Background: Anthropogenic activities occurring throughout the Sonoran Desert are replacing and fragmenting habitat and reducing landscape connectivity for the Sonoran desert tortoise (Gopherus morafkai). Understanding how the structure of the landscape influences tortoise habitat use and movement can help develop strategies for mitigating the impacts of these landscape alterations, which are conservation actions needed to support the species' long-term persistence. However, how natural and anthropogenic features influence fine-scale habitat use and movement of Sonoran desert tortoises remains unclear. Methods: The goals of this study were to (1) understand how characteristics of the landscape shape tortoise habitat use and movement in order to (2) identify factors that may reduce habitat use or threaten landscape connectivity for the species by discouraging or restricting movement. We collected GPS telemetry data from 17 adult tortoises tracked for two summer monsoon seasons, when tortoises are most active, in a U.S. National Monument along the international border between Arizona, USA and Sonora, Mexico. We used Hidden Markov Models (HMMs) to assign GPS locations to an encamped or a moving state. We used the moving state data in integrated Step Selection Analyses (iSSA) to examine how range-resident Sonoran desert tortoises select habitat and respond to landscape features while moving. Results: Tortoises selected to move through areas of intermediate vegetation cover and terrain ruggedness and avoided areas far from desert washes and close to low-traffic roads. Tortoises increased their speed when approaching or crossing low-traffic roads but showed no detectable response to a highway. Conclusion: Bare earth or high vegetation cover, flat or extremely rugged terrain, areas far from desert washes, and low-traffic roads may discourage or restrict tortoise movement. Therefore, preventing the development of roads, activities that degrade washes, and activities that thin, remove, or greatly increase vegetation cover may encourage tortoise habitat use and movement within those habitats. Simple Summary: This study explored the morphological differences in the carapaces of three tortoise species—Leopard tortoises (Stigmochelys pardalis), African spurred tortoises (Centrochelys sulcata), and Greek tortoises (Testudo graeca)—raised in captivity in Türkiye. Using 3D scanning and geometric morphometric analysis, we identified the vital shape variations and dimensional features across species. Principal component analysis revealed that PC1 and PC3 were significant in distinguishing species, with Leopard tortoises showing higher carapace shapes and African spurred tortoises having shorter carapaces. An allometric effect indicated that smaller tortoises had higher shells. These findings provide valuable insights for taxonomy and clinical practice, emphasizing the need to consider shape variations in tortoise care and species identification. The carapace morphology of tortoises is a crucial characteristic used for species identification, with features such as shell shape, roughness, and color patterns varying among species. Using 3D scanners, the carapaces were modeled, and a 3D geometric morphometric method was employed to analyze shape variations and dimensional features, with landmarks applied automatically. Among the species studied, African spurred tortoises had the largest carapace size. Principal component analysis (PCA) identified PC1 and PC3 as critical factors in distinguishing between species based on morphological characteristics. Positive PC1 values, associated with a shorter carapace height, indicated a flatter or more compact shell shape. A higher PC3 value corresponded to a raised shape at the back of the shell, while a lower PC3 value indicated a raised shape at the front. Specifically, Leopard tortoises exhibited a higher carapace shape than the other species, while African spurred tortoises had shorter carapaces. An allometric effect was observed in the carapaces, where smaller specimens tended to be proportionately higher-domed, whereas larger shells displayed a lower height in shape. These findings highlight the significance of shape variations in tortoise shells, which emerge during adaptation and have important implications for taxonomy and clinical practice. Hyaena faeces and mineral licks represent highly concentrated nutrient resources for leopard tortoises (Stigmochelys pardalis) living in nutrient‐poor environments. Here, we provide direct evidence for the consumption of hyaena faeces and artificial mineral licks by leopard tortoises in the Kalahari Desert. Tortoises aggregated at hyaena latrines and lick sites during the wet season between 10 AM and 6 PM in groups of 1–5 individuals. Where both salt and mineral licks were available, tortoises displayed a preference for salt. We suggest that calcium, phosphorus and sodium deficits are likely driving coprophagy and geophagy behaviours by tortoises in the Kalahari. [ABSTRACT FROM AUTHOR]

Natal dispersal is a crucial period for raptors with serious implications for individuals' survival and population demography. In this study we analyzed data from 18 GPS-tracked golden eagles in order to describe their dispersal ecology in northern Greece, where the species feeds mostly on tortoises. Young eagles in our population dispersed at 176 days post fledging, spent their first year of independence relatively close (40–60 kms) to their natal ranges and exhibited a variable temporary settlement behavior. Overall dispersal range sizes did not differ seasonally, but temporary settlement area range sizes were significantly larger in winter. Three eagles survived to territorial settlement and occupied ranges 20–60 kms from their natal areas. The application and refinement of the Scottish GET dispersal ranging model suggested that eagles used areas that had higher topographical relief and lower canopy cover during their natal dispersal. Habitat heterogeneity seems to also be influential during temporary settlement. Our study is the first to provide both such insights for golden eagles in southern eastern Europe and a method for delineating temporary settlement areas for the species. Our findings can be explained in terms of food and habitat availability. We highlight the importance of conserving heterogeneous open areas of complex topography and applying proactive management measures within temporary settlement areas for our population's conservation. Reproduction of the pancake tortoise in the wild has remained poorly known over the decades. This study fills the knowledge gap by investigating the reproductive ecology of the species in its natural habitats in Kenya. Data were collected using the time‐constrained search‐and‐seize method, group‐level scan sampling, radio tracking, and camera‐trapping. Our observations revealed a total of 19 mating events that occurred between December and April, predominantly during the rainy seasons. Mating duration averaged 15.2 ± 2.4 min. Male–male aggression was observed during the mating season, with the dominant male keeping off all other males and mating with the resident females. Nest preparation events occurred between April and June. Nine nesting events were recorded, but in only five of them was an egg deposited. The nests were dug in loose soil, appeared almost circular, and measured 8.4 ± 0.9 cm wide and 7.0 ± 0.1 cm deep (n = 7). The average clutch size was one egg. The eggs were white, hard‐shelled, oval, and elongated measuring 4.4 ± 0.4 cm long, 2.7 ± 0.04 cm wide and weighed 17 ± 0.6 g (n = 3). Incubation period lasted 177 ± 5.7 days (n = 2), and hatching coincided with the onset of the short rainy season in November, aligned with the availability of abundant food for the tortoises. The hatchlings were minimally wider than long, having an average straight carapace length of 4.5 ± 0.6 cm, an average width of 4.5 ± 0.4 cm, and on average weighed 14.7 ± 3.7g, (n = 9). They remained near the nest site for 4–5 days before relocating to tiny rock crevices away from the adult tortoise crevices. Egg and hatchling predation was high, with four of the total of six eggs, observed after they were laid, being destroyed. Three juveniles, out of nine, were also eaten by predators. The Spur-thighed tortoise has a large geographic distribution and can be infested by several tick species, mainly Hyalomma aegyptium. This tick has recently been confirmed as a natural carrier of numerous pathogenic, including zoonotic agents. The purpose of this study was to estimate the level of hard tick infestation in Mediterranean tortoises living in some regions of the west of Algeria (Oran, Mostaganem, and Tiaret) during the spring and summer of the year 2019. Among 39 turtles collected from the different study regions, 22 tested positive, which represents an infestation rate of 56.41 %. A total of 224 adult ticks and one nymph were collected, which can display a tick infestation intensity of 10.23 %. All of these ticks belong to a single species, which is Hyalomma aegyptium. In terms of abundance, this study showed 5.78 ticks/tortoise. Regarding sex, we have collected 198 male and 26 female ticks with a sex ratio of 7.62. The preferred attachment sites of Hyalomma aegyptium ticks were the inguinal, the limbs, and to a lesser degree, the neck. A few rare cases have been collected from the carapace. Further studies are needed to assess the impact of the intensity of H. aegyptium infestation on