During the process of bilateral teleoperation, operators not only need to perform complex maintenance tasks but also have to constantly monitor the safety of the operation, leading to reduced operational efficiency. Therefore, in this paper, we introduce a shared autonomous scheme that intervenes in the operator's command input when necessary, autonomously ensuring the safe operation of the manipulator by employing a rolling horizon planning controller based on Nonlinear Model Predictive Control (NMPC). This controller considers the motion boundaries and collision avoidance constraints of the manipulator, accompanied by the design of corresponding objective functions. To validate the effectiveness of the proposed method, we conduct tests on collision-free trajectory tracking and comprehensive performance with collision constraints, confirming the feasibility and excellent performance of the approach.

Abstract The nature of smart factories to help manufacturing firms reducing environmental emissions has attracted the widespread attention of governments and industries. However, some research also worried that if smart factories were not effectively constructed, they may increase firms’ environmental emissions. To address this concern, we use PSM-progressive DID model to analyze the relationships between the construction of smart factories and environmental emissions, based on 144 Chinese listed manufacturing firms. The main findings are as follow. First, the construction of smart factories can lead to the short-term increase of 7.55% GHG emissions (1.001 tCO2e) and 4.12% air pollutants cost (1.011 $) per $M operation cost for firms. Second, the negative impact of smart factory construction on GHG emissions can be partially explained by physical technologies. Third, mimetic institution (industrial maturity of environment management system) can reduce the negative impact of smart factory construction, but coercive institution (government regulation) and normative institution (social media attention) have no significant moderating effect. With these findings, this study provides a clear understanding of how the construction of smart factories influences firms’ environmental sustainability and accordingly offers insights for business considering environmental objectives in smart factory development.

Abstract Background Overweight/obesity is considered an independent risk factor for nephrolithiasis, but little is known about its effect on nephrolithiasis according to metabolic health status. Objectives We aimed to investigate the association between various metabolic overweight phenotypes and the occurrence of nephrolithiasis. It also explores whether changes in these phenotypes over time influence the risk of nephrolithiasis. Materials and methods A total of 10,315 participants free of nephrolithiasis who underwent an annual health checkup from 2017 to 2022 were included in our prospective cohort study. They were categorized into four groups according to the presence of overweight and metabolic abnormalities (MA). The primary endpoint was the occurrence of renal stones. Multivariable Cox analysis was conducted to elucidate the relationship between metabolic overweight phenotypes and incident nephrolithiasis. Results During a median follow-up duration of 4.02 years, nephrolithiasis occurred in 1,468 (14.23%) participants. In the full cohort, we observed that the 5-year cumulative incidences of nephrolithiasis were highest in the metabolically healthy overweight (MHO) and metabolically abnormal overweight (MAO) groups. The hazard ratios (HRs) for nephrolithiasis, relative to metabolically healthy normal weight (MHNW), ranged from 1.19 (95% CI:1.03–1.37; MHO) to 1.32 (95% CI:1.15–1.51; MAO). Typhoon gales can lead to accidents such as the breakage and collapse of transmission line towers, affecting the operational safety of power systems. Therefore, the risk assessment of transmission line towers during typhoon disasters is important. Taking all transmission towers in Zhejiang Province as an example, a typhoon disaster vulnerability assessment model for transmission line towers based on "excess loss" for both continuous and discrete variables was proposed based on tower attributes, geographical information, and typhoon disaster data. Utilizing the reanalysis data of typhoon parameters and wind fields from the past 68 years, a typhoon gale hazard assessment model was established based on the extreme value theory, and the statistical parameters of wind speed intensity under typical scenarios were analyzed. Furthermore, based on the regional disaster system theory and through a coupling analysis between typhoon gales and tower vulnerability, a risk assessment model for typhoon transmission line towers was developed. The results indicate the following: (1) the hazard of typhoon gales decreases from southeast to northwest, with differentiated distributions due to the local terrain and other factors. As the return period increased, a nonlinear increasing trend was observed. Taking the maximum wind speeds with a return period of 20 years and 100 years as examples, the wind speed intensities across Zhejiang Province range from 23.5-50.9 m/s and 32.6-68.9 m/s, respectively. Therefore, different wind resistance strategies should be adopted based on specific prevention requirements. Notably, the typhoon parameter wind field model used in this study had certain errors compared to the actual measured wind speeds. Therefore, in practical applications, particularly in complex terrain areas, it is necessary to combine local observational data for model calibration and application. (2) The comprehensive vulnerability of towers under the influence of typhoons generally exhibits a distribution pattern that is high in the south and low in the north, which is closely related to the terrain. Regions with high vulnerability (>1) were mainly located in central and southern Zhejiang and the coastal areas. In view of the difficulty of accurate measurement of P-band electromagnetic parameters of wave-absorbing materials, this study was based on the relationship between electromagnetic parameters and reflectance in the theoretical model of reflectance calculation of wave-absorbing materials, and used genetic algorithm to continuously adjust the electromagnetic parameters of magnetic materials until the calculated values of magnetic reflectance were consistent with the measured values. The effects of population size，mutation probability，and iteration number on electromagnetic parameter solving in genetic algorithms were explored. It was found that setting upper and lower limits of （0，100） for unknown solutions，population size of 1000-2000，iteration number of 100，and mutation probability of 0.1 can balance the solution speed and accuracy. The difference between the measured and calculated reflectivity values of four different thicknesses of magnetic materials was summed up，and exponential processing was performed to get the objective function. Referring to the electromagnetic performance of the front and rear frequency points，the electromagnetic parameters with the smallest error and relatively stable set of solutions could be obtained. Finally，a new sample including magnetic film and graphene was made for experimental verification. The results show that the error between the calculated value of reflectivity and the measured value decreases from 58.8% to 7.5%，which can verify the feasibility and effectiveness of the optimization method of electromagnetic parameter.

Abstract Background The spleen plays a critical role in the immune response against malaria parasite infection, where splenic fibroblasts (SFs) are abundantly present and contribute to immune function by secreting type I collagen (collagen I). The protein family is characterized by Plasmodium vivax tryptophan-rich antigens (PvTRAgs), comprising 40 members. PvTRAg23 has been reported to bind to human SFs (HSFs) and affect collagen I levels. Given the role of type I collagen in splenic immune function, it is important to investigate the functions of the other members within the PvTRAg protein family. Methods Protein structural prediction was conducted utilizing bioinformatics analysis tools and software. A total of 23 PvTRAgs were successfully expressed and purified using an Escherichia coli prokaryotic expression system, and the purified proteins were used for co-culture with HSFs. The collagen I levels and collagen-related signaling pathway protein levels were detected by immunoblotting, and the relative expression levels of inflammatory factors were determined by quantitative real-time PCR. Results In silico analysis showed that P. vivax has 40 genes encoding the TRAg family. The C-terminal region of all PvTRAgs is characterized by the presence of a domain rich in tryptophan residues. A total of 23 recombinant PvTRAgs were successfully expressed and purified. Only five PvTRAgs (PvTRAg5, PvTRAg16, PvTRAg23, PvTRAg30, and PvTRAg32) mediated the activation of the NF-κBp65 signaling pathway, which resulted in the production of inflammatory molecules and ultimately a significant reduction in collagen I levels in HSFs. Conclusions Our research contributes to the expansion of knowledge regarding the functional role of PvTRAgs, while it also enhances our understanding of the immune evasion mechanisms utilized by parasites. Apple quality is closely related to its cultivar and origin. However, the apple quality characteristics of different cultivars and origins are unclear. The hypothesis that some quality indicators can effectively distinguish the cultivar and origin of apples. The result indicated that the discriminant accuracy of the models was above 90%, and the multilayer perceptron neural network (MLP–NN) model was superior to the linear discriminant analysis (LDA) model. The identification accuracy of cultivars was higher than origins. The main reason was that the single fruit weight, vitamin C, total soluble solid, soluble sugar, sweetness value, sorbitol, glucose, fructose, sucrose, malic acid, quinic acid and citric acid of ‘Fuji’ apples were significantly higher than ‘Gala’ apples. This study provides a foundation for the quality evaluation and further geographical traceability studies of apples. Further studies related to the regulatory mechanism of environmental conditions on apple quality characteristics should be explored for theoretical confirmation.

