10. Enhanced protective effect of selenium-biofortified peptide RYNA(Se)MNDYT compared with its native peptide RYNAMNDYT in lipopolysaccharide-injured murine gut microbiota

Abstract

Selenopeptides may be a valuable bioactive compound to promote gut microbiota-targeted therapeutic methods for intestinal disease and hepatopathy. However, limited information is available on the utilization of selenopeptides by gut microbiota, especially Selenium (Se) function. For this purpose, the present study aimed to investigate the protective effect of selenopeptide (RYNA(Se)MNDYT, Se-P2, purity of ≥ 95 %) and its original peptide (RYNAMNDYT, P2, purity of ≥ 95 %) in vivoby the microbiota-metabolite axis and further analyze the potential contribution of Se biofortification to Se-P2 bioactivity. The results showed that Se-P2 exhibits a higher protective effect on lipopolysaccharide (LPS)-induced inflammation than P2, including pathology of the colon and liver, which suggested that the bioactivity of P2 was promoted by the organic combination of Se. Notably, gut microbiota composition tended to be a healthy structure by Se-P2 pretreatment in LPS-injured mice, which had a positive effect on LPS-induced gut microbiota dysbacteriosis. Additionally, only Se-P2 promoted an increase in the relative abundance of Lactobacillus, Alistipes, and Roseburiaand a decrease in the relative abundance of Akkermansia, Erysipelatoclostridium, and Bacteroidesin LPS-injured mice. The changes in gut microbiota were obviously correlated with the changes in metabolites and affected the metabolic pathways of valine, leucine, isoleucine, phenylalanine, tyrosine, and tryptophan biosynthesis and phenylalanine metabolism. This may be one of the key reasons for Se-P2 to exert bioactivity through the microbiota-metabolite axis. Furthermore, Se-biofortification in Se-enriched Cordyceps militarisaffected the parental proteins of Se-P2 to modulate mitogen-activated protein kinase, GPI anchored protein, and carbohydrate metabolism, translation, folding, sorting and degradation, which may contribute to the bioactivity of Se-P2. Our study provides information on the effect of Se on selenopeptides in vivo, which further promotes the prospective applications of selenopeptides as dietary supplements.

Abstract

The purification of the histone nuclear factor M has been achieved and it is found to be a protein with molecular mass of 48K which is the same as interferon regulatory factor 2 (IRF-2). The transcription of the human histone gene reaches a maximum limit during the early S phase. IRF-2 has an oncogenic action and there is a link between IRF-2 and a gene that is linked to DNA replication and the transition from G1 to S phase of cell cycle.

Abstract

Gene expression data from microarrays are being applied to predict preclinical and clinical endpoints, but the reliability of these predictions has not been established. In the MAQC-II project, 36 independent teams analyzed six microarray data sets to generate predictive models for classifying a sample with respect to one of 13 endpoints indicative of lung or liver toxicity in rodents, or of breast cancer, multiple myeloma or neuroblastoma in humans. In total, >30,000 models were built using many combinations of analytical methods. The teams generated predictive models without knowing the biological meaning of some of the endpoints and, to mimic clinical reality, tested the models on data that had not been used for training. We found that model performance depended largely on the endpoint and team proficiency and that different approaches generated models of similar performance. The conclusions and recommendations from MAQC-II should be useful for regulatory agencies, study committees and independent investigators that evaluate methods for global gene expression analysis.

Abstract

Gene expression data from microarrays are being applied to predict preclinical and clinical endpoints, but the reliability of these predictions has not been established. In the MAQC-II project, 36 independent teams analyzed six microarray data sets to generate predictive models for classifying a sample with respect to one of 13 endpoints indicative of lung or liver toxicity in rodents, or of breast cancer, multiple myeloma or neuroblastoma in humans. In total, >30,000 models were built using many combinations of analytical methods. The teams generated predictive models without knowing the biological meaning of some of the endpoints and, to mimic clinical reality, tested the models on data that had not been used for training. We found that model performance depended largely on the endpoint and team proficiency and that different approaches generated models of similar performance. The conclusions and recommendations from MAQC-II should be useful for regulatory agencies, study committees and independent investigators that evaluate methods for global gene expression analysis.

Abstract

Pleurotus geesteranus is a promising source of bioactive compounds. However, knowledge of the antioxidant behaviors of P. geesteranus protein hydrolysates (PGPHs) is limited. In this study, PGPHs were prepared with papain, alcalase, flavourzyme, pepsin, and pancreatin, respectively. The antioxidant properties and cytoprotective effects against oxidative stress of PGPHs were investigated using different chemical assays and H2O2 damaged PC12 cells, respectively. The results showed that PGPHs exhibited superior antioxidant activity. Especially, hydrolysate generated by alcalase displayed the strongest 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity (91.62%), 2,2-azino-bis (3-ethylbenzothia zoline-6-sulfonic acid) (ABTS) radical scavenging activity (90.53%), ferric reducing antioxidant power, and metal ion-chelating activity (82.16%). Analysis of amino acid composition revealed that this hydrolysate was rich in hydrophobic, negatively charged, and aromatic amino acids, contributing to its superior antioxidant properties. Additionally, alcalase hydrolysate showed cytoprotective effects on H2O2-induced oxidative stress in PC12 cells via diminishing intracellular reactive oxygen species (ROS) accumulation by stimulating antioxidant enzyme activities. Taken together, alcalase hydrolysate of P. geesteranus protein can be used as beneficial ingredients with antioxidant properties and protective effects against ROS-mediated oxidative stress. Abstract

Background: Metastatic castration-resistant prostate cancer (mCRPC) has a poor prognosis, necessitating the investigation of novel treatments and targets. This study evaluated JNJ-70218902 (JNJ-902), a T-cell redirector targeting transmembrane protein with epidermal growth factor-like and 2 follistatin-like domains 2 (TMEFF2) and cluster of differentiation 3, in mCRPC., Patients and Methods: Patients who had measurable/evaluable mCRPC after at least one novel androgen receptor-targeted therapy or chemotherapy were eligible. Participants received subcutaneous JNJ-902 0.3, 1.0, 1.5, 3.0, or 6.0 mg once weekly (QW) or 2.0, 3.0, 4.0, or 6.0 mg biweekly (Q2W). Study objectives included assessment of safety, pharmacokinetics, immunogenicity, and preliminary efficacy., Results: Eighty-two participants were enrolled to receive at least one dose of JNJ-902 (QW; n = 38; Q2W; n = 44). Median duration of treatment was 1.91 (0.0-19.4) months across dosing groups. All participants experienced at least one treatment-emergent adverse event (TEAE) and 76 (92.7%) experienced treatment-related TEAEs. Fourteen participants (17.1%) experienced a TEAE that led to study discontinuation, of which 3 (3.7%) were related to JNJ-902. Dose-limiting toxicities were observed in 2 participants (2.4%). Five participants (15.2%) with measurable disease had a confirmed partial response and 10 participants (12.2%) had ≥50% decrease from baseline prostate-specific antigen levels. Clinical activity was not dose related and no clear exposure-response relationship was observed., Conclusions: In this study, dose escalation was limited by emerging dose-limiting toxicities. Although a recommended phase II dose was not determined, findings indicate TMEFF2 to be a potential target in mCRPC that warrants further investigation., (© The Author(s) 2025.