the tortoise's health status and to assess the potential transmission of zoonotic pathogens. The diverse climatic and geological history of Southeast Asia has been a driving force behind significant Pleistocene and Holocene range dynamics and the consequent biological diversification in the region. Among the species exhibiting wide distributions, extending across several well-known zoogeographic barriers, the Elongated Tortoise (Indotestudo elongata) is noted for its extensive variability in size, shape, and colouration. To examine phylogeographic differentiation within I. elongata, we analysed three mitochondrial gene fragments (ND4, cyt b, and COI) for 52 individuals including fourteen historic specimens and 25 sequences obtained from Gen- Bank. Phylogenetic analyses of the concatenated alignment revealed genetic homogeneity across the range, supported by a shallow mean uncorrected p distance of 1.2% in cyt b. This corresponds to a surprising lack of biogeographic structuring. Concordantly, multivariate statistical analyses of morphometric and colouration-related characters of 166 adult tortoises (79 males and 87 females) from Myanmar, Thailand, Cambodia, and Vietnam showed no statistically significant differences between biogeographic units. The lack of any phylogeographic signal and morphological differentiation despite the presence of significant biogeographic barriers is unusual for a terrestrial vertebrate from this region. Archaeological evidence suggests that Elongated Tortoises have been used as a food item for millennia. It could be that their current wide distribution and the absence of genetic differentiation are the result of a significant range expansion caused by extensive human-mediated dispersal over the prehistoric and historic tortoise trade. Background: Obtaining de novo chromosome-level genome assemblies greatly enhances conservation and evolutionary biology studies. For many research teams, long-read sequencing technologies (that produce highly contiguous assemblies) remain unaffordable or unpractical. For the groups that display high synteny conservation, these limitations can be overcome by a reference-guided assembly using a close relative genome. Among chelonians, tortoises (Testudinidae) are considered one of the most endangered taxa, which calls for more genomic resources. Here we make the most of high synteny conservation in chelonians to produce the first chromosome-level genome assembly of the genus Testudo with one of the most iconic tortoise species in the Mediterranean basin: Testudo graeca. Results: We used high-quality, paired-end Illumina sequences to build a reference-guided assembly with the chromosome-level reference of Gopherus evgoodei. We reconstructed a 2.29 Gb haploid genome with a scaffold N50 of 107.598 Mb and 5.37% gaps. We sequenced 25,998 protein-coding genes, and identified 41.2% of the assembly as repeats. Demographic history reconstruction based on the genome revealed two events (population decline and recovery) that were consistent with previously suggested phylogeographic patterns for the species. This outlines the value of such reference-guided assemblies for phylogeographic studies. Conclusions: Our results highlight the value of using close relatives to produce de novo draft assemblies in species where such resources are unavailable. Our annotated genome of T. graeca paves the way to delve deeper into the species' evolutionary history and provides a valuable resource to enhance direct conservation efforts on their threatened populations. Background: The popularity of tortoises kept in captivity is increasing and has caused concern regarding the necessity to establish safe and straightforward anaesthesia for those reptiles. Aim: This study aimed to compare four protocols using levobupivacaine in spinal anaesthesia for the blockade of the caudal neuraxis of red-footed tortoises (Chelonoidis carbonarius). Methods: Twenty-four tortoises were randomly assigned into four groups: G1, levobupivacaine 0.75% (1.15 mg kg−1); G2, levobupivacaine 0.37% (1.15 mg kg−1); G3, levobupivacaine 0.75% (2.3 mg kg−1); and G4, levobupivacaine 0.75% (0.1 ml 5 cm−1 of straight carapace length). Tortoises were evaluated for respiratory rate, muscle relaxation, response to hindlimb or tail pinch, and cloacal reflex. Results: A 1.15 mg kg−1 dose of levobupivacaine 0.37% appears adequate for shorter procedures, whereas a 1.15 mg kg−1 dose of levobupivacaine 0.75% should be appropriate for longer procedures in red-footed tortoises. Conclusion: Our results are the first to show the effects of levobupivacaine on spinal anaesthesia in reptiles. Weight-based doses presented more intense and more homogeneous effects than carapace length-based doses in red-footed tortoises. Simple Summary: Some trace elements are essential for the health of vertebrates, but only limited data are available on blood levels in tortoises. The aim of this study was therefore to measure trace elements in heparinized blood plasma of Hermann's tortoises (Testudo hermanni) (n = 520) from March to September 2022 using inductively coupled plasma mass spectrometry (ICP-MS) and to establish specific reference intervals. Additionally, the influence of sex, season, and region of sample collection on the measured values were evaluated. Significant (p ≤ 0.05) sex-specific, seasonal, and regional differences were found for a number of trace elements indicating that all of these factors should be considered when establishing and interpreting blood trace element levels in tortoises. Oracle bones (Obs) are a significant carrier of the shang dynasty civilization, primarily consisting of tortoise shells and animal bones, through the study of which we can gain a deeper understanding of the political, economic, religious, and cultural aspects of the shang dynasty. The oracle bone drill chisel (Obdc) is considered an essential non-textual material. The segmentation of Obdc assists archaeologists determine the approximate age of the Obs, which possesses considerable research value. However, the breakage of thousands of years of underground buried Obs, the blurring of the edges of the area burned by the Obdc, the different shapes, and the inconsistent number have brought challenges to the accurate segmentation of the Obdc. In this article, we propose a group convolutional attention and pvt dual-branch network (GCA-PVT-Net) for Obdc segmentation. To our knowledge, this paper is the first to research the automatic segmentation of Obdc. It is a hybrid Convolutional neural network (CNN) and Transformer framework. The work offers the following contributions: (1) The Obdc images are labeled based on the delineation criteria of different drill chisel (DC) shapes to create the Obdc dataset. (2) A convolutional attention module (CAM) is proposed as both an encoder and decoder. The feature extraction process, which effectively integrates global and local information, ensures better modeling of long-term correlations in images while preserving details. (3) A channel feature aggregation module (CFAM) is designed to enhance the effective integration of channel features, enabling feature fusion across various branches and at different levels. (4) The edge deep supervision strategy is applied to smooth the jagged edge of the predicted images at the decoder's end. Extensive experiments on the Obdc dataset show that GCA-PVT-Net outperforms other state-of-the-art (SOTA) methods. Background: Hepatozoon fitzsimonsi (Dias, 1953) is a frequently found haemogregarine of southern African tortoises. At the time of this species' reassignment from the genus Haemogregarina to Hepatozoon, developmental stages such as sporocysts and sporozoites were observed in ticks associated with H. fitzsimonsi parasitised and non-parasitised tortoises. It was thus suggested that ticks may act as the potential vectors for this parasite. However, this earlier research was unable to confirm the identity of these sporogonic stages using molecular markers. In a separate study aimed at identifying tick species parasitising South African reptiles and molecularly screening these for the presence of Hepatozoon, that study identified H. fitzsimonsi in tortoise-associated ticks. Thus, the present study aimed to revisit the potential of ticks to act as vectors for H. fitzsimonsi in tortoises using a combined microscopy and molecular approach. Methods: Specimens of Kinixys natalensis, Kinixys spekii, Kinixys zombensis and Stigmochelys pardalis were collected from Bonamanzi and Ndumo Game Reserve, South Africa. Upon capture, animals were examined for ticks, and these were collected along with blood and other tissues. Adult ticks were dissected and visceral impression slides were prepared along with thin blood and tissue smears on clean microscope slides. Smears and impression slides were stained with Giemsa, screened and micrographs of parasites were captured. Two primer sets were employed to target fragments of the 18S rRNA gene of parasites found in both tortoises and ticks and the resulting sequences were then compared with other known H. fitzsimonsi and haemogregarine sequences from the GenBank database. Results: Peripheral blood gamont and liver merogonic stages were observed in S. pardalis, while the sporogonic stages were observed in the haemocoel of Amblyomma ticks. Gamont and sporocyst stages compared morphologically with previous descriptions of H. fitzsimonsi, identifying them as this species. Phylogenetic analysis revealed that