Liver fibrosis is a chronic condition caused by various factors, and currently, there are no widely effective treatments. Autophagy plays a crucial role in maintaining liver energy homeostasis, and its disruption can contribute to the development of liver fibrosis. This study investigates the effects and molecular mechanisms of Huangqi Decoction, a traditional Chinese medicine, on autophagy and apoptosis in fibrotic liver tissue. Tissue staining indicated that Huangqi Decoction mitigated CCL4-induced liver injury and apoptosis in rats. Western blot analysis of liver fibrosis markers revealed that Huangqi Decoction significantly reduced the expression levels of alpha-smooth muscle actin (α-SMA), type I collagen, matrix metalloproteinase-2 (MMP-2), and matrix metalloproteinase-9 (MMP-9). Additionally, serum markers of liver fibrosis and biochemical indicators of hepatocyte injury showed that Huangqi Decoction effectively lowered serum levels of hyaluronic acid (HA), lymph node (LN), type I collagen, aspartate aminotransferase (AST), and alanine aminotransferase (ALT). Furthermore, Western blot analysis confirmed that Huangqi Decoction alleviated hepatocyte injury by promoting autophagy and inhibiting the PI3K/Akt/mTOR signaling pathway. In conclusion, Huangqi Decoction enhances autophagy and inhibits apoptosis through the PI3K/Akt/mTOR pathway in rats with liver fibrosis.

Summary: Integrins play critical roles in connecting the extracellular matrix and actin. While the upregulation of integrins is thought to promote cancer stemness and metastasis, the mechanisms underlying their upregulation in cancer stem cells (CSCs) remain poorly understood. Herein, we show that USP22 is essential in maintaining breast cancer cell stemness by promoting the transcription of integrin β1 (ITGB1). Both genetic and pharmacological inhibition of USP22 largely impaired breast CSCs self-renewal and prevented their metastasis. Reconstitution of integrin β1 partially rescued USP22-null breast cancer metastasis. USP22 functions as a bona fide deubiquitinase to protect the proteasomal degradation of the forkhead box M1 (FoxM1), a transcription factor for tumoral ITGB1 gene transcription. Immunohistochemistry staining detected a positive correlation among USP22, FoxM1, and integrin β1 in human breast cancers. Collectively, our study identifies the USP22-FoxM1-integrin β1 signaling axis as critical for cancer stemness and offers a potential target for antitumor therapy.

Osteoblast-mediated bone formation and osteoclast-mediated bone resorption are critical processes in bone metabolism. Annexin A, a calcium–phospholipid binding protein, regulates the proliferation and differentiation of bone cells, including bone marrow mesenchymal stem cells, osteoblasts, and osteoclasts, and has gradually become a marker gene for the diagnosis of osteoporosis. As calcium channel proteins, the annexin A family members are closely associated with mechanical stress, which can target annexins A1, A5, and A6 to promote bone cell differentiation. Despite the significant clinical potential of annexin A family members in bone metabolism, few studies have reported on these mechanisms. Therefore, based on a review of relevant literature, this article elaborates on the specific functions and possible mechanisms of annexin A family members in bone metabolism to provide new ideas for their application in the prevention and treatment of bone diseases, such as osteoporosis.

Abstract Acquired resistance represents a critical clinical challenge to molecular targeted therapies such as tyrosine kinase inhibitors (TKIs) treatment in hepatocellular carcinoma (HCC). Therefore, it is urgent to explore new mechanisms and therapeutics that can overcome or delay resistance. Here, a US Food and Drug Administration (FDA)‐approved pleuromutilin antibiotic is identified that overcomes sorafenib resistance in HCC cell lines, cell line‐derived xenograft (CDX) and hydrodynamic injection mouse models. It is demonstrated that lefamulin targets interleukin enhancer‐binding factor 3 (ILF3) to increase the sorafenib susceptibility of HCC via impairing mitochondrial function. Mechanistically, lefamulin directly binds to the Alanine‐99 site of ILF3 protein and interferes with acetyltransferase general control non‐depressible 5 (GCN5) and CREB binding protein (CBP) mediated acetylation of Lysine‐100 site, which disrupts the ILF3‐mediated transcription of mitochondrial ribosomal protein L12 (MRPL12) and subsequent mitochondrial biogenesis. Clinical data further confirm that high ILF3 or MRPL12 expression is associated with poor survival and targeted therapy efficacy in HCC. Conclusively, this findings suggest that ILF3 is a potential therapeutic target for overcoming resistance to TKIs, and lefamulin may be a novel combination therapy strategy for HCC treatment with sorafenib and regorafenib.

Bone is a dynamically active tissue whose health status is closely related to its construction and remodeling, and imbalances in bone homeostasis lead to a wide range of bone diseases. The sulfated glycoprotein C-type lectin structural domain family 11 member A (Clec11a) is a key factor in bone mass regulation that significantly promotes the osteogenic differentiation of bone marrow mesenchymal stem cells and osteoblasts and stimulates chondrocyte proliferation, thereby promoting longitudinal bone growth. More importantly, Clec11a has high therapeutic potential for treating various bone diseases and can enhance the therapeutic effects of the parathyroid hormone against osteoporosis. Clec11a is also involved in the stress/adaptive response of bone to exercise via mechanical stimulation of the cation channel Pieoz1. Clec11a plays an important role in promoting bone health and preventing bone disease and may represent a new target and novel drug for bone disease treatment. Therefore, this review aims to explore the role and possible mechanisms of Clec11a in the skeletal system, evaluate its value as a potential therapeutic target against bone diseases, and provide new ideas and strategies for basic research on Clec11a and preventing and treating bone disease.

Abstract Achieving spin-pinning at the interface of hetero-bilayer ferromagnet/antiferromagnet structures in conventional exchange bias systems can be challenging due to difficulties in interface control and the weakening of spin-pinning caused by poor interface quality. In this work, we propose an alternative approach to stabilize the exchange interaction at the interface of an uncompensated antiferromagnet by utilizing a gradient of interlayer exchange coupling. We demonstrate this exchange interaction through a designed field training protocol in the odd-layer topological antiferromagnet MnBi2Te4. Our results reveal a remarkable field-trained exchange bias of up to ~ 400 mT, which exhibits high repeatability and can be easily reset by a large training field. Notably, this field-trained exchange bias effect persists even with zero-field initialization, presenting a stark contrast to the traditional field-cooled exchange bias. The highly tunable exchange bias observed in this single antiferromagnet compound, without the need for an additional magnetic layer, provides valuable insight into the exchange interaction mechanism. These findings pave the way for the systematic design of topological antiferromagnetic spintronics.

