Im Rahmen dieser Arbeit wurden einzelne Antennenkomplexe von Purpurbakterien, einzelne Antennenkomplexe höherer Pflanzen sowie einzelne Farbstoffmoleküle in einer Polymermatrix mittels konfokaler Mikroskopie untersucht. Die Chromophore sowohl im Proteingerüst als auch im Polymer dienen hierbei als hochempfindliche Sonden der jeweiligen Umgebung. So zeigen typische Laserfarbstoffe in einer PVB-Matrix eine ausgeprägte spektrale Diffusion, die durch eine Konformationsdynamik der Umgebung bewirkt wird. Korrelationen, die diese Dynamik beschreiben, zerfallen mit Zeitkonstanten, die eine bimodale Verteilung aufweisen. Antennenkomplexe, deren Funktion das Lichtsammeln und Weiterleiten der absorbierten Energie zum Reaktionszentrum ist, spielen eine wesentliche Rolle innerhalb der Photosynthese von Bakterien und Pflanzen. Sie zeigen aufgrund ihrer Pigmentanordnung oft erstaunliche quantenmechanische Eigenschaften. So bildet der Antennenkomplex LH2 von Purpurbakterien ein ringförmiges Aggregat aus 18 Bakteriochlorophyll-Molekülen, die stark gekoppelt sind. Die optischen Eigenschaften werden durch Sprünge in der Symmetrie bestimmt und es zeigt sich, dass bei Temperaturen unterhalb 200 K die Komplexe permanent elliptisch deformiert sind. Im Gegensatz dazu sind Antennenkomplexe höherer Pflanzen (LHC-II) unsymmetrisch aufgebaut. Bei tiefen Temperaturen (1.8 K) wird die Emission von LHC-II durch starke spektrale Diffusion geprägt. Dennoch lässt sich schließen, dass monomerische LHC-II einen emittierenden Zustand, trimerische LHC-II drei Zustände (einen pro monomerischer Untereinheit) aufweisen. Erstaunlicherweise ist die Emission bei Raumtemperatur linear polarisiert. Offenbar exsistiert hier ebenfalls nur ein Emitter pro Monomer. Durch Polarisationsmessungen konnte gezeigt werden, dass räumlich verschieden orientierte Emitter abwechselnd emittieren, jedoch immer nur ein Emitter zur selben Zeit., In this work single antenna complexes of purple bacteria, single antennas of higher plants, and single dye molecules in polymer matrices were investigated using confocal microscopy. Both the chromophores in the protein assembly and within the polymer serve as high sensitive probes of the environment. Typical laser dyes embedded in a PVB film show a pronounced spectral diffusion caused by conformational dynamics of the environment. Correlations which are attributed to this dynamics decays with time constants expelling a bimodal distribution. Antenna complexes harvest light and transfer the absorbed energy to the reaction center thus playing a key role within the photosynthesis of bacteria and plants. Due to the arrangement of the pigments antenna complexes often show amazing quantum mechanical properties. The antenna complex LH2 of purple bacteria forms a ring shaped aggregate of 18 strongly coupled bacteriochlorophyll molecules. The optical properties of LH2 complexes are determined by jumps of the symmetry and it could be shown that at temperatures below 200 K the complex is permanently elliptically deformed. In contrast, antenna complexes of plants (LHC-II) possess an unsymmetrical structure. At low temperatures (1.8 K) the emission is determined by strong spectral diffusion. Nevertheless, it could be shown that monomeric LHC-II has one emitting state, trimeric LHC-II three emitting states one per monomeric subunit. Astonishingly, the emission of monomeric LHC-II at room temperature is linearly polarized indicating one emitter per monomer at higher temperatures, too. Additionally, polarization studies showed that spatially different orientated emitters alternate in emission, but there is only one state emitting at the same time.
