IgG and IgM, and inflammatory factors interleukin-2 (IL-2), C-reactive protein (CRP), tumor necrosis factor-α (TNF-α) and IL-6 were detected via enzyme-linked immunosorbent assay (ELISA). The level of adiponectin was determined using the quantitative kit. In addition, the changes in intestinal flora were analyzed.The overall survival time of NSCLC patients was significantly affected by IL-2 (p=0.0026), CRP (p=0.03), TNF-α (p=0.014) and IL-6 (p=0.00018). It can be seen that these inflammatory factors may play important roles in the progression of NSCLC. The levels of TNF-α (p=0.037), IL-2 (p=0.043) and CRP (p=0.000) in the peripheral blood serum were significantly higher in disease group than control group. Meanwhile, the levels of IgA (p=0.040) and IgG (p=0.000) in the peripheral blood serum were significantly higher in disease group than control group. However, no significant difference was observed in the level of IgM between the two groups (p0.05). The expression of adiponectin gene (ADIPOQ) could remarkably affect the overall survival rate of NSCLC patients, and patients with high expression of ADIPOQ exerted significantly better prognosis (p=0.017). The level of serum adiponectin was evidently higher in control group than that in disease group (p0.05). According to the linear discriminant analysis (LDA) score of the intestinal flora in both groups, the abundance of some intestinal flora (Enterobacter and Lachnospiraceae) was markedly higher in disease group than control group (p0.05). However, the abundance of Bifidobacteria, Pediococcus and Lactobacillus was remarkably higher in control group than disease group (p0.05). Correlation analysis indicated that Lactobacillus was positively correlated with Bifidobacteria (r=0.44, p=0.000), whereas was negatively correlated with Enterobacter (r=-0.22, p=0.024). Furthermore, Enterobacter was negatively associated with Bifidobacteria (r=-0.15, p=0.038) and Streptococcus (r=-0.12, p=0.046).Serum inflammatory factors, adiponectin, intestinal flora and immunity may play important roles in the development of NSCLC.

16. Remifentanil protects neurological function of rats with cerebral ischemia-reperfusion injury via NR2B/CaMKIIα signaling pathway

Abstract

The effects of remifentanil were studied on cerebral ischemia-reperfusion injury (IRI) in rats, and its regulatory effect was explored on the N-methyl D-aspartate receptor subtype 2B (NR2B)/calcium/calmodulin-dependent protein kinase type II subunit alpha (CaMKII) signaling pathway in cerebral tissues. A total of 40 Sprague-Dawley rats were randomly assigned into sham group, model group, low-dose group (remifentanil injected into the caudal veins at 2 μg/kg) and high-dose group (remifentanil injected into the caudal veins at 10 μg/kg). Then, in the model, low-dose and high-dose groups, the rat model of cerebral IRI was established through middle cerebral artery occlusion (namely, ischemia for 1 h and reperfusion for 2 h), while no thread was inserted into the rats in the sham group. Neurological function of rats in each group was evaluated, and the cerebral infarct size was measured using 2,3,5-triphenyltetrazolium chloride (TTC) staining. Moreover, terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) staining was performed to determine the neuronal apoptosis level, and mitochondrial membrane potential was measured via JC-1 assay. Moreover, the reactive oxygen species (ROS) level in neurons and the malondialdehyde (MDA) content in cerebral tissues were determined using 2',7'-dichlorofluorescin diacetate (DCFH-DA) assay and enzyme-linked immunosorbent assay (ELISA), respectively. Finally, the expression levels of apoptosis-associated proteins and the NR2B/CaMKIIα signaling pathway-associated proteins in cerebral tissues were measured by Western blotting. Remifentanil preconditioning substantially decreased the neurological score of rats (p0.01), cerebral infarct size (p0.01), neuronal apoptosis level (p0.01), ROS level in neurons (p0.01), MDA content (p0.01) and expression levels of cysteinyl aspartate specific proteinase-3 (Caspase-3), NR2B, phosphorylated CaMKIIα (p-CaMKIIα) and p-cAMP responsive element binding protein (p-CREB) (p0.01), but it increased the mitochondrial membrane potential (p0.01) and B-cell lymphoma 2 (Bcl-2)/Bcl-2-associated X protein (Bax) ratio (p0.01). Remifentanil can repress the NR2B/ CaMKIIα signaling pathway in the neurons of rats with cerebral IRI to decrease the p-CREB expression, ROS level and MDA content in neurons, neuronal apoptosis level and cerebral infarct size, and increase the mitochondrial membrane potential, thereby protecting the neurological function.