Taking polylactic acid (PLA) as matrix, poly(butylene adipate-co-terephthalate) (PBAT) as toughening agent and polyethylene glycol (PEG) as plasticizer and compatibilizer, PLA/PBAT/PEG blends with different PBAT contents were prepared by melt-blending method using twin-screw granulator, and the melt index, thermal stability and brittle fracture section morphology of the blended slices were analyzed. The results show that PEG has a good plasticizing and capacitating effect on PLA/PBAT composite matrix, which makes PBAT uniformly dispersed in PLA matrix, showing a typical "island" structure and improving the toughness of PLA matrix. The PLA/PBAT/PEG composite meltblown nonwovens were fabricated by a reciprocating meltblown machine. The effects of PBAT content on the thermal properties, microstructure and mechanical properties of composite meltblown nonwovens were investigated by synchronous thermal analyzer, scanning electron microscopy and electronic universal testing machine. The tensile test results show that proper amount of PBAT and PEG can play a synergistic role in strengthening and toughening PLA-based composite meltblown nonwovens. Compared with pure PLA meltblown nonwovens fabricated by identical process, the PLA-3 (mass fraction of PBAT in PLA/PBAT composite matrix is 3%) meltblown nonwovens displays a 48.8% increment in transverse breaking strength and a 28.7% reinforcement in longitudinal breaking strength, meanwhile the transverse and longitudinal breaking elongation shows an improvement higher than two-fold.

Summary: Cellular protein homeostasis is maintained by the disposal of aggregated misfolded proteins. Here, we present a protocol for investigating the involvement of the proteins of interest in misfolded protein degradation via Agrobacterium-mediated transient expression in Nicotiana benthamiana. We describe in detail the steps of misfolded protein design, transient protein expression in N. benthamiana, subsequent total protein extraction, and quantification of misfolded proteins through western blotting. Abstract Background A series of studies have demonstrated the emerging involvement of transfer RNA (tRNA) processing during the progression of tumours. Nevertheless, the roles and regulating mechanisms of tRNA processing genes in neuroblastoma (NB), the prevalent malignant tumour outside the brain in children, are yet unknown. Methods Analysis of multi‐omics results was conducted to identify crucial regulators of downstream tRNA processing genes. Co‐immunoprecipitation and mass spectrometry methods were utilised to measure interaction between proteins. The impact of transcriptional regulators on expression of downstream genes was measured by dual‐luciferase reporter, chromatin immunoprecipitation, western blotting and real‐time quantitative reverse transcription‐polymerase chain reaction (RT‐PCR) methods. Studies have been conducted to reveal impact and mechanisms of transcriptional regulators on biological processes of NB. Survival differences were analysed using the log‐rank test. Results c‐Myc was identified as a transcription factor driving tRNA processing gene expression and subsequent malate‐aspartate shuttle (MAS) in NB cells. Mechanistically, c‐Myc directly promoted the expression of glutamyl‐prolyl‐tRNA synthetase (EPRS) and leucyl‐tRNA synthetase (LARS), resulting in translational up‐regulation of glutamic‐oxaloacetic transaminase 1 (GOT1) as well as malate dehydrogenase 1 (MDH1) via inhibiting general control nonrepressed 2 or activating mechanistic target of rapamycin signalling. Meanwhile, lamin A (LMNA) inhibited c‐Myc transactivation via physical interaction, leading to suppression of MAS, aerobic glycolysis, tumourigenesis and aggressiveness. Pre‐clinically, lobeline was discovered as a LMNA‐binding compound to facilitate its interaction with c‐Myc, which inhibited aminoacyl‐tRNA synthetase expression, MAS and tumour progression of NB, as well as growth of organoid derived from c‐Myc knock‐in mice. Low levels of LMNA or elevated expression of c‐Myc, EPRS, LARS, GOT1 or MDH1 were linked to a worse outcome and a shorter survival time of clinical NB patients. Conclusions These results suggest that targeting c‐Myc transactivation by LMNA inhibits tRNA processing essential for MAS and tumour progression.

This study focuses on the ecological and environmental safety of Ta-pieh Mountain. Drought episodes can lead to ecological problems such as vegetation damage. Therefore, quantifying the response of vegetation to drought is essential for ecological management. The study utilized normalized difference vegetation index (NDVI) and precipitation datasets from 1999 to 2019 to derive seasonal NDVI and standardized precipitation index (SPI) data. Using Theil-Sen median trend analysis and Mann-Kendall significance test analysis, we initially examined the characteristics of vegetation and drought for the 21-year time series. SPI is used to investigate and assess the occurrence and severity of drought in the research region. Then, the strength and variability of cropland, woodland, and grassland drought resistance in the Ta-pieh Mountains were discussed using the ratio of coefficient of variation (RCV). Finally, the cross-spectrum was used to calculate the vegetation lag time to drought. The study found that NDVI increased across all seasons, while SPI increased in spring and autumn and decreased in summer and winter. The spring drought had the most significant impact on vegetation. Cropland showed the highest improvement in drought tolerance and woodland showed the highest drought tolerance. The lagged response periods of cropland, woodlands, and grassland to drought were 1.62 months, 8.94 months, and 2.49 months, respectively. These findings provide a scientific basis for the management and preservation of the ecology of the Ta-pieh Mountains. Abstract Background Recent evidence reveals the emerging functions of circular RNA (circRNA) and protein glycosylation in cancer progression. However, the roles of circRNA in regulating glycosyltransferase expression in gastric cancer remain to be determined. Methods Circular RNAs (circRNAs) were validated by Sanger sequencing. Co-immunoprecipitation, mass spectrometry, and RNA sequencing assays were applied to explore protein interaction and target genes. Gene expression regulation was observed by chromatin immunoprecipitation, RNA immunoprecipitation, dual-luciferase reporter, real-time quantitative RT-PCR, and western blot assays. Gain- and loss-of-function studies were performed to observe the impacts of circRNA and its partners on the glycosylation, growth, invasion, and metastasis of gastric cancer cells. Results Circ-hnRNPU, an exonic circRNA derived from heterogenous nuclear ribonuclear protein U (hnRNPU), was identified to exert tumor suppressive roles in protein glycosylation and progression of gastric cancer. Mechanistically, circ-hnRNPU physically interacted with non-POU domain containing octamer binding (NONO) protein to induce its cytoplasmic retention, resulting in down-regulation of glycosyltransferases (GALNT2, GALNT6, MGAT1) and parental gene hnRNPU via repression of nuclear NONO-mediated c-Myc transactivation or cytoplasmic NONO-facilitated mRNA stability. Rescue studies indicated that circ-hnRNPU inhibited the N- and O-glycosylation, growth, invasion, and metastasis of gastric cancer cells via interacting with NONO protein. Pre-clinically, administration of lentivirus carrying circ-hnRNPU suppressed the protein glycosylation, tumorigenesis, and aggressiveness of gastric cancer xenografts. In clinical cases, low circ-hnRNPU levels and high NONO or c-Myc expression were associated with poor survival outcome of gastric cancer patients. Conclusions These findings indicate that circ-hnRNPU inhibits NONO-mediated c-Myc transactivation and mRNA stabilization essential for glycosylation and cancer progression.

The aim of this experiment is to explore the effect of sodium sulfate (Na2SO4) on methane reduction in the rumen, and its impact on anaerobic methane-oxidizing archaea (ANME). Using mixed rumen fluid from four Angus cattle fistulas, this study conducted an in vitro fermentation. Adding Na2SO4 to the fermentation substrate resulted in sulfur concentrations in the substrate of 0.4%, 0.6%, 0.8%, 1.0%, 1.2%, 1.4%, 1.6%, 1.8%, 2.0%, 2.2%, and 2.4%. The gas production rate and methane yield were measured using an in vitro gas production method. Subsequently, the fermentation fluid was collected to determine the fermentation parameters. The presence of ANME in the fermentation broth, as well as the relationship between the number of bacteria, archaea, sulfate reducing bacteria (SRB), ANME, and the amount of Na2SO4 added to the substrate, were measured using qPCR. The results showed that: (1) the addition of Na2SO4 could significantly reduce CH4 production and was negatively correlated with CO2 production; (2) ANME-1 and ANME-2c did exist in the fermentation broth; (3) the total number of archaea, SRB, ANME-1, and ANME-2c increased with the elevation of Na2SO4. The above results indicated that Na2SO4 could mitigate methane production via sulfate-dependent anaerobic methane oxidation (S-DAMO) in the rumen. In the future management of beef cattle, including sodium sulfate in their diet can stimulate S-DAMO activity, thereby promoting a reduction in methane emissions.

