Abstract: In this work we try to search for signals generated by ultra-heavy dark matter at the Large High Altitude Air Shower Observatory (LHAASO) data. We look for possible gamma-ray by dark matter annihilation or decay from 16 dwarf spheroidal galaxies in the field of view of LHAASO. Dwarf spheroidal galaxies are among the most promising targets for indirect detection of dark matter which have low fluxes of astrophysical $\gamma$-ray background while large amount of dark matter. By analyzing more than 700 days observational data at LHAASO, no significant dark matter signal from 1 TeV to 1 EeV is detected. Accordingly we derive the most stringent constraints on the ultra-heavy dark matter annihilation cross-section up to EeV. 2. Ab initio calculations with a new local chiral N3LO nucleon-nucleon force

Abstract: Ab initio calculations have achieved remarkable success in nuclear structure studies. Numerous works highlight the pivotal role of three-body forces in nuclear ab initio calculations. Concurrently, efforts have been made to replicate these calculations using only realistic nucleon-nucleon (NN) interactions. A novel local chiral next-to-next-to-next-to-leading order (N3LO) NN interaction, distinct due to its weaker tensor force, has recently been established. This paper applies this local NN interaction in ab initio frameworks to calculate the low-lying spectra of p-shell light nuclei, particularly 10B, ground-state energies and shell evolution in oxygen isotopes. Results are compared with calculations utilizing nonlocal chiral N3LO NN and chiral NN +3N interactions. The ab initio calculations with the local N N potential accurately describe the spectra of p-shell nuclei, notably the 10B. Additionally, the neutron drip line for oxygen isotopes, with 24O as the drip line nucleus, is accurately reproduced in ab initio calculations with the local NN interaction. Calculations with the local NN interaction also reproduce the subshell closure at N = 14 and 16, albeit with a stronger shell gap compared to experimental data. However, the calculated charge radii based on the local NN interaction are underestimated compared with experimental data, which is similar to results from the nonlocal NN interaction. Consequently, the present ab initio calculations further indicate significant spin-orbit splitting effects with the new local NN potential, suggesting that 3N forces remain an important consideration., Comment: 9 pages, 7 figures

3. Ab initio valence-space in-medium similarity renormalization group calculations for neutron-rich P, Cl, and K isotopes

Abstract: Neutron-rich P, Cl, and K isotopes, particularly those with neutron numbers around $N=28$, have attracted extensive experimental and theoretical interest. We utilize the \textit{ab initio} valence-space in-medium similarity renormalization group approach, based on chiral nucleon-nucleon and three-nucleon forces, to investigate the exotic properties of these isotopes. Systematic calculations of the low-lying spectra are performed. A key finding is the level inversion between $3/2_1^+$ and $1/2_1^+$ states in odd-$A$ isotopes, attributed to the inversion of $\pi 0d_{3/2}$ and $\pi 1s_{1/2}$ single-particle states.\textit{Ab initio} calculations, which incorporate the three-nucleon forces, correlate closely with existing experimental data. Further calculations of effective proton single-particle energies provide deeper insights into the shell evolution for $Z=14$ and $16$ sub-shells. Our results indicate that the three-body force plays important roles in the shell evolution for $Z=14$ and $16$ sub-shells with neutron numbers ranging from 20 to 28. Additionally, systematic \textit{ab initio} calculations are conducted for the low-lying spectra of odd-odd nuclei. The results align with experimental data and provide new insights for future research into these isotopes, up to and beyond the drip line., Comment: 12 pages, 9 figures, 1 table

Abstract: The KM2A is the largest sub-array of the Large High Altitude Air Shower Observatory (LHAASO). It consists of 5216 electromagnetic particle detectors (EDs) and 1188 muon detectors (MDs). The data recorded by the EDs and MDs are used to reconstruct primary information of cosmic ray and gamma-ray showers. This information is used for physical analysis in gamma-ray astronomy and cosmic ray physics. To ensure the reliability of the LHAASO-KM2A data, a three-level quality control system has been established. It is used to monitor the status of detector units, stability of reconstructed parameters and the performance of the array based on observations of the Crab Nebula and Moon shadow. This paper will introduce the control system and its application on the LHAASO-KM2A data collected from August 2021 to July 2023. During this period, the pointing and angular resolution of the array were stable. From the observations of the Moon shadow and Crab Nebula, the results achieved using the two methods are consistent with each other. According to the observation of the Crab Nebula at energies from 25 TeV to 100 TeV, the time averaged pointing errors are estimated to be $-0.003^{\circ} \pm 0.005^{\circ}$ and $0.001^{\circ} \pm 0.006^{\circ}$ in the R.A. and Dec directions, respectively., Comment: 15 pages, 9 figures

Abstract: The first source catalog of Large High Altitude Air Shower Observatory reported the detection of a very-high-energy gamma ray source, 1LHAASO J1219+2915. In this paper a further detailed study of the spectral and temporal behavior of this point-like source have been carried. The best-fit position of the TeV source ($\rm{RA}=185.05^{\circ}\pm0.04^{\circ}$, $\rm{Dec}=29.25^{\circ}\pm0.03^{\circ}$) is compatible with NGC 4278 within $\sim0.03$ degree. Variation analysis shows an indication of the variability at a few months level in the TeV band, which is consistent with low frequency observations. Based on these observations, we report the detection of TeV $\gamma$-ray emissions from this low-luminosity AGN NGC 4278. The observations by LHAASO-WCDA during active period has a significance level of 8.8\,$\sigma$ with best-fit photon spectral index $\varGamma=2.56\pm0.14$ and a flux $f_{1-10\,\rm{TeV}}=(7.0\pm1.1_{\rm{sta}}\pm0.35_{\rm{syst}})\times10^{-13}\,\rm{photons\,cm^{-2}\,s^{-1}}$, or approximately $5\%$ of the Crab Nebula. The discovery of VHE from NGC 4278 indicates that the compact, weak radio jet can efficiently accelerate particles and emit TeV photons., Comment: 11 pages, 5 figures

Abstract: KM2A is one of the main sub-arrays of LHAASO, working on gamma ray astronomy and cosmic ray physics at energies above 10 TeV. Detector simulation is the important foundation for estimating detector performance and data analysis. It is a big challenge to simulate the KM2A detector in the framework of Geant4 due to the need to track numerous photons from a large number of detector units (>6000) with large altitude difference (30 m) and huge coverage (1.3 km^2). In this paper, the design of the KM2A simulation code G4KM2A based on Geant4 is introduced. The process of G4KM2A is optimized mainly in memory consumption to avoid memory overffow. Some simpliffcations are used to signiffcantly speed up the execution of G4KM2A. The running time is reduced by at least 30 times compared to full detector simulation. The particle distributions and the core/angle resolution comparison between simulation and experimental data of the full KM2A array are also presented, which show good agreement.

