BACKGROUND AND AIMS: SH2 domain-containing protein tyrosine phosphatase 2 (Shp2) is the first identified pro-oncogenic tyrosine phosphatase that acts downstream of receptor tyrosine kinases (RTKs) to promote Ras-extracellular signal-regulated kinase signaling. However, this phosphatase was also shown to be antitumorigenic in HCC. This study is aimed at deciphering paradoxical Shp2 functions and mechanisms in hepatocarcinogenesis and at exploring its value as a pharmaceutical target in HCC therapy. APPROACHES AND RESULTS: We took both genetic and pharmaceutical approaches to examine the effects of Shp2 inhibition on primary liver cancers driven by various oncogenes and on metastasized liver tumors. We show here that the catalytic activity of Shp2 was essential for relay of oncogenic signals from RTKs in HCC and that chemical inhibition of Shp2 robustly suppressed HCC driven by RTKs. However, in contrast to a tumor-promoting hepatic niche generated by genetically deleting Shp2 in hepatocytes, treatment with a specific Shp2 inhibitor had a tumor-suppressing effect on metastasized liver tumor progression. Mechanistically, the Shp2 inhibitor enhanced antitumor innate immunity by down-regulating inflammatory cytokines, suppressing the chemokine (C-C motif) receptor 5 signaling axis, but up-regulating interferon-β secretion. CONCLUSIONS: These results unveil complex mechanisms for the tumor-suppressing effect of pharmaceutical Shp2 inhibition in the liver immune environment. We provide a proof of principle for clinical trials with specific Shp2 inhibitors in patients with primary and metastasized liver cancer.

ObjectiveRecent reports have demonstrated that a wider pulse pressure upon admission is correlated with heightened in-hospital mortality following spontaneous supratentorial intracerebral hemorrhage (ssICH). However, the underlying mechanism remains ambiguous. We investigated whether a wider pulse pressure was associated with hematoma expansion (HE).MethodsDemographic information, clinical features, and functional outcomes of patients diagnosed with ssICH were retrospectively collected and analyzed. Multivariate logistic regression was conducted to identify independent predictors of HE. Weighted logistic regression, restricted cubic spline models, and propensity score matching (PSM) were employed to estimate the association between pulse pressure and HE.ResultsWe included 234 eligible adult ssICH patients aged 60 (51–71) years, and 55.56% were male. The mean pulse pressure was 80.94 ± 23.32 mmHg. Twenty-seven patients (11.54%) developed early HE events, and 116 (49.57%) experienced a poor outcome (modified Rankin scale 3–6). A wider mean pulse pressure as a continuous variable was a predictor of HE [odds ratios (OR) 1.026, 95% confidence interval (CI) 1.007–1.046, p = 0.008] in multivariate analysis. We transformed pulse pressure into a dichotomous variable based on its cutoff value. BACKGROUND AND AIMS: Immunotherapy with programmed cell death 1 (PD-1)/programmed death ligand 1 (PD-L1) blockade has shown low response rates in liver cancer patients, with the underlying mechanisms unclear. To decipher a specific impact of the liver microenvironment, we compared the effects of anti-PD-L1 antibody (αPD-L1) blockade on the same tumor grown s.c. or in the liver. APPROACH AND RESULTS: We generated s.c. tumors in mice by inoculating MC38 colorectal cancer (CRC) cells under the skin and metastatic liver tumors by portal vein or splenic injection of CRC cells. Tumor-bearing mice were treated by i.p. injection of αPD-L1, polyinosinic:polycytidylic acid (poly[I:C]), or both. αPD-L1 monotherapy significantly suppressed s.c. tumor growth, but showed no effect on metastatic liver tumors. However, the combination of αPD-L1 with poly(I:C), an innate immunity-stimulating reagent, robustly inhibited tumor progression in liver. The combination therapy effectively down-regulated myeloid-derived suppressor cells (MDSCs), but up-regulated ratios of M1/M2 macrophages, CD8/CD4, and CD8/regulatory T (Treg) cells infiltrated into liver tumors and whole liver. A group of long-lasting T-bet+ Eomes- PD-1- cytotoxic T cells was maintained in the combo-treated liver, leading to resistance to tumor recurrence. Depleting macrophages or blocking type Ⅰ interferon signaling abrogated the synergistic antitumor effect of αPD-L1 and poly(I:C), indicating a requirement of boosting innate immunity for optimized activation of cytotoxic T cells by PD-1/PD-L1 blockade. CONCLUSIONS: The poor response of liver cancers to αPD-L1 therapy is largely attributable to a unique hepatic immunotolerant microenvironment, independent of tumor origins or types. The success of a combinatorial immunotherapy relies on coordinated inhibition or activation of various innate and adaptive immune cell activities.

