Low-molecular-weight (LMW) gels are a versatile class of soft materials that gained increasing interest over the last few decades. They are made of a small percentage, often lower than 1.0 %, of organic molecules called gelators, dispersed in a liquid medium. Such molecules have a molecular weight usually lower than 1 kDa. The gelator molecules start to interact after the addition of a trigger, and form fibres, whose entanglement traps the solvent through capillary forces. A plethora of LMW gelators have been designed, including short peptides. Such gelators present several advantages: the synthesis is easy and can be easily scaled up; they are usually biocompatible and biodegradable; the gelation phenomenon can be rationalised by making small variation on the peptide scaffold; they find application in several fields. In this thesis, an overview of several peptide based LMW gels is presented. In each study, the gelation conditions were carefully studied, and the final materials were thoroughly investigated. First, the gelation ability of a fluorinated phenylalanine was assessed, to understand how the presence of a rigid moiety and the presence of fluorine may influence the gelation. In this context, a method for the dissolution of sensitive gelators was studied. Then, the control over the gel formation was studied both over time and space, taking advantage of either the pH-annealing of the gel or the reaction-diffusion of a hydrolysing reagent. Some gels were probed for various applications. Due to their ability of trapping water and organic solvents, we used gels for trapping pollutants dissolved in water, as well as a medium for the controlled release of either fragrances or bioactive compounds. Finally, the interaction of the gel matrix with a light-responsive molecule was assessed to understand wether the gel properties or the interaction of the additive with light were affected.

Low molecular weight gelators (LMWGs) based on pseudo-peptides are here studied for the preparation of supramolecular materials. These compounds can self-assemble through non-covalent interactions such as hydrogen bonds and π-π stacking, forming fibres and gels. A wide variety of materials can be prepared starting from these building blocks, which can be tuned and functionalised depending on the application. In this work, derivatives of the three aromatic amino acids L-Phenylalanine, L-Tyrosine and L-DOPA (3,4-dihydroxiphenylalanine) were synthesised and tested as gelators for water or organic solvents. First, the optimal gelating conditions were studied for each compound, varying concentration, solvent and trigger. Then the materials were characterised in terms of mechanical properties and morphology. Water remediation from dye pollution was the first focus of this work. Organogels were studied as absorbent of dyes from contaminated water. Hydrogels functionalised with TiO2 nanoparticles and graphene platelets were proposed as efficient materials for the photo-degradation of dyes. An efficient method for the incorporation of graphene inside hydrogels using the gelator itself as dispersant was proposed. In these materials a high storage modulus coexists with good self-healing and biocompatibility. The incorporation of a mineral phase inside the gel matrix was then investigated, leading to the preparation of composite organic/inorganic materials. In a first study, the growth of calcium carbonate crystals was achieved inside the hydrogel, which preserved its structure after crystal formation. Then the self-assembled fibres made of LMWGs were used for the first time instead of the polymeric ones as reinforcement inside calcium phosphate cements (CPCs) for bone regeneration. This thesis describes the synthesis, the conformational analysis and the applications of pseudopetide foldamers containing the 4-carboxy-5-methyl-oxazolidin-2-one moiety or the pyroglutamic acid unit. These molecules mimic a proline group and have been applied to the formation of oligomers that in solution may easily adopt a stable secondary structure. These pseudoprolines block the peptide bond always in the trans conformation, because the nitrogen atom of the ring is adjacent to both an exocyclic and an endocyclic carbonyl group, forcing them to a strict trans conformation. This remarkable property induces a constrain in the pseudopeptide chain that may help the formation of supramolecular materials. Following a simple methodology we have efficiently prepared a variety of foldamers, whose proprieties may be changed choosing different amino acids, thus providing several secondary structures.