Abstract: We present the measurements of all-particle energy spectrum and mean logarithmic mass of cosmic rays in the energy range of 0.3-30 PeV using data collected from LHAASO-KM2A between September 2021 and December 2022, which is based on a nearly composition-independent energy reconstruction method, achieving unprecedented accuracy. Our analysis reveals the position of the knee at $3.67 \pm 0.05 \pm 0.15$ PeV. Below the knee, the spectral index is found to be -$2.7413 \pm 0.0004 \pm 0.0050$, while above the knee, it is -$3.128 \pm 0.005 \pm 0.027$, with the sharpness of the transition measured with a statistical error of 2%. The mean logarithmic mass of cosmic rays is almost heavier than helium in the whole measured energy range. It decreases from 1.7 at 0.3 PeV to 1.3 at 3 PeV, representing a 24% decline following a power law with an index of -$0.1200 \pm 0.0003 \pm 0.0341$. This is equivalent to an increase in abundance of light components. Above the knee, the mean logarithmic mass exhibits a power law trend towards heavier components, which is reversal to the behavior observed in the all-particle energy spectrum. Additionally, the knee position and the change in power-law index are approximately the same. These findings suggest that the knee observed in the all-particle spectrum corresponds to the knee of the light component, rather than the medium-heavy components., Comment: 8 pages, 3 figures

Abstract: The ground-state mass excess of the $T_{z}=-2$ drip-line nucleus $^{22}$Al is measured for the first time to be $18103(10)$ keV using the newly-developed B$\rho$-defined isochronous mass spectrometry method at the cooler storage ring in Lanzhou. The new mass excess value allowed us to determine the excitation energies of the two low-lying $1^+$ states in $^{22}$Al with significantly reduced uncertainties of 51 keV. Comparing to the analogue states in its mirror nucleus $^{22}$F, the mirror energy differences of the two $1^+$ states in the $^{22}$Al-$^{22}$F mirror pair are determined to be $-625(51)$ keV and $-330(51)$ keV, respectively. The excitation energies and the mirror energy differences are used to test the state-of-the-art \textit{ab initio} valence-space in-medium similarity renormalization group calculations with four sets of interactions derived from the chiral effective field theory. The mechanism leading to the large mirror energy differences is investigated and attributed to the occupation of the $\pi s_{1/2}$ orbital., Comment: 8 pages, 5 figures

Abstract: For decades, supernova remnants (SNRs) have been considered the prime sources of Galactic Cosmic rays (CRs). But whether SNRs can accelerate CR protons to PeV energies and thus dominate CR flux up to the knee is currently under intensive theoretical and phenomenological debate. The direct test of the ability of SNRs to operate as CR PeVatrons can be provided by ultrahigh-energy (UHE; $E_\gamma \geq 100$~TeV) $\gamma$-rays. In this context, the historical SNR Cassiopeia A (Cas A) is considered one of the most promising target for UHE observations. This paper presents the observation of Cas A and its vicinity by the LHAASO KM2A detector. The exceptional sensitivity of LHAASO KM2A in the UHE band, combined with the young age of Cas A, enabled us to derive stringent model-independent limits on the energy budget of UHE protons and nuclei accelerated by Cas A at any epoch after the explosion. Abstract: The highest energy gamma-rays from gamma-ray bursts (GRBs) have important implications for their radiation mechanism. Here we report for the first time the detection of gamma-rays up to 13 TeV from the brightest GRB 221009A by the Large High Altitude Air-shower Observatory (LHAASO). The LHAASO-KM2A detector registered more than 140 gamma-rays with energies above 3 TeV during 230$-$900s after the trigger. The intrinsic energy spectrum of gamma-rays can be described by a power-law after correcting for extragalactic background light (EBL) absorption. Such a hard spectrum challenges the synchrotron self-Compton (SSC) scenario of relativistic electrons for the afterglow emission above several TeV. Observations of gamma-rays up to 13 TeV from a source with a measured redshift of z=0.151 hints more transparency in intergalactic space than previously expected. 11. Multi-wavelength temporal variability of the blazar PKS 1510-089

Abstract: We perform correlation and periodicity search analyses on long-term multi-band light curves of the FSRQ 1510-089 observed by the space-based Fermi--Large Area Telescope in gamma-rays, the SMARTS and Steward Observatory telescopes in optical and near-infrared (NIR) and the 13.7 m radio telescope in Metsahovi Radio Observatory between 2008 and 2018. The z-transform discrete correlation function method is applied to study the correlation and possible time lags among these multi band light curves. Among all pairs of wavelengths, the gamma-ray vs. optical/NIR and optical vs. NIR correlations show zero time lags; however, both the gamma-ray and optical/NIR emissions precede the radio radiation. The Generalized Lomb-Scargle periodogram, Weighted Wavelet Z-transform, and REDFIT techniques are employed to investigate the unresolved-core-emission dominated 37 GHz light curve and yield evidence for a quasi-period around 1540 days, although given the length of the whole data set it cannot be claimed to be significant. We also investigate the optical/NIR color variability and find that this source shows a simple redder-when-brighter behavior over time, even in the low flux state., Comment: Accepted for publication in ApJ; 20 pages, 9 figures, 4 tables

Abstract: Nuclei in the vicinity of driplines have been receiving a lot of attention in nuclear structure studies. In the nuclei, the continuum coupling is crucial in reproducing weakly-bound and unbound phenomena. To calculate observables of the nuclei as open quantum systems, we have developed valence-space effective operators in the complex-energy Berggren basis using many-body perturbation theory. We focus on the Gamow-Teller $\beta$ decay in the {\it sd} shell. The two- plus three-nucleon force from the chiral effective field theory (EFT), named EM1.8/2.0, has been used. The Gamow shell model which takes the continuum coupling into account can properly reproduce experimental observations of weakly-bound and unbound states. The $\beta$-decay isospin asymmetry between the dripline nucleus $^{22}\rm Si$ and its mirror partner $^{22}\rm O$ is reproduced, in which the $s_{1/2}$ continuum plays a key role. Significant Thomas-Ehrman shift is seen through mirror energy differences between the mirror daughters $^{22}\rm Al$ and $^{22}\rm F$, in which the continuum effect plays an important role.