Abstract

To investigate whether human umbilical cord mesenchymal stem cells (UMSCs) derived exosomes (exosome) can repair the heart after myocardial infarction (MI) by delivering circ-0001273 and its mechanism.Through the Sprague Dawley (SD) rat MI model was established, at the same time, we designed si-circ-0001273. Phosphate-buffered saline (PBS), exosome and si-circ-0001273-exosome were transplanted into ischemic hearts of rat, respectively. Through the echocardiography, hematoxylin-eosin staining (HE) method to detect the rat heart recovery. Meanwhile, H9c2 was treated with hypoxic serum-free serum to construct an in vitro apoptosis model to further explore the effect of circ-0001273 on myocardial cell apoptosis.Compared with the exosome-treated group, the left ventricular ejection fraction (EF) and shortened fraction (FS) of the rat heart was remarkably reduced and the cardiac structure was more disordered in the si-circ-0001273-exosome-treated group. Meanwhile, in vitro TUNEL staining and flow cytometry detection, results showed that compared with the exosome co-culture group, the incidence of H9C2 cell apoptosis in the si-circ-0001273-exosome co-culture group was obviously increased.Circ-0001273 can remarkably inhibit the occurrence of myocardial cell apoptosis in ischemic environment, promote MI repair, and provide a good reference for clinical treatment.

18. Clinical features and survival impact of EBV-positive diffuse large B-Cell lymphoma with different age cutoffs

Abstract

In 2016 WHO classification, EBV +DLBCL of the elderly was replaced by EBV+ DLBCL NOS. This is due to the fact that many young patients of EBV+ DLBCL were found in recent years.In this study, we retrospectively analyzed clinical features and survival outcomes of EBV positive DLBCL patients in different age groups. All the patients treated at a single center.When we use different ages (40, 50 and 60 years old) as cutoffs, the prevalence of EBV positive DLBCL was 12.0%, 12.3% and 13.0% in younger patients and 19.0%, 15.4% and 13.8% in elder patients respectively. Whatever the age cutoff was, EBV positive associated with unfavorable clinical prognosis in elder groups. When we use 40 and 50 years old as age cutoffs, poor impacts of EBV positive on overall survival and progression-free survival were observed only in elder patients, but not in younger patients. It should be noted that when we use 60 years old as age cutoff, the results were the opposite.EBV+ DLBCL patients with age of 40 to 60 years old showed poorer prognostic features than EBV- DLBCL patients; however, patients in other age groups did not show evident differences in prognosis between EBV+ DLBCL patients and EBV- DLBCL patients. This finding was not reported before.

The aim of this study was to explore the effect of micro ribonucleic acid (miR)-126 on the apoptosis of retinal ganglion cells in glaucoma rats via the vascular endothelial growth factor (VEGF)-Notch signaling pathway.A total of 36 Sprague-Dawley (SD) rats were randomly divided into three groups, including normal group (n=12), model group (n=12) and miR-126 antagomir group (n=12). Rats in normal group did not receive any treatment. In model group and miR-126 antagomir group, the rats were used to establish glaucoma models and intervened with normal saline and miR-126 antagomir, respectively. Intraocular pressure was detected at the completion of modeling and the last intervention, at 7 days after which samples were taken. Western blotting was adopted to detect the relative protein expressions of Notch1 and Notch2. The content of VEGF was examined via enzyme-linked immunosorbent assay (ELISA). Quantitative polymerase chain reaction (qPCR) was carried out to detect the messenger RNA (mRNA) expressions of VEGF, Notch1 and Notch2. Terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay was performed to detect cell apoptosis.After modeling, intraocular pressure in model group and miR-126 antagomir group was significantly higher than that in normal group (p0.05). At the end of the intervention, intraocular pressure in miR-126 antagomir group was notably lower than that in model group (p0.05). VEGF content in model group and miR-126 antagomir group was notably higher than that in normal group (p0.05). However, it was markedly lower in miR-126 antagomir group than model group (p0.05). Model group exhibited remarkably higher protein expressions of Notch1 and Notch2 than normal group (p0.05). However, the protein expressions of Notch1 and Notch2 in miR-126 antagomir group were evidently reduced (p0.05). Besides, the mRNA expressions of VEGF, Notch1 and Notch2 in model group were significantly higher than those in normal group (p0.05). However, they were significantly lower in miR-126 antagomir group than those in model group (p0.05). Furthermore, the apoptosis rate in model group was distinctly higher than that in normal group (p0.05). However, it was notably lower in miR-126 antagomir group than model group (p0.05).MiR-126 facilitates the apoptosis of retinal ganglion cells in glaucoma rats by promoting the VEGF-Notch signaling pathway.

