Brassica oleracea is one of the important vegetable crops in China. Its seeds are primarily imported. Hence, developing innovative varieties belonging to China is an urgent requirement. Since broccoli is a cross-pollinated crop systematic selection and cross-breeding are challenging, and the application of radiation mutagenesis breeding in broccoli has not been reported. In this study, the seeds of Brassica oleracea were irradiated using carbon ion beams to investigate the biological effects of these beams on broccoli. The growth indices, antioxidant enzyme activity, photosynthetic indices, and chlorophyll fluorescence were detected at the seedling stage. The results revealed that irradiation with 100–500 Gy exhibited no significant effect on the germination of Brassica oleracea, while the germination of seeds was significantly inhibited at 600 Gy. After irradiation with 100–600 Gy, the root length, shoot length, seedling height, and leaf area decreased with the increase in irradiation dose. The Median lethal dose (LD50) of Brassica oleracea irradiated using a carbon ion beam was 415.89 Gy, and the dose that halved the root length was 495.12 Gy. After irradiation, the activities of superoxide dismutase (SOD) and peroxidase (POD) were higher than that of the control plants, while the activity of catalase (CAT) was lower than that of the control plants. The content of malondialdehyde (MDA) increased significantly at a dose of 400 Gy. The photosynthetic pigments (chlorophyll a, chlorophyll b, and carotenoids) showed an increasing trend initially, and then a decreasing trend with an increasing irradiation dose, and the highest value was detected at a dose of 300 Gy. Net photosynthesis, transpiration rate, and stomatal conductance exhibited a negative correlation with irradiation dose, while the non-photosynthetic quenching coefficient exhibited a significant increase after irradiation. The findings indicated that heavy ion beam radiation inhibited the growth of broccoli plants and affected the activity of antioxidant enzymes and photosynthesis. The study provided basic data for the radiation mutation breeding of Brassica oleracea.

Irreversible cardiomyopathy was caused by the therapeutic of anthracyclines in the chemotherapy of cancers. The cell apoptosis and autophagy were induced by anthracyclines in AC16 cells. MiR-223-3p ascends in anthracycline-treated AC16, but the expression of nuclear factor I-A (NFIA) was specifically down-regulated. However, the underlying molecular mechanism between NFIA and miR-223-3p is unclear now in AC16 cells. In our research, NFIA expression was dampened in AC16 cells by miR-223-3p mimics. Additionally, miR-223-3p knockdown hindered the apoptosis and autophagy in anthracycline-treated AC16. Furthermore, NFIA was predicted and verified as a miR-223-3p’s downstream target and rescued the functions of miR-223-3p. These findings illustrated that miR-223-3p advances anthracycline-stimulated cardiomyocyte damage progression by targeting NFIA, implying the promising therapeutic function of miR-223-3p on cardiomyocyte damage in cancer patients.

The most common subtype of lung cancer is non-small cell lung cancer (NSCLC) , accounting for 80% to 85% of lung cancer. During the development process, the tumor is easily metastasized to the brain, seriously affecting the treatment prognosis and quality of life of patients. At present, the main treatment options for brain metastasis of lung cancer are surgery, chemotherapy, radiotherapy, targeted therapy, and immunotherapy. In recent years, breakthroughs have been made in the application of immune checkpoint inhibitors, and some combination immunotherapies have been clinically approved. According to the Chinese Experts Consensus on Immune Checkpoint Inhibitors for Non-small Cell Lung Cancer, there have been certain studies showing that immunotherapy can be applied to the treatment of advanced NSCLC. Immunotherapy is like a double-edged sword, and its practical clinical significance still needs further exploration. This article reviews the progress of immunotherapy for brain metastasis of NSCLC and explores more reasonable clinical treatment plans for patients with brain metastasis of lung cancer.

Pairwise-based methods such as the free energy perturbation (FEP) method have been widely deployed to compute the binding free energy differences between two similar host-guest complexes. The calculated pairwise free energy difference is either directly adopted or transformed to absolute binding free energy for molecule rank ordering. We investigated, through both analytic derivations and simulations, how the selection of pairs in the experiment could impact the overall prediction precision. Our studies showed that (1) the estimated absolute binding free energy ( ΔG^ ) derived from calculated pairwise differences (ΔΔG) through weighted least squares fitting is more precise in prediction than the pairwise difference values when the number of pairs is more than the number of ligands and (2) prediction precision is influenced by both the total number of pairs and the specifically selected pairs, the latter being critically important when the number of calculated pairs is limited. Furthermore, we applied optimal experimental design in pair selection and found that the optimally selected pairs can outperform randomly selected pairs in prediction precision. In an illustrative example, we showed that, upon weighing ligand structure similarity into design optimization, the weighted optimal designs are more efficient than the literature reported designs. This work provides a new approach to assess retrospective pairwise-based prediction results, and a method to design new prospective pairwise-based experiments for molecular lead optimization. © 2019 Wiley Periodicals, Inc.

The study aimed to reveal the function of LXY30 peptide‐modified bone marrow mesenchymal stem cell‐derived exosomes (LXY30‐Exos) in NSCLC. LXY30 peptide is a peptide ligand targeting α3β1 integrin, and LXY30 specifically binds to Exos derived from different cells. We use transmission electron microscopy to identify LXY30‐Exos and tracking analysis for particles, and the LXY30‐Exos internalized by NSCLC cells in vitro and targeted NSCLC tumours in vivo were verified by multiple molecular technologies. The functions of LXY30‐Exos‐encapsulated miR‐30c, miR‐181b or miR‐613 were assessed using cell proliferation, migration and cell apoptosis assays. Meanwhile, the safety of the above engineered Exos was evaluated in vivo. After LXY30‐Exos were isolated and identified, LXY30‐Exos were confirmed to be internalized by NSCLC cells in vitro and specifically targeted NSCLC tumours in vivo. Functionally, LXY30‐Exos‐encapsulated miR‐30c, miR‐181b or miR‐613 weakened the proliferation, migration and cell cycle of NSCLC cells induced cellular apoptosis in vitro and restrained the tumour progression in vivo. Meanwhile, the safety of LXY30‐Exos‐encapsulated miR‐30c, miR‐181b or miR‐613 was confirmed in vivo. Overall, miR‐30c, miR‐181b and miR‐613 encapsulated in LXY30 peptide‐modified BMSC‐Exos relieved NSCLC. [ABSTRACT FROM AUTHOR]

The mechanisms governing gene regulation in domestic Yuzhong pigeon breast muscle development remain largely elusive. Here, we conducted a comparative analysis using Iso-seq and RNA-seq data from domestic Yuzhong pigeons and European meat pigeons to uncover signaling pathways and genes involved in breast muscle development. The Iso-seq data from domestic Yuzhong pigeons yielded 131,377,075 subreads, resulting in 16,587 non-redundant high-quality full-length transcripts post-correction. Furthermore, utilizing pfam, CPC, PLEK, and CPAT, we predicted 5575, 4973, 2333, and 4336 lncRNAs, respectively. Notably, several genes potentially implicated in breast muscle development were identified, including tropomyosin beta chain, myosin regulatory light chain 2, and myosin binding protein C. KEGG enrichment analysis revealed critical signaling pathways in breast muscle development, spanning carbon metabolism, biosynthesis of amino acids, glycolysis/gluconeogenesis, estrogen signaling, PI3K-AKT signaling, protein processing in the endoplasmic reticulum, oxidative phosphorylation, pentose phosphate pathway, fructose and mannose metabolism, and tight junctions. These findings offer insights into the biological processes driving breast muscle development in domestic Yuzhong pigeon, contributing to our understanding of this complex phenomenon.