the blood and tick sequences obtained in this study clustered in a monophyletic clade comprising known H. fitzsimonsi. Conclusions: The present study provides further support for ticks acting as the vectors of H. fitzsimonsi by molecularly identifying and linking observed developmental stages in tortoises (S. pardalis) with those in the invertebrate host (Amblyomma spp.). The present investigation examined the oropharyngeal roof of two turtles having different feeding behaviors: the landform Greek tortoise (Testudo graeca graeca) primarily herbivores and the semi-aquatic red-eared slider turtle (Trachemys scripta elegans) lives in freshwater that opportunistic omnivorous grossly and by scanning and light microscopes. Grossly, the Greek tortoise had a V-shaped roof consisting of the upper rhamphotheca, peri-palatine region, upper alveolar ridge, peripheral palatine ridge, median palatine ridge, vomer, choanae, caudal palatine part, and pharynx. At the same time, the red-eared slider had a semilunar roof consisting of upper rhamphotheca, two peripheral palatine ridges, core of palatine ridges, upper alveolar band, vomer, choanae, caudal palatine part, and pharynx. SEM revealed that the red-eared slider roof appeared more straightforward. The upper rhamphotheca is sharp, with a median premaxillary notch in the red-eared slider that gives a powerful bite for cutting to compensate absence of the teeth. Additionally, the red-eared slider's upper alveolar band is interrupted by a single upper alveolar ridge that appears spiky, pointed, and longer as it needs powerful chewing of prey and there are two types of teeth-like projections at its peri-palatine area for food-crushing and chewing. The Greek tortoise palatine region had numerous ridges and folds to provide roughness for food processing. Greek tortoises had small-sized choanae with two choanal folds to minimize choanal openings when eating dusty grasses. Histologically, Greek tortoise palate was rostrally thicker and more keratinized than caudally, and the caudal palatine region was characterized by a single pair of circumvallate-like papilla with multiple mucous openings and secretions, while red-eared slider palate was slightly keratinized at the peri-choanal region, and the rest of the palate was non-keratinized with few mucous openings. Migratory, long‐lived animals are an important focus for life‐history theory because they manifest extreme trade‐offs in life‐history traits: delayed maturity, low fecundity, variable recruitment rates, long generation times, and vital rates that respond to variation across environments. Galapagos tortoises are an iconic example: they are long‐lived, migrate seasonally, face multiple anthropogenic threats, and have cryptic early life‐history stages for which vital rates are unknown. From 2012 to 2021, we studied the reproductive ecology of two species of Galapagos tortoises (Chelonoidis porteri and C. donfaustoi) along elevation gradients that coincided with substantial changes in climate and vegetation productivity. Specifically, we (1) measured the body and reproductive condition of 166 adult females, (2) tracked the movements of 33 adult females using global positioning system telemetry, and monitored their body condition seasonally, (3) recorded nest temperatures, clutch characteristics, and egg survival from 107 nests, and (4) used radiotelemetry to monitor growth, survival, and movements of 104 hatchlings. We also monitored temperature and rainfall from field sites, and remotely sensed primary productivity along the elevation gradient. Our study showed that environmental variability, mediated by elevation, influenced vital rates of giant tortoises, specifically egg production by adult females and juvenile recruitment. Adult females were either elevational migrants or year‐round lowland residents. Migrants had higher body condition than residents, and body condition was positively correlated with the probability of being gravid. Nests occurred in the hottest, driest parts of the tortoise's range, between 6 and 165 m elevation. Clutch size increased with elevation, whereas egg survival decreased. Hatchling survival and growth were highest at intermediate elevations. Hatchlings dispersed rapidly to 100–750 m from their nests before becoming sedentary (ranging over <0.2 ha). Predicted future climates may impact the relationships between elevation and vital rates of Galapagos tortoises and other species living across elevation gradients. Abstract The anatomy of North American tortoises is poorly understood, despite a rich fossil record from the Eocene and younger strata. Stylemys nebrascensis is a particularly noteworthy turtle in this regard, as hundreds of specimens are known from Oligocene deposits, and as this species is one of the earliest fossil turtles to have been described in the scientific literature. Since its initial description based on a shell, many specimens with more complete material have been referred to Stylemys nebrascensis. Here, we review and confirm the referral of an important historic specimen to Stylemys nebrascensis, which includes shell, non-shell postcranial, and skull material. This allows us to document unique skull features of Stylemys nebrascensis (e.g., an unusual ‘poststapedial canal’ that connects the posterior skull surface with the cavum acustico-jugulare) and to refer another well-preserved skull to the species. Based on computed-tomography scanning of these two skulls, we provide a detailed description of the cranial and mandibular osteology of Stylemys nebrascensis. Stylemys nebrascensis has a combination of plesiomorphic skull characteristics (e.g., retention of a medial jugal process) and derived traits shared with extant gopher tortoises (e.g., median premaxillary ridge) that suggest it may be a stem-representative of the gopher tortoise lineage. This supports the hypothesis that extant and fossil tortoises from North America form a geographically restricted clade that split from Asian relatives during the Paleogene.

Clinical assessment of body condition is crucial in captive and free-ranging reptiles, since a large percentage of diseases result from inadequate nutrition. However, preventive health care is restricted by the lack of a practical method for the assessment in tortoises. Pre-existing evaluation systems based on weight and shell measurements are laborious and ignore the clinical presentation of the animal. The present study aimed to facilitate the assessment by establishing a body condition score. A total of 373 Hermann's Tortoises (Testudo hermanni) (n = 281 tortoises kept as pets in Germany and n = 92 tortoises originating from a free-ranging population (68) or a rearing station (24) in France) were examined and data (weight (g), carapace length (cm), width (cm), height (cm)) were recorded in a standard protocol between October 2020 and October 2021. A modified version of a body condition score for Mojave Desert Tortoises (Gopherus agassizii) (1 = cachectic, 3 = ideal, 5 = obese) was utilized and tested against pre-existing shell measurement systems (Jackson's ratio, body condition index, volume condition index, circumferential product). German captive tortoises were significantly heavier and larger than French specimens. In the Spearman's correlation matrix, the body condition score showed a statistically significant correlation with all measurement methods in the total population of captive tortoises (Testudo hermanni boettgeri), with a medium correlation strength, and a lack of correlation in free-ranging tortoises (Testudo hermanni hermanni). However, individual animal data suggested misleading results of mathematical equations in terms of body condition. Clinical evaluation of tortoises, including a body condition score, should be considered essential to provide good healthcare and should be an integral part of general examination. Taking the accidental death of a giant tortoise in the zoo in 1969 as its starting point, the paper studies the movement of animals, dead and alive. It traces the acquisition history of two new Galápagos giant tortoises by following the animals' trajectory from Ecuador to West Berlin during the Cold War. At the same time, it reconstructs the itineraries of the dead animals' remains through different natural history institutions, leading into the shared history of trade and traffic between zoos and natural history museums and into the political geographies of the Cold War. To put these episodes into a broader historical context, the article looks back on the expansion of global networks in animal trade through the lens of transport infrastructures, trading networks, and the accompanying red tape, tracing this back to the period around 1900 in order to reveal genealogies, thereby historicizing current logistical practices, policies, and problems. Taking logistics as a critical analytical category for natural history and thus examining natural history as logistics allows us to gain new insights into how logistics not only enabled but also shaped and reshaped natural history's material, economic, and epistemic frameworks. Anaerobic gut fungi (AGF, Neocallimastigomycota) reside in the alimentary tract of herbivores. While their presence in mammals is well documented, evidence for their occurrence in non-mammalian hosts is currently sparse. Culture-independent surveys of AGF in tortoises identified a unique community, with three novel deep-branching genera representing >90% of sequences in