Abstract

The aim of this study was to investigate the effect of leptin (Lep) on the proliferation, invasion and apoptosis of prostate cancer cells through the extracellular regulated protein kinase 1/2 (ERK1/2) signaling pathway.Prostate cancer DU145 cells in the logarithmic growth phase were randomly divided into Lep (10, 20, 40, 80, 160 and 320 ng/mL) groups and blank control (Con) group. After culture, the cells were treated for 6 h, 12 h and 24 h, respectively. The effects of Lep on the proliferation and invasion of DU145 cells were detected via methyl thiazolyl tetrazolium (MTT) assay and transwell chamber assay, respectively. Quantitative reverse transcription-polymerase chain reaction (qRT-PCR) was carried out to examine the messenger ribonucleic acid (mRNA) expressions of ERK1/2, b-cell lymphoma 2 (Bcl-2) and Bcl-2-associated X protein (Bax) in DU145 cells after Lep treatment for 24 h. Thereafter, immunofluorescence assay was performed to detect the localization of ERK1/2 protein in prostate cancer DU145 cells. In addition, the expressions of phosphorylated (p)-ERK, ERK1/2 and apoptosis-related proteins, Bcl-2, Bax and cleaved cysteinyl aspartate specific proteinase (c-Caspase 3) in prostate cancer DU145 cells after treatment with different concentrations of Lep for 24 h were examined by Western blotting.MTT assay results showed that the proliferation rate of DU145 cells increased significantly at 6 h, 12 h and 24 h after 5-320 ng/mL of Lep treatment (p0.05). Transwell assay manifested that the number of invasive cells was significantly raised after Lep treatment for 24 h (p0.05). Meanwhile, the invasion ability of cells increased gradually with the elevation of Lep concentration. Subsequent qRT-PCR results demonstrated that after treatment with different concentrations of Lep, the mRNA expressions of ERK1/2 and Bcl-2 rose markedly (p0.05). However, the mRNA expression of Bax was remarkably down-regulated (p0.05) with the increase of Lep concentration in a concentration-dependent manner. According to the detection using a laser scanning confocal microscope, ERK1/2 red fluorescence showed punctiform aggregation, which was gradually raised with the increase of Lep concentration for 24 h. Moreover, Western blotting results denoted that with the increase of Lep concentration, the protein expressions of p-ERK, ERK1/2 and Bcl-2 were notably elevated (p0.05), while those of Bax and c-Caspase 3 were distinctly reduced (p0.05).Lep activation induces the proliferation, promotes the invasion and inhibits the apoptosis of prostate cancer cells through the ERK1/2 signaling pathway.

21. MiR-200c inhibits proliferation and promotes apoptosis of Wilms tumor cells by regulating akt signaling pathway

Abstract

The aim of this study was to explore the effect of micro ribonucleic acid (miR)-200c on the proliferation and apoptosis of Wilms tumor cells, and to further elucidate its potential mechanisms.Reverse Transcription-Polymerase Chain Reaction (RT-PCR) was used to detect the expression level of miR-200c in cancer tissues and adjacent normal tissues of 20 patients with Wilms tumor. Human primary Wilms tumor cells were taken as research objects, and were divided into Control group and miR-200c mimic group. In miR-200c mimic group, miR-200c was overexpressed in Wilms tumor cells using liposome transfection technology. Subsequently, the proliferation and apoptosis of cells in each group were observed by functional assays. The expression levels of phosphorylated protein kinase B (p-Akt), total Akt (T-Akt) and glucose transporter protein type 1 (GLUT1) in each group of cells were finally detected by Western blotting.In Wilms tumor patients, the expression level of miR-200c in cancer tissues was notably lower than that in adjacent normal tissues (p0.05). Wilms tumor cells were cultured in vitro, and were transfected with miR-200c mimic. Subsequent cell counting kit-8 (CCK-8) assay results showed that the proliferation ability of cells in miR-200c mimic group was remarkably weakened (p0.05). Colony formation assay indicated the number of formed colonies in miR-200c mimic group was remarkably less than that in Control group (p0.05). Western blotting results manifested that overexpression of miR-200c markedly increased the ratio of B-cell lymphoma protein 2 (Bcl-2) to Bcl-2-associated X protein (Bax) (p0.05). Flow cytometry results revealed that miR-200c overexpression significantly elevated the apoptosis rate of Wilms tumor cells (p0.05). In addition, it was discovered that the overexpression of miR-200c could prominently reduce the phosphorylation level of intracellular Akt and the expression of its downstream protein GLUT1. Finally, immunohistochemical staining results verified that the expression levels of p-Akt and GLUT1 in cancer tissues of Wilms tumor patients were significantly higher than those in adjacent normal tissues (p0.05).MiR-200c was lowly expressed in cancer tissues of Wilms tumor patients. Besides, overexpression of miR-200c inhibited the proliferation and promoted the apoptosis of cells through targeted inhibition of the Akt/GLUT1 signaling pathway.