However, its mechanism of action remains unclear. Here, we evaluated the efficacy and molecular mechanism of BMSC-derived NVs carrying the curcumin analog H8 (H8-BMSCs-NVs) on hepatic glucose and lipid metabolism in type 2 diabetes (T2D).Subjects and Methods: Mouse BMSCs were isolated by collagenase digestion and H8-BMSCs-NVs were prepared by microvesicle extrusion. The effects of H8-BMSCs-NVs on hepatic glucose and lipid metabolism were observed in a T2D mouse model and a HepG2 cell insulin resistance model. To evaluate changes in potential signaling pathways, the PI3K/AKT/AMPK signaling pathway and expression levels of G6P and PEPCK were assessed by Western blotting.Results: H8-BMSCs-NVs effectively improved lipid accumulation in liver tissues and restored liver dysfunction in T2D mice. Meanwhile, H8-BMSCs-NVs effectively inhibited intracellular lipid accumulation in the insulin resistance models of HepG2 cells. Mechanistic studies showed that H8-BMSCs-NVs activated the PI3K/AKT/AMPK signaling pathway and decreased the expression levels of G6P and PEPCK.Conclusion: These findings demonstrate that H8-BMSCs-NVs improved hepatic glucose and lipid metabolism in T2D mice by activating the PI3K/AKT/AMPK signaling pathway, which provides novel evidence suggesting the potential of H8-BMSCs-NVs in the clinically treatment of T2D patients.Keywords: bone mesenchymal stem cell, nanovesicles, curcumin analogue H8, type 2 diabetes

Abstract: We present the first catalog of very-high energy and ultra-high energy gamma-ray sources detected by the Large High Altitude Air Shower Observatory (LHAASO). The catalog was compiled using 508 days of data collected by the Water Cherenkov Detector Array (WCDA) from March 2021 to September 2022 and 933 days of data recorded by the Kilometer Squared Array (KM2A) from January 2020 to September 2022. This catalog represents the main result from the most sensitive large coverage gamma-ray survey of the sky above 1 TeV, covering declination from $-$20$^{\circ}$ to 80$^{\circ}$. In total, the catalog contains 90 sources with an extended size smaller than $2^\circ$ and a significance of detection at $> 5\sigma$. Based on our source association criteria, 32 new TeV sources are proposed in this study. Among the 90 sources, 43 sources are detected with ultra-high energy ($E > 100$ TeV) emission at $> 4\sigma$ significance level. Abstract: The diffuse Galactic $\gamma$-ray emission, mainly produced via interactions between cosmic rays and the interstellar medium and/or radiation field, is a very important probe of the distribution, propagation, and interaction of cosmic rays in the Milky Way. In this work we report the measurements of diffuse $\gamma$-rays from the Galactic plane between 10 TeV and 1 PeV energies, with the square kilometer array of the Large High Altitude Air Shower Observatory (LHAASO). Diffuse emissions from the inner ($15^{\circ}10$~TeV). The energy spectrum in the inner Galaxy regions can be described by a power-law function with an index of $-2.99\pm0.04$, which is different from the curved spectrum as expected from hadronic interactions between locally measured cosmic rays and the line-of-sight integrated gas content. Furthermore, the measured flux is higher by a factor of $\sim3$ than the prediction. A similar spectrum with an index of $-2.99\pm0.07$ is found in the outer Galaxy region, and the absolute flux for $10\lesssim E\lesssim60$ TeV is again higher than the prediction for hadronic cosmic ray interactions. The latitude distributions of the diffuse emission are consistent with the gas distribution, while the longitude distributions show clear deviation from the gas distribution. Abstract: Recent observations of the light component of the cosmic-ray spectrum have revealed unexpected features that motivate further and more precise measurements up to the highest energies. The Dark Matter Particle Explorer is a satellite-based cosmic-ray experiment that has been operational since December 2015, continuously collecting data on high-energy cosmic particles with very good statistics, energy resolution, and particle identification capabilities. In this work, the latest measurements of the energy spectrum of proton+helium in the energy range from 46 GeV to 464 TeV are presented. Among the most distinctive features of the spectrum, a spectral hardening at 600 GeV has been observed, along with a softening at 29 TeV measured with a 6.6{\sigma} significance. Moreover, the detector features and the analysis approach allowed for the extension of the spectral measurement up to the sub-PeV region. Abstract: Isospin-symmetry breaking is responsible for the energy difference of excited states in mirror nuclei. It also influences the coefficient of the isobaric multiplet mass equation. In the present work, we extensively investigate isospin-symmetry breaking in medium mass nuclei within ab initio frameworks. For this, we employ the ab initio valence-space in-medium similarity renormalization group approach, in which charge-symmetry and charge-independence breakings are included in the adopted nuclear force. The mirror energies of sd- and pf- shell nuclei are computed for that matter. The effects of single-particle states on weakly bound and unbound nuclear states, especially those of the s-wave, are discussed. Predictions are also made concerning proton drip-line nuclei bearing large mirror energy difference. Finally, the coefficient of the isobaric multiplet mass equation in T = 1/2 and T = 1 isospin multiplets for A = 18 to A = 76 is calculated.

