This study assessed the presence of potentially toxic elements (PTEs) in China's northeastern black soil belt, an area with limited prior research. We collected 304 soil samples (0–20 cm) from Gonghe Town, Hailun City, and analyzed the PTE contamination degree using the single-factor pollution index and Nemerow pollution index. The results demonstrated that the mean concentrations of arsenic (As), cadmium (Cd), chromium (Cr), copper (Cu), mercury (Hg), nickel (Ni), lead (Pb), and zinc (Zn) were 11.16, 0.11, 65.29, 22.56, 0.03, 27.07, 26.09, and 66.01 mg/kg, respectively. Source apportionment was conducted via correlation analysis, principal component analysis, and positive matrix factorization, identifying four main sources: natural (33.2%), irrigation (29.5%), fuel (23.4%), and fertilizer (13.2%). The ecological risk index indicated a slight ecological risk, while the human health risk showed that non-carcinogenic risks were negligible and carcinogenic risks were acceptable. Our findings emphasize the need to prioritize controlling PTEs from fertilizer, particularly cadmium, and to a lesser extent, irrigation and fuel sources, focusing on As, Pband Hg. Remote sensing provides us with an approach for the rapid identification and monitoring of spatiotemporal changes in the urban ecological environment at different scales. This study aimed to construct a remote sensing assessment index for urban ecological livability with continuous fine spatiotemporal resolution data from Landsat and MODIS to overcome the dilemma of single image-based, single-factor analysis, due to the limitations of atmospheric conditions or the revisit period of satellite platforms. The proposed Ecological Livability Index (ELI) covers five primary ecological indicators – greenness, temperature, dryness, water-wetness, and atmospheric turbidity – which are geometrically aggregated by non-equal weights based on an entropy method. Considering multisource time-series data of each indicator, the ELI can quickly and comprehensively reflect the characteristics of the Ecological Livability Quality (ELQ) and is also comparable at different time scales. Based on the proposed ELI, the urban ecological livability in the central urban area of Wuhan, China, from 2002 to 2017, in the different seasons was analyzed every 5 years. The ELQ of Wuhan was found to be generally at the medium level (ELI ≈0.6) and showed an initial trend of degradation but then improved. Moreover, the ecological livability in spring and autumn and near rivers and lakes was found to be better, whereas urban expansion has led to the outward ecological degradation of Wuhan, but urban afforestation has enhanced the environment. In general, this paper demonstrates that the ELI has an exemplary embodiment in urban ecological research, which will support urban ecological protection planning and construction. The accumulation of potentially toxic elements (PTEs) in agricultural soils is of particular concern in China, while its status, ecological risks, and human health hazards have been little studied in the permafrost areas of Northeast China. In this study, 75 agricultural soil samples (0–20 cm) were collected from the Arctic Village, Mo'he City, in the northernmost part of China. The average concentration (mean ± standard deviation) of As, Cd, Cr, Cu, Hg, Ni, Pb, and Zn were 12.11 ± 3.66 mg/kg, 0.11 ± 0.08 mg/kg, 52.50 ± 8.83 mg/kg, 12.08 ± 5.12 mg/kg, 0.05 ± 0.02 mg/kg, 14.90 ± 5.35 mg/kg, 22.38 ± 3.04 mg/kg, and 68.07 ± 22.71 mg/kg, respectively. Correlation analysis, cluster analysis, and principal component analysis indicated that As, Cu, Ni, and Zn likely originated from geogenic processes, Hg and Pb from long-range atmospheric transport, Cd from planting activities, and Cr from Holocene alluvium. The geo-accumulation index and enrichment factor showed that As, Cd, Hg, and Zn are enriched in soils. The Nemerow pollution index showed that 66.67%, 24%, and 1.33% of soil samples were in slight, moderate, and heavy pollution levels, respectively, with Hg being the most important element affecting the comprehensive pollution index. The potential ecological risk index showed that 48.00% and 1.33% of soil samples were in the moderate ecological risk and high potential ecological