Abstract

We aimed at studying the correlation between TGIF2 expression and clinicopathological features of cervical cancer (CCa). The relationship between TGIF2 and FCMR and its influence on the proliferation and metastasis of tumor cells were investigated using molecular biology techniques, so as to reveal the pathogenesis of CCa and provide a new target for clinical treatment.TGIF2 expression in 60 pairs of cervical tumors and paracancerous tissues samples collected from CCa patients of our hospital was studied by quantitative real-time polymerase chain reaction (qPCR) analysis, and the association between TGIF2 expression and the clinical indicators or prognosis of CCa patients were analyzed. CCa cells with TGIF2 knockdown were constructed using transfection technology. Changes in the biological phenotypes (proliferation, migration, invasion) of CCa cells C33-A and HeLa after TGIF2 knockdown were determined by Cell Counting Kit-8 (CCK-8) and transwell assays. In addition, the effects of TGIF2/FCMR axis on CCa metastasis were further explored in nude mice in vivo.Our data revealed a significant increase in TGIF2 mRNA expression in CCa tissue specimens compared to adjacent ones, and the increasing degree was positively correlated with the incidence of lymph node or distant metastasis of CCa patients. The results of CCK-8 and transwell suggested that knocking down TGIF2 effectively attenuated the proliferative ability and invasiveness of CCa cells. Luciferase assay confirmed that TGIF2 can directly bind to the DNA promoter of its target gene FCMR. Simultaneous transfection of sh-TGIF2 and sh-FCMR partially reversed the inhibitory effect of single transfection of TGIF2 knockdown on the malignant progression of CCa. Experiments in nude mice also suggested that TGIF2 could promote CCa tumorigenesis through the modulation of FCMR expression.In summary, TGIF2 can promote the migration and proliferation ability of cervical cancer cells via down-regulating FCMR. Our study provides a new therapeutic target for the clinical treatment of cervical cancer.

Abstract

To study the effect of strontium ranelate (SR) on steroid-induced osteonecrosis of the femoral head (SIONFH) in rabbits and its regulatory mechanism.The ONFH model was established in 30 rabbits using steroid and they were randomly divided into Control group, Model group, and SR group. After SR intervention, the rabbits were sacrificed and sampled. The pathological injury of the femoral head in each group was detected via hematoxylin-eosin (HE) staining, the level of vascular endothelial growth factor (VEGF) in the femoral head in each group was detected via enzyme-linked immunosorbent assay (ELISA). The messenger ribonucleic acid (mRNA) and protein expression levels of transforming growth factor-β1 (TGF-β1), as well as the bone morphogenetic protein 2 (BMP2) in the femoral head in each group, were determined using Reverse Transcription-Polymerase Chain Reaction (RT-PCR) and Western blotting.The rabbit model of SIONFH was successfully established. Compared with Control group, the Model group had a severer pathological injury of the femoral head, a lower level of VEGF in the femoral head, significantly decreased mRNA and protein levels of TGF-β1 and BMP2. Compared with Model group, the SR group had markedly improved pathological injury of the femoral head, a higher level of VEGF in the femoral head, significantly increased mRNA and protein levels of TGF-β1, as well as BMP2.SR can remarkably improve the pathological injury of the femoral head and increase the expression of VEGF in SIONFH rabbits, whose potential mechanism may be related to the activation of the TGF-β1/BMP2 signaling pathway.

To investigate the relationship between urate iransporter-1 gene polymorphism and the uric acid-lowering effect of losartan in hypertensive patients complicated with hyperuricemia, polymerase chain reaction-restriction fragment length polymorphism was used to identify the genotypes of 100 hypertensive patients with hyperuricemia and 100 healthy controls. The experimental group patients received oral administration of 100 mg losartan for 14 d. Blood pressure, uric acid, creatinine, blood urea nitrogen, triglyceride, total cholesterol, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, small and dense low-density lipoprotein cholesterol and fasting plasma glucose were measured before and after treatment. There was no significant difference in the frequency distribution of alleles and genotypes of rs7929627, rs75786299 and rs3825017 between the two groups. Abstract

A new type of bar-cropping process, termed the pre-shearing cropping process, which can improve billet cross-section quality has been proposed in this study. The new cropping process was realized by using a servo-controlled press. A low-carbon steel 20Cr as-received and a medium-carbon steel SCM435 as-annealed were used to test the process. A group of pre-shearing cropping experiments was carried out with different pre-shearing values of 6.67, 13.33, 20.00, and 26.67% under a pre-shearing speed of 10 mm/s and a cropping speed of 600 mm/s, and conventional cropping experiments with different cropping speeds of 200, 400, and 600 mm/s were conducted for comparison. The quality of the cropped billets was evaluated quantitatively using the indices of surface flatness, ovality, and incline angle. The effect of cropping speed on the conventional process and the effect of pre-shearing on the pre-shear cropping process were investigated in detail. The experimental results showed that the pre-shearing cropping process could improve the quality of the cropped billets and reduce the maximum cropping load slightly compared with the conventional cropping process. Finally, the mechanism of the pre-shearing cropping process was furtherly analyzed by finite element simulation and tried to be summarized.