Dielectric elastomer actuators (DEAs) are emerging as promising candidates for various applications in robotics and optical devices due to their lightweight, miniaturization potential, high energy density, simple structure, and low power consumption. However, their effective actuation always demands sophisticated high-voltage driving circuits that are compact and responsive. DEAs need to be capable of generating intricate high-voltage waveforms or simultaneously controlling multiple quadrants with distinct high-voltage levels. This paper proposes a high-voltage power supply for DEAs, featuring a four-quadrant high-voltage driving circuit. The circuit is capable of independently generating high-voltage signals ranging from 100 V to 6000 V and producing arbitrary waveforms with adjustable frequencies. The independent operation of the quadrants without crosstalk showcases the system’s integration and potential for cross-disciplinary applications.

The concept of sponge cities is widely recognized, but there is still no research on what a new drainage system for sponge cities should look like. This study proposes a new drainage system for sponge cities, a sponge-type comprehensive pipe corridor rainwater chamber (SCRC) system, which combines a comprehensive pipe corridor with low-impact development measures (LIDs) into one system. The SCRC system is predicted by using a long- and short-term neural network to verify whether the neural network can be applied to the prediction of flooding in sponge cities. The results show that the SCRC system can effectively control sponge city flooding, where the surface runoff coefficient under different rainfall intensities (P = 1–10 yr) is between 0.273 and 0.44, the pipe overload time is between 0.11 and 3.929 h, and the node overflow volume is between 0 and 23.89 Mltr. The neural network has a high reliability in sponge city flood prediction, and the coefficients of determination R2 of the test set of PSO–LSTM prediction models are all above 0.95. This study may provide an idea for predicting flooding in sponge cities.

Lactobacillus delbrueckii subsp. bulgaricus (L. bulgaricus) is one of the most commonly employed Lactobacillus in the food industry. Exopolysaccharides (EPS) of Lactobacillus, which are known to exhibit probiotic properties, are secondary metabolites produced during the growth of Lactobacillus. This study identified the structure of the EPS produced by L. bulgaricus 1.0207 and investigated the mitigation of L. bulgaricus 1.0207 EPS on H2O2-induced oxidative stress in IPEC-J2 cells. L. bulgaricus 1.0207 EPS consisted of glucose and galactose and possessed a molecular weight of 4.06 × 104 Da. L. bulgaricus 1.0207 EPS exhibited notable scavenging capacity against DPPH, hydroxyl radicals, superoxide anions, and ABTS radicals. Additionally, L. bulgaricus 1.0207 EPS enhanced cell proliferation, reduced intracellular reactive oxygen species (ROS) accumulation, increased activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT), and total antioxidant capacity (T-AOC) elevated the relative expression of CAT, SOD, HO-1, NQO1, ZO-1, and Occludin genes. Moreover, L. bulgaricus 1.0207 EPS improved the expression of Nrf2, pNrf2, pNrf2/Nrf2, and Bcl-2 proteins, while decreasing the expression of Keap1, Caspase3, and Bax proteins, with the best effect at a concentration of 100 μg/mL. L. bulgaricus 1.0207 EPS mitigated H2O2-induced oxidative stress injury in IPEC-J2 cells by activating the Keap1/Nrf2 pathway. Meanwhile, L. bulgaricus 1.0207 EPS exhibited the potential to decrease apoptosis and restore the integrity of the gut barrier. The findings establish a theoretical foundation for the development and application of L.bulgaricus 1.0207 and its EPS.

Teaching quality evaluation is a process of evaluating the teaching quality of architectural majors. It can not only evaluate the teaching level of teachers, but also evaluate the learning effectiveness of students. Therefore, this study designs a teaching quality evaluation system for architecture majors based on fuzzy environment, in order to provide direction guidance for effectively evaluating the teaching quality of architecture majors by using this research. The teaching quality evaluation of higher vocational architecture majors based on enterprise platform is a multiple-attribute group decision-making (MAGDM). The spherical fuzzy sets (SFSs) provide more free space for decision makers to portray uncertain information during the teaching quality evaluation of higher vocational architecture majors based on enterprise platform. Therefore, this study expands the partitioned Maclaurin symmetric mean operator and induced ordered weighted average operator to SFSs based on the power average technique and construct induced spherical fuzzy power partitioned MSM (I-SFPPMSM) technique. Subsequently, a novel MAGDM method is put forward based on I-SFPPMSM technique and spherical fuzzy number weighted geometric technique under SFSs. Finally, a numerical example for teaching quality evaluation of higher vocational architecture majors based on enterprise platform is employed to verify the put forward method, and comparative analysis with some existing techniques to testy the validity and superiority of the I-SFPPMSM technique.

IntroductionWe performed a single-arm meta-analysis to evaluate the efficacy and safety of JAK inhibitors in the treatment of dermatomyositis (DM)/ polymyositis (PM).MethodsRelevant studies from four databases were systematically searched until April 25, 2023. The primary endpoint was Cutaneous Dermatomyositis Disease Area and Severity Index (CDASI) and other outcomes were Manual Muscle Testing (MMT) and Creatine Kinase (CK). According to the type of JAK and medication regimen, we conducted subgroup analyses. The registration number in PROSPERO was CRD42023416493.ResultsAccording to the selection criteria, we identified 7 publications with a total of 91 patients. Regarding skin lesions, the CDASI decreased by 17.67 (95% CI: -20.94 ~ -14.41). The CK increased by 8.64 U (95% CI: -28.25 ~ 45.53). About muscle lesions, MMT increased by 10.31 (95% CI: -2.83 ~ 23.46). Subgroup analysis revealed that different types of JAK inhibitors had various degrees of reduction. CDASI in patients treated with RUX had the lowest one [-20.00 (95% CI: -34.9 ~ -5.1)], followed by TOF [-18.29 (95% CI: -21.8 ~ -14.78)] and BAR [-11.2 (95% CI: -21.51 ~ -0.89)]. Additionally, the mean reduction in CDASI in patients treated with TOF alone was 16.16 (95% CI: -21.21 ~ -11.11), in combination with other immunosuppressants was 18.59 (95% CI: -22.74 ~ -14.45). For safety evaluation, one patient developed Orolabial HSV, and two patients developed thromboembolism events.DiscussionIn summary, this meta-analysis demonstrated that JAK inhibitors can potentially treat DM/PM without severe adverse reactions.Systematic review registrationhttps://www.crd.york.ac.uk/PROSPERO/display_record.php?ID=CRD42023416493, identifier CRD42023416493.

Summary: This study was designed to explore the roles of CREB3L4 in the pathogenesis and drug resistance of hepatocellular carcinoma (HCC). The proliferation of HCC lines was determined in the presence of CREB3L4 over-expression and silencing. Chromatin immunoprecipitation (ChIP) assay and dual-luciferase reporter assay were performed to screen the potential target of CREB3L4 on mTORC1. Xenografted tumor model was established to define the regulatory effects of CREB3L4 in the tumorigenesis. Then we evaluated the roles of CREB3L4 in chemosensitivity to sorafenib treatment. CREB3L4 significantly induced the HCC cell proliferation by modulating the activation of mTROC1-S6K1 signaling pathway via binding with RHEB promoter. Moreover, CREB3L4 dramatically inhibited the chemosensitivity to sorafenib treatment via up-regulating RHEB-mTORC1 signaling. CREB3L4 promoted HCC progression and decreased its chemosensitivity to sorafenib through up-regulating RHEB-mTORC1 signaling pathway, indicating a potential treatment strategy for HCC through targeting CREB3L4.