This thesis deals with the synthesis and the conformation analysis of hybrid foldamers containing the 4-carboxyoxazolidin-2-one unit or related molecules, in which an imido-type function is obtained by coupling the nitrogen of the heterocycle with the carboxylic acid moiety of the next unit. The imide group is characterized by a nitrogen atom connected to an endocyclic and an exocyclic carbonyl, which tend always to adopt the trans conformation. As a consequence of this locally constrained disposition effect, these imide-type oligomers are forced to fold in ordered conformations. The synthetic approach is highly tuneable with endless variations, so, simply by changing the design and the synthesis, a wide variety of foldamers with the required properties may be prepared “on demand”. Thus a wide variety of unusual secondary structures and interesting supramolecular materials may be obtained with hybrid foldamers. The behaviour in the solid state of some of these compounds has been analyzed in detail, thus showing the formation of different kinds of supramolecular materials that may be used for several applications. A winning example is the production of a bolaamphiphilic gelators that may also be doped with small amounts of dansyl containing compounds, needed to show the cellular uptake into IGROV-1 cells, by confocal laser scanning microscopy. These gels are readily internalized by cells and are biologically inactive, making them very good candidates in the promising field of drug delivery. In the last part of the thesis, a particular attention was directed to the search of new scaffolds that behave as constrained amino acid mimetics, showing that tetramic acids derivatives could be good candidates for the synthesis and applications of molecules having an ordered secondary structure., Il lavoro di ricerca svolto nel triennio di frequenza della Scuola di Dottorato in Scienze Chimiche ha avuto come obiettivo la sintesi e l’analisi conformazionale di molecole sintetiche, appartenenti alla classe dei foldameri pseudopeptidici. La maggior parte delle molecole sintetizzate hanno come nucleo centrale un’unità eterociclica derivante dalla treonina detta ossazolidinone. Gli acil-derivati di questo scaffold sintetico hanno la particolarità di assumere strutture secondarie stabili e ben definite. In questo ambito è stata messa a punto la sintesi di etero‐oligomeri (contenenti due o più diversi amminoacidi naturali o sintetici), sia lineari sia ciclici per mimare strutture secondarie come eliche, turns o sheets. Alcune classi di oligomeri tendono ad assumere una struttura ordinata in fase solida, presentandosi sotto forma di fibrille osservabili al microscopio ottico. Alcuni composti hanno mostrato interessanti proprietà come gelator, cioè composi in grado di formare gel sia con acqua che con solventi organici. Tali gel si sono dimostrati anche buoni complessanti per sali metallici. Sia i monomeri che gli oligomeri sono stati sintetizzati utilizzando metodi di coupling in soluzione. Una particolare attenzione è stata riposta nella purificazione delle varie molecole. Oltre ai composti di natura ossazolidinonica, è stata valutata la sintesi di altri scaffold sintetici che, una volta inseriti in strutture molecolari più complesse, danno origine a molecole in grado di essere classificate come foldameri. Una parte consistente dell’attività di laboratorio è stata volta alla sintesi di acidi tetrammici, eterocicli ricorrenti in natura. Un’analisi conformazionale delle molecole sintetizzate ha dimostrato come anche questo tipo di molecole sia in grado di auto assemblarsi in strutture secondarie ordinate.

Chemists have long sought to extrapolate the power of biological catalysis and recognition to synthetic systems. These efforts have focused largely on low molecular weight catalysts and receptors; however, biological systems themselves rely almost exclusively on polymers, proteins and RNA, to perform complex chemical functions. Proteins and RNA are unique in their ability to adopt compact, well-ordered conformations, and specific folding provides precise spatial orientation of the functional groups that comprise the “active site”. These features suggest that identification of new polymer backbones with discrete and predictable folding propensities (“foldamers”) will provide a basis for design of molecular machines with unique capabilities. The foldamer approach complements current efforts to design unnatural properties into polypeptides and polynucleotides. The aim of this thesis is the synthesis and conformational studies of new classes of foldamers, using a peptidomimetic approach. Moreover their attitude to be utilized as ionophores, catalysts, and nanobiomaterials were analyzed in solution and in the solid state. This thesis is divided in thematically chapters that are reported below. It begins with a very general introduction (page 4) which is useful, but not strictly necessary, to the expert reader. It is worth mentioning that paragraph I.3 (page 22) is the starting point of this work and paragraph I.5 (page 32) isrequired to better understand the results of chapters 4 and 5. In chapter 1 (page 39) is reported the synthesis and conformational analysis of a novel class of foldamers containing (S)-β3-homophenylglycine [(S)-β3-hPhg] and D- 4-carboxy-oxazolidin-2-one (D-Oxd) residues in alternate order is reported. The experimental conformational analysis performed in solution by IR, 1HNMR, and CD spectroscopy unambiguously proved that these oligomers fold into ordered structures with increasing sequence length.