The postnatal development and maturation of the liver, the major metabolic organ, are inadequately understood. We have analyzed 52,834 single-cell transcriptomes and identified 31 cell types or states in mouse livers at postnatal days 1, 3, 7, 21, and 56. We observe unexpectedly high levels of hepatocyte heterogeneity in the developing liver and the progressive construction of the zonated metabolic functions from pericentral to periportal hepatocytes, which is orchestrated with the development of sinusoid endothelial, stellate, and Kupffer cells. Trajectory and gene regulatory analyses capture 36 transcription factors, including a circadian regulator, Bhlhe40, in programming liver development. Remarkably, we identified a special group of macrophages enriched at day 7 with a hybrid phenotype of macrophages and endothelial cells, which may regulate sinusoidal construction and Treg-cell function. This study provides a comprehensive atlas that covers all hepatic cell types and is instrumental for further dissection of liver development, metabolism, and disease.

Abstract Background This systematic review and meta-analysis aimed to study the evidence on the efficacy and safety of omitting axillary lymph node dissection (ALND) for patients with clinically node-negative but sentinel lymph node (SLN)-positive breast cancer using all the available evidence. Methods The Embase, Medline, and Cochrane Library databases were searched through February 25, 2023. Original trials that compared only the sentinel lymph node biopsy (SLNB) with ALND as the control group for patients with clinically node-negative but SLN-positive breast cancer were included. The primary outcomes were axillary recurrence rate, total recurrence rate, disease-free survival (DFS), and overall survival (OS). Meta-analyses were performed to compare the odds ratio (OR) in rates and the hazard ratios (HR) in time-to-event outcomes between both interventions. Based on different study designs, tools in the revised Cochrane risk of bias tool were used for randomized trials and the risk of bias in nonrandomized studies of interventions to assess the risk of bias for each included article. Funnel plots and Egger's test were used for the publication’s bias assessment. Results In total, 30 reports from 26 studies were included in the systematic review (9 reports of RCTs, 21 reports of retrospective cohort studies). According to our analysis, omitting ALND in patients with clinically node-negative but SLN-positive breast cancer had a similar axillary recurrence rate (OR = 0.95, 95% confidence interval (CI): 0.76–1.20), DFS (HR = 1.02, 95% CI: 0.89–1.16), and OS (HR = 0.97, 95% CI: 0.92–1.03), but caused a significantly lower incidence of adverse events and benefited in locoregional recurrence rate (OR = 0.76, 95% CI: 0.59–0.97) compared with ALND. Conclusion For patients with clinically node-negative but SLN-positive breast cancer (no matter the number of the positive SLN), this review showed that SLNB alone had a similar axillary recurrence rate, DFS, and OS, but caused a significantly lower incidence of adverse events and showed a benefit for the locoregional recurrence compared with ALND. An OS benefit was found in the Macro subset that used SLNB alone versus complete ALND. Therefore, omitting ALND is feasible in this setting. Trial registration CRD 42023397963

ObjectiveThe objective was to explore the impact of WeChat follow-up management on the psychological distress, care burden, and quality of life of parents of infants with bronchopulmonary dysplasia (BPD) receiving in-home care.MethodsThis was a retrospective cohort study. A total of 101 parents of infants with BPD who were followed up from January 2016 to January 2022 were included in this study. According to different follow-up methods, these patients were classified into the WeChat group and the routine group. The Depression, Anxiety, and Stress Scale-21 (DASS-21), Zarit Caregiver Burden Interview (ZBI), and WHOQOL-BREF were used. The data on the psychological distress, care burden, and quality of life of the parents in the two groups were analyzed and compared at discharge and at the 3-month follow-up.ResultsThere was no significant difference in the DASS-21 and ZBI scores at discharge between the parents in the two groups. During the 3-month follow-up, the scores of the DASS-21 anxiety and stress subscale and the ZBI of parents in the WeChat group were significantly lower than those of parents in the routine group (P 0.05). A comparison of the WHOQOL-BREF score between the two groups showed that the total quality of life score in the WeChat group was significantly higher than that in the routine group (P