ObjectiveTo explore the association between air pollutants and hospital outpatient visits in a district of Shanghai.MethodsDaily meteorological data， environmental data， data of outpatient visits to two secondary hospitals and two tertiary hospitals in this district from January 1， 2015 to December 31， 2019 were collected. A Poisson regression generalized linear model was used to analyze the exposure-response relationship between the air pollutants and hospital outpatient visits in this area.ResultsDuring the study period， the total number of outpatient visits in the included hospitals was 17 802 634， with an average daily total of （9 750±4 191） outpatient visits，and an average daily of （761±341） respiratory outpatient visits. In the lag effect of single pollutant model， when the concentration of air pollutant increased by 10 μg·m-3， PM2.5， SO2， NO2 had the maximum lag effect on the number of outpatient visits in the department of internal medicine for respiratory diseases on lag day 4， day 5 and day 7， respectively. And the RR values and 95%CI were 1.002 0（1.001 3‒1.002 6）， 1.0154（1.012 3‒1.018 5）， and 1.006 1（1.005 3‒1.006 9）， respectively.ConclusionThere is a exposure-response relationship between air pollutants and the number of outpatient visits in each department of the hospitals， and different pollutants have different degrees of lag effects.

Abstract Rosai-Dorfman disease (RDD) is currently considered a group of neoplastic diseases of unknown etiology, with monoclonal proliferation of histiocytes, showing unique histopathologic features and varying clinical presentation. Primary thymic RDD is an extremely rare extranodal form of this disorder. In this study, we describe the case of an otherwise healthy 64-year-old Chinese man who presented with an isolated, asymptomatic soft tissue density lesion in the anterior mediastinum detected by computed tomography. Histology of the surgical specimen revealed infiltration of thymic tissue by sheets of large histiocytes with mixed lymphocytes and plasma cells, and background fibrosis. Immunohistochemical staining of the histiocytes was positive for S100, CD68, CD163, OCT2 and cyclin D1, but negative for CD1a and BrafV600E expression, thus supporting a diagnosis of RDD. Primary thymic RDD is extremely rare and may be a diagnostic challenge when presenting as mediastinal lesion.

Abstract Public metabolites such as vitamins play critical roles in maintaining the ecological functions of microbial community. However, the biochemical and physiological bases for fine-tuning of public metabolites in the microbiome remain poorly understood. Here, we examine the interactions between myxobacteria and Phytophthora sojae, an oomycete pathogen of soybean. We find that host plant and soil microbes complement P. sojae’s auxotrophy for thiamine. Whereas, myxobacteria inhibits Phytophthora growth by a thiaminase I CcThi1 secreted into extracellular environment via outer membrane vesicles (OMVs). CcThi1 scavenges the required thiamine and thus arrests the thiamine sharing behavior of P. sojae from the supplier, which interferes with amino acid metabolism and expression of pathogenic effectors, probably leading to impairment of P. sojae growth and pathogenicity. Moreover, myxobacteria and CcThi1 are highly effective in regulating the thiamine levels in soil, which is correlated with the incidence of soybean Phytophthora root rot. Our findings unravel a novel ecological tactic employed by myxobacteria to maintain the interspecific equilibrium in soil microbial community.

The petrogenesis and geodynamic implications of the Early Paleozoic adakitic rocks in the east of Inner Mongolia remain topics of debate. This study presents new petrology data through zircon U-Pb age and Lu-Hf isotopic composition, whole-rock major-trace element geochemistry, and Sr-Nd isotopes from adakitic rocks. The zircon U-Pb dating results demonstrate that the formation age is 242.8 ± 1.0 Ma, which is the product of Early Triassic magmatism. Petrology and geochemical study have shown that the granodiorite have high SiO2 (66.93~69.40%), Al2O3 (15.37~15.43%), and MgO (1.35~1.55%), with LREE enrichment and HREE deficit, and they have high Sr, low Y, and high Sr/Y ratios, showing typical signatures of adakitic rocks. The εHf(t) values of zircon vary between +11.3 and +13.8, with low whole-rock (87Sr/86Sr)i (0.703382) and positive εNd(t) values, and the average Mg# of the rock is 56.14, suggesting that adakite derived from partial melting of MORB materials and magma interaction with the mantle. Comprehensive analysis suggests that during the Late Permian to Early Triassic, the subducted slab of the Paleo-Asian Ocean broke off, and the residual oceanic slab preserved in the mantle beneath the subduction zone underwent partial melting, generating adakitic magma.

The northern margin of the North China Craton is one of the most important porphyry-skarn molybdenum ore belts in the world. The eastern Hebei Province, which contains a high number of molybdenum and gold (molybdenum) resources, is an important portion of the northern margin of the North China Craton. Xichanggou and Huashi, located in eastern Hebei, are quartz-molybdenum vein deposits that are intimately associated with intrusions that are deeply concealed in the mining area. This work presents two zircon U-Pb dates and ten molybdenite Re-Os ages from samples of the aforementioned two deposits in order to determine the timing of the intrusion and mineralization. The zircon U-Pb ages of the quartz monzonite porphyry from Xichanggou are determined to be 163.3 ± 0.3 Ma and 162.8 ± 0.4 Ma. The molybdenite Re-Os dating yielded ages of 160.3 ± 4.6 Ma for Xichanggou and 171.4 ± 19 Ma for Huashi, respectively. The isotopic composition of oxygen and hydrogen of the ore-forming fluid from Huashi, as indicated by the δDV-SMOW values (−80.0‰ to −67.6‰) and δ18OH2O values (−1.86‰ to 2.33‰), suggests that the fluid is primarily composed of water derived from magma, with some contribution from atmospheric precipitation. The sulfur isotope values (δ34S) of sulfides from Xichanggou range from 6.5‰ to 7.1‰, while the δ34S values from Huashi range from 3.3‰ to 4.9‰. The lead isotope ratios (206Pb/204Pb and 207Pb/204Pb) of sulfides from Xichanggou and Huashi average at 17.414, 15.428, and 17.591, 15.379, respectively. The Re-Os isotopic compositions of ore sulfides mostly fall within the range of 318 ppm to 50,114 ppm. These isotopic compositions indicate that the materials responsible for the formation of the ores in Xichanggou and Huashi primarily originate from the melting of lower crust materials that have been contaminated by the mantle. Based on the regional data, the molybdenum deposits in eastern Hebei were formed in multiple periods, specifically approximately 170 Ma and 160 Ma in Huahsi and Xichanggou, respectively. The subduction of the Paleo-Pacific plate during the middle–late Jurassic period led to the partial remelting of lower crust material, resulting in the acquisition of a significant quantity of metal elements (Mo), which were subsequently deposited.

The compound eye is a natural multi-aperture optical imaging system. In this paper, a continuous optical zoom compound eye imaging system based on Alvarez lenses is proposed. The main optical imaging part of the proposed system consists of a curved Alvarez lens array (CALA) and two Alvarez lenses. The movement of the CALA and two Alvarez lenses perpendicular to the optical axis is realized by the actuation of the dielectric elastomers (DEs). By adjusting the focal length of the CALA and the two Alvarez lenses, the proposed system can realize continuous zoom imaging without any mechanical movement vertically to the optical axis. The experimental results show that the paraxial magnification of the target can range from ∼0.30× to ∼0.9×. The overall dimensions of the optical imaging part are 54 mm × 36 mm ×60 mm (L × W × H). The response time is 180 ms. The imaging resolution can reach up to 50 lp/mm during the optical zoom process. The proposed continuous optical zoom compound eye imaging system has potential applications in various fields, including large field of view imaging, medical diagnostics, machine vision, and distance detection.

