In recent years, the world has entered a period of vigorous development in intelligent computing. As deep learning models with huge parameters and complex structures, large model training requires fast synchronization of training parameters between multiple cards and servers, which imposes higher requirements on the bandwidth, latency, reliability, scalability and security of datacenter networks. The requirements and related key technologies of intelligent computing networks for large model training were studied, and the standard specifications, academic research, and case practices of intelligent computing networks were analyzed, in order to promote the development of intelligent computing networks.

Background IgA nephropathy (IgAN) is the most common primary glomerulonephritis in China and worldwide, approximately 25%-30% of patients will progress to end-stage renal disease within 20 years after diagnosis. Currently, there is no effective and safe treatment specifically for IgAN. In recent years, there has been a rapid progress in the research of new drugs for IgAN, among which the targeted delayed-release budesonide capsules is the first allopathic drug for IgAN globally. Objective To investigate the mechanism of corticosteroid budesonide capsules in the treatment of IgAN based on network pharmacology. Methods Chemical Book platform was used to screen the targets of budesonide; GeneCards and CTD databases were utilized to obtain the relevant targets of IgAN. The intersection of budesonide targets and IgAN targets was obtained through a Venn diagram. A protein-protein interaction (PPI) network map was constructed, and gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis were performed on the intersecting targets. Results A total of 242 targets for budesonide, 1 443 candidate targets for IgAN, and 146 intersecting targets were selected. The 15 core targets in the PPI network included interleukin-6 (IL-6), tumor necrosis factor (TNF), interleukin-10 (IL-10), vascular endothelial growth factor A (VEGFA), epidermal growth factor receptor (EGFR), interleukin-1B (IL-1B), interleukin-4 (IL-4), interleukin-8 (CXCL8), gene on chromosome 1 (JUN), interleukin-13 (IL-13), interleukin-2 (IL-2), chemokine 2 (CCL2), toll-like receptor 4 (TLR4), colony-stimulating factors (CSF2), and albumin (ALB). Enrichment analysis revealed 1 646 GO enrichment results and 174 KEGG signaling pathways. The biological processes (BP) mainly involved positive regulation of phosphorylation, inflammatory response, and positive regulation of cell movement. The cellular components (CC) mainly involved cytoplasmic vesicle lumen, cyst cavity, and secretory granule lumen. The molecular functions (MF) mainly involved receptor signaling activity, receptor regulator activity, and receptor ligand activity. The KEGG signaling pathways mainly included interleukin 17 signaling pathway, cytokine-cytokine receptor interaction, pathways in cancer, and tumor necrosis factor signaling pathway. Conclusion This study provides preliminary verified that budesonide can treat IgAN by targeting IL-6, TNF, IL-10, VEGFA, EGFR, and other targets, through multiple signaling pathways, like cytokine-cytokine receptor interaction, interleukin-17 signaling pathway, pathways in cancer, and tumor necrosis factor signaling pathway, providing a theoretical basis for further research and clinical practice of budesonide.

Synchronous dynamics (fluctuations that occur in unison) are universal phenomena with widespread implications for ecological stability. Synchronous dynamics can amplify the destabilizing effect of environmental variability on ecosystem functions such as productivity, whereas the inverse, compensatory dynamics, can stabilize function. Here we combine simulation and empirical analyses to elucidate mechanisms that underlie patterns of synchronous versus compensatory dynamics. In both simulated and empirical communities, we show that synchronous and compensatory dynamics are not mutually exclusive but instead can vary by timescale. Our simulations identify multiple mechanisms that can generate timescale-specific patterns, including different environmental drivers, diverse life histories, dispersal, and non-stationary dynamics. We find that traditional metrics for quantifying synchronous dynamics are often biased toward long-term drivers and may miss the importance of short-term drivers. Our findings indicate key mechanisms to consider when assessing synchronous versus compensatory dynamics and our approach provides a pathway for disentangling these dynamics in natural systems.

Electrochemistry intersected nanoscience 25 years ago when it became possible to control the flow of electrons through single molecules and nanostructures. Many surprises and a wealth of understanding were generated by these experiments. Professor Nongjian Tao was among the pioneering scientists who created the methods and technologies for advancing this new frontier. Achieving a deeper understanding of charge transport in molecules and low-dimensional materials was the first priority of his experiments, but he also succeeded in discovering applications in chemical sensing and biosensing for these novel nanoscopic systems. In parallel with this work, the investigation of a range of phenomena using novel optical microscopic methods was a passion of his and his students. This article is a review and an appreciation of some of his many contributions with a view to the future.

Hexanucleotide GGGGCC repeat expansion in C9ORF72 is the most prevalent genetic cause of amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). One pathogenic mechanism is the aberrant accumulation of dipeptide repeat (DPR) proteins produced by the unconventional translation of expanded RNA repeats. Here, we performed genome-wide CRISPR-Cas9 screens for modifiers of DPR protein production in human cells. We found that DDX3X, an RNA helicase, suppresses the repeat-associated non-AUG translation of GGGGCC repeats. DDX3X directly binds to (GGGGCC)n RNAs but not antisense (CCCCGG)n RNAs. Its helicase activity is essential for the translation repression. Reduction of DDX3X increases DPR levels in C9ORF72-ALS/FTD patient cells and enhances (GGGGCC)n-mediated toxicity in Drosophila. Elevating DDX3X expression is sufficient to decrease DPR levels, rescue nucleocytoplasmic transport abnormalities, and improve survival of patient iPSC-differentiated neurons. This work identifies genetic modifiers of DPR protein production and provides potential therapeutic targets for C9ORF72-ALS/FTD.

Steel Shear Walls (SSWs) are known as structures that dampen the seismic loads like earthquakes in every construction, especially in tall buildings. The SSW contains three critical elements; beam, column, and infill plate. These elements have a significant effect on the seismic performance of the SSWs. The infill plate of SSW is important due to the absorption of the induced external energies. The current research paper studied different configurations of the sandwich panels with trapezoidal corrugated cores as the stiffeners inside the infill plate of the SSWs numerically in the presence of cyclic loading conditions. Two directions the horizontal and vertical are considered for the stiffeners. Also, various numbers of rows and columns are assumed for the stiffeners. The initial model was validated with the published experimental data. According to the average values that obtained, the horizontally and vertically oriented sandwich panels increase the plastic dissipation energy of the SSWs by around 156% and 196%, respectively. The outputs derived from this study will help to increase the SSWs' strength in the built constructions by using sandwich panels inside them. The volatile compounds in the fruits of 24 apricot cultivars were quantitatively and qualitatively determined via headspace solid-phase microextraction and gas chromatography–mass spectrometry (HS-SPME-GC–MS). A total of 429 volatile compounds were detected in these fruits, and the greatest number of detected terpenoids was 77. Significant differences were found among the cultivars in terms of the total volatile compound content of the fruits, with variation from 112.76 ('ZSHYX') to 317.36 µg/g ('JNL'). Using relative odor activity value (rOAV) analysis, 42 key aroma compounds were identified. The rOAVs of (2S,4R)-4-methyl-2-(2-methylprop-1-enyl)oxane, (E)-non-2-enal, (3-methyl-3-sulfanylbutyl) formate, and thiophen-2-ylmethanethiol were above 1000, and most had green, fruity, and woody odors; these results indicated that these substances were important contributors to the overall aroma of the apricot fruits. Our study provides a comprehensive analysis of the volatile compounds from 24 representative apricot cultivars and can aid in the further scientific understanding of the metabolites and aroma in apricots.