Abstract High‐energy‐density lithium metal batteries (LMBs) are widely accepted as promising next‐generation energy storage systems. However, the safety features of practical LMBs are rarely explored quantitatively. Herein, the thermal runaway behaviors of a 3.26 Ah (343 Wh kg−1) Li | LiNi0.5Co0.2Mn0.3O2 pouch cell in the whole life cycle are quantitatively investigated by extended volume‐accelerating rate calorimetry and differential scanning calorimetry. By thermal failure analyses on pristine cell with fresh Li metal, activated cell with once plated dendrites, and 20‐cycled cell with large quantities of dendrites and dead Li, dendrite‐accelerated thermal runaway mechanisms including reaction sequence and heat release contribution are reached. Suppressing dendrite growth and reducing the reactivity between Li metal anode and electrolyte at high temperature are effective strategies to enhance the safety performance of LMBs. These findings can largely enhance the understanding on the thermal runaway behaviors of Li metal pouch cells in practical working conditions.

Glioblastoma is acknowledged as the most aggressive cerebral tumor in adults. However, the efficacy of current standard therapy is seriously undermined by drug resistance and suppressive immune microenvironment. Ferroptosis is a recently discovered form of iron-dependent cell death that may have excellent prospect as chemosensitizer. The utilization of ferropotosis inducer Erastin could significantly mediate chemotherapy sensitization of Temozolomide and exert anti-tumor effects in glioblastoma. In this study, a combination of hydrogel-liposome nanoplatform encapsulated with Temozolomide and ferroptosis inducer Erastin was constructed. The αvβ3 integrin-binding peptide cyclic RGD was utilized to modify codelivery system to achieve glioblastoma targeting strategy. As biocompatible drug reservoirs, cross-linked GelMA (gelatin methacrylamide) hydrogel and cRGD-coated liposome realized the sustained release of internal contents. In the modified intracranial tumor resection model, GelMA-liposome system achieved slow release of Temozolomide and Erastin in situ for more than 14 d. The results indicated that nanoplatform (T+E@LPs-cRGD+GelMA) improved glioblastoma sensitivity to chemotherapeutic temozolomide and exerted satisfactory anti-tumor effects. It was demonstrated that the induction of ferroptosis could be utilized as a therapeutic strategy to overcome drug resistance. Furthermore, transcriptome sequencing was conducted to reveal the underlying mechanism that the nanoplatform (T+E@LPs-cRGD+GelMA) implicated in. It is suggested that GelMA-liposome system participated in the immune response and immunomodulation of glioblastoma via interferon/PD-L1 pathway. Collectively, this study proposed a potential combinatory therapeutic strategy for glioblastoma treatment.

Immunotherapy with checkpoint inhibitors for liver cancer, while active in many clinical trials worldwide, may have uncertain outcomes due to the unique immunotolerant microenvironment of the liver. In previous experiments, we unexpectedly identified a robust liver tumor-preventive effect of a synthetic double-stranded RNA, polyinosinic-polycytidylic acid (polyIC), in mice. Herein we further demonstrate that polyIC given at the precancer stage effectively prevented liver tumorigenesis by activating natural killer cells, macrophages, and some T-cell subsets; no inhibitory effect was observed on tumor progression if injected after tumor initiation. Nevertheless, polyIC administration potently induced programmed death ligand 1 (PD-L1) expression in liver sinusoid endothelial cells, which prompted us to test a combined treatment of polyIC and PD-L1 antibody (Ab). Although injecting PD-L1 Ab alone did not show any therapeutic effect, injection of polyIC sensitized the hepatic response to PD-L1 blockade. Combination of polyIC and PD-L1 Ab resulted in sustained accumulation of active cluster of differentiation 8 cytotoxic T cells and robust liver tumor suppression and conferred a survival advantage in mice. These preclinical data in animal models suggest that, despite the low efficacy of PD-L1/PD-1 blockade alone, careful design of mechanism-based combinatorial immunotherapeutic protocols may shift the paradigm in liver cancer treatment by coordinating maximal activation of multiple innate and adaptive immune functions. Conclusion: We provide proof of principle for the development of an efficient prevention strategy of liver tumorigenesis and a powerful combination immunotherapy for primary liver cancer.