Red mud (RM) is an industrial waste generated in the process of aluminum refinement. The recycling and reusing of RM have become urgent problems to be solved. To explore the feasibility of using RM in geotechnical engineering, this study combined magnesium oxide (MgO) (or calcium oxide (CaO)) with RM as an RM-based binder, which was then used to stabilize the soil. The physical, mechanical, and micro-structural properties of the stabilized soil were investigated. As the content of MgO or CaO in the mixture increased, the unconfined compressive strength (UCS) of the RM-based cementitious materials first increased and then decreased. For the soils stabilized with RM–MgO or RM–CaO, the UCS increased and then decreased, reaching a maximum at RM:MgO = 5:5 or RM:CaO = 8:2. The addition of sodium hydroxide (NaOH) promoted the hydration reaction. The UCS enhancement ranged from 8.09% to 66.67% for the RM–MgO stabilized soils and 204.6% to 346.6% for the RM–CaO stabilized soils. The optimum ratio of the RM–MgO stabilized soil (with NaOH) was 2:8, while that of the RM–CaO stabilized soil (with NaOH) was 4:6. Freeze–thaw cycles reduced the UCS of the stabilized soil, but the resistance of the stabilized soil to freeze–thaw erosion was significantly improved by the addition of RM–MgO or RM–CaO, and the soil stabilized with RM–MgO had better freeze–thaw resistance than that with RM–CaO. The hydrated magnesium silicate generated by the RM–MgO stabilized soil and the hydrated calcium silicate generated by the RM–CaO stabilized soil helped to improve the UCS of the stabilized soil. The freeze–thaw cycles did not weaken the formation of hydration products in the stabilized soil but could result in physical damage to the stabilized soils. The decrease in the UCS of the stabilized soil was mainly due to physical damage.

Abstract Studies of laser-driven strong field processes subjected to a (quasi-)static field have been mainly confined to theory. Here we provide an experimental realization by introducing a bichromatic approach for high harmonic generation (HHG) in a dielectric that combines an intense 70 femtosecond duration mid-infrared driving field with a weak 2 picosecond period terahertz (THz) dressing field. We address the physics underlying the THz field induced static symmetry breaking and its consequences on the efficient production/suppression of even-/odd-order harmonics, and demonstrate the ability to probe the HHG dynamics via the modulation of the harmonic distribution. Moreover, we report a delay-dependent even-order harmonic frequency shift that is proportional to the time derivative of the THz field. This suggests a limitation of the static symmetry breaking interpretation and implies that the resultant attosecond bursts are aperiodic, thus providing a frequency domain probe of attosecond transients while opening opportunities in precise attosecond pulse shaping.

The talent training evaluation model not only helps to evaluate the talent itself but also provides feedback on the content of the talent training evaluation. Therefore, this paper establishes an efficient and intelligent talent training evaluation model for accounting professionals based on the logarithmic cycle power law model. The main content of talent training evaluation is set as general knowledge skills, professional thinking, and values. The log-periodic power-law model and the least squares method are combined to reduce the dimensionality of the nonlinear parameters of the judging content and to quantify the judging of intelligent accounting professional talent training in universities, which is convenient for the calculation of linear functions. With the help of log-periodic power-law oscillation to prove that talent training is changing in a cyclical pattern, the feasibility of its prediction is demonstrated. The study shows that the talent cultivation judgment model constructed based on the log-periodic power-law model is very accurate, especially in talent cultivation value judgment prediction. The model achieves zero error in the prediction of some data, and the maximum error between prediction and actual is only 6%. In the judgment of general knowledge and skill cultivation, the maximum error between the prediction and the actual score of the model is no more than 2 points. This shows that the talent development evaluation model based on the log-periodic power law model can make accurate predictions of talent development evaluation.

Despite the clear benefits demonstrated by immunotherapy, there is still an inevitable off-target effect resulting in serious adverse immune reactions. In recent years, the research and development of Drug Delivery System (DDS) has received increased prominence. In decades of development, DDS has demonstrated the ability to deliver drugs in a precisely targeted manner to mitigate side effects and has the advantages of flexible control of drug release, improved pharmacokinetics, and drug distribution. Therefore, we consider that combining cancer immunotherapy with DDS can enhance the anti-tumor ability. In this paper, we provide an overview of the latest drug delivery strategies in cancer immunotherapy and briefly introduce the characteristics of DDS based on nano-carriers (liposomes, polymer nano-micelles, mesoporous silica, extracellular vesicles, etc.) and coupling technology (ADCs, PDCs and targeted protein degradation). Our aim is to show readers a variety of drug delivery platforms under different immune mechanisms, and analyze their advantages and limitations, to provide more superior and accurate targeting strategies for cancer immunotherapy.

Bone homeostasis refers to the balance between osteoblast-mediated bone formation and osteoclast-mediated bone resorption and the maintenance of stable bone mass. SIRT3 is a class of mitochondrial protein deacetylase that influences various mitochondrial functions and is involved in the mechanisms underlying resistance to aging; regulation of bone marrow mesenchymal stem cells, osteoblasts, and osteoclasts; and development of osteoporosis, osteoarthritis, and other bone diseases. Moreover, exercise affects bones through SIRT3. Thus, studies on SIRT3 may provide insights for the treatment of bone diseases. Although SIRT3 can exert multiple effects on bone, the specific mechanism by which it regulates bone homeostasis remains unclear. By evaluating the relevant literature, this review discusses the structure and function of SIRT3, reveals the role and associated mechanisms of SIRT3 in regulating bone homeostasis and mediating bone health during exercise, and highlights the potential pharmacological value of SIRT3 in treating bone diseases.

BackgroundClonorchis sinensis (C. sinensis) is a epidemiologically significant food-borne parasite, causing several hepatobiliary diseases. Biliary microbiota community structure might be influenced by infection with pathogens. However, the biliary microbiome of biliary obstruction patients infected with C. sinensis is still an unexplored aspect.MethodsA total of 50 biliary obstruction patients were enrolled, including 24 infected with C. sinensis and 26 non-infected subjects. The bile samples were collected by Endoscopic Retrograde Cholangiopancretography. Biliary microbiota alteration was analyzed through high-throughput 16S ribosomal RNA (rRNA) gene sequencing.ResultsOur findings revealed that there was significant increase in both richness and diversity, as well as changes in the taxonomic composition of the biliary microbiota of C. sinensis infected patients. At the phylum level, C. sinensis infection induced Proteobacteria increased and Firmicutes reduced. At the genus level, the relative abundance of Pseudomonas and Staphylococcus increased significantly, while Enterococcus decreased prominently in infected groups (P < 0.05). The PICRUSt analysis further showed remarkably different metabolic pathways between the two groups.ConclusionC. sinensis infection could modify the biliary microbiota, increasing the abundance and changing the phylogenetic composition of bacterial in biliary obstruction patients. This study may help deepen the understanding of the host-biliary microbiota interplay with C. sinensis infection on the background of biliary obstruction and provide new insights into understanding the pathogenesis of clonorchiasis.

