Abstract

Unmanned aerial vehicles (UAVs) play a crucial role in various domains such as military, civil, industrial, and so on. However, the coordination and task allocation of multiple UAVs in engineering practices face numerous challenges. As the scale of battlefields expands, and the diversity of UAV missions and constraints increases, the existing task allocation methods suffer from issues such as a mismatch between theoretical models and real‐world applications, low task execution efficiency, and poor responsiveness in dynamic environments. To address these challenges, this paper proposes an improved genetic algorithm (GA)‐based approach for multi‐UAV cooperative task allocation. By collecting battlefield information, decomposing tasks, and considering UAV resource types, an optimization model for multi‐UAV cooperative task allocation is constructed. The proposed method, using an improved GA, generates a set of Pareto‐optimal task solutions for decision‐makers. Case studies demonstrate that this approach effectively enhances task execution efficiency and reduces the total flight distance cost of UAVs. [ABSTRACT FROM AUTHOR]

Abstract

Human operating error is one of the important factors affecting the reliable operation of unmanned aerial vehicle (UAV) components, however, people always neglect human-operating-error of the existing unmanned aircraft systems and components reliability assessment methods. This article set the problem to the most important position to search, divided the scene of UAV components assignment to the Status debugging monitoring model and Preventive maintenance type. First, analysis of two scenarios UAV components made qualitative analysis of human error using the method of failure mode and effect analysis; and then put forward the two scenarios of human reliability assessment model, used to quantify the probability of human error. In example analysis, it considers the primary factors those affect humans reliability, to calculate and compare the proportion of human error at different situation. Results show that the proposed method can quantify human error probability, and quantitative characterization of human error can affect the reliability of the UAV components.

Abstract

Summary Chromatin immunoprecipitation and sequencing (vs ChIP‐seq) is an essential tool for epigenetic and molecular genetic studies. Although being routinely used, ChIP‐seq is expensive, requires grams of plant materials, and is challenging for samples that enrich fatty acids such as seeds. Here, we developed an Ultrasensitive Plant ChIP‐seq (UP‐ChIP) method based on native ChIP‐seq combined with Tn5 tagmentation‐based library construction strategy. UP‐ChIP is generally applicable for profiling both histone modification and Pol II in a wide range of plant samples, such as a single Arabidopsis seedling, a few Arabidopsis seeds, and sorted nuclei. Compared with conventional ChIP‐seq, UP‐ChIP is much less labor intensive and only consumes 1 μg of antibody and 10 μl of Protein‐A/G conjugated beads for each IP and can work effectively with the amount of starting material down to a few milligrams. By performing UP‐ChIP in various conditions and genotypes, we showed that UP‐ChIP is highly reliable, sensitive, and quantitative for studying histone modifications. Detailed UP‐ChIP protocol is provided. We recommend UP‐ChIP as an alternative to traditional ChIP‐seq for profiling histone modifications and Pol II, offering the advantages of reduced labor intensity, decreased costs, and low‐sample input. Abstract

Wheat is one of the major sources of food for much of the world. However, because bread wheat's genome is a large hybrid mix of three separate subgenomes, it has been difficult to produce a high-quality reference sequence. Using recent advances in sequencing, the International Wheat Genome Sequencing Consortium presents an annotated reference genome with a detailed analysis of gene content among subgenomes and the structural organization for all the chromosomes. Examples of quantitative trait mapping and CRISPR-based genome modification show the potential for using this genome in agricultural research and breeding. Ramírez-González et al. exploited the fruits of this endeavor to identify tissue-specific biased gene expression and coexpression networks during development and exposure to stress. These resources will accelerate our understanding of the genetic basis of bread wheat.

Abstract

The yellow seed characteristic in Brassica napus L. is desirable because of its association with higher oil content and better quality of oil-extracted meal. YN01-429 is a yellow-seeded canola-quality germplasm developed in Canada arising from several years of research. Seed-coat pigmentation is due to oxidized proanthocyanidins (PA; condensed tannins) derived from phenylpropanoids and malonyl CoA. We found PA accumulation to be most robust in young seed coats (20 d post anthesis; dpa) of a related black-seeded line N89-53 and only very little PA in YN01-429, which also contained much less extractable phenolics. The flavonol content, however, did not show as great a difference between these two lines. Furthermore, sinapine, a product of the general phenylpropanoid metabolism, was present at comparable levels in the embryos of both lines. Dihydroflavonol reductase (DFR) activity that commits phenolics to PA synthesis was lower in YN01-429 seed coats. The results of Southern blot and in silico analyses were indicative of two copies of the DFR gene in B. napus. Both copies were functional in YN01-429, ruling out homeoallelic repression or silencing, but together they showed very low expression levels (17-fold fewer transcripts) relative to DFR activity in N89-53 seed coats. These results collectively suggest that YN01-429 differs in regulatory circuits that impact the PA synthesis branch much more than the flavonol synthesis branch in the seed coats and such circuits do not impinge upon general phenylpropanoid metabolism in the embryos.

Abstract

A mitochondrial glycerol-3-phosphate (G-3-P) shuttle that channels cytosolic reducing equivalent to mitochondria for respiration through oxidoreduction of G-3-P has been extensively studied in yeast and animal systems. Here, we report evidence for the operation of such a shuttle in Arabidopsis thaliana. We studied Arabidopsis mutants defective in a cytosolic G-3-P dehydrogenase, GPDHc1, which, based on models described for other systems, functions as the cytosolic component of a G-3-P shuttle. We found that the gpdhc1 T-DNA insertional mutants exhibited increased NADH/NAD+ ratios compared with wild-type plants under standard growth conditions, as well as impaired adjustment of NADH/NAD+ ratios under stress simulated by abscisic acid treatment. The altered redox state of the NAD(H) pool was correlated with shifts in the profiles of metabolites concerning intracellular redox exchange. The impairment in maintaining cellular redox homeostasis was manifest by a higher steady state level of reactive oxygen species under standard growth conditions and by a significantly augmented hydrogen peroxide production under stress. Loss of GPDHc1 affected mitochondrial respiration, particularly through a diminished capacity of the alternative oxidase respiration pathway. We propose a model that outlines potential involvements of a mitochondrial G-3-P shuttle in plant cells for redox homeostasis.