18. Ab initio descriptions of $A=16$ mirror nuclei with resonance and continuum coupling

Abstract: We have used an {\it ab initio} Gamow shell model to study the isospin symmetry breaking in the $A=16$ mirror nuclei of $^{16}$F, $^{16}$N, $^{16}$Ne and $^{16}$C. Starting from a chiral interaction with two-nucleon force (2NF) at N$^3$LO and three-nucleon force (3NF) at N$^2$LO, a complex-momentum ${\it psd}$-shell Hamiltonian was constructed by employing the many-body perturbation theory in the Gamow Hartree-Fock basis which includes bound, resonant and continuum states self-consistently. Such an elaborated {\it ab initio} Gamow shell model with both continuum coupling and 3NF included can properly treat the many-body correlations of weakly bound and unbound nuclei. The mirror partners of $^{16}$F and $^{16}$N exhibit different level orders in their excitation spectra, which can be well explained by the inclusion of 3NF in the calculation. The isospin asymmetry between the mirror partners $^{16}$Ne and $^{16}$C was studied in detail by insight into their configuration structures. This study aims to evaluate the levels and explore the associated factors with KAP on antibiotic use and AMR in Chinese nursing students.Methods: A cross-sectional survey using a self-administered questionnaire consisting of demographics and selected features and KAP on antibiotic use and AMR was conducted to measure KAP levels among nursing students at various universities in Hubei Province, China. The logistic regression analyses were performed to analyze the potential factors associated with the KAP.Results: The survey eventually included a total of 1959 nursing students. The mean scores for KAP were 57.89 ± 26.32, 55.00 ± 12.50, and 71.88 ± 15.63, respectively. Regarding knowledge, 54.3% of participants were unaware that antibiotic was ineffective against viral infections. Regarding attitude, 36% of participants agreed that current antibiotic abuse existed; 96.2% of participants thought it necessary to set up a special course on antibiotics. Regarding practice, only 48.4% of participants usually purchased antibiotics with a prescription. Multivariable analyses indicated that lack of discussion on AMR in school courses was an independent risk factor against KAP, respectively. The main knowledge sources of antibiotic being outside the classroom was an independent risk factor related to knowledge and practice. The average score > 80 points was an independent protective factor related to knowledge and practice.Conclusion: The KAP level on antibiotic use and AMR among Hubei nursing students was general and required further strengthening. Nursing students with risk factors should be prioritized in educational interventions. Abstract: More than a century after the performance of the oil drop experiment, the possible existence of fractionally charged particles FCP still remains unsettled. The search for FCPs is crucial for some extensions of the Standard Model in particle physics. Most of the previously conducted searches for FCPs in cosmic rays were based on experiments underground or at high altitudes. However, there have been few searches for FCPs in cosmic rays carried out in orbit other than AMS-01 flown by a space shuttle and BESS by a balloon at the top of the atmosphere. In this study, we conduct an FCP search in space based on on-orbit data obtained using the DArk Matter Particle Explorer (DAMPE) satellite over a period of five years. Unlike underground experiments, which require an FCP energy of the order of hundreds of GeV, our FCP search starts at only a few GeV. An upper limit of $6.2\times 10^{-10}~~\mathrm{cm^{-2}sr^{-1} s^{-1}}$ is obtained for the flux. Our results demonstrate that DAMPE exhibits higher sensitivity than experiments of similar types by three orders of magnitude that more stringently restricts the conditions for the existence of FCP in primary cosmic rays., Comment: 19 pages, 6 figures, accepted by PRD

Abstract: The Large High Altitude Air Shower Observatory (LHAASO) has three sub-arrays, KM2A, WCDA and WFCTA. The flux variations of cosmic ray air showers were studied by analyzing the KM2A data during the thunderstorm on 10 June 2021. The number of shower events that meet the trigger conditions increases significantly in atmospheric electric fields, with maximum fractional increase of 20%. The variations of trigger rates (increases or decreases) are found to be strongly dependent on the primary zenith angle. The flux of secondary particles increases significantly, following a similar trend with that of the shower events. To better understand the observed behavior, Monte Carlo simulations are performed with CORSIKA and G4KM2A (a code based on GEANT4). We find that the experimental data (in saturated negative fields) are in good agreement with simulations, assuming the presence of a uniform upward electric field of 700 V/cm with a thickness of 1500 m in the atmosphere above the observation level. Due to the acceleration/deceleration and deflection by the atmospheric electric field, the number of secondary particles with energy above the detector threshold is modified, resulting in the changes in shower detection rate., Comment: 18 pages, 11 figures

22. Efficient Second Harmonic Generation from Silicon Slotted Nanocubes with Bound States in the Continuum

Abstract: Optical materials with centrosymmetry, such as silicon and germanium, are unfortunately absent of second-order nonlinear optical responses, hindering their developments in efficient nonlinear optical devices. Here, a design with an array of slotted nanocubes is proposed to realize remarkable second harmonic generation (SHG) from the centrosymmetric silicon, which takes advantage of enlarged surface second-order nonlinearity, strengthened electric field over the surface of the air-slot, as well as the resonance enhancement by the bound states in the continuum. Compared with that from the array of silicon nanocubes without air-slots, SHG from the slotted nanocube array is improved by more than two orders of magnitude. The experimentally measured SHG efficiency of the silicon slotted nanocube array is high as 1.8*10^-4 W^-1, which is expected to be further engineered by modifying the air-slot geometries. 23. High-Q Resonances Governed by the Quasi-Bound States in the Continuum in All-Dielectric Metasurfaces

Abstract: The realization of high-Q resonances in a silicon metasurface with various broken-symmetry blocks is reported. Theoretical analysis reveals that the sharp resonances in the metasurfaces originate from symmetry-protected bound states in the continuum (BIC) and the magnetic dipole dominates these peculiar states. A smaller size of the defect in the broken-symmetry block gives rise to the resonance with a larger Q factor. Importantly, this relationship can be tuned by changing the structural parameter, resulting from the modulation of the topological configuration of BICs. Consequently, a Q factor of more than 3,000 can be easily achieved by optimizing dimensions of the nanostructure. At this sharp resonance, the intensity of the third harmonic generation signal in the patterned structure can be 368 times larger than that of the flat silicon film. The proposed strategy and underlying theory can open up new avenues to realize ultrasharp resonances, which may promote the development of the potential meta-devices for nonlinearity, lasing action, and sensing., Comment: 12 pages,5 figures

Abstract: We have developed an {\it ab initio} deformed in-medium similarity renormalization group (IMSRG) for open-shell nuclei. This is a single-reference IMSRG in deformed Hartree-Fock (HF) basis. Deformed wave functions are more efficient in describing deformed nuclei. The broken spherical symmetry needs to be restored by angular momentum projection, which is computational expensive. The angular momentum mainly capture the static correlations and can be estimated by the projection of the HF state. In this work, we do deformed IMSRG calculation and add the correlation energy from projected HF as a leading order approximation. As the test ground, we have calculated the deformed $^{8,10}\rm Be$ isotopes with the optimized chiral interaction NNLO$_{\rm opt}$. The results are benchmarked with the no-core shell model and valence space IMSRG calculations. Then we systematically investigated the ground-state energies and charge radii of even-even isotopes from light beryllium to medium-mass magnesium. The calculated energies are extrapolated to infinite basis space by an exponential form, and compared with the extrapolated valence-space IMSRG results and experimental data available.