Objective To investigate the effect of gelsemine on the proliferation, migration and invasion of rheumatoid arthritis endothelial cells (RASFs) and its possible mechanism. Methods RASFs were isolated and cultured. After RASFs were intervened with 50, 100, 200 μg/mL gelsemine for 24 h or small interfering RNA or over-expression vector of circular RNA circ_001680 was transfected into RASFs, cell proliferation was measured by CCK-8 method and clone formation test, cell migration was detected by scratch test, cell invasion was examined by Transwell, the protein expression of E-cadherin and N-cadherin was detected by Western blot. The protein expression of circ_001680 was examined by RT-qPCR. Results Gelsemine or interference with circ_001680 expression reduced the A value, the number of clones, the healing rate of scratches and the number of invaded cells of RASFs and the protein expression of N-cadherin in cells (P＜0.05), and promoted the protein expression of E-cadherin (P＜0.05). Gelsemine reduced the expression of circ_001680 in RASFs (P＜0.05), and the over-expression of circ_001680 reduced the inhibitory effect of gelsemine on the proliferation, migration and invasion of RASFs. Conclusions Gelsemine may inhibit the proliferation, migration and invasion of RASFs, and its mechanism is potentially related to the down-regulation of circ_001680 expression.

Abstract The pursuit of sustainable energy has a great request for advanced energy storage devices. Lithium metal batteries are regarded as a potential electrochemical storage system because of the extremely high capacity and the most negative electrochemical potential of lithium metal anode. Dead lithium formed in the stripping process significantly contributes to the low efficiency and short lifespan of rechargeable lithium metal batteries. This review displays a critical review on the current research status about the stripping electrochemistry of lithium metal anode. The significance of stripping process to a robust lithium metal anode is emphasized. The stripping models in different electrochemical scenarios are discussed. Specific attention is paid to the understanding for the electrochemical principles of atom diffusion, electrochemical reaction, ion diffusion in solid electrolyte interphase (SEI), and electron transfer with the purpose to strengthen the insights into the behavior of lithium electrode stripping. The factors affecting stripping processes and corresponding solutions are summarized and categorized as follows: surface physics, SEI, operational and external factors. This review affords fresh insights to explore the lithium anode and design robust lithium metal batteries based on the comprehensive understanding of the stripping electrochemistry.

Summary: Low-temperature solid oxide fuel cells (LT-SOFCs) are a promising next-generation fuel cell due to their low cost and rapid start-up, posing a significant challenge to electrode materials with high electrocatalytic activity. Herein, we reported the bimetallic nanoparticles encapsulated in carbon nanotubes (NiFe@CNTs) prepared by carefully controlling catalytic pyrolysis of waste plastics. Results showed that plenty of multi-walled CNTs with outer diameters (14.38 ± 3.84 nm) were observed due to the smallest crystalline size of Ni-Fe alloy nanoparticles. SOFCs with such NiFe@CNTs blended in anode exhibited remarkable performances, reaching a maximum power density of 885 mW cm−2 at 500°C. This could be attributed to the well-dispersed alloy nanoparticles and high graphitization degree of NiFe@CNTs to improve HOR activity. Our strategy could upcycle waste plastics to produce nanocomposites and demonstrate a high-performance LT-SOFCs system, addressing the challenges of sustainable waste management and guaranteeing global energy safety simultaneously.