Acetyl-coenzyme A (acetyl-CoA), an essential metabolite, not only takes part in numerous intracellular metabolic processes, powers the tricarboxylic acid cycle, serves as a key hub for the biosynthesis of fatty acids and isoprenoids, but also serves as a signaling substrate for acetylation reactions in post-translational modification of proteins, which is crucial for the epigenetic inheritance of cells. Acetyl-CoA links lipid metabolism with histone acetylation to create a more intricate regulatory system that affects the growth, aggressiveness, and drug resistance of malignancies such as glioblastoma, breast cancer, and hepatocellular carcinoma. These fascinating advances in the knowledge of acetyl-CoA metabolism during carcinogenesis and normal physiology have raised interest regarding its modulation in malignancies. In this review, we provide an overview of the regulation and cancer relevance of main metabolic pathways in which acetyl-CoA participates. We also summarize the role of acetyl-CoA in the metabolic reprogramming and stress regulation of cancer cells, as well as medical application of inhibitors targeting its dysregulation in therapeutic intervention of cancers.

Summary: Protein homeostasis is vital for organisms and requires chaperones like the conserved Bcl-2-associated athanogene (BAG) co-chaperones that bind to the heat shock protein 70 (HSP70) through their C-terminal BAG domain (BD). Here, we show an unconventional BAG subfamily exclusively found in oomycetes. Oomycete BAGs feature an atypical N-terminal BD with a short and oomycete-specific α1 helix (α1′), plus a C-terminal small heat shock protein (sHSP) domain. In oomycete pathogen Phytophthora sojae, both BD-α1′ and sHSP domains are required for P. sojae BAG (PsBAG) function in cyst germination, pathogenicity, and unfolded protein response assisting in 26S proteasome-mediated degradation of misfolded proteins. PsBAGs form homo- and heterodimers through their unique BD-α1′ to function properly, with no recruitment of HSP70s to form the common BAG-HSP70 complex found in other eukaryotes. Our study highlights an oomycete-exclusive protein homeostasis mechanism mediated by atypical BAGs, which provides a potential target for oomycete disease control.

Apelin, a novel endogenous ligand of the G-protein-coupled receptor APJ, is encoded by the APLN gene and can be hydrolyzed into multiple subtypes, with Apelin-13 being one of the most active subtypes of the Apelin family. Recent studies have revealed that Apelin-13 functions as an adipokine that participates in the regulation of different biological processes, such as oxidative stress, inflammation, apoptosis, and energy metabolism, thereby playing an important role in the prevention and treatment of various metabolic diseases. However, the results of recent studies on the association between Apelin-13 and various metabolic states remain controversial. Furthermore, Apelin-13 is regulated or influenced by various forms of exercise and could therefore be categorized as a new type of exercise-sensitive factor that attenuates metabolic diseases. Thus, in this review, our purpose was to focus on the relationship between Apelin-13 and related metabolic diseases and the regulation of response movements, with particular reference to the establishment of a theoretical basis for improving and treating metabolic diseases.

BackgroundInflammatory bowel disease (IBD) is a chronic immune-mediated disorder affecting millions worldwide. Due to the complexity of its pathogenesis, the treatment options for IBD are limited. This study focuses on ELF4, a member of the ETS transcription factor family, as a target to elucidate its role in IBD and investigate its mechanism of action in alleviating IBD symptoms by activating IL1RN transcription to suppress the activity of inflammatory TH17 cells.MethodsUsing the GEO database, this study examined LPS-induced intestinal inflammatory genes and their regulation mechanisms. We examined the colon length of LPS-treated mice and derived the Disease Activity Index (DAI). H&E staining, ELISA, and flow cytometry were used to detect mice colon tissue damage, inflammatory factor levels in mouse serum, mouse macrophage types and inflammatory TH17 cell activity. RT-qPCR and Western blot detected ELF4, IL1RN, M1, and M2 polarization markers. In Vitro, using dual-luciferase and ChIP assays, we tested mouse bone marrow-derived macrophages (BMDMs) and mouse intestinal epithelial cells for IL1RN promoter activity and ELF4 enrichment.ResultsBioinformatics showed that LPS-induced colitis animals have reduced ELF4 expression in their colon tissue. In vivo tests confirmed reduced ELF4 expression in mice with LPS-induced colitis. ELF4 overexpression reduced mouse intestinal inflammation. ELF4 activated IL1RN transcription in bioinformatics and in vitro tests. ELF4 promoted IL1RN transcription and macrophage M2 polarization to limit intestinal epithelial cell death and inflammation and reduce mouse intestinal inflammation in vitro. ELF4 also reduced the Th17/Treg ratio by increasing IL1RN transcription.ConclusionELF4 activates IL1RN transcription, suppresses inflammatory TH17 cells, and induces macrophage M2 polarization to treat IBD.

Objective: Preparation of GYLEQ-Ca chelate with antioxidant activity and calcium supplement function. Methods: On the basis of solid-phase synthesis of GYLEQ antioxidant peptide, the conditions of chelation reaction between casein GYLEQ antioxidant peptide and Ca2+ were optimized by single factor and orthogonal experiment. The structure of GYLEQ-Ca chelate was characterized by IR and SEM. Meanwhile, the in vitro antioxidant activity, calcium retention in simulated gastrointestinal juice of GYLEQ-Ca chelate and its effect on cell viability were analyzed. Results: The optimum conditions for GYLEQ-Ca chelation reaction were temperature 50 ℃, time 40 min, pH 7.5, and peptide-calcium mass ratio 4∶1. The influence of chelation temperature on the chelation rate reached a very significant level, and the pH and peptide-calcium mass ratio reached a significant level. The infrared absorption peak of the GYLEQ-Ca chelate was changed compared with GYLEQ peptide, and the SEM observation showed that GYLEQ peptide was granular, while GYLEQ-Ca chelate was a network structure adhered together, indicating that GYLEQ peptide reacted with Ca2+ to form a new compound. The DPPH, hydroxyl radical scavenging rate and reducing power of GYLEQ-Ca chelate were higher than those of GYLEQ peptide. The calcium retention rate of GYLEQ-Ca chelate in simulated intestinal fluid was maintained at more than 70%, and GYLEQ-Ca chelate has no cytotoxicity. Conclusion: GYLEQ-Ca chelate has good antioxidant activity and calcium maintenance rate.

Abstract Background The triglyceride glucose-body mass index (TyG-BMI index) has been considered a reliable surrogate measure of insulin resistance; however, its ability to predict the incidence of cardiovascular disease in individuals with coronary artery disease (CAD) remains uncertain. The aim of this study was to demonstrate the correlation between the TyG-BMI index and cardiovascular incidence. Methods A total of 2533 consecutive participants who underwent percutaneous coronary intervention (PCI) and drug-eluting stent (DES) implantation were included. Data from 1438 patients was analyzed in the study. The endpoint was defined as a composite of acute myocardial infarction, repeat revascularization, stroke, and all-cause mortality (major adverse cardiac and cerebrovascular events, MACCEs) at 34-month follow-up. The formula for calculating the TyG-BMI index is ln [fasting triglyceride (mg/dL) × fasting blood glucose (mg/dL)/2] × BMI. Results Among the 1438 participants, 195 incident patient cases of MACCEs were ascertained. The incidence of MACCEs showed no statistically significant differences in the TyG-BMI index tertiles in the overall population. Further exploratory subgroup analysis and multivariable logistic regression analysis revealed a linear relationship between the TyG-BMI index (per 1 SD increased) and MACCEs in the elderly patients (OR = 1.22, 95% CI 1.011–1.467, p = 0.038) and in the female patients (OR = 1.33, 95% CI 1.004–1.764, p = 0.047). The addition of the TyG-BMI index to traditional risk factor models in elderly and female patients did not improve risk prediction for MACCEs. Conclusion A higher TyG-BMI index was proportionally related to an increased incidence of MACCEs in the elderly or female patients. However, the inclusion of the TyG-BMI index did not provide better predictive performance for MACCEs in the elderly, specifically in female patients.