25. Role of the companion lensing galaxy in the CLASS gravitational lens B1152+199

Abstract: We reinvestigate the Cosmic Lens All-Sky Survey (CLASS) gravitational lens B1152+199 using archived Hubble Space Telescope (HST) data and Very Long Baseline Interferometry (VLBI) data. A consistent luminosity ratio within effective radius between the host galaxy and the X-galaxy is measured from HST tri-band images, which leads to a mass ratio between the two galaxies as $r_b\sim 2$. To determine the role of the X-galaxy in the lens system, we modelled the dual-lens system with constraints from the VLBI-resolved jet components and the HST images. The 8.4-GHz global-VLBI data currently provide the most stringent constraints to the mass model, especially to the radial power-law slope. The optimized models for this two-image three-component radio lens favour a steeper-than-isothermal inner slope. The jet bending in image B was also investigated and it turned out to be rather a misalignment than a curvature. The goodness of fit indicates that the role of the X-galaxy is crucial in the lens system if three pairs of resolved jet components are to be fitted. When we imported the optimal model from radio constraints to optical modelling with the HST tri-band data, the optimization kept the consistency of the optimal model and successfully reproduced the features observed in the HST images. This implies that the diffuse emission discovered in the HST images is actually a detection of the secondary lensing effects from the companion lens., Comment: Published in MNRAS; 20 pages, 14 figures

Abstract: With both three-nucleon force and continuum coupling included, we have developed a self-consistent {\it ab initio} Gamow shell model within the Gamow Hartree-Fock (GHF) basis obtained by the realistic interaction itself. With the chiral two-nucleon N$^3$LO and three-nucleon N$^2$LO interactions, the Gamow shell model has been applied to the mirror systems of $Z=8$ neutron-rich isotopes and $N=8$ proton-rich isotones, giving good agreements with data in binding energies, dripline positions and excitation spectra. The GHF calculated that the $0d_{3/2}$, $1s_{1/2}$ and $1p_{3/2}$ orbitals are resonances. The resonance states and their interplay with nonresonant continua play a crucial role in the descriptions of nuclei around driplines. Excitation spectra and Thomas-Ehrman shifts observed can be better described when both three-nucleon force and continuum coupling are considered in calculations. The three-nucleon force and continuum coupling produce a combined effect on the Thomas-Ehrman shift, e.g., for the ${1/2}^+$ resonance level of $^{19}$Na. Differentially expressed RNA modification-related genes were identified and then functionally annotated. Univariate logistic regression and lasso regression analysis were used to identify primary disease genes for OA and RA. Validation was done using scRNA-seq analysis and immunohistochemistry (IHC) in human knee synovial tissues and a murine destabilization of the medial meniscus (DMM) model. Through WGCNA analysis, genes associated with cell pyroptosis or autophagy in OA and RA were identified, which were then combined with differentially expressed RNA modification-related genes to construct a PPI interaction network. Furthermore, hub genes were selected for ceRNA interaction network analysis, correlation analysis with OA and RA molecular subtypes, as well as correlation analysis with 22 immune cells.Results: Six RNA modification-related genes (ADAMDEC1, IGHM, OGN, TNFRSF11B, SCARA3 and PTN) were identified as potential OA and RA pathogenesis biomarkers. Their expression was validated in human knee synovial tissues and a murine DMM model. Functional enrichment of differentially expressed RNA modification-related genes between RA and OA was analyzed using GO, KEGG, GSEA, and GSVA. Based on WGCNA and PPI analysis, the six hub genes related to pyroptosis and RNA modification (CXCL10, CXCL9, CCR7, CCL5, CXCL1, and CCR2) were identified as central nodes for ceRNA interaction, correlation with OA and RA molecular subtypes, and association with 22 immune cells.Conclusion: Our research revealed the significance of RNA modification-related genes in the development of OA and RA pathogenesis, thereby providing a novel research direction for understanding the mechanisms, diagnosis, and treatment of OA and RA.Keywords: m6A, m5C, m1A, osteoarthritis, rheumatoid arthritis, single-cell sequencing

Abstract: The Crab pulsar and the surrounding nebula powered by the pulsar's rotational energy through the formation and termination of a relativistic electron-positron wind is a bright source of gamma-rays carrying crucial information about this complex conglomerate. We report the detection of $\gamma$-rays with a spectrum showing gradual steepening over three energy decades, from $5\times 10^{-4}$ to $1.1$ petaelectronvolt (PeV). The ultra-high-energy photons exhibit the presence of a PeV electron accelerator (a pevatron) with an acceleration rate exceeding 15% of the absolute theoretical limit. Assuming that unpulsed $\gamma$-rays are produced at the termination of the pulsar's wind, we constrain the pevatron's size, between $0.025$ and $0.1$ pc, and the magnetic field $\approx 110 \mu$G. The production rate of PeV electrons, $2.5 \times 10^{36}$ erg $\rm s^{-1}$, constitutes 0.5% of the pulsar's spin-down luminosity, although we do not exclude a non-negligible contribution of PeV protons to the production of the highest energy $\gamma$-rays., Comment: 43 pages, 13 figures, 2 tables; Published in Science

Abstract: The measurement of the energy spectrum of cosmic ray helium nuclei from 70 GeV to 80 TeV using 4.5 years of data recorded by the DArk Matter Particle Explorer (DAMPE) is reported in this work. A hardening of the spectrum is observed at an energy of about 1.3 TeV, similar to previous observations. In addition, a spectral softening at about 34 TeV is revealed for the first time with large statistics and well controlled systematic uncertainties, with an overall significance of $4.3\sigma$. The DAMPE spectral measurements of both cosmic protons and helium nuclei suggest a particle charge dependent softening energy, although with current uncertainties a dependence on the number of nucleons cannot be ruled out., Comment: 11 pages, 13 figures, published in Phys. Abstract: The Wide Field-of-View Cherenkov Telescope Array (WFCTA) and the Water Cherenkov Detector Arrays (WCDA) of LHAASO are designed to work in combination for measuring the energy spectra of various cosmic ray species over a very wide energy range from a few TeV to 10 PeV. The energy calibration of WCDA can be achieved with a proven technique of measuring the westward shift of the Moon shadow of galactic cosmic rays due to the geomagnetic field. This deflection angle $\Delta$ is inversely proportional to the energy of the cosmic rays. The precise measurements of the shifts by WCDA allows us to calibrate its energy scale for energies as high as 35 TeV. The energy scale measured by WCDA can be used to cross calibrate the energy reconstructed by WFCTA, which spans the whole energy range up to 10 PeV. In this work, we will demonstrate the feasibility of the method using the data collected from April 2019 to January 2020 by the WFCTA array and WCDA-1 detector, the first of the three water Cherenkov ponds, already commissioned at LHAASO site.