Abstract

Dihadron azimuthal correlations containing a high transverse momentum ( ) trigger particle are sensitive to the properties of the nuclear medium created at RHIC through the strong interactions occurring between the traversing parton and the medium, i.e. jet-quenching. Previous measurements revealed a strong modification to dihadron azimuthal correlations in Au+Au collisions with respect to p+p and d+Au collisions. The modification increases with the collision centrality, suggesting a path-length or energy density dependence to the jet-quenching effect. This paper reports STAR measurements of dihadron azimuthal correlations in mid-central (20%-60%) Au+Au collisions at GeV as a function of the trigger particle's azimuthal angle relative to the event plane, . The azimuthal correlation is studied as a function of both the trigger and associated particle . The subtractions of the combinatorial background and anisotropic flow, assuming Zero Yield At Minimum (ZYAM), are described. The correlation results are first discussed with subtraction of the even harmonic (elliptic and quadrangular) flow backgrounds. The away-side correlation is strongly modified, and the modification varies with , with a double-peak structure for out-of-plane trigger particles. The near-side ridge (long range pseudo-rapidity correlation) appears to drop with increasing while the jet-like component remains approximately constant. The correlation functions are further studied with the subtraction of odd harmonic triangular flow background arising from fluctuations. It is found that the triangular flow, while responsible for the majority of the amplitudes, is not sufficient to explain the -dependence of the ridge or the away-side double-peak structure. The dropping ridge with could be attributed to a -dependent elliptic anisotropy; however, the physics mechanism of the ridge remains an open question. Even with a -dependent elliptic flow, the away-side correlation structure is robust. These results, with extensive systematic studies of the dihadron correlations as a function of , trigger and associated particle , and the pseudo-rapidity range , should provide stringent inputs to help understand the underlying physics mechanisms of jet-medium interactions in high energy nuclear collisions.

Abstract

This paper proposes an automatic approach for residential areas revision by means of analysing the correlation between the residential area and the topographic and geographical factors. The approach consists of four major steps: the extracting of missing residential area from the remote sensing images with high resolution; the statistic analysing on the size changes of missing residential area in each grade of the elevation, slope, distance from the road and other impact factors; modelling of residential area modification in the urban and rural region; testing the methods using 100 counties data which are located in the middle part of China North-South Seismic Belt and comparing the result to the Land Use in map scale 1:100000. The experimental results present the accuracy of urban residents by 70% increased to 89.4%, rural residents by 47% up to 81.9%, rural residents from 8% increased to 78.5%. Therefore, there is available risk exposure information in a sparsely populated area because the spatial grid distributions of population and buildings are based on the residential areas. The proposed approach in this paper will improve the accuracy of the seismic risk assessment if it is applied to the national or the whole world.

Abstract

Hybrid Au-nanoparticles (NPs)/β-Ga2O3 thin films were fabricated by laser molecular beam epitaxy of high quality β-Ga2O3 thin films on c-plane sapphire substrates followed by spin coating and post annealing of Au NPs. Photodetectors (PDs) made of hybrid Au-NPs/β-Ga2O3 thin films and bare β-Ga2O3 thin films are comparatively studied. It is found that Au-NPs can obviously improve the performances of PDs. Lower dark current, higher photoresponse and faster switching time under 254 nm light illumination are obtained by introducing Au-NPs. The localized surface plasmonic resonance from Au-NPs could be attributed to the enhanced performance. Meanwhile, a photoelectric response to 532 nm light illumination is observed in an Au NPs/β-Ga2O3 based PD. It is suggested that our hybrid Au-NPs/β-Ga2O3 PD could serve as a dual-band detector in one device.

Abstract

One of the primary goals of nuclear physics is to understand the force between nucleons, which is a necessary step for understanding the structure of nuclei and how nuclei interact with each other. Rutherford discovered the atomic nucleus in 1911, and the large body of knowledge about the nuclear force since acquired was derived from studies made on nucleons or nuclei. Although antinuclei up to antihelium-4 have been discovered and their masses measured, we have no direct knowledge of the nuclear force between antinucleons. Here, we study antiproton pair correlations among data taken by the STAR experiment at the Relativistic Heavy Ion Collider and show that the force between two antiprotons is attractive. In addition, we report two key parameters that characterize the corresponding strong interaction: namely, the scattering length (f0) and effective range (d0). As direct information on the interaction between two antiprotons, one of the simplest systems of antinucleons, our result provides a fundamental ingredient for understanding the structure of more complex antinuclei and their properties., 25 pages, 4 figures. 32. Mechanical Characteristics Generated by Solidification Crack During High Strength Steel Welding

Abstract

A multiscale method was developed to elaborate weld solidification crack mechanical field. A sequential thermo-mechanical finite element method (FEM) of butt-joint was performed to provide both of geometry and loading condition of submoel. Then mechanical behaviours generated by impurity was achieved by submodelling with embedded solidification cracks in terms of elasto-plastic FEM. The solidification initial cracks were geometrical discontinuity in nature and modelled as mechanical property discontinuity. Results show that additive stress and back stress are built-up at impurity-weld matrix interface. The developed modelling method was validated by solidification crack tests, X-ray tests together with fractography. The underlying mechanisms in suppression solidification cracking by adjustment of alloy contents in flux cored wire aiming to diminish the impurity size and improve the ductility of weld metal were revealed by developed method. Results show that, given a 9 percent increase of the weld metal ductility, a 29.6 percent decrease of the additive stress is achieved; and the longitudinal oriented impurity readily propagates crossing section rather than along impurity longitudinal tip. The susceptibility to cracking is distance-dependent, heavily on fields near to impurity and little on far field.