Objectives: Obstructive sleep apnea (OSA) is an independent risk factor for metabolic syndrome (MetS). Recent studies have indicated that circadian clock genes were dysregulated in OSA. In addition, it is clear that the impairment of circadian clocks drives the progression of MetS. Therefore, we hypothesized that circadian rhythm disruption links OSA with MetS.Methods: A total of 118 participants, who underwent polysomnography (PSG) and were diagnosed as healthy snorers (control, n = 29) or OSA (n = 89) patients based on the apnea–hypopnea index (AHI), were enrolled in the present study. General information, anthropometric data, blood biochemical indicators, clock gene expressions, and levels of oxidative and inflammatory indicators were collected, determined, and compared in all the participants.Results: We found that Brain and muscle aryl hydrocarbon receptor nuclear translocator-like protein 1 (Bmal1) and Differentiated embryo chondrocyte 1 (Dec1) were upregulated, while Period 1 (Per1) was reduced in OSA patients. In addition, these changing trends were closely associated with the hypoxia indicator of AHI and have a significant impact on the presence of MetS components, such as hyperglycemia (Dec1 and Per1, p < 0.05 and 0.001, respectively), hypertension (Bmal1 and Dec1, p < 0.001 and 0.01, respectively), hyperlipidemia (Dec1, p < 0.01), and obesity (Dec1, p < 0.05). Notably, expressions of Dec1 correlated with IR and predicted the presence of MetS in OSA patients. Finally, we also observed that Dec1 expression was interrelated with levels of both oxidative indicators and inflammatory biomarkers (IL-6) in OSA.Conclusion: This study concluded that circadian clock disruptions, especially Dec1, link OSA with MetS in an oxidative and inflammatory-related manner. Circadian clock Dec1 can be used as a specific biomarker (p < 0.001) and therapeutic target in OSA combined with Mets patients.

Objective: Ovarian cancer is a deadly gynecologic malignancy with a poor prognosis. It is essential to evaluate the early detection and screening programs of ovarian cancer via timely assessment of long-time survival, particularly in China where those data are incredibly limited. Here, we aimed to provide timely and accurately assessment of long-term survival estimate of ovarian cancer patients from eastern China. Methods: Data of 770 ovarian cancer patients diagnosed between 2004–2018 were obtained from four cancer registries in Taizhou, eastern China, were included. We used period analysis to calculate five-year relative survival (RS) of aforementioned ovarian cancer patients for overall and the stratification by age at diagnosis and region. Results: Our findings demonstrated that the overall five-year RS for ovarian cancer patients in Taizhou between 2014 and 2018 was 69.2%, while urban areas were higher compared to rural areas (77.6% vs. 64.9%). We also observed a significant age gradient with the five-year RS decreasing from 79.6% for age group < 55 years to 66.9% for age group > 74 years. Furthermore, we identified a clear upward trend of five-year RS over the study period, both overall and stratified by region and age at diagnosis. Conclusion: This is the first study in China using period analysis to provide the most up-to-date five-year RS for ovarian cancer patients from Taizhou, eastern China, which reaches 69.2% during 2014–2018. Our results provide valuable information for timely assessment of early detection and screening programs for ovarian cancer in eastern China.

Summary: High-dimensional cellular and molecular profiling of biological samples highlights the need for analytical approaches that can integrate multi-omic datasets to generate prioritized causal inferences. Current methods are limited by high dimensionality of the combined datasets, the differences in their data distributions, and their integration to infer causal relationships. Here, we present Essential Regression (ER), a novel latent-factor-regression-based interpretable machine-learning approach that addresses these problems by identifying latent factors and their likely cause-effect relationships with system-wide outcomes/properties of interest. ER can integrate many multi-omic datasets without structural or distributional assumptions regarding the data. It outperforms a range of state-of-the-art methods in terms of prediction. ER can be coupled with probabilistic graphical modeling, thereby strengthening the causal inferences. The utility of ER is demonstrated using multi-omic system immunology datasets to generate and validate novel cellular and molecular inferences in a wide range of contexts including immunosenescence and immune dysregulation. The bigger picture: Multi-omic technologies for deep cellular and molecular profiling from model organisms or humans have rapidly expanded. However, existing analytical approaches are constrained by the high dimensionality of these datasets, differences in data distributions, and the inability to generate causal inference beyond predictive biomarkers. To address these issues, we developed a novel interpretable machine-learning framework, Essential Regression (ER). ER integrates high-dimensional multi-omic datasets without distributional assumptions regarding the data and identifies significant latent factors and their causal relationships with system-wide outcomes/properties of interest. ER uses higher-order relationships encapsulated in the latent factors, rather than the individual observables, to home in on novel mechanistic insights. Our approach outperforms a range of state-of-the-art methods in terms of prediction and generates novel immunological inferences, consistent with evidence in model organisms.

