Bivalent chromatin is characterized by occupation of both activating and repressive histone modifications. Here, we develop a mathematical model that involves antagonistic histone modifications H3K4me3 and H3K27me3 to capture the key features of bivalent chromatin. Three necessary conditions for the emergence of bivalent chromatin are identified, including advantageous methylating activity over demethylating activity, frequent noise conversions of modifications, and sufficient nonlinearity. The first condition is further confirmed by analyzing the existing experimental data. Investigation of the composition of bivalent chromatin reveals that bivalent nucleosomes carrying both H3K4me3 and H3K27me3 account for no more than half of nucleosomes at the bivalent chromatin domain. We identify that bivalent chromatin not only allows transitions to multiple states but also serves as a stepping stone to facilitate a stepwise transition between repressive chromatin state and activating chromatin state and thus elucidate crucial roles of bivalent chromatin in mediating phenotypical plasticity during cell fate determination.

Surface glint significantly reduces the measurement accuracy of remote sensing reflectance of water, Rrs, making it difficult to effectively use field measurements for studying water optical properties, accurately retrieving water quality parameters, and validating satellite remote sensing products. To accurately assess the effectiveness of various glint removal methods and enhance the accuracy of water reflectance measurements, a portable multiprobe high-resolution System (PMHRS) is designed. The system is composed of a spectrometer, fiber bundles, an irradiance probe, and three radiance probes. The reliability and measurement accuracy of the PMHRS are ensured through rigorous laboratory radiometric calibration and temperature correction. The comprehensive uncertainty of laboratory calibration ranges from 1.29% to 1.43% for irradiance calibration and from 1.47% to 1.59% for radiance calibration. Field measurement results show a strong correlation with both synchronous ASD data, and Sen2Cor-atmospherically corrected Sentinel-2B data (R2 = 0.949, RMSE = 0.013; R2 = 0.926, RMSE = 0.0105). The water-leaving radiance measurements obtained under different solar elevation angles using three methods (M99 method, polarization method, and SBA) demonstrate that the improved narrow field-of-view polarization probe effectively removes surface glint across various solar elevation angles (with overall better performance than the traditional M99 method). At a solar elevation angle of 69.7°, the MAPD and MAD between the measurements of this method and those of the SBA are 5.8% and 1.4 × 10−4, respectively. The results demonstrate that the PMHRS system outperforms traditional methods in sun glint removal, significantly enhancing the accuracy of water remote sensing reflectance measurements and improving the validation quality of satellite data. This work provides a crucial technical foundation for the development of high-resolution continuous observation platforms in complex aquatic environments. It holds significant implications for improving the accuracy of field-based water remote sensing reflectance measurements and for enhancing the quality of water ecological monitoring data and satellite validation data. Anxiety disorder (AD) is a common disabling disease. The prolonged disease course may lead to impaired cognitive performance, brain function, and a bad prognosis. Few studies have examined the effect of disease course on brain function by electroencephalogram (EEG). Methods: Resting-state EEG analysis was performed in 34 AD patients. The 34 patients with AD were divided into two groups according to the duration of their illness: anxious state (AS) and generalized anxiety disorder (GAD). Then, EEG features, including univariate power spectral density (PSD), fuzzy entropy (FE), and multivariable functional connectivity (FC), were extracted and compared between AS and GAD. These features were evaluated by three previously validated machine learning methods to test the accuracy of classification in AS and GAD. Results: Significant decreased PSD and FE in GAD were detected compared with AS, especially in the Alpha 2 band. In addition, FC analysis indicated that GAD patients' connection between the left and right hemispheres decreased. Based on machine learning, AS and GAD are classified on a six-month criterion with the highest classification accuracy of up to 0.99 ± 0.0015. Conclusion: The brain function of patients is more severely impaired in AD patients with longer illness duration. Resting-state EEG demonstrated to be a promising examination in the classification in GAD and AS using machine learning methods with better classification accuracy. [ABSTRACT FROM AUTHOR]

Background: The semiactive or inactive probiotics or their extracts used in dermatology have interesting properties to ameliorate signs of irritated skin and enhance the skin barrier. Bifidobacterium, as the most common probiotics, which has been found to be effective in reducing acne and improving the skin barrier function of atopic dermatitis. Bifida Ferment Lysate (BFL) can be obtained from Bifidobacterium by fermentation and extraction. Purpose: In this study, we investigated the effect of a topically used BFL on the skin using in vitro evaluation methods. Results: The results showed that upregulation of skin physical barrier gene (FLG, LOR, IVL, TGM1, and AQP3) and antimicrobial peptide gene (CAMP and hBD‐2) in HaCaT cells by BFL might be responsible for skin barrier resistance. In addition, BFL had strong antioxidant properties representing a dose‐dependent increasing of the scavenging capacity of DPPH, ABTS, hydroxyl, and superoxide radicals. BFL treatment also fundamentally inhibited the intracellular ROS and MDA production and improved the activities of antioxidant enzymes (CAT and GSH‐Px) in H2O2‐stimulated HaCaT cells. As a good immunomodulatory factor, BFL efficiently decreased the secretion of IL‐8 and TNF‐α cytokines, and COX‐2 mRNA expression in LPS‐induced THP‐1 macrophages. Conclusion: BFL can strengthen the skin barrier function and stimulate skin barrier resistance, to reinforce the skin against oxidative stress and inflammatory stimuli. [ABSTRACT FROM AUTHOR]

Porcine viral diarrhea diseases affect the swine industry, resulting in significant economic losses. Porcine epidemic diarrhea virus (PEDV) genotypes G1 and G2, and groups A and C of the porcine rotavirus, are major etiological agents of severe gastroenteritis and profuse diarrhea, particularly among piglets, with mortality rates of up to 100%. Based on the high prevalence rate and frequent co-infection of PEDV, RVA, and RVC, close monitoring is necessary to avoid greater economic losses. We have developed a multiplex TaqMan probe-based real-time PCR for the rapid simultaneous detection and differentiation of PEDV subtypes G1 and G2, RVA, and RVC. This test is highly sensitive, as the detection limits were 20 and 100 copies/μL for the G1 and G2 subtypes of PEDV, respectively, and 50 copies/μL for RVA and RVC, respectively. Eighty-eight swine clinical samples were used to evaluate this new test. The results were 100% in concordance with the standard methods. Since reassortment between porcine and human rotaviruses has been reported, this multiplex test not only provides a basis for the management of swine diarrheal viruses, but also has the potential to impact public health as well.