most samples. Representatives of all genera were successfully isolated under strict anaerobic conditions. Transcriptomics-enabled phylogenomic and molecular dating analyses indicated an ancient, deep-branching position in the AGF tree for these genera, with an evolutionary divergence time estimate of 104-112 million years ago (Mya). Such estimates push the establishment of animal-Neocallimastigomycota symbiosis from the late to the early Cretaceous. Further, tortoise-associated isolates (T-AGF) exhibited limited capacity for plant polysaccharides metabolism and lacked genes encoding several carbohydrate-active enzyme (CAZyme) families. Finally, we demonstrate that the observed curtailed degradation capacities and reduced CAZyme repertoire is driven by the paucity of horizontal gene transfer (HGT) in T-AGF genomes, compared to their mammalian counterparts. This reduced capacity was reflected in an altered cellulosomal production capacity in T-AGF. Our findings provide insights into the phylogenetic diversity, ecological distribution, evolutionary history, evolution of fungal-host nutritional symbiosis, and dynamics of genes acquisition in Neocallimastigomycota. Blood gas analysis reflects the exchange of oxygen and carbon dioxide in the lungs. This test provides important information, since the relationship between these gases has a direct impact on the acid-basic balance in the body. Given the significance of blood gas analysis in Brazilian reptiles, this study set out to establish temperature-corrected and uncorrected reference intervals for venous blood gas measurements in Chelonoidis carbonarius, and to compare values between females and males. In this study, 19 animals were used, 8 males and 11 females. Blood samples were collected from the dorsal coccygeal vein, and the analyses were performed immediately after blood sample collection. The following parameters were measured: pH, PO2, HCO3-, TCO2, BEecf, Na, K, ICa, and Glu, and were compared between females and males. Additionally, pH, pCO2, and pO2 values were compared with and without temperature correction. Oxygen saturation and Na levels were significantly higher (p<0.05) in males. Furthermore, it was possible to infer that the lower the body temperature relative to the environmental temperature, the larger the difference in pH following temperature correction. Aeromonas dhakensis (A. dhakensis) is becoming an emerging pathogen worldwide, with an increasingly significant role in animals and human health. It is a ubiquitous bacteria found in terrestrial and aquatic milieus. However, there have been few reports of reptile infections. In this study, a bacterial strain isolated from a dead Aldabra giant tortoise was identified as A. dhakensis HN-1 through clinical observation, matrix-assisted laser desorption ionization–time of flight mass spectrometry (MALDI-TOF/MS), and gene sequencing analysis. Subsequently, to evaluate its pathogenicity, the detection of virulence genes and mice infection experiments were performed. A. dhakensis HN-1 was found to contain seven virulence genes, including alt, ela, lip, act, aerA, fla, and hlyA. Mice infected with A. dhakensis HN-1 exhibited hemorrhage of varying degrees in multiple organs. The half-maximal lethal dose (LD50) value of A. dhakensis HN-1 for mice was estimated to be 2.05 × 107 colony forming units (CFU)/mL. The antimicrobial susceptibility test revealed that A. dhakensis HN-1 was resistant to amoxicillin, penicillin, ampicillin and erythromycin. This is the first report of A. dhakensis in Aldabra giant tortoises, expanding the currently known host spectrum. The anatomy of North American tortoises is poorly understood, despite a rich fossil record from the Eocene and younger strata. Stylemys nebrascensis is a particularly noteworthy turtle in this regard, as hundreds of specimens are known from Oligocene deposits, and as this species is one of the earliest fossil turtles to have been described in the scientific literature. Since its initial description based on a shell, many specimens with more complete material have been referred to Stylemys nebrascensis. Here, we review and confirm the referral of an important historic specimen to Stylemys nebrascensis, which includes shell, non-shell postcranial, and skull material. This allows us to document unique skull features of Stylemys nebrascensis (e.g., an unusual 'poststapedial canal' that connects the posterior skull surface with the cavum acustico-jugulare) and to refer another well-preserved skull to the species. Based on computed-tomography scanning of these two skulls, we provide a detailed description of the cranial and mandibular osteology of Stylemys nebrascensis. Stylemys nebrascensis has a combination of plesiomorphic skull characteristics (e.g., retention of a medial jugal process) and derived traits shared with extant gopher tortoises (e.g., median premaxillary ridge) that suggest it may be a stem-representative of the gopher tortoise lineage. Invasive alien species are among the most pervasive threats to biodiversity. Invasive species can cause catastrophic reductions in populations of native and endemic species and the collapse of ecosystem function. A second major global conservation concern is the extirpation of large‐bodied mobile animals, including long‐distance migrants, which often have keystone ecological roles over extensive spatial extents. Here, we report on a potentially catastrophic synergy between these phenomena that threatens the endemic biota of the Galapagos Archipelago. We used GPS telemetry to track 140 migratory journeys by 25 Western Santa Cruz Island Galapagos tortoises. We plotted the spatial interaction between tortoise migrations and recently established non‐native forest dominated by the invasive tree Cedrela odorata (Cedrela forest). We qualified (a) the proportion of migratory journeys that traversed Cedrela forest, and (b) the probability that this observed pattern occurred by chance. Tortoise migrations were overwhelmingly restricted to small corridors between Cedrela forest blocks, indicating clear avoidance of those blocks. Just eight of 140 migrations traversed extensive Cedrela stands. Tortoises avoid Cedrela forest during their migrations. Further expansion of Cedrela forest threatens long‐distance migration and population viability of critically endangered Galapagos tortoises. The prickly pear (Opuntia megasperma var. orientalis), a pivotal species for the ecological balance of Española Island in Galapagos, has witnessed a severe decline in its population due to the enduring presence of introduced feral goats over several decades. Additionally, the inherent slow recovery of this species, requiring several years of development, has contributed to its population decline. Several attempts were carried out to restore this species, but they were not successful due to the island's extreme arid conditions. Subsequently, innovative water-saving technologies were introduced to ensure the survival and growth of the Opuntia species. Two water-saving technologies, Groasis Waterboxx® and Hydrogel, were applied in two distinct treatments, the first involving Waterboxx solely, and the second combining Waterboxx with Hydrogel, alongside a control group. Planting involved two types of cacti: cladodes and seedlings. To safeguard against potential damage from giant tortoises and local birds, protective mesh fencing was installed around the plants. Each monitoring session recorded plant survival and growth, evaluating the impact of water-saving technologies on cactus survival, maximum plant height reached, age at the time of plant death, and growth achieved since planting. Additionally, the study assessed the influence of climate on plant survival and growth. Unfortunately, the employment of protective mesh fences and Waterboxx containers resulted in the unintended loss of specific bird species. Consequently, a decision was taken to remove these protective measures, resulting in a substantial rise in herbivorous activity, and the subsequent mortality of nearly all plants. Our findings underscore the efficacy of water-saving technologies in Opuntia restoration. However, successful application necessitates a better understanding of these technologies within the unique conditions of the island. Simple Summary: This article describes the diagnostic evaluation and therapy of a tortoise with a bulging oral lesion of unknown origin. Initially, the animal owner indicated that a similar oral mass at the same location had been surgically removed four years ago. At that time, the abnormal oral structure had not been evaluated further. This time, the mass was removed surgically and further examined by both veterinary pathologists (veterinarians specialised in the examination of animal diseases and body structures) and virologists. No causal agents, which have been described before to trigger comparable lesions in tortoises, were found. Parts of the removed lesion were examined in further virological studies to scan the tissue material for potential new infectious agents, which might be connected to the abnormal oral lesion. Indeed, the authors were able to detect virus material within the tissue mass belonging to a comparably new virus family. The exact influence of these agents on the origin of the lesion, though, remains unclear. An adult male Bell's hinge-back tortoise (Kinixys belliana) was admitted to a veterinary clinic due to a swelling in the oral cavity. Physical examination revealed