Environmental microvibrations, often originating from unidentified sources, pose a significant challenge for predicting and controlling their complex wave fields, potentially leading to measurement errors of sensitive instruments in high-precision laboratories and impacting the accuracy of experimental outcomes. Therefore, investigating effective control measures for environmental microvibrations under passive conditions is key to addressing such engineering issues. This paper presents a finite element analysis method tailored to address environmental microvibrations in the absence of apparent sources. This method involves obtaining the vibration time history at specific ground surface points through field measurements and combining the Rayleigh wave velocity attenuation character with depth at the center frequencies of one-third octave bands within the 1–100 Hz frequency range; the vibration time history at any depth in the soil is calculated. These calculated vibrations are then applied as input loads to the corresponding nodes on one boundary of the foundation–soil model, serving as the source of environmental microvibrations. The predicted results are compared with measured data and the empirical point source input method, indicating that this approach is more precise and efficient, providing valuable reference for the prediction and analysis of environmental microvibrations. In addition, utilizing this method, the study examines the effects of pile foundation parameters such as the pile length, burial depth, and concrete baseplate thickness on the vibration isolation performance of environmental microvibrations, providing guidance for designing pile foundation isolation.

The pressure distribution and the force on tunnel segments of synchronous grouting in the shield tail gap channel of shield tunnels are key to controlling the stability and surface settlement of the strata surrounding such tunnels. Based on the basic principles of fluid mechanics and the limit equilibrium method, this study establishes a mathematical model of synchronous grouting in shield tunnels, derives the expressions of the grouting pressure and the force on tunnel segments in the shield tail gap channel, and verifies them using an engineering case study. Studies have shown that the force on tunnel segments and the speed of shield excavation are increasing. An excessive shield excavation speed will cause the load on tunnel segments to increase, which exacerbates the uneven distribution of the grouting pressure. The force on tunnel segments and the grouting pressure also have a positive relationship with the thickness of the shield tail gap, but the impact is limited to a certain range. With an increase in the tunnel radius, the number of grouting holes should be appropriately increased to balance the water and soil pressure in the surrounding strata. These research results can provide a theoretical reference for the design of synchronous grouting for shield tunnels in the future.

This study investigates the impact of varying concentrate levels in the diets of Angus beef cattle on their performance, nutrient digestion, and metabolism during the growth (7 to 12 months) and fattening (13 to 30 months) phases. Fifteen Angus steers were continuously fed and divided into low-concentrate (L) and high-concentrate (H) groups based on the fattening period and dietary formulations. Throughout each 9-week trial phase, a comprehensive range of parameters was systematically measured, including dry matter intake (DMI), average daily gain (ADG), gain-to-feed ratio (G/F), blood parameters, rumen fluid composition, and microbial diversity. In the fattening phases, an increase in concentrate levels resulted in a significant rise in the cattle’s DMI. Although there was a minor increase in ADG compared to the growing phases, this increase was not statistically significant. The efficiency of nitrogen (N) utilization in the cattle decreased, accompanied by a significant reduction in the apparent digestibility of nutrients. Ruminal fermentation produced more energy substances; however, there was a notable decrease in the abundance of fiber-decomposing microbes (such as the NK4A214_group, Ruminococcus, Papillibacter, and Acetitomaculum) and a significant increase in the abundance of starch-degrading microbes (including Bacteroidota and Prevotellaceae). Additionally, there was a significant reduction in the abundance of immune system-related functional pathways. This suggests that high-concentrate fattening does not necessarily lead to improved growth performance and may negatively affect metabolic health and nutrient digestion.

Enterohemorrhagic Escherichia coli (EHEC) serotype O157:H7 is a commonly encountered foodborne pathogen that can cause hemorrhagic enteritis and lead to hemolytic uremic syndrome (HUS) in severe cases. Bifidobacterium is a beneficial bacterium that naturally exists in the human gut and plays a vital role in maintaining a healthy balance in the gut microbiota. This study investigated the protective effects of B. longum K5 in a mouse model of EHEC O157:H7 infection. The results indicated that pretreatment with B. longum K5 mitigated the clinical symptoms of EHEC O157:H7 infection and attenuated the increase in myeloperoxidase (MPO) activity in the colon of the mice. In comparison to the model group, elevated serum D-lactic acid concentrations and diamine oxidase (DAO) levels were prevented in the K5-EHEC group of mice. The reduced mRNA expression of tight junction proteins (ZO-1, Occludin, and Claudin-1) and mucin MUC2, as well as the elevated expression of virulence factors Stx1A and Stx2A, was alleviated in the colon of both the K5-PBS and K5-EHEC groups. Additionally, the increase in the inflammatory cytokine levels of TNF-α and IL-1β was inhibited and the production of IL-4 and IL-10 was promoted in the K5-EHEC group compared with the model group. B. longum K5 significantly prevented the reduction in the abundance and diversity of mouse gut microorganisms induced by EHEC O157:H7 infection, including blocking the decrease in the relative abundance of Roseburia, Lactobacillus, and Oscillibacter. Meanwhile, the intervention with B. longum K5 promoted the production of acetic acid and butyric acid in the gut. This study provides insights into the use of B. longum K5 for developing probiotic formulations to prevent intestinal diseases caused by pathogenic bacterial infections.

High-power quantum cascade lasers (QCLs) have a wide application prospect. In this paper, a high-power high-beam-quality device with a large ridge width is demonstrated. The effect of different ridge widths on mode loss was studied, and the results showed that the mode loss decreased as the ridge width increased. Furthermore, as the width of the ridge increased, the temperature of the active region rose. In the experiment, the wafers were grown by metal–organic chemical vapor deposition (MOCVD), and the ridge width of the device was controlled by wet etching. A laser with a ridge width of 15 µm and a length of 5 mm achieved an output of 2.2 W under 288 K continuous wave (CW) operation, with a maximum slow-axis divergence angle of 27.2° and a device wavelength of 5 μm. The research results of this article promote the industrial production of base transverse mode QCL.

Introduction: Yanling Yinbiancha, a cultivar of Camellia sinensis (L.) O. Kuntze, is an evergreen woody perennial with characteristic albino leaves. A mutant variant with green leaves on branches has been recently identified. The molecular mechanisms underlying this color variation remain unknown.Methods: We aimed to utilize omics tools to decipher the molecular basis for this color variation, with the ultimate goal of enhancing existing germplasm and utilizing it in future breeding programs.Results and discussion: Albinotic leaves exhibited significant chloroplast degeneration and reduced carotenoid accumulation. Transcriptomic and metabolomic analysis of the two variants revealed 1,412 differentially expressed genes and 127 differentially accumulated metabolites (DAMs). Enrichment analysis for DEGs suggested significant enrichment of pathways involved in the biosynthesis of anthocyanins, porphyrin, chlorophyll, and carotenoids. To further narrow down the causal variation for albinotic leaves, we performed a conjoint analysis of metabolome and transcriptome and identified putative candidate genes responsible for albinism in C. sinensis leaves. 12, 7, and 28 DEGs were significantly associated with photosynthesis, porphyrin/chlorophyll metabolism, and flavonoid metabolism, respectively. Chlorophyllase 2, Chlorophyll a-Binding Protein 4A, Chlorophyll a-Binding Protein 24, Stay Green Regulator, Photosystem II Cytochrome b559 subunit beta along with transcription factors AP2, bZIP, MYB, and WRKY were identified as a potential regulator of albinism in Yanling Yinbiancha. Moreover, we identified Anthocyanidin reductase and Arabidopsis Response Regulator 1 as DEGs influencing flavonoid accumulation in albino leaves. Identification of genes related to albinism in C. sinensis may facilitate genetic modification or development of molecular markers, potentially enhancing cultivation efficiency and expanding the germplasm for utilization in breeding programs.