The barrier function of nasal mucosal epithelial cells plays an irreplaceable role in the spread and expansion of viruses in the body. This study found that influenza A virus H1N1 could induce apoptosis of nasal mucosal epithelial progenitor cells, cause an inflammatory response, and trigger the maturation and recruitment of nasal submucosal dendritic cells (DCs), but the mechanism remained unclear. Therefore, we used RNA sequencing and high-resolution untargeted metabolomics to sequence and perform combined bioinformatic analysis of H1N1 virus-infected nasal mucosal epithelial cells from 6 different patients. The abnormal arginine metabolism signaling pathway caused by H1N1 virus infection was screened out, and arginase inhibitors were used to interfere with the abnormal arginine metabolism and the maturation and recruitment of submucosal DCs caused by the H1N1 virus in vitro and in vivo. We conclude that H1N1 influenza virus promotes the recruitment and maturation of submucosal DCs by causing abnormal arginine metabolism in nasal mucosal epithelial cells, thereby triggering respiratory mucosal immunity.

Abstract Lithium ion battery has achieved great success in portable electronics and even recently electronic vehicles since its commercialization in 1990s. However, lithium‐ion batteries are confronted with several issues in terms of the sustainable development such as the high price of raw materials and electronic products, the emerging safety accidents, etc. The recent progresses are herein emphasized on lithium batteries for energy storage to clearly understand the sustainable energy chemistry and emerging energy materials. The Perspective presents novel lithium‐ion batteries developed with the aims of enhancing the electrochemical performance and sustainability of energy storage systems. First, revolutionary material chemistries, including novel low‐cobalt cathode, organic electrode, and aqueous electrolyte, are discussed. Then, the characteristics of safety performance are analyzed and strategies to enhance safety are subsequently evaluated. Battery recycling is considered as the key factor for a sustainable society and related technologies are present as well. Finally, conclusion and outlook are drawn to shed lights on the further development of sustainable lithium‐ion batteries.

Abstract We report an experimental demonstration of positive–negative sub-wavelength interference without correlation. Typically, people can achieve sub-wavelength effects with correlation measurement no matter by using bi-photon or thermal light sources. In this paper, we adopt a thermal light source, and we count the realizations in which the intensities of the definite symmetric points are above or below a certain threshold. The distribution of numbers of these realizations which meet the restriction will show a sub-wavelength effect. With proper constrictions, positive and negative interference patterns are demonstrated.

The problem of quantum first detection time has been extensively investigated in recent years. Employing the stroboscopic measurement protocol, we consider such a monitored quantum walk on sequentially or periodically perturbed rings and focus on the statistics of first detected return time, namely, the time it takes a particle to return to the initial state for the first time. Using time-independent perturbation theory, we obtain the general form of the eigenvalues and eigenvectors of the Hamiltonian. For the case of a sequentially perturbed ring system, we find steplike behaviors of ∑_{n=1}^{N}nF_{n} (→〈n〉 as N→∞) when N increases, with two plateaus corresponding to integers, where F_{n} is the first detected return probability at the nth detection attempt. Meanwhile, if the initial condition preserves the reflection symmetry, the mean return time is the same as the unperturbed system. For the periodically perturbed system, similar results can also appear in the case where the symmetry is preserved; however, the size of the ring, the interval between adjacent perturbations, and the initial position may change the mean return time in most cases. In addition, we find that the decay rate of the first detection probability F_{n} decreases with the increase in perturbation amplitude. More profoundly, the symmetry-preserving setup of the initial conditions leads to the coincidence of F_{n}. The symmetry of the physical systems under investigation is deeply reflected in the quantum detection time statistics.