an approximately 2.5 × 1.5 cm sized, irregularly shaped tissue mass with villiform projections extending from its surface located in the oropharyngeal cavity. An initial biopsy was performed, and the lesion was diagnosed as squamous papilloma. Swabs taken for virological examination tested negative with specific PCRs for papillomavirus and herpesvirus. Further analysis of the oropharyngeal mass via metagenomic sequencing revealed sequence reads corresponding to a member of the family Adintoviridae. The tissue mass was removed one week after the initial examination. The oral cavity remained unsuspicious in follow-up examinations performed after one, five and twenty weeks. However, a regrowth of the tissue was determined 23 months after the initial presentation. The resampled biopsy tested negative for sequence reads of Adintoviridae. Conclusively, this report presents the diagnostic testing and therapy of an oral cavity lesion of unknown origin. Roads have often been identified as barriers to the movement of free-ranging animals. However, whether restoration of landscape connectivity across roadways can mitigate barriers to movement is insufficiently understood in light of indirect effects of roads on wildlife movement. We GPS-tagged free-ranging Mojave desert tortoises (Gopherus agassizii) to quantify movement behavioral states using hidden Markov models in relation to a major highway and to document use of existing, permeable culverts. We then used the observed movement behaviors to parameterize simulations of tortoise movement to evaluate alternative culvert designs and placements for enhancing connectivity across the roadway. Tortoises were most active during mid-day, in warm temperatures, and when close to the highway. The highway affected transition probabilities between movement states, as females were more likely than males to switch to an energy-demanding traveling movement state, remain in that state, and move farther than usual within that state. In contrast, males were more likely than females to continue in the low-energy resting state when close to the highway, but if traveling, to travel farther than usual. We observed two highway crossings by a tagged tortoise, which was a higher rate of crossing than in simulated tortoises. Simulated crossing rates increased with culvert size and culvert density, and size and density appeared more important for crossing than if culverts were placed singly or in pairs. Existing culvert densities across the region appeared potentially sufficient for long-term genetic connectivity, but only if retrofitted to allow for tortoise access and passing. We concluded that existing highway traffic may indirectly depress tortoise populations adjacent to the highway, particularly via negative impacts to female movements, and that existing culverts in washes should be retrofitted to allow for periodic tortoise crossings to improve structural connectivity for occasional passage. Background: The Zarudny's spur-thighed tortoise or Iranian tortoise (Testudo graeca zarudnyi) has just been reported from Iran so far. Therefore, the purpose of this study was to determine the effects of season and gender on serum biochemical parameters of this valuable species in Iran. Results: This study was performed on 20 clinically healthy adult Zarudny's spur-thighed tortoises. Blood samples were collected from the jugular vein and then serum biochemical parameters and body weight were measured in autumn and winter. The following biochemical parameters were measured: total cholesterol, triglyceride, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, very low-density lipoprotein, total protein, creatinine, urea, glucose, calcium, inorganic phosphorus, total bilirubin, uric acid, alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase and lactate dehydrogenase. Urea concentration increased significantly at emergence from hibernation. Also, in winter, total protein, phosphorous, creatinine, total bilirubin concentrations and alkaline phosphatase activity decreased significantly compared to autumn, but aspartate aminotransferase and lactate dehydrogenase activities were significantly higher than in autumn. There was no significant difference for the parameters mentioned above by gender, except for phosphorous. Phosphorous concentration in females was significantly higher than that of males in autumn. Cholesterol and low-density lipoprotein cholesterol levels in females were significantly higher than males in autumn and winter. Conclusions: The present study is the first study that monitors the serum biochemical parameters of adult Zarudny's spur-thighed tortoises based on season and gender. Seasonal reference intervals should be used for biochemical parameters in this valuable species. Also, sex-specific reference intervals for phosphate and cholesterol are necessary. Objective: The literature does not clearly specify the locations inhabited by the spurthighed tortoise (Testudo graeca) from the eastern Black Sea coast of Türkiye. This study thus aimes to reveal new locality records for T. graeca in Trabzon province. Materials and Methods: Two adult male specimens were caught from the Darıca and Konaklar neighborhoods in the respective Akçaabat and Ortahisar districts. Some of the morphological characteristics of these specimens have been recorded using a digital caliper. After taking morphometric measurements, the tortoises were returned to their habitat. No anesthetic procedure was performed on the turtles. Results: Both the Darıca and Konaklar specimens have five vertebral scutes, 11 pairs of marginal scutes, and four pairs of costal scutes on their carapace. In addition, both specimens were seen to have one undivided supracaudal scute and one nuchal scute on their carapace. The Darıca specimen has a straight carapace length (SCL) of 208.19 mm, and plastron length (PL) of 188.08 mm while the Konaklar specimen has an SCL of 216.33 mm and a PL of 196.28 mm. Conclusion: The study compared its specimens' the pholidosis and morphometric characteristics and color-pattern features with those of specimens reported in the literature. The morphological features of the Darıca and Konaklar specimens are similar to those for the samples of Testudo graeca ibera in the literature. Jalthal Forest (62.6 km2) is a remnant forest patch of a once continuous, lush and dense area of sal Shorea robusta and mixed deciduous vegetation in eastern lowland Nepal that is now surrounded by a dense human population. The forest is one of the last remnants of suitable habitat for the Critically Endangered yellow-headed tortoise Indotestudo elongata. This study explored the distribution of and conservation threats facing the yellow-headed tortoise in Jalthal Forest through field surveys in 20 community forests and structured interviews with 80 members of 22 community forest user groups and local people during September–November 2021. We surveyed 11.3 km2 of the forest and observed only four individuals of I. elongata , indicating a population density of 0.35/km2. Local people perceived that the species is decreasing mainly because of hunting (42%) and habitat deterioration from incursion of invasive species (30%). Based on the interviews with local people involved in hunting, an annual mean of 6.8 tortoises were killed during 2017–2021. The purposes of hunting were consumption (70%) and ethnomedicinal uses (30%) by the Meche and Sathar communities. Despite being protected under the law, illegal trade in tortoises and freshwater turtles is common in India, with different species being trafficked for different markets. Indian species of tortoises and hard-shell turtles are predominantly trafficked for the pet trade and soft-shell turtles for the meat trade. Given their distinct markets, the operation of trade may vary between these different groups of tortoises and freshwater turtles, thereby necessitating different types of interventions. However, a systematic examination of illegal trade in tortoises and freshwater turtles that takes into account the differences between these markets is currently lacking. Here we compare the supply networks of tortoises/hard-shell turtles (in demand for pet trade) vs soft-shell turtles (meat trade), using information from 78 and 64 seizures, respectively, that were reported in the media during 2013–2019. We used social network analysis to compare the two networks and the role of individual nodes (defined as locations at the district or city scale) within these networks. We found that the tortoise/hard-shell turtle network had a larger geographical scale, with more international trafficking links, than the soft-shell turtle network. We recorded convoluted smuggling routes in tortoise/hard-shell turtle trafficking, whereas soft-shell turtle trafficking was uni-directional from source to destination. Within both networks, we found that a few nodes played disproportionately important roles as key exporting, importing or transit nodes. Our study provides insights into the similarities and differences in the illegal supply networks of different groups of tortoises and freshwater turtles, in demand for different markets. In this study, we report the complete mitochondrial genome of the red-footed tortoise (Chelonoidis carbonarius). The compete mitochondrial genome of C. carbonarius is 16,639 bp in length, including 13 protein-coding genes, 22 tRNA genes, and two rRNA genes. The mitogenome was deposited in NCBI GenBank under the accession number