A zebrafish xenograft model was used to determine the anti-cancer efficacy of QBD. Cell counting kit-8 assay, terminal deoxyribonucleotide transferase-mediated-dUTP nick-end labeling assay; immunofluorescence, and flow cytometry were used to evaluate the in vitro anti-proliferative and pro-apoptotic effects of QBD and ART B on H1299 cells. Subsequently, the related targets and action mechanisms of both QBD and ART B predicted by network pharmacological analyses were experimentally validated by real-time PCR and Western blot assays on H1299 cells.Results: UPLC-QTOF-MS/MS identified a total of 69 compounds (such as ART B, mangiferin, and artemisinic acid) in QBD. The in vivo data showed that QBD significantly inhibited the growth of H1299 cells in xenograft larval zebrafish from 125 to 500 μg/mL. The in vitro data showed that QBD induced apoptosis of H1299 cells, accompanied by down-regulating the expression of BCL-2 and up-regulating the expression of BIM, PUMA, BAX, c-PARP, γ-H2A.X, c-CASP3, and c-CASP8. Alike QBD, ART B exerted similar anti-proliferative and pro-apoptotic effects on H1299 cells. Moreover, ART B inhibited expressions of BCL2L1, AKT1, AKT2, MMP-2, and EGFR, and up-regulated ALB expression. Mechanistically, ART B promoted apoptosis of H1299 cells by inhibiting PI3K/Akt signaling pathway.Conclusion: This study revealed the anti-NSCLC efficacy of QBD. ART B, the effective component of QBD, plays an anti-NSCLC role by down-regulating the PI3K-Akt signaling pathway. Modern pharmacological studies suggest that some natural monomeric compounds have a significant effect on inhibiting tumor growth. However, poor stability and solubility, and side effects are the main factors limiting the clinical application of natural monomeric compounds.Methods: In this paper, drug-co-loaded nanoself-assemblies were selected as a delivery system to improve the chemical stability and solubility of Tanshinone II A and Glycyrrhetinic acid, and to produce a synergetic anti-hepatoma effect.Results: The study suggested that the drug co-loaded nanoself-assemblies showed high drug loading capacity, good physical and chemical stability, and controlled release. In vitro cell experiments verified that the drug-co-loaded nanoself-assemblies could increase the cellular uptake and cell inhibitory activity. In vivo studies verified that the drug co-loaded nanoself-assemblies could prolong the MRT0-∞, increase accumulation in tumor and liver tissues, and show strong synergistic anti-tumor effect and good bio-safety in H22 tumor-bearing mice.Conclusion: This work indicates that natural monomeric compounds co-loaded nanoself-assemblies would be a potential strategy for the treatment of hepatoma.Graphical Abstract: Keywords: nanoself-assemblies, drug combination, glycyrrhetinic acid, tanshinone IIA, liver cancer

Abstract: A kinematically complete quasi-free $(p,pn)$ experiment in inverse kinematics was performed to study the structure of the Borromean nucleus $^{17}$B, which had long been considered to have neutron halo. By analyzing the momentum distributions and exclusive cross sections, we obtained the spectroscopic factors for $1s_{1/2}$ and $0d_{5/2}$ orbitals, and a surprisingly small percentage of 9(2)$\%$ was determined for $1s_{1/2}$. Our finding of such a small $1s_{1/2}$ component and the halo features reported in prior experiments can be explained by the deformed relativistic Hartree-Bogoliubov theory in continuum, revealing a definite but not dominant neutron halo in $^{17}$B. The present work gives the smallest $s$- or $p$-orbital component among known nuclei exhibiting halo features, and implies that the dominant occupation of $s$ or $p$ orbitals is not a prerequisite for the occurrence of neutron halo., Comment: Accepted for publication in Physical Review Letters,8 pages, 4 figures

Abstract: The focal plane camera is the core component of the Wide Field-of-view Cherenkov/fluorescence Telescope Array (WFCTA) of the Large High-Altitude Air Shower Observatory (LHAASO). Because of the capability of working under moonlight without aging, silicon photomultipliers (SiPM) have been proven to be not only an alternative but also an improvement to conventional photomultiplier tubes (PMT) in this application. Eighteen SiPM-based cameras with square light funnels have been built for WFCTA. The telescopes have collected more than 100 million cosmic ray events and preliminary results indicate that these cameras are capable of working under moonlight. The characteristics of the light funnels and SiPMs pose challenges (e.g. dynamic range, dark count rate, assembly techniques). In this paper, we present the design features, manufacturing techniques and performances of these cameras. Abstract: As a sub-array of the Large High Altitude Air Shower Observatory (LHAASO), KM2A is mainly designed to cover a large fraction of the northern sky to hunt for gamma-ray sources at energies above 10 TeV. Even though the detector construction is still underway, a half of the KM2A array has been operating stably since the end of 2019. In this paper, we present the pipeline of KM2A data analysis and the first observation on the Crab Nebula, a standard candle in very high energy gamma-ray astronomy. We detect gamma-ray signals from the Crab Nebula in both energy ranges of 10$-$100 TeV and $>$100 TeV with high significance, by analyzing the KM2A data of 136 live days between December 2019 and May 2020. With the observations, we test the detector performance including angular resolution, pointing accuracy and cosmic ray background rejection power. The energy spectrum of the Crab Nebula in the energy range 10-250 TeV fits well with a single power-law function dN/dE =(1.13$\pm$0.05$_{stat}$$\pm$0.08$_{sys}$)$\times$10$^{-14}$$\cdot$(E/20TeV)$^{-3.09\pm0.06_{stat}\pm0.02_{sys}}$ cm$^{-2}$ s$^{-1}$ TeV$^{-1}$. It is consistent with previous measurements by other experiments. The types of cells involved in OA include chondrocytes, osteoblasts, osteoclasts, synovial fibroblasts, T cells, macrophages, and mesenchymal stem cells (MSCs). There are many effects of immune cells in OA, and innate immune cells such as dendritic cells and macrophages have significant roles in the pathogenesis of OA. On the other hand, the role of adaptive immune cells such as T-cell subsets, B-cell subsets, and NK cells is important in OA pathogenesis. MSCs not only play a key role in the dynamic balance and repair of OA tissue but also inhibit the immune system. Therefore, MSCs are a new potential therapeutic agent for OA. Considering the difference between the clinical behaviour of BM and LM, there is the need for addressing the role of these biomarkers in LM.Methods: The present study retrospectively analyzed 95 consecutive patients with LM from lung cancer who were diagnosed at the National Cancer Center, Cancer Hospital of Chinese Academy of Medical Sciences between January 2016 and December 2019. Baseline NLR, platelet-to-lymphocyte ratio (PLR), systemic immunoinflammation index (SII), and lymphocyte‐to-monocyte ratio at diagnosis of LM were calculated based on complete blood count and correlated, along with other characteristics, with overall survival (OS) using univariate and multivariate analyses. The best cutoff values for systemic immunoinflammation biomarkers were derived using the surv_cutpoint function in R software, which optimized the significance of the split between Kaplan–Meier survival curves.Results: Median OS of patients with LM was 12 months (95% CI 9– 17 months). On univariate analysis, NLR, PLR, SII, LMR, sex, smoking history, ECOG performance status (PS) scores, histological subtypes and targeted therapy were all significantly associated with OS. Only NLR (P=0.034, 95% CI 1.060– 4.578) and ECOG PS scores (P=0.019, 95% CI 0.137– 0.839) maintained a significant association with OS on multivariate analysis. Furthermore, patients with baseline NLR > 3.57 had significantly worse OS than patients with NLR ≤ 3.57 (median OS 7 vs 17 months), as did patients with ECOG PS scores > 2 vs ≤ 2 (median OS 4 vs 15 months).Conclusion: Both baseline NLR and PS scores at the time of LM diagnosis are helpful and available prognostic biomarkers for patients with LM from lung cancer.Keywords: systemic immunoinflammation index, neutrophil-to-lymphocyte ratio, platelet-to-lymphocyte ratio, lymphocyte‐to-monocyte ratio, leptomeningeal metastases, lung cancer