Abstract Background Psoas hematoma rarely occurs in patients with spondylolisthesis who undergo posterior lumbar interbody fusion (PLIF) surgery. Case presentation Here we reported a case of a 57-year-old male patient diagnosed with spondylolisthesis who underwent PLIF at the local hospital. Seven days post-surgery, abdominal pain occurred, and the pain in the right lower limb gradually increased. The computerized tomography (CT) indicated a formation of hematoma around the psoas muscle. Digital-subtraction angiography (DSA) suggested a vascular injury, a rupture of the right segmental artery of the lumbar vertebral level 4. The patient then received DSA vascular embolization, after which the lower lumbar segmental artery active bleeding was stopped. One month after discharge, the abdominal hematoma was gradually absorbed, and the pain in the waist, leg, and abdomen disappeared. Conclusion Symptoms such as abdominal pain, abdominal distension, and exacerbation of lower limb pain, may suggest the occurrence of psoas hematoma after PLIF. DSA vascular embolization is suggested as the first treatment approach for this type of complication.

Epithelial-mesenchymal transition (EMT) is thought to be involved in the tissue remodeling and long-term inflammatory process of chronic sinusitis (CRS), but the driving mechanism is still unclear. Using high-resolution mass spectrometry, we performed a proteomic screen of CRS nasal mucosal tissue to identify differentially expressed proteins. Data are available via ProteomeXchange with identifier PXD030884. Specifically, we identified S100 calcium binding protein A4 (S100A4), an effective factor in inflammation-related diseases, and its downstream protein closely related to tissue fibrosis collagen type I alpha 1 chain (COL1A1), which suggested its involvement in nasal mucosal tissue remodeling. In addition, stimulation of human nasal epithelial cells (HNEpCs) with lipopolysaccharide (LPS) mimicked the inflammatory environment of CRS and showed that S100A4 is involved in regulating EMT and thus accelerating tissue remodeling in the nasal mucosa, both in terms of increased cell motility and overexpression of mesenchymal-type proteins. Additionally, we further investigated the regulation mechanism of S100A4 involved in EMT in CRS. Our research results show that in the inflammatory environment of CRS nasal mucosal epithelial cells, TCF-4 will target to bind to S100A4 and regulate its transcription. The transcription of S100A4 in turn affects the execution of the important signaling pathway in EMT, the Wnt/GSK-3β/β-catenin pathway, through the TCF-4/β-catenin complex. In conclusion, this study confirmed that the expression of S100A4 was significantly increased during the progressive EMT process of CRS mucosal epithelial cells, and revealed that the transcriptional regulation of S100A4 plays an important role in the occurrence and development of EMT. This finding will help us to better understand the pathogenesis behind the remodeling in CRS patients, and identify target molecules for the treatment of CRS.

High-current pulsed electron beam sources are the core components of high-power microwave systems. In order to meet the requirements of future applications, one needs to improve the performance of electron beam sources in terms of vacuum insulation, beam transportation, and thermal management. In this paper, we report about our recent progress in the development of high-current vacuum electron beam sources. In order to meet the vacuum maintenance requirements of high-power microwave tubes, a high-electric field ceramic vacuum interface is designed and fabricated based on the ceramic metal brazing technique. In our experiments, a stable operation of the ceramic vacuum interface is demonstrated in the 10 Hz repetition mode with withstand voltage of larger than 600 kV and pulse width of about 100 ns. Besides, a cold cathode is developed using SiC nanowires, and an average beam current density of 1.2 kA/cm2 is achieved under the electric field strength of 90 kV/cm. Compared with traditional velvet cathodes, the characteristics of the SiC nanowire cathode, such as macro-electrical stability, emission uniformity, and operating life have been significantly improved. Furthermore, a high-current electron beam collector has been developed for relativistic backward wave oscillator tubes. A spiral flume is designed in the collector to meet the requirement of both high specific energy and low flow rate. It shows that the withstand heat flow density is in the order of 1012 W/m2, which is suitable for the long pulse and repetitive operation of the system. These results represent a significant step towards the practical application of long-life high-power microwave systems.