OQ789392. Furthermore, we also constructed a phylogenetic tree of Chelonoidis using eight species. Freshwater turtles and tortoises are highly threatened by illegal and unsustainable wildlife trade. The trade has been thriving online, particularly on social media platforms, in recent decades. We conducted a two-year online survey on 20 Facebook groups in 2019-2020 and reviewed CITES trade data from 1990-2019 (30 yr) to elucidate chelonian trade dynamics in the Philippines. A total of 5,801 individuals representing 77 taxa were documented for sale on Facebook in 2019-2020. The 10 most commonly traded species represented 79% of the total quantity, and Centrochelys sulcata (n = 1,211) was the most offered for sale. The analysis of 307 CITES trade records showed that the Philippines imported between 3,365 (importer-reported) and 4,279 (exporter-reported) live chelonians of 39 species from 21 countries and territories during the 30-yr period. There were 23 CITES Appendix-listed species, including six species listed on Appendix I, documented for sale on Facebook but without import records from the CITES Trade Database such as Astrochelys radiata (n = 47), Astrochelys yniphora (n = 2), Geochelone platynota (n = 30), Geoclemys hamiltonii (n = 30), Pangshura tecta (n = 42), and Pyxis arachnoides (n = 3). Whereas the origins of these species were uncertain (e.g. pre-CITES convention; smuggled), some individuals may be poached from the wild, recently smuggled, and illegally offered in the country. Our results show that the wildlife trade continues to thrive on Facebook. Greater cooperation among conservation groups, wildlife authorities, and social media platforms and appropriate actions are urgently needed to curtail illegal online wildlife trade proliferation. Eating owl pellets by non-predator vertebrates is a rare and difficult-to-prove food chain relationship. In this paper, I reported the first record of a Spur-thighed Tortoise (Testudo graeca) eating a Eurasian Eagle-Owl (Bubo bubo) pellet with remains of a right Wood Pigeon (Columba palumbus) wing. Simple Summary: Anaesthesia in chelonians poses a challenge due to their resistance, making injectable anaesthesia highly valuable for this order of reptiles. In this study, we evaluated the effects of a dexmedetomidine–midazolam–ketamine–morphine (DMKM) combination delivered subcutaneously (SC) or intramuscularly (IM) in twenty-one Aldabra giant tortoises (Aldabrachelys gigantea) as a chemical restraint for a minimally invasive procedure for celioscopic sex identification. The intramuscular administration of a DMKM combination seems to be a good option to be used as a safe, reliable, short-lasting anaesthesia for minimally invasive procedures such as celioscopy. Sex identification through coelioscopy is a minimally invasive surgical technique used to determine the sex of chelonians by directly visualizing their internal reproductive organs. An adequate anaesthesiologic plan is essential to guarantee patient immobilization and proper analgesia during the entire surgical procedure. In this study, we evaluated the effects of a combination of dexmedetomidine (0.05 mg/kg), midazolam (1 mg/kg), ketamine (8 mg/kg), and morphine (1 mg/kg) (DMKM) randomly delivered intramuscularly (IM) or subcutaneously (SC) in twenty-one Aldabra giant tortoise (Aldabrachelys gigantea) into the right antebrachium for celioscopic sex identification. Heart rate (HR), respiratory rate (RR), and body temperature (BT) were measured, along with the skeletal muscle tone of the thoracic and pelvic limbs, neck retraction reflex, palpebral reflex, and jaw tone every 15 min. The anaesthesiologic plan was considered to be adequate at the loss of the thoracic and pelvic limb retraction reflexes. After a 45 min interval, if the anaesthetic plan was deemed insufficient for the celioscopic procedure, a 5 mg/kg dose of propofol was administered intravenously into the subcarapacial venous plexus. At the end of the procedure, atipamezole (0.5 mg/kg) and flumazenil (0.05 mg/kg) were administered intramuscularly into the left antebrachium as reversal agents. Both HR and RR decreased from baseline to both 15 and 30 min. Due to the persistence of thoracic and pelvic limb retraction reflexes 45 min after DMKM administration, 6/11 (55%) cases in the SC group required the additional administration of propofol, in contrast to only 1/10 (10%) cases in the IM group (p = 0.05). The recovery times were comparable between the successfully induced animals in the IM and SC groups. Although studies on Testudo hermanni boettgeri and T. graeca ibera have been conducted for more than a century in Bulgaria, little or no data have been published on some aspects of their ecology. Moreover, most of the local populations have still not been examined. The purpose of the study was to examine two local populations of the Hermann's and the Spur-thighed Tortoise occurring together. The study was carried out during two different periods from 2012 to 2023. Here we present data on the numbers, age structure, and sex ratio of the two populations. We also provide information on their home range and real and potential threats. The results indicated that T. hermanni boettgeri is much more numerous than T. graeca ibera. The age structure of both species was very similar - the adults predominated over subadults and juveniles. The sex ratio was slightly in favor of males in the Hermann's Tortoise, whereas it was female-biased in the Spur-thighed Tortoise. The mean home range of the Hermann's Tortoise was smaller compared to other populations of the species. Females had a larger home range than males. The mean home range of the Spur-thighed Tortoise was larger than that of the Hermann's Tortoise and similar to the home range of other populations on a species level. The biggest threat identified was related to partial deforestation within the habitat of the tortoises. The accuracy of posttranslocation population monitoring methods is critical to assessing long-term success in translocation programs. Translocation can produce unique challenges to monitoring efforts; therefore, it is important to understand the flexibility and robustness of commonly used monitoring methods. In Florida, USA, thousands of gopher tortoises Gopherus polyphemus have been, and continue to be, translocated from development sites to permitted recipient sites. These recipient sites create a broad range of potential monitoring scenarios due to variability in soft-release strategies, habitat conditions, and population demographics. Line transect distance sampling is an effective method for monitoring natural tortoise populations, but it is currently untested for translocated populations. We therefore produced 3,024 individual-based, spatially explicit scenarios of translocated tortoise populations that differed in recipient site and tortoise population properties, based on real-world examples, literature review, and expert opinion. We virtually sampled simulated tortoise populations by using line transect distance sampling methods and built a Bayesian hierarchical model to estimate the population density for each simulation, which incorporated individual-level covariates (i.e., burrow width and burrow occupancy). Line transect distance sampling was largely appropriate for the conditions that typify gopher tortoise recipient sites, particularly when detection probability on the transect lines was greater than or equal to 0.85. Designing the layout of transects relative to the orientation of soft-release pens, to avoid possible sampling biases that lead to extreme outliers in estimates of tortoise densities, resulted in more accurate population estimates. We also suggest that use of individual-level covariates, applied using a Bayesian framework as demonstrated in our study, may improve the applicability of line transect distance sampling surveys in a variety of contexts and that simulation can be a powerful tool for assessing survey design in complex sampling situations. Very large specimens of T. graeca ibera were found in Bulgaria, but mostly in the 20th century. Presently, such tortoises are almost absent in the country. Here we summarise data about the largest spur-thighed tortoises registered in Bulgaria and provide information about five new large-sized individuals. We also draw attention to the fact that large specimens can hardly be found in the country today and discuss some possible negative effects of the extinction of these specimens on the existing populations. The maximum straight carapace length of the largest T. graeca ibera ever found in Bulgaria was ≈ 389 mm, but the maximum straight carapace length of the newly measured tortoises was 298 mm. Considering the rarity of such tortoises in the country today, the genes that determine the potential to reach larger sizes may gradually disappear. Thus, the institutions responsible for the conservation of nature should pay attention to areas where the presence of large-sized individuals has been established. This might help preserve the natural genome of T. graeca ibera in the country and therefore the existence of large individuals.