However, while the rapid development of cross-border mergers and acquisitions, many Chinese enterprises encountered lots of setbacks and failures, which are closely related to the lack of experience of the cross-border mergers and acquisitions. This paper is intended to analyze cross-border mergers and acquisitions, finds out the problems for the Chinese companies in the process of cross-border M & A, and finally proposes some recommendations accordingly.

Abstract

Materials science and engineering is one of the hot research topics in the world, among which mechanical properties of materials play a critical role in application of the new materials. Based on this, a special session Mechanical Properties of Materials was held within the 2nd Global Conference on Materials Science and Engineering, Nov. 20-22, 2013. This special issue contains a selection of twenty scientific papers, which are focused on the structure, mechanical properties, and strength of materials. In this review, the selected papers from the special session are summarized. Introduction. The Global Conference on Materials Science and Engineering (CMSE) is an annual event since 2012. The CMSE 2013 was held by Hubei University of Science and Technology in the City of Xianning (Hubei, China), November 20-22, 2013. Within this event, a special session named Mechanical Properties of Materials was held. In this session, 31 papers were selected for publication, of them 20 papers are published in this special issue (1-20), two more (21, 22) in the next issue, and 9 papers are published by Materials Research Innovations (23-31). All papers underwent the standard peer-review process of the journal and were accepted for publication based on the evaluation of at least two independent anonymous reviewers. Highlighted Papers. Results of the study (1) indicate that crack tip stress distribution characteristics and crack propagation dynamics are closely related to the microstructure evolution caused by the change of a strain rate and temperature. At a lower strain rate and temperature, the crack propagated by a brittle mechanism without the change in atomic configuration near the crack tip. The stress concentration occurred at the crack tip of a growing crack. The crack propagation exhibited a gradual brittle-to-ductile transition with increasing temperature and strain rate, the peak stress was accompanied by the appearance of the microstructure evolution ahead of the crack tip. In (3), uniformly distributed TiC nanoparticle-reinforced iron-based composites were successfully prepared by planetary milling in argon and subsequent hot pressing. Nearly full density composite specimens could be obtained via milling for 6 h followed by hot pressing at 1100C under 50 MPa. Comparatively spherical TiC particles and fine fibrous Fe3C phases were observed in the iron matrix composite. Microstructural analysis results show that the average diameter of TiC particles and the length of Fe3C phases tend to decrease with an increase in the TiC volume content. Her and Wu (7) studied the annealing effect on the microstructure and mechanical properties of titanium nitride (TiN) films. Abstract

Polycomb protein histone methyltransferase enhancer of Zeste homologe 2 (EZH2) is frequently overexpressed in human malignancy and is implicated in cancer cell proliferation and invasion. However, it is largely unknown whether EZH2 has a role in modulating DNA damage response. Here, we show that EZH2 is an important determinant of cell fate decision in response to genotoxic stress. EZH2 depletion results in abrogation of both cell cycle G1 and G2/M checkpoints, directing DNA damage response toward predominant apoptosis in both p53-proficient and p53-deficient cancer cells, but not in normal cells. Mechanistically, EZH2 regulates DNA damage response in p53 wild-type cells mainly through transcriptional repression of FBXO32, which binds to and directs p21 for proteasome-mediated degradation, whereas it affects p53-deficient cells through regulating Chk1 activation by a distinct mechanism. Furthermore, pharmacological depletion of EZH2 phenocopies the effects of EZH2 knockdown on cell cycle checkpoints and apoptosis. These data unravel a crucial role of EZH2 in determining the cancer cell outcome following DNA damage and suggest that therapeutic targeting oncogenic EZH2 might serve as a strategy for improving conventional chemotherapy in a given malignancy.

Abstract

Deregulation of the pRB/E2F pathway, which occurs frequently in human malignancy, is often associated with inappropriate proliferation and/or apoptosis. While the role of E2F1 in apoptosis induction has been well-established, it remains unclear how this pro-apoptotic activity is regulated in cancer. Here we describe EZH2, an oncogenic polycomb histone methyltransferase and an E2F1 target, as an important regulator of E2F1-dependent apoptosis. We show that E2F1 induces EZH2 expression, which in turn antagonizes the induction of E2F1 pro-apoptotic target Bim expression. RNAi-mediated gene depletion of EZH2 enhances E2F1-dependent Bim expression, thereby promoting the pro-apoptotic activity of E2F1. Hence, the concomitant induction of EZH2 and Bim by E2F1 constitutes a fail-safe mechanism to allow tumor cells with aberrant E2F1 activity to evade apoptosis. These findings reveal a novel mechanism by which the apoptotic activity of E2F1 is restrained in human cancer and also provide the first evidence that EZH2 directly regulates apoptotic process in cancer cells.