Abstract: We probe nuclear multipole resonances in the framework of the random-phase approximation by using the interaction obtained from the chiral effective field theory. The three-nucleon force is included in a form of the in-medium two-nucleon interaction which was derived from the chiral three-nucleon force. The isoscalar monopole, isoscalar dipole, isovector dipole and isoscalar quadrupole resonances of the closed-shell $^{56,68,78}$Ni have been investigated. The calculations reasonably reproduce the experimental multipole resonances of $^{56,68}$Ni, and well describe the pygmy dipole resonance and dipole polarizability measured in $^{68}$Ni. The multipole resonances of $^{78}$Ni, including pygmy dipole resonance and dipole polarizability, are predicted. The detailed effects of the tensor force and three-body force are analyzed by dissecting the chiral interaction. We find that in general the tensor force effect on electric giant resonances is not as significant as the effect from the three-body force, although the tensor force provides more than half of the binding energy. Abstract: The precise measurement of the spectrum of protons, the most abundant component of the cosmic radiation, is necessary to understand the source and acceleration of cosmic rays in the Milky Way. This work reports the measurement of the cosmic ray proton fluxes with kinetic energies from 40 GeV to 100 TeV, with two and a half years of data recorded by the DArk Matter Particle Explorer (DAMPE). This is the first time an experiment directly measures the cosmic ray protons up to ~100 TeV with a high statistics. The measured spectrum confirms the spectral hardening found by previous experiments and reveals a softening at ~13.6 TeV, with the spectral index changing from ~2.60 to ~2.85. Abstract: The isotropic chemical shifts can be calculated either by full-electron configuration, or by hybrid functionals, which costs a large amount of computational resources. To save the time, DFT+U could be employed to calculate the isotropic chemical shifts. However, the calculated properties are very sensitive to the Hubbard correction value $U_{eff}$. Here the double Fermi-contact-shift verification approach with DFT+U method is proposed with much higher computational efficiency, that is, simultaneously calculate the Fermi-contact shifts on two nuclei ($^{6}Li$ and $^{17}O$) to predict the optimal $U_{eff}$. The optimal $U_{eff}$ is also helpful to the calculations of quadrupolar coupling constant $C_{Q}$, $g$-factor, band structure and density of states.

Abstract: The DArk Matter Particle Explorer (DAMPE), a satellite-based cosmic ray and gamma-ray detector, was launched on December 17, 2015, and began its on-orbit operation on December 24, 2015. In this work we document the on-orbit calibration procedures used by DAMPE and report the calibration results of the Plastic Scintillator strip Detector (PSD), the Silicon-Tungsten tracKer-converter (STK), the BGO imaging calorimeter (BGO), and the Neutron Detector (NUD). The results are obtained using Galactic cosmic rays, bright known GeV gamma-ray sources, and charge injection into the front-end electronics of each sub-detector. The determination of the boundary of the South Atlantic Anomaly (SAA), the measurement of the live time, and the alignments of the detectors are also introduced. The calibration results demonstrate the stability of the detectors in almost two years of the on-orbit operation.