Our study was focused on the external features differentiation of two adult species of turtles, including a land tortoise (Testudo graeca graeca) and a freshwater turtle (Trachemys scripta elegans), by gross morphological description of head, carapace, plastron, and tail. Generally, turtles show gross morphological differences according to the ecology and the environment. The carapace and legs are the fascinating noticeable parts showing significant differences between tortoise and turtle as T. graeca graeca shows a small carapace length with noticeable convexity. In contrast, the freshwater turtle has a flattened broad carapace adapted to swimming. At the same time, the legs of T. graeca show multiple keratinized spurs with rigid claws, but T. scripta has legs covered with soft, moist skin and thin claws. The plastron is relatively the same in both species, while the head is unique with a large size and red flap in T. scripta. We conclude that the flat body and legs with broad head are a modification for swimming, while the rigid claws with keratinized outer leg skin are signs of land walking adaptation. Roadways and railways can reduce wildlife movements across landscapes, negatively impacting population connectivity. Connectivity may be improved by structures that allow safe passage across linear barriers, but connectivity could be adversely influenced by low population densities. The Mojave desert tortoise is threatened by habitat loss, fragmentation, and population declines. The tortoise continues to decline as disturbance increases across the Mojave Desert in the southwestern United States. While underground crossing structures, like hydrological culverts, have begun receiving attention, population density has not been considered in tortoise connectivity. Our work asks a novel question: How do culverts and population density affect connectivity and potentially drive genetic and demographic patterns? To explore the role of culverts and population density, we used agent-based spatially explicit forward-in-time simulations of gene flow. We constructed resistance surfaces with a range of barriers to movement and representative of tortoise habitat with anthropogenic disturbance. We predicted connectivity under variable population densities. Simulations were run for 200 non-overlapping generations (3400 years) with 30 replicates using 20 microsatellite loci. We evaluated population genetic structure and diversity and found that culverts would not entirely negate the effects of linear barriers, but gene flow improved. Our results also indicated that density is important for connectivity. Low densities resulted in declines regardless of the landscape barrier scenario (> 75% population census size, > 97% effective population size). Results from our simulation using current anthropogenic disturbance predicted decreased population connectivity over time. Genetic and demographic effects were detectable within five generations (85 years) following disturbance with estimated losses in effective population size of 69%. The pronounced declines in effective population size indicate this could be a useful monitoring metric. We suggest management strategies that improve connectivity, such as roadside fencing tied to culverts, conservation areas in a connected network, and development restricted to disturbed areas. The threatened Mojave desert tortoise (Gopherus agassizii) exhibits temperature-dependent sex determination, and individuals appear externally sexually monomorphic until sexual maturity. A non-surgical sex identification method that is suitable for a single in situ encounter with hatchlings is essential for minimizing handling of wild animals. We tested (1) whether plasma testosterone quantified by enzyme-linked immunosorbent assay differentiated males from females in 0–3 month old captive hatchlings, and (2) whether an injection of follicle-stimulating hormone (FSH) differentially elevates testosterone in male hatchlings to aid in identifying sex. We validated sex by ceolioscopic (laparoscopic) surgery. We then fit the testosterone concentrations to lognormal distributions and identified the concentration below which individuals are more likely female, and above which individuals are more likely male. Using a parametric bootstrapping procedure, we estimated a 0.01–0.04% misidentification rate for naïve testosterone samples, and a 1.26–1.39% misidentification rate for challenged (post-FSH injection) testosterone samples. Quantification of plasma testosterone concentration from small volume (0.1 mL) blood samples appears to be a viable, highly accurate method to identify sex of 0–3 month old hatchlings and could be a valuable tool for conservation measures and investigation of trends and variation in sex ratios for in situ wild nests. Multidisciplinary approaches for the conservation of endangered species have great importance in preparing management plans. In addition to ecological and population demographic parameters, genetic data provide vital information for conservation management plans for a species. The Hermann's tortoise (Testudo hermanni) suffers from illegal trade, fires, habitat fragmentation and destruction, and therefore, the genetic diversity of the regional populations has been affected. It was aimed herein to impart knowledge on the population genetic structure of T. hermanni boettgeri, listed as near threatened by the International Union for Conservation of Nature Red List as a subspecies and has limited distribution in European Türkiye. A study was conducted of 15 microsatellite loci of 221 tortoises from 15 different localities in the Thrace region in Türkiye. All of the examined loci were polymorphic, and the number of alleles varied from 2 to 13. The quantity of private alleles (Pa) at the localities ranged from 0 to 6. The average gene diversity was 0.31 (range: 0.25-0.38). The highest levels of allelic richness, private alleles, and genetic diversity (Ar, Pa, He) were observed at localities 3 and 7, close to each other. The total population (p < 0.001) and 12 of the 15 studied localities diverged from the Hardy--Weinberg equilibrium. Of the 15 localities studied, 6 had significantly different inbreeding coefficients. Furthermore, a 2-phased model of mutation (TPM) (p < 0.001) detected a recent bottleneck in the population. We observed out‐of‐crevice behaviours on seven pancake tortoises (Malacochersus tornieri Siebenrock, 1903) at six sites in the Tarangire–Manyara Ecosystem. Most out‐of‐crevice activities were observed during the wet season and late morning, which included basking, walking, foraging and suspected courtship fighting. Predation was evidenced on six live tortoises and two carcasses, with suspects being mesocarnivores, birds and lizards. Future studies should understand the effects of seasonal resource availability on behaviours and life history aspects, as well as predators and predation rates experienced by pancake tortoises. Herpesviruses can be significant reptile pathogens. Herpesviral infection in a wild-caught, male spider tortoise (Pyxis arachnoides) under human care was detected during a routine wellness examination prior to transition between zoologic organizations. The tortoise had no clinical signs of illness. Oral swabs obtained during a physical examination as part of pre-shipment risk mitigation for infectious disease were submitted for consensus herpesvirus PCR assay and sequencing. Based on comparative sequence analysis, the novel herpesvirus identified is a member of the subfamily Alphaherpesvirinae. Studies of herpesviral phylogeny in chelonian species support branching patterns of turtle herpesviruses that closely mirror those of their hosts. The symmetry of these patterns is suggestive of close codivergence of turtle herpesviruses with their host species. The distribution of these viruses in both tortoises and emydids suggests a phylogenetic duplication event in the herpesviruses after host divergence of the Pleurodira and basal to the divergence of Americhelydia. Herpesviral infections have been documented to cause higher morbidity when introduced to aberrant host species, and significant consideration must be given to the presence of herpesviruses in the management of tortoise collections, particularly collections that include various species of testudines. Turtles and tortoises are one of the world's most threatened vertebrates; more than half of the 352 currently recognised species are threatened. To implement the IUCN CPSG's One Plan Approach to Conservation, we herein analyse the available information from the Zoological Information Management System (ZIMS) to provide an overview of the species already held in zoos. A total of 252 species (71.6%) are currently kept in ZIMS institutions, with 138 of them listed as threatened (76.7% of all threatened turtles). Additional 26 (15 threatened) species are listed in the database Zootierliste (Zoo Animal List). Zoos keep 152 (84.4%) of 180 threatened and 110 out of 150 not threatened species and show a preference for keeping threatened species. Concerning threatened turtle species, nine are represented with more than 500 individuals in zoo collections, while 25 are only kept as single individuals or in same-sex-groups. More than half of the held species are only represented in one to ten zoos. Most species are kept in North America, Europe and Asia, where most of the ZIMS institutions are located. A total of 92 (59 threatened) species (37.1 % of all zoo-kept species) were successfully bred in 140 zoos (15.8 % of 888 ZIMS-institutions keeping turtles) in the last 12 months. There already exists a tendency towards breeding threatened species. Still, zoos could improve both conservation breeding networking and establish further conservation breeding programs to create reserve populations, which