Abstract

Dihadron correlations are analyzed in sNN=200 GeV d+Au collisions classified by forward charged particle multiplicity and zero-degree neutral energy in the Au-beam direction. It is found that the jetlike correlated yield increases with the event multiplicity. After taking into account this dependence, the non-jet contribution on the away side is minimal, leaving little room for a back-to-back ridge in these collisions.

Abstract

We present ΛΛ correlation measurements in heavy-ion collisions for Au+Au collisions at sqrt[s_{NN}]=200 GeV using the STAR experiment at the Relativistic Heavy-Ion Collider. The Lednický-Lyuboshitz analytical model has been used to fit the data to obtain a source size, a scattering length and an effective range. Implications of the measurement of the ΛΛ correlation function and interaction parameters for dihyperon searches are discussed.

Abstract

The emergence of drug-resistant strains of Mycobacterium tuberculosis is a serious public health problem. Many of the specific gene mutations that cause drug resistance in M. tuberculosis are point mutations. We are developing a PCR-peptide nucleic acid (PNA)-based ELISA as a diagnostic method to recognize point mutations in genes associated with isoniazid and rifampin resistance in M. tuberculosis. Specific point mutation-containing sequences and wild-type sequences of cloned mycobacterial genes were PCR-amplified, denatured, and hybridized with PNA probes bound to microplate wells. Using 15-base PNA probes, we established the hybridization temperatures (50°C–55°C) and other experimental conditions suitable for detecting clinically relevant point mutations in the katG and rpoB genes. Hybridization of PCR-amplified sequences that contained these point mutations with complementary mutation-specific PNAs resulted in significant increases in ELISA response compared with hybridization using wild-type-specific PNAs. Conversely, PCR-amplified wild-type sequences hybridized much more efficiently with wild-type PNAs than with the mutation-specific PNAs. Using the M. tuberculosis cloned genes and PCR-PNA-ELISA format developed here, M. tuberculosis sequences containing point mutations associated with drug resistance can be identified in less than 24 h.

Abstract

The capacity of UV-catalyzed Fenton oxidation to destroy mono-, di- and trinitrotoluenes (MNT, DNT, TNT) in aqueous solution was evaluated. Mononitrotoluenes were rapidly destroyed by Fenton oxidation in the dark at pH 3.0 or by UV/Fenton oxidation without pH adjustment. UV/Fenton oxidation at pH 3.0 was most effective for DNT and TNT destruction. The number and position of nitro substitutions influenced oxidation rate: 2-nitrotoluene > 4-nitrotoluene > 2,4-dinitrotoluene > 2,6-dinitrotoluene > 2,4,6-trinitrotoluene. UV/Fenton oxidation mineralized more than 95% of the TNT in aqueous extracts of contaminated soil. While dissolved humic and fulvic acids differentially influenced destruction rate, total TNT destroyed after 4 h was not greatly affected. Our results demonstrated that UV-catalyzed Fenton oxidation effectively destroyed nitrotoluenes in water and aqueous extracts of contaminated soil.

Abstract

We present measurements of three-dimensional correlation functions of like-sign, low-transverse-momentum kaon pairs from root sNN = 200 GeV Au + Au collisions. A Cartesian surface-spherical harmonic decomposition technique was used to extract the kaon source function. The latter was found to have a three-dimensional Gaussian shape and can be adequately reproduced by Therminator event-generator simulations with resonance contributions taken into account. Compared to the pion one, the kaon source function is generally narrower and does not have the long tail along the pair transverse momentum direction. The kaon Gaussian radii display a monotonic decrease with increasing transverse mass m(T) over the interval of 0.55

Abstract

Breast cancer remains a major health problem even with all the recent technological advancements. Large-scale gene expression analysis has offered great ease for both biological characterization and therapeutic planning of breast cancer. Previous studies mostly used variance/regression analysis, which becomes fundamentally flawed when there are unaccounted array specific factors. Here we aim to investigate the underlying mechanism of breast cancer through partial least squares (PLS) based gene expression profile analysis.With a gene expression profile data set downloaded from the Gene Expression Omnibus database, we performed PLS based analysis.We acquire 932 and 771 differentially expressed genes (DEGs) in breast cancer metastasis of estrogen-receptor (ER)-positive and ER-negative patients, respectively. For ER-positive patients, 32 pathways were found to be enriched with DEGs, including immune related pathways, cellular processes and environmental information processing pathways. Survival analysis demonstrated that 18 of them were closely related with non-recurrence rate along time after surgery. For ER negative patients, only three pathways including the folate biosynthesis pathway were enriched with DEGs and none of them overlapped with those of ER positive patients. Only the cholinergic synapse pathway was significantly associated with the non-recurrence rate according to the survival analysis.Our findings shed light on pathways involved in breast cancer relapse with the hope to give some theoretical supports for further therapeutic study.