risk levels, respectively. The non-carcinogenic and carcinogenic human health risk index for adults and children were both less than 1, which was within the acceptable range. This study revealed the accumulation pattern of PTEs in agricultural soils of permafrost regions and provided a scientific basis for research on ecological security and human health. The consistency of geospatial data is of great significance for the application and updating of geographic information in web maps. Due to the multiple data sources and different temporal versions, the tile web maps usually meet the inconsistency question across different layers. This study tries to develop a method to detect this kind of inconsistency utilizing a raster-based scaling approach. Compared with vector-based handling, this method can be directly available for multi-level tile images in a pixel representation form. The proposed cross-layer raster tile map rendering method (CRTMRM) consists of four primary aspects: geographic object separation, consistency rendering rules, data scaling and derivation with super-pixel segmentation, and inconsistency detection. The scale transformation strategy with the super-pixel attempts to obtain a simplified representation. Taking the scale lifespan variation and geometric consistency rules into account, the inconsistency detection of tile maps is conducted between temporal versions, multi-sources, and different scales through actual and derived data overlay analysis. The experiment focuses on features of cross-layer water or vegetation areas with Level 9 to Level 14 in Baidu Maps, Amap, and Google Maps. Objective: Rheumatoid arthritis (RA), as a chronic autoimmune disorder, seriously threatens human health. However, no study has thoroughly illustrated the etiology of RA. The present work focused on investigating the biological functions of STAT6 and the upstream miRNAs that regulate its expression., Methods: Synovial tissues from rheumatoid arthritis (RA) patients and normal participants were acquired. Cell viability, proliferation, apoptosis, concentrations of cytokines, miRNA and protein levels, and relative luciferase activities were detected., Results: WB and qRT-PCR showed that STAT6 was obviously up-regulated in synovial tissues of RA patients as well as RA fibroblast-like synoviocytes (RA FLSs). Functionally, down-regulation of STAT6 significantly inhibited the growth of RA FLSs as indicated by EdU and CCK-8 assays. In addition, inhibition of STAT6 remarkably promoted apoptosis of RA FLSs. Besides, silence of STAT6 notably suppressed inflammatory cytokine levels, such as TNF-α, IL-6 and IL-1β. Mechanistically, STAT6 was predicted to be the direct target of and negatively regulated by miR-135a-5p. Moreover, STAT6 was involved in the regulation of miR-135a-5p on cell growth, apoptosis and inflammatory response of RA FLSs., Conclusion: miR-135a-5p/STAT6 is a potential novel therapeutic target for RA treatment., Competing Interests: None., (AJTR Copyright © 2022.)

Objectives: To explore the expression levels and the potential regulatory mechanism of miR-21-5p in LPS-treated H9c2 cells., Methods: The secretions of the inflammatory cytokines induced by LPS in H9c2 cells were evaluated using ELISA. We used RT-RCR and western blot to measure the relative mRNA and protein expression levels in LPS-treated H9c2 cells. CCK-8 and EdU assays showed the viability and proliferation profiles of the H9c2 cells. TUNEL assays demonstrated the apoptotic behaviors of the H9c2 cells, and a luciferase reporter analysis was used to investigate the interactions between miR-21-5p and programmed cell death protein 4 (PDCD4)., Results: LPS induced damage to the H9c2 cells by reducing the cell viability and down-regulating miR-21-5p. On the other hand, miR-21-5p overexpression inhibited the LPS-induced inflammatory damage in the H9c2 cells. Moreover, PDCD4 was verified as a downstream target gene of miR-21-5p, and its expression was inhibited by the higher miR-21-5p content. Finally, miR-21-5p inhibited septic processes, and the PDCD4 overexpression rescued the miR-21-5p effect in the LPS-treated H9c2 cells., Conclusion: Our findings suggest that miR-21-5p inhibits the LPS-induced progression of sepsis in H9c2 cells.