Readmission for severe exacerbations is a crucial event for COPD patients. However, factors associated with readmission for severe exacerbations are incomplete. The study aimed to investigate different characteristics between the severe and non-severe exacerbation groups.Patients and Methods: Patients hospitalized for severe AECOPD were included in multi-centers, and their exacerbations in next 12 months after discharge were recorded. According to exacerbations, patients were separated into the severe-exacerbation group and the non-severe exacerbation group. Propensity-score matching (PSM) and multivariable analyses were performed to compare the baseline characteristics of two groups. The Hosmer-Lemeshow test and receiver operating characteristic curve were applied to evaluate how well the model could identify clusters.Results: The cohort included 550 patients with severe AECOPD across 27 study centers in China, and 465 patients were finally analyzed. A total of 41.5% of patients underwent readmission for AECOPD within 1 year. There were no significant differences in baseline characteristics between groups after PSM. Severe exacerbations in the 12 months were related to some factors, eg, the duration of COPD (13 vs 8 years, P< 0.001), the COPD Assessment Test (CAT) score (20 vs 17, P< 0.001), the blood eosinophil percentage (1.5 vs 2.0, P< 0.05), and their inhaler therapies. Patients readmitted with AECOPD had a longer time of diagnosis (≥ 9 years), more symptoms (CAT ≥ 10), and lower blood eosinophils (Eos < 2%). A clinical model was derived to help identify patients at risk of readmission with severe exacerbations.Conclusion: These analyses confirmed the relevance of COPD at admission with future severe exacerbations. A lower blood eosinophils percentage appears to be related to readmission when combined with clinical history. The pathological mechanisms underlying EoE are largely unknown.Objective: We sought to understand the mechanisms underlying aeroallergen-induced EoE in Sharpin gene deficient (Sharpin-/-) mice that is prone to inflammatory response.Methods: Sharpin-/-mice were exposed with Aspergillus fumigatus and ovalbumin intranasally every alternate day for 4 weeks. Wild type (WT) naïve mice, WT exposed, and un-exposed Sharpin-/- mice were controls. Histopathological analysis was performed by H&E, trichrome and major basic protein staining. Total and specific IgE, IgG, and IgA levels were measured by ELISA and Th2 cytokine and CCL11 chemokine gene expression were determined.Results: Airborne allergen exposed Sharpin-/- mice showed severe eosinophilic inflammation in the esophagus (p < 0.001), and markedly increased epithelial thickening (p < 0.0001) compared to WT normal controls, whereas airborne allergen exposed WT mice and unexposed Sharpin-/- mice only showed mild eosinophilic inflammation in the esophagus. These exposed Sharpin-/- mice also showed over 7-fold increase in blood eosinophils (p < 0.0001), 60-fold increase in eosinophils in bronchoalveolar lavage fluid (p < 0.0001) and 4-fold increase in eosinophils in the skin (p < 0.0001) compared to normal controls. Surprisingly, exposed Sharpin-/- mice did not show elevation of serum total or antigen-specific IgE levels but reduced total IgA and IgG levels than normal controls There was a marked increase in IL-4, IL-13 and CCL11 gene expression in esophageal tissue (p < 0.001) in exposed Sharpin-/- mice compared to WT normal mice.Conclusion: Th2 cytokines and chemokines, but not IgE may play an important pathologic role in aeroallergen-induced EoE. An animal model of lupus nephritis was established by pristane to verify the interventional effect of LP-HFY15 on mouse lupus nephritis by regulating the transforming growth factor-β 1 (TGF-β 1) signaling pathway.Materials and Methods: Indexes in mouse serum and tissues were detected by kits, pathological changes in mouse kidney were observed by hematoxylin-eosin (H&E) staining, and quantitative polymerase chain reaction (qPCR) was used to detect TGF-β 1-related expression in mouse kidney tissue, which further elucidated the mechanism of LP-HFY15.Results: LP-HFY15 decreased the elevation of urinary protein and the levels of interleukin-6 (IL-6), IL-12, tumor necrosis factor alpha (TNF-α), and interferon γ (IFN-γ) in serum and kidney tissue. LP-HFY15 also reduced serum creatinine (SCr), blood urea nitrogen (BUN), total cholesterol (TC), triglyceride (TG), and raised total protein (TP), and albumin (ALB) levels in mice with nephritis. In addition, LP-HFY15 inhibited the positive rate of double-stranded deoxyribonucleic acid (dsDNA) antibodies in mice with nephritis. The observation of H&E sections showed that LP-HFY15 alleviated the glomerulus morphological incompleteness and inflammatory infiltration caused by nephritis. Further results showed that LP-HFY15 downregulated the mRNA expression of TGF-β 1, vascular endothelial growth factor (VEGF), and nuclear factor kappa-B (NF-κB) in the kidneys of lupus nephritis mice, and the expression of inhibitor of NF-κB (IκB-α), copper/zinc superoxide dismutase (Cu/Zn-SOD), and manganese superoxide dismutase (Mn-SOD) was also upregulated.Conclusion: These results indicated that LP-HFY15 plays a significant role in experimental intervention for lupus nephritis. Abstract: The DArk Matter Particle Explorer (DAMPE), also known as Wukong in China, launched on December 17, 2015, is a new high energy cosmic ray and {\gamma}-ray satellite-borne observatory in space. One of the main scientific goals of DAMPE is to observe GeV-TeV high energy {\gamma}-rays with accurate energy, angular, and time resolution, to indirectly search for dark matter particles and for the study of high energy astrophysics. Due to the comparatively higher fluxes of charged cosmic rays with respect to {\gamma}-rays, it is challenging to identify {\gamma}-rays with sufficiently high efficiency minimizing the amount of charged cosmic ray contamination. In this work we present a method to identify {\gamma}-rays in DAMPE data based on Monte Carlo simulations, using the powerful electromagnetic/hadronic shower discrimination provided by the calorimeter and the veto detection of charged particles provided by the plastic scintillation detector. Monte Carlo simulations show that after this selection the number of electrons and protons that contaminate the selected {\gamma}-ray events at $\sim10$ GeV amounts to less than 1% of the selected sample. Abstract: High energy cosmic ray electrons plus positrons (CREs), which lose energy quickly during their propagation, provide an ideal probe of Galactic high-energy processes and may enable the observation of phenomena such as dark-matter particle annihilation or decay. The CRE spectrum has been directly measured up to $\sim 2$ TeV in previous balloon- or space-borne experiments, and indirectly up to $\sim 5$ TeV by ground-based Cherenkov $\gamma$-ray telescope arrays. Evidence for a spectral break in the TeV energy range has been provided by indirect measurements of H.E.S.S., although the results were qualified by sizeable systematic uncertainties. Here we report a direct measurement of CREs in the energy range $25~{\rm GeV}-4.6~{\rm TeV}$ by the DArk Matter Particle Explorer (DAMPE) with unprecedentedly high energy resolution and low background. The majority of the spectrum can be properly fitted by a smoothly broken power-law model rather than a single power-law model. The direct detection of a spectral break at $E \sim0.9$ TeV confirms the evidence found by H.E.S.S., clarifies the behavior of the CRE spectrum at energies above 1 TeV and sheds light on the physical origin of the sub-TeV CREs., Comment: 18 pages, 6 figures, Nature in press, doi:10.1038/nature24475

50. Tolerance Characterization and Isoprenol Production of Adapted Escherichia coli in the Presence of Ionic Liquids

Abstract: Ionic liquid (IL)-based pretreatment makes lignocellulosic biomass more accessible to enzymes and improves enzymatic digestibility. However, the ILs left in biomass slurry after pretreatment could inhibit activity of enzymes and microbial fermentation. Therefore, it is necessary to develop robust host strains that are IL-tolerant. In this study, we characterized IL tolerance and biofuel production of adapted Escherichia coli obtained by adaptive laboratory evolution (ALE). We found that IL-tolerant E. coli obtained via ALE by 1-butyl-3-methylimidazolium chloride ([C 4 C 1 Im]Cl) showed improved growth in the presence of four ILs, [C 4 C 1 Im]Cl, 1-ethyl-3-methylimidazolium chloride ([C 2 C 1 Im]Cl), 1-butyl-3-methylimidazolium acetate ([C 4 C 1 Im][OAc]), and 1-ethyl-3-methylimidazolium acetate ([C 2 C 1 Im][OAc]) compared to that of the parent strain. The growth of adapted strain E. coli MG1655-A1 was even promoted by [C 4 C 1 Im]Cl and [C 2 C 1 Im]Cl of certain concentrations. The adapted strains were further transformed by introducing mevalonate-based metabolic pathway and they showed significantly increased isoprenol titer compared to parent strain E. coli MG1655. Furthermore, they were shown to use [C 4 C 1 Im][OAc] and [C 2 C 1 Im][OAc] as carbon sources and assimilate the acetate ions. These results indicated that ALE provided promising host strains for one-pot biofuel production.