Abstract

We report measurements of the third harmonic coefficient of the azimuthal anisotropy, upsilon(3), known as triangular flow. The analysis is for charged particles in Au + Au collisions at root s(NN) = 200 GeV, based on data from the STAR experiment at the BNL Relativistic Heavy Ion Collider. Two-particle correlations as a function of their pseudorapidity separation are fit with narrow and wide Gaussians. Measurements of triangular flow are extracted from the wide Gaussian, from two-particle cumulants with a pseudorapidity gap, and also from event plane analysis methods with a large pseudorapidity gap between the particles and the event plane. These results are reported as a function of transverse momentum and centrality. A large dependence on the pseudorapidity gap is found. Results are compared with other experiments and model calculations.

Abstract

Measurements of the elliptic flow, upsilon(2), of identified hadrons (pi(+/-), K-+/-, K-s(0), p, (p) over bar, phi, Lambda, (Lambda) over bar, Xi(-), (Xi) over bar (+), Omega(-), (Omega) over bar (+)) in Au + Au collisions at root s(NN) = 7.7, 11.5, 19.6, 27, 39, and 62.4 GeV are presented. The measurements were done at midrapidity using the time-projection chamber and the time-of-flight detectors of the Solenoidal Tracker at RHIC experiment during the beam-energy scan program at Relativistic Heavy Ion Collider. A significant difference in the upsilon(2) values for particles and the corresponding antiparticles was observed at all transverse momenta for the first time. The difference increases with decreasing center-of-mass energy, root s(NN) (or increasing baryon chemical potential, mu(B)), and is larger for the baryons as compared to the mesons. This implies that particles and antiparticles are no longer consistent with the universal number-of-constituent quark (NCQ) scaling of upsilon(2) that was observed at root s(NN) = 200 GeV. However, for the selected group of particles (p(+), K+, K-s(0), p, Lambda, Xi(-), Omega(-)) NCQ scaling at (m(T) - m(0))/n(q) > 0.4 GeV/c(2) is not violated within +/- 10%. The upsilon(2) values for f mesons at 7.7 and 11.5 GeV are approximately two standard deviations from the trend defined by the other hadrons at the highest measured p(T) values.

Abstract

We present a study of the average transverse momentum (${p}_{t}$) fluctuations and ${p}_{t}$ correlations for charged particles produced in Cu+Cu collisions at midrapidity for $\sqrt{{s}_{NN}}=$ 62.4 and 200 GeV. These results are compared with those published for Au+Au collisions at the same energies, to explore the system size dependence. In addition to the collision energy and system size dependence, the ${p}_{t}$ correlation results have been studied as functions of the collision centralities, the ranges in ${p}_{t}$, the pseudorapidity $\ensuremath{\eta}$, and the azimuthal angle $\ensuremath{\phi}$. The square root of the measured ${p}_{t}$ correlations when scaled by mean ${p}_{t}$ is found to be independent of both colliding beam energy and system size studied. Transport-based model calculations are found to have a better quantitative agreement with the measurements compared to models which incorporate only jetlike correlations.

Abstract

Jet-medium interactions are studied via a multihadron correlation technique ( called "2 + 1"), where a pair of back-to-back hadron triggers with large transverse momentum is used as a proxy for a di-jet. This work extends the previous analysis for nearly symmetric trigger pairs with the highest momentum threshold of trigger hadron of 5 GeV/c with the new calorimeter-based triggers with energy thresholds of up to 10 GeV and above. The distributions of associated hadrons are studied in terms of correlation shapes and per-trigger yields on each trigger side. In contrast with di-hadron correlation results with single triggers, the associated hadron distributions for back-to-back triggers from central Au + Au data at root S-NN = 200 GeV show no strong modifications compared to d + Au data at the same energy. An imbalance in the total transverse momentum between hadrons attributed to the near-side and away-side of jetlike peaks is observed. The relative imbalance in the Au + Au measurement with respect to d + Au reference is found to increase with the asymmetry of the trigger pair, consistent with the expectation from medium-induced energy-loss effects. In addition, this relative total transverse momentum imbalance is found to decrease for softer associated hadrons. Such evolution indicates that the energy missing at higher associated momenta is converted into softer hadrons. DOI:10.1103/PhysRevC.87.044903

Abstract

Direct PCR detection of bacteria in clinical samples is often hindered by the presence of compounds that inhibit the PCR. To improve and accelerate the diagnosis of Mycobacterium avium-M. intracellulare complex infections, an immunomagnetic PCR (IM-PCR) assay was developed. This IM-PCR procedure combines the separation of mycobacteria by antimycobacterial monoclonal antibody coupled to magnetic beads with an M. avium-M. intracellulare complex-specific PCR protocol based on 16S rRNA gene sequences. As few as 10 M. avium bacilli were detected in spiked human stool samples, a clinical specimen usually refractory to conventional PCR analysis, by the IM-PCR method. Moreover, M. avium organisms were detected in about 24 h in 18 of 22 culture-confirmed fecal samples from AIDS patients. This IM-PCR protocol should allow for the rapid and sensitive detection of M. avium isolates in clinical specimens.