would be in broad fulfilment with the IUCN's One Plan Approach to Conservation. Online sales are increasingly a route by which exotic animals are sold in the global pet trade. There are numerous types of online platforms and transaction types, and dedicated classified advertisement sites are a popular means of buying and selling animals. Despite their large and increasing use, we have a relatively poor understanding of the number of, and taxonomic variation in, the animals sold online. This information may be key in efforts to optimise the welfare of the animals being sold, and the ethics and sustainability of the trade via that platform. To fill this knowledge gap, we monitored and analysed the advertisements of chelonians (turtles and tortoises) placed on one of the United Kingdom's largest dedicated classified ads sites, www.pets4homes.co.uk, over the course of a year, from July 2020 until June 2021. We analysed temporal, taxonomic, and advertiser related trends in the volumes of advertisements placed and compared the prices and the sentiment of language within adverts for different species. We found that the species advertised, the prices requested, and infrequent use of the site by most advertisers is consistent with most adverts being for animals being resold by casual users. Further, we found that turtles were consistently advertised for lower prices and in multiples than tortoises, and that the language with which they were advertised was less positive. We conclude that on this website the online trade reflects the broader trade, rather than drives the sales of chelonians in the UK, and that any interventions aiming to improve welfare and sustainability would be better placed earlier in the supply chain. Simple Summary: Animal health is directly linked to viability of the population, which may be affected by anthropogenic activities and diseases. Biomarkers such as serum chemistry and parameters of oxidative balance are good indicators of an overall biological status, providing information on the effects of contaminants on the organism. The objective of this work was to analyze biochemical and molecular indicators and their correlations to biogenic and risk elements in Testudo hermanni. Biochemical parameters were analyzed using the commercial kit DiaSys and biochemical analyzer Randox RX Monza. Sodium, potassium, and chlorides were measured using the EasyLite analyzer. Oxidative stress was evaluated using colorimetric and luminometric methods. Quantification of chemical elements in the animal blood was carried out using inductively coupled plasma mass spectrometry. Biochemical values of analyzed samples from Hermann's tortoises were almost the same as referential values described by multiple authors, with minor aberrations in the total protein parameter. Values of arsenic (As) and nickel (Ni) showed correlation with biochemical parameters and the parameters of oxidative stress. Cadmium (Cd) exhibited correlation with the biochemical parameter aspartate aminotransferase (AST). Finally, this study detected heavy metals and their significant correlations with selected biochemical and molecular parameters in Hermann's tortoises. Trophic rewilding is increasingly being used to promote megafauna reintroductions to island ecosystems, yet ecosystem response to population restoration once megafauna reintroduction occurs remains understudied. In this study of a population of Galapagos giant tortoises reintroduced to an arid island, tortoise exclosures monitored over an 8‐year‐long period revealed that, in response to the presence of tortoises, herbaceous plant cover and numbers of regenerating woody plants decreased, whereas extent of grass cover increased. Vegetation mapping over a 15‐year‐long period across the island indicated a threshold density of 1–2 tortoises per hectare halted incursion of woody plants and triggered a shift in this savannah‐type ecosystem toward more grasses. Restoration of this giant tortoise population has shaped plant communities at both local and landscape scales with cascading effects on many components of biodiversity on the island. There are more species of lizards and snakes (squamates) alive today than any other order of land vertebrates, yet their fossil record has been poorly documented compared with other groups. Here, we describe a gigantic Pleistocene skink from Australia based on extensive material that includes much of the skull and postcranial skeleton, and spans ontogenetic stages from neonate to adult. Tiliqua frangens substantially expands the known ecomorphological diversity of squamates. At approximately 2.4 kg, it was more than double the mass of any living skink, with an exceptionally broad, deep skull, squat limbs and heavy, ornamented body armour. It probably filled the armoured herbivore niche that land tortoises (testudinids), absent from Australia, occupy on other continents. Tiliqua frangens and other giant Plio-Pleistocene skinks suggest that small-bodied groups that dominate vertebrate biodiversity might have lost their largest and often most morphologically extreme representatives in the Late Pleistocene, expanding the scope of these extinctions. Mojave desert tortoises (Gopherus agassizii), a threatened species under the US Endangered Species Act, are long‐lived reptiles that experience a chronic respiratory disease. The virulence of primary etiologic agent, Mycoplasma agassizii, remains poorly understood, but it exhibits temporal and geographic variability in causing disease outbreaks in host tortoises. Multiple attempts to culture and characterize the diversity of M. agassizii have had minimal success, even though this opportunistic pathogen chronically persists in nearly every population of Mojave desert tortoises. The current geographic range and the molecular mechanisms of virulence of the type‐strain, PS6T, are unknown, and the bacterium is thought to have low‐to‐moderate virulence. We designed a quantitative polymerase chain reaction (qPCR) targeting three putative virulence genes annotated on the PS6T genome as exo‐α‐sialidases, enzymes which facilitate growth in many bacterial pathogens. We tested 140 M. agassizii‐positive DNA samples collected from 2010 to 2012 across the range of Mojave desert tortoises. We found evidence of multiple‐strain infections within hosts. We also found the prevalence of these sialidase‐encoding genes to be highest in tortoise populations surrounding southern Nevada, the area from which PS6T was originally isolated. We found a general pattern of loss or reduced presence of sialidase among strains, even within a single host. However, in samples that were positive for any of the putative sialidase genes, one particular gene (528), was positively associated with bacterial loads of M. agassizii and may act as a growth factor for the bacterium. Our results suggest three evolutionary patterns: (1) high levels of variation, possibly due to neutral changes and chronic persistence, (2) a trade‐off between moderate virulence and transmission, and (3) selection against virulence in environmental conditions known to be physiologically stressful to the host. Roads are important drivers of habitat loss, degradation, and fragmentation that affect global biodiversity. Detrimental effects of roads include direct mortality of individual animals, spread of habitat-altering invasive plants, and loss of demographic and genetic connectivity of wildlife populations. Various measures address the negative effects of roads on wildlife. However, most strategies for minimizing or mitigating the effects of roads are focused on the actual roads themselves rather than on the collective travel network across landscapes. We summarized a growing body of literature that has documented the effects of road density on wildlife populations and the benefits associated with lower densities. This literature supports the application of limits on road density as a viable tool for managing cumulative effects. Based on these examples, we recommend road densities, including all linear features used for travel, of less than 0.6 km/km2 as a general target for travel management in areas where wildlife conservation is a priority. Lower densities may be necessary in particularly sensitive areas, whereas higher densities may be appropriate in areas less important to landscape-level conservation and wildlife connectivity. Public policy and funding also are needed to address challenges of enforcing off-highway vehicle regulations. In applying this general overview to a case study of the Mojave desert tortoise Gopherus agassizii, we found that all management plans across the species' range lack considerations of road density and that tortoise populations declined within all conservation areas with road densities of more than 0.75 km/km2. From this, we provide several travel management recommendations specific to Mojave desert tortoise conservation beginning with identifying the entire travel network within management areas. Specific actions for managing or setting limits for road density depend on the site-specific biological or management context, for instance relative to habitat quality or proximity to designated tortoise conservation areas. In addition, increasing law enforcement and public outreach will improve enforcement and compliance of travel regulations, and installing tortoise-exclusion fencing along highways will reduce road kills and allow tortoise populations to reoccupy depleted areas adjacent to highways.
